Research background. An innovative integrated bioprocess system for bioethanol production from raw sugar beet cossettes (SBC) and arabitol from remaining exhausted sugar beet cossettes (ESBC) was studied. This integrated three-stage bioprocess system is an example of the biorefinery concept to maximise the use of raw SBC for the production of high value-added products such as sugar alcohols and bioethanol. Experimental approach. The first stage of the integrated bioprocess system was simultaneous sugar extraction from SBC and its alcoholic fermentation to produce bioethanol in an integrated bioreactor system (vertical column bioreactor and stirred tank bioreactor) containing a high-density suspension of yeast Saccharomyces cerevisiae (30 g/L). The second stage was the pretreatment of ESBC with dilute sulfuric acid to release fermentable sugars. The resulting liquid hydrolysate of ESBC was used in the third stage as a nutrient medium for arabitol production by non-Saccharomyces yeasts (Spathaspora passalidarum CBS 10155 and Spathaspora arborariae CBS 11463). Results and conclusions. The obtained results show that the efficiency of bioethanol production increased with increasing temperature and prolonged residence time in the integrated bioreactor system. The maximum bioethanol production efficiency (87.22 %) was observed at a time of 60 min and a temperature of 36 °C. Further increase in residence time (above 60 min) did not result in the significant increase of bioethanol production efficiency. Weak acid hydrolysis was used for ESBC pretreatment and the highest sugar yield was reached at 200 °C and residence time of 1 min. The inhibitors of the weak acid pretreatment were produced below bioprocess inhibition threshold. The use of the obtained liqiud phase of ESBC hydrolysate for the production of arabitol in the stirred tank bioreactor under constant aeration clearly showed that S. passalidarum CBS 10155 with 8.48 g/L of arabitol (YP/S=0.603 g/g and bioprocess productivity of 0.176 g/(L.h)) is a better arabitol producer than Spathaspora arborariae CBS 10155. Novelty and scientific contribution. An innovative integrated bioprocess system for the production of bioethanol and arabitol was developed based on the biorefinery concept. This three-stage bioprocess system shows great potential for maximum use of SBC as a feedstock for bioethanol and arabitol production and it could be an example of a sustainable ‘zero waste’ production system.

Water kefir is a product obtained through the fermentation of sucrose solution, usually with some kind of dried fruit addition, by a combined culture of micro-organisms which are contained within kefir grains. Its popularity is rising because of the simplicity of its preparation and its anti-inflammatory, antioxidant, probiotic, and antibacterial effects. In this research, the water kefir production was studied in 250 mL jars, as well as in a horizontal rotating tubular bioreactor (HRTB). The first part of the research was conducted in smaller-scale (jars), wherein the optimal fruit and fruit portions were determined. These experiments included the addition of dried plums, apricots, raisins, dates, cranberries, papaya, and figs into 150 mL of initial sugar solution. Also, the optimal ratio between dried fruit and sucrose solution (0.2) at the beginning of the bioprocess was determined. The second part of this research was conducted using HRTB. The experiments in the HRTB were carried out by using different operational modes (constant or interval bioreactor rotation). A total of six different bioreactor setups were used, and in all experiments, figs were added at the beginning of the bioprocess (0.2 ratio between dried figs and sucrose solution). On the basis of the obtained results, the interval bioreactor rotation mode proved to be the better HRTB mode for the production of the water kefir, as the yield of the main fermentation products was higher, and their ratios were the most adequate for the quality of water kefir drink. The optimal results were obtained via HRTB setup 3/57 (3 min rotation, 57 min pause within 1 h) and rotation speed of 3 rpm. Furthermore, it is clear that HRTB has great potential for water kefir production due to the fact that HRTB experiments showed shorter fermentation times (at least five times) than water kefir production in jars.

Glavni proizvod anaerobne digestije je bioplin, koji je obnovljivo gorivo, a sporedni proizvod ovog procesa je digestat, koji se koristi kao gnojivo bogato hranjivim tvarima. Dodatni pozitivni učinci anaerobne digestije su razgradnja organskog otpada te smanjenje neugodnih mirisa i koncentracije patogenih mikroorganizama. Bioplin se uglavnom koristi za proizvodnju električne energije i topline, a u nekim slučajevima se pročišćava da bi se dobio biometan koji se koristi u mreži prirodnog plina, kao gorivo za motore s unutarnjim sagorijevanjem ili kao polazna kemikalija za kemijsku industriju. Zbog svega navedenog, razvoj proizvodnje bioplina ima pozitivne društveno-ekonomske i ekološke učinke. Bioplin proizveden u Hrvatskoj većinom se koristi za proizvodnju električne i toplinske energije u kogeneracijskim postrojenjima. Iako su u Hrvatskoj dostupne različite obnovljive sirovine koje bi se mogle iskoristiti za proizvodnju bioplina, njihov je potencijal do sada bio nedovoljno iskorišten. Kao sirovine za proizvodnju bioplina u nas se pretežno koriste gnojovka i nusproizvodi poljoprivrede, klaonica i prehrambene industrije. Racionalnijim korištenjem zemljišta i razvojem prehrambene industrije mogla bi se povećati količina poljoprivrednih ostataka i nusproizvoda koji nastaju preradom hrane. Usmjeravanjem i poticanjem korištenja ovih nusproizvoda za anaerobnu digestiju može se stimulirati brži razvoj proizvodnje bioplina u Hrvatskoj. Pored mogućeg povećanja vlastite proizvodnje električne energije i goriva, radi se o ekološki povoljnoj tehnologiji koja ima pozitivan društveno-ekonomski učinak.

