Marine renewable energy has broad application prospects. Compared with the single energy utilization device, the floating multi-energy combined power generation device can better realize the optimal allocation of resources and reduce the development cost of energy. In order to fully understand the development prospects of multi-energy combined power generation devices, this paper starts with the multi-energy combined power generation integration scheme, analyzes the differences between different types of combined power generation devices, and summarizes their technical characteristics. Next, the research progress of each module analysis method and multi-module coupling dynamics analysis method of the floating multi-energy combined power generation device is clarified and summarized in such aspects as theoretical research and simulation. Finally, the applicable scenarios of the various methods are analyzed and the development trends, problems, and challenges of floating multi-energy combined power generation devices are discussed, aiming to provide references for the subsequent construction of economically feasible floating multi-energy combined power generation devices.

The contact morphology change caused by high-current ablation will seriously affect the electric field distribution in the interrupter chamber, which in turn affects the closing prebreakdown arc duration, indicating that the prebreakdown arc duration can be used as one of the indicators to measure the contact ablation state. A circuit breaker simulated ablation test platform was established, and the voltage and current signals, electromagnetic field signals, and vibration signals in the process of circuit breaker closing were measured, and the closing prebreakdown duration was calculated. The results show that under the same size of the ablation current, with the ablation of the contacts, the closing prebreakdown duration shows an overall trend of increasing and then decreasing, and the larger the ablation current is, the larger the change in the closing prebreakdown duration is. At the same time, simulation verification was carried out, and the results show that the electric field distortion on the surface of the static arc contact inside the arc extinguishing chamber is the largest, and the ablation of the contact will further increase the degree of distortion of the electric field inside the arc extinguishing chamber. As the degree of ablation increases, the prebreakdown moment of the circuit breaker closing is advanced, and the prebreakdown duration increases.

Summary: Artificial intelligence (AI) has been found to assist in optical differentiation of hyperplastic and adenomatous colorectal polyps. We investigated whether AI can improve the accuracy of endoscopists’ optical diagnosis of polyps with advanced features. We introduced our AI system distinguishing polyps with advanced features with more than 0.870 of accuracy in the internal and external validation datasets. All 19 endoscopists with different levels showed significantly lower diagnostic accuracy (0.410–0.580) than the AI. Abstract The integration of nano-semiconductors into electromagnetic wave absorption materials is a highly desirable strategy for intensifying dielectric polarization loss; achieving high-attenuation microwave absorption and realizing in-depth comprehension of dielectric loss mechanisms remain challenges. Herein, ultrafine oxygen vacancy-rich Nb2O5 semiconductors are confined in carbon nanosheets (ov-Nb2O5/CNS) to boost dielectric polarization and achieve high attenuation. The polarization relaxation, electromagnetic response, and impedance matching of the ov-Nb2O5/CNS are significantly facilitated by the Nb2O5 semiconductors with rich oxygen vacancies, which consequently realizes an extremely high attenuation performance of − 80.8 dB (> 99.999999% wave absorption) at 2.76 mm. As a dielectric polarization center, abundant Nb2O5–carbon heterointerfaces can intensify interfacial polarization loss to strengthen dielectric polarization, and the presence of oxygen vacancies endows Nb2O5 semiconductors with abundant charge separation sites to reinforce electric dipole polarization. Moreover, the three-dimensional reconstruction of the absorber using microcomputer tomography technology provides insight into the intensification of the unique lamellar morphology regarding multiple reflection and scattering dissipation characteristics. Additionally, ov-Nb2O5/CNS demonstrates excellent application potential by curing into a microwave-absorbing, machinable, and heat-dissipating plate. This work provides insight into the dielectric polarization loss mechanisms of nano-semiconductor/carbon composites and inspires the design of high-performance microwave absorption materials.

Wall bushing is the core insulator of 40.5 kV air insulation switchgear, and its insulation performance determines the insulation level and service life of the switchgear. Wall bushing often appears partial discharge, insulation aging, grounded breakdown, and other problems after a period of operation. The main reason is the partial discharge of the equipotential line for the wall bushing. In this paper, the influence of the wall bushing equipotential line on the electric field distribution is systematically analyzed to improve the operating reliability and service life of the 40.5 kV wall bushing. Based on the electric field simulation of the equipotential line, the feasibility of the traditional live detection method is given. The characteristic gas detection method is proposed through air electrolysis, thermal decomposition, early warning of the development trend of the internal suspension defect state. Finally, it proposes corresponding preventive measures to avoid the development of internal suspension defect of wall bushing into a sudden grounding, and ensure the reliability of the system operation.

The Xihu Depression is the largest hydrocarbon-bearing depression of the East China Sea Shelf Basin (also referred to as the ECSSB). However, the depositional systems and reservoir distribution of the Oligocene Huagang Formation in the Xihu Depression are still controversial. Under the guidance of sedimentology and stratigraphy, this study documented a marine-terrestrial transitional environment in the restricted bay setting of the Oligocene Huagang Formation through core description, well logging, and seismic data analysis. This study also revealed that the Oligocene Huagang Formation is dominated by tidal delta, estuary, and gravity flow deposits in the central anticline zone of the Xihu Depression. The new understanding of the sedimentary systems and the discovery of the transgressive gap in the eastern Diaoyu Islands uplift explain the origin of fine-grained sediments and the EW-trending sand bodies in the central depression and the sand bodies parallel to shoreline in the west slope belt, which cannot be explained by previous study results, such as southern transgression or fluvial deltas and even lacustrine deposition. Moreover, the tidal channels, tidal sand flats, and gravity flow sand bodies formed by the transgressive tides are high-quality reservoirs. The study will provide a basis for well placement and serve as guidance for the selection of favorable hydrocarbon exploration areas in the Xihu Depression.

