7. Optical and infrared studies of cometary dust

Abstract

In 1968, Finson and Probstein outlined a theory which could be used to analyse cometary dust tails. This theory is applied to a series of P/Halley images, obtained using the UK Schmidt Telescope. The theory is successful and shows that P/Halley underwent a series of major outbursts during early 1986. The duration of the outbursts is approximately 2 days. This value coincides with the 2.2day nuclear rotation period, estimated by other observers. The size of the dust grains varies from submicron to several hundred microns and their calculated orbital parameters indicate that a considerable fraction of the larger grains contibute to the Zodiacal Dust Cloud. A range of CCD and UK Schmidt images reveal dust features in the comae of comets P/Crommelin, P/Giacobini-Zinner and P/Halley. Intensity contours for many images exhibited a "swinging round" effect i.e. for each image there was a systematic change in the photometric axis from the central condensation outwards. This was attributed to radiation pressure effects. Astrometric images of P/Halley show sharp parabolic hoods. These hoods are discussed with reference to near-nuclear dust grain orbits. Preliminary results indicate that the rotation period of Hailey is at least a few days. Mie (1908), developed a method by which the general properties of scattering and emission by dust grains, can be used to model cometary thermal spectra. Such a method is applied to several P/Halley spectra, taken during April, 1986. The results indicate that the dust grains could be composed of both an absorbing (e.g. magnetite) and a dielectric (e.g. silicate) material. The interrelations between decaying comets and Earth-crossing asteroids are discussed. Optical and infrared observations indicate that comet P/Neujmin 1 may be a transitionary object. P/Arend-Rigaux however, is still very active, displaying a large dust coma, with anisotropic emission.

8. Investigating the Performance of the Jason-2/OSTM Radar Altimeter Over Lakes and Reservoirs

Abstract

Many inland water investigations utilize archival and near-real time radar altimetry data to enable observation of the variation in surface water level. A multi-altimeter approach allows a more global outlook with improved spatial resolution, and combined long-term observations improve statistical analyses. Central to all programs is a performance assessment of each instrument. Here, we focus on data quantity and quality pertaining to the Poseidon-3 radar altimeter onboard the Jason-2/OSTM satellite.Utilizing an interim data set (IGDR), studies show that the new on-board DIODE/median and DIODE/DEM tracking modes are performing well, acquiring and maintaining the majority of lake and reservoir surfaces in varying terrains. The 20-Hz along-track resolution of the data, and particularly the availability of the range output from the ice-retracker algorithm, also improves the number of valid height measurements. Based on test-case lakes and reservoirs, output from the ice-retracker algorithm is also seen to have a clear advantage over the ocean-retracker having better height stability across calm and icy surfaces, a greater ability to gain coastline waters, and less sensitivity to loss of water surface when there is island contamination in the radar echo. Such on-board tracking and postprocessing retracking enables the lake waters to be quickly gained after coastline crossing. Values can range from <0.1 s to 2.5 s, but the majority of measurements are obtained in less than 0.4 s or <2.3 km from the coast. Validation exercises reveal that targets of 150 km2 surface area and 0.8 km width are able to be monitored offering greater potential to acquire lakes in the 100 C300 km2 size-category. Time series of height variations are also found to be accurate to 3 to 33 cm rms depending on target size and the presence of winter ice. These findings are an improvement over the IGDR/GDR results from the predecessor Jason-1 and TOPEX/Poseidon missions and can satisfy the accuracy requirements of both the science-related and operational lake study programs.

Abstract

The strain of agent from cattle infected with bovine spongiform encephalopathy (BSE) gives a typical pattern of disease in mice that remains in place after experimental passage through several intermediate species. There is strong evidence that the same agent strain plays a role in both BSE and 'new variant' Creutzfeldt-Jakob disease (vCJD). It seems that the agent interacts with genetic factors in the host to control the timing and neuropathology of the disease with extreme precision. Transmissible spongiform encephalopathies carry some form of information that details strain-specific properties.