Microalgal biomass is an excellent platform for producing food, feed, nutraceuticals, pharmaceuticals, and biofuels. This study aimed to investigate the effect of the trophic mode of cultivation (phototrophic, heterotrophic, and mixotrophic) on the growth and biomass composition of Chlorella vulgaris S2. The contents of lipids and carbohydrates, as well as the fatty acid composition of total lipids, were studied. The effects of the carbon-to-nitrogen ratio (C:N) and the organic carbon concentration of the growth media under mixotrophic and heterotrophic conditions were also investigated. The C:N ratio of 30 mol mol−1 favoured lipid synthesis, and the C:N ratio of 10 mol mol−1 favoured carbohydrate synthesis. Maximal lipid and biomass productivities (2.238 and 0.458 g L−1 d−1, respectively) were obtained under mixotrophic conditions at the C:N ratio of 50 mol mol−1 and glucose concentration of 50 g L−1. Fed-batch cultivation conducted in a stirrer tank bioreactor under heterotrophic growth conditions increased biomass (2.385 g L−1 d−1, respectively) and lipid (0.339 L−1 d−1) productivities ~50 and ~60 times compared to the fed-batch phototrophic cultivation, respectively. The trophic mode, growth phase, and growth medium composition significantly influenced the fatty acid composition. Under mixotrophic and heterotrophic growth conditions, lipid accumulation is associated with an increase in oleic acid (C18:1) content. Mixotrophically grown biomass of Chlorella vulgaris S2 under optimised conditions is a suitable source of lipids for biodiesel production.

Significant amounts of various residues (i.e. lignocellulosic biomass) are generated in agriculture, food industry and forestry. Therefore, it is important to raise awareness about the possibilities of using such materials, which nowadays should not be treated as waste, but can be used as renewable biotechnological raw materials for the production of chemicals, other high-value products and biofuels. Lignocellulosic material consists mainly of cellulose, hemicellulose and lignin. The subject of interest of this paper is lignocellulosic waste from agriculture and the food industry as a possible raw material basis for the progress of sustainable biotechnological production in the Republic of Croatia. The available amounts of these lignocellulosic raw materials, the types of bioprocesses in which they can be used, the pretreatment procedures that need to be carried out before the implementation of the bioprocess itself, and the types of biotechnological products that can be obtained have been considered.

Značajne količine raznovrsnih ostataka (odnosno lignocelulozne biomase) nastaju u poljoprivredi, prehrambenoj industriji i šumarstvu. Stoga je važno podići svijest o mogućnostima primjene takvih materijala koji se u današnje vrijeme ne bi trebali tretirati kao otpad, već se mogu koristiti kao obnovljive biotehnološke sirovine za proizvodnju kemikalija, drugih visokovrijednih proizvoda i biogoriva. Lignocelulozni materijal uglavnom sadrži celulozu, hemicelulozu i lignin. Predmet interesa ovog rada je lignocelulozni otpad iz poljoprivrede i prehrambene industrije kao mogući sirovinski temelj za napredak održive biotehnološke proizvodnje u Republici Hrvatskoj. Razmotrene su dostupne količine ovih lignoceluloznih sirovina, tipovi bioprocesa u kojima se one mogu koristiti, postupci predobrade koje je neophodno provesti prije provedbe samog bioprocesa te vrste biotehnoloških proizvoda koje je moguće dobiti.

The growing commercial application of microalgae in different industry sectors, including the production of bioenergy, pharmaceuticals, nutraceuticals, chemicals, feed, and food, demands large quantities of microalgal biomass with specific compositions produced at reasonable prices. Extensive studies have been carried out on the design of new and improvement of current cultivation systems and the optimisation of growth medium composition for high productivity of microalgal biomass. In this study, the concentrations of the main macronutrients, silicon, nitrogen and phosphorus, essential for the growth of diatom Nitzschia sp. S5 were optimised to obtain a high biomass concentration. The effect of main macronutrients on growth kinetics and cell composition was also studied. Silicon had the most significant effect on diatom growth during batch cultivation. The concentration of biomass increased 5.45-fold (0.49 g L−1) at 1 mM silicon concentration in modified growth medium compared to the original Guillard f/2 medium. Optimisation of silicon, nitrogen, and phosphorus quantities and ratios further increased biomass concentration. The molar ratio of Si:N:P = 7:23:1 mol:mol:mol yielded the highest biomass concentration of 0.73 g L−1. Finally, the fed-batch diatom cultivation of diatom using an optimised Guillard f/2 growth medium with four additions of concentrated macronutrient solution resulted in 1.63 g L−1 of microalgal biomass. The proteins were the most abundant macromolecules in microalgal biomass, with a lower content of carbohydrates and lipids under all studied conditions.

