BackgroundWidespread exposure to organophosphate ester (OPE) flame retardants with potential reproductive toxicity raises concern regarding the impacts of gestational exposure on birth outcomes. Previous studies of prenatal OPE exposure and birth outcomes had limited sample sizes, with inconclusive results.ObjectivesWe conducted a collaborative analysis of associations between gestational OPE exposures and adverse birth outcomes and tested whether associations were modified by sex.MethodsWe included 6,646 pregnant participants from 16 cohorts in the Environmental influences on Child Health Outcomes (ECHO) Program. Nine OPE biomarkers were quantified in maternal urine samples collected primarily during the second and third trimester and modeled as log2-transformed continuous, categorized (high/low/nondetect), or dichotomous (detect/nondetect) variables depending on detection frequency. We used covariate-adjusted linear, logistic, and multinomial regression with generalized estimating equations, accounting for cohort-level clustering, to estimate associations of OPE biomarkers with gestational length and birth weight outcomes. Secondarily, we assessed effect modification by sex.ResultsThree OPE biomarkers [diphenyl phosphate (DPHP), a composite of dibutyl phosphate and di-isobutyl phosphate (DBUP/DIBP), and bis(1,3-dichloro-2-propyl) phosphate] were detected in >85% of participants. In adjusted models, DBUP/DIBP [odds ratio (OR) per doubling=1.07; 95% confidence interval (CI): 1.02, 1.12] and bis(butoxyethyl) phosphate (OR for high vs. nondetect=1.25; 95% CI: 1.06, 1.46), but not other OPE biomarkers, were associated with higher odds of preterm birth. We observed effect modification by sex for associations of DPHP and high bis(2-chloroethyl) phosphate with completed gestational weeks and odds of preterm birth, with adverse associations among females. In addition, newborns of mothers with detectable bis(1-chloro-2-propyl) phosphate, bis(2-methylphenyl) phosphate, and dipropyl phosphate had higher birth weight-for-gestational-age z-scores (β for detect vs. nondetect=0.04-0.07); other chemicals showed null associations.DiscussionIn the largest study to date, we find gestational exposures to several OPEs are associated with earlier timing of birth, especially among female neonates, or with greater fetal growth. https://doi.org/10.1289/EHP13182.

Isocitrate Dehydrogenase-1 (IDH1) is commonly mutated in lower-grade diffuse gliomas. The IDH1R132H mutation is an important diagnostic tool for tumor diagnosis and prognosis; however, its role in glioma development, and its impact on response to therapy, is not fully understood. We developed a murine model of proneural IDH1R132H-mutated glioma that shows elevated production of 2-hydroxyglutarate (2-HG) and increased trimethylation of lysine residue K27 on histone H3 (H3K27me3) compared to IDH1 wild-type tumors. We found that using Tazemetostat to inhibit the methyltransferase for H3K27, Enhancer of Zeste 2 (EZH2), reduced H3K27me3 levels and increased acetylation on H3K27. We also found that, although the histone deacetylase inhibitor (HDACi) Panobinostat was less cytotoxic in IDH1R132H-mutated cells (either isolated from murine glioma or oligodendrocyte progenitor cells infected in vitro with a retrovirus expressing IDH1R132H) compared to IDH1-wild-type cells, combination treatment with Tazemetostat is synergistic in both mutant and wild-type models. These findings indicate a novel therapeutic strategy for IDH1-mutated gliomas that targets the specific epigenetic alteration in these tumors.

Removing polychlorinated biphenyls (PCBs) from subsurface water, soils, and transformer oil is crucial to save the environment from these pollutant materials. Hydrodechlorination (HDC) of PCBs consists of numerous chemical reactions and the simple kinetic models may not provide details for the process. To gain more awareness of the reaction mechanism, in the proposed approach, the isoconversional methods of the Friedman were investigated paralleling other kinetic models of Langmuir‐Hinshelwood (L‐H), Eley‐Rideal (E‐R), pseudo‐first‐order, and pseudo‐second‐order methods. The analysis was validated by laboratory results of HDC of contaminated transformer oil in front of Pd/MWCNTs. The most reactivity was observed for biphenyls with a higher number of chlorines. Finding a suitable model, Akaike Information Criteria were applied. It was attained that Friedman model was the most suitable for monitoring of HDC of PCBs in front of catalyst. Besides, E‐R reaction was appropriate to elucidate the theoretical interpretations of the adsorption and desorption of reactants and chlorinated benzene. [ABSTRACT FROM AUTHOR]

Inspired by the remarkable photochemical and photophysical properties of novel 2-(2'-hydroxyphenyl)benzothiazole (HBT) derivatives that could be potentially applied across various disciplines, in this work, effects of atomic electronegativity of chalcogen elements on excited state hydrogen bond effects and excited state intramolecular proton transfer (ESIPT) reaction of the biphenyl-modified HBT derivatives (i.e. HBT-HH-O, HBT-HH-S and HBT-HH-Se) are focused. By comparing the structural changes and infrared (IR) vibrational spectra of the HBT-HH fluorophores in toluene solvent, combined with the preliminary detection of hydrogen bond interaction by core-valence bifurcation (CVB) index, we can conclude that the hydrogen bond could be strengthened in S1 state, which is favourable for the occurrence of ESIPT reactions. The charge recombination behaviour of hydrogen bond induced by photoexcitation also further illustrates this point. Via constructing potential energy curves (PECs) based on restrictive optimisation and searching transition state (TS) form, we confirm the variations of atomic electronegativity of chalcogen have the regulatory effect on the ESIPT behaviour for HBT-HH derivatives, that is, the lower the atomic electronegativity is more conducive to the ESIPT reaction. Two new sesquiterpenoid glycosides, 8α (H)-eudesmane-1,3,11 (13)-triene-2-one -12-O-β-D-glucopyranoside (1) and dmetelisproside B (2), together with ten known compounds (3–12) were isolated from calyces of Physalis alkekengi L. var. franchetii (Mast.) Makino (PAF). Their structures were unambiguously elucidated through HR-ESI-MS, UV, IR, and NMR spectral data. Compounds 1, 10, and 12 exhibited DPPH scavenging ability with IC50 values of 33.69 ± 6.65, 6.29 ± 0.06, and 25.66 ± 3.06 μM, respectively. Additionally, 10 and 12 demonstrated weak α-glucosidase inhibition activity with IC50 values of 250.9 ± 6.60 and 347.6 ± 2.48 μM, respectively. [ABSTRACT FROM AUTHOR]

