Georgia's rich biodiversity, as a key part of the Caucasus hotspot, makes it an ideal destination for scientific research. Nevertheless, knowledge about the species diversity of different animal groups in this area remains limited. In this article we provide information on the nine centipede species (Henia brevis (Silvestri, 1896), H. hirsuta Verhoeff, 1928, H. taurica (Sseliwanoff, 1884), Strigamia caucasia (Verhoeff, 1938), S. pusilla (Sseliwanoff, 1884), Harpolithobius spinipes Folkmanová, 1958, Lithobius antipai Matic, 1969, L. foviceps Muralevitch, 1926, and L. micropodus (Matic, 1980)) newly recorded in Georgia. Three of these species (H. brevis, H. hirsuta, and L. micropodus) are also new records for the Caucasus region. Data on their distribution, maps of the localities, and photos of the specimens studied are also given.

The purpose of the research is to investigate subacute cutaneous toxicity in rats, allergenic properties in guinea pigs and tolerance of increased doses of drugs based on imidacloprid, moxidectin and pyriproxyfen by dogs and cats of different age groups.Materials and methods. Toxicological studies (subacute cutaneous toxicity, study of allergenic properties) were conducted in the vivarium of the All-Russian Research Institute of Plant Protection, a branch of the Federal Scientific Center for Experimental Veterinary Medicine of the Russian Academy of Sciences, on male rats and guinea pigs. Experiments on the tolerance of drugs by target animal species (dogs, cats) were conducted in the Podolsk department of the All-Russian Research Institute of Plant Protection. The choice of doses, frequency and methods of administration of the studied drugs were determined in accordance with the Guidelines for Experimental (Preclinical) Study of New Pharmacological Substances. All doses were calculated for the drug taking into account the density (1.08 g/cm3).Results and discussion. When studying the acute cutaneous toxicity of the drug on rats, LD50 exceeded the maximum possible dose of 10 000 mg/kg (hazard class 4 according to GOST 12.1.007-76). In a subacute experiment on rats, it was established that doses of 1000 and 500 mg/kg are ineffective (safe), the threshold and toxic doses could not be established. These drugs, when treating animals 8 times with an interval of 3 days in a three- and five-fold increase in the maximum therapeutic dose during the experimental period, did not have a negative effect on the general condition of the animals, their physiological status and behavior; no statistically significant changes in the morphological and biochemical parameters of the blood, physicochemical parameters of urine were noted in animals from the experimental groups compared to the control. However, in one adult dog from the second experimental group, which was used a five-fold increase in the maximum therapeutic dose of the drug, depression was noted on the 20th day of the experiment. The physiological state of the animal returned to normal within 24 hours voluntarily without the use of symptomatic therapy.

The purpose of the research is to extend information across the distribution of zooanthroponotic helminth infections transmitted through freshwater fish in the Magadan Region.Materials and methods. The study area is located in the Upper Kolyma basin on the Magadan Region and the Republic of Sakha (Yakutia) border. The Omulevsky Park is also located here, one of the clusters of the Chersky National Park named after A. V. Andreev. The existing lake and river systems (Darpir, Momontai, Malyk, Urultun and Ui) are inhabited by 7 fish species: Arctic char (Salvelinus alpinus), east-Siberian grayling (Thymallus arcticus pallasi), round fish (Prosopium cylindraceum), Siberian sucker (Catostomus catostomus rostratus), burbot (Lota lota leptura), common minnow (Phoxinus phoxinus) and sculpin from the Kolyma (Cottus kolymensis). 685 specimens of fish were undergone parasitological survey using established procedures. The found helminths were fixed in 70% ethanol and clarified in glycerol.Results and discussion. In the study of the fish helminth fauna in the lake and river systems of the Upper Kolyma Highlands, three helminth species that posed a danger to humans and animals were identified, Dibothriocephalus dendriticus, D. ditremus (Diphyllobothriidae) and Eustrongylides excisus (Dioctophymatidae). Information was provided on fish infection with these parasites in the lake and river systems, and their distribution in the Magadan Region, and a brief description of these types of helminths was given. In domestic scientific and methodological publications, the problem of eustrongylidiasis as a zooanthroponotic disease is not sufficiently covered, therefore, emphasis is placed on nematodes of the genus Eustrongylides. The research results may attract the attention of sanitary and veterinary service specialists to this issue, especially given the creation of a national park in the Magadan Region. Data on the freshwater fish infection with Diphyllobothriidae in the Upper Kolyma Lake and river systems expand the diphyllobothriasis nosoarea in the Magadan Region and allows the Susumansky District to be included in its boundaries.

The purpose of the research is to study the parasite fauna in laboratory rats and mice in SPF-status and open-type vivariums and to compare the infection rate depending on the maintenance type within each type of samples.Materials and methods. The studies were conducted using life-time diagnosis (flotation method and Scotch-tape test), and the material were fecal samples from laboratory rodents and litter samples. Ten percent of the vivarium population and all newly arrived mice and rats in quarantine were examined. Fifty-four individual samples from pure line mice, and 24 litter samples were examined. A total of 234 samples were collected from the laboratory rats: 93 individual samples, 55 combined samples, and 17 sawdust samples; 69 samples were examined by Scotch-tape test.Results and discussion. Twenty six percent of the laboratory SPF vivarium mice were found to have protozoa: Giardia muris (11.1%) and Trichomonas sp. (20.4%). Nematodes Aspiculuris tetraptera (51.0%) and Syphacia obvelata (20.6%), and cestode Rodentolepis nana (12.0%) were recorded in the open-type vivarium mice. The laboratory open-type vivarium rats were found to have nematodes S. muris (up to 60.9%), A. tetraptera (5.4%), Trichosomoides crassicauda (1.8%), cestode R. nana (27.3%), protozoa Eimeria sp. (7.2%) and Giardia sp. (9.0%). The compared infection of individual with combined mouse fecal samples showed statistically significant differences for all parasites in general and for individual species. Pairwise comparisons of the infection showed that the combined sample detected R. nana statistically significantly more often versus the individual sample (27.3 vs. 5.4%, P < 0.001). The comparison of the infection depending on the type of rat maintenance within each sample type did not show statistically significant differences.

The purpose of the research is to determine the physiological characteristics of the single-host tick Rhipicephalus (Boophilus) annulatus depending on environmental factors, including the season of the year.Materials and methods. The study of the gonadotrophic cycle and fertility of B. annulatus was carried out during their seasonal activity from March 2022 to November 2023. The work consisted of two stages: field and laboratory. Ticks were collected manually from cattle in different settlements. Material was collected in natural biotopes according to the generally accepted method using a standard flag and drag. Nourished females were selected for oviposition and kept in laboratory conditions while maintaining optimal temperature and humidity. A total of 1,743 specimens were collected and identified. The sex ratio was 1: 7 with a predominance of females.Results and discussion. The gonadotrophic cycle and reproductive potential of B. annulatus vary by season. In autumn, the oviposition period, taking into account the incubation period, extends to an average of 28 days. The reproductive potential of females varied from 23 to 2194 eggs/ind. In spring, the cycle duration is comparatively shorter and, on average, does not exceed 25 days. Tick fertility can range from 19 to 5503 eggs/ind. which determines their abundance in summer and autumn months. The data obtained confirm the need for acaricidal treatment of animal grazing areas in the foothill zone in late autumn in order to reduce the number of B. annulatus both after winter diapause and throughout the year.

The purpose of the research is to study the efficacy of a macrocyclic lactone drug using the spot-on method against passalurosis and psoroptic mange in rabbits.Materials and methods. The study was conducted at the Department of Parasitology and Veterinary-Sanitary Expertise of the MVA named after K. I. Skryabin in 2022–2023 to determine the efficacy of antiparasitic drops (active ingredients: phenylpyrazole derivative, pyrazinoisoquinoline derivative, macrocyclic lactone, insect growth regulator, manufactured by AVZ S-P, LLC) against passalurosis and psoroptic mange. The antiparasitic drops were applied by the spot-on method to rabbits of different age and sex groups in doses of 0.5 mL per animal for body weight of up to 4 kg and 1 mL per animal for body weight of 4 to 8 kg against passalurosis and psoroptic mange. Rabbit fecal samples were examined using the Fülleborn method. Skin scrapings from the inner auricle surface were studied for Psoroptes cuniculi mites as per D. A. Priselkova.Results and discussion. The antiparasitic drug was found to have 100 % efficacy against ticks Psoroptes cuniculi and nematodes Passalurus ambigus.