At present, the architecture modeling method of fluvial reservoirs are still developing. Traditional methods usually use grids to characterize architecture interbeds within the reservoir. Due to the thin thickness of this type of the interlayers, the number of the model grids must be greatly expanded. The number of grids in the tens of millions often makes an expensive computation; however, upscaling the model will generate a misleading model. The above confusion is the major reason that restricts the large-scale industrialization of fluvial reservoir architecture models in oilfield development and production. Therefore, this paper explores an intelligent architecture modeling method for multilevel fluvial reservoirs based on architecture interface and element. Based on the superpositional relationship of different architectural elements within the fluvial reservoir, this method uses a combination of multilevel interface constraints and non-uniform grid techniques to build a high-resolution 3D geological model for reservoir architecture. Through the grid upscaling technology of heterogeneous architecture elements, different upscaling densities are given to the lateral-accretion bedding and lateral-accretion bodies to simplify the model gridding. This new method greatly reduces the number of model grids while ensuring the accuracy of lateral-accretion bedding models, laying a foundation for large-scale numerical simulation of the subsequent industrialization of the architecture model. This method has been validated in A layer of X oilfield with meandering fluvial channel sands as reservoirs and B layer of Y oilfield with braided river sands as reservoirs. The simulation results show that it has a higher accuracy of production history matching and remaining oil distribution forecast of the targeted sand body. The numerical simulation results show that in the actual development process of oilfield, the injected water will not displace oil in a uniform diffusive manner as traditionally assumed, but in a more complex pattern with oil in upper part of sand body being left behind as residual oil due to the influences of different levels of architecture interfaces. This investigation is important to guiding reservoir evaluation, remaining oil analysis, profile control and potential tapping and well pattern adjustment.

Perinatal exposure to bifenthrin (BF) alters neurodevelopment. However, the most susceptible time period to BF exposure and the possible mechanisms are not clear. In the current study, pregnant female mice were treated with BF (0.5 mg/kg/d) at three different stages [gestational day (GD) 0–5, 6–15 and 16-birth (B)] and neurologic deficits were evaluated in offspring mice. BF exposure at GD 16-B significantly altered the locomotor activity and caused learning and memory impairments in 6-week-old offspring. Gestational BF exposure also caused neuronal loss in the region of cornu ammonis of hippocampi of 6-week-old offspring. Interestingly, neurobehavioral impairments and neuronal loss were not observed in offspring at 10-week-old. BF exposure at GD 16-B also decreased protein levels of VGluT1, NR1 and NR2A while increased the protein levels of NR2B and VGAT1, as well as the gene levels of Il-1β, Il-6 and Tnf-α in hippocampi of 6-week-old offspring. Collectively, these data demonstrate that gestational exposure to a low dose BF causes neurodevelopmental deficits that remit with the age and the late-stage of pregnancy is the most susceptible time window to BF exposure. Imbalance in excitatory/inhibitory neuronal transmission, altered expression levels of NMDA receptors and increased neural inflammation may be associated with BF prenatal exposure-triggered neurobehavioral impairments.

The neuroprotective effects of Er-Zhi-Wan (EZW), a well-known traditional Chinese formulation, in MPTP-induced Parkinson’s disease (PD) models are poorly understood and require evaluation. A model of PD induced by MPTP was used to evaluate the neuroprotective effects of EZW in mice. The underlying pharmacological mechanisms of EZW for the prevention and treatment of PD were then explored using a combination of multilevel databases, network pharmacology, biological experiments, and LCMS/MS. In vivo data showed that pretreatment with EZW can be neuroprotective against MPTP-induced motor dysfunction and can effectively rescue dopaminergic neurons from MPTP-induced degeneration in mice. Furthermore, data from combined multilevel databases and network pharmacology analysis strategies suggested that the neuroprotective activity of EZW in the treatment of PD is mediated by a complicated multicomponent, multitarget network. Genes such as Grm2, Grm5, Drd2, and Grik2 were identified as important therapeutic targets. Subsequent experimental validation showed that EZW can broadly regulate the mRNA levels of these receptor genes as well as BDNF, and consequently increase the phosphorylation levels of CREB to stimulate CREB signaling. These targets and signaling systems may be responsible for the reversal of neuronal death by EZW after MPTP exposure. The LC-MS/MS results also identified a wide range of chemical components of EZW, including at least 53 precise compounds, further demonstrating the complexity of the network in which EZW exerts its neuroprotective activity. Our work provides evidence for the mechanism of EZW in MPTP-PD models and supports the neuroprotective function of EZW in neurodegenerative diseases.

In the Laozi, it is said, “Heaven and earth are long and enduring because they do not arise from themselves, so they can live forever.” In Zhuangzi - Zai You, it is also said, “Keep your mental peace, and your body will be healthy. Don't strain your body and don't consume your spirit to live a long life.” It is clear that people attach importance to anti-aging and the desire for longevity.Throughout human history, we have treated aging as an inevitable process, but with the development of medical science, we have become more aware of the various molecular changes in the human body. In an aging society, more people are suffering from age-related diseases such as osteoporosis, Alzheimer's disease, and cardiovascular disease, which has led to a search for anti-aging. However, by ‘living longer’ we mean not only living but also living longer in good health. The mechanisms of aging are still unclear and there is a great deal of interest and curiosity in how to combat aging effectively.Some potential criteria exist for the determination of anti-aging drugs: the first criterion is the ability to exert life-extending effects in model organisms, preferably in mammals; the second criterion is the ability to prevent or delay several age-related diseases in mammals; and the third criterion is the ability to inhibit the transition of cells from a quiescent to a senescent state. Based on these criteria, the current anti-aging drugs often involved are rapamycin, metformin, curcumin and other polyphenols, polysaccharides, resveratrol, etc.The most studied and relatively well-understood pathways and factors of aging are currently known to include seven enzymes, six biological factors, and one chemical, which mainly involve more than ten pathways such as Nrf2/SKN-1; NFκB; AMPK; P13K/AKT; IGF; and NAD.

