Recent decades have seen growing attention on viruses in the environment and their potential impacts as a result of global epidemics. Due to the diversity of viral species along with the complexity of environmental and host factors, virus extraction and detection methods have become key for the study of virus ecology. This review systematically summarises the methods for extracting and detecting pathogens from different environmental samples (e.g., soil, water, faeces, air) and biological samples (e.g., plants, animals) in existing studies, comparing their similarities and differences, applicability, as well as the advantages and disadvantages of each method. Additionally, this review discusses future directions for research in this field. The aim is to provide a theoretical foundation and technical reference for virus ecology research, facilitating further exploration and applications in this field.

The phenomenon of glacial freezing and thawing involves microbial sequestration, release, and colonization, which has the potential to impact ecosystem functioning through changes in microbial diversity and interactions. In this study, we examined the structural features of microbial communities of the Dongkemadi glacier, including bacteria, fungi, and archaea, in four distinct glacial environments (snow, ice, meltwater, and frontier soil). The sequestration, release, and colonization of glacial microbes have been found to significantly impact the diversity and structure of glacial microbial communities, as well as the complexity of microbial networks. Specifically, the complexity of bacterial networks has been observed to increase in a sequential manner during these processes. Utilizing the Inter-Domain Ecological Network approach, researchers have further explored the cross-trophic interactions among bacteria, fungi, and archaea. The complexity of the bacteria-fungi-archaea network exhibited a sequential increase due to the processes of sequestration, release, and colonization of glacial microbes. The release and colonization of glacial microbes led to a shift in the role of archaea as key species within the network. Additionally, our findings suggest that the hierarchical interactions among various microorganisms contributed to the heightened complexity of the bacteria-fungi-archaea network. The primary constituents of the glacial microbial ecosystem are unclassified species associated with the Polaromonas. It is noteworthy that various key species in glacial ecosystems are influenced by the distinct environmental factors. Moreover, our findings suggest that key species are not significantly depleted in response to abrupt alterations in individual environmental factors, shedding light on the dynamics of microbial cross-trophic interactions within glacial ecosystems.

Salt stress significantly impairs plant growth, presenting a challenge to agricultural productivity. Exploring the regulatory mechanisms underlying salt stress responses is critically important. Here, we identified a significant role for the maize LESION-SIMULATING DISEASE transcription factor, ZmLSD1, in enhancing salt stress response. Subcellular localization analysis indicated that ZmLSD1-GFP was localized in the nucleus in the maize protoplast. Overexpressing ZmLSD1 in maize obviously enhanced the tolerance of plants to salt stress. Physiological analysis indicated that overexpressed ZmLSD1 in maize could mitigate the accumulation of H2O2 and MDA content exposed to salt stress. RNA-seq and qPCR-PCR analyses showed that ZmLSD1 positively regulated ZmWRKY29 expression. ChIP-qPCR and EMSA experiments demonstrated that ZmLSD1 could directly bind to the promoter of ZmWRKY29 through the GTAC motif both in vitro and in vivo. Overall, our findings suggest that ZmLSD1 plays a positive role in enhancing the tolerance of maize to salt by affecting ZmWRKY29 expression.

Radiation protection is an important field of study, as it relates to human health and environmental safety. Radiation-resistance mechanisms in extremophiles are a research hotspot, as this knowledge has great application value in bioremediation and development of anti-radiation drugs. Mount Everest, an extreme environment of high radiation exposure, harbors many bacterial strains resistant to radiation. However, owing to the difficulties in studying them because of the extreme terrain, many remain unexplored. In this study, a novel species (herein, S7-12T) was isolated from the moraine of Mount Everest, and its morphology and functional and genomic characteristics were analyzed. The strain S7-12T is white in color, smooth and rounded, non-spore-forming, and non-motile and can survive at a UV intensity of 1000 J/m2, showing that it is twice as resistant to radiation as Deinococcus radiodurans. Radiation-resistance genes, including IbpA and those from the rec and CspA gene families, were identified. The polyphasic taxonomic approach revealed that the strain S7-12T (=KCTC 59114T =GDMCC 1.3458T) is a new species of the genus Knoellia and is thus proposed to be named glaciei. The in-depth study of the genome of strain S7-12T will enable us to gain further insights into its potential use in radiation resistance. Understanding how microorganisms resist radiation damage could reveal potential biomarkers and therapeutic targets, leading to the discovery of potent anti-radiation compounds, thereby improving human resistance to the threat of radiation.

