10. Microvesicles: ROS scavengers and ROS producers

Abstract

This review analyzes the relationship between microvesicles and reactive oxygen species (ROS). This relationship is bidirectional; on the one hand, the number and content of microvesicles produced by the cells are affected by oxidative stress conditions; on the other hand, microvesicles can directly and/or indirectly modify the ROS content in the extra- as well as the intracellular compartments. In this regard, microvesicles contain a pro-oxidant or antioxidant machinery that may produce or scavenge ROS: direct effect. This mechanism is especially suitable for eliminating ROS in the extracellular compartment. Endothelial microvesicles, in particular, contain a specific and well-developed antioxidant machinery. On the other hand, the molecules included in microvesicles can modify (activate or inhibit) ROS metabolism in their target cells: indirect effect. This can be achieved by the incorporation into the cells of ROS metabolic enzymes included in the microvesicles, or by the regulation of signaling pathways involved in ROS metabolism. 18. In vitro effect of albendazole and albendazole-sulphoxide on 70 and 60 KDa heat shock proteins in Echinococcus granulosus protoscolices

Abstract

Las proteínas del estrés o Heat Shock Proteins (HSPs) (HSP70 y HSP60) de protoescolex de Echinococcus granulosus fueron estudiadas mediante inmunohistoquimica tras incubación in vitro con albendazol, albendazol sulfóxido o la combinación de ambos, y bajo tratamiento térmico. La especificidad de los anticuerpos fue comprobada mediante inmunoblotting. Se observó un descenso en la inmunoreactividad de la HSP60 y HSP70 tras la incubación con los antihelminticos. Por el contrario, los protoescolex incubados con tratamiento hipertérmico mostraron una mayor inmunoreactividad en ambas proteínas. Heat shock proteins (HSP70 and HSP60) were immunohistochemically studied in Echinococcus granulosus protoscolices incubated in vitro with albendazole, or albendazole sulphoxide, or both, and under hyperthermic conditions. Antibody specifity against E. granulosus protoscolices was tested by immunoblotting technique. A significant loss of HSP70 and HSP60 immunoreactivity was observed after the benzimidazole carbamate treatment. In vitro hyperthermic treatments of E. granulosus protoscolices exhibited higher immunoreactivity levels than control protoscolices. Fil: Perez Serrano, J.. Abstract

Abstract The response of astroglial cells in the hippocampus to long-term portacaval anastomosis (PCA), an experimental model of hepatic encephalopathy, was studied in adult male rats and compared with controls. Six months after PCA, the rat hippocampi were processed for glial fibrillary acidic protein (GFAP). GFAP-immunopositive astroglial profiles were observed in all hippocampal layers in PCA rats, but GFAP distribution differed in PCA rats and controls. In PCA rats, cell bodies and cell processes immunostained with GFAP were observed mainly in the CA1-CA3 layers in relation to pyramidal neurons, whereas the number and length of the astroglial processes decreased in the rest of the hippocampal regions. In addition, decreased GFAP immunoreactivity in the perivascular processes was observed in PCA rats compared with controls. These results show that long-term PCA elicited different responses in GFAP expression in different hippocampal regions, which might suggest a regional variation in glial sensitivity.