Objective: To investigate the effect of honokiol (HON) and the role of high-mobility group protein B1 (HMGB1) on the pathogenesis of severe acute pancreatitis (SAP). Methods: Thirty mice were numbered according to weight, and randomly divided into 5 groups using a random number table, including control, SAP, SAP and normal saline (SAP+NS), SAP and ethyl pyruvate (SAP+EP), or SAP+HON groups, 6 mice in each group. Samples of pancreas, intestine, and blood were collected 12 h after SAP model induction for examination of pathologic changes, immune function alterations by enzyme linked immunosorbent assay (ELISA), and Western blot. In vitro experiments, macrophages were divided into 5 groups, the control, lipopolysaccharide (LPS), LPS+DMSO (DMSO), LPS+anti-HMGB1 monoclonal antibody (mAb), and LPS+ HON groups. The tight connection level was determined by transmission electron microscopy and fluorescein isothiocyanate-labeled. The location and acetylation of HMGB1 were measured by Western blot. Finally, pyridone 6 and silencing signal transducer and activator of the transcription 1 (siSTAT1) combined with honokiol were added to determine whether the Janus kinase (JAK)/ STAT1 participated in the regulation of honokiol on HMGB1. The protein expression levels of HMGB1, JAK, and STAT1 were detected using Western blot. Results: Mice with SAP had inflammatory injury in the pancreas, bleeding of intestinal tissues, and cells with disrupted histology. Mice in the SAP+HON group had significantly fewer pathological changes. Mice with SAP also had significant increases in the serum levels of amylase, lipase, HMGB1, tumor necrosis factor- α, interleukin-6, diamine oxidase, endotoxin-1, and procalcitonin. Mice in the SAP+HON group did not show these abnormalities (P<0.01). Studies of Caco-2 cells indicated that LPS increased the levels of occludin and claudin-1 as well as tight junction permeability, decreased the levels of junctional adhesion molecule C, and elevated intercellular permeability (P<0.01). HON treatment blocked these effects. Studies of macrophages indicated that LPS led to low nuclear levels of HMGB1, however, HON treatment increased the nuclear level of HMGB1 (P<0.01). HON treatment also inhibited the expressions of JAK1, JAK2, and STAT1 (P<0.01) and increased the acetylation of HMGB1 (P<0.05). A novel class of diazonium salts is introduced for the photochemical aryl‐aryl coupling to produce (substituted) biphenyls. As common diazonium tetrafluoroborate salts fail, soluble and safe aryl diazonium trifluoroacetates are applied. In this mild synthesis route no catalysts are required to generate an aryl‐radical by irradiation with UV‐A light (365 nm). This reactive species undergoes direct C−H arylation at an arene, forming the product in reasonable reaction times. With the implementation of a continuous flow setup in a capillary photoreactor 13 different biphenyl derivatives are successfully synthesized. By integrating an inline 19F‐NMR benchtop spectrometer, samples are reliably quantified as the fluorine‐substituents act as a probe. Here, real‐time NMR spectroscopy is a perfect tool to monitor the continuously operated system, which produces fine chemicals of industrial relevance even in a multigram scale. An efficient and stable palladium species catalyst immobilized on functionalized hyper–cross-linked polymers (HCPs-Pd) has been successfully developed and applied in the Suzuki–Miyaura coupling reaction of diverse types of aromatic halides with aryl boronic acid in this work. The results demonstrated that HCPs-Pd exhibited high catalytic activity, which benefited from the microporous structure of the catalyst guaranteed high dispersibility of active palladium, and high specific surface area, large pore volume, good chemical stability. Under optimal reaction conditions, 93.0% of biphenyl yield in the Suzuki–Miyaura reaction with bromobenzene (C6H5Br) and phenylboronic acid (C6H7BO2) as raw material was obtained. The good stability of the HCPs-II-Pd catalyst was verified by five cycles experiments. Perhaps this work provides new insights into the high-yield preparation of aromatic biphenyl compounds employing HCPs-Pd as an efficient and stable catalyst in the Suzuki–Miyaura reaction. This study presents an eco‐conscious approach to enhance the efficiency of the Suzuki–Miyaura cross‐coupling reaction. We first synthesized graphene oxide nanosheets using the Hummers method and then coated them to incorporate metallic copper on their surface. Following this, we conducted various analyses, including Fourier transform infrared spectroscopy (FT‐IR), scanning electron microscopy (SEM), transmission electron microscopy (TEM), Brunauer–Emmett–Teller (BET) analysis, cyclic voltammetry (CV), and energy‐dispersive X‐ray spectroscopy (EDS) identification, to characterize these modified nanosheets. Subsequently, we utilized Cu‐modified graphene oxide nanosheets as cathode catalysts in an electro‐oxidation synthesis setup. To verify the effectiveness of this novel approach, we utilized bromobenzene and phenylboronic acid as model substrates to synthesize biphenyl compounds. The reaction yielded impressive product yields ranging from 87% to 93%. Operating under environmentally friendly conditions, this electro‐oxidation synthesis not only enhances selectivity but also significantly reduces the environmental impact of the reaction. Our findings highlight the potential of this green chemistry strategy, offering a promising avenue for sustainable and efficient organic synthesis, as evidenced by the successful coupling of bromobenzene and phenylboronic acid with consistently high yields. Honokiol, a bioactive component found in Magnolia officinalis, has shown in protecting against ischemic stroke in animal models. However, its poor water solubility has limited its clinical applications. In this study, we introduced a hydrophilic building block on the aromatic ring of honokiol, resulting in the synthesis of four new compounds (HH-A, -B, -C and -D) with significantly improved water solubility. We then investigated the neuroprotective effects of these compounds in mouse and rat models of transient middle cerebral artery occlusion/reperfusion (tMCAO/R) brain injury. Among the compounds tested, HH-A, also known as (S)-6-((3′,5-diallyl-2,4′-dihydroxy-[1,1′-biphenyl]-3-yl)amino)-6-oxohexane-1,5-diaminium chloride, showed the most promising results. HH-A was found to significantly reduced the infarct volume and brain edema in mice. It also outperformed the other three compounds and honokiol, even surpassing the effects of edaravone dexborneol. Additionally, HH-A demonstrated dose-dependent improvements in body weight, neurological deficits, and infarct volume. Further analysis in tMCAO/R rat model revealed that HH-A treatment led to significant upregulations of Nrf2 and HO-1 in the brain. HH-A also significantly reduced the expression of HNE, and exhibited anti-apoptotic effects by decreasing the expression of Bax and increasing the expression of Bcl-2. This was further supported by a decrease in the number of TUNEL positive cells. Taken together, the neuroprotective effects of HH-A may be attributed to its ability to target the Nrf2/HO-1 signaling pathway, leading to reduced oxidative stress and apoptosis in the brain. Background: Polychlorinated biphenyls (PCBs) are persistent organic pollutants classified as endocrine disruptors related to prediabetes and diabetes. Polybrominated biphenyls are similar in structure to PCBs and are used as flame retardants. Due to the increased worldwide prevalence of diabetes, there is increased interest in understanding the role of environmental and occupational pollutants in its development. The study aims to assess the relation between PCBs and PBBs in the serum of electronic workers and glycated hemoglobin level as an early indicator of prediabetes and type 2 diabetes mellitus among occupationally exposed workers. Methods: Blood samples were collected from 152 workers to assess PCBs (by GCMS), random blood sugar (RBS), and glycated hemoglobin (HbA1c). Participants were classified into two groups according to the presence or absence of PCBs in their serum and were compared for RBS and HbA1c levels. Results: Only two participants had detectable PCB derivate in their serum by GCMS, PCB 1 with methyl and benzole side chains. Regarding PBBs, 18 participants (12%) had detectable PBBs in their serum by GCMS. All participants had RBS and HbA1c levels within the normal range. No statistically significant difference was found between mean levels of RBS and HbA1c between participants with detected biphenyls and those without. Conclusion: The banning of PCB use in industry and modern automated techniques have prevented exposure to PCBs among electronics workers. Daily dietary exposure estimates from beef, pork, chicken, turkey and siluriform fish were calculated using toxic equivalency (TEQ) data from the U.S. Department of Agriculture's survey of dioxins and dioxin-like compounds (DLCs) in the domestic meat supply and consumption data. Exposure estimates for the whole population and age groups were based on mean consumption of a commodity and mean or 90th percentile TEQ dioxin levels from the survey. Ratios of the exposure estimates to the U.S. EPA oral reference dose (RfD) of 0.7 pg TEQ/kg bw/day were calculated to determine if domestic meat might contribute materially to consumer exposure. In general, normal consumption of lean beef, pork, chicken and turkey will not cause exposures exceeding the RfD. Non-lean meats will have higher dioxin levels as dioxins accumulate in fat, therefore consumption of non-lean meat might cause higher exposure than compared to lean meat. However, on a mean basis, none of the exposure estimates for non-lean beef, pork, chicken or turkey exceeded the RfD for any of the age groups. For some age groups, especially toddlers, there are commodities such as non-lean beef in the 90th percentile of dioxin TEQs and siluriform fish that might exceed the RfD on occasion. Bicyclol, an innovative hepatoprotective drug, was approved by the Chinese National Medical Products Administration (NMPA) in 2001 to treat Hepatitis B and drug-induced liver injury. Two active metabolites of bicyclol have been identified as M2 and M3. To evaluate the impact on drug safety and efficacy of possible drug-drug interactions (DDIs) associated with these metabolites, a sufficient quantity of these metabolites is required. Herein, we report a concise novel route for the synthesis of M2 and M3 using the Suzuki-Miyaura coupling as the key step. Furthermore, we complete the gram-scale syntheses of M2 and M3. New sulfur-containing mono- and bis-1,4-naphthoquinone thioethers, which are thiolated analogs of vitamin K, were synthesized. The molecular structure in the crystal-line state of the thioether with a pyrazinylethyl substituent at the sulfur atom was determined by X-ray diffraction analysis. The electrochemical transformations and spectral properties in the UV-vis range of the spectrum were studied for the obtained compounds. Most bis-1,4-naphthoquinone thioethers are characterized by two reversible two-electron reduction processes. The compound with a diphenyl sulfide bridging linker was found to have four cathodic stages corresponding to the sequential reduction of naphthoquinone fragments. Sulfides containing two naphthoquinone fragments showed the highest radical scavenging activity. The studied compounds exerted an anti/prooxidant effect in the process of lipid peroxidation of rat liver homogenate in vitro depending on the time of incubation. The thioethers exhibited antibacterial activity (9.76–78.13 µg mL−1) against Gram-positive bacteria (S. aureus ATCC 6538 and ATCC 43300, E. faecium ATCC 3576 and ATCC 28212). [ABSTRACT FROM AUTHOR]

