In the last eight years, it has become possible to image chemical reactivity at the single-molecule and -particle level with fluorescence microscopy. This Perspective describes one of the imaging techniques that enabled this state-of-the-art application: imaging by the location change of molecules and particles. In this method, the microscope and experiment are configured to produce a signal when an individual molecule or particle changes location or changes mobility concurrently with a chemical change. This imaging technique has enabled observation of single chemical reactions and unraveled mechanisms of complex chemical and physical processes in transition metal and polymerization systems. This Perspective has three major goals: (1) to unify studies of different chemical processes or of different chemical questions, which, in spite of these differences, employ a similar microscopy detection method, (2) to explain the technique to nonexperts and those who might be interested in joining this nascent field, and (3) to highlight unique information available through this cross-disciplinary technique and the value this information has for chemical reaction development generally and catalysis specifically. To this end, application of the location change method to the investigation of polymerization reactions with radical initiators and separately with metal catalysts, and to ligand exchange reactions at platinum complexes are described.

Aiming to provide a portable research platform to develop algorithms for neuro-steered hearing aids, a joint hearing aid - EEG measurement setup was implemented in this work. The setup combines the miniaturized electroencephalography sensor technology cEEGrid with a portable hearing aid research platform - the Portable Hearing Laboratory. The different components of the system are connected wirelessly, using the lab streaming layer framework for synchronization of audio and EEG data streams. Our setup was shown to be suitable for simultaneous recording of audio and EEG signals used in a pilot study (n=5) to perform an auditory Oddball experiment. The analysis showed that the setup can reliably capture typical event-related potential responses. Furthermore, linear discriminant analysis was successfully applied for single-trial classification of P300 responses. The study showed that time-synchronized audio and EEG data acquisition is possible with the Portable Hearing Laboratory research platform.

Activating agents enable the efficient preparation of organozinc complexes from zinc metal and organohalides, but their mechanisms had been obscured by the heterogeneous nature of these systems. Fluorescence microscopy, with the sensitivity to detect surface reaction intermediates, reveals distinct activating mechanisms of widely used activation strategies: trimethylsilyl chloride, LiCl, DMSO, and Rieke zinc powder. The resulting development of mechanistic models provides a better understanding of the oxidative-addition-solubilization sequence in organozinc reagent formation and contains lessons for methods development.

A thioarylation method is developed for the synthesis of 2,3-dihydrothiopheniums through an electrophilic-cyclization-cross-coupling mechanism, harnessing the gold(I)/(III) cycle of the recently developed MeDalPhosAuCl catalyst. Single-crystal X-ray crystal structural analysis of the dihydrothiophenium products characterized the antiaddition of the sulfur and Csp 2 group to the alkyne and a preference for 5-endo dig cyclization. The dihydrothiophenium products are demonstrated as synthetic building blocks for stereodefined acyclic tetrasubstituted alkenes upon ring-opening reaction with amines. Intramolecular competition experiments show the favorability of Csp 3 tether cyclizations over Csp 2 tethers, preferentially generating dihydrothiopheniums over thiopheniums. Intermolecular competition experiments of alkyne aryl groups and an intermolecular aryl iodide competition suggest a rate-determining reductive elimination step in the gold(I)/gold(III) catalytic cycle. This rate-determining step is further supported by HRMS analysis of reaction intermediates that identify the catalyst resting state under turnover conditions. Catalyst poisoning experiments provide evidence of substrate inhibition, further consistent with these conclusions.

Fluorescence lifetime imaging microscopy (FLIM) provides spatiotemporal resolution of the changing composition of emulsion droplets during aqueous-surfactant Suzuki coupling. In contrast to previous investigations, the present experiments characterize the full course of a catalytic chemical reaction, addressing key questions about reaction species buildup and correlating these microscale behaviors with bench-scale product yields. At low concentrations of (active) catalyst, droplet environments are stable; however, at higher concentrations, emulsion droplet environments change markedly. These changes are consistent with the buildup and consumption of reaction species inside the droplets. A combination of FLIM and bright-field imaging pinpoints limitations in catalyst solubility as controlling rate and degree of buildup of species in droplets. These solubility limitations are also identified as the cause of a reaction induction period and an origin of the rate-and-reproducibility advantage obtained by adding THF cosolvent. The subsequent mechanistic model from these data led to a bench-scale reaction optimization, wherein premixing the catalyst components bypasses the catalyst induction period, resulting in a faster reaction. The understanding generated by FLIM thus provides an early example of how visualizing changes in droplet compositions on the microscale during ongoing aqueous-organic reactions can be leveraged for enhancing efficiencies in bench-scale reactions.

A fluorescence lifetime imaging microscopy (FLIM) technique characterizes surfactant-dependent partitioning of organics in a system that mimics a Negishi-like cross-coupling reaction in water, under synthetic concentrations, with emulsion droplets. Experimental partitioning data were not predictable from simple hydrophilic-lipophilic balances. The ionic surfactant cetrimonium chloride suppressed the reactivity of the metallic zinc surface, presumably through competitive chloride binding and concurrent cetrimonium coating, a finding that may contribute to the reduced performance of ionic surfactants in the bench-scale coupling reaction.