Bioethanol is the most widely used alternative transportation fuel to petrol. Bioethanol is considered a clean, renewable, and environmentally friendly fuel that can contribute to climate change mitigation, decreased environmental pollution, and enhanced energy security. Commercial bioethanol production is based on traditional agricultural crops such as corn, sugarcane, and sugarbeet, primarily used as food and feed. In order to meet the growing demand for this fuel and decrease competition in the food and biofuel sectors for the same feedstock, other raw materials and process technologies have been intensively studied. Lignocellulosic biomass is one of the most abundant renewable resources, with it being rich in compounds that could be processed into energy, transportation fuels, various chemical compounds, and diverse materials. Bioethanol production from lignocellulosic biomass has received substantial attention in recent decades. This review gives an overview of bioethanol production steps from lignocellulosic biomass and challenges in the production process. The following aspects of bioethanol production are covered here, including pretreatment methods, process strategies, strain development, ethanol isolation and purification, and technical hurdles.

The main product of anaerobic digestion is biogas, which is a renewable fuel, and the by-product of this process is digestate, which is used as a nutrient-rich fertilizer. Additional positive effects of anaerobic digestion are the decomposition of organic waste and the reduction of unpleasant odors and the concentration of pathogenic microorganisms. Biogas is mainly used for the production of electricity and heat, and in some cases it is purified to obtain biomethane which is used in the natural gas network, as a fuel for internal combustion engines or as a starting chemical for the chemical industry. Due to all of the above, the development of biogas production has positive socio-economic and ecological effects. Biogas produced in Croatia is mostly used for the production of electricity and thermal energy at cogeneration plants. Although various renewable raw materials are available in Croatia that could be used for biogas production, their potential has been underutilized until now. Manure and by-products of agriculture, slaughterhouses and the food industry are mainly used as raw materials for the production of biogas in our country. More rational use of land and development of the food industry could increase the amount of agricultural residues and by-products resulting from food processing. Directing and encouraging the use of these by-products for biogas production can stimulate a faster development of biogas production in our Croatia. In addition to the possible increase in own production of electricity and fuel, it is an environmentally friendly technology that has a positive socio-economic effect.

The underutilized biomass and different organic waste streams are nowadays in the focus of research for renewable energy production due to the effusive use of fossil fuels and greenhouse gas emission. In addition, one of the major environmental problems is also a constant increase of the number of organic waste streams. In a lot of countries, sustainable waste management, including waste prevention and reduction, has become a priority as a means to reduce pollution and greenhouse gas emission. Application of biogas technology is one of the promising methods to provide solutions for both actual energy-related and environmental problems. This review aims to present conventional and novel biogas production systems, as well as purification and upgrading technologies, nowadays applicable on a large scale, with a special focus on the CO2 and H2S removal. It also gives an overview of feedstock and the parameters important for biogas production, together with digestate utilization and application of molecular biology in order to improve the biogas production.

Brewers’ spent grains (BSG) are a by-product of the brewing industry that is mainly used as feedstock; otherwise, it has to be disposed according to regulations. Due to the high content of glucose and xylose, after pretreatment and hydrolysis, it can be used as a main carbohydrate source for cultivation of microorganisms for production of biofuels or biochemicals like 2,3-butanediol or lactate. 2,3-Butanediol has applications in the pharmaceutical or chemical industry as a precursor for varnishes and paints or in the food industry as an aroma compound. So far, Klebsiella pneumoniae, Serratia marcescens, Clostridium sp., and Enterobacter aerogenes are being used and investigated in different bioprocesses aimed at the production of 2,3-butanediol. The main drawback is bacterial pathogenicity which complicates all production steps in laboratory, pilot, and industrial scales. In our study, a gram-positive GRAS bacterium Paenibacillus polymyxa DSM 742 was used for the production of 2,3-butanediol. Since this strain is very poorly described in literature, bacterium cultivation was performed in media with different glucose and/or xylose concentration ranges. The highest 2,3-butanediol concentration of 18.61 g l–1 was achieved in medium with 70 g l–1 of glucose during 40 h of fermentation. In contrast, during bacterium cultivation in xylose containing medium there was no significant 2,3-butanediol production. In the next stage, BSG hydrolysates were used for bacterial cultivation. P. polymyxa DSM 742 cultivated in the liquid phase of pretreated BSG produced very low 2,3-butanediol and ethanol concentrations. Therefore, this BSG hydrolysate has to be detoxified in order to remove bacterial growth inhibitors. After detoxification, bacterium cultivation resulted in 30 g l–1 of lactate, while production of 2,3-butanediol was negligible. The solid phase of pretreated BSG was also used for bacterium cultivation after its hydrolysis by commercial enzymes. In these cultivations, P. polymyxa DSM 742 produced 9.8 g l–1 of 2,3-butanediol and 3.93 g l–1 of ethanol. On the basis of the obtained results, it can be concluded that different experimental setups give the possibility of directing the metabolism of P. polymyxa DSM 742 toward the production of either 2,3-butanediol and ethanol or lactate.