In this work, we have prepared some common diphenyl diselenide compounds and evaluated the solvent effect on the 77Se NMR chemical shift. Thus, 77Se NMR spectra of diphenyl diselenides containing electron-withdrawing or electron-donating groups were recorded in eight different deuterated solvents with distinct polarities. The effects of temperature and concentration were also assessed and were found to play a minor role in the precise values of 77Se chemical shift. Finally, a computational study was performed to rationalize the 77Se NMR chemical shift behavior and to highlight the limits of standard approaches. [ABSTRACT FROM AUTHOR]

A new biphenyl compound: 5-aldehyde-4′-hydroxy-2,2′-dimethoxybiphenyl (1), along with 11 known phenolic compounds (2–12) were isolated from the stem of Ficus pumila Linn. Their structures were established by spectroscopic methods, including MS, UV, IR, 1 D and 2 D NMR. Compounds 4 and 10 showed weakly cytotoxicity against MCF-7, with IC50 values of 32.5 and 37.3 µM, respectively. [ABSTRACT FROM AUTHOR]

An investigation based on synthesis of 6‐arylated‐pyridine‐3‐yl)methanol derivatives: 6‐(2‐benzofuranyl)pyridin‐3‐yl)methanol (BFPM), (6‐([biphenyl]‐4‐yl)3‐pyridinyl methanol (BPPM), 6‐(3‐Flourophenyl)‐4‐pyridine‐methanol (FPPM), and 6‐(4‐Chlorophenyl)‐4‐pyridine‐methanol (CPPM) compounds is presented in this study. Furthermore, a comparison of experimental spectroscopic data (NMR, FTIR, UV‐Vis), structural factors, nonlinear optical characteristics with theoretical frontier molecular orbital analysis, natural population analysis, natural bond orbitals analysis of pyridine derivatives is drawn using density functional theory (DFT) and time‐dependent density functional theory (TD‐DFT) at the B3LYP/6‐311+G (2d,p) level of theory was used. Therefore, the DFT‐based findings and experimental data were in perfect accord. By using NBO analysis, it has been possible to understand how charge delocalization and HP interactions contribute to molecule stability. Moreover, the FMO energy levels were used to calculate global reaction characteristics, which showed that FPPM with the highest ▵ELUMO – HOMO of 4.91 eV, was less reactive and more stable than BFPM, BPPM, and CPPM. Compared to the prototype compound, NLO studies revealed that BFPM, BPPM, FPPM, and FPPM exhibit better characteristics, revealing their potential for use in photoelectric technology. Magnoliae Officinalis Cortex (MOC), an herbal drug, contains polyphenolic lignans mainly magnolol (MN) and honokiol (HK). Methotrexate (MTX), a critical drug for cancers and autoimmune deseases, is a substrate of multidrug resistance-associated protein 2 (MRP2) and breast cancer resistance protein (BCRP). This study investigated the effect of coadministration of MOC on the pharmacokinetics of MTX and relevant mechanisms. SpragueeDawley rats were orally administered MTX alone and with single dose (2.0 and 4.0 g/kg) and repeated seven doses of MOC (2.0 g/kg thrice daily for 2 days, the 7th dose given at 0.5 h before MTX). The serum concentrations of MTX were determined by a fluorescence polarization immunoassay. The results showed that a single dose of MOC at 2.0 g/kg significantly increased the AUC0-t and MRT of MTX by 352% and 308%, and a single dose at 4.0 g/kg significantly enhanced the AUC0-t and MRT by 362% and 291%, respectively. Likewise, repeated seven doses of MOC at 2.0 g/kg significantly increased the AUC0-t and MRT of MTX by 461% and 334%, respectively. Mechanism studies indicated that the function of MRP2 was significantly inhibited by MN, HK and the serum metabolites of MOC (MOCM), whereas BCRP was not inhibited by MOCM. In conclusion, coadministration of MOC markedly enhanced the systemic exposure and mean residence time of MTX through inhibiting the MRP2-mediated excretion of MTX. [ABSTRACT FROM AUTHOR]

