Abstract

Research was conducted to examine the effect of a mixture of yeast extract and bioTrypcase (YE+bT) on the growth and physiology of lactic acid producing bacteria. Results show that YE+bT increased lactic acid yield and also influenced the ratio between cellular synthesis and energy production.

Abstract

International audience; LC-MS-MS combined with Selected Reaction Monitoring (SRM) mode is a gold standard method to assay pesticides. As the number of targets increases, specific analyte transitions are monitored only around their expected retention time (RT) in order to ensure correct definition of peak shape. In this case, the analysis is secured applying RT windows much larger than the peak width to as far as possible take into account RT drift. However, any additional, even minor, modification of the LC configuration (extra-column volume change, flow rate, column length or diameter…) requires adjustment of this RT window. Because of independency towards RT, the new scout-MRM software alleviates all these limitations and expands the multiplexing capability as exemplified by multi-residue analysis of 215 pesticides. A multiplexed assay was build for 215 pesticides (174 from AB Sciex standard kit and 41 from IRSTEA, Lyon, France). LC-MS/MS was performed using Agilent 1290 system coupled to a QTRAP® 6500 system equipped with a TurboV® source. Separation was achieved on a C18 reverse phase column (Xbridge, 2.1x100x3.5µm) using a linear gradient consisting of A: water with 2mM ammonium formate and 0.05% formic acid and B: methanol with 2mM ammonium formate and 0.05% formic acid. Data were acquired with a modified version of Analyst® 1.6.2 software to implement scout-MRM. In the context of multi-residue pesticide assay, this preliminary study focused on the merit of new scout-MRM software towards other multiplexed monitoring modes in term of repeatability and accuracy. Scout signals are played by 5 spiked-in deuterated pesticides triggering 5 groups of transitions (2 transitions per pesticide, 430 transitions at all). Repeatability of scout-MRM and scheduled-MRM methods were assessed by calculating CVs from triplicate analysis of the 174 pesticide mix standard. Average CVs is about 3% for both methods, showing that they have equivalent performances. Finally, scout-MRM was used for multi-residue screening and quantification of pesticides in true surface water samples by applying an external calibration procedure. Accuracy of scout-MRM quantification results was ultimately compared to ones obtained with conventional scheduled-MRMTM approach.

Abstract

The European Space Agency's Planck satellite, launched on 14 May 2009, is the third-generation space experiment in the field of cosmic microwave background (CMB) research. It will image the anisotropies of the CMB over the whole sky, with unprecedented sensitivity (${{\Delta T}\over T}$ ~ 2 × 10-6) and angular resolution (~5 arcmin). Planck will provide a major source of information relevant to many fundamental cosmological problems and will test current theories of the early evolution of the Universe and the origin of structure. It will also address a wide range of areas of astrophysical research related to the Milky Way as well as external galaxies and clusters of galaxies. The ability of Planck to measure polarization across a wide frequency range (30-350 GHz), with high precision and accuracy, and over the whole sky, will provide unique insight, not only into specific cosmological questions, but also into the properties of the interstellar medium. This paper is part of a series which describes the technical capabilities of the Planck scientific payload. It is based on the knowledge gathered during the on-ground calibration campaigns of the major subsystems, principally its telescope and its two scientific instruments, and of tests at fully integrated satellite level. It represents the best estimate before launch of the technical performance that the satellite and its payload will achieve in flight. In this paper, we summarise the main elements of the payload performance, which is described in detail in the accompanying papers. In addition, we describe the satellite performance elements which are most relevant for science, and provide an overview of the plans for scientific operations and data analysis.

It will also address a wide range of areas of astrophysical research related to the Milky Way as well as external galaxies and clusters of galaxies. The ability of Planck to measure polarization across a wide frequency range (30-350 GHz), with high precision and accuracy, and over the whole sky, will provide unique insight, not only into specific cosmological questions, but also into the properties of the interstellar medium. This paper is part of a series which describes the technical capabilities of the Planck scientific payload. It is based on the knowledge gathered during the on-ground calibration campaigns of the major subsystems, principally its telescope and its two scientific instruments, and of tests at fully integrated satellite level. It represents the best estimate before launch of the technical performance that the satellite and its payload will achieve in flight. In this paper, we summarise the main elements of the payload performance, which is described in detail in the accompanying papers. Abstract