Intermittent partial discharge has become an essential threat to the safety and stability of high voltage electrical equipment. In order to reduce the sudden insulation breakdown failure due to the intermittent and latent partial discharge of the operating equipment, this paper analyzes the discharge characteristics of the intermittent partial discharge in the constant voltage mode. The UHF partial discharge spectrum of typical intermittent partial discharges is obtained, and the intermittent discharges are divided into low-risk and high-risk categories by adopting PCA and FCM cluster algorithms. Aiming at the characteristics of intermittent partial discharge and the on-line monitoring method of partial discharge, a partial discharge signal collection method based on dynamic frequency is proposed. An intelligent monitoring system of intermittent partial discharge is designed to acquire partial discharge signals initially locate defects. Then the BP neural network method is applied to automatically evaluate and early warn the internal insulation status. The test concluded that the designed monitoring system for intermittent partial discharge could dynamically track and warn the low-risk and high-risk, and evaluate the insulation status of the equipment.

Large-capacity impulse test of distribution transformer requires a large amount of instantaneous energy, while the impulse mode of conventional dedicated high-voltage line and generator requires synchronous switch, adjusted impedance, generator, etc., which has low operability and the risk of affecting system stability. To solve the problems in the large-capacity impulse test of distribution transformers, this paper proposes a high-current impulse test method based on energy storage technology, power electronics technology and intelligent control technology. The stepless continuous adjustable voltage of large capacity short circuit test, precise control of test current peak factor and stable output are realized, and the corresponding smart energy storage power supply for the large capacity test of a distribution transformer is developed. Through simulation and live test, the design of the energy storage intelligent power supply proposed in this paper greatly reduces the power supply side capacity requirements of large capacity impulse test of power distribution, and has excellent operability in field test.

Breaking–closing of the circuit breaker inevitably produces micron metal dust, significantly affecting the internal insulation state of the GIS. This paper first analyzes the force characteristics of micron metal dust and its separation characteristics inside the circuit breaker, then builds a test platform supporting 520 kV power frequency withstand voltage and 1175 kV lightning impulse withstand voltage based on true type 363 kV GIS to investigate the influence of micron foreign matter on the insulation performance of GIS. Subsequently, ANSOFT is used to simulate the electric field distortion when there is micron metal dust inside the circuit breaker, and the influence of foreign matter on the degree of electric field distortion under the action of the flow field. The characteristics of the influence of micron foreign matter on the insulation performance of foreign matter inside the GIS are summarized. Secondly, using artificial micron foreign matter, the pulse current and the UHF methods are adopted to detect the discharge amount. The sensitivity of different detection methods to the discharge detection of the micron foreign matter is analysed, plus the partial discharge characteristics of the micron foreign matter under different voltages. Afterwards, based on the test results of micron foreign matter, two improvement measures are proposed, including the treatment of foreign matter inside the GIS and the dual-light source angiography inspection, so as to reduce the influence of micron foreign matter on the insulation status of the GIS. Finally, through the AC withstand voltage test of artificial embedded foreign matter and the insulation margin check under 1.1 times lightning impact, the effectiveness of the measures proposed in this paper is verified, thus providing technical support for certifying the impact of micron foreign matter on the insulation state during equipment operation and maintenance.

Thermal analysis is an important method to evaluate the thermal stability of polymer materials; however, efficiently describing the complex pyrolysis process of polymers remains a great challenge. In this paper, a new dual-competitive parallel-reaction kinetics model is proposed for describing the thermogravimetric lines of material pyrolysis processes. In the proposed method, peak analysis, dual-competitive reaction form, and parallel reaction mechanism are combined to build the kinetic model of resin polymer pyrolysis. The model includes peak-determined reaction components, and each reaction component is divided into two competitive reactions. The two competitive reactions are obtained by conducting initial peak analysis and subsequent global parameter optimization. The rebuilt thermogravimetric line via the proposed model agrees well with the experimental data, with few reaction compositions and low computational cost. In addition, the pyrolysis of synthetic polymers and natural biomass are also successfully described by the model with high accuracy. This work provides an updated kinetic model for describing polymer pyrolysis accurately, which may advance the thermal analysis in developing polymers of complex pyrolysis behavior.

The Ordovician reservoirs in the Tahe oilfield are dominated by fractured-vuggy carbonate reservoirs, of which fault-karst reservoirs are a hot topic in recent years. Fault-karst reservoirs feature high production, large burial depth, and strong heterogeneity under the control of faulting and karstification. Based on geological, logging, and seismic data, this study classified the Ordovician fault-karst reservoirs in the Yuejin block of the Tahe oilfield into three types, namely karst-cave, dissolved-vug, and fractured types, and established the integrated identification criteria of the three types of reservoirs. This study characterized karst caves, dissolved vugs, and multi-scale faults through seismic wave impedance inversion and frequency-domain detection of multi-scale faults. 3D geological models of different types of reservoirs were built using the combined deterministic and stochastic methods and characterized the spatial distribution of multi-scale faults, karst caves, dissolved vugs, and physical property parameters of reservoir. This study established the method for the geological modeling of fault-karst reservoirs, achieved the quantitative characterization and revealed the heterogeneity of fault-karst reservoirs. The karst-cave and dissolved-vug types are high in porosity and act as reservoirs, while the fractured type is high in permeability and act as flow pathway. This study lays the foundation for the development index prediction, well emplacement, and efficient development of the fault-karst carbonate reservoirs.

Adhesive bonding plays a pivotal role in structural connections, yet the bonding strength is notably affected by the presence of pore defects. However, the invisibility of interior pores severely poses a challenge to understanding their influence on tensile failure behaviors under loading. In this study, we present a pioneering investigation into the real-time micro-failure mechanisms of adhesively bonded structures using in situ X-ray micro-CT. Moreover, the high-precision finite element analysis (FEA) of stress distribution is realized by establishing the real adhesive layer model based on micro-CT slices. The findings unveil that pores induce stress concentration within the adhesive layer during the tensile process, with stress levels significantly contingent upon pore sizes rather than their specific shapes. Consequently, larger pores initiate and propagate cracks along their paths, ultimately culminating in the failure of adhesively bonded structures. These outcomes serve as a significant stride in elucidating how pore defects affect the bonding performance of adhesively bonded structures, offering invaluable insights into their mechanisms.