The present work combines experiment and theory to reveal the behavior of bromo-substituted-biphenyls after an electron attachment. We experimentally determine anion lifetimes using an electron attachment–magnetic sector mass spectrometer instrument. Branching ratios of dissociative electron attachment fragments on longer timescales are determined using the electron attachment–quadrupole mass spectrometer instrument. In all cases, fragmentation is low: Only the Br− and [M–Br]− ions are detected, and [M–H]− is observed only in the case of 4-Br-biphenyl and parent anion lifetimes as long as 165 µs are observed. Such lifetimes are contradictory to the dissociation rates of 2- and 4-bromobiphenyl, as measured by the pulse radiolysis method to be 3.2 × 1010 and >5 × 1010 s−1, respectively. The discrepancy is plausibly explained by our calculation of the potential energy surface of the dissociating anion. Isolated in vacuum, the bromide anion can orbit the polarized aromatic radical at a long distance. A series of local minima on the potential energy surface allows for a roaming mechanism prolonging the detection time of such weakly bound complex anions. Background: Telmisartan (TLT) is a prototypic angiotensin receptor blocker largely used to treat hypertension worldwide. In addition to its cardioprotective effects, TLT presents pleiotropic activities and notably displays noticeable anti-inflammatory and antitumor effects. The repression of the programmed cell death-1 (PD-1)/programmed death-ligand 1 (PD-L1) immune checkpoint may be implicated antitumor action of TLT, as it is the case with many other compounds equipped with a biphenyl moiety. We have used molecular modeling to compare the interaction of TLT and derivatives with the PD-L1 dimer protein. Results: Two molecules, TLT-dimer and TLT-acylglucuronide, were found to form more stable complexes with PD-L1 than TLT itself. In parallel, the docking analysis performed with a series of 12 sartans led to the identification of Olmesartan as a potential PD-L1 binder. The stacked biphenyl unit of Olmesartan positions the molecule along the groove delimited by the two protein monomers. The flanking tetrazole and imidazole moieties, on each side of the biphenyl unit of Olmesartan, contribute favorably to the protein interaction via specific hydrogen bonding interactions. Conclusions: The computational analysis suggests a possible binding of Olmesartan to PD-L1 dimer and thus offers novel perspectives for the design of small molecules capable of interrupting the PD-1/PD-L1 immune checkpoint. The reversible hydrogenation/dehydrogenation of aromatic molecules, known as liquid organic hydrogen carriers, is considered as an attractive option for the safe storage and release of elemental hydrogen. The recently reported efficient synthetic routes to obtain methoxy-biphenyls in high yield make them promising candidates for hydrogen storage. In this work, a series of methoxy-substituted biphenyls and their structural parent compounds were studied. The absolute vapour pressures were measured using the transpiration method and the enthalpies of vaporisation/sublimation were determined. We applied a step-by-step procedure including structure–property correlations and quantum chemical calculations to evaluate the quality of thermochemical data on the enthalpies of phase transitions and enthalpies of formation of the studied methoxy compounds. The data sets on thermodynamic properties were evaluated and recommended for calculations in chemical engineering. A thermodynamic analysis of chemical reactions based on methoxy-biphenyls in the context of hydrogen storage was carried out and the energetics of these reactions were compared with the energetics of reactions of common LOHCs. The investigation of chemical constituents from the rhizomes of Ruscus aculeatus resulted in the isolation of two new biphenyl derivatives, aculebiphenyls A and B (1-2), together with two known analogs (3-4). Their chemical structures were elucidated based on extensive spectroscopic interpretation and HR-ESI-MS analysis. Compounds 3-4 were isolated from the Ruscus genus for the first time. The isolated compounds were tested for anti-inflammatory activities and antibacterial activities. Compound 1 exhibited significant inhibitory effects on LPS-induced NO production and COX-2 with IC50 values of 10.8 µM and 0.4 µM. Compound 1 also significantly down-regulated the levels of inflammatory cytokines IL-1β, IL-6, and TNF-α. Compound 1 showed moderate antibacterial activities. [ABSTRACT FROM AUTHOR]

BACKGROUND: Polybrominated biphenyls (PBB) and polychlorinated biphenyls (PCB) are persistent organic pollutants with potential endocrine-disrupting effects linked to adverse health outcomes. OBJECTIVES: In this study, we utilize high-resolution metabolomics (HRM) to identify internal exposure and biological responses underlying PCB and multigenerational PBB exposure for participants enrolled in the Michigan PBB Registry. METHODS: HRM profiling was conducted on plasma samples collected from 2013 to 2014 from a subset of participants enrolled in the Michigan PBB Registry, including 369 directly exposed individuals (F0) who were alive when PBB mixtures were accidentally introduced into the food chain and 129 participants exposed to PBB in utero or through breastfeeding, if applicable (F1). Metabolome-wide association studies were performed for PBB- 153 separately for each generation and ΣPCB (PCB-118, PCB-138, PCB-153, and PCB-180) in the two generations combined, as both had direct PCB exposure. Metabolite and metabolic pathway alterations were evaluated following a well-established untargeted HRM workflow. RESULTS: Mean levels were 1.75 ng/mL [standard deviation (SD). 13.9] for PBB-153 and 1.04 ng/mL (SD. 0.788) for ΣPCB. Sixty-two and 26 metabolic features were significantly associated with PBB-153 in F0 and F1 [false discovery rate (FDR) 푝<0.2], respectively. There were 2,861 features associated with ΣPCB (FDR 푝<0.2). Metabolic pathway enrichment analysis using a bioinformatics tool revealed perturbations associated with ΣPCB in numerous oxidative stress and inflammation pathways (e.g., carnitine shuttle, glycosphingolipid, and vitamin B9 metabolism). Metabolic perturbations associated with PBB-153 in F0 were related to oxidative stress (e.g., pentose phosphate and vitamin C metabolism) and in F1 were related to energy production (e.g., pyrimidine, amino sugars, and lysine metabolism). Using authentic chemical standards, we confirmed the chemical identity of 29 metabolites associated with ΣPCB levels (level 1 evidence). CONCLUSIONS: Our results demonstrate that serum PBB-153 is associated with alterations in inflammation and oxidative stress-related pathways, which differed when stratified by generation. We also found that ΣPCB was associated with the downregulation of important neurotransmitters, serotonin, and 4-aminobutanoate. Background Sacubitril/Valsartan has been highly efficacious in randomized trials of heart failure with reduced ejection fraction (HFrEF). However, the effectiveness of sacubitril/valsartan in older patients hospitalized for HFrEF in real-world US practice is unclear. Methods and Results This study included Medicare beneficiaries age ≥65 years who were hospitalized for HFrEF ≤40% in the Get With The Guidelines-Heart Failure registry between October 2015 and December 2018, and eligible for sacubitril/valsartan. Associations between discharge prescription of sacubitril/valsartan and clinical outcomes were assessed after inverse probability of treatment weighting and adjustment for other HFrEF medications. Overall, 1551 (10.9%) patients were discharged on sacubitril/valsartan. Of those not prescribed sacubitril/valsartan, 7857 (62.0%) were prescribed an