Abstract For biogas production, different renewable feedstocks, e.g., feces, manure, silage, industrial by-products, and municipal waste, can be used. Biogas production from various renewable feedstocks has positive socioeconomic and environmental impact. In Europe, biogas is mainly used for generating heat and electricity. It consists of methane (55–70% by volume), carbon dioxide (30–45% by volume), and small amounts of other compounds. In some cases, biogas is upgraded to pure biomethane and utilized as vehicle fuel, instead of fossil fuels, thus reducing the emissions of greenhouse gases. Biomethane can also serve as a platform chemical in chemical and biochemical synthesis to produce value-added products. The additional positive effects of anaerobic digestion of animal manure and slurries are organic waste degradation, reducing odors, and pathogens. Digestate, obtained as a by-product of anaerobic digestion, is rich in nutrients and therefore is applied as fertilizer in agriculture. Biogas production in Croatia is mainly based on manure and by-products from agriculture, food industry, and slaughterhouses. The obtained biogas is mostly used for electricity and heat generation. Potential for large-scale biogas production in Croatia is still insufficiently used, although various renewable feedstocks are available. More rational and focused management of lignocellulosic residues, animal excrements, food processing by-products, and biodegradable fraction of municipal waste could contribute to the development of Croatian biogas sector. Biogas production in Croatia can be affected by the changes of animal breeding capacity due to the struggle to cope with the European Union (EU) standards and prices. Concerning large unused agricultural areas, great potential lies in their rational exploitation for fast-growing biomass, e.g., for energy crops or perennial grasses. This review will discuss the potential of biogas in the industrial and farming sector, current state of biogas production, and various key drivers and barriers influencing biogas production in Croatia.

In the present study, water extracts from banana and red beetroot peels were evaluated as a potential source of biologically active compounds for the formulation of edible films. Using spectrophotometric and HPLC-DAD methodologies, banana peel extract was found to be a valuable source of dopamine (156.08 mg L−1), while red beetroot peel extract was abundant in red-violet pigments betacyanins (90.1 mg betanin L−1). The biological activity of the extracts was studied by determining their effects on macromolecular models, including DNA (plasmid phiX RF1 DNA), protein (bovine serum albumin), and lipid (linoleic acid) models, as well as on continuous human cell lines of colon cancer Caco-2 and hepatocellular liver cancer Hep G2 at concentrations of 0.2 and 1 mg mL−1. Results showed that the extracts had no adverse effects and both were further used for the formulation of edible films using alginate in combination with three types of plant proteins—rice, peanut, and pumpkin. In general, edible films based on banana peel extract were characterized by better bioactive properties compared with the films based on red beetroot peel extract. The addition of peanut proteins into the formulations resulted in the most desirable bioactive profile of the formulated edible films, including total phenolic content and antioxidant capacity. Aside from the control sample prepared only with the alginate, the highest dopamine content was determined in the film with incorporated pumpkin proteins (10.72 mg g−1 dw), while the sample prepared with peanut proteins was richest in betacyanins (175.58 mg betanin g−1 dw).

Paramylon is a valuable intracellular product of the microalgae Euglena gracilis, and it can accumulate in Euglena cells according to the cultivation conditions. For the sustainable production of paramylon and appropriate cell growth, different bioreactor processes and industrial byproducts can be considered as substrates. In this study, a complex medium with corn steep solid (CSS) was used, and various bioreactor processes (batch, fed batch, semicontinuous and continuous) were performed in order to maximize paramylon production in the microalgae Euglena gracilis. Compared to the batch, fed batch and repeated batch bioprocesses, during the continuous bioprocess in a stirred tank bioreactor (STR) with a complex medium containing 20 g/L of glucose and 25 g/L of CSS, E. gracilis accumulated a competitive paramylon content (67.0%), and the highest paramylon productivity of 0.189 g/Lh was observed. This demonstrated that the application of a continuous bioprocess, with corn steep solid as an industrial byproduct, can be a successful strategy for efficient and economical paramylon production.

Bioethanol production from lignocellulosic hydrolysates requires a producer strain that tolerates both the presence of growth and fermentation inhibitors and high ethanol concentrations. Therefore, we constructed heterozygous intraspecies hybrid diploids of Saccharomyces cerevisiae by crossing two natural S. cerevisiae isolates, YIIc17_E5 and UWOPS87-2421, a good ethanol producer found in wine and a strain from the flower of the cactus Opuntia megacantha resistant to inhibitors found in lignocellulosic hydrolysates, respectively. Hybrids grew faster than parental strains in the absence and in the presence of acetic and levulinic acids and 2-furaldehyde, inhibitors frequently found in lignocellulosic hydrolysates, and the overexpression of YAP1 gene increased their survival. Furthermore, although originating from the same parental strains, hybrids displayed different fermentative potential in a CO2 production test, suggesting genetic variability that could be used for further selection of desirable traits. Therefore, our results suggest that the construction of intraspecies hybrids coupled with the use of genetic engineering techniques is a promising approach for improvement or development of new biotechnologically relevant strains of S. cerevisiae. Moreover, it was found that the success of gene targeting (gene targeting fidelity) in natural S. cerevisiae isolates (YIIc17_E5α and UWOPS87-2421α) was strikingly lower than in laboratory strains and the most frequent off-targeting event was targeted chromosome duplication.

This work investigates the methodology of producing a 3D-printed microreactor from the acrylic resin by PolyJet Matrix process. The PolyJet Matrix technology employs different materials or their combinations to generate 3D-printed structures, from small ones to complex geometries, with different material properties. Experimental and numerical methods served for the evaluation of the geometry and production of the microreactor and its hydrodynamic characterization. The operational limits of the single-phase flow in the microchannels, further improvements and possible applications of the microreactor were assessed based on the hydrodynamic characterization.