Abstract

International audience; Large-scale climate patterns influenced temperature and weather patterns around the globe in 2011. In particular, a moderate-to-strong La Nina at the beginning of the year dissipated during boreal spring but reemerged during fall. The phenomenon contributed to historical droughts in East Africa, the southern United States, and northern Mexico, as well the wettest two-year period (2010-11) on record for Australia, particularly remarkable as this follows a decade-long dry period. Precipitation patterns in South America were also influenced by La Nina. Heavy rain in Rio de Janeiro in January triggered the country's worst floods and landslides in Brazil's history. The 2011 combined average temperature across global land and ocean surfaces was the coolest since 2008, but was also among the 15 warmest years on record and above the 1981-2010 average. The global sea surface temperature cooled by 0.1 degrees C from 2010 to 2011, associated with cooling influences of La Nina. Global integrals of upper ocean heat content for 2011 were higher than for all prior years, demonstrating the Earth's dominant role of the oceans in the Earth's energy budget. In the upper atmosphere, tropical stratospheric temperatures were anomalously warm, while polar temperatures were anomalously cold. This led to large springtime stratospheric ozone reductions in polar latitudes in both hemispheres. Ozone concentrations in the Arctic stratosphere during March were the lowest for that period since satellite records began in 1979. An extensive, deep, and persistent ozone hole over the Antarctic in September indicates that the recovery to pre-1980 conditions is proceeding very slowly. Atmospheric carbon dioxide concentrations increased by 2.10 ppm in 2011, and exceeded 390 ppm for the first time since instrumental records began. Other greenhouse gases also continued to rise in concentration and the combined effect now represents a 30% increase in radiative forcing over a 1990 baseline. Most ozone depleting substances continued to fall. The global net ocean carbon dioxide uptake for the 2010 transition period from El Nino to La Nina, the most recent period for which analyzed data are available, was estimated to be 1.30 Pg C yr(-1), almost 12% below the 29-year long-term average. Relative to the long-term trend, global sea level dropped noticeably in mid-2010 and reached a local minimum in 2011. The drop has been linked to the La Nina conditions that prevailed throughout much of 2010-11. Global sea level increased sharply during the second half of 2011. Global tropical cyclone activity during 2011 was well-below average, with a total of 74 storms compared with the 1981-2010 average of 89. Similar to 2010, the North Atlantic was the only basin that experienced above-normal activity. For the first year since the widespread introduction of the Dvorak intensity-estimation method in the 1980s, only three tropical cyclones reached Category 5 intensity level-all in the Northwest Pacific basin. The Arctic continued to warm at about twice the rate compared with lower latitudes. Below-normal summer snowfall, a decreasing trend in surface albedo, and above-average surface and upper air temperatures resulted in a continued pattern of extreme surface melting, and net snow and ice loss on the Greenland ice sheet. Warmer-than-normal temperatures over the Eurasian Arctic in spring resulted in a new record-low June snow cover extent and spring snow cover duration in this region. In the Canadian Arctic, the mass loss from glaciers and ice caps was the greatest since GRACE measurements began in 2002, continuing a negative trend that began in 1987. New record high temperatures occurred at 20 m below the land surface at all permafrost observatories on the North Slope of Alaska, where measurements began in the late 1970s. Arctic sea ice extent in September 2011 was the second-lowest on record, while the extent of old ice (four and five years) reached a new record minimum that was just 19% of normal. On the opposite pole, austral winter and spring temperatures were more than 3 degrees C above normal over much of the Antarctic continent. However, winter temperatures were below normal in the northern Antarctic Peninsula, which continued the downward trend there during the last 15 years. In summer, an all-time record high temperature of -12.3 degrees C was set at the South Pole station on 25 December, exceeding the previous record by more than a full degree. Antarctic sea ice extent anomalies increased steadily through much of the year, from briefly setting a record low in April, to well above average in December. The latter trend reflects the dispersive effects of low pressure on sea ice and the generally cool conditions around the Antarctic perimeter.