angiotensin-converting enzyme inhibitor/angiotensin II receptor blocker. Over 12-month follow-up, compared with a discharge prescription of angiotensin-converting enzyme inhibitor/angiotensin II receptor blocker, sacubitril/valsartan was independently associated with lower all-cause mortality (adjusted hazard ratio [HR], 0.82; 95% CI, 0.72-0.94; P=0.004) but not all-cause hospitalization (adjusted HR, 0.97; 95% CI, 0.89-1.07; P=0.55) or heart failure hospitalization (adjusted HR, 1.04; 95% CI, 0.91-1.18; P=0.59). AimsImproving the health status (symptoms, function, and quality of life) of patients with heart failure with reduced ejection fraction (HFrEF) is a primary treatment goal. Angiotensin receptor neprilysin inhibitors (ARNI) improve short-term health status in clinical practice, but the sustainability of these improvements is unknown.Methods and resultsIn CHAMP-HF, a multicentre observational study of outpatients with HFrEF, patients initiated on ARNI were propensity score matched 1:2 to patients not using ARNI with Cox regression modelling time to ARNI initiation, adjusted for sociodemographic and clinical variables, medical history, medications, and baseline Kansas City Cardiomyopathy Questionnaire (KCCQ) scores. Repeated measures models for the overall KCCQ score and each domain compared the health status trajectories of patients initiated on ARNI vs. not. Among 3930 participants, 746 (19.0%) began ARNI, of whom 576 were matched to 1152 non-ARNI patients. Prior to matching, participants initiated on ARNI were younger, non-Hispanic, had lower EFs, more commonly had a history of ventricular arrhythmia, were less likely to be taking an ACEI/ARB, and more likely to be treated with beta-blockers and mineralocorticoid receptor antagonists. We have analyzed the nature and role of functional groups on the self‐assembly modes and the physical properties of multicomponent gels with structurally similar individual components. The gelation properties of individual and mixed enantiomeric compounds of biphenyl bis‐(amides) of alanine (BPA) or phenylalanine (BPP) methyl ester were analyzed in various solvent/solvent mixtures. Multicomponent gels were formed by mixing the enantiomeric BPP compounds at a lower concentration, but a higher concentration was required for mixed alanine‐based BPA gels. The comparison of the mechanical strength of the individual and mixed BPP compounds indicated that the mixed BPP gels displayed enhanced mechanical strength (∼2‐fold increase) in p‐xylene, but a weaker gel was observed in DMSO/water. However, a reverse trend was observed for BPA gels, indicating the role of functional groups in the gel network formation. X‐ray diffraction analysis of the gelator and the xerogels in the solid state confirmed the formation of co‐assembled networks in mixed enantiomeric gels. The stability of the gels towards anions was evaluated by analyzing the anion induced stimuli‐responsive properties. The influence of origin, depositional environment and thermal maturity of organic matter on the occurrence of aromatic hydrocarbons in source rocks from the central and northwestern Niger Delta was investigated. Eighty two source rock samples from four oil wells in the central and northwestern Niger Delta were analyzed for the aromatic hydrocarbon content using gas chromatography-mass spectrometry (GC-MS). The results of analysis of aromatic hydrocarbon fractions of the source rock extracts show the presence of many classes of aromatic hydrocarbons, which include naphthalenes, phenanthrenes, biphenyls, dibenzothiophenes and fluorenes. Trimethylnaphthalene (TMN) is the most abundant among the naphthalenes while dimethylphenanthrene (DMP) is the most abundant among the phenanthrenes. Among the biphenyls, 3-methylbiphenyl (3 MB) is the most abundant while 4-methyldibenzothiophene (4MDBT) is the most abundant among the dibenzothiphenes. Only two fluorenes were detected, and fluorene is more in abundance than 1-methylfluorene (1 MF). Depositional environment indicators of aromatic hydrocarbon organic matter reveal that the organic materials in the source rocks in these wells were deposited in marine-to-swamp depositional environments under reducing to suboxic conditions. Thermal maturity indicators calculated from the abundance of the aromatic hydrocarbons indicate that the source rock samples are thermally mature. Most of the source rocks are at the peak of oil window while a few at the early oil window. Background and Aim: In recent years, there has been an increasing interest in determining the antibacterial effect of honey from different regions. Yet, the data available on the pollen profile and the antibacterial activity of Algerian honey are very few compared to Algeria's significant botanical, climatic, and geographical diversity. Therefore, this research aims to study the qualitative melissopalynology analysis, Glucose oxidase (GOX) activity, and the antibacterial effect of honey samples from different botanical and geographical origins. Materials and Methods: Five natural honey samples were collected from local beekeepers. The antibacterial effect was carried out towards pathogenic bacteria, including Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. The antibacterial activity of honey samples was evaluated using six dilutions (80%, 40%, 20%, 10%, 5%, and 2.5%). The bacterial susceptibility to honey was evaluated by the agar well diffusion and broth dilution assays. GOX activity was determined using hydrogen peroxide as a standard with peroxidase and o-dianisidine. Results: The results showed that two honey samples could be classified as monofloral (H1: Hedysarum coronarium, H4: Ziziphus spp), whereas honey samples H2, H3, and H5 are multifloral honey. All honey exhibits a good bactericidal effect against pathogenic bacteria. Statistical analysis showed that there was a correlation between GOX level and the antibacterial of honey samples. Conclusion: It can be concluded that honey is an important natural product which has an antibacterial effect towards pathogenic bacteria. Therefore, honey should be used as an alternative therapy for the treatment of wounds infected by multidrug-resistant bacteria. AimsThe aim of our study was to investigate heterogeneity of health status treatment response of sacubitril/valsartan in patients with heart failure with reduced ejection fraction (HFrEF).Methods and resultsWe leveraged data from CHAMP-HF, an observational registry of 140 US clinics and 5026 outpatients with chronic HFrEF, where health status was serially assessed using the Kansas City Cardiomyopathy Questionnaire (KCCQ)-12 Overall Summary Scale (range from 0 to 100; ≥20-point improvement is a very large improvement). In 334 patients newly initiated on sacubitril/valsartan, we used hierarchical multivariable logistic regression (13 patient-level characteristics as well as baseline KCCQ-12 score) to calculate the odds ratio (OR) of any characteristic being associated with a very large health status improvement. A total of 104/334 (31.1%) of patients achieved the primary endpoint, where only worse baseline health status [KCCQ-12 score of 0-60 points had an OR = 0.86/5-point higher score (CI 0.79, 0.93)], and those with a KCCQ-12 score of 60-80 points had an OR = 0.61/5-point higher score (0.45-0.82), which was associated with a very large benefit. No other patient characteristic was associated with a very large health status improvement (P > 0.05).ConclusionsWe found that, after initiation of sacubitril/valsartan, only worse baseline health status was associated with very large health status improvement. Accordingly, a trial of therapy-particularly in those with worse symptoms, function, and quality of life-and assessing treatment response are likely to be the best prospective strategy.