Commercial zinc metal powder requires activation for consistent and reliable use as a reductant in the formation of organozinc reagents from organohalides, and for the avoidance of supplier and batch-to-batch variability. However, the impact of activation methods on the reaction environments of subsequent intermediates has been unknown. Herein, a fluorescence lifetime imaging microscopy (FLIM) method is developed to bridge this knowledge gap, by imaging and examining reaction intermediates on zinc metal that has been activated by pretreatment through different common methods (i. e., by chemical activation with TMSCl, dibromoethane, or HCl; or by mechanical activation). The group of chemical activating agents, previously thought to act similarly by removing oxide layers, are here shown to produce markedly different reaction environments experienced by subsequent oxidative-addition intermediates from organohalides - data uniquely available through FLIM's ability to detect small quantities of intermediates in situ coupled with its microenvironmental sensitivity. These different microenvironments potentially give rise to different rates of formation, subsequent solubilization, and reactivity, despite the shared "[RZnX]" molecular structure of these intermediates. Different reaction selectivity occurs with the Lewis acids B -chlorocatecholborane (ClBcat), B -bromocatecholborane (BrBcat), and BBr 3 , favoring either alkyne haloboration, electrophilic cyclization of a tethered nucleophilic sulfur onto the alkyne, or group transfer of the nucleophile. This reaction selectivity also depends on the chain length of the tethered nucleophile, revealing a subtle interplay of relative kinetics and thermodynamics. In all cases, BBr 3 reacts readily with alkynes to form haloborated products; however, this process is reversible, and this reversibility can be harnessed to ultimately access regio- and stereodefined cyclic sulfonium zwitterions via the slower but thermodynamically favored electrophilic cyclization pathway. Reversibility was noted by following the reaction by NMR spectroscopy, and by characterizing the kinetic and thermodynamic products by a combination of 2D NMR spectroscopy and single-crystal X-ray diffraction. The "mixed" reagent bromocatechol borane (BrBcat) displayed reactivity between ClBcat and BBr 3 , producing bromoboration in some cases and electrophilic cyclization in others. With this enhanced understanding of the reaction dynamics, it becomes possible to use boron Lewis acids in a predictable manner in cases where haloboration is the kinetic product but in which the reversibility of this reaction maintains access to eventual alternative reactivity leading to desired building blocks in organic synthesis.

Trimethylsilyl chloride (TMSCl) is commonly used to "activate" metal(0) powders toward oxidative addition of organohalides, but knowledge of its mechanism remains limited by the inability to characterize chemical intermediates under reaction conditions. Here, fluorescence lifetime imaging microscopy (FLIM) overcomes these prior limitations and shows that TMSCl aids in solubilization of the organozinc intermediate from zinc(0) metal after oxidative addition, a previously unknown mechanistic role. This mechanistic role is in contrast to previously known roles for TMSCl before the oxidative addition step. To achieve this understanding, FLIM, a tool traditionally used in biology, is developed to characterize intermediates during a chemical reaction-thus revealing mechanistic steps that are unobservable without fluorescence lifetime data. The regio- and stereodefined synthesis of trisubstituted alkenes remains a significant synthetic challenge. Herein, a method is developed for producing regio- and stereodefined trisubstituted iodoalkenes by diverting intermediates from an iodination-electrophilic-cyclization mechanism. Specifically, cyclized sulfonium ion-pair intermediates are diverted to alkenes by ring-opening with nucleophilic iodide. Alternatively, scavenging of the iodide by AgOTf prevents ring-opening, enabling isolation of the sulfonium ion-pair intermediate. Isolation of the ion pair enables access to complementary reactivity, including ring-opening by alternative nucleophiles (i.e., amines), yielding trisubstituted acyclic alkenes and an example acyclic tetrasubstituted alkene. X-ray crystallographic determination of reaction intermediates and products confirms that the initial electrophilic-cyclization step sets the stereo- and regiochemistry of the product. The products serve as synthetic building blocks by readily participating in downstream functionalization reactions, including oxidation, palladium-catalyzed cross-coupling, and nucleophilic displacement.

An autofluorescence technique to characterize polymerization progress in real time/in line was developed, which functioned in the absence of typical fluorogenic groups on the monomer or polymer. The monomer dicyclopentadiene and polymer polydicyclopentadiene are hydrocarbons that lack traditional functional groups for fluorescence spectroscopy. Here, the autofluorescence of formulations containing this monomer and polymer during ruthenium-catalyzed ring-opening metathesis polymerization (ROMP) was harnessed for reaction monitoring. The methods fluorescence recovery after photobleaching (FRAP) and here-developed fluorescence lifetime recovery after photobleaching (FLRAP) characterized polymerization progress in these native systems-without requiring exogenous fluorophore. (Auto)fluorescence lifetime recovery changes during polymerization correlated linearly to degree of cure, providing a quantitative link with reaction progress. These changing signals also provided relative rates of background polymerization, enabling comparison of 10 different catalyst-inhibitor-stabilized formulations. Multiple-well analysis demonstrated suitability for future high-throughput evaluation of formulations for thermosets. Fluorescence lifetime imaging microscopy (FLIM) reveals vesicle sizes, structures, microenvironments, reagent partitioning, and system evolution with two chemical reactions for widely used surfactant-water systems under conditions relevant to organic synthesis, including during steps of Negishi cross-coupling reactions. In contrast to previous investigations, the present experiments characterize surfactant systems with representative organohalide substrates at high concentrations (0.5 M) that are reflective of the preparative-scale organic reactions performed and reported in water. In the presence of representative organic substrates, 2-iodoethylbenzene and 2-bromo-6-methoxypyridine, micelles swell into emulsion droplets that are up to 20 μm in diameter, which is 3-4 orders of magnitude larger than previously measured in the absence of an organic substrate (5-200 nm). The partitioning of reagents in these systems is imaged through FLIM─demonstrated here with nonpolar, amphiphilic, organic, basic, and oxidative-addition reactive compounds, a reactive zinc metal powder, and a palladium catalyst. FLIM characterizes the chemical species and/or provides microenvironment information inside micelles and vesicles. These data show that surfactants cause surfactant-dictated microenvironments inside smaller micelles (<200 nm) but that addition of a representative organic substrate produces internal microenvironments dictated primarily by the substrate rather than by the surfactant, concurrent with swelling. Addition of a palladium catalyst causes the internal environments to differ between vesicles─information that is not available through nor predicted from prior analytical techniques. Together, these data provide immediately actionable information for revising reaction models of surfactant-water systems that underpin the development of sustainable organic chemistry in water.

