Abstract In our study, we employed an interdisciplinary approach to study the diverse parts of the Missale Nidrosiense, published in 1519. Our aim was a thorough investigation of the materials used and the manufacturing methods that may give indications on dating and provenance of the components of the book and where the book was bound. Initially, visual and multispectral methods were employed to investigate the books’ components, printing technology and bookbinding structure. Subsequently, other methods were applied: the composition of metallic components was determined by X-ray fluorescence spectroscopy (XRF). Pigments, printing inks and binders were characterised by using a combination of XRF and synchrotron-based infrared microscopy. Non-invasive dendrochronology based on X-ray tomography was utilised, to indicate date and provenance of the wooden boards of the book. Additionally, we used a biocodicological approach to identify the species of animal used in the parchment. This resulted in a complete biography of the book. We were able to acquire new information about the materials used and their provenance. This provides new information about craft, economy, trade and commercial exchange in the beginning of the sixteenth century in North-west Scandinavia, despite the lack of written documentation from this period.

Recent global events have underscored the need for broad access to digitized library special collections. At the same time, a burgeoning field of scientific and historical inquiry is finding a goldmine of data in the physical old books and manuscripts stored for centuries on library shelves. This article gives an overview of some of the interesting studies employing library materials in the new field of biocodicology, which expands the field of codicology (learning about book history through studying a copy's physical attributes, sometimes referred to as "archaeology of the book") to interrogate physical books with proteomic, genomic, and microbiomic tools. Historical health sciences collections provide rich, new research avenues for budding biocodicologists, and biocodicology and other interdisciplinary fields focused on material culture present an unforeseen justification for institutions' continued preservation and access to individual physical copies.

Microbiological and biomolecular approaches to cultural heritage research have expanded the established research horizon from the prevalent focus on the cultural objects’ conservation and human health protection to the relatively recent applications to provenance inquiry and assessment of environmental impacts in a global context of a changing climate. Standard microbiology and molecular biology methods developed for other materials, specimens, and contexts could, in principle, be applied to cultural heritage research. However, given certain characteristics common to several heritage objects—such as uniqueness, fragility, high value, and restricted access, tailored approaches are required. In addition, samples of heritage objects may yield low microbial biomass, rendering them highly susceptible to cross-contamination. Therefore, dedicated methodology addressing these limitations and operational hurdles is needed. Here, we review the main experimental challenges and propose a standardized workflow to study the microbiome of cultural heritage objects, illustrated by the exploration of bacterial taxa. The methodology was developed targeting the challenging side of the spectrum of cultural heritage objects, such as the delicate written record, while retaining flexibility to adapt and/or upscale it to heritage artifacts of a more robust constitution or larger dimensions. A new trend is molecular analysis, which has given rise to the emerging field of biocodicology, comprising protein and DNA analysis for the identification of the biological origin of the skins that are used for their manufacture. In addition, DNA analysis can identify the microbiome that is present in the object under investigation, which adds value by providing information on its history and state of preservation. In any case, it is important to complement the biomolecular investigations with microscopical and physicochemical analyses. Some of the complementary analytical techniques that are reviewed here, such as elemental analysis by X-ray fluorescence (XRF) with compound-specific analytical methods such as Fourier transform infrared (FTIR) and Raman spectroscopy are advantageous as they can be applied in a non-invasive way and without inducing any changes in the objects. Abstract Historic legal deeds are one of the most abundant resources in British archives, but also one of the most neglected. Despite the millions that survive, we know remarkably little about their manufacture, including the species of animal on which they were written. Here we present the species identification of 645 sixteenth–twentieth century skins via peptide mass fingerprinting (ZooMS), demonstrating the preferential use of sheepskin parchment. We argue that alongside their abundance and low cost, the use of sheepskins over those of other species was motivated by the increased visibility of fraudulent text erasure and modification afforded by the unique structure of their skin.

Non-Invasive Physico-Chemical and Biological Analysis of Parchment Manuscripts - An Overview. [Extracted from the article]

Abstract Biocodicology, the study of the biological information stored in manuscripts, offers the possibility of interrogating manuscripts in novel ways. Exploring the biological data associated to parchment documents will add a deeper level of understanding and interpretation to these invaluable objects, revealing information about book production, livestock economies, handling, conservation and the historic use of the object. As biotechnological methods continue to improve we hope that biocodicology will become a highly relevant discipline in manuscript studies, contributing an additional perspective to the current scholarship. We hope that this review will act as a catalyst enabling further interactions between the heritage science community, manuscript scholars, curators and conservators.

This interdisciplinary research on the Kiev folia, a 10th century Slavonic manuscript, shows how material and biological analyses as well as imaging methods can be applied to answer relevant questions that contribute to a better understanding of the history and conservation of ancient manuscripts. By applying non-invasive analytical methods, including X-ray Fluorescence analysis (XRF) and Fourier Transform Infrared spectroscopy in reflection mode (rFTIR), calcium oxalate related to the degradation phenomena of the ink material was detected, showing rFTIR bands at 1644 cm−1 and 1327 cm−1. Moreover, potassium ferrocyanide was identified on the peculiar blue stains visible on some folios. Multispectral images of these stains improved the legibility of the underlying letters in the near infrared range. Finally, shotgun metagenomic analyses gave insight into the possible animal origin of the skin(s) used for the manufacture of the codex and the microbiome colonizing its surface. The microbiome consisted mainly of viruses (53%) and bacteria (47%) represented by halotolerant/halophilic bacteria as well as bacteria and viruses typical of the human microbiome. In contrast, the proportion of eukaryotes (0.05%) and archaea (0.07%) was very low. This information served to determine the conservation status and the risk of deterioration of the manuscript. • This interdisciplinary approach answers scientific questions related to ancient manuscripts. • XRF and rFTIR show the application of minium, iron gall and carbon black inks. • The composition of the peculiar blue stains was identified as potassium ferrocyanide. • Shotgun metagenomic analysis gave insight into the possible animal origin of the skin. • The microbiome showed halotolerant and commensal bacteria of the human microbiome.