The purpose of the study therapeutic efficacy of combined anthelmintic drug on dogs and cats of different age groups naturally infected with cestodes and nematodes.Materials and methods. The study drug in the suspension form contains oxantel pamoate, pyrantel pamoate, praziquantel, and additives as active ingredients. The therapeutic efficacy of the drug was evaluated on 228 animals naturally infected with nematodes or cestodes in the Podolsk Experimental Production Base of the VNIIP – FSC VIEV. The animals were divided into experimental and control groups of 6 animals each. The experimental dogs and cats were administered the study drug while the control animals were not given the drug. Clinical examinations and laboratory tests of fecal samples were performed on days 10, 20 and 30 after the start of the experiment. The method of helminthoscopy was used to detect segments and helminthoovoscopy as per Fülleborn to detect helminth eggs/cocoons in animal fecal samples with their subsequent differentiation. The results were statistically processed by the Student method using Microsoft Excel 2016.Results and discussion. It was found that anthelmintic drug based on oxantel pamoate, pyrantel pamoate and praziquantel had high therapeutic efficacy against parasitism in dogs and cats of nematodes Toxocara spp., Toxascaris leonina, Trichuris vulpis, Uncinaria stenocephala, Ancylostoma spp. and cestodes Echinococcus spp. (except for cats), Mesocestoides spp., Taenia spp., Dipylidium caninum, and Diphyllobothrium latum. When the drug was used in the animals of different age groups, no side effects or complications were recorded.

The purpose of the research is to study the probiotic Subalin effects on body weight gain in broiler chickens with spontaneous eimeriosis.Materials and methods. On a poultry farm in the Moscow Region, the body weight gain dynamics was studied in 120 floormanaged broiler chickens aged 9 to 40 days spontaneously infected with eimeria after administration of different doses of the probiotic Subalin vs. control. The chickens selected for the experiment were divided into four similar groups of 30 birds each. Subalin was administered to the first group chickens at a dose of 1.5 mL per 1 L of drinking water, while the broilers of the second and third groups were administered Subalin at a dose of 2 and 2.5 mL per 1 L of water, respectively. Subalin doses were given to the chickens of groups 1–3 in three stages: on 11–12; 18–19 and 26–27 days of age for consecutive 48 hours. The broilers of group 4 did not receive the drug and served as a control. The chickens of all groups underwent clinical, hematological, and coprological examinations; they were kept in similar conditions on the floor, in isolation, and fed as per zootechnical standards. Control weighing of the chickens was done on days 10 and 40 of their growth.Results and discussion. The total weight gain of the group 1 chickens was 1985 g in the experiment; 2203 g and 2214 g in groups 2 and 3, respectively, and 1803 g in the control group. The productivity of the experimental chickens was 10.1; 22.2 and 22.8% higher vs. control. The Subalin optimal dose was 2 mL per 1 L of drinking water according to the above scheme. The tested doses of Subalin had positive effects on the number of enterobacteria in the chicken intestines. Blood tests showed no statistically significant changes in clinical or hematological parameters in the experimental and control chickens.

The purpose of the research is to apply molecular genetic research methods to identify the taxonomic affiliation of gastrointestinal parasitic sheep nematodes of the family Trichostrongylidae using nested PCR followed by the restriction fragment length polymorphism (RFLP) analysis.Materials and methods. Parasitic nematodes, L3 Strongylata larvae obtained from incubated fecal samples of sheep. The genomic DNA was isolated using a commercial kit for DNA extraction from micro-quantities of tissues (Synthol, Moscow) as per the manufacturer’s guidelines. For DNA amplification, a T-100 Bio-Rad thermal cycler and a commercial Eurogen Master Mix reagent kit were used. The PCR regime was performed according to the WAAVP guidelines, 2006. The restriction endonuclease Rsa I of amplified Trichostrongylidae fragments was performed according to guidelines of the enzyme manufacturer (Sibenzyme, Novosibirsk).Results and discussion. To determine the taxonomic affiliation of Strongylata larvae isolated after incubation of feces from sheep, molecular genetic studies were performed using nested PCR followed by the restriction fragment length polymorphism (RFLP) analysis. This method makes it possible to identify, with the least effort, the genotypes of three species of Strongylata Haemonchus contortus, Trichostrongylus colubriformis, and Teladorsagia circumcincta at the larval stage.

The purpose of the research is to evaluate the comparative efficacy of two veterinary drugs («Inspector ear drops» and «Rolf Club 3D ear drops») to treat dogs and cats for otodectic mange complicated by otitis media of bacterial etiology.Materials and methods. A total of 36 animals participated in the studies. The studies were carried out at the VNIIP – FSC VIEV. The diagnosis was confirmed comprehensively from epidemiological data, clinical signs and laboratory tests. Microscopy of scrapings taken from the inner surface of the ears was performed to detect Otodectes cynotis. Bacteriological and mycological examination of ear canal discharge was conducted according to established procedures. Ear canal otoscopic examination was performed using a veterinary otoscope. For therapeutic purposes, the experimental animals were treated with «Inspector ear drops» and «Rolf Club 3D ear drops» as per instructions for use.Results and discussion. The initial comprehensive clinical examination diagnosed otodectic mange in all animals as complicated by acute otitis media of bacterial etiology. On treatment day 8, the animals showed a positive effect of the studied drugs; clinical signs of the disease reduced significantly. In a repeated clinical examination on day 11, most animals showed clinical recovery that was confirmed by negative bacteriological and mycological studies. Acarological studies showed no mites on days 8 and 14-15. However, the course of treatment was extended to 14 days for two dogs, after which their complete clinical recovery was observed. After the end of the study, the animals from the control group were treated with «Rolf Club 3D ear drops». Thus, an assessment of the results revealed that the drugs for veterinary use, «Inspector ear drops» and «Rolf Club 3D ear drops» showed an identical therapeutic effect in dogs and cats against otodectic mange complicated by acute bacterial otitis media.

The purpose of the research is to study the anthelmintic efficacy of suspensions based on a combined solid dispersion of Fenbendazole and Niclosamide.Materials and methods. Mechanochemistry methods were used to prepare suspensions of Fenbendazole (FBZ) and Niclosamide (NSM) substances, in particular, liquid phase grinding of substances with additives (licorice extract, polyvinylpyrrolidone (PVP)) in water or in aqueous sodium carboxymethylcellulose (NaCMC) solution. The grinding was conducted in LE-101 roller mill at energy intensity of 1 g for 1 hour at a roll rotation speed of 60–70 rpm. The resulting suspension samples of combined FBZ and NSM dispersions were studied for cestodocidal activity on a laboratory model of hymenolepiosis of white mice that were infected at a dose of 200 infective Hymenolepis nana eggs per animal.Results and discussion. The 100% efficacy of a combined NaCMC-based drug suspension containing FBZ 13 mg and NSM 130 mg in 1 g of suspension was achieved at a dose of 20 mg/kg of NSM and 2 mg/kg of FBZ. The suspension based on licorice extract, NaCMC, and dioctyl sulfosuccinate sodium salt containing FBZ 18.6 mg and NSM 186 mg in 1 g of suspension at the same dose, i.e. 20 mg/kg of NSM and 2 mg/kg of FBZ showed 71.43% efficacy against H. nana. The suspension based on licorice extract and NaCMC containing FBZ 24.6 mg and NSM 246 mg in 1 g of suspension at the same dose showed a 62.0% effect against H. nana. The activity of the basic drug, the FBZ and NSM suspension was 28.58% at this dose without using mechano-chemical treatment.

The purpose of the research is to study the acute oral and dermal toxicity of a veterinary drug (fipronil, pyriproxyfen) in mice and rats.Materials and methods. The studies were performed according to the Methodical Guidelines of the State Pharmacological Committee in the vivarium of the VNIIP – FSC VIEV from February to April 2021. White outbred male mice and male rats were used as a test system to study toxicological characteristics of the study drug. The veterinary drug Insektal Shampoo is a topical solution that contains fipronil and pyriproxyfen as active ingredients. In studying the acute oral toxicity and acute dermal toxicity of the study drug on laboratory animals, established procedures were used.Results and discussion. The median lethal dose (LD50) was 12 500 (11,532÷13,468) mg/kg when administered to the white male mice intragastrically as per Miller-Tainter method. According to the generally accepted hygiene classification, the drug was classified as hazard class 4 according to GOST 12.1.007-76. For the white male rats, LD50 was more than 20,800 mg/ kg (hazard class 4). When applied to intact rat skin, LD50 was more than 10 400 mg/kg, which corresponds to hazard class 4.

