Abstract Background Regorafenib, a multi-targeted kinase inhibitor, has been used in the treatment of Hepatocellular carcinoma (HCC). The purpose of this study is to investigate the mechanism of Regorafenib in HCC. Methods Regorafenib’s impact on the sensitivity of HCC cells was assessed using CCK8. Differential gene expression analysis was performed by conducting mRNA sequencing after treatment with Regorafenib. The m6A methylation status of CHOP and differential expression of m6A methylation-related proteins were assessed by RIP and Western Blot. To explore the molecular mechanisms involved in the therapeutic effects of Regorafenib in HCC and the impact of METTL14 and CHOP on Regorafenib treatment, we employed shRNA/overexpression approaches to transfect METTL14 and CHOP genes, as well as conducted in vivo experiments. Results Treatment with Regorafenib led to a notable decrease in viability and proliferation of SK-Hep-1 and HCC-LM3 cells. The expression level of CHOP was upregulated after Regorafenib intervention, and CHOP underwent m6A methylation. Among the m6A methylation-related proteins, METTL14 exhibited the most significant downregulation. Mechanistic studies revealed that Regorafenib regulated the cell cycle arrest in HCC through METTL14-mediated modulation of CHOP, and the METTL14/CHOP axis affected the sensitivity of HCC to Regorafenib. In vivo, CHOP enhanced the anticancer effect of Regorafenib. Conclusion The inhibition of HCC development by Regorafenib is attributed to its modulation of m6A expression of CHOP, mediated by METTL14, and the METTL14/CHOP axis enhances the sensitivity of HCC to Regorafenib. These findings provide insights into the treatment of HCC and the issue of drug resistance to Regorafenib.

Abstract N 6-methyladenosine (m6A) is an RNA modification that can be removed by demethylases [fat mass and obesity-associated protein (FTO) and AlkB homolog 5 (ALKBH5)], which regulate gene expression and cell function. We show that m6A levels and m6A demethylase levels are altered in nasopharyngeal carcinoma (NPC) tissues vs. normal tissues. High FTO and ALKBH5 predict a poor prognosis in NPC patients. Silencing FTO and ALKBH5 inhibited the malignant behavior of patient-derived NPC cells in a short time. However, as time progressed, the inhibitory effect of FTO or ALKBH5 was weakened, and the cosilencing of FTO and ALKBH5 maintained a better inhibitory effect. Combined transcriptome and m6A-seq analysis revealed a downstream target gene that was jointly regulated by FTO and ALKBH5 in NPC, and ARHGAP35 was chosen to do further study. The synergistic silencing of FTO and ALKBH5 increased the methylation level on the mRNA CDS of a new transcription factor (ARHGAP35) and positively regulate the protein coding capacity and mRNA stability of ARHGAP35, thus leading to increased expression of ARHGAP35 and inhibition of the malignant phenotype of tumor cells. Our study revealed that the growth and metastasis of NPC can be stably inhibited through synergistic silencing of the demethylases FTO and ALKBH5, which play a positive role in the treatment of NPC by regulating the downstream transcript ARHGAP35 and increasing its m6A level.

Due to the heavy burden on human drivers when remotely controlling hexapod robots in complex terrain environments, there is a critical need for robot intelligence to assist in generating control commands. Therefore, this study proposes a mapping process framework that generates a combination of human-robot commands based on decision target values, focusing on the task of robot intelligence assisting drivers in generating human-robot command combinations. Furthermore, human-robot state constraints are quantified as geometric constraints on robot motion and driver fatigue constraints. By optimizing and filtering the feasible set of human-robot commands based on human-robot state constraints, instruction combinations are formed and recommended to the driver in real-time, thereby enhancing the efficiency and safety of human-machine coordination. To validate the effectiveness of the proposed method, a remote human-robot collaborative driving control system based on wearable devices is designed and implemented. Experimental results demonstrate that drivers utilizing the human-robot command recommendation system exhibit significantly improved robot walking stability and reduced collision rates compared to individual driving.

Atmospheric volatile organic compounds (VOCs), such as olefins and aromatics, released from synthetic chemical pesticide sprays can increase regional air pollution, public health risks, and food security risks. However, significant uncertainties remain regarding the measurement methods and chemical profiles of VOC emissions. Using an agricultural city, Changchun City in Northeast China, as a case study, we quantified real-time concentration and composition data based on online monitoring instruments for the year 2023. This study optimized data collection methods for emission factors and activity levels and developed a high-precision emission inventory of VOCs in pesticides at the city scale. The emission factors for VOCs from the seven categories of pesticides were estimated as follows: 78 g/kg (nicosulfuron and atrazine, oil-dispersible [OD] and suspension emulsion [SE], respectively), 4 g/kg (chlorpyrifos and indoxair conditioningarb, suspension concentrate [SC]), 5 g/kg (fluopicolide and propamocarb hydrochloride, SC), 217 g/kg (MCPA-dimethylammonium, aqueous solution [AS]), 34 g/kg (glyphosate, AS), 575 g/kg (beta-cypermethrin and malathion, emulsifiable concentrate [EC]), and 122 g/kg (copper abietate, emulsion in water [EW]), depending on the pesticide formulation components and formulation types. The orchard insecticide exhibited the highest emission factors among all pesticides owing to its emulsifiable concentrate formulation and 80% content of inactive ingredients (both factors contribute to the high content of organic solvents in the pesticide). The major components of VOC emissions from pesticide spraying were halocarbons (27–44%), oxygenated VOCs (OVOCs) (25–38%), and aromatic hydrocarbons (15–28%). The total VOC emissions from pesticide spraying in the Changchun region accounted for 10.6 t, with Yushu City contributing 28% of the VOC emissions and Gongzhuling City and Dehui City contributing 18.7% and 16.0%, respectively. Herbicides were the main contributors to VOC emissions because of their high emission factors and extensive use in fields (used for spraying maize and rice, the main crops in Changchun City). May and June exhibited the highest VOC emissions from pesticide application, with May accounting for 57.0% of annual pesticide emissions, predominantly from herbicides (95.1%), followed by insecticides (4.9%). June accounted for 30.1% of the annual pesticide emissions, with herbicides being the largest contributor of VOC emissions. An emission inventory of VOC with a monthly scale and spatial grid resolutions of 0.083° and 0.5° in 2023 was developed. These emission factors and inventories of pesticide applications provide valuable information for air quality modeling. This study also provides an important scientific basis for enhancing regional air quality and mitigating the environmental impact of pesticide use in major grain-producing areas.

Abstract Distant metastasis is currently the main factor affecting the prognosis of nasopharyngeal carcinoma (NPC), and understanding the mechanisms of metastasis and identifying reliable therapeutic targets are critical for improving prognosis and achieving clinical translation. Macrophages, as important immune cells in the tumor microenvironment (TME), have been shown to regulate metastasis. And extracellular vesicles (EVs) secreted by stromal cells and tumor cells play the important role in intercellular communication in the tumor microenvironment. However, the role of NPC-EVs on macrophages and their function in regulating macrophages to affect metastasis has not been fully clarified. In this study, we report that NPC-EVs can be uptake by macrophages and alter macrophage polarization, for the first time, we identified the genes implicated in these regulatory functions: SCARB1, HAAO, and CYP1B1. Moreover, we found that SCARB1 was positively associated with metastasis and poor prognosis of NPC. Interestingly, we found that SCARB1-rich EVs promoted M1 macrophages ferroptosis to decrease M1 macrophages infiltration by upregulating the HAAO level while decreasing phagocytosis of M2 macrophages by upregulating the CYP1B1 level. Finally, we identified the SCARB1-binding gene KLF9, which is involved in the transcription of HAAO and CYP1B1. Our findings showed that SCARB1-EVs promoted metastasis by co-regulating M1 and M2 macrophage function. The related mechanism will provide a new therapeutic strategy to help patients with NPC improve their prognosis.