Understanding individual-block solvation in self-assembled block copolymer systems is experimentally difficult, but this solvation underpins the assembly and disassembly observed at the bulk scale. Here, covalently attached viscosity-sensitive molecular rotors for fluorescence lifetime imaging microscopy uncover and quantitatively elucidate previously undisclosed differential block-selective responses toward solvation changes upon addition of DMSO and THF to self-assembled ROMP-based amphiphilic block copolymers. The sensitivity of this method provides unique information on block-selective solvent-triggered assembly and disassembly mechanisms, revealing behaviors invisible to or with superior sensitivity to traditional 1 H NMR spectroscopy. These experiments demonstrate an analytical method and provide a granular mechanistic understanding, both suitable for fine tuning block copolymer assembly and disassembly processes.

Control of polymer molecular weight is critical for tailoring structure-function properties; however, traditional molecular weight characterization techniques have limited ability to determine the molecular weight of polymers in real time without sample removal from the reaction mixture, with spatial resolution, and of insoluble polymers. In this work, a fluorescence lifetime imaging microscopy (FLIM) method was developed that overcomes these limitations. The method is demonstrated with polynorbornene and polydicyclopentadiene, polymers derived from ruthenium-catalyzed ring-opening metathesis polymerization (ROMP). The polymer M w , ranging from 35 to 570 kg/mol as determined by gel-permeation chromatography, was quantitatively correlated with the fluorescence lifetime. The revealed correlation then enabled time-resolved measurement of M w during an ongoing ROMP reaction, requiring only 1 s per measurement (of a 45 μm × 45 μm polymer sample area), and provided spatial resolution, resulting in simultaneous characterization of polymer morphology. To provide the fluorescence signal, the initial reaction solutions contained a very low doping of a reactive norbornene monomer labeled with fluorescent boron dipyrromethene (BODIPY), such that 1 in every 10 7 monomers contained a fluorophore. The resulting FLIM visualization method enables the rapid determination of the molecular weights of growing polymers without removal from the reaction mixture and regardless of polymer solubility.

A demethylative silyl radical cascade cyclization of 2-isocyanothioanisoles toward 2-silylated benzothiazole building blocks has been developed. The development of a "radical initiator-inhibitor symbiosis" system solves the challenge of otherwise dominant methyl radical-triggered side reactions brought about by kinetically unfavored generation of reactive silyl radical species. The products accessed in this protocol are amendable to various downstream functionalization reactions, including the quick construction of a topoisomerase II inhibitor via a Hiyama cross-coupling reaction and of an antiviral agent via a fluoride-/hydroxide-free nucleophilic substitution to acyl chloride.

The ability to directly observe chemical reactions at the single-molecule and single-particle level has enabled the discovery of behaviors otherwise obscured by ensemble averaging in bulk measurements. However powerful, a common restriction of these studies to date has been the absolute requirement to surface tether or otherwise immobilize the chemical reagent/reaction of interest. This constraint arose from a fundamental limitation of conventional microscopy techniques, which could not track molecules or particles rapidly diffusing in three dimensions, as occurs in solution. However, many chemical processes occur entirely in the solution phase, leaving single-particle/-molecule analysis of this critical area of science beyond the scope of available technology. Here, we report the first kinetics studies of freely diffusing and actively growing single polymer-particles at the single-particle level freely diffusing in solution. Active-feedback single-particle tracking was used to capture three-dimensional (3D) trajectories and real-time volumetric images of freely diffusing polymer particles ( D ≈ 10 -12 m 2 /s) and extract the growth rates of individual particles in the solution phase. The observed growth rates show that the average growth rate is a poor representation of the true underlying variability in polymer-particle growth behavior. These data revealed statistically significant populations of faster- and slower-growing particles at different depths in the sample, showing emergent heterogeneity while particles are still freely diffusing in solution. These results go against the prevailing premise that chemical processes in freely diffusing solution will exhibit uniform kinetics. We anticipate that these studies will launch new directions of solution-phase, nonensemble-averaged measurements of chemical processes.

Polymer growth induces physical changes to catalyst microenvironments. Here, these physical changes are quantified in real time and are found to influence microscale chemical catalysis and the polymerization rate. By developing a method to "peer into" optically transparent living-polymer particles, simultaneous imaging of both viscosity changes and chemical activity was achieved for the first time with high spatiotemporal resolution through a combination of fluorescence intensity microscopy and fluorescence lifetime imaging microscopy techniques. Specifically, an increase in microenvironment viscosity led to a corresponding local decrease in the catalytic molecular ruthenium ring-opening metathesis polymerization rate, plausibly by restricting diffusional access to active catalytic centers. Consistent with this diffusional-access model, these viscosity changes were found to be monomer-dependent, showing larger changes in microenvironment viscosity in cross-linked polydicyclopentadiene compared to non-crosslinked polynorbornene. The sensitivity and high spatial resolution of the imaging technique revealed significant variations in microviscosities between different particles and subparticle regions. These revealed spatial heterogeneities would not be observable through alternative ensemble analytical techniques that provide sample-averaged measurements. The observed spatial heterogeneities provide a physical mechanism for variation in catalytic chemical activity on the microscale that may accumulate and lead to nonhomogeneous polymer properties on the bulk scale.

Contrary to prevailing thought, the salt content of supernatants is found to dictate reactivity differences of different preparation methods of Rieke zinc toward oxidative addition of organohalides. This conclusion is established through combined single-particle microscopy and ensemble spectroscopy experiments, coupled with careful removal or keeping of the supernatants during Rieke zinc preparations. Fluorescence microscopy experiments with single-Rieke-zinc-particle resolution determined the microscale surface reactivity of the Rieke zinc in the absence of supernatants, thus pinpointing its inherent reactivity independent of the convoluting supernatant composition. In parallel experiments, scanning electron microscopy, energy-dispersive spectroscopy, X-ray photoelectron spectroscopy, and inductively coupled plasma-mass spectrometry characterized the zinc metal chemical composition at the bulk and single-particle levels. Proton nuclear magnetic resonance spectroscopy kinetics characterized bench-scale Rieke zinc reactivity in the presence and absence of different supernatants and exogenous salt additives. Together, these experiments show that the differences in reactivity from sodium-reduced vs lithium-reduced Rieke zinc arise from the residual salts in the supernatant rather than the differing salt compositions of the solids. This supernatant salt also determines the structure of the ultimate organozinc product, generating either the diorganozinc or monoorganozinc halide complex. That different organozinc complexes formed upon direct insertion to different preparations of Rieke zinc was not previously reported, despite Rieke zinc's widespread use. These findings impact organozinc-reagent and nanomaterial synthesis by showing that, unexpectedly, desired Rieke zinc reactivity can be achieved through simple addition of soluble salts to solutions that were used to prepare the metals─a substantially easier synthetic manipulation than solid composition and morphology control.