The purpose of the research is to evaluate some toxicological parameters of antiparasitic selamectin-based drug.Materials and methods. The study drug is a solution for external use, 1 mL of which contains the active component selamectin (120 mg), as well as additives. The experiments used a total of 66 white outbred male rats, 42 white outbred male mice and 3 guinea pigs. The parameters of acute oral toxicity (in mice and rats), acute dermal toxicity (in rats), and dermal and mucous membrane irritation (in rats and guinea pigs) of antiparasitic selamectin-based drug were evaluated. LD50 was calculated and the irritation of the drug was evaluated using established procedures in toxicology.Results and discussion. The maximum tolerated dose of the selamectin-based drug was 2550 mg/kg in the mice and 4250 mg/kg in the rats. The absolutely lethal dose for the mice and the rats was 6800 and 8500 mg/kg, respectively. It was established that the drug belonged to hazard class 3 (moderately hazardous substances) when administered intragastrically to the animals: LD50 for the mice was 4816.7 mg/kg, and LD50 for the rats was 6091.7 mg/kg. In determining acute dermal toxicity in the rats, the drug was classified as hazard class 4 (low-hazardous substances): LD50 more than 8500 mg/kg. The drug irritant effect on the skin of the rats at doses of 4250, 6375 and 8500 mg/kg was not established. The drug had a mild effect on the mucous membrane of the eyes of the guinea pigs with its recovery to normal within 24 hours.

The purpose of the research is to study the effects of ivermectin-containing drug in recommended and increased doses on adult dogs and cats.Materials and methods. The drug tolerance on dogs and cats was studied on 24 clinically healthy animals that were divided into experimental groups of 6 animals each, according to the principle of analogs, at the Podolsk Experimental Production Base of the VNIIP – FSC VIEV. The drug was administered seven times with a 3-day interval at a therapeutic dose of 0.2 mg of ivermectin per 1 kg of body weight to the first experimental group; and 0.6 mg of ivermectin per 1 kg of body weight to the second experimental group. The overall health status of the dogs and the cats was monitored daily, and blood samples were taken on days 20 and 34 of the experiment to study some morphological and biochemical parameters. Statistical data was processed in Microsoft Excel 2016 using the paired Student's t-test for dependent samples.Results and discussion. The studied ivermectin-containing drug had no negative effects on the adult dogs and cats’ overall health status, physiological status and behavior, or morphological and biochemical blood parameters. When using the drug in a threefold increased dose, three dogs and one cat showed depression which spontaneously disappeared within 48 hours. In addition, the animals from the second experimental group developed an allergic reaction, decreased functional activity of the liver and kidneys, and metabolic processes intensity, and red blood system destabilization. On day 34 of the experiment, the above changes were absent in the dogs and the cats, which indicates the restored physiological and biochemical status in the experimental animals.

São Tomé and Príncipe oceanic islands, in Central West Africa, are characterised by exceptional levels of endemism. Since human colonisation in the mid-15th century, São Tomé and Príncipe have lost 74% and 67% of their native habitat, respectively. Today, these islands are mainly covered by remaining old-growth forests, secondary regrowth forests, shaded plantations (mostly of cocoa), oil palm plantations (in the case of São Tomé), small-scale agricultural areas and urban areas. Yet, little is known about how species on these islands are coping with land-use changes. Island ecosystems are particularly important for bats, with about 25% of the world’s bat species being entirely restricted to island systems. São Tomé and Príncipe Islands comprise six and four native insectivorous bats, respectively. Two species, Chaerephon tomensis and Macronycteris thomensis, are island-endemics in São Tomé; Pseudoromicia principis is an island-endemic in Príncipe; and Miniopterus newtoni is endemic from both São Tomé and Príncipe. Here, we present a dataset comprising a comprehensive compilation of occurrence records derived from acoustic sampling of insectivorous bats across the predominant land-use types of both the São Tomé and Príncipe Islands. In each sampling site, standardised surveys consisted of deploying one Audio Moth device that recorded for 1 minute every 5 minutes over a 48-hour period. We identified a total of 19,437 bat-passes across the 115 sites surveyed in São Tomé Island and 17,837 bat-passes across the 50 sites surveyed in Príncipe Island.Based on a sampling effort of 1,584 hours of recordings manually processed to identify all the contained bat passes, this dataset, publicly available on GBIF, provides comprehensive information on the activity of insectivorous bats across two endemic-rich oceanic islands in the Gulf of Guinea. For each bat pass identified, we report the identified species, geographic coordinates, land-use type, altitude, date and time. This is the first public dataset providing detailed information on species-level habitat use for insectivorous bats on oceanic islands in Africa.

The cosmopolitan terrestrial slug, Deroceras laeve (O. F. Müller, 1774), is reported in the Philippines for the first time and characterized through morphology, morphometrics, and cytochrome oxidase subunit I (COI) gene analysis. Slug samples were recovered from two administrative regions in Mindanao, Philippines. In Region X, there were two sites: Misamis Oriental (Gingoog, 664 m a.s.l.; Claveria, 937 m a.s.l.) with farms planted with cabbage (Brassica oleracea), radish (Raphanus sativus), and eggplant (Solanum melongena); and Bukidnon (Talakag, 1410 m a.s.l.) planted with cabbage. In Region XI, specimens were collected from potted ornamentals in five nurseries along the Kapatagan road, Davao del Sur, 1000-1200 m a.s.l. The external morphology of the specimens matched the published descriptions, and their identity was further confirmed by their partial COI sequences. The obtained COI sequence of the specimen in Region X showed 99-100% similarity with the voucher specimens from Mexico (KX959495, KX959496, KX959497, KX959498, and KX495499); while that of the specimen from Region XI is 100% identical to specimens collected from Japan (MW507142), Canada (MT680918 and MT941436), UK (KF894311), and Vietnam (MT941435 and MT941436). Moreover, D. laeve from Region X and Region XI shared 98% similarity with each other. Preliminary surveys show that slug occurrence is prevalent mainly in highland regions of the southern Philippines where specialty crops/high value crops like vegetables and ornamentals are cultivated. Further surveys are essential to confirm any damage that they may cause, their distribution, associated parasites, and pest status in the Philippines.

Systematic studies on the biodiversity of bryophytes along elevational gradients have been conductuted within the native vegetation of the Azores, using the MOVECLIM framework. The primary objective of this study was to inventory the bryophytes present within preserved areas of native vegetation in Terceira Island (Azores). From 25 to 28 September 2012, an inventory of the bryoflora was carried out along an elevational gradient, starting near Serreta lighthouse (38.76658 Latitude; -27.37539 Longitude; 40 m a.s.l.) and culminating on the top of Santa Bárbara Mountain (38.73064 Latitude; -27.32164 Longitude; 1000 m a.s.l.). The study followed the adapted MOVECLIM standardised protocol, as follows: i) six sites were selected along an elevational transect, each site spaced at 200 m elevation intervals; ii) within each site, two 10 m x 10 m plots were established in close proximity from each other (10-15 m); iii) within these plots, three 2 m x 2 m quadrats were randomly selected and sampled for bryophytes. The following substrates were surveyed in each quadrat: rock, soil, humus, organic matter, tree bark at three different heights and leaves/fronds. For each available and bryophyte-colonised substrate, three replicate microplots of 10 cm x 5 cm were collected, resulting in a maximum of 24 microplots per quadrat.Nearly three-quarters of the maximum expected number of microplots (636 out of 864; eventID) were found across the six sites on Terceira Island, resulting in a total of 3677 records (occurrenceID). A high proportion of the specimens could be identified to the species rank (n = 3661; 99.6%), representing 38 families, 60 genera and 92 species, including 58 species of liverworts (Marchantiophyta) and 34 species of mosses (Bryophyta). The inventory included several endemic species: two liverwort species endemic to the Azores, five species endemic to Macaronesia (three mosses and two liverworts) and 11 European endemic species (three mosses and eight liverworts). The elevations with the highest species richness, the highest number of endemic species and the highest number of conservation concern species, spanned between 600 and 1000 m a.s.l. above sea level, coinciding with the best preserved forest vegetation. Overall, tree-dwelling and ground-dwelling substrates showed similar levels of bryophyte occupation (75% vs. 72%). However, the 636 events were unevenly distributed across substrates: leaves and rocks had the fewest replicates (n = 54; 50.0%), while humus and the lowest tree height had the highest values (n = 106; 98.1% and n = 98; 90.7%, respectively).The study contributed to expanding knowledge about the diversity and distribution of the Azorean Bryoflora, both on a local and a regional scale.