Highlights The crystal facets featured with facet-dependent physical and chemical properties can exhibit varied electrocatalytic activity toward hydrogen evolution reaction (HER) and oxygen evolution reaction (OER) attributed to their anisotropy. The highly active exposed crystal facets enable increased mass activity of active sites, lower reaction energy barriers, and enhanced catalytic reaction rates for HER and OER. The formation mechanism and control strategy of the crystal facet, significant contributions as well as challenges and perspectives of facet-engineered catalysts for HER and OER are provided.

Improving fuel cell technologies based on Pt-based alloys is important for efficient fuel cells. Here, the authors report a hybrid PtCo alloy electrocatalyst for acidic oxygen reduction at high current densities and H2/air fuel cell power densities.

Interictal epileptiform spike (referred to as spike) detected from electroencephalograms lasting only 20- to 200-ms can provide a reliable evidence-based indicator for clinical seizure type diagnosis. Recent feature representation approaches focus either on the concrete-level or abstract-level information mining of the spike, thus demonstrating suboptimal detection performance. Additionally, existing abstract-level information mining methods of the spike based deep learning networks have not realized the effective feature representation of long-term dependent distinguished information within similar waveform cycles caused by morphological heterogeneity, which affects detection performance. Thus, a multilevel feature learning method for accurate spike detection was proposed in this study. Specifically, the spatio-temporal-frequency multidomain information in concrete-level first are inferred the common mimetic properties of the spike using the multidomain feature extractors. Then, the effective feature representation of long-term dependent distinguished information within similar waveform cycles caused by morphological heterogeneity is suitably captured using the temporal convolutional network. Finally, the spatio-temporal-frequency multidomain long-term dependent feature representation of spike is calculated using the element-wise manner to fuse the feature representation in concrete- and abstract-levels. The experimental results indicate that the proposed method can achieve an accuracy of 90.62±1.38%, sensitivity of 90.38±1.52%, specificity of 91.00±1.60%, precision of 90.33±4.71%, and the false detection rate per minute is $0.148\pm 0.020\text{m}^{-1}$ , which are higher than when using the feature representation in the concrete-or abstract-level alone. Additionally, the detection results indicate that the proposed method avoids the subjectivity and inefficiency of visual inspection, and it enables a highly accurate detection of the spike.

Summary: The nasal polyps (NPs) microenvironment comprises multiple cell types, including mesenchymal stromal cells (MSCs). Insulin-like growth factor binding protein 2 (IGFBP2) plays crucial roles in cell proliferation, differentiation and more. However, the role of NPs-derived MSCs (PO-MSCs) and IGFBP2 in NPs pathogenesis remains poorly defined. Herein, primary human nasal epithelial cells (pHNECs) and MSCs were extracted and cultured. Extracellular vesicles (EVs) and soluble proteins were isolated to investigate the role of PO-MSCs on epithelial-mesenchymal transition (EMT) and epithelial barrier function in NPs. Our data showed that IGFBP2, but not EVs from PO-MSCs (PO-MSCs-EVs), exhibited a crucial role in EMT and barrier destruction. Moreover, focal adhesion kinase (FAK) signaling pathway is necessary for IGFBP2 to exert its functions in human and mice nasal epithelial mucosa. Altogether, these findings may improve the current understanding of the role of PO-MSCs in NPs microenvironment and ultimately contribute to the prevention and treatment of NPs.

Abstract Objectives We aimed to construct an induction system for polyploid giant cancer cells (PGCCs), as well as to investigate PGCC features and clinical significance. Methods A laryngeal neoplasm‐PGCC induction system was constructed using paclitaxel liposomes (PTX). We used western blots to compare expression of epithelial‐mesenchymal transition‐related proteins, stem cell interrelated proteins, and cyclin‐associated proteins. We then measured PGCC count in tissue samples of patients with laryngeal neoplasms and analyzed its relationship with prognosis. Statistical significance was determined using t‐tests. Results PTX successfully induced PGCCs. Western blotting showed that CyclinB1, CDC25C, CDK1, E‐cadherin, and EIF‐4A expression decreased in PGCCs compared with normal cancer cells, whereas vimentin and CD133 expression increased. Abstract The design and production of active, durable, and nonprecious electrocatalysts toward alkaline hydrogen oxidation and evolution reactions (HOR/HER) are extremely appealing for the implementation of hydrogen economy, but remain challenging. Here, we report a facile electric shock synthesis of an efficient, stable, and inexpensive NiCoCuMoW multi‐element alloy on Ni foam (NiCoCuMoW) as a bifunctional electrocatalyst for both HOR and HER. For the HOR, the current density of NiCoCuMoW could reach ∼11.2 mA cm–2 when the overpotential is 100 mV, higher than that for commercial Pt/C (∼7.2 mA cm–2) and control alloy samples with less elements, along with superior CO tolerance. Moreover, for the HER, the overpotential at 10 mA cm−2 for NiCoCuMoW is only 21 mV, along with a Tafel slope of low to 63.7 mV dec−1, rivaling the commercial Pt/C as well (35 mV and 109.7 mV dec−1). Density functional theory calculations indicate that alloying Ni, Co, Cu, Mo, and W can tune the electronic structure of individual metals and provide multiple active sites to optimize the hydrogen and hydroxyl intermediates adsorption, collaboratively resulting in enhanced electrocatalytic activity.

Gaseous organic compounds, mainly volatile organic compounds (VOCs), have become a wide concern in various indoor environments where we spend the majority of our daily time. The sources, compositions, variations, and sinks of indoor VOCs are extremely complex, and their potential impacts on human health are less understood. Owing to the deployment of the state-of-the-art real-time mass spectrometry during the last two decades, our understanding of the sources, dynamic changes and chemical transformations of VOCs indoors has been significantly improved. This review aims to summarize the key findings from mass spectrometry measurements in recent indoor studies including residence, classroom, office, sports center, etc. The sources and sinks, compositions and distributions of indoor VOCs, and the factors (e.g., human activities, air exchange rate, temperature and humidity) driving the changes in indoor VOCs are discussed. The physical and chemical processes of gas-particle partitioning and secondary oxidation processes of VOCs, and their impacts on human health are summarized. Finally, the recommendations for future research directions on indoor VOCs measurements and indoor chemistry are proposed.

High-rise teaching buildings are complex public buildings that combine the evacuation risks of school buildings and high-rise buildings. In this regard, studying fire product transport patterns and personnel evacuation characteristics of high-rise school buildings is crucial for safe and rapid evacuation. In this paper, we applied Pyrosim2018 software to build a model of a teaching building and performed numerical fire simulation to analyze temperature, CO gas, and visibility to determine the available evacuation time ASET; meanwhile, we performed evacuation simulation by Pathfinder 2019 to determine the required evacuation time and analyze the congestion problem during evacuation. By improving the evacuation route, secondary simulations were conducted and compared with the previous results. The results show that visibility is the main factor affecting evacuation in of the event of a fire in this school building. Based on the visibility analysis, it is recommended that personnel evacuate from floors four and above within 709.2 S when the fire location is at a specific position on the third floor. While the original safety exits of the school building can avoid a large number of casualties, they cannot guarantee the safe evacuation of all people, and planning a reasonable evacuation route can obviously relieve the evacuation pressure in the high-rise corridor.