The motion of single molecular ruthenium catalysts during and after single turnover events of ring-opening metathesis polymerization is imaged through single-molecule superresolution tracking with a positional accuracy of ±32 nm. This tracking is achieved through the real-time incorporation of spectrally tagged monomer units into active polymer chain ends during living polymerization; thus, by design, only active-catalyst motion is detected and imaged, without convolution by inactive catalysts. The catalysts show diverse individualistic diffusive behaviors with respect to time that persist for up to 20 s. Catalysts occupy three mobility populations: quasi-stationary (23%), intermediate (53%, 65 nm), and large (24%, 145 nm) step sizes. Differences in catalyst mobility populations also exist between individual aggregates ( p < 0.001). Such differential motion indicates widely different local catalyst microenvironments during the catalytic turnover. These mobility differences are uniquely observable through single-catalyst microscopy and are not measurable through traditional ensemble analytical techniques for characterizing the behavior of molecular catalysts, such as nuclear magnetic resonance spectroscopy. The measured distributions of active molecular catalyst motions would not be readily predictable through modeling or first-principles, and the range likely impacts individual catalyst turnover rate and selectivity. This range plausibly contributes to property distributions observable in bulk polymers, such as molecular weight polydispersity (e.g., 1.9 in this system), leading to a revised understanding of the mechanistic, microscale origins of macroscale polymer properties.

Micelles on the surfaces of individual metallic zinc particles are imaged by fluorescence microscopy with sensitivity up to single micelles. These micelles are made fluorescent to enable imaging, through the incorporation of boron dipyrromethene fluorophores as representative organic molecular "cargo". Highlighting an advantage of this in situ and sensitive fluorescence technique, the same micelles are not visible by ex situ scanning electron microscopy/energy dispersive X-ray spectroscopy analysis. Examination of micellar solutions with zinc reveals an aging process: micelles do not immediately adhere to the zinc surfaces upon mixing but rather build up over time. Furthermore, at longer times, smaller zinc particles become fully encased in micelle "shells". Once adhered, micelles remain in the local regions of the zinc surface for the duration of the imaging experiments (>2 h). Single micelles are imaged in solution, and their molecular contents are characterized. Two-color fluorescence crossover experiments show that micelles adhered to the surface of the zinc exchange molecular contents with micelles in solution, achieving molecular exchange equilibrium in ∼2.5 h. Unique (non-ensemble averaged) exchange kinetics are displayed by micelles at different locations on the zinc surface, consistent with exchange kinetics of single micelles or small local clusters of micelles. The aging of the micellar solutions and the rate of exchange while on the surface of the zinc suggest that micelle mass transport processes may contribute to overall reaction barriers in sustainable organozinc cross-coupling reactions in micellar water. The observed aging of the system suggests routes for improvement of preparative, bench-scale synthetic reactions involving micellar preparations of organozinc compounds.

Purpose of Review: This review is meant to describe the genetic associations with pediatric severe COVID-19 pneumonia and the postinfectious complication of the multisystem inflammatory syndrome in children (MIS-C). Multiple genetic approaches have been carried out, primarily in adults with extrapolation to children, including genome-wide association studies (GWAS), whole exome and whole genome sequencing (WES/WGS), and target gene analyses., Recent Findings: Data from adults with severe COVID-19 have identified genomic regions (human leukocyte antigen locus and 3p21.31) as potential risk factors. Genes related to viral entry into cells (ABO blood group locus, ACE2, TMPRS22) have been linked to severe COVID-19 patients by GWAS and target gene approaches. Type I interferon (e.g. IFNAR2) and antiviral gene (e.g. TLR7) associations have been identified by several genetic approaches in severe COVID-19. WES has noted associations with several immune regulatory genes (e.g. SOCS1). Target gene approaches have identified mutations in perforin-mediated cytolytic pathway genes in children and adults with severe COVID-19 and children with MIS-C., Summary: Several genetic associations have been identified in individuals with severe COVID-19 and MIS-C via various genetic approaches. A metal-free regio- and stereocontrolled group-transfer route toward the synthesis of trisubstituted alkenes is described. In this route, an electrophilic heterocyclization is followed by ring-opening group transfer. Specifically, a thioboration reaction transforms readily available alkynyl sulfide precursors into alkenyl boronates and alkenyl sulfides with defined regio- and stereochemistry in one synthetic step using commercially available B-chlorocatecholborane (ClBcat). Mechanistic studies identified the likely pathway as proceeding through zwitterionic rather than haloborated intermediates. The regio- and stereochemistry set in the initial cyclization step is preserved in the final acyclic alkene product, producing alkenes with up to four modifiable substituents with predictable regio- and stereochemistry. Downstream functionalization reactions showcase the versatility of the substitutions of the resulting alkenes. The chemoselectivity of molecular catalysts underpins much of modern synthetic organic chemistry. However, little is known about the selectivity of individual catalysts because this single-catalyst-level behavior is hidden by the bulk catalytic behavior. Here, for the first time, the selectivity of individual molecular catalysts for two different reactions is imaged in real time at the single-catalyst level. This imaging is achieved through fluorescence microscopy paired with spectral probes that produce a snapshot of the instantaneous chemoselectivity of a single catalyst for either a single-chain-elongation or a single-chain-termination event during ruthenium-catalyzed polymerization. Solvent effects are often difficult to understand in cases where reaction intermediates, and thus their differential behavior in different solvents, are not directly observable by traditional ensemble analytical techniques. Herein, the sensitivity of single-particle fluorescence microscopy uniquely enables direct observation of organozinc intermediates and solvent effects on their build-up and persistence. When combined with NMR spectroscopy, these imaging data pinpoint the previously elusive mechanistic origin of solvent effects in the synthesis of widely used organozinc reagents. These findings characterize the acceleration of oxidative addition of the starting organoiodide to the surface of zinc metal in DMSO relative to THF, but once formed, surface intermediates display similar persistence in either solvent. In contrast to previously reported borylative heterocyclization methods, a reaction here proceeds without air-free techniques to access synthetically useful borylated thiophenes, benzothiophenes, and isocoumarins. A comparison of stability/decomposition rates in air of several catecholboronic ester (Bcat) compounds derived from different heterocycle cores showed a strong dependence on the heterocycle structure. Lessons learned from this comparison were then harnessed for the development of borylative heterocyclization reactions under ambient-atmosphere conditions and with wet solvent. In contrast to literature reports suggesting general moisture sensitivity, a subset of Bcat products resulting from this technique were chromatography-stable and directly isolable, obviating the requirement for an extra synthetic transformation into more stable boron species, such as pinacolboronic esters (Bpin), for isolation. The isolated Bcat products were amenable to various downstream functionalization reactions, including reactions that were not accessible with their better-known Bpin counterparts, showing the complementarity of Bcat reaction partners and expanding their known chemistry. These results suggest the value of conceptual revisitation of substitution and solvent influence on stability and isolability of organo-Bcat compound classes and lay the groundwork for development of additional practical borylative methods in air.

