Seventy-two primiparous and multiparous sows (36 per dietary treatment) farrowed in 4 groups were used to evaluate the effects of spray-dried plasma protein (SDP) on sow and litter performance during lactation. Dietary lactation treatments consisted of a corn-soybean meal control and a corn-soybean meal diet containing 0.5% SDP. Both diets were formulated to contain 1.0% total Lys and 3.46 Mcal/kg of ME and were fed from d 107 [+ or -] 1.2 of gestation to weaning. Sows were allotted to dietary treatment based on breed, parity, and the date of d 107 of gestation. Litters were standardized within diet, and pigs were weaned at an average age of 19 [+ or -] or 2.1 d. Sows were fed 3 times daily during lactation. After weaning, sows were fed a common gestation diet and checked twice daily for estrus. Sows were grouped by parity (young sows, [less than or equal to] 3; mature sows, >3) for statistical analysis. The data were analyzed as a 2 x 2 factorial arrangement of treatments; the factors were parity ([less than or equal to] 3 or >3) and SDP (0 or 0.5%). Treatment differences were considered significant at P < 0.10. Mature sows had a greater BW on d 107 of gestation, on d 1 postfarrowing, and at weaning; greater lactation ADFI; and greater litter BW after cross-fostering, but pig survival to weaning was decreased. Sows fed SDP had a greater gestation interval, litter BW at weaning, and litter ADG, with 1 less lactation day. The effect of SDP addition was dependent on sow parity, as noted by numerous SDP x parity interactions. The addition of SDP increased lactation ADFI in mature sows but decreased ADFI in young sows. Mature sows fed SDP had a greater numbet of pigs weaned per litter, litter and pig weaning weights, pig survival to weaning, and number of pigs weaned per litter weighing more than 3.6 kg, but the SDP diet had little to no effect on these responses in young sows. Subsequent farrowing data were collected, but no dietary treatment effects (P > 0.10) were observed. We have previously shown that plasma protein supplementation prevents the activation of lymphocyte populations of Peyer's patches and mesenteric lymph nodes, which is known as organized gut-associated lymphoid tissue (GALT). Here, we examined the effects of spray-dried plasma proteins (SDAP) and Ig concentrate (IgC) supplements on lamina propria and intraepithelial lymphocytes (diffuse GALT) in a model of mild intestinal inflammation induced by the intraperitoneal administration of Staphylococcus aureus enterotoxin B (SEB). Wistar-Lewis rats were fed diets supplemented with SDAP (8% wt:wt), IgC (1.5% wt:wt), or bovine milk proteins (control diet) from weaning (d 21) to d 34 after birth. On d 30 and 33, rats were given SEB (0.5 mg/kg body weight) or PBS (control). Experimental groups were designated control, SEB, SEB-SDAP, and SEB-IgC. Lymphocyte populations were analyzed by immunohistochemistry. In lamina propria, SEB increased the cytotoxic lymphocyte populations of T-[gamma][delta] cells (38%; P < 0.001 ) and natural killer cells (59%; P < 0.05) and the number of activated T lymphocytes (148%; P < 0.001 ). Both SDAP and IgC decreased the effects of SEB on these lymphocyte subsets (P < 0.05). In the epithelium, SEB induced a 117% increase in intraepithelial-activated lymphocytes that was reduced by SDAP supplementation (P < 0.01). The effects of plasma supplements on intestinal lymphocyte populations suggest that oral plasma proteins can modulate the degree of activation of diffuse GALT.

The high-density lipoprotein (HDL) constituent apolipoprotein CI (apoCI) protects mice against mortality in bacterial sepsis. We assessed whether high plasma apoCI levels protect against mortality from infection in humans. We determined plasma levels of apoCI, lipids, and C-reactive protein in 85-year-old participants of the prospective population-based Leiden 85-Plus Study (n 561). Participants were followed for specific causes of death. High apoCI levels were associated with 40% reduced risk of mortality from infection (hazard ratio [HR], 0.60; 95% confidence interval [CI], 0.42-0.86; p = .005) for every increase of 1 standard deviation in apoCI level. A similar association was observed for high HDL cholesterol (HR, 0.65; 95% CI, 0.46-).94; p = .022), but not for LDL cholesterol, triglycerides, and C-reactive protein levels. The association of apoCI was independent of HDL cholesterol, as multivariate analysis did not alter the association for apoCI (HR, 0.63; 95% CI, 0.44-0.90; p = .013), whereas for HDL cholesterol significance was lost. We conclude that high apoCI levels are associated with reduced mortality from infection, in line with experimental evidence in rodents. The innate immune response is a conserved trait shared by invertebrates and vertebrates. In crustaceans, circulating hemocytes play significant roles in the immune response, including the release of prophenoloxidases. Activated phenoloxidase (tyrosinase) participates in encapsulation and melanization of foreign organisms as well as sclerotization of the new exoskeleton after wound-repair or molting. Hemocyanin functions as a phenoloxidase under certain conditions and thus also participates in the immune response and molting. The relative contributions of hemocyte phenoloxidase and hemocyanin in the physiological ratio at which they occur in hemolymph have been investigated in the crab Cancer magister. Differences in activity, substrate affinity, and catalytic ability between the two enzymes indicate that hemocytes are the predominant source of phenoloxidase activity in crabs. In contrast, hemocyanin is the primary source of phenoloxidase activity in isopods and chelicerates whose hemocytes show no phenoloxidase activity. Quantitative PCR studies on the distribution of prophenoloxidase mRNA in the tissues of Carcinus maenas showed little effect relative to salinity stress. Phylogenetic analysis of hemocyanin, phenoloxidase, and other members of this arthropod gene family are consistent with the possibility that a common ancestral molecule had both phenoloxidase and oxygen-binding capabilities.

A study reveals that glutamate-gated calcium flux in the turtle brain is blocked by a protein which is present in anoxic turtle plasma, and that glutamate-mediated calcium fluxes are reduced by anoxia. To determine the activation of calcium channels in cerebrocortical brain slices from turtles which is glutamate-induced, elevations in intracellular calcium, measured with fluorescent dye fura 2, were used. Reduction glutamate-mediated calcium flux in the turtle brain was shown after 14-18 hours of anoxia, and blockade of glutamate-mediated calcium flux in turtle brain slices was produced by plasma protein from turtles in anoxic conditions.