Abstract Ferromagnetic alloys/carbon composites with excellent electrical and magnetic properties are highly desirable as electromagnetic wave absorption materials, but achieving high‐attenuation performance in C‐band (4–8 GHz) remains a challenge. Herein, a direct carbothermal reduction of organic gluconate solid‐solution precursor method is developed to synthesize ferromagnetic Fe‐Co‐Ni alloy/carbon composites, which realize high‐attenuation electromagnetic wave absorption in C‐band. By virtue of Fe, Co, and Ni elements homogeneously dispersing at the molecular level in the solid‐solution precursor with the regulated mole ratio, serial FeCo2Ni/C, Co7Fe3/C, FeNi3/C, and Co3Ni/C composites can be deliberately prepared. First‐principles calculations and off‐axis electron holograms can clearly unravel that these Fe‐Co‐Ni alloys could perform as excellent dielectric‐magnetic complementary loss units to trigger synergistic electronic dipole polarization oscillation, magnetic moment resonance, and magnetic coupling interaction. Meanwhile, the rich alloy–carbon interfaces and conductive carbon skeleton can facilitate delightful interfacial polarization and conductive loss. Combining these positive electromagnetic energy dissipation characteristics, FeCo2Ni/C with strengthened dipole polarization oscillation and outstanding impedance matching realizes an extremely high‐attenuation absorption performance with a minimum reflection loss of −82.2 dB at 5.21 GHz. This work provides a feasible and flexible insight into producing magnetic‐dielectric complementary Fe‐Co‐Ni alloys/carbon composites with various alloy compositions as excellent electromagnetic wave absorbers.

Here the authors report a general synthetic method to achieve ultrafine metal oxide - reduced graphene oxide nanocomposites for dye filtration, achieving high water permeability and selectivity.

Abstract Neural tube defects (NTDs) remain one of the most life-threatening birth defects affecting infants. Most patients with NTDs eventually develop lifelong disability, which cause significant morbidity and mortality and seriously reduce the quality of life. Our previous study has found that ethionine inhibits cell viability by disrupting the balance between proliferation and apoptosis, and preventing neural stem cells from differentiating into neurons and astrocytes. However, how ethionine participates in the pathogenesis of neural tube development through N6-methyladenosine (m6A) modification remains unknown. This study aims to investigate METTL3- and ALKBH5-mediated m6A modification function and mechanism in NTDs. Herein, our results demonstrate that SAM play not only a compensatory role, it also leads to changes of m6A modification in neural tube development and regulation. Additionally, these data implicate that METTL3 is enriched in HT-22 cells, and METTL3 knockdown reduces cell proliferation and increases apoptosis through suppressing Wnt/β-catenin signaling pathway. Significantly, overexpression of ALKBH5 can only inhibit cell proliferation, but cannot promote cell apoptosis. This research reveals an important role of SAM in development of NTDs, providing a good theoretical basis for further research on NTDs. This finding represents a novel epigenetic mechanism underlying that the m6A modification has profound and lasting implications for neural tube development.

Pre-insertion resistors (PIR) are critical components for suppressing the closing inrush current and operating overvoltage during the closing process of the circuit breaker. And it is widely adopted in 800kV and above transmission lines and alternating current filters of converter substations. These special application environments lead to higher failure rates than other electrical components. However, in the daily operation and fault diagnosis of PIR, there is no systematic method to analyze whether there are defects in PIR and the cause of failure after the PIR fails. Based on two PIR failure in series and parallel structures, this paper proposes methods such as dynamic resistance fitting, acoustic and vibration signal detection, insulation performance test, and heat capacity test during operation. In the aspect of fault diagnosis, methods such as PIR splicing based on Iterative Closest Point , stress analysis, and electric field simulation are proposed. Finally, proposed methods can be adjusted to estimate the operating state of PIR through the disassembly inspection to avoid the GIS breakdown accident during the operation of the circuit breaker.

This work presents a silicon-based capacitively transduced width extensional mode (WEM) MEMS rectangular plate resonator with quality factor (Q) of over 10,000 at a frequency of greater than 1 GHz. The Q value, determined by various loss mechanisms, was analyzed and quantified via numerical calculation and simulation. The energy loss of high order WEMs is dominated by anchor loss and phonon-phonon interaction dissipation (PPID). High-order resonators possess high effective stiffness, resulting in large motional impedance. To suppress anchor loss and reduce motional impedance, a novel combined tether was designed and comprehensively optimized. The resonators were batch fabricated based on a reliable and simple silicon-on-insulator (SOI)-based fabrication process. The combined tether experimentally contributes to low anchor loss and motional impedance. Especially in the 4th WEM, the resonator with a resonance frequency of 1.1 GHz and a Q of 10,920 was demonstrated, corresponding to the promising f × Q product of 1.2 × 1013. By using combined tether, the motional impedance decreases by 33% and 20% in 3rd and 4th modes, respectively. The WEM resonator proposed in this work has potential application for high-frequency wireless communication systems.

Market competition is considered to have a significant impact on product diversification, but related issues are rarely discussed on a city scale. To analyze the diversification of export commodities and export market of 270 Chinese cities, this study uses data from 2000 to 2017 based on the commodity concentration index, market concentration index, export similarity index, spatial stratified heterogeneity, and the Almon lag model. The study’s findings are: (1) The diversification of exports in most Chinese cities increased, which was higher in more developed cities in the southeast than in less developed ones in the northwest. With time, the degree of commodity and market diversification in some developed southeast Chinese cities (e.g., Shanghai) declined. This indicates the difference in and complexity of the evolution of export development in hundreds of Chinese cities between 2000 and 2017. (2) The export competition between cities became increasingly fierce, which effectively urged most of them to improve their export diversification levels. Facing increasing export competition pressure, 81.11% of the total number of cities will enhance the diversification of export commodities to cope with challenges posed by other cities. But only 56.67% will further expand the export market when the pressure of export competition increases. The biggest contribution of this study is the finding that for most Chinese cities, when export competition from other domestic cities increases, increasing diversification of products becomes a wise choice. However, the influence of competition on the diversification degree of the city’s export market is relatively weak. This study provides not only a new perspective for existing research on urban export trade, but also valuable information for cities to form a more profitable and robust trading system.