Yields of organoindium reagents synthesized from indium metal were previously reported to be highly dependent on metal batch and supplier due to the presence or absence of anticaking agent. Here, single-particle fluorescence microscopy established that MgO, an additive in some batches nominally for anticaking, significantly increased the physisorption of small-molecule organics onto the surface of the resulting MgO-coated indium metal particles. An inert and relatively nonpolar boron dipyrromethene fluorophore with a hydrocarbon tail provided a sensitive probe for this surface physisorption. SEM images revealed markedly different surface properties of indium particles either with or without MgO, consistent with their different physisorption properties observed by fluorescence microscopy. We further documented incomplete commercial bottle labeling regarding the presence and composition of this anticaking agent, both within our laboratory and previously in the literature, which may complicate reproducibility between laboratories. Trimethylsilyl chloride pretreatment, a step employed in a subset of reported synthetic procedures, removed the anticaking agent and produced particles with similar physisorption properties as commercial batches of indium powder distributed without the anticaking agent. These data indicate the possibility of an additional substrate/catalyst physisorption mechanism by which the anticaking agent may be influencing synthetic procedures that generate organoindium reagents from indium metal, in addition to simple anticaking., Competing Interests: The authors declare no competing financial interests.

Mechanistic studies have historically played a key role in the discovery and optimization of reactions in organic and organometallic chemistry. However, even apparently simple organic and organometallic transformations may have surprisingly complicated multistep mechanisms, increasing the difficulty of extracting this mechanistic information. The resulting reaction intermediates often constitute a small fraction of the total reaction mixture, for example, creating a long-term analytical challenge of detection. This challenge is particularly pronounced in cases where the positions of intermediates on the reaction energy surface mean that they do not "build up" to the quantities needed for observation by traditional ensemble analytical tools. Thus, their existence and single-step elementary reactivity cannot be studied directly. New approaches for obtaining this otherwise-missing mechanistic information are therefore needed. Single-turnover, single-molecule, single-particle, and other subensemble fluorescence microscopy techniques are ideally suited for this role because of their sensitivity and spatiotemporal resolution. Inspired by the robust development of single-molecule fluorescence microscopy tools for studying enzyme catalysis, our laboratory has developed analogous fluorescence microscopy techniques to overcome mechanistic challenges in synthetic chemistry, with sensitivity as high as the single-complex, single-turnover, and single-molecule level. These techniques free the experimenter from the previous restriction that intermediates must "build up" to quantities needed for detection by ensemble analytical tools and are suited to systems where synchronization through flash photolysis or stopped flow would be inconvenient or inaccessible. In this process, the techniques transform certain previously "unobservable" intermediates and their elementary single-step reactivities into "observable" ones through sensitive and selective spectral handles. Our program has focused on imaging reactions in small-molecule, organic, and polymer synthetic chemistry with an accent on the reactivity of molecular transition metal complexes and catalysts. Our laboratory initiated studies in this area in 2008 with the imaging of individual palladium complexes that were tagged with spectator fluorophores. To enable imaging, we started with fluorophore selection and development, overcame challenges with imaging in organic solvents, and developed strategies compatible with air-sensitive chemistry and concentrations of reagents generally used in small-molecule synthesis. These studies grew to include characterization of previously unknown organometallic intermediates in the synthesis of organozinc reagents and the direct study of their elementary-step reactivity. The ability to directly observe this behavior generated predictive power for selecting salts that accelerated organozinc reagent formation in synthesis, including salts that had not yet been reported synthetically. In 2017 we also developed the first single-turnover imaging of molecular (chemo)catalysts, which through the technique's spatiotemporal resolution revealed abruptly time-variable polymerization kinetics wherein molecular ruthenium ring-opening metathesis polymerization (ROMP) catalysts changed rates independently from other catalysts less than 1 μm away. Individual catalytic turnovers, each corresponding to one single-chain-elongation reaction arising from insertion of single ROMP or enyne monomers at individual Grubbs II molecular ruthenium catalysts, were spatiotemporally resolved as green flashes in growing polymers. In this Account, we discuss the development of this technique from idea to application, including challenges overcome and strategies created to image synthetic organic and organometallic molecular chemistry at the highest levels of detection sensitivity. We also describe challenges not yet solved and provide an outlook for this growing field at the intersection of microscopy and synthetic/molecular chemistry.