Non-conventional yeasts are increasingly being investigated and used as producers in biotechnological processes which often offer advantages in comparison to traditional and well-established systems. Most biotechnologically interesting non-conventional yeasts belong to the Saccharomycotina subphylum, including those already in use (Pichia pastoris, Yarrowia lypolitica, etc.), as well as those that are promising but as yet insufficiently characterized. Moreover, for many of these yeasts the basic tools of genetic engineering needed for strain construction, including a procedure for efficient genetic transformation, heterologous protein expression and precise genetic modification, are lacking. The first aim of this study was to construct a set of integrative and replicative plasmids which can be used in various yeasts across the Saccharomycotina subphylum. Additionally, we demonstrate here that the electroporation procedure we developed earlier for transformation of B. bruxellensis can be applied in various yeasts which, together with the constructed plasmids, makes a solid starting point when approaching a transformation of yeasts form the Saccharomycotina subphylum. To provide a proof of principle, we successfully transformed three species from the Schwanniomyces genus (S. polymorphus var. polymorphus, S. polymorphus var. africanus and S. pseudopolymorphus) with high efficiencies (up to 8 × 103 in case of illegitimate integration of non-homologous linear DNA and up to 4.7 × 105 in case of replicative plasmid). For the latter two species this is the first reported genetic transformation. Moreover, we found that a plasmid carrying replication origin from Scheffersomyces stipitis can be used as a replicative plasmid for these three Schwanniomyces species.

Microbial lipids produced from lignocellulosic biomass are sustainable alternative feedstock for biodiesel production. In this study, corn cobs were used as a carbon source for lipid production and growth of oleaginous yeast Trichosporon oleaginosus. Lignocellulosic biomass was subjected to alkali and acid pretreatment using sulfuric acid and sodium hydroxide under different temperatures, catalyst concentrations and treatment times. Pretreatment of corn cobs was followed by cellulase hydrolysis. Hydrolysis of alkali pretreated (2% NaOH at 50 °C for 6 h, 1% NaOH at 50 °C for 16 h, 2% NaOH at 121 °C for 1 h, 1% NaOH at 121 °C for 2 h) and acid pretreated (1% H2SO4 120 °C for 20 min, and 2% H2SO4 120 °C for 10 min) corn cobs resulted in more than 80% of the theoretical yield of glucose. The effect of substrate (5, 10, 15 and 20%, g g−1) and cellulase loading (15 and 30 Filter Paper Units per gram of glucan, FPU g−1) on fermentable sugar yield was also studied. The maximal glucose concentration of 81.64 g L−1 was obtained from alkali-pretreated corn cobs (2% NaOH at 50 °C for 6 h) at 20% substrate loading and 30 FPU of Cellic CTec2 g−1 of glucan. Enzymatic hydrolysates of pretreated biomasses and filtrates of lignocellulosic slurries obtained after pretreatment were used for growth and lipid synthesis by T. oleaginosus. The highest lipid concentration of 18.97 g L−1 was obtained on hydrolysate of alkali-pretreated corn cobs (with 1% NaOH at 50 °C for 16 h) using a 15% (g g−1) substrate loading and 15 FPU g−1 of cellulase loading. Significant lipid accumulation was also achieved using undetoxified filtrates of pretreated slurries as substrates. Results showed that pretreated corn cobs and undetoxified filtrates are suitable carbon sources for the growth and efficient accumulation of lipids in T. oleaginosus.

Marine microalgae and cyanobacteria are sources of diverse bioactive compounds with potential biotechnological applications in food, feed, nutraceutical, pharmaceutical, cosmetic and biofuel industries. In this study, five microalgae, Nitzschia sp. S5, Nanofrustulum shiloi D1, Picochlorum sp. D3, Tetraselmis sp. Z3 and Tetraselmis sp. C6, and the cyanobacterium Euhalothece sp. C1 were isolated from the Adriatic Sea and characterized regarding their growth kinetics, biomass composition and specific products content (fatty acids, pigments, antioxidants, neutral and polar lipids). The strain Picochlorum sp. D3, showing the highest specific growth rate (0.009 h−1), had biomass productivity of 33.98 ± 0.02 mg L−1 day−1. Proteins were the most abundant macromolecule in the biomass (32.83–57.94%, g g−1). Nanofrustulum shiloi D1 contained significant amounts of neutral lipids (68.36%), while the biomass of Picochlorum sp. D3, Tetraselmis sp. Z3, Tetraselmis sp. C6 and Euhalothece sp. C1 was rich in glycolipids and phospholipids (75%). The lipids of all studied microalgae predominantly contained unsaturated fatty acids. Carotenoids were the most abundant pigments with the highest content of lutein and neoxanthin in representatives of Chlorophyta and fucoxanthin in strains belonging to the Bacillariophyta. All microalgal extracts showed antioxidant activity and antimicrobial activity against Gram-negative E. coli and S. typhimurium and Gram-positive S. aureus.