The fractal dimension is widely used in many fields as a parameter to characterize the geometric complexity and geometric distribution relationship of research objects. To study the surface characteristics of sulfide ore particles, the fractal theory was applied to quantitatively characterize the surface fractal dimension Ds of sulfide ore microparticles in three particle size ranges, 60–100 mesh, 100–140 mesh and >200 mesh, based on the area–perimeter method. Using an optical microscope, grain projection images of the particles were obtained. The grain shape and characteristics of sulfide ore particles were studied by means of an image processing system. The results demonstrate that the grain shape of sulfide ore particles can be expressed by fractal dimension, and the particle surface fractal dimension ranges from 2.4392 to 2.5492. It was found that the fractal properties begin to decrease due to the increasing of the particle size. The larger the fractal dimension, the finer the particles are. The fractal dimension of sulfide ore particles can be used as an important indicator of their particle shape distribution characteristics, which can provide important information for further study of the relevant physical and chemical properties of sulfide ore particles and provide a new theoretical method and basis for the adhesion and removal of sulfide ore dust. With the quantitative description of the fractal distribution of sulfide ore particles, a new way to study the adhesive force between particles is offered for further research.

This paper proposes a lightweight detection model based on machine vision, YOLOv5-GC, to improve the efficiency and accuracy of detecting and classifying surface defects in preforming materials. During this process, clear images of the entire surface are difficult to obtain due to the stickiness, high reflectivity, and black resin of the thermosetting plain woven prepreg. To address this challenge, we built a machine vision platform equipped with a linescan camera and high-intensity linear light source that captures surface images of the material during the preforming process. To solve the problem of defect detection in the case of extremely small and imbalanced samples, we adopt a transfer learning approach based on the YOLOv5 neural network for defect recognition and introduce a coordinate attention and Ghost Bottleneck module to improve recognition accuracy and speed. Experimental results demonstrate that the proposed approach achieves rapid and high-precision identification of surface defects in preforming materials, outperforming other state-of-the-art methods. This work provides a promising solution for surface defect detection in preforming materials, contributing to the improvement of composite material quality.

Introduction: Emerging evidence indicates that physiological and pathological conditions of the nose are posttranscriptionally regulated by microRNAs, a class of small noncoding RNAs. Recently, microRNA-223-3p has been increasingly implicated in the modulation of allergic rhinitis Objective: This study aimed to assess the role and mechanism of microRNA-223-3p in a mouse model of allergic rhinitis. Methods: The expression level of miR-223-3p was measured in the serum of 41 allergic rhinitis patients and 39 healthy controls using quantitative real time polymerase chain reaction. BALB/c mice were used to establish an allergic rhinitis model by intraperitoneal sensitization and intranasal challenge with ovalbumin. MicroRNA-223-3p agomir/antagomir was then intranasally administered to mice after ovalbumin challenge for another week. The symptoms of nasal rubbing and sneezing were recorded. Serum ovalbumin-specific immunoglobulin E concentration, microRNA-223-3p expression and proinflammatory cytokine (IL-4, IL-5, IFN-γ) levels in nasal mucosa were measured by ELISA and quantitative real time polymerase chain reaction, respectively. Histopathologic changes were evaluated using hematoxylin and eosin staining. Results: MicroRNA-223-3p levels increased significantly in both allergic rhinitis patients and allergic rhinitis mice. In addition, upregulation of microRNA-223-3p levels by nasal administration of microRNA-223-3p agomir also markedly increased the concentration of ovalbumin -specific IgE, the frequencies of nasal rubbing and sneezing, the levels of proinflammatory cytokines (IL-4, IL-5, IFN-γ) and eosinophil infiltration in the nasal mucosa of allergic rhinitis mice. Moreover, microRNA-223-3p antagomir appeared to strongly ameliorate the symptoms and pathology in nasal mucosa. Subsequently, we demonstrated for the first time that microRNA-223-3p negatively regulated INPP4A expression by binding with the 3′ untranslated region (3′UTR) of INPP4A. Conclusions: These findings indicate that microRNA-223-3p plays an important role in regulating the pathology and symptoms of allergic rhinitis by targeting INPP4A.

Hexapod robots are widely used for resource exploration, post-disaster rescue, and military equipment. They typically travel on rugged and complex roads. The robot itself has a high probability of failure owing to mechanical failure, driving motor failure, or external environmental interference. To improve the adaptability of the robot to a complex environment, a motion control method for fault-tolerant gait was designed. The trajectory generator based on zero-moment point information can generate a smooth desired trajectory for the body’s center of mass, thereby improving the robot’s zero-moment point trajectory tracking effect and motion stability. The force-distribution algorithm based on torque optimization selects the minimum square sum of the driving force as the objective function and reduces the number of constraint equations through QR decomposition to increase the speed at which the expected contact force at the foot is calculated. A CoppeliaSim and MATLAB/Gurobi joint simulation platform were built to simulate and verify the fault-tolerant motion planning of the hexapod robot and foot contact force control algorithm. The feasibility and effectiveness of the fault-tolerant motion planning and foot force control algorithm applied to a hexapod robot with a single-foot failure are verified.

Abstract Distant metastasis accompanied by angiogenesis is the main cause of nasopharyngeal carcinoma (NPC)-related death. Nuclear exosomes (nEXOs) are potential tumour biomarkers. High mobility group box 3 (HMGB3), a nuclear protein, is known to be overexpressed in cancers. However, its role in NPC has not been elucidated. Here, we explore for the first time the function of nEXO HMGB3 in tumour angiogenesis involved in NPC metastasis using a series of in vitro experiments with NPC cell lines and clinical specimens and in vivo experiments with tumour xenograft zebrafish angiogenesis model. We found a high expression of HMGB3 in NPC, accompanied by the formation of micronuclei, to be associated with metastasis. Furthermore, the NPC-secreted HMGB3 expression was associated with tumour angiogenesis. Moreover, HMGB3-containing nEXOs, derived from the micronuclei of NPC cells, were ingested by the human umbilical vein endothelial cells (HUVECs), and accelerated angiogenesis in vitro and in vivo. Importantly, western blotting and flow cytometry analysis showed that circulating nEXO HMGB3 positively correlated with NPC metastasis. In summary, nEXO HMGB3 can be a significant biomarker of NPC metastasis and provide a novel basis for anti-angiogenesis therapy in clinical metastasis.

Outdoor autonomous mobile robots heavily rely on GPS data for localization. However, GPS data can be erroneous and signals can be interrupted in highly urbanized areas or areas with incomplete satellite coverage, leading to localization deviations. In this paper, we propose a SLAM (Simultaneous Localization and Mapping) system that combines the IESKF (Iterated Extended Kalman Filter) and a factor graph to address these issues. We perform IESKF filtering on LiDAR and inertial measurement unit (IMU) data at the front-end to achieve a more accurate estimation of local pose and incorporate the resulting laser inertial odometry into the back-end factor graph. Furthermore, we introduce a GPS signal filtering method based on GPS state and confidence to ensure that abnormal GPS data is not used in the back-end processing. In the back-end factor graph, we incorporate loop closure factors, IMU preintegration factors, and processed GPS factors. We conducted comparative experiments using the publicly available KITTI dataset and our own experimental platform to compare the proposed SLAM system with two commonly used SLAM systems: the filter-based SLAM system (FAST-LIO) and the graph optimization-based SLAM system (LIO-SAM). The experimental results demonstrate that the proposed SLAM system outperforms the other systems in terms of localization accuracy, especially in cases of GPS signal interruption.

