17. Role of Quercetin in the Mitigation of Lindane Induced Toxicity in Terms of Oxidative Responses and Metabolic Status in Mice Brain

Abstract

Background: The current study evaluated the protective potential of quercetin against lindane induced toxicity in mice brain. For investigation, mice were allocated into four groups; the first group was the control; the second group was administered with an oral dose of lindane (25 mg/kg bw) for 4 consecutive days; the third group was exposed to quercetin (40 mg/kg bw) and to the fourth group, quercetin was administered 1 hour prior to the exposure of lindane. Objective: Two major objectives of the study were set . The first objective was to create lesions in the brain by lindane and; the second was to evaluate the neuroprotective potential of quercetin. Methods: To study oxidative responses, the levels of thiobarbituric Acid Reactive Substances (TBARS), Protein Carbonyl Content (PCC), Reduced Glutathione (GSH), Superoxide Dismutase (SOD), Catalase (CAT), and Glutathione Peroxidase (GPx) were measured in brain homogenates. Three key steps regulating enzymes of the Tricarboxylic Acid (TCA) cycle viz citrate synthase (CS), pyruvate dehydrogenase (PDH) and fumarase were also assayed. Results: Lindane treatment significantly enhanced the levels of TBARS (P Conclusion: Pretreatment with quercetin in lindane treated group not only restored previously altered biochemical parameters after lindane treatment and also significantly improved them, which suggests that quercetin is not only safe rather is neuroprotective against lindane intoxication.

Abstract

Thiamine (vitamin B1), cofactor for various multi-enzyme complexes in energy metabolism, and plays a major role in the synthesis of cholinesterases such as acetylcholinesterase (AChE); butyrylcholinesterase (BChE). Present study deals with the changes in the cholinesterases, γ-aminobutyric acid (GABA) and serotonin in mice brain following thiamine deficiency. Experimental mice (6–8 week old) were made thiamine deficient by intraperitoneal injection of pyrithiamine hydrobromide and fed with thiamine-deficient diet. Animals were divided into three groups, Group I (Control), Group II (thiamine deficient mice for 8 days), and Group III (thiamine deficient mice for 10 days). The higher serotonin level whereas significant decreases in the AChE, BChE and GABA level were recorded in treated groups as compared to control. Hence, vitamin B1 deficiency disturbs the cholinergic system and neurotransmitters levels in brain which may lead to neurodegenerative diseases. [ABSTRACT FROM AUTHOR]

46. A Comparison of Antibiotics' Resistance Patterns of E. coli and B. Subtilis in their Biofilms and Planktonic Forms.

Abstract

Background: A biofilm refers to a community of microbial cells that adhere to surfaces that are surrounded by an extracellular polymeric substance. Bacteria employ various defence mechanisms, including biofilm formation, to enhance their survival and resistance against antibiotics., Objective: The current study aims to investigate the resistance patterns of Escherichia coli (E. coli) and Bacillus subtilis (B. subtilis) in both biofilms and their planktonic forms., Methods: E. coli and B. subtilis were used to compare resistance patterns in biofilms versus planktonic forms of bacteria. An antibiotic disc diffusion test was performed to check the resistance pattern of biofilm and planktonic bacteria against different antibiotics such as penicillin G, streptomycin, and ampicillin. Biofilm formation and its validation were done by using quantitative (microtiter plate assay) and qualitative analysis (Congo red agar media)., Results: A study of surface-association curves of E. coli and B. subtilis revealed that surface adhesion in biofilms was continuously constant as compared to their planktonic forms, thereby confirming the increased survival of bacteria in biofilms. Also, biofilms have shown high resistance towards the penicillin G, ampicillin and streptomycin as compared to their planktonic form., Conclusion: It is safely inferred that E. coli and B. subtilis, in their biofilms, become increasingly resistant to penicillin G, ampicillin and streptomycin., (Copyright© Bentham Science Publishers; For any queries, please email at epub@benthamscience.net.)

Abstract

Alzheimer's disease (AD) is a specific brain disease that gradually worsens due to dementia over a long period. AD accounts for almost 60% to 80% of cases of dementia. Any damage to neurons affects their ability to communicate, leading to alteration in thinking, behaviour and feelings. Besides mental, motor abilities of an individual may also be affected due to AD. Therefore, it is cardinal to understand the key mechanisms by which either AD progression can be ceased or, after the onset of the disease it could be reverted. Both of these steps need the identification of a particular receptor or a molecular marker through which a drug can enter the neurons. Cholinergic transporters are such potential targets of AD, which regulate the movement of acetylcholine and thus regulate the nerve impulse conduction in the brain.