The south of Benin, a country in West Africa, is still home to remnants of dense forests that benefit from a particularly rainy sub-equatorial climate, with annual rainfall of up to 1,200 mm. These forest ecosystems are an integral part of the West African forest block, which stretches from Liberia to Togo. However, despite their richness and ecological importance, these forests are unfortunately subject to strong human pressures, particularly from slash-and-burn agriculture, intensive logging and the growing urbanisation of coastal areas. Preserving these forests is crucial, however, as they are home to remarkable plant and animal biodiversity, with many endemic species. What's more, these forests play an essential role in regulating the local climate, protecting soil and water resources, as well as providing local populations with a vital source of energy wood, non-timber forest products and support for their traditional agricultural practices. Faced with these conservation challenges, identifying and characterising the main tree species found in the forests of southern and central Benin, forest species and their ecology is an essential prerequisite for implementing sustainable management and restoration strategies for these threatened forest ecosystems in southern Benin. This work aims to identify and draw attention to the different forest species, specially tree forest species present in southern and central Benin.The dataset provides information on forest species found in southern and central Benin, West Africa. This dataset is extremely useful for forestry research, as it focuses mainly on the various forest species of major importance. It can be used as a basis for characterising individuals or populations of species, based on their abundance in relation to anthropogenic pressure and changes in environmental conditions.These species are characteristic of forests and, above all, are of particular interest both to populations and to managers of protected areas. Field collections were initiated in 2007 as part of natural forest inventory work. The data collected have been completed by various field works that followed this work on forest species in southern Benin. The latest version of the dataset is publicly and freely accessible on the GBIF website at the address https://www.gbif.org/dataset/aff3a10a-a86b-4eff-98e4-d63f92fd6f7e.It should be noted that the fact that the collection and monitoring were carried out in southern Benin, a region known for its great diversity of species, over a period of 10 years making these data particularly relevant information to study the effects of climate change and human pressure on ecosystems in this area.

The continent of Antarctica has remained relatively free of the impacts of invasive species to date. However, Antarctica is under increasing anthropogenic pressure from human activity and climate change, elevating the risk of alien species introductions. Scientific research and the maintenance of research stations by Antarctic Treaty Parties requires the transfer of large amounts of equipment and cargo, which can harbour biosecurity risk material. Here, we assess two decades of data collected by the Australian Antarctic Division on the detection of biosecurity risk material in its facilities and vessels, both during transport and in Antarctica. We use these data to identify emerging risk species or pathways, to compare the variability in detections over time and to construct a consequence table to facilitate effective responses and resource allocation to future detections, translating our research findings into guidance for decision-makers. We find that, despite the development of policy instruments, monitoring and management for the prevention of alien species introductions to Antarctica, the risk of introductions is ongoing. We highlight areas of concern, including the transport of live spiders and the continuing potential for cargo to harbour biosecurity risk material and the benefit of ongoing training and investment and support for staff and expeditioners in the reporting of non-native species detections. Finally, we provide tools and recommendations for decision-makers and on-ground managers in the Antarctic biosecurity space, based on our research. Future studies on the establishment risk of commonly transported species would assist in improving these tools.

Abstract Ischemic heart disease is the most prevalent cause of death worldwide affecting both the gender of all age groups. The high mortality rate is due to damage of myocardial tissue that emanates at the time of myocardial ischemia and re-oxygenation, thus averting reperfusion injury is recognized as a potential way to reduce acute cardiac injury and subsequent mortality. Flavonoids are polyphenol derivatives of plant origin and empirical shreds of evidence substantiate their numerous activities such as antioxidant, anti-inflammatory, anti-apoptotic, and anti-thrombotic activity, leading to their role in cardio protection. Recent investigations have unveiled the capacity of flavonoids to impede pivotal regulatory enzymes, signaling molecules, and transcription factors that orchestrate the mediators participating in the inflammatory cascade. The present comprehensive review, dwells on the preclinical studies on the effectiveness of flavonoids from the year 2007 to 2023, for the prevention and therapeutics for myocardial ischemia-reperfusion injury. Abstract Objective Alternative polyadenylation (APA) is a co-transcriptional process that leads to isoform diversity in the 3’ ends of mRNAs. APA is known to occur during differentiation, and its dysregulation is observed in diseases like cancer and autoimmune disorders. It has been previously reported that differentiation of 3T3-L1 cells to adipocytes leads to an overall lengthening of mRNAs, but the proteins involved in this regulation have not been identified. The expression levels of subunits of the cleavage and polyadenylation (C/P) complex can regulate the choice of poly(A) site, which in turn can affect different cellular activities. In this paper, we studied the change in levels of C/P proteins during 3T3-L1 differentiation. Results We observed that while the RNA expression of these proteins is unchanged during differentiation, the protein levels of some subunits do change, including a decrease in levels of CPSF73, the nuclease that cuts at the poly(A) site. However, overexpression of CPSF73 alone does not affect the efficiency and rate of differentiation.

Abstract Background In Nigeria, since 2002, Imatinib mesylate (glivec®) has been available freely to chronic myeloid leukaemia (CML) patients but only at a tertiary health care centre in the southwestern part of the country. Despite this, it is not readily accessible to many patients due to the distance and other challenges including low socioeconomic status and political problems, preventing timely access to specialist care. Abstract Objective Sago palm (Metroxylon sagu Rottb.) is one of the most important economic crops abundantly found in Mukah, Sarawak, Malaysia. The robustness of the palm triggered the Sarawak government’s selection as one of the state’s commodity crops, with the opening of several sago palm plantations. However, stunted (non-trunking) palms were reported in several sago palm plantations despite attaining a maturity period of more than ten years after cultivation. Research targeting this problem has been conducted in various fields, yet information on molecular mechanisms is still scarce. This study aimed to determine the genes responsible for sago palm’s normal phenotype (trunking) by attaining leaf transcriptomes from samples of all trunking sago palms from different sago palm plantations. Data description The conventional CTAB method was employed in the present investigation to extract total RNA from leaf tissues. Transcriptome sequencing was conducted on the Illumina NovaSeq 6000 platform. Differential expression analysis was performed using the DESeq2 package. A total of 6,119 differentially expressed genes, comprising 4,384 downregulated and 1,735 upregulated genes, were expressed in all three sago palm datasets. The datasets provide insights into the commonly expressed genes among trunking sago palms.

Abstract Background and objectives Medical students experience different types of challenges during their MD program, which become more challenging when it comes to the internship, putting too much pressure on them. This study aimed to explore the challenges of medical interns of Shiraz University of Medical Sciences (SUMS), Shiraz, Iran. Methods In this qualitative research, which was conducted from September 2023 March 2024, medical interns of SUMS were selected using purposeful sampling method. The data were collected through deep and semi-structured interviews, and it was continued until data saturation was achieved. The data analysis was performed through thematic content analysis. Results A total of 13 participants (6 men and 7 women) with a mean ± SD age of 25 ± 1.6 years were interviewed. The challenges of medical interns consisted of 423 meaning units, 79 open codes, 14 subthemes, and 6 main themes; the main themes consisted of educational challenges, challenges following university regulations, health and behavioral issues, economic/employment-related issues, sociocultural and recreational challenges, and ethical issues. Conclusion The results of this study highlight the necessity for reforms in the medical internship phase aiming to tackle interns’ issues and establish a supportive and nurturing environment for future medical doctors.

Abstract The study aimed to investigate the effects of acute ingestion of saffron (SAF) on physiological (i.e., heart rate and blood lactate) and perceptual (i.e., ratings of perceived exertion [RPE] and feeling scale) measures in response to a repeated-sprint ability test (RSS) in healthy young males (N = 22; mean ± SD: age, 21.7 ± 1.24 yrs.). All participants completed two experimental trials with a one-week washout period using a double-blind, placebo-controlled, crossover design. In each session, the participants were randomly chosen to receive either a capsule of saffron (300 mg) (SAF session) or a capsule of lactose (PLB session) two hours before performing the RSS. No significant differences (p > 0.05) were found for heart rate, RPE, and feeling scale between the SAF or PLB sessions at pre- and post-RSS. There were no significant changes (p > 0.05) in peak time, total time, fatigue index, and blood lactate in either the SAF or PLB sessions. Acute SAF ingestion did not significantly improve RSS performance nor physiological and perceptual measures in active young males. Future trials should address the topic by using shortened/prolonged higher doses of SAF on biological, physical, physiological, and perceptual responses to acute and chronic exercise.