Lithium chloride is known to promote the direct insertion of metallic zinc powder into organohalides in the practical synthesis of organozinc reagents, but the reason for its special ability is poorly understood. Pioneering a combined approach of single-metal-particle fluorescence microscopy with 1 H NMR spectroscopy, we herein show that the effectiveness of different lithium salts toward solubilizing intermediates on the surface of zinc metal establishes a previously unknown reactivity correlation that predicts the propensity of that salt to promote macroscale reagent synthesis and also predicts the solution structure of the ultimate organozinc reagent. A salt-free pathway is also identified. These observations of an organometallic surface intermediate, its elementary-step reactivity, and the impact of various synthetic conditions (salt, salt-free, temperature, stirring, and time) on its persistence, are uniquely available from the sensitivity and spatial localization ability of the microscopy technique. These studies unify previously disparate observations under a single unified mechanistic framework. This framework enables the rational prediction of salt effects on multiple steps in organozinc reagent synthesis and reactivity. This is an early example of single-particle microscopy characterization of elementary steps providing predictive power in reaction development by gaining a sensitive and selective spectral handle on an important intermediate, highlighting the role of this next generation of analytical tools in the development of synthetic chemistry.

Herein we report an aminoboration reaction that employs inexpensive, Earth-abundant, and commercially available Cu(OTf) 2 as an effective catalyst in the direct addition of B-N σ bonds to C-C π bonds, generating borylated pyrazoles, which are useful building blocks for drug discovery. By nature of the mechanism, the reaction produces exclusively one regioisomer and tolerates groups incompatible with alternative lithiation/borylation and iridium-catalyzed C-H activation/borylation methods. The reaction can be scaled up, and the resulting isolable pyrazole pinacol boronates can be further functionalized through palladium-catalyzed Suzuki cross-coupling reactions.

A simple, regiocontrolled, and transition-metal-free approach to access exclusively 3-borylated thiophene derivatives is reported. The commercially available B-chlorocatecholborane reagent (ClBcat) acts as a carbophilic Lewis acid to activate the alkyne in readily synthesized ( Z)-organylthioenyne substrates. This boron-induced activation initiates the formal thioboration and subsequent sulfur dealkylation, leading to the formation of 3-borylated thiophenes in good yields. The resulting borylated thiophenes are isolable as boronic esters (Bpin) and boronamides (Bdan). These borylated products are amenable to diverse downstream functionalization reactions, i.e., C-C bond formation through cross-coupling, azidation, bromination, and C-H activation.

An oxyboration reaction that employs B-O σ bonds as addition partners to C-C π bonds to form borylated dihydrofurans and isochromenes has been developed. By nature of the mechanism, the reaction produces exclusively one borylated regioisomer, in contrast to and/or complementary to alternative routes that produce these borylated heterocycles via C-H activation. Access to the borylative heterocyclization route is demonstrated from alcohols directly or from a hydroboration-oxyboration sequence starting from the corresponding ketone, forming the heterocyclic core and installing the boron in one synthetic step. Catechol boronates were directly used as coupling partners in the in situ Suzuki cross-coupling reactions without transesterification to pinacol boronates.

Essentially no information is known about the behavior of individual molecular catalysts under reaction conditions. This is a result of the averaging inherent to traditional analytical techniques. Herein, a combined fluorescence microscopy and 1 H NMR spectroscopy study reveals that unique (that is, non-ensemble averaged) distributions and time-variable kinetics from molecular ruthenium catalysts within growing polynorbornene occur and are detectable between 10 -9  m and 10 -6  m of substrate, surprisingly just 1000-fold less concentrated than a typical laboratory bench-scale reaction. Catalytic cycles are typically depicted as possessing time-invariant steps with fixed rates. Yet the true behavior of individual catalysts with respect to time is unknown, hidden by the ensemble averaging inherent to bulk measurements. Evidence is presented for variable chemical kinetics at individual catalysts, with a focus on ring-opening metathesis polymerization catalyzed by the second-generation Grubbs' ruthenium catalyst. Fluorescence microscopy is used to probe the chemical kinetics of the reaction because the technique possesses sufficient sensitivity for the detection of single chemical reactions. Multiple active individual molecular ruthenium catalysts have been pinpointed within growing polynorbornene, thereby revealing information on the reaction dynamics and location that is unavailable through traditional ensemble experiments. This is the first single-turnover imaging of a molecular catalyst by fluorescence microscopy and allows detection of individual monomer reactions at an industrially important molecular ruthenium ring-opening metathesis polymerization (ROMP) catalyst under synthetically relevant conditions (e.g. unmodified industrial catalyst, ambient pressure, condensed phase, ca. 0.03 m monomer). Organoboron compounds and heterocycles are powerful building blocks and precursors for organic synthesis, including for drug discovery and agrochemical and material synthesis. The common strategy for the synthesis of borylated heterocycles involves two separate synthetic steps: first, synthesis of the heterocyclic core, and second, borylation of the core through established methods such as transition-metal-catalyzed C-H or C-X activation/borylation or lithiation/borylation. In this Account, we describe our laboratory's development of borylative heterocyclization reactions that access the heterocyclic core and install boron in one synthetic step. These methods provide complementary bond disconnections, regiochemistry, and functional-group compatibility to current methods. We describe our methods with two categories: a direct borylation method that refers to addition reactions starting from a preformed B-element σ bond, which is essential in the mechanistic route to product formation, and a formal borylation method that refers to addition reactions that do not require formation of a B-element bond but instead proceed through carbon-carbon π-bond activation by an electrophilic boron source followed by dealkylation or deacylation. Through electrophilic activation of the alkyne rather than activation of the B-element bond, formal borylation provides a complementary strategy toward neutral organoboron reagents. We first studied direct oxyboration toward the formation of borylated benzofurans, where a preformed boron-oxygen σ bond is added across an alkyne activated by a carbophilic gold catalyst. We describe detailed mechanistic and kinetic studies of this class of reactions. Application of the knowledge gained from these studies aided in the future development of additional direct borylation reactions involving boron-nitrogen and boron-oxygen σ bonds to form borylared indoles and isoxazoles, respectively. Formal addition of boron/oxygen equivalents to effect oxyboration to form borylated lactones from o-alkynyl esters is then described. This class of reactions takes advantage of bifunctional ClBcat as a carbophilic carbon-carbon π-bond activator and eventual dealkylating agent. We describe our motivation in developing this new class of catalyst-free borylation reactions and subsequently applying the formal borylation strategy to the thioboration of o-alkynylthioanisole substrates to form borylated benzothiophenes. We then proceed to describe our investigations into the details of the mechanism of the formal thioboration reaction. These collaborative mechanistic studies included experimental and computational findings that elucidated the rate-determining step and intermediates of the reaction. These studies further compared different boron sources as electrophiles, including those used in other known reactions, providing fundamental knowledge about the capabilities of commercially available boron reagents toward borylative heterocyclization. Our findings provide guiding principles for reaction design and information leading toward the design of a diverse set of boron-heteroatom addition reactions and their formal equivalents that proceed through borylative heterocyclization.