The scale-up of the coupling of water electroreduction (HER) with organic electrooxidation remains challenging. Here the authors address this challenge by coupling HER with electrooxidation of raw biomass chitin, cogenerating acetate and green hydrogen safely at high current density.

Abstract Background Nasopharyngeal carcinoma (NPC) is one of the most common malignancy in head and neck. With the development of treatments, the prognosis has improved these years, but metastasis is still the main cause of treatment failure. The endoplasmic reticulum (ER) resident protein 44 is a UPR-induced ER protein of the protein disulphide isomerase (PDI) family. This study investigated the role of ERp44 in NPC progression. Methods Firstly, immunohistochemistry, western blot and qRT-PCR were used to investigate the expression of ERp44 in NPC samples and cell lines. We analyzed 44 NPC samples for ERp44 expression and investigated the association between its expression level with clinicopathologic parameters. Then we took CCK8, Transwell migration assay and used the zebrafish model to access the role of ERp44 on the malignant phenotype in NPC cells. Secondly, we used co-IP to gain the proteins that interact with ERp44 and took proteomic analysis. Furthermore, we successfully constructed the mutant variants of ERp44 and found the interaction domain with ATP citrate lyase(ACLY). Lastly, we subcutaneously injected NPC cells into nude mice and took immunohistochemistry to exam the expression of ACLY and ERp44. Then we used western blot to detect the expression level of epithelial-mesenchymal transition (EMT) markers. Results In the present study, we found ERp44 was elevated in NPC tissues and correlated with clinical stages and survive state of the patients. In vitro, the downregulation of ERp44 in NPC cells (CNE2, 5-8F) could suppress cells proliferation and migration. After that, we recognized that ACLY might be a potential target that could interact with ERp44. We further constructed the mutant variants of ERp44 and found the interaction domain with ACLY. The promotion of ERp44 on cell migration could be inhibited when ACLY was knocked down. More importantly, we also observed that the interaction of ERp44 with ACLY, especially the thioredoxin region in ERp44 play a vital role in regulating EMT. Lastly, we found ERp44 was positively correlated with the expression of ACLY and could promote NPC cells growth in nude mice. Conclusion Our data indicated that ERp44 participates in promoting NPC progression through the interaction with ACLY and regulation of EMT.

BackgroundAcute kidney injury (AKI) is a morbid complication and the main cause of multiple organ failure and death in severely burned patients. The objective of this study was to explore epidemiology, risk factors, and outcomes of AKI for severely burned patients.MethodsThis retrospective study was performed with prospectively collected data of severely burned patients from the Institute of Burn Research in Southwest Hospital during 2011–2017. AKI was diagnosed according to Kidney Disease Improving Global Outcomes (KDIGO) criteria (2012), and it was divided into early and late AKIs depending on its onset time (within the first 3 days or >3 days post burn). The baseline characteristics, clinical data, and outcomes of the three groups (early AKI, late AKI and non-AKI) were compared using logistic regression analysis. Mortality predictors of patients with AKI were assessed.ResultsA total of 637 adult patients were included in analysis. The incidence of AKI was 36.9% (early AKI 29.4%, late AKI 10.0%). Multiple logistic regression analysis revealed that age, gender, total burn surface area (TBSA), full-thickness burns of TBSA, chronic comorbidities (hypertension or/and diabetes), hypovolemic shock of early burn, and tracheotomy were independent risk factors for both early and late AKIs. However, sepsis was only an independent risk factor for late AKI. Decompression escharotomy was a protective factor for both AKIs. The mortality of patients with AKI was 32.3% (early AKI 25.7%, late AKI 56.3%), and that of patients without AKI was 2.5%. AKI was independently associated with obviously increased mortality of severely burned patients [early AKI, OR = 12.98 (6.08–27.72); late AKI, OR = 34.02 (15.69–73.75)]. Abstract Tumour‐associated angiogenesis plays a critical role in metastasis, the main cause of malignancy‐related death. Extracellular vesicles (EVs) can regulate angiogenesis to participate in tumour metastasis. Our previous study showed that EVs rich in HAX1 are associated with in metastasis of nasopharyngeal carcinoma (NPC). However, the mechanism by which HAX1 of EVs promotes metastasis and angiogenesis is unclear. In this study, we demonstrated that EVs rich in HAX1 promote angiogenesis phenotype by activating the FAK pathway in endothelial cells (ECs) by increasing expression level of ITGB6. The expression level of HAX1 is markedly correlated with microvessel density (MVDs) in NPC and head and neck cancers based on an analysis of IHC. In addition to a series of in vitro cellular analyses, in vivo models revealed that HAX1 was correlated with migration and blood vessel formation of ECs, and metastasis of NPC. Using ribosome profiling, we found that HAX1 regulates the FAK pathway to influence microvessel formation and promote NPC metastasis by enhancing the translation efficiency of ITGB6. Our findings demonstrate that HAX1 can be used as an important biomarker for NPC metastasis, providing a novel basis for antiangiogenesis therapy in clinical settings.

The human mediodorsal thalamic nucleus (MD) is crucial for higher cognitive functions, while the fine anatomical organization of the MD and the function of each subregion remain elusive. In this study, using high-resolution data provided by the Human Connectome Project, an anatomical connectivity-based method was adopted to unveil the topographic organization of the MD. Four fine-grained subregions were identified in each hemisphere, including the medial (MDm), central (MDc), dorsal (MDd), and lateral (MDl), which recapitulated previous cytoarchitectonic boundaries from histological studies. The subsequent connectivity analysis of the subregions also demonstrated distinct anatomical and functional connectivity patterns, especially with the prefrontal cortex. To further evaluate the function of MD subregions, partial least squares analysis was performed to examine the relationship between different prefrontal-subregion connectivity and behavioral measures in 1012 subjects. The results showed subregion-specific involvement in a range of cognitive functions. Specifically, the MDm predominantly subserved emotional-cognition domains, while the MDl was involved in multiple cognitive functions especially cognitive flexibility and inhibition. The MDc and MDd were correlated with fluid intelligence, processing speed, and emotional cognition. In conclusion, our work provides new insights into the anatomical and functional organization of the MD and highlights the various roles of the prefrontal-thalamic circuitry in human cognition.

The tractive capability of Multi-wheeled mobile robots (WMRs) depends heavily on the performance of the driving control of their steerable wheels. The recently proposed concept of a pseudo-driven wheel (PDW) provides a simple, versatile, and effective way to improve the tractive capability. However, its focus is to eliminate the internal forces between the wheels and save energy; therefore, it cannot provide sufficient driving force to the body on rough terrains. This paper proposes a control strategy to adjust the power output of a PDW along the directions of two degrees of freedom, an approach that improves the trajectory tracking control of WMRs, especially on rough terrains. To achieve the desired active following control of the PDW, an active disturbance rejection controller is used to compensate the disturbances arising from the wheel-terrain interaction. The Adams and Simulink are used for a joint simulation of which results show that the proposed control strategy is necessary for accurate trajectory tracking, for which the feasibility and effectiveness were verified by the experimental results. In summary, the trajectory tracking accuracy of the WMR and its tractive capabilities on rough, sandy terrain were improved.

