Differential object marking (DOM) is a common feature of many languages, including Standard Turkish and Urum, a Turkish variety spoken by ethnic Greeks in Georgia. This article investigates the interaction between case marking and word order in both varieties of Turkish. While Standard Turkish shows a strong dependence of the case marking possibilities from word order, i.e. bare objects may only appear in immediately preverbal position of a clause, the analysis reveals that bare objects in Urum may occur in any position of a clause. This provides evidence that the verb in Urum can freely move within the VP.

Cell growth and proliferation are processes in the cell that must be tightly regulated. Transcription of ribosomal RNA and ribosomal biogenesis are directly linked to cell growth and proliferation, since the ribosomal RNA encodes for the majority of transcription in a cell and ribosomal biogenesis influences directly the number of proteins that are synthesized. In the work presented in this thesis, we have investigated the ribosomal RNA genes, namely the ribosomal DNA genes and the 5S rRNA genes, and their transcriptional regulation. One protein complex that is involved in RNA polymerase I and III transcription is the chromatin remodelling complex B‑WICH (WSTF, SNF2h, NM1). RNA polymerase I transcribes the rDNA gene, while RNA polymerase III transcribes the 5S rRNA gene, among others. In Study I we determined the mechanism by which B‑WICH is involved in regulating RNA polymerase I transcription. B‑WICH is associated with the rDNA gene and was able to create a more open chromatin structure, thereby facilitating the binding of HATs and the subsequent histone acetylation. This resulted in a more active transcription of the ribosomal DNA gene. In Study II we wanted to specify the role of NM1 in RNA polymerase I transcription. We found that NM1 is not capable of remodelling chromatin in the same way as B‑WICH, but we demonstrated also that NM1 is needed for active RNA polymerase I transcription and is able to attract the HAT PCAF. In Study III we investigated the intergenic part of the ribosomal DNA gene. We detected non-coding RNAs transcribed from the intergenic region that are transcribed by different RNA polymerases and that are regulated differently in different stress situations. Furthermore, these ncRNAs are distributed at different locations in the cell, suggesting that they have different functions. In Study IV we showed the involvement of B‑WICH in RNA Pol III transcription and, as we previously had shown in Study I, that B‑WICH is able to create a more open chromatin structure, in this case by acting as a licensing factor for c-Myc and the Myc/Max/Mxd network. Taken together, we have revealed the mechanism by which the B‑WICH complex is able to regulate RNA Pol I and Pol III transcription and we have determined the role of NM1 in the B‑WICH complex. We conclude that B‑WICH is an important factor in the regulation of cell growth and proliferation. Furthermore, we found that the intergenic spacer of the rDNA gene is actively transcribed, producing ncRNAs. Different cellular locations suggest that the ncRNAs have different functions. Beim fortgeschrittenen malignen Melanom ist nicht selten eine radikale axilläre oder ilioinguinale Lymphadenektomie erforderlich. Intraoperativ wird eine Saugdrainage in den Situs gelegt, um die Lymphe abzuleiten. Nach der Operation treten häufig Lymphfisteln über die Drainage oder punktionsbedürftige Serome auf. Üblicherweise werden die Drainagen erst entfernt, wenn sie weniger als 50 ml pro Tag fördern. Im Rahmen einer Studie sollte überprüft werden, ob die Rate an Seromen und Wundinfektionen reduziert werden kann, wenn die Drainage länger in situ belassen wird. Dazu wurden zwei Zeitintervalle miteinander verglichen. Von 2003 bis 2007 wurden die subkutanen Saugdrainagen spätestens am 8. postoperativen Tag unabhängig von der drainierten Flüssigkeitsmenge entfernt und von 2008 bis 2011 blieben sie unbegrenzt liegen, bis die Lymphfistel sistierte. Im Zeitraum von 2003 bis 2011 wurden 553 Lymphadenektomien bei einem Mela-nom erfasst. Es wurden 221 axilläre und 153 ilioinguinale Lymphknotendissektionen