Bioetanol je proizveden procesom istodobne saharifikacije i fermentacije (eng. Simultaneous saccharification and fermentation, SSF) s kvascem Saccharomyces cerevisiae na enzimskom hidrolizatu predobrađenih kukuruznih oklasaka. Istraživan je učinak trajanja predhidrolize kukuruznih oklasaka na učinkovitost procesa proizvodnje bioetanola te je uspoređen s konvencionalnim procesom SSF. Dvije smjese komercijalno dostupnih enzima rabljene su za hidrolizu sirovine; prva je sadržavala Celluclast 1.5L (Sigma) i β-glukozidazu (Carl Roth), a druga smjesa Celluclast 1.5L (Sigma) i Viscozyme L (Sigma). Dva dana predhidrolize imala su pozitivan učinak na prinos bioetanola, dok je dulje trajanje predhidrolize smanjilo prinos bioetanola. Najveća koncentracija etanola od 41,24 g dm−3 postignuta je u procesu SSF uz dva dana predhidrolize koja je iznosila 63,15 % teoretskog iskorištenja izračunatog na neobrađeni kukuruzni oklasak. Najveća ukupna produktivnost procesa od 0,36 g dm−3 h−1 postignuta je konvencionalnim SSF-om uz enzimsku smjesu koja je sadržavala Celluclast 1.5L (Sigma) i Viscozyme L (Sigma). Koncentracija etanola iznosila je 36,6 g dm−3, što je 56,02 % teoretskog prinosa etanola na neobrađeni kukuruzni oklasak.

Abstract Background Lytic polysaccharide monooxygenases (LPMO) release a spectrum of cleavage products from their polymeric substrates cellulose, hemicellulose, or chitin. The correct identification and quantitation of these released products is the basis of MS/HPLC-based detection methods for LPMO activity. The duration, effort, and intricate analysis allow only specialized laboratories to measure LPMO activity in day-to-day work. A spectrophotometric assay will simplify the screening for LPMO in culture supernatants, help monitor recombinant LPMO expression and purification, and support enzyme characterization. Results Based on a newly discovered peroxidase activity of LPMO, we propose a fast, robust, and sensitive spectrophotometric activity assay using 2,6-dimethoxyphenol (2,6-DMP) and H2O2. The fast enzymatic assay (300 s) consists of 1 mM 2,6-DMP as chromogenic substrate, 100 µM H2O2 as cosubstrate, and an adequate activity of LPMO in a suitable buffer. The high molar absorption coefficient of the formed product coerulignone (ε 469 = 53,200 M−1 cm−1) makes the assay sensitive and allows reliable activity measurements of LPMO in concentrations of approx. 0.5–50 mg L−1. Conclusions The activity assay based on 2,6-DMP detects a novel peroxidase activity of LPMO. This activity can be accurately measured and used for enzyme screening, production, and purification, and can also be applied to study binding constants or thermal stability. However, the assay has to be used with care in crude extracts, because other enzymes such as laccase or peroxidase will interfere with the assay. We also want to stress that the peroxidase activity is a homogeneous reaction with soluble substrates and should not be correlated to heterogeneous LPMO activity on polymeric substrates.

Biodiesel and biogas are two very important sources of renewable energy worldwide, and particularly in the EU countries. While biodiesel is almost exclusively used as transportation fuel, biogas is mostly used for production of electricity and heat. The application of more sophisticated purification techniques in production of pure biomethane from biogas allows its delivery to natural gas grid and its subsequent use as transportation fuel. While biogas is produced mostly from waste materials (landfills, manure, sludge from wastewater treatment, agricultural waste), biodiesel in the EU is mostly produced from rapeseed or other oil crops that are used as food, which raises the ‘food or fuel’ concerns. To mitigate this problem, considerable efforts have been made to use non-food feedstock for biodiesel production. These include all kinds of waste oils and fats, but recently more attention has been devoted to production of microbial oils by cultivation of microorganisms that are able to accumulate high amounts of lipids in their biomass. Promising candidates for microbial lipid production can be found among different strains of filamentous fungi, yeast, bacteria and microalgae. Feedstocks of interest are agricultural waste rich in carbohydrates as well as different lignocellulosic raw materials where some technical issues have to be resolved. In this work, recovery and purification of biodiesel and biogas are also considered.

Production of biofuels from renewable feedstocks has captured considerable scientific attention since they could be used to supply energy and alternative fuels. Bioethanol is one of the most interesting biofuels due to its positive impact on the environment. Currently, it is mostly produced from sugar- and starch-containing raw materials. However, various available types of lignocellulosic biomass such as agricultural and forestry residues, and herbaceous energy crops could serve as feedstocks for the production of bioethanol, energy, heat and value-added chemicals. Lignocellulose is a complex mixture of carbohydrates that needs an efficient pretreatment to make accessible pathways to enzymes for the production of fermentable sugars, which after hydrolysis are fermented into ethanol. Despite technical and economic difficulties, renewable lignocellulosic raw materials represent low-cost feedstocks that do not compete with the food and feed chain, thereby stimulating the sustainability. Different bioprocess operational modes were developed for bioethanol production from renewable raw materials. Furthermore, alternative bioethanol separation and purification processes have also been intensively developed. This paper deals with recent trends in the bioethanol production as a fuel from different renewable raw materials as well as with its separation and purification processes.