In order to study the air volume distribution regularity of mine ventilation clothing, the influence of ventilation clothing of different pipeline structures on air flow distribution is discussed. By establishing the simulation models of three different structures and using software simulation, the air volume distribution under different ventilation air volume and different pipeline structures is analyzed; the relationship between the air volume of the pipelines and the frictional resistance along the course is studied. The results demonstrated that the air volume of the pipelines farther from the ventilation inlet in the transverse type and longitudinal type is larger, while the air volume of the pipelines close to the ventilation inlet is smaller, and the air volume of the intermediate pipelines is medium and relatively stable; regularity of air volume distribution in pipelines: transverse type > spiral type > longitudinal type. Total air volume in the ventilation tubes: spiral type > longitudinal type > transverse type, with the increase of ventilation volume, the air volume in the spiral type tube increases greatly, and a small increase in air volume for transverse type and longitudinal type. Based on the data obtained from the transverse type simulation, the air volume of the pipelines airflow is inversely proportional to the frictional resistance along the course.

The expansion of preform and the optimization of preform have become important steps in the molding process. At present, there are some questions in the expansion of thermoset composite material preform and precompression, for example, the inaccurate dimensions, cracks, and wrinkles. For the expansion of preform, the finite element inverse algorithm is used as the expansion algorithm, and then the initial solution is optimized by the arc length mapping method, the expansion of preform is realized by the iterative equation which is solved by the ABAQUS solver. The effectiveness of the expansion of preform is verified through the comparison between the finite element inverse algorithm with DYNAFORM. The optimization of the precompression process is researched in order to solved the problems of cracks and wrinkles in the integral precompression method of preform. Firstly, the precompression sequence is adjusted by the precompression method, and then the precompression direction is optimized by the genetic algorithm. Through numerical simulation, the maximum thinning rate is reduced to 13%, and the maximum thickening rate is reduced to 6%, which improve the problems of cracks and wrinkles of preform, and the effectiveness of the optimization method is verified.

Renewable energy-driven hydrogen generation from water electrolysis has been widely recognized as a promising approach to utilize sustainable energy resources, reduce our dependence on legacy fossil fuels and alleviate net carbon dioxide emissions. However, conventional water electrolyzers suffer from the high overpotentials, mainly due to the sluggish kinetics of anodic oxygen evolution reaction (OER). This reaction also generates reactive oxygen species that could degrade the proton exchange membrane and oxygen that may mix with the cathodic hydrogen to form explosive gaseous mixtures. To address these issues, an innovative hybrid water electrolysis strategy which involves a certain alternative oxidation reaction to replace OER has been developed, and has led to a burgeoning area that sparks much research interest in finding available alternative reactions and their corresponding electrocatalysts. Herein, we summarize the alternative reactions into three groups: (1) the reagent-sacrificing type that can generate H2 with an ultra-low potential while the substrates are oxidized to valueless products; (2) the pollutant-degrading type at which environmental pollutants are used as substrates; (3) the value-added type that produces valuable products at the anode. Catalyst and electrolyzer designs for hybrid electrolysis are also briefly discussed, with an emphasis on the catalyst reconstruction phenomenon. Finally, the present challenges and perspectives are put forward.

Abstract Zinc‐air batteries (ZABs) could be an ideal candidate for challenging energy conversion and storage technology. Usually, the crucial oxygen reduction reaction and oxygen evolution reaction are naturally lethargic and require efficient catalysts to kinetically boost the energy conversion processes to achieve the practicable ZABs application. Transition‐metal/carbon hybrid catalysts show excellent potentials in electrocatalysis, and they have been developed into cost‐effective and scalable oxygen electrocatalyst alternatives for expensive and scarcer noble metal ones in rechargeable ZABs. Herein we summarize recent achievements in transition metal/carbon hybrids for oxygen electrocatalysis. These catalysts mainly fall into two categories by the integrated metal/metal oxides nanoparticles and atomically dispersed transition metal/carbon matrix. We focus on the relationship between structure and performance with their synthesis strategies. Their application in rechargeable ZABs are also comprehensively discussed. Finally, the perspectives on transition metal/carbon hybrids for oxygen electrocatalysis are presented for the future challenges in rechargeable ZABs.

This paper considers observer design for discrete-time descriptor systems with packet losses. By taking packet loss into consideration, the error dynamic of the proposed observer becomes a stochastic switched system. Consequently, the proposed observer is synthesized in a stochastic switched system framework. Sufficient conditions for the stochastic stability with a prescribed robust performance of the error dynamic system are derived and converted into linear matrix inequalities. Not only can the proposed observer deal with packet losses, but it also attenuates the effect of process disturbance and measurement noise. A numerical simulation of a truck-trailer is given to demonstrate the effectiveness of the proposed method.

Coal and gas outburst is one of the major disasters in the safety production of coal mine. According to the mechanism of coal and gas outburst, based on the comprehensive analysis of various influencing factors of coal and gas outburst, with the principles of selected early warning indicator, the basic information database of coal and gas outburst warning is constructed, and the information data query function is realized. The mathematical model of coal and gas outburst warning is established by the logistic regression analysis based on the gas concentration, the gas desorption index of drill cuttings, and the initial velocity of gas emission from the borehole. The multivariate information coupled warning was conducted according to the selected early warning indicator system, and the early warning level was divided with the result of early warning. The design and research of the coal and gas outburst warning system are carried out based on the geographic information system (GIS). The coal and gas outburst warning system was verified by taking the Tunliu mine of Lu’an Group as an example. The establishment of the early warning system is a new technical way to the early warning management of coal and gas outburst and can provide a guarantee for coal and gas outburst prevention.

Due to the harsh operating environment of aero-engines, a surface structure that provides excellent aerodynamic performance is urgently required to save energy and reduce emissions. In this study, microgroove polyurethane coatings fabricated by chemical synthesis are investigated in terms of their effect on aerodynamic performance, which is a new attempt to investigate the impact on aerodynamic performance of compressor cascade at transonic speeds. This method reduces manufacturing and maintenance cost significantly compared with traditional laser machining. Wake measurements are conducted in the high-speed linear compressor cascade wind tunnel to evaluate the performance of cascade attached with different microgroove polyurethane coatings. Compared with the Blank case, the microgroove polyurethane coatings have the characteristic of reducing flow loss, with a maximum reducing rate of 5.87% in the area-averaged total pressure loss coefficient. The mechanism of flow loss control is discussed through analyzing the correlation between the total pressure distribution and turbulence intensity distribution. The results indicate that a large quantity of energy loss in the flow field due to turbulence dissipation and the reduction in viscous drag by microgroove polyurethane coatings relates to its effect on turbulence control. This paper demonstrates a great perspective on designing micro-nano surface structure for aero-engine applications.