getrennt ausgewertet und die Ergebnisse beider Zeitintervalle miteinander verglichen. Nach axillärer Lymphadenektomie unterschieden sich die Patienten in den beiden Zeitintervallen hinsichtlich grundlegender Charakteristika nicht. Die Saugdrainagen wurden im ersten Zeitintervall im Median nach 6 Tagen gezogen und im zweiten nach 20 Tagen (p, In patients with advanced melanoma radical lymphadenectomy of the axilla or groin is often necessary for cure. A suction drain is usually placed after complete resection to remove all lymphatic fluid from the wound. Lymphatic fistula and seroma formation do often occur so that the drainage is only removed if less than 50 cc is secreted. The purpose of study was to prove that seroma and wound infections are decreased if the drain is in situ as long as lymphatic fluid is produced. Two time intervals of time were compared. From 2003 to 2007 the suction drain is removed after at least 8 days, irrespective of the amount of fluid secreted. This concept was changed and from 2008 to 2011 the drain was kept in place until lymphorrhea was less than 50 cc per day. 553 lymphadenectomy of the melanoma from 2003 to 2011 were documented in a prospective study. 221 axillary lymphadenectomies and 153 lymphadenectomies were included and separately analyzed. The two groups of patients of both time intervals were not different after axillary lymphadenectomy. The operative time was 101±33 minutes and not varying between time intervals. At the second time interval, more lymph nodes were examined (p=0,001) and more lymph node metastasis were found (p=0,002). Dendritic cells (DC) are known to be important orchestrators of the immune response, resulting in an effective adaptive immunity against pathogens. In therapeutic cancer vaccination, antigen (Ag)-loaded DC are used to initiate T-cell responses against tumors. An optimal cancer vaccine should induce a strong CD8+ CTL response and also promote T-cell help to support an effective CD8+ T-cell memory formation. However, the underlying cellular interactions of DC, T-helper cells, and CD8+ T cells are incompletely understood, notably in humans. Hence, the aim of this study was to generate human autologous Ag-specific in-vitro-systems to examine the communication between these cells and the effects of T-cell help on CTL priming vs. subsequent expansion. To make the control of antigen-specific interactions between DC and CD4+ T cells feasible, the specificity of the T cells was redirected by TCR-RNA electroporation. To obtain TCRs for equipping T cells with a new specificity, a fast and robust PCR-based method to clone functional TCRs was established. By the addition of a defined 5’ end, the cloning of TCR-chains without previous knowledge of the TCR-chain subtypes was enabled. The subsequent functional validation in a newly developed Jurkat T-cell assay showed that this assay is indicative for the functionality of the newly cloned TCRs in ex vivo isolated T cells, since the results correlated with the cytokine release, functional avidity, proliferative capacity, and tumor-cell recognition of freshly isolated TCR-transfected primary T cells. Consequently, the PCR-based cloning strategy in combination with the informative Jurkat T-cell assay provides a fast and low-cost method to clone and validate a TCR derived from T-cell clones. The Ag-specific cross-talk between human resting CD4+ T cells, reprogrammed by TCR-RNA transfection, and autologous peptide-loaded DC initiated Th1 cytokine secretion and induced T-cell activation and DC maturation. Moreover, the two suggested T-cell help models, i.e. the two-cell interaction model and the three-cell interaction model, were mimicked to elucidate the role of T-cell help in CD8+ T-cell priming and subsequent expansion. The sequential interaction of the DC, first with specific helper T cells to elicit DC licensing, and then, after these had been removed from the co-culture, with CD8+ T cells, barely improved CTL priming or expansion. In sharp contrast, a simultaneous interaction of all three cell types resulted in a significantly improved CTL expansion upon re-stimulation. Yet, this was only detectable, when both epitopes – the