Laser diodes based on organic semiconductor materials have high threshold current densities that require the suppression of various inherent loss processes. One way to study such loss processes is to analyze the external quantum efficiency (EQE) roll-off in organic light-emitting diodes (OLEDs). In this work, we used electrical simulations to analyze the origin of the experimental EQE roll-off of an OLED based on 4,4′-bis[(N-carbazole)styryl]biphenyl (BSBCz) under extremely high current injection (∼1 kA/cm2). We considered various singlet exciton annihilations and quenching processes (i.e., singlet-singlet annihilation, singlet-triplet annihilation, singlet-polaron annihilation, singlet-heat quenching, and electric field quenching of singlet excitons). These results showed that the EQE roll-off can be attributed to Joule heating and/or singlet-triplet annihilation and/or the dissociation of singlet excitons under a high applied electric field. The electric field quenching of singlet excitons was confirmed by a field-induced photoluminescence (PL) quenching experiment. By applying an electric field-induced charge dissociation model to both the EQE and field-induced PL quenching, we estimated the singlet exciton binding energy of a BSBCz film to be in the range of 0.64–0.71 eV. Diaryl-furanones are specific analytical reagents for the biochemical detection of primary amines by fluorescence techniques. Well-known reagents are fluorescamine (Fluram) and 2-methoxy-2,4-diphenyl-3(2H)-furanone (MDPF), yielding fluorescent products with λem at 480–490 nm. Although the reaction products claim to be pyrrolinones, recent studies show that they are really 3-oxopyrrole (pyrrolone) derivatives. Both reagents have been used for the cytochemical demonstration of primary amines. In this work, we have applied the fluorescent products of MDPF with amines (n-butylamine, BA; glucosamine, GA; and spermine, Sp), which showed interesting fluorescence reactions with chromatin DNA. 2,4-diphenyl-3-oxopyrrole products (diPOPy) can be easily synthesized according to well-known procedures, by mixing solutions of MDPF in acetone with water at pH 9 containing the amino compounds. DiPOPy derivatives of BA, GA, and Sp were used for spectroscopic, microscopic, and molecular modeling studies, showing a bright and selective blue–green fluorescence on DNA substrates, mainly chromatin, kinetoplast DNA, and stretched chromatin fibers. The cationic diPOPy fluorophore is planar, with a high partial positive charge in the N atom, and suitable for intercalative binding to DNA. A mechanism of fluorescamine fluorescence due to an inner-salt isomeric form is proposed, and an astonishing correlation between adenine–thymine-rich centromeric heterochromatin in mouse metaphase chromosomes after reaction of the fluorescamine reagent with protein amino groups is also discussed. Introduction: Phenols are a large family of natural and synthetic compounds with known antioxidant activity. The aim of this study was to perform in vitro screening of natural and natural-like phenol monomers and their C2-symmetric dimers (hydroxylated biphenyls) in order to identify those representatives whose pharmacophores have the strongest antioxidant and the lowest prooxidant activity. Material and methods: Antioxidative properties of 36 compounds (monomers and their C2-symmetric dimers) were evaluated in vitro. Different (red/ox) assays were used to measure their total oxidative potential (TOP), their total antioxidative capacity (TAC), the pro-oxidative-antioxidant balance (PAB) and total SH-group content (SHG) in a biologically relevant environment. The Pro-oxidative Score, Antioxidative Score and the Oxy Score were also calculated. Trolox, a water soluble analogue of α-tocopherol, was used as a positive control. Results: In an assay consisting of pooled human serum, 6 of the 36 compounds showed significant antioxidant activity (compounds 6, 7, 12, 13, 26, and 27), whereas 4 showed extremely weak antioxidant activity (compounds 2, 29, 30, and 31). Within the 36 compounds comprising zingerone, dehydrozingerone, aurone, chalcone, and magnolol derivatives, in both monomeric and dimeric forms, the 2 compounds that indicated the highest antioxidant activity were dehydrozingerone derivatives (compounds 6 and 12). Trolox's activity was found between the strong and weak antioxidant compounds analysed in our study. Conclusions: In this study selected dehydrozingerones were identified as good candidates for in-depth testing of their biological behaviour and for possible precursors for synthesis of novel polyphenolic molecules with potential therapeutic applications. South Pars Industrial Energy Zone, located in the southwest of Iran along the Persian Gulf coast, encompasses many industrial units in the vicinity of urban areas. This research study investigated the effects of polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) on human health and the environment. Suspended particulate matters (SPM) in the air sampled, in summer and winter 2019, from ten stations next to industrial units and residential areas. The samples were analyzed by gas chromatography-mass spectrometry (GC–MS). Spatial distribution maps of pollutants in the region were prepared using GIS software. The highest carcinogenic risk due to PAHs and PCBs measured as ( 6.07 × 10 - 6 ± 1.85822 × 10 - 6 ) and ( 8.80 × 10 - 8 ± 2.76786 × 10 - 8 ) , respectively. According to the US Environmental Protection Agency limit ( 1 × 10 - 6 ), the cancer risks from PAH compounds were significant and need further investigation. The PCB cancer risks were within acceptable ranges. The highest adsorption ratios for PAHs were obtained through skin and PCBs by ingestion. The maximum measured non-carcinogenic hazard indexes (HI) turned out to be 0.037 and 0.023 for PAH and PCB, respectively, and were reported as acceptable risks. The predominant source of PAH in industrial areas was liquid fossil combustion, and in urban areas replaced by coal-wood-sugarcane combustion. Petrochemical complexes, flares, power plants (69%), electric waste disposal sites, and commercial pigments (31%) were reported as PCB sources. Industries activities were the most effective factors in producing the highest level of carcinogenic compounds in the region, and it is necessary to include essential measures in the reform programs. BACKGROUND: Amniogenesis is a key event in biochemical pregnancy, and its failure may result in human embryonic death. However, whether and how environmental chemicals affect amniogenesis remain largely unknown. OBJECTIVES: The objective of the present study was to screen chemicals that may disrupt amniogenesis in an amniotic sac embryoid model and to investigate the potential mechanism of amniogenesis failure, with a focus on organophosphate flame retardants (OPFRs). METHODS: This study developed a high-throughput toxicity screening assay based on transcriptional activity of octamer-binding transcription factor 4 (Oct4). For the two positive OPFR hits with the strongest inhibitory activity, we used time-lapse and phase-contrast imaging to assess their effects on amniogenesis. Associated pathways were explored by RNA-sequencing and western blotting, and potential binding target protein was identified through a competitive binding experiment RESULTS: Eight positive hits exhibiting Oct4 expression were identified, with 2-ethylhexyl-diphenyl phosphate (EHDPP) and isodecyl diphenyl phosphate (IDDPP) showing the strongest inhibitory activity. EHDPP and IDDPP were found to disrupt the rosette-like structure of the amniotic sac or inhibit its development. Functional markers of squamous amniotic ectoderm and inner cell mass were also found disrupted in the EHDPP- and IDDPPexposed embryoids. Mechanistically, embryoids exposed to each chemical exhibited abnormal accumulation of phosphorylated nonmuscle myosin (p-MLC-II) and were able to bind to integrin β1 (ITGβ1). CONCLUSION: The amniotic sac embryoid models suggested that OPFRs disrupted amniogenesis likely by inhibiting the ITGb1 pathway, thus providing direct in vitro evidence associating OPFRs with biochemical miscarriage. Polystyrene anchored palladium (II) complex of 2-acetylbenzimidazole was synthesized and characterized by elemental analysis, FT-IR, UV–Vis/DRS spectroscopic and thermogravimetric analysis. Based on the analytical studies, square planar geometry was assigned to Pd (II) ion in the polymer supported palladium complex. The novel polymer supported palladium complex exhibited excellent catalytic activity towards Suzuki–Miyaura cross-coupling reaction of 4-bromoanisole with differently substituted boronic acids in H2O/EtOH solvent system, to obtain coupled products in excellent yields. Supported complex exhibited better activity and recyclability as compared to unsupported palladium complex. Based on the results obtained, a plausible mechanism for the Suzuki–Miyaura cross-coupling reaction has been proposed. BackgroundPulmonary hypertension (PH) results in increased right ventricular (RV) afterload and ventricular remodeling. Sacubitril/valsartan (sac/val) is a dual acting drug, composed of the neprilysin inhibitor sacubitril and the angiotensin receptor blocker valsartan, that has shown promising outcomes in reducing the risk of death and hospitalization for chronic systolic left ventricular heart failure. In this study, we aimed to examine if angiotensin receptor-neprilysin inhibition using sac/val attenuates RV remodeling in PH.Methods and ResultsRV pressure overload was induced in Sprague-Dawley rats via banding the main pulmonary artery. Three different cohorts of controls, placebo-treated PH, and sac/val-treated PH were studied in a 21-day treatment window. Terminal invasive hemodynamic measurements, quantitative histological analysis, biaxial mechanical testing, and constitutive modeling were employed to conduct a multiscale analysis on the effects of sac/val on RV remodeling in PH. Sac/val treatment decreased RV maximum pressures (29% improvement, P=0.002), improved RV contractile (30%, P=0.012) and relaxation (29%, P=0.043) functions, reduced RV afterload (35% improvement, P=0.016), and prevented RV-pulmonary artery uncoupling. Furthermore, sac/val attenuated RV hypertrophy (16% improvement, P=0.006) and prevented transmural reorientation of RV collagen and myofibers (P=0.011). The combined natriuresis and vasodilation resulting from sac/val led to improved RV biomechanical properties and prevented increased myofiber stiffness in PH (61% improvement, P=0.032).ConclusionsSac/val may prevent maladaptive RV remodeling in a pressure overload model via amelioration of RV pressure rise, hypertrophy, collagen, and myofiber reorientation as well as tissue stiffening both at the tissue and myofiber level.