Abstract

At present diagnosis of BSE in cattle and sheep scrapie routinely requires post-mortem confirmation which is traditionally performed by neurohistological examination for Spongiform Vacuolation. Detection of disease specific forms of the host protein PrP, called PrPSc, offers alternatives. Deposits can be detected immunohistochemically, by searching tissue extracts by electron microscopy for the fibrils (scrapie associated fibrils: SAF) which are comprised of PrPSc, or immunoblotting of tissue extracts for PrPSc. In this paper we examine the possibilities of using the immunochemical detection of PrPSc diagnostically. We have raised antibody reagents to PrP, used them to determine the kinetics of PrPSc deposition in murine models of the disease and to determine if PrPSc can be detected in ruminant tissues from BSE or scrapie infected animals.

Abstract

Flow regime and total suspended solids (TSS), along with chemical and biological parameters associated with urbanization and intensive agriculture/aquaculture, impact benthic organisms, which represents the food chain foundation in aquatic systems. We designed this experiment to begin the integration of physical flow regime effects along with chemical and biological indicators on periphyton, an important benthic organism indicator, towards a goal of using periphyton (live biomass containing chlorophyll a) and pheophytin (dead chlorophyll-containing biomass) in assessments of stream health. Physical flow regimes in a laboratory flume were created using multiple roughness conditions and an in-channel weir Results suggested that one could model the hydraulic regime with a hydraulic model, HEC-RAS, to within 2% to 11% of measured velocity values. Thus, one may roughly but not precisely move computed velocities from the flume to the field. Significant interactions between biological response and hydraulic and TSS factors were observed, and the study suggested several indicators of periphyton-pheophytin response that are potentially relevant to ecological engineering applications featuring a channel. Increased mean velocities significantly reduced (P < 0.05) live periphyton and filament length. The 200 mg L-1 TSS level significantly reduced (P < 0.05) biomass and filament development. The effect of TSS was least where velocity was highest and depth was most shallow, which had the least effect on light absorption and resulted in the least sediment deposition. The intermediate TSS level (100 mg L-1) appeared to stimulate a growth response based on periphyton and filament length, although the effect was less noticeable where velocities were higher. Pheophytin tended to be highest in conditions resulting in lower periphyton values, consistent with the notion that regimes imparting physical stress would harbor the highest concentration of pheophytin. The periphyton/filament length ratio tended to be lowest in velocities less than O. 75 m s-1 except in the high TSS case, where both periphyton and filament length were low. Low velocities and low to moderate TSS would provide the most biomass for grazing organisms and would result in the most effective nutrient filtering due to long filament length. High periphyton/pheophytin ratios were associated with high TSS, velocities exceeding ∼0.5 m s-1, or both. 34. Synthesis of Analogues of Congo Red and Evaluation of Their Anti-Prion Activity

Abstract

No cure as of yet exists for any of the transmissible spongiform encephalopathies. In this paper, we describe the synthesis of analogues of Congo red and evaluation against a cellular model of infection, the SMB (scrapie mouse brain) persistently infected cell line, for their ability to inhibit the infectivity of the abnormal form of prion protein (PrP-res). The compounds have also been tested for their ability to inhibit the polymerization of PrPC by PrP-res. A number of analogues showed inhibition of PrP-res infectivity at nanomolar concentrations. Several analogues show promise; the most active compound, 2a, inhibits the formation of PrP-res in SMB cells with an EC50 of 25−50 nM.

35. Generation of a high-density rat EST map.

Abstract

We have developed a high-density EST map of the rat, consisting of >11,000 ESTs. These ESTs were placed on a radiation hybrid framework map of genetic markers spanning all 20 rat autosomes, plus the X chromosome. The framework maps have a total size of approximately 12,400 cR, giving an average correspondence of 240 kb/cR. The frameworks are all LOD 3 chromosomal maps consisting of 775 radiation-hybrid-mapped genetic markers and ESTs. To date, we have generated radiation-hybrid-mapping data for >14,000 novel ESTs identified by our Rat Gene Discovery and Mapping Project (http://ratEST.uiowa.edu), from which we have placed >11,000 on our framework maps. To minimize mapping errors, ESTs were mapped in duplicate and consensus RH vectors produced for use in the placement procedure. This EST map was then used to construct high-density comparative maps between rat and human and rat and mouse. These maps will be a useful resource for positional cloning of genes for rat models of human diseases and in the creation and verification of a tiling set of map order for the upcoming rat-genome sequencing.