Abstract Objective Effective time management is crucial for the survival of all patients, particularly those with cardiovascular conditions. This is especially true in the context of pre-hospital emergency services, where prompt intervention can significantly impact outcomes. This study delves into the timeliness of emergency services and the subsequent outcomes for hospitalized cardiovascular patients in EMS center in Fasa University of Medical Sciences, southern Iran. Results A total of 4972 emergency calls related to cardiac diagnoses were received between 2020 and 2023. The transport time was significantly correlated with age, location of the mission, and type of mission. Of the total, 86 underwent angioplasty within the standard time of less than 90 min, of which 81 were discharged and 5 died. 51 patients underwent angioplasty after more than 90 min, of which 47 were discharged and 4 died. In addition, 124 of these patients experienced cardiopulmonary resuscitation, of which 63 were successful and 61 were unsuccessful.

Abstract Objectives The Ishikawa cell line is the most widely used model system for investigating implantation and endometrial cancer. Understanding the biology of this cell line is essential for developing effective interventional strategies. To gain a deeper understanding of its cellular protein profile, we extracted cellular proteins from Ishikawa cells and analyzed the peptides using mass spectrometry. Our goal was to create a proteomic resource specifically tailored for Ishikawa cells. This data set is of particular significance in the realm of targeted drug delivery. Liposomes are synthetic spherical vesicles composed of hydrophobic bilayer phospholipids and have received immense recognition as highly effective carriers for the delivery of pharmaceutical drugs and essential nutrients to the endometrium. Phosphatidylcholine and phosphatidylethanolamine are often combined to create functional liposomal systems. To discern any potential interfering effects originating from the liposome backbone, our investigation involved direct effects of phospholipid liposomes on endometrial epithelial cells. Data description The data set includes peptide spectra derived from the intracellular proteomes of Ishikawa endometrial cancer cell isolates and their phospholipid-treated counterparts. Representing a proteome-wide profile, this dataset aims to contribute to a broader understanding of the physiology of endometrial epithelial cells. Proteomic analysis identified key proteins involved in the intricate regulation of cellular metabolism, cell cycle progression, and signaling. Between-group analysis revealed no differentially expressed proteins after adjusting for multiple testing using the applied thresholds (p-value 1). Abstract Objectives Apples are one of the most frequently consumed fruits and are effective in preventing lifestyle-related and other diseases. However, few studies have been conducted to evaluate health benefits of processed apple products such as juice. In this study, we analyzed the health benefits of consuming apple juice, focusing on changes in the gut microbiota, which plays an important role in maintaining human health. Results Rats were fed apple juice ad libitum, and the relative abundances of various gut microbiota in fecal samples were analyzed. In addition, rats treated apple juice were fed with a high-fat diet, and body weight, plasma triglyceride, glucose, and cholesterol levels were measured. The relative abundance of Clostridium cluster XIV did not change with the treatment of apple juice, but the relative abundance of Clostridium cluster IV was significantly decreased. In contrast, the relative abundances of Lactobacillus and Bifidobacterium, which provide benefits to the human body, were significantly increased by 3-fold and 10-fold, respectively, with apple juice consumption. When apple juice-treated rats were fed a high-fat diet, the increase in body weight, liver fat, and blood lipid parameters were all suppressed compared to high-fat alone group. Conculusion This study suggests that the consumption of apple juice changes the gut microbiota, exerts a prebiotic effect, and is effective in improving lifestyle-related diseases.

Abstract Background and purpose The relationship between dietary patterns and cirrhosis is undeniable. The present study aimed to investigate the association between the Dietary Approaches to Stop Hypertension (DASH) diet and the risk of mortality in patients with cirrhosis prospectively. Methods In this cohort study, 121 cirrhotic patients were enrolled and followed up annually for four years. Nutritional status and dietary intakes were assessed initially, and the DASH score was calculated accordingly. Crude and multivariable-adjusted hazard ratios (HR) with 95% confidence intervals (CI) were estimated using Cox proportional hazard analyses. Results DASH components including fruits, vegetables, legumes, nuts and seeds, and low-fat dairy products were significantly associated with lower mortality risk in cirrhotic patients. Also, a higher DASH score was significantly associated with a reduction in the risk of mortality in patients with cirrhosis, so that after adjusting for all confounders, the risk of mortality in the upper tertile was 89% lower than the first tertile (HR = 0.11, 95% CI: 0.03–0.42, P trend

Abstract Objective The integration of artificial intelligence (AI) in healthcare education is inevitable. Understanding the proficiency of generative AI in different languages to answer complex questions is crucial for educational purposes. The study objective was to compare the performance ChatGPT-4 and Gemini in answering Virology multiple-choice questions (MCQs) in English and Arabic, while assessing the quality of the generated content. Both AI models’ responses to 40 Virology MCQs were assessed for correctness and quality based on the CLEAR tool designed for evaluation of AI-generated content. The MCQs were classified into lower and higher cognitive categories based on the revised Bloom’s taxonomy. The study design considered the METRICS checklist for the design and reporting of generative AI-based studies in healthcare. Results ChatGPT-4 and Gemini performed better in English compared to Arabic, with ChatGPT-4 consistently surpassing Gemini in correctness and CLEAR scores. ChatGPT-4 led Gemini with 80% vs. 62.5% correctness in English compared to 65% vs. 55% in Arabic. For both AI models, superior performance in lower cognitive domains was reported. Both ChatGPT-4 and Gemini exhibited potential in educational applications; nevertheless, their performance varied across languages highlighting the importance of continued development to ensure the effective AI integration in healthcare education globally.

Abstract Objective Cardiovascular diseases (CVDs) are the most common cause of death worldwide. Diet plays an important role among many risk factors for CVDs. The present study aimed to investigate the relationship between carbohydrate quality index (CQI) and conventional risk factors of CVDs in Iranian adults. Results A higher CQI was related to a higher intake of energy, fiber, whole grains, fruits, vegetables, nuts, legumes, and dairy products. Additionally, a significant negative association was observed between CQI and triglycerides (TG) (odds ratio (OR) = 0.85; 95% confidence interval (CI): 0.73–0.98, highest versus the lowest tertile, p for trend = 0.026) and non-high density lipoprotein cholesterol (non-HDL-C) (OR = 0.85; 95% CI: 0.75–0.96, highest versus the lowest tertile, p for trend = 0.012). No significant correlation was shown between CQI and other cardiovascular risk factors. The findings indicate that the CQI is inversely associated with TG and non-HDL-C. Further studies are proposed to confirm these findings.

Abstract Objective This study validates a direct multiplex real-time reverse transcription polymerase chain reaction (rRT-PCR) assay which was previously established for enabling rapid and simultaneous detection of African swine fever (ASF) virus (ASFV) and classical swine fever virus. The assay eliminates the need for viral nucleic acid purification using a buffer system for crude extraction and an impurity-tolerant enzyme. However, the assay had not yet been validated using field samples of ASFV-infected pigs. Therefore, to address this gap, we tested 101 samples collected from pigs in Vietnam during 2018 and 2021 for validation. Results The rRT-PCR assay demonstrated a diagnostic sensitivity of 98.8% and a specificity of 100%. Remarkably, crude samples yielded results comparable to those of purified samples, indicating the feasibility of using crude samples without compromising accuracy in ASFV detection. Our findings emphasize the effectiveness of the rRT-PCR assay for the prompt and accurate diagnosis of both swine fever viruses, which is essential for effective disease prevention and control in swine populations.