Although researchers have investigated a variety of approaches to the development of three-dimensional (3D) point cloud matching algorithms, the results have been limited by low accuracy and slow speed when registering large numbers of point cloud data. To address this problem, a new fast point cloud registration algorithm based on a 3D neighborhood point feature histogram (3DNPFH) descriptor is proposed for fast point cloud registration. With a 3DNPFH, the 3D key-point locations are first transformed into a new 3D coordinate system, and the key points generated from similar 3D surfaces are then close to each other in the newly generated space. Subsequently, a neighborhood point feature histogram (NPFH) was designed to encode neighborhood information by combining the normal vectors, curvature, and distance features of a point cloud, thus forming a 3DNPFH (3D + NPFH). The descriptor searches radially for 3D key point locations in the new 3D coordinate system, reducing the search coordinate system for the corresponding point pairs. The “NPFH” descriptor is then coarsely aligned using the random sample consensus (RANSAC) algorithm. Experiment results show that the algorithm is fast and maintains high alignment accuracy on several popular benchmark datasets, as well as our own data.

Thermal fatigue is the main failure mode for chromium hot-work steels. In this study, pre-alloyed chromium hot-work steel powders with three different Cr, Mo, and V addition levels (low content (LH), medium content (MH), and high content (HH)) were used for selective laser melting (SLM). The microstructure and thermal fatigue properties of these SLM-processed materials were investigated. After thermal fatigue tests, LH possessed the lowest hardness (approximately 573 HV5) and longest crack length, MH possessed the highest hardness (approximately 688 HV5) and HH (with the hardness of approximately 675 HV5) possessed the shortest crack length. It can be concluded that the increase of V content in MH is the main reason for the refined grains which result in an enhanced hardness and thermal fatigue resistance compared to LH. The further increase of the Cr and Mo content in HH leads to the grain coarsening and hardness decreasing, which is supposed to degrade the thermal fatigue resistant properties according to the conventional theory. However, HH exhibited an enhanced thermal fatigue resistance compared to MH. That is because the higher stored energy in MH deteriorated its thermal fatigue resistance compared to HH.

Non-human primate models are widely used in studying the brain mechanism underlying brain development, cognitive functions, and psychiatric disorders. Neuroimaging techniques, such as magnetic resonance imaging, play an important role in the examinations of brain structure and functions. As an indispensable tool for brain imaging data analysis, brain atlases have been extensively investigated, and a variety of versions constructed. These atlases diverge in the criteria based on which they are plotted. The criteria range from cytoarchitectonic features, neurotransmitter receptor distributions, myelination fingerprints, and transcriptomic patterns to structural and functional connectomic profiles. Among them, brainnetome atlas is tightly related to brain connectome information and built by parcellating the brain on the basis of the anatomical connectivity profiles derived from structural neuroimaging data. The pipeline for building the brainnetome atlas has been published as a toolbox named ATPP (A Pipeline for Automatic Tractography-Based Brain Parcellation). In this paper, we present a variation of ATPP, which is dedicated to monkey brain parcellation, to address the significant differences in the process between the two species. The new toolbox, MonkeyCBP, has major alterations in three aspects: brain extraction, image registration, and validity indices. By parcellating two different brain regions (posterior cingulate cortex) and (frontal pole) of the rhesus monkey, we demonstrate the efficacy of these alterations. The toolbox has been made public (https://github.com/bheAI/MonkeyCBP_CLI, https://github.com/bheAI/MonkeyCBP_GUI). It is expected that the toolbox can benefit the non-human primate neuroimaging community with high-throughput computation and low labor involvement.

In order to study the effects of stress and water pressure on the permeability of fractured sandstone, an ultra-deep gas layer of fractured tight sandstone in WengFu Mine in GuiZhou province, was selected for experimental design and research, and the effects of confining pressure, internal pressure, effective stress, and water pressure on gas permeability were studied. The results show that the effective stress and slippage effect compete with each other when the confining pressure is kept fixed and the internal pressure is increased in the fractured tight sandstone reservoir. At the beginning of the experiment, the gas slippage effect is strong while the rock stress sensitivity is weak. The gas permeability increasingly decreases with the increase of internal pressure. The effective stress is stronger than slippage effect, and the gas permeability increases with the increase of internal pressure when crossing the low point of permeability. The greater the confining pressure is, the greater the internal pressure needed to reach the low permeability point will be. With the increase of axial stress, horizontal contact hydraulic hole increasingly closed. Horizontal hydraulic holes are more sensitive to axial stress than vertical ones.

Abstract Background In the early stage of severe burn, patients often exhibit a high level of inflammatory mediators in blood and are likely to develop sepsis. High-volume haemofiltration (HVHF) can eliminate these inflammatory mediators. We hypothesised that early application of HVHF may be beneficial in reducing sepsis and improving the prognosis of patients with severe burns. Methods Adults patients with burns ≥ 50% total burn surface area (TBSA) and in whom the sum of deep partial and full-thickness burn areas was ≥ 30% were enrolled in this randomised prospective study, and they were divided into control (41 cases) and HVHF (41 cases) groups. Patients in the control group received standard management for major burns, whereas the HVHF group additionally received HVHF treatment (65 ml/kg/h for 3 consecutive days) within 3 days after burn. The incidence of sepsis and mortality, some laboratory data, levels of inflammatory cytokines in the blood, HLA-DR expression on CD14+ peripheral blood monocytes, the proportion of CD25+Foxp3+ in CD4+ T lymphocytes, and the counts of CD3+, CD4+ and CD8+ T lymphocytes were recorded within 28 days post-burn. Results The incidence of sepsis, septic shock and duration of vasopressor treatment were decreased significantly in the HVHF group. In addition, in the subgroup of patients with burns ≥ 80% TBSA, the 90-day mortality showed significant decreases in the HVHF group. The ratio of arterial oxygen partial pressure to the fraction of inspiration oxygen was improved after HVHF treatment. In the patients who received HVHF treatment, the blood levels of inflammatory cytokines, including tumour necrosis factor-α, interleukin (IL)-1β, IL-6 and IL-8, as well as the blood level of procalcitonin were found to be lower than in the control group. Moreover, higher HLA-DR expression on CD14+ monocytes and a lower proportion of CD25+Foxp3+ in CD4+ T lymphocytes were observed in the patients in the HVHF group. Conclusions Early application of HVHF benefits patients with severe burns, especially for those with a greater burn area (≥ 80% TBSA), decreasing the incidence of sepsis and mortality. This effect may be attributed to its early clearance of inflammatory mediators and the recovery of the patient’s immune status. Background/Aims: N-acetylcysteine (NAC) is a novel and promising agent with activity against bacterial biofilms. Human serum also inhibits biofilm formation by some bacteria. We tested whether the combination of NAC and human serum offers greater anti-biofilm activity than either agent alone. Methods: Microtiter plate assays and confocal laser scanning microscopy were used to evaluate bacterial biofilm formation in the presence of NAC and human serum. qPCR was used to examine expression of selected biofilm-associated genes. Extracellular matrix (ECM) was observed by transmission electron microscopy. The antioxidants GSH or ascorbic acid were used to replace NAC, and human transferrin, lactoferrin, or bovine serum albumin were used to replace serum proteins in biofilm formation assays. A rat central venous catheter model was developed to evaluate the effect of NAC on biofilm formation in vivo. Results: NAC and serum together increased biofilm formation by seven different bacterial strains. In Staphylococcus aureus, expression of genes for some global regulators and for genes in the ica-dependent pathway increased markedly. In Pseudomonas aeruginosa, transcription of las, the PQS quorum sensing (QS) systems, and the two-component system GacS/GacA increased significantly. ECM production by S. aureus and P. aeruginosa was also enhanced. The potentiation of biofilm formation is due mainly to interaction between NAC and transferrin. Intravenous administration of NAC increased colonization by S. aureus and P. aeruginosa on implanted catheters. Conclusions: NAC used intravenously or in the presence of blood increases bacterial biofilm formation rather than inhibits it.