Based on the nonlinear coupled wave equations, the nonreciprocal response of the third-harmonic generation process has been theoretically investigated in nonlinear photonic crystals with a defect embedded. The simulation results have shown that complete nonreciprocity for third-harmonic generation can be achieved under specific conditions, and the arbitrary nonreciprocal response can be realized by adjusting the structure parameters, including the crystal length, the defect position and the defect width. These findings hold great potential for implementing practical nonlinear optical isolating devices.

An index profile renormalization method is proposed to study the effective refraction index increase in a high-index contrast waveguide bend such as a silicon waveguide bend. This method transforms a waveguide bend to an equivalent straight waveguide (ESW). The simulation results show that the ESW method can calculate the effective refraction index increase in a three-dimensional (3D) bend, costing much less time than the traditional finite difference time domain method and the index profile renormalization (IPR) method developed for small radius cases. In addition, the field center shift is studied. The relationships are established between the effective refraction index increase and field center shift from the bend axis along the radial direction for estimations based on the approximated IPR method. These results can lead to great convenience in estimating the effective refraction index increase in 3D bend, which could be used in conditions requiring critical phase control.

Nocardioides, a genus belonging to Actinomycetes, can endure various low-nutrient conditions. It can degrade pollutants using multiple organic materials such as carbon and nitrogen sources. The characteristics and applications of Nocardioides are described in detail in this review, with emphasis on the degradation of several hard-to-degrade pollutants by using Nocardioides, including aromatic compounds, hydrocarbons, haloalkanes, nitrogen heterocycles, and polymeric polyesters. Nocardioides has unique advantages when it comes to hard-to-degrade pollutants. Compared to other strains, Nocardioides has a significantly higher degradation rate and requires less time to break down substances. This review can be a theoretical basis for developing Nocardioides as a microbial agent with significant commercial and application potential.

Ecosystems away from human disturbance provide an ideal paradigm for microbial ecology research. The Burqin glacier No. 18 in the Chinese Altay Mountains is such an ecosystem; however, there are no prior studies on the microbiology in the area. Here, we isolated 902 bacterial strains on the Burqin glacier No. 18 to determine the diversity and distribution characteristics of microorganisms. Isolated strains belonged to six phyla (in the order of dominance: Proteobacteria, Actinobacteria, Bacteroidetes, Cyanobacteria, Firmicutes, and Deinococcus-Thermus) and 90 genera. Our results also demonstrated the presence of a high proportion of potential new species (43%) in the Burqin glacier No. 18, and 67% of the potential new species were isolated at 25 °C. Species diversity varied among habitats, with the lowest diversity in surface ice and the highest diversity in the soil farthest from the glacier terminus. The pigmented colonies made up 52.7% of all isolates, with yellow-colored colonies being the most abundant (18.8%). This study indicates that the Burqin glacier No. 18 hosts rich bacterial strain diversity, and may represent a significant potential source of new functional and pigmented bacteria for the development of critical pharmaceuticals.