helper and the CD8 epitope – were presented by the same DC. In addition, the Ag-specific CD8+ T cells, generated in the presence of help provided in this three-cell interaction setting, showed a sustained CD27 expression, indicating memory formation. In conclusion, these findings might have several implications for DC vaccination strategies, since this human in vitro model system can be used to gain new insights in the molecular mechanisms involved in T-cell help, which then can be simulated by optimized DC vaccines. Dendritische Zellen (DC) sind für ihre Funktion als wichtige Organisatoren für Immunantworten bekannt, was in eine effiziente Einleitung der adaptiven Immunität gegen Pathogene resultiert. Im therapeutischen Ansatz der Krebsimpfung werden antigenbeladene DCs verwendet um T-Zell-Antworten gegen Tumore zu induzieren. Eine optimale Krebsimpfung sollte sowohl eine starke CD8+ CTL (zytotoxische T-Lymphozyten) Antwort induzieren, als auch T-Zell-Hilfe für eine effektive Generierung von CD8+ Gedächtniszellen fördern. Die zu Grunde liegenden zellulären Mechanismen der Interaktion zwischen DCs, T-Helferzellen und CD8+ T-Zellen sind jedoch insbesondere im humanen System nur unvollständig erfasst. Demzufolge war das Ziel dieser Arbeit die Generierung von humanen autologen antigenspezifischen In-vitro-Systemen, um die Kommunikation zwischen diesen Zellen und die Effekte der T-Zell-Hilfe auf das Primen von CTLs gegenüber der darauf folgenden Expansion zu untersuchen. Um die Kontrolle der antigenspezifischen Interaktion zwischen DCs und CD4+ T-Zellen zu ermöglichen, wurde die T-Zell-Spezifität durch Elektroporation von TCR (T-Zell-Rezeptor)-RNA umprogrammiert. Eine schnelle und stabile PCR-basierte Methode zur Klonierung funktioneller TCRs wurde etabliert um TCRs für die Ausstattung der T-Zellen mit einer neuen Spezifität zu erhalten. Durch das Anhängen eines definierten 5’ Endes wurde die Klonierung von TCR-Ketten ohne vorherige Kenntnis der Subtypen dieser TCR-Ketten ermöglicht. Die anschließende Validierung im neu entwickelten Jurkat T-Zell-System ergab, dass das Testsystem indikativ für die Funktionalität der neu klonierten TCRs in ex vivo isolierten T-Zellen ist, da dessen Ergebnisse mit der Zytokinsekretion, der funktionellen Avidität, der Proliferationskapazität und Tumorzellerkennung von frisch isolierten TCR-transfizierten primären T-Zellen korrelierten. Folglich stellt die PCR-basierte Klonierstrategie, in Kombination mit dem informativen Jurkat T-Zell-System, eine schnelle und kostengünstige Methode zur Klonierung und Validierung von TCRs, die von T-Zell-Klonen abstammen, dar. Der antigenspezifische Cross-Talk zwischen humanen ruhenden CD4+ T-Zellen, die durch TCR-RNA Transfektion umprogrammiert wurden, und autologen peptidbeladenen DCs initiierte eine Th1-Zytokinsekretion und induzierte sowohl T-Zell-Aktivierung, als auch DC-Reifung. Zudem wurden die zwei T-Zell-Hilfe-Modelle, d.h. das Zwei-Zell-Interaktions-Modell und das Drei-Zell-Interaktions-Modell, nachgeahmt, um die Rolle der T-Zell-Hilfe für das Primen der CD8+ T-Zellen und die anschließende Expansion aufzuklären. Die sequentielle Interaktion der DC, zuerst mit der spezifischen T-Helferzelle um eine DC Lizenzierung hervorzurufen und danach mit der CD8+ T-Zelle, nachdem die T-Helferzelle bereits aus der Ko-Kultur entfernt wurde, verbesserte das Primen und die Expansion der CTLs kaum. Im scharfen Gegensatz dazu resultierte die gleichzeitige Interaktion aller drei Zelltypen in eine signifikante Verbesserung der CTL Expansion nach Restimulation. Jedoch war dies nur detektierbar, wenn beide Epitope - das Helfer- und das CD8-Epitop - auf derselben DC präsentiert wurden. Zudem zeigten die CD8+ T-Zellen, die unter Bereitstellung von Hilfe im Drei-Zell-Interaktions-Setting generiert wurden, eine anhaltende CD27-Expression, was auf eine Gedächtniszellbildung hinweist. Zusammenfassend haben diese Ergebnisse etliche Auswirkungen auf DC-Vakzinationsstrategien, da dieses humane In-vitro-Modell verwendet werden kann, um neue Einblicke in die molekularen Mechanismen, die in der T-Zell-Hilfe involviert sind, zu gewinnen. Diese können dann in optimierten DC-Impfungen simuliert werden.