Aiming at the noise control of the HVDC converter station, a one-dimensional two-port metamaterial muffler based on the acoustic slow-wave effect is designed and manufactured. The metamaterial muffler achieves a broadband quasi-perfect absorption of noise from 600 to 900 Hz while ensuring a certain ventilation capacity. In addition, the internal equivalent sound velocity curve and the sound pressure and velocity field of the muffler are used to reveal the mechanism of its broadband quasi-perfect sound absorption. The performance of the muffler was verified by theoretical, numerical, and experimental models. The work in this paper is of guiding significance for solving the noise problem in HVDC converter stations.

As the only renewable carbon resource on Earth, lignocellulosic biomass is abundant in reserves and has the advantages of environmental friendliness, low price, and easy availability. The pyrolysis of lignocellulosic biomass can generate solid biochar with a large specific surface area, well-developed pores, and plentiful surface functional groups. Therefore, it can be considered as a catalyst for upgrading the other two products, syngas and liquid bio-oil, from lignocellulosic biomass pyrolysis, which has the potential to be an alternative to some non-renewable and expensive conventional catalysts. In addition, as another carbon resource, waste plastics can also use biochar-based catalysts for catalytic pyrolysis to solve the problem of accumulation and produce fuels simultaneously. This review systematically introduces the formation mechanism of biochar from lignocellulosic biomass pyrolysis. Subsequently, the activation and modification methods of biochar catalysts, including physical activation, chemical activation, metal modification, and nonmetallic modification, are summarized. Finally, the application of biochar-based catalysts for lignocellulosic biomass and waste plastics pyrolysis is discussed in detail and the catalytic mechanism of biochar-based catalysts is also investigated.

The inositol polyphosphate-5-phosphatase E (Inpp5e) gene is located on chromosome 9q34.3. The enzyme it encodes mainly hydrolyzes the 5-phosphate groups of phosphatidylinositol (3,4,5)-trisphosphate (PtdIns (3,4,5) P3) and phosphatidylinositol (4,5)-bisphosphate (PtdIns (4,5)P2), which are closely related to ciliogenesis and embryonic neurodevelopment, through mechanisms that are largely unknown. Here we studied the role of Inpp5e gene in ciliogenesis during embryonic neurodevelopment using inositol-deficiency neural tube defects (NTDs) mouse and cell models. Confocal microscopy and scanning electron microscope were used to examine the number and the length of primary cilia. The dynamic changes of Inpp5e expression in embryonic murine brain tissues were observed during Embryonic Day 10.5–13.5 (E 10.5–13.5). Immunohistochemistry, western blot, polymerase chain reaction (PCR) arrays were applied to detect the expression of Inpp5e and cilia-related genes of the embryonic brain tissues in inositol deficiency NTDs mouse. Real-time quantitative PCR (RT-qPCR) was used to validate the candidate genes in cell models. The levels of inositol and PtdIns(3,4) P2 were measured using gas chromatography-mass spectrometry (GC-MS) and enzyme linked immunosorbent assay (ELISA), respectively. Our results showed that the expression levels of Inpp5e gradually decreased in the forebrain tissues of the control embryos, but no stable trend was observed in the inositol deficiency NTDs embryos. Inpp5e expression in inositol deficiency NTDs embryos was significantly decreased compared with the control tissues. The expression levels of Inpp5e gene and the PtdIns (3,4) P2 levels were also significantly decreased in the inositol deficient cell model. A reduced number and length of primary cilia were observed in NIH3T3 cells when inositol deficient. Three important cilia-related genes (Ift80, Mkks, Smo) were down-regulated significantly in the inositol-deficient NTDs mouse and cell models, and Smo was highly involved in NTDs. In summary, these findings suggested that down-regulation of Inpp5e might be associated with abnormal ciliogenesis during embryonic neurodevelopment, under conditions of inositol deficiency.

Based on well logging responses, sedimentary patterns and sandstone thickness, the distribution characteristics of meandering river sedimentary sand body of Neogene Minghuazhen Formation NmIII2 layer in the west of Shijiutuo Bulge, Chengning Uplift, Bohai Bay Basin were investigated. A new approach to calculate the occurrence of the sand-mudstone interfaces using resistivity log of horizontal well was advanced to solve the multiple solution problem of abandoned channel's orientation. This method uses the trigonometric function relationship between radius, dip and length of the resistivity log to calculate the occurrence qualitatively – quantitatively to help determine the true direction of the abandoned channels. This method can supplement and improve the architecture dissection technique for meandering river sandbodies. This method was used to study the dip angle and scale of the lateral accretion layers in point bar quantitatively to help determine the spatial distribution of lateral accretion layers. The fine architecture model of underground meandering river reservoir in the study area has been established. Different from traditional grids, different grid densities for lateral accretion layers and bodies were used in this model by non-uniform upscaling to establish the inner architecture model of point-bars and realize industrial numerical simulation of the whole study area. The research results can help us predict the distribution of remaining oil, tap remaining oil, and optimize the waterflooding in oilfields. The reservoir architecture analysis of braided rivers, especially falling-silt seam forms, has played a key role in predicting remaining oil distributions. However, no studies have used architecture analyses that document braided river outcrops and researched the tapping of the few remaining oil distributions based on outcrops in the Songliao basin, northeast China. In this paper, the architecture characteristics and remaining oil distribution of braided river reservoirs are studied using a combination of an outcrop, modern deposition and subsurface well data. The new 8–13 m thick Lower Cretaceous Quantou Formation outcrop of the Songliao basin is a braided fluvial succession arranged in one large fining-upward cycle. Eight facies (Gt, St, Sm, Sh, Sp, Sw, Fl and Fm), four architecture elements (CH, DA, LV, and FF), and three orders of bounding surfaces (third-, fourth-, and fifth-order) are recognized. A new distribution pattern of falling-silt seams and a braided river architecture model are presented according to the analysis of the outcrop. In the mid-channel bar, the falling-silt seams thin from the mid-bar to the bar tail following the flow direction. Each falling-silt seam is oriented tangentially to the basal surface of the mid-channel bar, and the upper falling-silt seam extends farther than the lower one. In a Daqing Oilfield exploitation block in the Songliao basin, while channels and bars are the main reservoir units, they have different remaining oil distribution patterns. For bars, water injection wells located at the mid-bar, zonal injection technology, the drilling of horizontal wells, and proper well patterns are proposed. Fourth-order bounding surfaces, single braided channels, stacking patterns, and the lateral blocking of levees and floodplains are the key factors affecting the remaining oil distribution in channels.