Background/Aims: As an “ESKAPE” pathogen, Acinetobacter baumannii is one of the leading causes of drug-resistant infections in humans. Phage therapy may be a useful strategy in treating infections caused by drug-resistant A. baumannii. Among 21 phage strains that were isolated and described earlier, we investigated the therapeutic efficacy of Abp1 because of its relatively wide host range. Methods: Phage stability assays were used to evaluate thermal and pH stability of Abp1. Abp1 was co-cultured with A. baumannii (AB1) over a range of multiplicities of infection to determine its bactericidal efficacy. HeLa or THP-1 cells were used in the cytotoxicity and protection assays. Finally, the therapeutic effects of Abp1 on local and systemic A. baumannii infection in mice were determined. Results: We found that Abp1 exhibits high thermal and pH stability and has a low frequency of lysogeny. Bacteriophage resistance also occurs at a very low frequency (3.51±0.46×10-8), and Abp1 can lyse almost all host cells at a MOI as low as 0.1. Abp1 has no detectable cytotoxicity to HeLa or THP-1 cells as determined by LDH release assay. Abp1 can rescue HeLa cells from A. baumannii infection, even if introduced 2 hours post infection. In both local and systemic A. baumannii infection mouse models, Abp1 treatment exhibits good therapeutic effects. Conclusion: Abp1 is an excellent candidate for phage therapy against drug-resistant A. baumannii infections.

Wheeled mobile robots are widely utilized for environment-exploring tasks both on earth and in space. As a basis for global path planning tasks for wheeled mobile robots, in this study we propose a method for establishing an energy-based cost map. Then, we utilize an improved dual covariant Hamiltonian optimization for motion planning method, to perform point-to-region path planning in energy-based maps. The method is capable of efficiently handling high-dimensional path planning tasks with non-convex cost functions through applying a robust active set algorithm, that is, non-monotone gradient projection algorithm. To solve the problem that the path planning process is locked in weak minima or non-convergence, we propose a randomized variant of the improved dual covariant Hamiltonian optimization for motion planning based on simulated annealing and Hamiltonian Monte Carlo methods. The results of simulations demonstrate that the final paths generated can be time efficient, energy efficient and smooth. And the probabilistic completeness of the method is guaranteed.

We propose a new local path planning approach based on optimization methods with probabilistic completeness in this article. This approach adds a linear constraint to the original covariant Hamiltonian optimization for motion planning problem with a new cost function. By deducing the dual form, the path planning problem is described as a box-constrained quadratic programming problem. The nonmonotone gradient projection algorithm is introduced to solve the dual problem, which makes the algorithm adaptable to non-convex cost functions. In order to prevent early convergence at local minima that can occur when applying optimization methods, this article introduces Hamiltonian Monte Carlo to the modification, which constantly forces the initial path to jump out of the local extremum, thus improving the robustness and success rate of the path planning approach. Compared with other methods through simulations, this approach is proven to provide balanced planning efficiency and path quality. The feasibility in a real environment is experimentally validated by applying the approach to a wheeled mobile robot.

ABSTRACT Acinetobacter baumannii is a growing threat, although lytic bacteriophages have been shown to effectively kill A. baumannii. However, the interaction between the host and the phage has not been fully studied. We demonstrate the global profile of transcriptional changes in extensively drug-resistant A. baumannii AB1 and the interaction with phage φAbp1 through RNA sequencing (RNA-seq) and bioinformatic analysis. Only 15.6% (600/3,838) of the genes of the infected host were determined to be differentially expressed genes (DEGs), indicating that only a small part of the bacterial resources was needed for φAbp1 propagation. Contrary to previous similar studies, more upregulated rather than downregulated DEGs were detected. Specifically, φAbp1 infection caused the most extensive impact on host gene expression at 10 min, which was related to the intracellular accumulation phase of virus multiplication. Based on the gene coexpression network, a middle gene (gp34, encoding phage-associated RNA polymerase) showed a negative interaction with numerous host ribosome protein genes. In addition, the gene expression of bacterial virulence/resistance factors was proven to change significantly. This work provides new insights into the interactions of φAbp1 and its host, which contributes to the further understanding of phage therapy, and provides another reference for antibacterial agents. IMPORTANCE Previous research has reported the transcriptomic phage-host interactions in Escherichia coli and Pseudomonas aeruginosa, leading to the detailed discovery of transcriptomic regulations and predictions of specific gene functions. However, a direct relationship between A. baumannii and its phage has not been previously reported, although A. baumannii is becoming a rigorous drug-resistant threat. We analyzed transcriptomic changes after φAbp1 infected its host, extensively drug-resistant (XDR) A. baumannii AB1, and found defense-like responses of the host, step-by-step control by the invader, elaborate interactions between host and phage, and elevated drug resistance gene expressions of AB1 after phage infection. These findings suggest the detailed interactions of A. baumannii and its phage, which may provide both encouraging suggestions for drug design and advice for the clinical use of vital phage particles.

Wheeled mobile robots (WMRs) in real complex environments such as on extraterrestrial planets are confronted with uncertain external disturbances and strong coupling of wheel-ground interactions while tracking commanded trajectories. Methods based on sliding mode control (SMC) are popular approaches for these situations. Traditional SMC has some potential problems, such as slow convergence, poor robustness, and excessive output chattering. In this paper, a kinematic-based feed-forward control model is designed for WMRs with longitudinal slippage and applied to the closed-loop control system for active compensation of time-varying slip rates. And a new adaptive SMC method is proposed to guide a WMR in trajectory tracking missions based on the kinematic model of a general WMR. This method combines the adaptive control method and a fast double-power reaching law with the SMC method. A complete control loop with active slip compensation and adaptive SMC is thus established. Simulation results show that the proposed method can greatly suppress chattering and improve the robustness of trajectory tracking. The feasibility of the proposed method in the real world is demonstrated by experiments with a skid-steered WMR on the loose-soil terrain.

Salinity is an important index of water quality in oilfield water injection engineering. To address the need for real-time measurement of salinity in water flooding solutions during oilfield water injection, a salinity measurement system that can withstand a high temperature environment was designed. In terms of the polarization and capacitance effects, the system uses an integrator circuit to collect information and fuzzy control to switch gears to expand the range. Experimental results show that the system can operate stably in a high-temperature environment, with an accuracy of 0.6% and an uncertainty of 0.2% in the measurement range of 1–10 g/L.

Background: CFTR is implicated in cutaneous wound healing although the underlying mechanisms are not fully understood. In other cell types, CFTR is reported to regulate MAPK/ NF-κB signaling. We undertook the present study to explore a possible role of CFTR in regulating MAPK/NF-κB during cutaneous wound healing. Methods& Results: The splint-excisional and incisional wound healing models were used in CFTR mutant (DF508) mice. The cell-scratch model was used in a human keratinocyte line, HaCaT, in conjunction with CFTR knockdown or overexpression. The epidermal inflammation, keratinocyte proliferation and differentiation, as well as MAPK/NF-κB signaling were examined. Inhibitors of MAPK/NF-κB were also used. Results: Both DF508 mice and HaCaT cells with CFTR knockdown exhibited delayed cutaneous wound healing with exuberant inflammation, increased proliferation and aberrant differentiation. Knockdown of CFTR in HaCaT cells resulted in phosphorylation of ERK, p38 and IκBα. The disturbance of inflammation, proliferation and differentiation in HaCaT cells were reversed by CFTR overexpression or inhibition of MAPK or NF-κB. Conclusion: CFTR plays a role in suppressing MAPK/NF-κB to relieve inflammation, reduce proliferation and promote differentiation of keratinocytes, and thus promotes cutaneous wound healing.