Adipocytes are critical for ovarian cancer cells to home to the omentum, but the metabolic changes initiated by this interaction are unknown. To this end, we carried out unbiased mass spectrometry-based metabolomic and proteomic profiling of cancer cells cocultured with primary human omental adipocytes. Cancer cells underwent significant proteo-metabolomic alteration(s), typified by changes in the lipidome with corresponding upregulation of lipid metabolism proteins. FABP4, a lipid chaperone protein, was identified as the critical regulator of lipid responses in ovarian cancer cells cocultured with adipocytes. Subsequently, knockdown of FABP4 resulted in increased 5-hydroxymethylcytosine levels in the DNA, downregulation of gene signatures associated with ovarian cancer metastasis, and reduced clonogenic cancer cell survival. In addition, clustered regularly interspaced short palindromic repeats (CRISPR)-mediated knockout of FABP4 in high-grade serous ovarian cancer cells reduced metastatic tumor burden in mice. Consequently, a small-molecule inhibitor of FABP4 (BMS309403) not only significantly reduced tumor burden in a syngeneic orthotopic mouse model but also increased the sensitivity of cancer cells toward carboplatin both in vitro and in vivo. Taken together, these results show that targeting FABP4 in ovarian cancer cells can inhibit their ability to adapt and colonize lipid-rich tumor microenvironments, providing an opportunity for specific metabolic targeting of ovarian cancer metastasis. Glioblastoma (GBM) responses to bevacizumab are invariably transient with acquired resistance. We profiled paired patient specimens and bevacizumab-resistant xenograft models pre- and post-resistance toward the primary goal of identifying regulators whose targeting could prolong the therapeutic window, and the secondary goal of identifying biomarkers of therapeutic window closure. Bevacizumab-resistant patient specimens and xenografts exhibited decreased vessel density and increased hypoxia versus pre-resistance, suggesting that resistance occurs despite effective therapeutic devascularization. Microarray analysis revealed upregulated mesenchymal genes in resistant tumors correlating with bevacizumab treatment duration and causing three changes enabling resistant tumor growth in hypoxia. First, perivascular invasiveness along remaining blood vessels, which co-opts vessels in a VEGF-independent and neoangiogenesis-independent manner, was upregulated in novel biomimetic 3D bioengineered platforms modeling the bevacizumab-resistant microenvironment. Second, tumor-initiating stem cells housed in the perivascular niche close to remaining blood vessels were enriched. Third, metabolic reprogramming assessed through real-time bioenergetic measurement and metabolomics upregulated glycolysis and suppressed oxidative phosphorylation. Single-cell sequencing of bevacizumab-resistant patient GBMs confirmed upregulated mesenchymal genes, particularly glycoprotein YKL-40 and transcription factor ZEB1, in later clones, implicating these changes as treatment-induced. Serum YKL-40 was elevated in bevacizumab-resistant versus bevacizumab-naïve patients. CRISPR and pharmacologic targeting of ZEB1 with honokiol reversed the mesenchymal gene expression and associated stem cell, invasion, and metabolic changes defining resistance. Honokiol caused greater cell death in bevacizumab-resistant than bevacizumab-responsive tumor cells, with surviving cells losing mesenchymal morphology. Employing YKL-40 as a resistance biomarker and ZEB1 as a target to prevent resistance could fulfill the promise of antiangiogenic therapy. SIGNIFICANCE: Bevacizumab resistance in GBM is associated with mesenchymal/glycolytic shifts involving YKL-40 and ZEB1. Targeting ZEB1 reduces bevacizumab-resistant GBM phenotypes. BackgroundIn PIONEER-HF (Comparison of Sacubitril/Valsartan Versus Enalapril on Effect on NT-pro BNP in Patients Stabilized From an Acute Heart Failure Episode), the in-hospital initiation of sacubitril/valsartan in patients hospitalized for acute decompensated heart failure (ADHF) was well-tolerated and led to improved outcomes. We aim to determine the representativeness of the PIONEER-HF trial among patients hospitalized for ADHF using real-world data.MethodsThe study population was derived from all patients discharged alive for ADHF in the Get With The Guidelines-HF registry from 2006 to 2018 with HF with reduced ejection fraction (HFrEF; all HFrEF with ADHF). We then determined the proportion of patients meeting PIONEER-HF eligibility criteria (PIONEER-HF eligible) and those meeting a set of limited eligibility criteria (actionable cohort). Rates of HF readmissions and all-cause mortality were then compared between the all HFrEF with ADHF, PIONEER-HF eligible, and actionable cohorts using linked Medicare claims data.ResultsA total of 99 767 patients with HFrEF in Get With The Guidelines-HF were hospitalized for ADHF. PIONEER-HF inclusion criteria were met by 71 633 (71.8%) patients, and both inclusion and exclusion criteria were met by 20 704 (20.8%) patients. Further, 68 739 (68.9%) patients met the criteria for the actionable cohort. Among the Centers for Medicare and Medicaid-linked patients, the HF rehospitalization rate at 1 year was 35.1% (95% CI, 34.5-35.8) for all HFrEF with ADHF patients, 32.6% (95% CI, 31.3-33.9) for the PIONEER-HF eligible cohort, and 33.1% (95% CI, 32.3-33.9) for the actionable cohort. The 1-year all-cause mortality was 36.7% (95% CI, 36.1-7.4) for all HFrEF with ADHF patients, 31.6% (95% CI, 30.3-32.9) for the PIONEER-HF eligible cohort, and 32.2% (95% CI, 31.4-33.0) for the actionable cohort.ConclusionsPatient characteristics and clinical outcomes for patients eligible for PIONEER-HF only modestly differ when compared with those encountered in routine practice, suggesting that the in-hospital initiation of sacubitril/valsartan should be routinely considered for patients with HFrEF hospitalized for ADHF.