Alternative to the use of fossil fuels are biofuels (e.g., bioethanol, biodiesel and biogas), which are more environmentally friendly and which can be produced from different renewable resources. In this investigation, bioethanol production from raw sugar beet cossettes (semi-solid substrate) by yeast Saccharomyces cerevisiae in a horizontal rotating tubular bioreactor (HRTB) was studied. Obtained results show that HRTB rotation mode (constant or interval) and rotation speed have considerable impact on the efficiency of bioethanol production in the HRTB. The main goal of this research was to develop a non-structural mathematical model of bioethanol production from raw sugar beet cossettes in the HRTB. The established mathematical model of bioethanol production in the HRTB describes substrate utilization and product formation (glycerol, ethanol and acetate) and presumes negative impact of high substrate concentration on the working microorganism (substrate inhibition) by using Andrews inhibition kinetics. All simulations of bioethanol production in the HRTB were performed by using Berkeley Madonna software, version 8.3.14 (Berkeley Madonna, Berkeley, CA, USA). The established non-structural bioprocess model describes relatively well the bioethanol production from raw sugar beet cossettes in the HRTB.

Microbial lipids have similar fatty acid composition to plant oils, and therefore, are considered as an alternative feedstock for biodiesel production. Oleaginous yeasts accumulate considerable amounts of lipids intracellularly during growth on low-cost renewable feedstocks such as lignocellulosic biomass. In this study, we cultivated yeast Trichosporon oleaginosus on hydrolysate of alkaline pretreated corn cobs. Different process configurations were evaluated and compared, including separate hydrolysis and fermentation (SHF) with cellulase recycle and simultaneous saccharification and fermentation (SSF) in batch and fed-batch mode. At low enzyme loading, the highest lipid concentration of 26.74 g L−1 was reached in fed-batch SSF fed with 2.5% (g g−1) substrate. Batch SHF was conducted for four rounds with recycling the cellulase adsorbed on unhydrolyzed lignocellulosic biomass. Thirty percent of cellulase saving was achieved for rounds 2–4 without compromising productivity and lipid yield. The addition of Tween 80 to lignocellulosic slurry improved the hydrolysis rate of structural carbohydrates in pretreated lignocellulosic biomass. Furthermore, supplementing the growth medium with Tween 80 improved lipid yield and productivity without affecting yeast growth. Oleaginous yeast T. oleaginosus is a promising strain for the sustainable and efficient production of lipids from renewable lignocellulosic feedstock.

Various fungal species can degrade lignocellulolytic materials with their enzyme cocktails composed of cellulolytic and lignolytic enzymes. In this work, seven fungal species (Mucor indicus DSM 2185, Paecilomyces variotii CBS 372.70, Myceliophthora thermophila CBS 663.74, Thielavia terrestris CBS 456.75, Botryosphaeria dothidea JCM 2738, Fusarium oxysporum f.sp. langenariae JCM 9293, and Fusarium verticillioides JCM 23107) and four nutrient media were used in the screening for effective lignocellulose degrading enzymes. From the seven tested fungi, F. oxysporum and F. verticilliodes, along with nutrient medium 4, were selected as the best medium and producers of lignocellulolytic enzymes based on the determined xylanase (>4 U mg−1) and glucanase activity (≈2 U mg−1). Nutrient medium 4 supplemented with pretreated corn cobs was used in the production of lignocellulolytic enzymes by sequential solid-state and submerged cultivation of F. oxysporum, F. verticilliodes, and a mixed culture of both strains. F. oxysporum showed 6 times higher exoglucanase activity (3.33 U mg−1) after 5 days of cultivation in comparison with F. verticillioides (0.55 U mg−1). F. oxysporum also showed 2 times more endoglucanase activity (0.33 U mg−1). The mixed culture cultivation showed similar endo- and exoglucanase activities compared to F. oxysporum (0.35 U mg−1; 7.84 U mg−1). Maximum xylanase activity was achieved after 7 days of cultivation of F. verticilliodes (≈16 U mg−1), while F. oxysporum showed maximum activity after 9 days that was around 2 times lower compared to that of F. verticilliodes. The mixed culture achieved maximum xylanase activity after only 4 days, but the specific activity was similar to activities observed for F. oxysporum. It can be concluded that both fungal strains can be used as producers of enzyme cocktails for the degradation of lignocellulose containing raw materials, and that corn cobs can be used as an inducer for enzyme production.

Povećanje emisije stakleničkih plinova, energetska ovisnost i nestabilnost isporuke energenata posljedica su ubrzane potrošnje fosilnih goriva zbog ubrzanog rasta svjetske populacije i industrijalizacije. Bioetanol je postao atraktivno zamjensko biogorivo jer se proizvodi iz obnovljivih sirovina i ekološki je prihvatljiv. Upotrebljava se kao pogonsko gorivo i to kao hidrirani (96 %) ili bezvodni (u mješavinama s benzinom). Bioetanol se proizvodi fermentacijom s kvascem S. cerevisiae ili nekim drugim mikroorganizmom iz ugljikohidrata kao što su jednostavni šećeri, škrob i celuloza. Nakon fermentacije, bioetanol se izdvaja i pročišćava destilacijom i dehidracijom. Sastav je propisan specifikacijama za primjenu u motornim gorivima. Uobičajeni usjevi, kao što su kukuruz, šećerna repa i trska, zasad su osnovne sirovine za proizvodnju bioetanola. Međutim, proizvodnja bioetanola iz ovih sirovina ne može zadovoljiti globalne potrebe za bioetanolom, zbog njihove primarne uloge u prehrani ljudi i životinja. Lignoceluloza je pogodna sirovina za proizvodnju bioetanola jer je široko rasprostranjena, obnovljiva i ne upotrebljava se u prehrani. Proizvodnja bioetanola iz lignoceluloznih sirovina je složen proces, koji se u mnogim aspektima razlikuje od proizvodnje iz šećernih ili škrobnih sirovina. U ovom radu prikazane su dosadašnje i nove tehnologije u proizvodnji bioetanola uključujući primjenu različitih sirovina, postupaka proizvodnje i radnih mikroorganizama. Nadalje, navedene su mogućnosti integracije osnovnih koraka u bioprocesima proizvodnje s ciljem povećanja produktivnosti i smanjenja troškova proizvodnje.