39. Acetylated alpha-tubulin in Physarum: immunological characterization of the isotype and its usage in particular microtubular organelles.

Abstract

We have used monoclonal antibodies specific for acetylated and unacetylated alpha-tubulin to characterize the acetylated alpha-tubulin isotype of Physarum polycephalum, its expression in the life cycle, and its localization in particular microtubular organelles. We have used the monoclonal antibody 6-11B-1 (Piperno, G., and M. T. Fuller, 1985, J. Cell Biol., 101:2085-2094) as the probe for acetylated alpha-tubulin and have provided a biochemical characterization of the monoclonal antibody KMP-1 as a probe for unacetylated tubulin in Physarum. Concomitant use of these two probes has allowed us to characterize the acetylated alpha-tubulin of Physarum as the alpha 3 isotype. We have detected this acetylated alpha 3 tubulin isotype in both the flagellate and in the myxameba, but not in the plasmodium. In the flagellate, acetylated tubulin is present in both the flagellar axonemes and in an extensive array of cytoplasmic microtubules. The extensive arrangement of acetylated cytoplasmic microtubules and the flagellar axonemes are elaborated during the myxameba-flagellate transformation. In the myxameba, acetylated tubulin is not present in the cytoplasmic microtubules nor in the mitotic spindle microtubules, but is associated with the two centrioles of this cell. These findings, taken together with the apparent absence of acetylated alpha-tubulin in the ephemeral microtubules of the plasmodium suggest a natural correspondence between the presence of acetylated alpha-tubulin and microtubule organelles that are intrinsically stable or cross-linked.

Abstract

Flavobacterium columnare causes columnaris disease in fish. Columnaris disease is incompletely understood, and adequate control measures are lacking. The type IX secretion system (T9SS) is required for F. columnare gliding motility and virulence. The T9SS and gliding motility machineries share some, but not all, components. GldN (required for gliding and for secretion) and PorV (involved in secretion but not required for gliding) are both needed for virulence, implicating T9SS-mediated secretion in virulence. The role of motility in virulence is uncertain. We constructed and analyzed sprB , sprF , and gldJ mutants that were defective for motility but that maintained T9SS function to understand the role of motility in virulence. Wild-type cells moved rapidly and formed spreading colonies. In contrast, sprB and sprF deletion mutants were partially defective in gliding and formed nonspreading colonies. Both mutants exhibited reduced virulence in rainbow trout fry. A gldJ deletion mutant was nonmotile, secretion deficient, and avirulent in rainbow trout fry. To separate the roles of GldJ in secretion and in motility, we generated gldJ truncation mutants that produce nearly full-length GldJ. Mutant gldJ 563 , which produces GldJ truncated at amino acid 563, was defective for gliding but was competent for secretion as measured by extracellular proteolytic activity. This mutant displayed reduced virulence in rainbow trout fry, suggesting that motility contributes to virulence. Fish that survived exposure to the sprB deletion mutant or the gldJ 563 mutant exhibited partial resistance to later challenge with wild-type cells. The results aid our understanding of columnaris disease and may suggest control strategies.IMPORTANCE Flavobacterium columnare causes columnaris disease in many species of freshwater fish in the wild and in aquaculture systems. Abstract

Flavobacterium columnare causes columnaris disease in freshwater fish in both natural and aquaculture settings. This disease is often lethal, especially when fish population density is high, and control options such as vaccines are limited. The type IX secretion system (T9SS) is required for F. columnare virulence, but secreted virulence factors have not been fully identified. Many T9SS-secreted proteins are predicted peptidases, and peptidases are common virulence factors of other pathogens. T9SS-deficient mutants, such as Δ gldN and Δ porV , exhibit strong defects in secreted proteolytic activity. The F. columnare genome has many peptidase-encoding genes that may be involved in nutrient acquisition and/or virulence. Mutants lacking individual peptidase-encoding genes, or lacking up to ten peptidase-encoding genes, were constructed and examined for extracellular proteolytic activity, for growth defects, and for virulence in zebrafish and rainbow trout.