Abstract Strain Y74T was an isolate from the sandy soil in the town of Huatugou, Qinghai–Tibet Plateau, China. An analysis of this strain's phenotypic, chemotaxonomic, and genomic characteristics established the relationship of the isolate with the genus Planococcus. Strain Y74T was able to grow between 4 and 42°C (with an optimum temperature of 28°C) at pH values of 6–8.5 and in 0%–7% (w/v) NaCl. The dominant quinones were MK‐8 and MK‐7. The polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, and an unknown phospholipid. The majority of the fatty acid content was anteiso‐C15:0 (28.8%) followed by C16:1 ω7c alcohol (20.9%) and iso‐C14:0 (13.4%). The 16S rRNA gene sequence similarity analysis demonstrated a stable branch formed by strain Y74T and Planococcus halotolerans SCU63T (99.66%). The digital DNA–DNA hybridization between these two strains was 57.2%. The G + C content in the DNA of Y74T was 44.5 mol%. In addition, the morphological, physiological, and chemotaxonomic pattern clearly differentiated the isolates from their known relatives. In conclusion, the strain Y74T (=JCM 32826T = CICC24461T) represents a novel member of the genus Planococcus, for which the name Planococcus antioxidans sp. nov. is proposed. Strain Y74T was found to have potent antioxidant activity via its hydrogen peroxide tolerance and its 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical‐scavenging activity. The DPPH radical‐scavenging activity was determined to be 40.2 ± 0.7%. The genomic analysis indicated that six peroxidases genes, one superoxide dismutase gene, and one dprA (DNA‐protecting protein) are present in the genome of Y74T.

Mount Everest provides natural advantages to finding radiation-resistant extremophiles that are functionally mechanistic and possess commercial significance. (1) Background: Two bacterial strains, designated S5-59T and S8-45T, were isolated from moraine samples collected from the north slope of Mount Everest at altitudes of 5700m and 5100m above sea level. (2) Methods: The present study investigated the polyphasic features and genomic characteristics of S5-59T and S8-45T. (3) Results: The major fatty acids and the predominant respiratory menaquinone of S5-59T and S8-45T were summed as feature 3 (comprising C16:1 ω6c and/or C16:1 ω7c) and ubiquinone-10 (Q-10). Phylogenetic analyses based on 16S rRNA sequences and average nucleotide identity values among these two strains and their reference type strains were below the species demarcation thresholds of 98.65% and 95%. Strains S5-59T and S8-45T harbored great radiation resistance. The genomic analyses showed that DNA damage repair genes, such as mutL, mutS, radA, radC, recF, recN, etc., were present in the S5-59T and S8-45T strains. Additionally, strain S5-59T possessed more genes related to DNA protection proteins. The pan-genome analysis and horizontal gene transfers revealed that strains of Sphingomonas had a consistently homologous genetic evolutionary radiation resistance. Moreover, enzymatic antioxidative proteins also served critical roles in converting ROS into harmless molecules that resulted in resistance to radiation. Further, pigments and carotenoids such as zeaxanthin and alkylresorcinols of the non-enzymatic antioxidative system were also predicted to protect them from radiation. (4) Conclusions: Type strains S5-59T (=JCM 35564T =GDMCC 1.3193T) and S8-45T (=JCM 34749T =GDMCC 1.2715T) represent two novel species of the genus Sphingomonas with the proposed name Sphingomonas qomolangmaensis sp. nov. and Sphingomonas glaciei sp. nov. The type strains, S5-59T and S8-45T, were assessed in a deeply genomic study of their radiation-resistant mechanisms and this thus resulted in a further understanding of their greater potential application for the development of anti-radiation protective drugs.

Laohugou Glacier No. 12 is located on the northern slope of the western Qilian Mountains with a temperate continental wet climate and an extremely cold winter. Bacteria in a newly exposed moraine have to cope with various pressures owing to deglaciation at the glacier snout. However, limited information is available regarding the high diversity and temporary survival of culturable heterotrophic bacteria under various environmental stresses. To examine the tolerance of extremophiles against varying environmental conditions in a newly exposed moraine, we simulated environmental stress in bacterial cultures. The results showed that the isolated strains belonged to actinobacteria, Proteobacteria, Bacteroidetes, Deinococcus-Thermus, and Firmicutes. Actinobacteria was the most abundant phylum, followed by Proteobacteria, at both high and low temperatures. Pseudarthrobacter was the most abundant genus, accounting for 14.2% of the total isolates. Although several microorganisms grew at 10 °C, the proportion of microorganisms that grew at 25 °C was substantially higher. In particular, 50% of all bacterial isolates grew only at a high temperature (HT), whereas 21.4% of the isolates grew at a low temperature (LT), and 38.6% of the isolates grew at both HT and LT. In addition, many radiation-resistant extremophiles were identified, which adapted to both cold and oxidative conditions. The nearest neighbors of approximately >90% of bacteria belonged to a nonglacial environment, such as oil-contaminated soil, rocks, and black sand, instead of glacial niches. This study provides insights into the ecological traits, stress responses, and temporary survival of culturable heterotrophic bacteria in a newly exposed moraine with variable environmental conditions and the relationship of these communities with the non-glacial environment. This study may help to understand the evolution, competition, and selective growth of bacteria in the transition regions between glaciers and retreats in the context of glacier melting and retreat owing to global warming.