Batch cultivation of monoculture of Lactobacillus sp. and two–strain mixed culture of Lactobacillus sp. and Lactobacillus amylovorus DSM 20531T was carried out with the aim of producing L-(+)- and D-(–)/L-(+)-lactic acid to be implemented in poly(lactic acid) polymer production. Metabolic capacity of two Lactobacillus strains to ferment different carbon sources (glucose, sucrose or soluble starch) during cultivation in MRS medium at 40 °C, in a laboratory-scale stirred tank bioreactor was defined. Lactobacillus sp. showed similar affinity towards mono- and disaccharide substrates, which were homofermentatively converted mostly to L-(+)-lactic acid. L. amylovorus DSM 20531T has been characterized as a D/L-lactate producer and it is capable of conducting simultaneous saccharification and fermentation. Due to the interaction of Lactobacillus sp. with L. amylovorus DSM 20531T, starch was hydrolysed and fermented to the mixture of L-(+)- and D-(–)-lactic acid. Modified Luedeking-Piret kinetics used for the description of substrate utilization, growth of mono- and mixed cultures and production of lactic acid stereoisomers showed good agreement with experimental data.

A three-step process consisting of biomass hydrolysis, fermentation and in-situ gas stripping by a vacuum assisted recovery system, was optimized to increase the ethanol production from sugar beet pulp. The process combines the advantages of stripping and vacuum separation and enhances the fermentation productivity through in-situ ethanol removal. Using the design of experiment and response surface methodology, the effect of major factors in the process, such as pressure, recycling ratio and solids concentration, was tested to efficiently remove ethanol after the combined hydrolysis and fermentation step. Statistical analysis indicates that a decreased pressure rate and an increased liquid phase recycling ratio enhance the productivity and the yield of the strip-vacuum fermentation process. The results also highlight further possibilities of this process to improve integrated bioethanol production processes. According to the statistical analysis, ethanol production is strongly influenced by recycling ratio and vacuum ratio. Mathematical models that were established for description of investigated processes can be used for the optimization of the ethanol production.

In this investigation, the production of ethanol from the raw sugar beet juice and raw sugar beet cossettes has been studied. For ethanol production from the raw sugar beet juice, batch and fed-batch cultivation techniques in the stirred tank bioreactor were used, while batch ethanol production from the raw sugar beet cossettes was carried out in horizontal rotating tubular bioreactor (HRTB). In both cases, Saccharomyces cerevisiae was used as a production microorganism. During batch ethanol production from the raw sugar beet juice, ethanol yield was 59.89 g/L and production efficiency 78.8 %, and in fed-batch process the yield was 92.78 g/L and efficiency 93.4 %. At the same time, ethanol production in HRTB from the raw sugar beet cossettes with inoculum of 16.7 % V/m (raw sugar beet cossettes) resulted in the highest ethanol yield of 54.53 g/L and production efficiency of 79.5 %. The obtained results clearly show that both intermediates of sugar beet processing can be successfully used for ethanol production.

Amylolytic lactic acid bacterium Lactobacillus amylovorus DSM 20531T utilised glucose, sucrose and starch as a sole carbon and energy source. The three substrates were completely depleted from MRS medium during batch cultivations carried out in a laboratory scale stirred tank bioreactor at constant temperature (40 °C) and pH value (5.5). Under the tested conditions, the bacterium was capable of conducting simultaneously starch hydrolysis and fermentation. A mixture of two stereoisomers, D-(–)- and L-(+)-lactic acid, was produced in all cases by highly efficient homofermentative bioprocess with 0.93 to 1 g of lactate produced per g of total (consumed) substrate. The effect of temperature on the kinetics of cell growth and lactic acid production by the amylolytic strain in the starch-containing medium was also investigated. Efficient simultaneous saccharification and fermentation (SSF) was obtained at 35, 40 and 45 °C with completely degraded complex carbohydrate in 8 to 12 h and the product yield coefficient in the range from 0.91 to 0.93 g/g. Maximum values for substrate consumption rate (0.89 h^–1), maximum specific growth rate (0.87 h^–1), product formation rate (2.01 h^–1), and productivity of lactic acid (1.45 g/(L·h)) were obtained at 45 °C, while maximum biomass concentration (4.38 g/L) was attained at 40 °C. The ratio of the two stereoisomeric forms of produced lactic acid was strongly affected by the temperature. Unstructured kinetic model was used to describe the consumption of the three substrates, bacterial biomass formation and lactic acid production by L. amylovorus DSM 20531T. The dependence of biokinetic parameters on temperature was described by cardinal temperature model. The applied models successfully predicted all experimental data.