Abstract Background Tumour dormancy, a resistance mechanism employed by cancer cells, is a significant challenge in cancer treatment, contributing to minimal residual disease (MRD) and potential relapse. Despite its clinical importance, the mechanisms underlying tumour dormancy and MRD remain unclear. In this study, we employed two syngeneic murine models of myeloid leukemia and melanoma to investigate the genetic, epigenetic, transcriptomic and protein signatures associated with tumour dormancy. We used a multiomics approach to elucidate the molecular mechanisms driving MRD and identify potential therapeutic targets. Results We conducted an in-depth omics analysis encompassing whole-exome sequencing (WES), copy number variation (CNV) analysis, chromatin immunoprecipitation followed by sequencing (ChIP-seq), transcriptome and proteome investigations. WES analysis revealed a modest overlap of gene mutations between melanoma and leukemia dormancy models, with a significant number of mutated genes found exclusively in dormant cells. These exclusive genetic signatures suggest selective pressure during MRD, potentially conferring resistance to the microenvironment or therapies. CNV, histone marks and transcriptomic gene expression signatures combined with Gene Ontology (GO) enrichment analysis highlighted the potential functional roles of the mutated genes, providing insights into the pathways associated with MRD. In addition, we compared “murine MRD genes” profiles to the corresponding human disease through public datasets and highlighted common features according to disease progression. Proteomic analysis combined with multi-omics genetic investigations, revealed a dysregulated proteins signature in dormant cells with minimal genetic mechanism involvement. Pathway enrichment analysis revealed the metabolic, differentiation and cytoskeletal remodeling processes involved in MRD. Finally, we identified 11 common proteins differentially expressed in dormant cells from both pathologies. Conclusions Our study underscores the complexity of tumour dormancy, implicating both genetic and nongenetic factors. By comparing genomic, transcriptomic, proteomic, and epigenomic datasets, our study provides a comprehensive understanding of the molecular landscape of minimal residual disease. These results provide a robust foundation for forthcoming investigations and offer potential avenues for the advancement of targeted MRD therapies in leukemia and melanoma patients, emphasizing the importance of considering both genetic and nongenetic factors in treatment strategies. Abstract Background Infertility is a growing global health concern affecting millions of couples worldwide. Among several factors, an extreme body weight adversely affects reproductive functions. Leptin is a well-known adipokine that serves as an endocrine signal between adiposity and fertility. However, the exact mechanisms underlying the effects of high leptin level on female reproduction remain unclear. Methods Transgenic pigs overexpressing leptin (♀) were produced by backcrossing and screened for leptin overexpression. The growth curve, fat deposition, reproductive performance, apoptosis, serum hormones and cholesterol production, RNA sequencing, and single-nucleus RNA sequencing (snRNA-seq) of the leptin-overexpressing pigs and wild-type group were evaluated. Results Transgenic pigs overexpressing leptin (♀) were obtained, which exhibited significantly reduced body weight, body size, and back fat thickness. These pigs manifested a late onset of puberty (330 ± 54.3 vs. 155 ± 14.7 days), irregular estrous behavior characterized by increased inter-estrous interval (29.2 ± 0 vs. 21.3 ± 0.7 days), and more number of matings until pregnancy (at least 3 times). This reproductive impairment in leptin pigs was related to hormonal imbalances characterized by increased levels of FSH, LH, prolactin, E2, P4, and TSH, altered steroidogenesis such as increased levels of serum cholesterol esters along with steroidogenic markers (StAR, CYP19A), and ovarian dysfunctions manifested by neutrophilic infiltration and low expression of caspase-3 positive cells in the ovaries. Moreover, bulk RNA sequencing of the ovaries also revealed neutrophilic infiltration followed by upregulation of inflammation-related genes. Furthermore, snRNA-seq reflected that leptin overexpression triggered immune response, suppressed follicle development and luteinization, resulting in metabolic dysfunction and hormone imbalance in the ovary. Conclusions Low body weight in leptin overexpressing pigs adversely affects the reproductive performance, causing delayed puberty, irregular estrous cycles, and reduced breeding efficiency. This is linked to metabolic imbalances, an increased immune response, and altered ovarian functions. This study provides a theoretical basis for the complex mechanisms underlying leptin, and infertility by employing leptin-overexpressing female pigs.

Abstract Sex hormones play a pivotal role as endocrine hormones that exert profound effects on the biological characteristics and vascular function of vascular smooth muscle cells (VSMCs). By modulating intracellular signaling pathways, activating nuclear receptors, and regulating gene expression, sex hormones intricately influence the morphology, function, and physiological state of VSMCs, thereby impacting the biological properties of vascular contraction, relaxation, and growth. Increasing evidence suggests that abnormal phenotypic changes in VSMCs contribute to the initiation of vascular diseases, including atherosclerosis. Therefore, understanding the factors governing phenotypic alterations in VSMCs and elucidating the underlying mechanisms can provide crucial insights for refining interventions targeted at vascular diseases. Additionally, the varying levels of different types of sex hormones in the human body, influenced by sex and age, may also affect the phenotypic conversion of VSMCs. This review aims to explore the influence of sex hormones on the phenotypic switching of VSMCs and the development of associated vascular diseases in the human body. Graphical Abstract

Abstract Chilean peach growers have achieved worldwide recognition for their high-quality fruit products. Among the main factors influencing peach fruit quality, sweetness is pivotal for maintaining the market's competitiveness. Numerous studies have been conducted in different peach-segregating populations to unravel SSC regulation. However, different cultivars may also have distinct genetic conformation, and other factors, such as environmental conditions, can significantly impact SSC. Using a transcriptomic approach with a gene co-expression network analysis, we aimed to identify the regulatory mechanism that controls the sugar accumulation process in an 'O × N' peach population. This population was previously studied through genomic analysis, associating LG5 with the genetic control of the SSC trait. The results obtained in this study allowed us to identify 91 differentially expressed genes located on chromosome 5 of the peach genome as putative new regulators of sugar accumulation in peach, together with a regulatory network that involves genes directly associated with sugar transport (PpSWEET15), cellulose biosynthesis (PpCSLG2), flavonoid biosynthesis (PpPAL1), pectin modifications (PpPG, PpPL and PpPMEi), expansins (PpEXPA1 and PpEXPA8) and several transcription factors (PpC3H67, PpHB7, PpRVE1 and PpCBF4) involved with the SSC phenotype. These results contribute to a better understanding of the genetic control of the SSC trait for future breeding programs in peaches.

Abstract Background Neisseria gonorrhoeae (Ng) causes the sexually transmitted disease gonorrhoea. There are no vaccines and infections are treated principally with antibiotics. However, gonococci rapidly develop resistance to every antibiotic class used and there is a need for developing new antimicrobial treatments. In this study we focused on two gonococcal enzymes as potential antimicrobial targets, namely the serine protease L,D-carboxypeptidase LdcA (NgO1274/NEIS1546) and the lytic transglycosylase LtgD (NgO0626/NEIS1212). To identify compounds that could interact with these enzymes as potential antimicrobials, we used the AtomNet virtual high-throughput screening technology. We then did a computational modelling study to examine the interactions of the most bioactive compounds with their target enzymes. The identified compounds were tested against gonococci to determine minimum inhibitory and bactericidal concentrations (MIC/MBC), specificity, and compound toxicity in vitro. Results AtomNet identified 74 compounds that could potentially interact with Ng-LdcA and 84 compounds that could potentially interact with Ng-LtgD. Abstract The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infects a variety of animal species. Susceptibility to SARS-CoV-2 is primarily determined by the utilization of the viral receptor, ACE2. SARS-CoV-2 can utilize a broad range of animal ACE2 isoforms in vitro, including the ACE2 from various camelid species. However, experimental infection of these animals does not lead to productive infection or seroconversion. In this study, we investigate the susceptibility of camelids to SARS-CoV-2 using novel well-differentiated camelid nasal organoids. We show that camelid nasal organoids are highly susceptible to Middle East respiratory syndrome coronavirus (MERS-CoV) infection, but not to infection with different SARS-CoV-2 variants (614G, BA.1 or EG.5.1.1). All viruses efficiently infected human airway organoids. Immunohistochemistry analysis revealed the absence of ACE2 on camelid nasal organoids and dromedary camel upper respiratory tract. In contrast, DPP4 was expressed in both camelid nasal organoids and the camel upper respiratory tract, which correlates with MERS-CoV infection. This study indicates that the camelid upper respiratory tract lacks expression of ACE2, which is associated with resistance to SARS-CoV-2 infection.

Abstract Nucleocapsid protein (N), or nucleoprotein (NP) coats the genome of most RNA viruses, protecting and shielding RNA from cytosolic RNAases and innate immune sensors, and plays a key role in virion biogenesis and viral RNA transcription. Often one of the most highly expressed viral gene products, N induces strong antibody (Ab) and T cell responses. N from different viruses is present on the infected cell surface in copy numbers ranging from tens of thousands to millions per cell, and it can be released to bind to uninfected cells. Surface N is targeted by Abs, which can contribute to viral clearance via Fc-mediated cellular cytotoxicity. Surface N can modulate host immunity by sequestering chemokines (CHKs), extending prior findings that surface N interferes with innate and adaptive immunity. In this review, we consider aspects of surface N cell biology and immunology and describe its potential as a target for anti-viral intervention.