Several formal heteroborylative cyclization reactions have been recently reported, but little physical-organic and mechanistic data are known. We now investigate the catalyst-free formal thioboration reaction of alkynes to gain mechanistic insight into B-chlorocatecholborane (ClBcat) in its new role as an alkynophilic Lewis acid in electrophilic cyclization/dealkylation reactions. In kinetic studies, the reaction is second-order globally and first-order with respect to both the 2-alkynylthioanisole substrate and the ClBcat electrophile, with activation parameters of ΔG ‡ = 27.1 ± 0.1 kcal mol -1 at 90 °C, ΔH ‡ = 13.8 ± 1.0 kcal mol -1 , and ΔS ‡ = -37 ± 3 cal mol -1 K -1 , measured over the range 70-90 °C. Carbon kinetic isotope effects supported a rate-determining Ad E 3 mechanism wherein alkyne activation by neutral ClBcat is concerted with cyclative attack by nucleophilic sulfur. A Hammett study found a ρ + of -1.7, suggesting cationic charge buildup during the cyclization and supporting rate-determining concerted cyclization. Studies of the reaction with tris(pentafluorophenyl)borane (B(C 6 F 5 ) 3 ), an activating agent capable of cyclization but not dealkylation, resulted in the isolation of a postcyclization zwitterionic intermediate. Kinetic studies via UV-vis spectroscopy with this boron reagent found second-order kinetics, supporting the likely relevancy of intermediates in this system to the ClBcat system. Computational studies comparing ClBcat with BCl 3 as an activating agent showed why BCl 3 , in contrast to ClBcat, failed to mediate the complete the cyclization/demethylation reaction sequence by itself. Overall, the results support a mechanism in which the ClBcat reagent serves a bifunctional role by sequentially activating the alkyne, despite being less electrophilic than other known alkyne-activating reagents and then providing chloride for post-rate-determining demethylation/neutralization of the resulting zwitterionic intermediate.

The first ring-forming thioboration reaction of C-C π bonds is reported. This catalyst-free method proceeds in the presence of a commercially available external electrophilic boron source (B-chlorocatecholborane) in good to high yields. The method is scalable and tolerates a variety of functional groups that are intolerant of other major borylation methods. The resulting borylated benzothiophenes participate in a variety of in situ derivatization reactions, showcasing that these borylated intermediates do not need to be isolated prior to downstream functionalization. This methodology has been extended to the synthesis of borylated dihydrothiophenes. The sensitivity provided by fluorescence microscopy enabled the observation of surface intermediates in the synthesis of soluble organozinc reagents by direct insertion of alkyl iodides to commercial zinc powder. Five hypotheses were examined for the mechanistic role of lithium chloride in enabling this direct insertion. The data are consistent with lithium chloride solubilizing organozinc reagents from the surface of the zinc after oxidative addition.

In the present study, the oxyboration reaction catalyzed by IPrAuTFA in the presence and absence of NaTFA has been examined with kinetic studies, mass spectrometry, and 1 H NMR and 11 B NMR spectroscopy. Data from monitoring the reactions over the temperature range from 30 to 70 °C, the catalyst range from 1.3 to 7.5 mol %, and the NaTFA additive range from 2.5 to 30 mol % suggests a mechanism that involves rate-determining catalyst generation. Data from additive studies that replaced NaTFA with NaBARF (BARF = tetrakis[3,5-bis(trifluoromethyl)phenyl]borate) or Bu 4 NTFA as an alternative additive suggest that catalyst quenching from residual NaCl remaining from a one-pot substrate synthesis/reaction method is the cause of this effect, despite the low solubility of this NaCl byproduct in toluene. A catalyst-free oxyboration reaction of alkynes is developed. The resulting borylated isocoumarins and 2-pyrones are isolated as boronic acids, pinacolboronate esters, or potassium organotrifluoroborate salts, providing a variety of bench-stable organoboron building blocks for downstream functionalization. This method has functional group compatibility, is scalable, and proceeds with readily available materials: B-chlorocatecholborane and methyl esters. Mechanistic studies indicate that the B-chlorocatecholborane acts as a carbophilic Lewis acid toward the alkyne, providing a mechanistically distinct pathway for oxyboration that avoids B-O σ bond formation and enables this catalyst-free route.

Herein we report an oxyboration reaction with activated substrates that employs B-O σ bond additions to C-C π bonds to form borylated isoxazoles, which are potential building blocks for drug discovery. Although this reaction can be effectively catalyzed by gold, it is the first example of uncatalyzed oxyboration of C-C π bonds by B-O σ bonds--and only the second example that is catalyzed. This oxyboration reaction is tolerant of groups incompatible with alternative lithiation/borylation and palladium-catalyzed C-H activation/borylation technologies for the synthesis of borylated isoxazoles.