The Qaidam Basin of the Qinghai–Tibet Plateau is a cold, hyper-arid desert that presents extreme challenges to microbial communities. As little is known about variations between surface and subsurface microbial communities, high-throughput DNA sequencing was used in this study to profile bacterial communities of the soil samples collected at different depths in three regions in the Qaidam Basin. The α-diversity indices (Chao, Shannon, and Simpson) indicated that bacterial abundance and diversity were higher in the east and the high-elevation regions compared to the west region. In general, Firmicutes was dominant in the west region, while Proteobacteria and Acidobacteria were dominant in the east and the high-elevation regions. The structure of the bacterial communities differed greatly across regions, being strongly correlated with total organic carbon (TOC) and total nitrogen (TN) content. The differences in bacterial communities between the surface and the subsurface soil samples were smaller than the differences across the regions. Network analyses of environmental factors and bacterial genera indicated significant positive correlations in all regions. Overall, our study provides evidence that TOC and TN are the best predictors of both surface and subsurface bacterial communities across the Qaidam Basin. This study concludes that the bacterial community structure is influenced by both the spatial distance and the local environment, but environmental factors are the primary drivers of bacterial spatial patterns in the Qaidam Basin.

The Taklimakan Desert located in China is the second-largest shifting sand desert in the world and is known for its harsh conditions. Types of γ-rays or UV radiation-resistant bacterial strains have been isolated from this desert. However, there is no information regarding the proportions of the radiation-resistant strains in the total culturable microbes. We isolated 352 bacterial strains from nine sites across the Taklimakan Desert from north to south. They belong to Actinobacteria, Firmicutes, Proteobacteria, and Bacteroidetes. The phylum Actinobacteria was the most predominant in abundance and Firmicutes had the highest species richness. Bacteroidetes had the lowest abundance and was found in four sites only, while the other three phyla were found in every site but with different distribution profiles. After irradiating with 1000 J/m2 and 6000 J/m2 UV-C, the strains with survival rates higher than 10% occupied 72.3% and 36.9% of all culturable bacteria, respectively. The members from Proteobacteria had the highest proportions, with survival rates higher than 10%. After radiation with 10 kGy γ-rays, Kocuria sp. TKL1057 and Planococcus sp. TKL1152 showed higher radiation-resistant capabilities than Deinococcus radiodurans R1. Besides obtaining several radiation-resistant extremophiles, this study measured the proportions of the radiation-resistant strains in the total culturable microbes for the first time. This study may help to better understand the origin of radioresistance, especially by quantitatively comparing proportions of radiation-resistant extremophiles from different environments in the future.

Actinobacteria are known for their metabolic potential of producing diverse secondary metabolites such as antibiotics. Actinobacteria also playimportant roles in biogeochemical cycling and how soils develop. However, little is known about the effect of the vegetation type on the actinobacterial community structures in soils from arid regions. For these reasons, we have analyzed the actinobacterial communities of five types of ecosystem (tree grove, shrub, meadow, desert, and farm) in the Heihe river basin. Using 16S rRNA high-throughput sequencing, we found 11 classes of Actinobacteria, with dominant classes of Actinobacteria (36.2%), Thermoleophilia (28.3%), Acidimicrobiia (19.4%). Five classes, 15 orders, 20 families and 36 genera were present in all samples. The dominant generalist genera were Gaiella, Solirubrobacter, Nocardioides, Mycobacterium, and Pseudonocardia. The actinobacterial community structures were significantly affected by the environment and vegetation type. The diversity of the actinobacterial community in the desert ecosystem was high, and this ecosystem harbored the highest proportion of unclassified sequences, representing rare Actinobacteria. Functional metagenomic prediction, using PICRUSt, indicated that Actinobacteria play an important role in nitrogen cycling in both desert and cultivated farm ecosystems.