Abnormal development of central nervous system (CNS) caused by neural tube defects (NTDs) is not only remained the major contributor in the prevalence of stillbirths and neonatal deaths, but also represents a significant cause of lifelong physical disability in the surviving infants. Ethionine is a non-proteinogenic amino acid and antagonist of methionine. Methionine cycle is essential for the elimination of reactive oxygen species (ROS), while lysosomes are involved in the initiation of autophagy. However, its role in ethionine-induced cell death in neural tube defects, still need to be explored. In this study, we investigated the effect of ethionine on NTDs as well as the underlying mechanism involved in this process. Following the establishment of NTDs model using ethionine-induced C57BL/6 mice, ethionine was intraperitoneally injected at a dose of 500 mg/kg in E7.5. Our study revealed that ethionine has induced mitochondrial apoptosis in NTDs by reducing mitochondrial autophagy both in vivo and in vitro. These results provided a possible molecular mechanism for redox regulation of autophagic process.

Abstract Background Human leukocyte antigen (HLA)‐DP is an HLA class II molecule. Overexpression of HLA class II molecules in placental trophoblast cells may induce pregnancy loss. However, the association between HLA‐DP and pregnancy loss remains unclear. HLA‐DPA1 is an HLA‐DP peptide chain. The objective of this study was to assess the association between HLA‐DPA1 genetic polymorphism and anembryonic pregnancy, a type of early pregnancy loss, in the Chinese population. Methods A case–control study was designed to compare the frequencies of HLA‐DPA1 gene polymorphisms in an anembryonic pregnancy group and a control group. Sixty‐eight cases and 122 controls were recruited. Statistical analysis was performed to assess the correlation between single‐nucleotide polymorphisms (SNPs) and anembryonic pregnancy susceptibility. MassARRAY high‐throughput DNA analysis was used to analyze 19 HLA‐DPA1 SNPs. To explore how HLA‐DPA1 polymorphism could affect anembryonic pregnancy, HLA‐DPA1 serum levels were analyzed by ELISA. Results Homozygous typing of rs1431403 (CC and TT) significantly increased the risk of anembryonic pregnancy in the case group (ORCC = 3.13, 95% CI: 1.50–6.53; ORTT = 2.96, 95% CI: 1.31–6.66; ORCC+TT = 3.06, 95% CI: 1.62–5.78). In samples with high HLA‐DPA1 levels (≥1,500 pg/ml), the homozygous rs1431403 genotypes (nCC = 21, 43.8%; nTT = 20, 57.1%) were observed more frequently than were heterozygous genotypes. Conclusion HLA‐DPA1 rs1431403 may be a risk factor for anembryonic pregnancy in the Chinese population. Homozygous rs1431403 genotypes (CC and TT) may increase the risk of anembryonic pregnancy by aberrantly increasing the HLA‐DPA1 levels.

IntroductionDiabetes is a metabolic disease with a high incidence and serious harm to human health. Islet β-cell function defects can occur in the late stage of type 1 diabetes and type 2 diabetes. Studies have shown that stem cell is a promising new approach in bioengineering regenerative medicine. In the study of stem cell differentiation, three-dimensional (3D) cell culture is more capable of mimicking the microenvironment of cell growth in vivo than two-dimensional (2D) cell culture. The natural contact between cells and cells, and cells and extracellular matrix can regulate the development process and promote the formation of the artificial regenerative organs and organization. Type IV, VI collagen and laminin are the most abundant extracellular matrix components in islets. Matrigel, a basement membrane matrix biomaterial rich in laminin and collagen IV.Materials and MethodsWe used Matrigel biomaterial to physically embed human dental pulp stem cells (hDPSCs) to provide vector and 3D culture conditions for cells, and we explored and compared the preparation methods and preliminary mechanisms of differentiation of hDPSCs into insulin-producing cells (IPCs) under 2D or 3D culture conditions.We first designed and screened the strategy by mimicking the critical events of pancreatogenesis in vivo, and succeeded in establishing a new method for obtaining IPCs from hDPSCs. Activin A, Noggin, and small molecule compounds were used to synergistically induce hDPSCs to differentiate into definitive endoderm-like cells, pancreatic progenitor like cells and IPCs step by step under 2D culture conditions. Then, we used Matrigel to simulate the microenvironment in vivo, induced hDPSCs to differentiate into IPCs in Matrigel, evaluated and compared the efficiency between 2D and 3D culture conditions.ResultsThe results showed that the synergistic combination of growth factors and small molecule compounds and 3D culture promoted the differentiation of hDPSCs into IPCs, significantly enhancing the release of insulin and C-peptide from IPCs.DiscussionSignificant support is provided for obtaining a large number of functional IPCs for disease modeling and final cell therapy in regenerative medicine.

In recent years, the impact of land-use systems on global climate change has become increasingly significant, and land-use change has become a hot issue of concern to academics, both within China and abroad. Urbanization, as an important socioeconomic factor, plays a vital role in promoting land-use transition, which also shows a significant spatial dependence on urbanization. This paper constructs a theoretical framework for the interaction relationship between urbanization and land-use transition, taking the Yangtze River Delta as an example, and measures the level of urbanization from the perspective of population urbanization, economic urbanization and social urbanization, while also evaluating the level of land-use morphologies from the perspective of dominant and recessive morphologies of land-use. We construct a PVAR model and coupled coordination model based on the calculated indexes for empirical analysis. The results show that the relationship between urbanization and land-use transition is not a simple linear relationship, but tends to be complex with the process of urbanization, and reasonable urbanization and land-use morphologies will promote further benign coupling in the system. By analyzing the interaction relationship between urbanization and land-use transition, this study enriches the study of land-use change and provides new pathways for thinking about how to promote high-quality urbanization.

