Abstract

Research was conducted to study the synthesis of a selective neuronal acetylcholine-gated ion channel agonist. Crude reaction mixtures containing intermediates were acidified with 6 M HCl while heterocyclic halides were coupled with substituted alkynes. Results showed that a dilution of the reaction mixture with water caused the intermediate ketoamide to precipitate out of solution. They also indicated that racemic nicotine was resolved by sequential crystallizations with d-tartaric acid and di-p-toluoyl-L-tartaric acid.

Abstract

A procedure was created for the racemization of (R)-enriched 5-bromonicotine. It involves utilization of chloroformate-induced ring opening and halogen exchange-mediated racemization of substituted pyrrolidine derivatives. The method, which can be used in the preparation of a wide array of novel nicotinic agonists and antagonists, is expected to improve yields that can be obtained in the resolution sequence. It is also useful in highlighting major differences in reactivity between unsubstituted nicotine derivatives and substituted pyridine ring.