Bacterial communities in cold-desert habitats play an important ecological role. However, the variation in bacterial diversity and community composition of the cold-desert ecosystem in Qinghai–Tibet Plateau remains unknown. To fill this scientific gape, Illumina MiSeq sequencing was performed on 15 soil samples collected from different cold-desert habitats, including human-disturbed, vegetation coverage, desert land, and sand dune. The abundance-based coverage estimator, Shannon, and Chao indices showed that the bacterial diversity and abundance of the cold-desert were high. A significant variation reported in the bacterial diversity and community composition across the study area. Proteobacteria accounted for the largest proportion (12.4–55.7%) of all sequences, followed by Actinobacteria (9.2–39.7%), Bacteroidetes (1.8–21.5%), and Chloroflexi (2.7–12.6%). Furthermore, unclassified genera dominated in human-disturbed habitats. The community profiles of GeErMu, HongLiangHe, and CuoNaHu sites were different and metagenomic biomarkers were higher (22) in CuoNaHu sites. Among the soil physicochemical variables, the total nitrogen and electric conductivity significantly influenced the bacterial community structure. In conclusion, this study provides information regarding variation in diversity and composition of bacterial communities and elucidates the association between bacterial community structures and soil physicochemical variables in cold-desert habitats of Qinghai–Tibet Plateau.

Different stakeholders have different concerns about the transportation of hazardous chemicals. Multi-objective optimization comprehensively considering transportation risks and transportation interests is the primary demand for the transportation of hazardous chemicals. For this, this paper studies the multi-objective combinatorial optimization problem of multiple hazardous chemicals in the same distribution range of transportation network. Firstly, according to the transportation characteristics of hazardous chemicals, the calculation methods of distribution paths in terms of physical dissimilarity and spatial dissimilarity were proposed. Then, a multi-objective optimization model of hazardous chemicals transportation was established with transportation risk, cost and time as optimization objectives, and the model was solved by the improved NSGA-II algorithm. Finally, the feasibility of the optimization method was verified in practical examples. The research results show that the proper increase of transportation risk value can greatly reduce the transportation cost and transportation time. When the calculation results only emphasize the optimal risk, there exist a large number of shared road sections in different transportation planning paths, so that the transportation risks are concentrated; however, after setting the threshold and increasing the constraint conditions, the phenomenon of the shared road section is significantly reduced, and the distribution of the hazardous chemicals transportation risks is relatively dispersed, to further control the potential hazards. This shall provide a theoretical reference for the risk regulation of hazardous chemicals transportation and the cost control of carriers.

The dissacharide trehalose is an important intracellular osmoprotectant and the OtsA/B pathway is the principal pathway for trehalose biosynthesis in a wide range of bacterial species. Scaffolding proteins and other cytoskeletal elements play an essential role in morphogenetic processes in bacteria. Here we describe how OtsA, in addition to its role in trehalose biosynthesis, functions as an osmotic stress sensor to regulate cell morphology in Arthrobacter strain A3. In response to osmotic stress, this and other Arthrobacter species undergo a transition from bacillary to myceloid growth. An otsA null mutant exhibits constitutive myceloid growth. Osmotic stress leads to a depletion of trehalose-6-phosphate, the product of the OtsA enzyme, and experimental depletion of this metabolite also leads to constitutive myceloid growth independent of OtsA function. In vitro analyses indicate that OtsA can self-assemble into protein networks, promoted by trehalose-6-phosphate, a property that is not shared by the equivalent enzyme from E. coli, despite the latter's enzymatic activity when expressed in Arthrobacter. This, and the localization of the protein in non-stressed cells at the mid-cell and poles, indicates that OtsA from Arthrobacter likely functions as a cytoskeletal element regulating cell morphology. Recruiting a biosynthetic enzyme for this morphogenetic function represents an intriguing adaptation in bacteria that can survive in extreme environments.