Bacillus Calmette-Guerin (BCG) is a potent agent for the prevention of tuberculosis. Current studies have regarded BCG as an immunomodulator. However, there is little information on whether it can be used to inhibit airway inflammation and airway remodeling caused by asthma. Therefore, in this study, we investigate the role of epithelial–mesenchymal transition (EMT) in airway inflammation and airway remodeling as well as the possible therapeutic mechanism of BCG for the treatment of asthma. Wistar rats were sensitized and challenged by ovalbumin for 2 weeks or 8 weeks. BCG was subcutaneously administered daily before every ovalbumin challenge to determine its therapeutic effects. The 2 weeks model group showed extensive eosinophilia, chronic inflammatory responses, bronchial wall thickening, airway epithelium damage, increased levels of transforming growth factor β 1 (TGF-β1) in both bronchoalveolar lavage fluid and sera, decreased expression of epithelial marker E-cadherin, and increased expressions of mesenchymal markers α-smooth muscle actin (α-SMA) and Fibronectin (Fn). Except for inflammatory responses, all responses were more significant in the 8 weeks model group which displayed characteristics of airway remodeling including subepithelial fibrosis, smooth muscle hypertrophy, and goblet cell hyperplasia. When compared with the model groups, BCG administration inhibited airway inflammation and airway remodeling, decreased TGF-β1 levels, upregulated expression of E-cadherin, and downregulated expression of α-SMA and Fn. The present study suggests for the first time that increased secretion of TGF- β1 induced by asthmatic chronic inflammation may result in EMT, which is one of the most important mechanisms of airway inflammation and airway remodeling seen with asthma. BCG alleviates airway inflammation and airway remodeling by preventing TGF-β1 induced EMT, therefore BCG may be a new therapy for treating asthma.

Abstract A reduction in the particle size is expected to improve the properties and increase the application potential of high-entropy alloys. Therefore, in this study, a novel sol–gel autocombustion technique was first used to synthesize high-entropy alloys. The average grain size of the prepared nanocrystalline CoCrCuNiAl high-entropy alloys showed was 14 nm with an excellent and uniform dispersion, exhibiting a distinct magnetic behavior similar to the superparamagnetic behavior. We show that the metal nitrates first form (Co,Cu,Mg,Ni,Zn)O high-entropy oxides, and then in situ reduce to CoCrCuNiAl high-entropy alloys by the reducing gases, and the chelation between citric acid and the metal ions and the in situ chemical reactions are the dominant reaction mechanisms. We demonstrate that the sol–gel autocombustion process is an efficient way to synthesize solid solution alloys eluding the restriction of a high mixing entropy.

Myo-inositol (MI) is one of the stereoisomers of hexahydroxycyclohexane, which plays an important role in intracellular signal pathway. Derivatization is an indispensable step in both external and internal standard method during the chromatography-mass spectrometer (GC–MS) detection, as MI can’t be ionized directly. It is valuable to optimize the derivative process and the detection volume for clinical detection. By single-gear pair contact analysis, this paper, based on the Hertz theory, gets the mesh density of finite element solver in convergence case and then studies contact spots with multi-gear pairs. Through simulation analysis, this paper studies the face gear transmission error in different conditions, and this paper studies the face gear transmission error in different conditions. Then come to a conclusion that: In a set of three different load cases, the greater the torque load is applied to the face gear, the greater the transmission error there is, but the speed of face gear almost has no effect on the gear transmission error. When the face gear is installed with shaft angle of 0.02°, its transmission error magnitude will be 2.0~2.3 times bigger compared with installed without any error. The study can provide guidance for the design, production and practical application of the face gear.

Neural tube defects (NTDs) are severe, common birth defects that result from failure of neural tube closure, but their pathological mechanisms are not yet fully understood. Histone modifications have an important role in gene regulation during fetal development. We therefore hypothesized that the human NTDs may be partly caused by an imbalance in metabolism, perhaps caused by nutritional deficiencies, that leads to aberrant histone modifications. Here, we report a screen of fetal brain histone modifications using 2D nano-LC strong cation exchange reverse phase (SCX/RP) MS/MS and the identification of 61 unique post-translational modification sites on histones H1, H2a, H2b, H3, and H4. Of these, 38 sites are novel (not already found in the Uniprot database). Furthermore, we compared the histone modification patterns between normal brains and NTD brains special of which maternal folate levels were lower than of normal control. The results showed that histone H3 lysine 79 dimethylation (H3K79me2) and a novel identified site, H2bK5 monomethylation (H2bK5me1), were completely absent in individuals with NTDs. Follow-up Western blotting validated the decreased H3K79me2 expression in brains with NTDs, but the amplified samples experiments displayed that decreased H3K79me2 expression was not suitable for all samples with NTDs. Furthermore, folate-free treated mouse embryonic stem cells induced the decreased H3K79me2 level. Subsequently, our ChIP results in normal fetal brain tissues showed that H3K79me2 binds to SUFU, RARA and ITGA3 which induce NTDs phenotype after knockout in mice, and in NTDs brain tissues the bindings of H3K79me2 to these three genes were significantly altered. Taken together, our study indicated that low folate treatment might attenuate H3K79 dimethylation, further affect its regulate activation on target genes, some of which are NTDs-resulting associated, lastly interrupt early embryo developing. Our study increases the understanding of normal fetal brain histone modifications and provides a platform for investigating histone modifications in neural disease and also has an insight into a potential role of aberrant histone modification in etiology of NTDs.