Background: Previous studies have established that estrogen is capable of accelerating cutaneous wound healing through multiple mechanisms, one of which involves affecting keratinocytes biological properties, such as migration, proliferation, etc. This study aims to reveal the underlying molecular mechanisms of estrogen promoting epidermal keratinocytes proliferation. Method & Results: We found that compared with female mice with a normal estrous cycle, female mice with their ovaries removed before puberty exhibited a delayed cutaneous wound healing, thinner epidermis, and significantly fewer proliferating cell nuclear antigen (PCNA)-positive keratinocytes. Moreover, a significant increase in HaCaT proliferation was detected by a CCK8 assay when treated with 17 β-estradiol compared with those treated with control vehicle. Consistent with the results of the CCK8 assay, flow cytometry indicated a high proportion of 17 β-estradiol-treated HaCaT cells in S phase compared with vehicle-treated cells. Western blot analysis demonstrated the activation of Akt, Erk and upregulation of PCNA in HaCaT cells treated with 17 β-estradiol. Interestingly, Erk activation occurred prior to Akt activation. Upregulation of PCNA expression, elevated proliferation and high S phase fraction of HaCaT cell by 17 β-estradiol could be reversed by an Akt or Erk inhibitor. Moreover, Erk inhibition reversed 17 β-estradiol-induced Akt activation, whereas an Akt inhibitor exhibited no effect on Erk, further suggesting that Erk was on the upstream while Akt on the downstream of the signaling pathway. Conclusion: This study demonstrates that one of the critical mechanisms underlying 17 β-estradiol promoting skin wound healing is through regulation of keratinocyte proliferation via Erk/Akt signaling pathway.

Staphylococcus argenteus is a novel staphylococcal species (also considered as a part of Staphylococcus aureus complex) that is infrequently reported on, and clinical S. argenteus infections are largely unstudied. Here, we report a persistent and recurrent hip joint infection case in which a S. argenteus strain and its small colony variants (SCVs) strain were successively isolated. We present features of the two S. argenteus strains and case details of their pathogenicity, explore factors that induce S. argenteus SCVs formation in the course of anti-infection therapy, and reveal potential genetic mechanisms for S. argenteus SCVs formation. S. argenteus strains were identified using phenotypic and genotypic methods. The S. argenteus strain XNO62 and SCV strain XNO106 were characterized using different models. S. argenteus SCVs were induced by the administration of amikacin and by chronic infection course based on the clinical case details. The genomes of both strains were sequenced and aligned in a pair-wise fashion using Mauve. The case details gave us important insights on the characteristics and therapeutic strategies for infections caused by S. argenteus and its SCVs. We found that strain XNO62 and SCV strain XNO106 are genetically-related sequential clones, the SCV strain exhibits reduced virulence but enhanced intracellular persistence compared to strain XNO62, thus promoting persistent infection. The induction experiments for S. argenteus SCVs demonstrated that high concentrations of amikacin greatly induce S. argenteus XNO62 to form SCVs, while a chronic infection of S. argenteus XNO62 slightly induces SCVs formation. Potential genetic mechanisms for S. argenteus SCVs formation were revealed and discussed based on genomic alignments. In conclusion, we report the first case of infection caused by S. argenteus and its SCVs strain. More attention should be paid to infections caused by S. argenteus and its SCVs, as they constitute a challenge to current therapeutic strategies. The problem of S. argenteus SCVs should be noticed, in particular when amikacin is used or in the case of a chronic S. argenteus infection.

Pseudomonas aeruginosa is the leading cause of infection in burn patients. The increasing carbapenem resistance of P. aeruginosa has become a serious challenge to clinicians. The present study investigated the molecular typing and carbapenem resistance mechanisms of 196 P. aeruginosa isolates from the bloodstream and wound surface of patients in our burn center over a period of 6 years. By multilocus sequence typing (MLST), a total of 58 sequence types (STs) were identified. An outbreak of ST111, a type that poses a high international risk, occurred in 2014. The isolates from wound samples of patients without bacteremia were more diverse and more susceptible to antibiotics than strains collected from the bloodstream or the wound surface of patients with bacteremia. Importantly, a large proportion of the patients with multisite infection (46.51%) were simultaneously infected by different STs in the bloodstream and wound surface. Antimicrobial susceptibility testing of these isolates revealed high levels of resistance to carbapenems, with 35.71% susceptibility to imipenem and 32.14% to meropenem. To evaluate mechanisms associated with carbapenem resistance, experiments were conducted to determine the prevalence of carbapenemase genes, detect alterations of the oprD porin gene, and measure expression of the ampC β-lactamase gene and the mexB multidrug efflux gene. The main mechanism associated with carbapenem resistance was mutational inactivation of oprD (88.65%), accompanied by overexpression of ampC (68.09%). In some cases, oprD was inactivated by insertion sequence element IS1411, which has not been found previously in P. aeruginosa. These findings may help control nosocomial P. aeruginosa infections and improve clinical practice.

Methicillin-resistant Staphylococcus aureus (MRSA) infections are prevalent in burn wards, and are especially serious in S. aureus bacteremia (SAB) patients. Glycopeptides and daptomycin are effective against MRSA infections, but MIC creeps can reduce their efficacy. Our object was to perform a molecular epidemiological investigation of S. aureus isolates in our burn center and to evaluate MICs for antimicrobials against SAB-associated MRSA isolates. A total of 259 S. aureus isolates, obtained from August 2011 to July 2016, were used in this study. Multiple molecular typing was used for molecular epidemiological analysis. E-tests were used to determine MICs of vancomycin, teicoplanin, and daptomycin for SAB-associated MRSA isolates. MIC values were stratified by collection date or source and compared. Spearman's test was used to analyze MICs correlations amongst tested antimicrobials. ST239-MRSA-III-t030-agrI clone was found to be dominant in both SAB and non-SAB patients, and significantly more in SAB patients (P < 0.0001). SAB-MRSA isolates exhibited decreased MICs for vancomycin, teicoplanin, and daptomycin during the 5-year period. Compared to those isolated from catheters or wounds, SAB-MRSA isolates from the bloodstream were less susceptible to vancomycin and daptomycin, but more susceptible to teicoplanin. MICs Correlation was found only between vancomycin and daptomycin in MRSA isolates from the bloodstream (rho = 0.250, P = 0.024). In conclusion, our results suggest that MRSA infections are still serious problems in burn centers. In contrast to most other studies, we observed increased susceptibility to glycopeptides and daptomycin against SAB-associated MRSA in our center from 2011 to 2016, suggesting the use of glycopeptides does not lead to MIC creeps. Isolates from different sites of the body may exhibit different levels of susceptibility and change trend over time for different antimicrobials, antimicrobials selection for MRSA infections should be considered comprehensively.