Waterlogging has increasingly become one of the major constraints to maize productivity in some maize production zones because it causes serious yield loss. Bulked segregant RNA-seq (BSR-seq) has been widely applied to profile candidate genes and map associated Single Nucleotide Polymorphism (SNP) markers in many species. In this study, 10 waterlogging sensitive and eight tolerant inbred lines were selected from 60 maize inbred lines with waterlogging response determined and preselected by the International Maize and Wheat Improvement Center (CIMMYT) from over 400 tropical maize inbred lines. BSR-seq was performed to identify differentially expressed genes and SNPs associated with waterlogging tolerance. Upon waterlogging stress, 354 and 1094 genes were differentially expressed in the tolerant and sensitive pools, respectively, compared to untreated controls. When tolerant and sensitive pools were compared, 593 genes were differentially expressed under untreated and 431 genes under waterlogged conditions, of which 122 genes overlapped. To validate the BSR-seq results, the expression levels of six genes were determined by qRT-PCR. The qRT-PCR results were consistent with BSR-seq results. Comparison of allelic polymorphism in mRNA sequences between tolerant and sensitive pools revealed 165 (normal condition) and 128 (waterlogged condition) high-probability SNPs. We found 18 overlapping SNPs with genomic positions mapped. Eighteen SNPs were contained in 18 genes, and eight and nine of 18 genes were responsive to waterlogging stress in tolerant and sensitive lines, respectively. Six alleles of the 18 originated from tolerant pool were significantly up-regulated under waterlogging, but not those from sensitive pool. Importantly, one allele (GRMZM2G055704) of the six genes was mapped between umc1619 and umc1948 on chromosome 1 where a QTL associated with waterlogging tolerance was identified in a previous research, strongly indicating that GRMZM2G055704 is a candidate gene responsive to waterlogging. Our research contributes to the knowledge of the molecular mechanism for waterlogging tolerance in maize.

Spikelet number per panicle (SPP), differentiated spikelet number per panicle (D-SPP), and preflowering aborted spikelet number per panicle (A-SPP) were examined in five rice cultivars at three planting densities (HD; high, MD; medium, LD; low planting density) in the field condition. Rice plants at LD produced a higher panicle number per plant but lower panicle number per unit area, accompanied by higher D-SPP and SPP, on average. A-SPP and the ratio of A-SPP to D-SPP (%A) showedno consistent trends. There was a broader range of D-SPP values at LD than at HD because of larger D-SPP in higher order panicles (panicles with a higher spikelet number). D-SPP was smaller in lower order panicles in all cultivars and years, whereas %A increased. D-SPP and SPP of each panicle were positively correlated with tiller size (tiller height, leaf area, and neck internode diameter). Spikelet production efficiency for D-SPP or for SPP (spikelet number per leaf area) of each tiller was higher in IR65564-44-51 (NPT65) and Akihikari than in the other cultivars, indicating a greater capacity of tillers to produce spikelets or support spikelet growth. In each cultivar except NPT65, spikelet production efficiency for D-SPP increased as panicle order decreased, whereas spikelet production efficiency for SPP remained constant or decreased. This finding indicates that irrespective of planting density, lower order panicles produce more spikelets than they can afford physiologically, but they were regulated downward to a nearly constant value in four cultivars. In NPT65 different from other cultivars, spikelet production efficiency for D-SPP was lower in lower order panicles.