The European Space Agency's Planck satellite, launched on 14 May 2009, is the third-generation space experiment in the field of cosmic microwave background (CMB) research. It will image the anisotropies of the CMB over the whole sky, with unprecedented sensitivity ( ~ 2 × 10-6) and angular resolution (~5 arcmin). Planck will provide a major source of information relevant to many fundamental cosmological problems and will test current theories of the early evolution of the Universe and the origin of structure. It will also address a wide range of areas of astrophysical research related to the Milky Way as well as external galaxies and clusters of galaxies. 24. Cryogenic Tests of a 0.1 K Dilution Cooler for Planck-HFI.

Abstract

Planck is the third medium-size scientific mission (M3) of the ESA HORIZON 2000 program and is to be launched in 2007. The bolometer-based Planck/HFI (High Frequency Instrument) is cooled down to 100 mK in order to measure the Cosmic Microwave Background (CMB) radiation with sensitivity close to the fundamental photon noise limit. We report the critical design drivers for the open-cycle dilution system and the solutions adopted. The system must be able to operate for 24 months in space with severe constraints on the temperature stability. The thermomechanical architecture is described with the strong integration of the dilution with the focal plane. Thin NbTi superconducting tubes are used to achieve both a mechanical stiffness and a low thermal conductivity for the supporting focal plane. A passive thermal filter based on the high heat capacity of the Holmium-Yttrium alloy is used to achieve the low $$ 20nk/\sqrt {Hz}$$ thermal fluctuation spectrum required. In order to survive the vibration level during launch, a locking mechanism, based on the shape memory alloy NiTi, has been designed to hold the focal plane while at ambient temperature. Control of the purity of the 3He and 4He isotopes is essential for long-term operation. In addition to the initial purity control, several filtering systems are implemented within the injection system. Abstract

This article is free to read on the publishers website A moderately thermophilic, facultatively anaerobic, amylolytic bacterium was isolated from palm wine, a tropical alcoholic beverage that was sampled in Senegal. The cells were gram positive, catalase positive, non-spore forming, rod shaped, and slightly motile with peritrichous flagella. The strain which we examined did not possess cytochrome and produced L-(+)-lactate, acetate, ethanol, and formate but not hydrogen during carbohydrate fermentation. Growth occurred at pH values ranging from 5.4 to 8.5, and optimum growth occurred at around pH 7.0. The optimum temperature for growth was around 50°C, and the upper temperature limit for growth was 58°C. The guanine-plus-cytosine content of the DNA was 38.8 ± 0.2 mol%. A sequence analysis of the 16S rRNA gene revealed that the new organism is closely related phylogenetically to members of genus Bacillus. Despite the lack of spores, we propose that on the basis of phylogenetic characteristics, the new isolate should be classified as a new Bacillus species, Bacillus thermoamylovorans. The type strain is strain DKP (= Collection of Institut Pasteur CNCM 1-1378).

Abstract

Introduction: In this study the effect of subthalamic nucleus neep brain Stimulation (STN-DBS) on the gait parameters of patients with Parkinson's disease (PD) was examined using an ambulatory gait analysis system based on miniature gyroscopes. The aim was twofold: to characterize gait with and without stimulation (ON and OFF states) and to understand how STN-DBS improves motor performance in PD patients.Methods: Ten PD patients, 20±3 months after implantation of bilateral STN-DBS were asked to walk 20m. Patients did the test twice: once STN-DBS was ON and once 180 minutes after turning it OFF. Six miniature gyroscopes were attached on each wrist, thigh and shank respectively. A light data-logger (Physilog®) carried by the patients recorded angular velocities measured by gyroscopes, during each walking trial. Spatio-temporal gait parameters were estimated using a method described earlier [1]. In addition, the range of the movements of lower limbs in the sagittal plane and hands in 3D were estimated based on the integration of angular velocity during each gait cycle. Mean and stride-stride variability of all parameters were obtained for each patients. The same protocol was used to measure ten normal, age matched control subjects. Unified Parkinson's Disease Score (UPDRS III) was obtained for each patient in the ON and OFF states.Results: Compared to controls, PD patients (OFF state) had significantly shorter normalized stride length and velocity (SL=0.40 vs. 0.79, p