Seit Jahrhunderten wird in Märchen die Kultur und Geschichte von Völkern überliefert, zunächst mündlich, später in schriftlicher Form und in der heutigen Zeit auch in Verfilmungen. Viele Märchen existieren in den verschiedensten Versionen und sind den unterschiedlichsten Kulturen gemein. Um die Analyse von Märchen möglich zu machen, hat Propp eine Vielzahl von Schritten herausgearbeitet, die jedem Zaubermärchen gemein sind. So stellte er fest, dass den Zaubermärchen stets der Initiationsritus zugrunde liegt, welcher den Übergang vom Jugendlichen zum Erwachsenen kennzeichnet. Metzeltin und Thir stellten noch zwei weitere Riten heraus, welche in Märchen vorkommen: Die Brautwerbung und die Herrscherersetzung. Eines der berühmtesten Märchen unserer Zeit ist Die Schöne und das Biest. In unzähligen Büchern und Verfilmungen wurde diese Geschichte immer wieder neu erzählt. Fünf verschiedene Versionen dieses Zaubermärchens, das Volksmärchen La Belle Rose, das Kunstmärchen La Belle et la Bête von Mme Leprince de Beaumont, sowie Verfilmungen von Jean Cocteau, Eugene Marner und Walt Disney, werden in dieser Arbeit anhand der von Propp, Metzeltin und Thir vorgestellten Modelle analysiert und verglichen. Aufgrund der Analyse ist eindeutig zu erkennen, dass es sich in allen Fällen um Zaubermärchen handelt und eine charakteristische Grundstruktur sogar in der stark abgewandelten Zeichentrickverfilmung von Walt Disney erhalten blieb. Allen Werken liegt der Initiationsritus zugrunde, vor Allem wird das Biest initiiert, das ähnlich wie die Initianden antiker Jägerkulturen einige Zeit in Gestalt eines Tieres verbringen muss. Mit der Zeit trat jedoch immer mehr der Ritus der Brautwerbung in den Vordergrund. Die Beobachtung zeigt, dass die Riten immer noch existieren, ihre Bedeutung und ihr Schwerpunkt sich aber mit der Zeit verschoben haben und vor Allem Riten reflektiert werden, die in der aktuellen Kultur vorkommen., For centuries, fairy tales have conveyed the culture and history of people, first in oral tradition, later in written form and nowadays even in films. Many of those tales exist in a variety of versions and have common roots throughout different cultures. Propp has provided a means to analyse those fairy tales, constituting a number of elements that are common for magical tales. He found that all magical tales are based on the rites of initiation, which mark the turning point from adolescence to adulthood. Metzeltin and Thir added two more rites, which frequently occur in magical tales: the rites of courtship and the rites of investiture. Beauty and the Beast is one of the most popular fairy tales in our times. Countless books and films retell the story of a beautiful girl who falls in love with a monster. This paper compares five versions of the tale, namely La belle rose, the originally French folk tale, La Belle et la Bête, a literary version written by Mme Leprince de Beaumont, and the films by Jean Cocteau, Eugene Marner and Walt Disney. The analysis is based on the models created by Propp, Metzeltin and Thir. This analysis proves that each of those versions, even the extremely modified Walt Disney version, fulfils the criteria of the magical tale and is structured characteristically. Each is based on the initiation of the beast, whose enchantment reminds us of the ancient hunter societies, where the adolescents had to dress in animal skins during initiation. Gradually the rites of courtship gained more importance and replaced the rites of initiation. Analysis shows that the rites still exist, but their meaning and their importance varies. The focus generally lies on those rites that are still valid in modern societies.