Abstract Background GALNTs (UDP-GalNAc; polypeptide N-acetylgalactosaminyltransferases) initiate mucin-type O-GalNAc glycosylation by adding N-GalNAc to protein serine/threonine residues. Abnormalities in O-GalNAc glycosylation are involved in various disorders such as Parkinson’s disease (PD), a neurodegenerative disorder. GALNT9 is potentially downregulated in PD patients. Methods To determine whether GALNT9 enrichment ameliorates cytotoxicity related to PD-like variations, a pcDNA3.1-GALNT9 plasmid was constructed and transfected into SH-SY5Y cells to establish a GALNT9-overexpressing cell model. Results Downregulation of GALNT9 and O-GalNAc glycosylation was confirmed in our animal and cellular models of PD-like variations. GALNT9 supplementation greatly attenuated cytotoxicity induced by MPP+ (1-Methyl-4-phenylpyridinium iodide) since it led to increased levels of tyrosine hydroxylase and dopamine, reduced rates of apoptosis, and significantly ameliorated MPP+-induced mitochondrial dysfunction by alleviating abnormal levels of mitochondrial membrane potential and reactive oxygen species. A long-lasting mPTP (mitochondrial permeability transition pores) opening and calcium efflux resulted in significantly lower activity in the cytochrome C-associated apoptotic pathway and mitophagy process, signifying that GALNT9 supplementation maintained neuronal cell health under MPP+ exposure. Additionally, it was found that glycans linked to proteins influenced the formation of protein aggregates containing α-synuclein, and GALNT9 supplement dramatically reduced such insoluble protein aggregations under MPP+ treatment. Glial GALNT9 predominantly appears under pathological conditions like PD-like variations. Conclusions GALNT9 enrichment improved cell survival, and glial GALNT9 potentially represents a pathogenic index for PD patients. This study provides insights into the development of therapeutic strategies for the treatment of PD.

Abstract Choriocarcinoma is a malignant cancer that belongs to gestational trophoblastic neoplasia (GTN). Herein, serum metabolomic analysis was performed on 29 GTN patients and 30 healthy individuals to characterize the metabolic variations during GTN progression. Ultimately 24 differential metabolites (DMs) were identified, of which, Equol was down-regulated in GTN patients, whose VIP score is the 3rd highest among the 24 DMs. As an intestinal metabolite of daidzein, the anticancer potential of Equol has been demonstrated in multiple cancers, but not choriocarcinoma. Hence, human choriocarcinoma cell lines JEG-3 and Bewo were used and JEG-3-derived subcutaneous xenograft models were developed to assess the effect of Equol on choriocarcinoma. The results suggested that Equol treatment effectively suppressed choriocarcinoma cell proliferation, induced cell apoptosis, and reduced tumorigenesis. Label-free quantitative proteomics showed that 136 proteins were significantly affected by Equol and 20 proteins were enriched in Gene Ontology terms linked to protein degradation. Tripartite motif containing 21 (TRIM21), a E3 ubiquitin ligase, was up-regulated by Equol. Equol-induced effects on choriocarcinoma cells could be reversed by TRIM21 inhibition. Annexin A2 (ANXA2) interacted with TRIM21 and its ubiquitination was modulated by TRIM21. We found that TRIM21 was responsible for proteasome-mediated degradation of ANXA2 induced by Equol, and the inhibitory effects of Equol on the malignant behaviors of choriocarcinoma cells were realized by TRIM21-mediated down-regulation of ANXA2. Moreover, β-catenin activation was inhibited by Equol, which also depended on TRIM21-mediated down-regulation of ANXA2. Taken together, Equol may be a novel candidate for the treatment for choriocarcinoma. Graphical abstract

Abstract Moyamoya disease, characterized by basal cerebral artery obstruction, was studied for differential protein expression to elucidate its pathogenesis. Proteomic analysis of cerebrospinal fluid from 10 patients, categorized by postoperative angiography into good and poor prognosis groups, revealed 46 differentially expressed proteins. Notably, cadherin 18 (CDH18) was the most significantly upregulated in the good prognosis group. In addition, the expression of cadherin 18 (CDH18) and phenotypic transformation-related proteins were measured by qRT-PCR and western blot. The effects of CDH18 in vascular smooth muscle cells were detected by CCK-8, EdU, transwell and wound healing assays. The overexpression of CDH18 in vascular smooth muscle cells (VSMCs) was found to inhibit proliferation, migration, and phenotypic transformation. These findings suggest CDH18 as a potential therapeutic target in moyamoya disease.

Abstract Intervertebral disc degeneration (IDD) is a major cause of discogenic pain, and is attributed to the dysfunction of nucleus pulposus, annulus fibrosus, and cartilaginous endplate (CEP). Osteopontin (OPN), a glycoprotein, is highly expressed in the CEP. However, little is known on how OPN regulates CEP homeostasis and degeneration, contributing to the pathogenesis of IDD. Here, we investigate the roles of OPN in CEP degeneration in a mouse IDD model induced by lumbar spine instability and its impact on the degeneration of endplate chondrocytes (EPCs) under pathological conditions. OPN is mainly expressed in the CEP and decreases with degeneration in mice and human patients with severe IDD. Conditional Spp1 knockout in EPCs of adult mice enhances age-related CEP degeneration and accelerates CEP remodeling during IDD. Mechanistically, OPN deficiency increases CCL2 and CCL5 production in EPCs to recruit macrophages and enhances the activation of NLRP3 inflammasome and NF-κB signaling by facilitating assembly of IRAK1-TRAF6 complex, deteriorating CEP degeneration in a spatiotemporal pattern. More importantly, pharmacological inhibition of the NF-κB/NLRP3 axis attenuates CEP degeneration in OPN-deficient IDD mice. Overall, this study highlights the importance of OPN in maintaining CEP and disc homeostasis, and proposes a promising therapeutic strategy for IDD by targeting the NF-κB/NLRP3 axis.

Abstract There are currently no targeted delivery systems to satisfactorily treat bone-related disorders. Many clinical drugs consisting of small organic molecules have a short circulation half-life and do not effectively reach the diseased tissue site. This coupled with repeatedly high dose usage that leads to severe side effects. With the advance in nanotechnology, drugs contained within a nano-delivery device or drugs aggregated into nanoparticles (nano-drugs) have shown promises in targeted drug delivery. The ability to design nanoparticles to target bone has attracted many researchers to develop new systems for treating bone related diseases and even repurposing current drug therapies. In this review, we shall summarise the latest progress in this area and present a perspective for future development in the field. We will focus on calcium-based nanoparticle systems that modulate calcium metabolism and consequently, the bone microenvironment to inhibit disease progression (including cancer). We shall also review the bone affinity drug family, bisphosphonates, as both a nano-drug and nano-delivery system for bone targeted therapy. The ability to target and release the drug in a controlled manner at the disease site represents a promising safe therapy to treat bone diseases in the future.

Abstract Osteoarthritis (OA) is the most common form of arthritic disease, and phenotypic modification of chondrocytes is an important mechanism that contributes to the loss of cartilage homeostasis. This study identified that Fascin actin-bundling protein 1 (FSCN1) plays a pivotal role in regulating chondrocytes phenotype and maintaining cartilage homeostasis. Proteome-wide screening revealed markedly upregulated FSCN1 protein expression in human OA cartilage. FSCN1 accumulation was confirmed in the superficial layer of OA cartilage from humans and mice, primarily in dedifferentiated-like chondrocytes, associated with enhanced actin stress fiber formation and upregulated type I and III collagens. FSCN1-inducible knockout mice exhibited delayed cartilage degeneration following experimental OA surgery. Mechanistically, FSCN1 promoted actin polymerization and disrupted the inhibition of Decorin on TGF-β1, leading to excessive TGF-β1 production and ALK1/Smad1/5 signaling activation, thus, accelerated chondrocyte dedifferentiation. Intra-articular injection of FSCN1-overexpressing adeno-associated virus exacerbated OA progression in mice, which was mitigated by an ALK1 inhibitor. Moreover, FSCN1 inhibitor NP-G2-044 effectively reduced extracellular matrix degradation in OA mice, cultured human OA chondrocytes, and cartilage explants by suppressing ALK1/Smad1/5 signaling. These findings suggest that targeting FSCN1 represents a promising therapeutic approach for OA.