The Maryland Zoo in Baltimore experienced an outbreak of Frog virus-3 (FV3)-like ranavirus during the summer of 2011, during which 14 of 27 (52%) of its captive eastern box turtles (Terrapene carolina carolina) survived. To assess survival, immunity, and viral shedding, an experimental challenge study was performed in which the surviving, previously infected turtles were reinfected with the outbreak strain of FV3-like ranavirus. Seven turtles were inoculated with virus intramuscularly and four control turtles received saline intramuscularly. The turtles were monitored for 8 wk with blood and oral swabs collected for quantitative polymerase chain reaction (qPCR). During that time, one of seven (14%) inoculated turtles and none of the controls (0%) died; there was no significant difference in survival. Clinical signs of the inoculated turtles, except for the turtle that died, were mild compared to the original outbreak. Quantitative PCR for FV3-like ranavirus on blood and oral swabs was positive for all inoculated turtles and negative for all controls. The turtle that died had intracytoplasmic inclusion bodies in multiple organs. Three inoculated and two control turtles were euthanized at the end of the study. No inclusion bodies were present in any of the organs. Quantitative PCR detected FV3-like ranavirus in the spleen of a control turtle, which suggested persistence of the virus. The surviving five turtles were qPCR-negative for FV3-like ranavirus from blood and oral swabs after brumation. Quantitative PCR for Terrapene herpesvirus 1 found no association between ranavirus infection and herpesvirus loads. In conclusion, previously infected eastern box turtles can be reinfected with the same strain of FV3-like ranavirus and show mild to no clinical signs but can shed the virus from the oral cavity.

This communication demonstrates the first catalytic aminoboration of C-C π bonds by B-N σ bonds and its application to the synthesis of 3-borylated indoles. The regiochemistry and broad functional group compatibility of this addition reaction enable substitution patterns that are incompatible with major competing technologies. This aminoboration reaction effects the formation of C-B and C-N bonds in a single step from aminoboronic esters, which are simple starting materials available on the gram scale. This reaction generates synthetically valuable N-heterocyclic organoboron compounds as potential building blocks for drug discovery. The working mechanistic hypothesis involves a bifunctional Lewis acid/base catalysis strategy involving the combination of a carbophilic gold cation and a trifluoroacetate anion that activate the C-C π bond and the B-N σ bond simultaneously.

Electronic effects in the transmetalation of an aryl group from gold to boron were investigated by NMR spectroscopy. The transmetalation reaction is more facile for increasingly electrophilic boron reagents and is in equilibrium under certain conditions. Observed tetracoordinate boronate compounds suggest a two-step, associative transmetalation reaction mechanism in which the organogold complex first delivers a nucleophilic phenyl group to the empty p orbital of boron. For certain substrates, this tetracoordinate intermediate decomposes to the tricordinate, final transmetallation product, and in others this tricordinate species remains in equilibrium with a tetracoordinate anionic boron compound. Experimental and theoretical investigations into the extension of this transmetalation reaction from a mechanistic step in our previously reported intramolecular gold-catalyzed addition of boron-oxygen σ bonds across alkynes to an intermolecular variant are discussed.2014 Elsevier Ltd. The chemo- and regioselectivity and functional group compatibility in gold and palladium cooperatively catalyzed cross-coupling reactions were determined in the synthesis of lactones; the selectivity in the gold and palladium dual-metal catalysis system was distinct from that available for the same class of substrates in systems with only gold catalysis or only palladium catalysis rather than dual catalysis. The dual-catalytic rearrangement reaction selectively promoted oxidative addition at the C-O bond over the C-Br bond, providing a useful C-Br bond handle for downstream functionalization showcased via Suzuki-Miyaura and Sonogashira coupling reactions. Product classes were expanded from isocoumarins to three previously unpublished ring classes: pyrone, indolepyrone, and furopyrone.

For nearly 70 years, the addition of boron-X σ bonds to carbon-carbon multiple bonds has been employed in the preparation of organoboron reagents. However, the significantly higher strength of boron-oxygen bonds has thus far precluded their activation for addition, preventing a direct route to access a potentially valuable class of oxygen-containing organoboron reagents for divergent synthesis. We herein report the realization of an alkoxyboration reaction, the addition of boron-oxygen σ bonds to alkynes. Functionalized O-heterocyclic boronic acid derivatives are produced using this transformation, which is mild and exhibits broad functional group compatibility. Our results demonstrate activation of this boron-O σ bond using a gold catalysis strategy that is fundamentally different from that used previously for other boron addition reactions.

Double-label crossover, modified-substrate, and catalyst comparison experiments in the gold and palladium dual-catalytic rearrangement/cross-coupling of allenoates were performed in order to probe the mechanism of this reaction. The results are consistent with a cooperative catalysis mechanism whereby 1) gold activates the substrate prior to oxidative addition by palladium, 2) gold acts as a carbophilic rather than oxophilic Lewis acid, 3) competing olefin isomerization is avoided, 4) gold participates beyond the first turnover and therefore does not serve simply to generate the active palladium catalyst, and 5) single-electron transfer is not involved. These experiments further demonstrate that the cooperativity of both gold and palladium in the reaction is essential because significantly lower to zero conversion is achieved with either metal alone in comparison studies that examined multiple potential gold, palladium, and silver catalysts and precatalysts. Notably, employment of the optimized cocatalysts, PPh 3 AuOTf and Pd 2 dba 3 , separately (i.e., only Au or only Pd) results in zero conversion to product at all monitored time points compared to quantitative conversion to product when both are present in cocatalytic reactions.

In recent years, single-molecule and single-particle fluorescence microscopy has emerged as a tool to investigate chemical systems. After an initial lag of over a decade with respect to biophysical studies, this powerful imaging technique is now revealing mechanisms of 'classical' organic reactions, spatial distribution of chemical reactivity on surfaces and the phase of active catalysts. The recent advance into commercial imaging systems obviates the need for home-built laser systems and thus opens this technique to traditionally trained synthetic chemists. We discuss the requisite photophysical and chemical properties of fluorescent reporters and highlight the main challenges in applying single-molecule techniques to chemical questions. The goal of this Perspective is to provide a snapshot of an emerging multidisciplinary field and to encourage broader use of this young experimental approach that aids the observation of chemical reactions as depicted in many textbooks: molecule by molecule.