OBJECTIVE To observe enhancement of anti-tumor immunity by gene vaccine using nucleofection technology.METHODS The technique of nucleofection was used to transfereffectively plasmid DNA into immature dendritic cells (iDCs); we studied immune responses regulated by DNA vaccine using real-time quantitative polymerase chain reaction (PCR) and Western-blotting to optimize the follow-up lymphocyte activation. The anti-tumor capacity of lymphocytes primed by DCs was analyzed using lactate dehydrogenase with a non-radioactive cytotoxicity assay.RESULTS Human monocyte-derived dendritic cells (hMoDCs) were induced by interleukin (IL)-4 and granulocyte-macrophage colony-stimulating factor (GM-CSF) in vitro from human monocytes for 5 or 6 days. DNA vaccine was transfected to iDCs with high transfection (35.73%) using nucleofection. Compared with the iDC group, the expression of Th1 cell cytokine IL-12, IL-18 and Th2 cell cytokine IL-4 increased after stimulation. CD86 and CD83 were upregulated compared with non-nucleofected groups 48 hours after nucleofection with DC-pVAX-PRA. The result of the cytotoxicity assay showed that DCs-pVAX-PRA primed non-adherent peripheral blood mononuclear cells (PBMCs) exhibit their highest cytotoxicity against target cells.CONCLUSION The results show that DNA vaccine was transfected to iDC with high transfection efficiency using nucleofection, priming autologous lymphocytes for anti-tumor immunity by upregulated expression of co-stimulatory molecules, adhesion molecules and cytokines. These results provided a basis to explore the molecular mechanism of DNA vaccine in vivo.

OBJECTIVE To observe anti-tumor effects of PVAX-PSMA gene vaccine. METHODS The PSMA gene was inserted into a mammalian expression vector, PVAX-1, to construct the DNA vaccine candidate, and was then used to vaccinate C57BL/6 mice. Animals vaccinated with PVAX-1 and NaCl were used as controls. Anti-PSMA antibody was detected in sera of the animals. The proliferation and cytotoxicity of the spleen cells were observed. The immunized mice were inoculated with RM-1 cells. The mice were inoculated with RM-1 cells, and then the mice were immunized. The anti-tumor efficacy of the gene vaccine was evaluated by the ratio of tumor formation, tumor volume, tumor mass before and a er gene vaccination and evaluated by survival rate of the immunized mice.RESULTS High level of anti-PSMA antibody was induced in the PVAX-PSMA group. The splenocytes from PVAX-PSMA group were stimulated to produce strong proliferation responses and significant cytotoxic T-cells (CTL) activity. After the mice were immunized with PVAX-PSMA gene, tumor occurrence was decreased, and the growth velocity of tumor was markedly reduced, resulting in prolonged tumor-free time (P < 0.05).CONCLUSION PVAX-PSMA gene vaccine has significant anti- tumor effects and provides an experimental basis for primary prevention and immunotherapy of prostate cancer.

DNA methylation is thought to be involved in the etiology of neural tube defects (NTDs). However, the exact mechanism between DNA methylation and NTDs remains unclear. Herein, we investigated the change of methylation in mouse model of NTDs associated with folate dysmetabolism by use of ultraperformance liquid chromatography tandem mass spectrometry (UPLC/MS/MS), liquid chromatography-electrospray ionization tandem mass spectrometry (LC-MS/MS), microarray, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and Real time quantitative PCR. Results showed that NTD neural tube tissues had lower concentrations of 5-methyltetrahydrofolate (5-MeTHF, P = 0.005), 5-formyltetrahydrofolate (5-FoTHF, P = 0.040), S-adenosylmethionine (SAM, P = 0.004) and higher concentrations of folic acid (P = 0.041), homocysteine (Hcy, P = 0.006) and S-adenosylhomocysteine (SAH, P = 0.045) compared to control. Inositol 1,3,4-trisphosphate 5/6-kinase (ITPK1) is a pivotal regulatory enzyme in inositol metabolic pathway. The objective was to assess the potential impact of the maternal ITPK1 genotypes on the inositol parameter and on the NTD risk in a NTD high-risk area in China. Human umbilical cord-derived mesenchymal stem cell (UC-MSC) transplantation has shown promise for the treatment of various diseases. For clinical applications, UC-MSCs have been stored in 0.9% saline, 5% dextrose, dextrose and sodium chloride injection, Plasma-Lyte A, 1% human serum albumin (1% HSA), or 5% HSA before administration, but the effect of storage conditions on the viability and biological function of the cells remains unknown. Freshly harvested UC-MSCs were resuspended and incubated in these solutions for 2, 4, or 6 h at 4°C or room temperature (24°C). Cell viability, apoptotic/necrotic fraction, poststorage growth potential, immunophenotype, immunosuppressive capacity, and differentiation capacity were analyzed. When stored in parenteral solutions, UC-MSCs showed progressive deterioration in survival viability and adhesion ability. After 6-h storage, the best viability and attachment rate of UC-MSCs decreased to 83.0 ± 1.6% and 71.8 ± 3.2%, respectively. Our results suggested that UC-MSCs in these conditions lose their viability in a short time. However, it seems that the other biological functions of the surviving UC-MSCs were little affected. Since UC-MSCs suspended in these mediums lose their survival viability in a short time to levels significantly below the permissible limits (70%) by FDA, precautions need to be taken on using these solutions as suspension medium and further studies on the optimal methods for preservation are urgent.

BACKGROUND: Neural tube defects (NTDs) are one of the most common birth defects caused by a combination of genetic and environmental factors. Currently, little is known about the genetic basis of NTDs although up to 70% of human NTDs were reported to be attributed to genetic factors. Here we performed genome-wide copy number variants (CNVs) detection in a cohort of Chinese NTD patients in order to exam the potential role of CNVs in the pathogenesis of NTDs. METHODS: The genomic DNA from eighty-five NTD cases and seventy-five matched normal controls were subjected for whole genome CNVs analysis. Non-DGV (the Database of Genomic Variants) CNVs from each group were further analyzed for their associations with NTDs. Gene content in non-DGV CNVs as well as participating pathways were examined. RESULTS: Fifty-five and twenty-six non-DGV CNVs were detected in cases and controls respectively. Among them, forty and nineteen CNVs involve genes (genic CNV).