Abstract Osteoporosis remains incurable. The most widely used antiresorptive agents, bisphosphonates (BPs), also inhibit bone formation, while the anabolic agent, teriparatide, does not inhibit bone resorption, and thus they have limited efficacy in preventing osteoporotic fractures and cause some side effects. Thus, there is an unmet need to develop dual antiresorptive and anabolic agents to prevent and treat osteoporosis. Hydroxychloroquine (HCQ), which is used to treat rheumatoid arthritis, prevents the lysosomal degradation of TNF receptor-associated factor 3 (TRAF3), an NF-κB adaptor protein that limits bone resorption and maintains bone formation. We attempted to covalently link HCQ to a hydroxyalklyl BP (HABP) with anticipated low antiresorptive activity, to target delivery of HCQ to bone to test if this targeting increases its efficacy to prevent TRAF3 degradation in the bone microenvironment and thus reduce bone resorption and increase bone formation, while reducing its systemic side effects. Unexpectedly, HABP-HCQ was found to exist as a salt in aqueous solution, composed of a protonated HCQ cation and a deprotonated HABP anion. Nevertheless, it inhibited osteoclastogenesis, stimulated osteoblast differentiation, and increased TRAF3 protein levels in vitro. HABP-HCQ significantly inhibited both osteoclast formation and bone marrow fibrosis in mice given multiple daily PTH injections. In contrast, HCQ inhibited marrow fibrosis, but not osteoclast formation, while the HABP alone inhibited osteoclast formation, but not fibrosis, in the mice. HABP-HCQ, but not HCQ, prevented trabecular bone loss following ovariectomy in mice and, importantly, increased bone volume in ovariectomized mice with established bone loss because HABP-HCQ increased bone formation and decreased bone resorption parameters simultaneously. In contrast, HCQ increased bone formation, but did not decrease bone resorption parameters, while HABP also restored the bone lost in ovariectomized mice, but it inhibited parameters of both bone resorption and formation. Our findings suggest that the combination of HABP and HCQ could have dual antiresorptive and anabolic effects to prevent and treat osteoporosis.

Abstract Background Dissection of complex plant cell wall structures demands a sensitive and quantitative method. FTIR is used regularly as a screening method to identify specific linkages in cell walls. However, quantification and assigning spectral bands to particular cell wall components is still a major challenge, specifically in crop species. In this study, we addressed these challenges using ATR-FTIR spectroscopy as it is a high throughput, cost-effective and non-destructive approach to understand the plant cell wall composition. This method was validated by analysing different varieties of mungbean which is one of the most important legume crops grown widely in Asia. Results Using standards and extraction of a specific component of cell wall components, we assigned 1050–1060 cm−1 and 1390–1420 cm−1 wavenumbers that can be widely used to quantify cellulose and lignin, respectively, in Arabidopsis, Populus, rice and mungbean. Also, using KBr as a diluent, we established a method that can relatively quantify the cellulose and lignin composition among different tissue types of the above species. We further used this method to quantify cellulose and lignin in field-grown mungbean genotypes. The ATR-FTIR-based study revealed the cellulose content variation ranges from 27.9% to 52.3%, and the lignin content variation ranges from 13.7% to 31.6% in mungbean genotypes. Conclusion Multivariate analysis of FT-IR data revealed differences in total cell wall (600–2000 cm−1), cellulose (1000–1100 cm−1) and lignin (1390–1420 cm−1) among leaf and stem of four plant species. Overall, our data suggested that ATR-FTIR can be used for the relative quantification of lignin and cellulose in different plant species. This method was successfully applied for rapid screening of cell wall composition in mungbean stem, and similarly, it can be used for screening other crops or tree species.

Abstract Background The proportion of nitrogen (N) derived from the atmosphere (Ndfa) is a fundamental component of the plant N demand in legume species. To estimate the N benefit of grain legumes for the subsequent crop in the rotation, a simplified N balance is frequently used. This balance is calculated as the difference between fixed N and removed N by grains. The Ndfa needed to achieve a neutral N balance (hereafter $$\theta$$ θ ) is usually estimated through a simple linear regression model between Ndfa and N balance. This quantity is routinely estimated without accounting for the uncertainty in the estimate, which is needed to perform formal statistical inference about $$\theta$$ θ . In this article, we utilized a global database to describe the development of a novel Bayesian framework to quantify the uncertainty of $$\theta$$ θ . This study aimed to (i) develop a Bayesian framework to quantify the uncertainty of $$\theta$$ θ , and (ii) contrast the use of this Bayesian framework with the widely used delta and bootstrapping methods under different data availability scenarios. Results The delta method, bootstrapping, and Bayesian inference provided nearly equivalent numerical values when the range of values for Ndfa was thoroughly explored during data collection (e.g., 6–91%), and the number of observations was relatively high (e.g., $$\ge 100$$ ≥ 100 ). When the Ndfa tested was narrow and/or sample size was small, the delta method and bootstrapping provided confidence intervals containing biologically non-meaningful values (i.e. 100%). However, under a narrow Ndfa range and small sample size, the developed Bayesian inference framework obtained biologically meaningful values in the uncertainty estimation. Conclusion In this study, we showed that the developed Bayesian framework was preferable under limited data conditions ─by using informative priors─ and when uncertainty estimation had to be constrained (regularized) to obtain meaningful inference. The presented Bayesian framework lays the foundation not only to conduct formal comparisons or hypothesis testing involving $$\theta$$ θ , but also to learn about its expected value, variance, and higher moments such as skewness and kurtosis under different agroecological and crop management conditions. This framework can also be transferred to estimate balances for other nutrients and/or field crops to gain knowledge on global crop nutrient balances.

Abstract Background Wheat (Triticum aestivum L.) is an important grain crops in the world, and its growth and development in different stages is seriously affected by saline-alkali stress, especially in seedling stage. Therefore, nondestructive detection of wheat seedlings under saline-alkali stress can provide more comprehensive technical support for wheat breeding, cultivation and management. Results This research focused on moisture signal prediction and classification of saline-alkali stress in wheat seedlings using fusion techniques. After collecting and analyzing transverse relaxation time and Multispectral imaging (MSI) information of wheat seedlings, four regression models were used to predict the moisture signal. K-Nearest Neighbor (KNN) and Gaussian-Naïve Bayes (GNB) models were combined with fivefold cross validation to classify the prediction of wheat seedling stress. The results showed that wheat seedlings would increase the bound water content through a certain mechanism to enhance their saline-alkali stress. Under the same Na concentration, the effect of alkali stress on moisture, growth and spectrum of wheat seedlings is stronger than salt stress. The Gradient Boosting Decision Regression Tree model performs the best in predicting wheat moisture signals, with a coefficient of determination (R2P) of 0.98 and a root mean square error of 109.60. It also had a short training time (1.48 s) and an efficient prediction speed (1300 obs/s). The KNN and GNB demonstrated significantly enhanced predictive performance when classifying the fused dataset, compared to using single datasets individually. In particular, the GNB model performing best on the fused dataset, with Precision, Recall, Accuracy, and F1-score of 90.30, 88.89%, 88.90%, and 0.90, respectively. Conclusions Under the same Na concentration, the effects of alkali stress on water content, spectrum, and growth of wheat were stronger than that of salt stress, which was more unfavorable to the growth of wheat. The fusion of low-field nuclear magnetic resonance and MSI technology can improve the classification of wheat stress, and provide an effective technical method for rapid and accurate monitoring of wheat seedlings under saline-alkali stress. Abstract The major drawback to the implementation of genomic selection in a breeding program lies in long-term decrease in additive genetic variance, which is a trade-off for rapid genetic improvement in short term. Balancing increase in genetic gain with retention of additive genetic variance necessitates careful optimization of this trade-off. In this study, we proposed an integrated index selection approach within the genomic inferred cross-selection (GCS) framework to maximize genetic gain across multiple traits. With this method, we identified optimal crosses that simultaneously maximize progeny performance and maintain genetic variance for multiple traits. Using a stochastic simulated recurrent breeding program over a 40-years period, we evaluated different GCS methods along with other factors, such as the number of parents, crosses, and progeny per cross, that influence genetic gain in a pulse crop breeding program. Across all breeding scenarios, the posterior mean variance consistently enhances genetic gain when compared to other methods, such as the usefulness criterion, optimal haploid value, mean genomic estimated breeding value, and mean index selection value of the superior parents. In addition, we provide a detailed strategy to optimize the number of parents, crosses, and progeny per cross that can potentially maximize short- and long-term genetic gain in a public breeding program.