A simple and efficient nitrile-directed meta-C-H olefination, acetoxylation, and iodination of biaryl compounds is reported. Compared to the previous approach of installing a complex U-shaped template to achieve a molecular U-turn and assemble the large-sized cyclophane transition state for the remote C-H activation, a synthetically useful phenyl nitrile functional group could also direct remote meta-C-H activation. This reaction provides a useful method for the modification of biaryl compounds because the nitrile group can be readily converted to amines, acids, amides, or other heterocycles. Notably, the remote meta-selectivity of biphenylnitriles could not be expected from previous results with a macrocyclophane nitrile template. DFT computational studies show that a ligand-containing Pd-Ag heterodimeric transition state (TS) favors the desired remote meta-selectivity. Control experiments demonstrate the directing effect of the nitrile group and exclude the possibility of non-directed meta-C-H activation. Substituted 2-pyridone ligands were found to be key in assisting the cleavage of the meta-C-H bond in the concerted metalation-deprotonation (CMD) process.

ObjectivesThis study sought to describe the short-term health status benefits of angiotensin-neprilysin inhibitor (ARNI) therapy in patients with heart failure and reduced ejection fraction (HFrEF).BackgroundAlthough therapy with sacubitril/valsartan, a neprilysin inhibitor, improved patients' health status (compared with enalapril) at 8 months in the PARADIGM-HF (Prospective Comparison of ARNI with ACE inhibitor to Determine Impact on Global Mortality and Morbidity in Heart Failure) study, the early impact of ARNI on patients' symptoms, functions, and quality of life is unknown.MethodsHealth status was assessed by using the 12-item Kansas City Cardiomyopathy Questionnaire (KCCQ) in 3,918 outpatients with HFrEF and left ventricular ejection fraction ≤40% across 140 U.S. centers in the CHAMP-HF (Change the Management of Patients with Heart Failure) registry. A comparative in vitro study of the antioxidant potential of natural phenols (zingerone, curcumin, raspberry ketone, magnolol) and their synthesized derivatives was performed. The antioxidant efficiency was evaluated in blood serum obtained from healthy individuals, by means of spectrophotometry, before and after the addition of pro-oxidant tert-butyl hydroperoxide (TBH). Moreover, the antioxidant effect of an equimolar mixture of curcumin and zingerone was investigated. Interpretation of our results reveals that in the blood serum of healthy individuals curcumin (C1), raspberry ketone (RK1), magnolol (M1) and synthesized derivative of zingerone (Z2) demonstrate remarkable antioxidant effects (p < 0.05). However, in the state of TBH-induced excessive oxidative stress natural magnolol and synthesized derivatives C1, Z1 and RK1 show powerful antioxidant activity and thus can be further investigated to obtain information about their metabolic transformations and their potential influence at the cellular level. Results obtained from measurements in an equimolar mixture of zingerone and curcumin indicate synergism (p < 0.05) between the two compounds. This combination is especially successful due to the fast and efficient neutralization of added pro-oxidant TBH. The commercial availability of turmeric and ginger and their frequent combined use in diet suggest ideas for further broader utilization of the beneficial synergistic effect of their phenolic components. A simple, readily prepared biphenyl bis‐amidinium compound (1⋅Cl2) is able to selectively precipitate sulfate from water. The precipitant is effective at concentrations as low as 1 mM and shows complete selectivity against monovalent anions, and high selectivity even against CO32− and HPO42−. It is highly effective (>90 % sulfate removed) in both seawater and highly acidic conditions relevant to mining waste‐streams. X‐ray crystallography reveals that 1⋅SO4 forms a tightly packed, anhydrous, structure where each sulfate anion receives eight hydrogen bonds from amidinium N−H hydrogen bond donors. Our recent studies for nonnucleoside reverse transcriptase inhibitors identified a highly potent compound JK-4b against WT HIV-1 (EC 50 = 1.0 nmol/L), but the poor metabolic stability in human liver microsomes (t 1/2 = 14.6 min) and insufficient selectivity (SI = 2059) with high cytotoxicity (CC 50 = 2.08 μmol/L) remained major issues associated with JK-4b. The present efforts were devoted to the introduction of fluorine into the biphenyl ring of JK-4b, leading to the discovery of a novel series of fluorine-substituted NH 2 -biphenyl-diarylpyrimidines with noticeable inhibitory activity toward WT HIV-1 strain (EC 50 = 1.8–349 nmol/L). The best compound 5t in this collection (EC 50 = 1.8 nmol/L, CC 50 = 117 μmol/L) was 32-fold in selectivity (SI = 66,443) compared to JK-4b and showed remarkable potency toward clinically multiple mutant strains, such as L100I, K103N, E138K, and Y181C. The metabolic stability of 5t was also significantly improved (t 1/2 = 74.52 min), approximately 5-fold higher than JK-4b in human liver microsomes (t 1/2 = 14.6 min). Also, 5t possessed good stability in both human and monkey plasma. No significant in vitro inhibition effect toward CYP enzyme and hERG was observed. The single-dose acute toxicity test did not induce mice death or obvious pathological damage. These findings pave the way for further development of 5t as a drug candidate. In the present study, we reported a series of fluorine-substituted NH 2 -biphenyl-diarylpyrimidines with noticeable anti-HIV activity, of which 5t exhibited significantly improved druggability. In this study, the synthesis and characterization of 2-(2-fluorobiphenyl-4-yl)-N'-[(substituted methylene]propanehydrazides (3a-s) and 2-(2-fluoro-[1,1'-biphenyl]-4-yl)-N-(5-methyl-2-(substituted aryl)-4-oxothiazolidin-3-yl)propanamides (4a-s) are described and also the antiproliferative effect of the compounds on HT-29, HeLa, A549 and MCF-7 cancer cell lines is investigated. Additionally, mouse embryonic fibroblast cells NIH3T3 were also evaluated to determine the selectivity. The results showed that the identified compounds did not cause any toxicity against NIH3T3 cell line. Moreover, N-(2-(3,5-Bis(trifluoromethyl)phenyl)-5-methyl-4-oxothiazolidin-3-yl)-2-(2-fluoro-[1,1'-biphenyl]-4-yl)propanamide (4h) had the most growth inhibitory effect (55.97% inhibition) on HT-29 colorectal adenocarcinoma cell line. Diphenyl phosphate (DPHP) is an aryl phosphate ester (APE) used as an industrial catalyst and chemical additive and is the primary metabolite of flame retardant APEs, including triphenyl phosphate (TPHP). Minimal DPHP-specific toxicity studies have been published despite ubiquitous exposure within human populations following metabolism of TPHP and other APEs. Therefore, the objective of this study was to determine the potential for DPHP-induced toxicity during embryonic development. Using zebrafish as a model, we found that DPHP significantly increased the distance between the sinus venosus and bulbus arteriosis (SV-BA) at 72 h postfertilization (hpf) following initiation of exposure before and after cardiac looping. Interestingly, pretreatment with d-mannitol mitigated DPHP-induced effects on SV-BA length despite the absence of DPHP effects on pericardial area, suggesting that DPHP-induced cardiac defects are independent of pericardial edema formation. Using mRNA-sequencing, we found that DPHP disrupts pathways related to mitochondrial function and heme biosynthesis; indeed, DPHP significantly decreased hemoglobin levels in situ at 72 hpf following exposure from 24 to 72 hpf. Overall, our findings suggest that, similar to TPHP, DPHP impacts cardiac development, albeit the potency of DPHP is significantly less than TPHP within developing zebrafish.

Background The angiotensin-receptor/neprilysin inhibitor ( ARNI ) sacubitril/valsartan reduces hospitalization and mortality for patients with heart failure with reduced ejection fraction. However, adoption of ARNI into clinical practice has been slow. Factors influencing use of ARNI have not been fully elucidated. Using data from the Get With The Guidelines-Heart Failure registry, Hospital Compare, Dartmouth Atlas, and the American Hospital Association Survey, we sought to identify hospital characteristics associated with patient-level receipt of an ARNI prescription. Methods and Results We analyzed patients with heart failure with reduced ejection fraction who were eligible for ARNI prescription (ejection fraction≤40%, no contraindications) and hospitalized from October 1, 2015 through December 31, 2016. We used logistic regression to estimate the associations between hospital characteristics and patient ARNI prescription at hospital discharge, accounting for clustering of patients within hospitals using generalized estimating equation methods and adjusting for patient-level covariates. Of 16 674 eligible hospitalizations from 210 hospitals, 1020 patients (6.1%) were prescribed ARNI at discharge. The median hospital-level proportion of patients prescribed ARNI was 3.3% (Q1, Q3: 0%, 12.6%). After adjustment for patient-level covariates, for-profit hospitals had significantly higher odds of ARNI prescription compared with not-for-profit hospitals (odds ratio, 2.53; 95% CI , 1.05-6.10; P=0.04), and hospitals located in the Western United States had lower odds of ARNI prescription compared with those in the Northeast (odds ratio, 0.33; 95% CI , 0.13-0.84; P=0.02). Conclusions Relatively few hospital characteristics were associated with ARNI prescription at hospital discharge, in contrast to what has been observed in early adoption in other disease areas. Additional evaluation of barriers to implementing new evidence into heart failure practice is needed.

Patients with worsening heart failure with reduced ejection fraction (HFrEF) spend a large proportion of time in the hospital and other health care facilities. The benefits of guideline-directed medical therapy (GDMT) in the outpatient setting have been shown in large randomized controlled trials. However, the decision to initiate, continue, switch, or withdraw HFrEF medications in the inpatient setting is often based on multiple factors and subject to significant variability across providers. Based on available data, in well-selected, treatment-naïve patients who are hemodynamically stable and clinically euvolemic after stabilization during hospitalization for HF, elements of GDMT can be safely initiated. Inpatient continuation of GDMT for HFrEF appears safe and well-tolerated in most hemodynamically stable patients. Hospitalization is also a potential time for switching from an angiotensin-converting enzyme inhibitor/angiotensin II receptor blocker to sacubitril/valsartan therapy in eligible patients, and is the subject of ongoing study. Therapy withdrawal or need for dose reduction is rarely required, but if needed identifies a particularly at-risk group of patients with progressive HF. If recurrent intolerance to neurohormonal blockers is observed, these patients should be evaluated for advanced HF therapies. There is an enduring need for using the teachable moment of HFrEF hospitalization for optimal initiation, continuation, and switching of GDMT to improve post-discharge patient outcomes and the quality of chronic HFrEF care.