A measles outbreak occurred in a western county of China in 2013, the year after China's historic nadir of measles. We conducted a field investigation to identify gaps in measles vaccination coverage and immunization program weaknesses, and to provide recommendations for measles outbreak response and immunization program improvement.We analyzed surveillance data from 2008 to 2013 to describe the measles epidemiology of the county. Measles-containing vaccine coverage was estimated using two methods: previously-reported administrative coverage and an estimation of coverage by clinic-kept vaccination records (n = 542). We conducted a rapid field coverage assessment in a migrant population village to evaluate coverage after emergency vaccination. We conducted a review of hospital records of measles cases to address the role hospital transmission played during the early stage of this outbreak.There were 153 cases in the outbreak, primarily among children too young to vaccinate, unvaccinated children less than 3 years old, and adults. Measles-containing vaccine coverage by the field assessment showed that 20% of children aged 8-17 months had zero doses, and 9% of ≥2 years old children had fewer than two doses. The vaccination statuses of all adult cases were either zero doses or unknown. At least 61% of cases had been hospitalized. The proportion of cases who had been hospital-exposed 7 to 21 days prior to rash onset decreased from 52% to 22% after hospitals strengthen their isolation measures.This outbreak was a result of measles vaccination coverage gaps among young children and adults, and insufficient hospital isolation of cases. The lower coverage seen in the field estimation compared with reported coverage showed that reported coverage could have been overestimated. Hospitals were sites of transmission in the early stage of the outbreak. A strict hospital isolation policy could decrease spread of measles. Emergency vaccination was associated with stopping measles transmission in low coverage areas.

Glacier forelands, where the initially exposed area is unvegetated with minimal human influence, are an ideal place for research on the distributions and biogeography of microbial communities. Actinobacteria produce many bioactive substances and have important roles in soil development and biogeochemical cycling. However, little is known about the distribution and biogeography of Actinobacteria in glacier forelands. Therefore, we investigated the patterns of diversity and the biogeography of actinobacterial communities of the inhabited forefields of 5 glaciers in China. Of the bacteria, the mean relative abundance of Actinobacteria was 13.1%, and 6 classes were identified in the phylum Actinobacteria. The dominant class was Actinobacteria (57%), which was followed in abundance by Acidimicrobiia (19%) and Thermoleophilia (19%). When combined, the relative abundance of the other three classes, the MB-A2-108, Nitriliruptoria and Rubrobacteria, was only 2.4%. A biogeographic pattern in the forelands of the 5 glaciers in China was not detected for actinobacterial communities. Compared with 7 other actinobacterial communities found in the forelands of glaciers globally, those in the Southern Hemisphere were significantly different from those in the Northern Hemisphere. Moreover, the communities were significantly different on the separate continents of the Northern Hemisphere. The dissimilarity of the actinobacterial communities increased with geographic distance ( r = 0.428, p = 0.0003). Because of environmental factors, the effect of geography was clear when the distance exceeded a certain continent-level threshold. With the analysis of indicator species, we found that each genus had a geographic characteristic, which could explain why the communities with greater diversity were more strongly affected by biogeography.

Drought stress is the major threat to maize development and production. Here the bulked segregant transcriptome analysis (BSTA) was applied to discover the molecular mechanisms of drought stress responses in maize. The evaluation of drought stress tolerance was contacted for 200 maize inbred lines. Two bulk pools were constructed by selecting 10 inbred lines that were tolerant to drought (TD) and other 10that were sensitive to drought (SD). BSTA was performed to identify the differentially expressed genes (DEGs) within two kinds of maize under drought treatment. A total of 4886 and 5274 DEGs were detected in TD and SD bulk pools, respectively. The functional annotations showed that most of the DEGs are involved in the hormone metabolism, transcription regulation and alternative splicing pathways. Comparisons of the DEGs between two bulk pools revealed that the number of up-regulated genes related to alternative splicing in TD bulk pool was significantly higher than that in SD bulk pool. Besides, 928 up-regulated genes and 346 down-regulated genes among 1274 DEGs were identified including protein kinase andtranscription factors only in TD bulk pool. This result indicates that many common mechanisms are involved in drought regulation, moreover alternative splicing may play dominant role in response to drought stress in maize.
