Abstract

Type I interferon (IFN-I) responses are critical for the control of RNA virus infections, however, many viruses, including Dengue (DENV) and Chikungunya (CHIKV) virus, do not directly activate plasmacytoid dendritic cells (pDCs), robust IFN-I producing cells. Herein, we demonstrated that DENV and CHIKV infected cells are sensed by pDCs, indirectly, resulting in selective IRF7 activation and IFN-I production, in the absence of other inflammatory cytokine responses. To elucidate pDC immunomodulatory functions, we developed a mouse model in which IRF7 signaling is restricted to pDC. Despite undetectable levels of IFN-I protein, pDC-restricted IRF7 signaling controlled both viruses and was sufficient to protect mice from lethal CHIKV infection. Early pDC IRF7-signaling resulted in amplification of downstream antiviral responses, including an accelerated natural killer (NK) cell-mediated type II IFN response. These studies revealed the dominant, yet indirect role of pDC IRF7-signaling in directing both type I and II IFN responses during arbovirus infections.

Abstract

BACKGROUND: Cardiac magnetic resonance (CMR) as advanced diagnostic tool for the heart has been introduced in our institution since September 2019. AIM: We report on the first fifty consecutive patients using this imaging modality. METHODS AND MATERIALS: Strict protocol for CMR procedure, imaging quality assessment, contraindications, and informed consent were established. Patients selected for CMR were enrolled in a prospective registry. Visualizing the heart chambers, heart muscle and heart valves, resulted in acquiring complex imaging of the heart structure and function. When applicable, patients received gadolinium contrast agent for Late Gadolinium Enhancement (LGE). Adenosine was used for stress induced myocardial perfusion study. In this study, we report on the initial CMR procedures in the first 15 months. RESULTS: The age of the patients ranges from 17 to 82 and the number of male and female patients was well balanced. No absolute contraindications were met in any patient. Relative contraindications were noted but did not prevent from performing the scan. Different cardiac pathologies were encountered in the examined patients. Most common was the ischemic heart disease – 19 (38%). We had 15 (30%) out of 46 (92%) CMR procedures with LGE showing fibrotic scaring. Quality image assessment was scaled from poor to excellent. Most of the assessments were graded very good and good (46% and 48%), no poor, and very poor noted. CONCLUSION: CMR has been successfully introduced in Kosovo as excellent imaging tool for diagnosing and characterizing a nearly exhaustive spectrum of heart diseases.

Abstract

Background Cancer patients are thought to have an increased risk of developing severe Coronavirus Disease 2019 (COVID-19) infection and of dying from the disease. In this work, predictive factors for COVID-19 severity and mortality in cancer patients were investigated. Patients and Methods In this large nationwide retro-prospective cohort study, we collected data on patients with solid tumours and COVID-19 diagnosed between March 1 and June 11, 2020. The primary endpoint was all-cause mortality and COVID-19 severity, defined as admission to an intensive care unit (ICU) and/or mechanical ventilation and/or death, was one of the secondary endpoints. Results From April 4 to June 11, 2020, 1289 patients were analysed. The most frequent cancers were digestive and thoracic. Altogether, 424 (33%) patients had a severe form of COVID-19 and 370 (29%) patients died. In multivariate analysis, independent factors associated with death were male sex (odds ratio 1.73, 95%CI: 1.18-2.52), ECOG PS ≥ 2 (OR 3.23, 95%CI: 2.27-4.61), updated Charlson comorbidity index (OR 1.08, 95%CI: 1.01-1.16) and admission to ICU (OR 3.62, 95%CI 2.14-6.11). The same factors, age along with corticosteroids before COVID-19 diagnosis, and thoracic primary tumour site were independently associated with COVID-19 severity. None of the anticancer treatments administered within the previous 3 months had any effect on mortality or COVID-19 severity, except cytotoxic chemotherapy in the subgroup of patients with detectable SARS-CoV-2 by RT-PCR, which was associated with a slight increase of the risk of death (OR 1.53; 95%CI: 1.00-2.34; p = 0.05). A total of 431 (39%) patients had their systemic anticancer treatment interrupted or stopped following diagnosis of COVID-19. Conclusions Mortality and COVID-19 severity in cancer patients are high and are associated with general characteristics of patients. We found no deleterious effects of recent anticancer treatments, except for cytotoxic chemotherapy in the RT-PCR-confirmed subgroup of patients. In almost 40% of patients, the systemic anticancer therapy was interrupted or stopped after COVID-19 diagnosis., Highlights • A total of 1289 patients with solid tumours and COVID-19 were analysed. • Mortality and COVID-19 severity were mainly driven by patient general characteristics. • Overall, we found no deleterious effects of recent anticancer treatments on mortality. . • Systemic anticancer treatment was interrupted or stopped in 39% of patients.

Abstract

The aim of this research is to present body mass index (BMI) data in children 3-5 years of age from Skopje and provide the information on the prevalence of different categories of nutritional status during the early childhood. Material and methods: The study included 420 preschool children (210 boys and 210 girls). Stature and body weight were measured, and the BMI value was calculated. Twelve anthropometric parameters were measured using standard equipment and measurement technique according to the International Biological Programme. Results: The majority of anthropometrical parameters have shown significant age and sex specific differences in favor of male subjects. Values at the 50th percentile in our male subjects aged 3, 4 and 5 years for the weight-for-age index were 19 kg, 19.1 and 21 kg, respectively whereas in the female subjects the corresponding values were 16.8 kg, 20 kg and 21 kg. The height-for-age index values corresponding to the 50th percentile showed slightly higher values in our male subjects (100 cm, 109.5 cm and 116 cm) than those in our female subjects (102 cm, 108.5 cm and 116 cm). The values of 50th percentile of BMI in our males subjects were 18.1 kg/m², 16.2 kg/m² and 16 kg/m² whereas in our females were 16.5 kg/m², 16.7 kg/m² and 16.4 kg/m². Conclusion: These results show that underweight is a health problem of the rising generation in Skopje and needs to be considered along with the problem of overweight. It is recommended that the detected values could be applied for evaluation of deviations in growth and nutritional status in children aged 3-5.

Abstract

1.1. Objectives: The main objective of the study was to assess the impact of socioeconomic factors on somatotype components in Macedonian adolescents at the age 11 to 18 years. 1.2. Methods:The study included 2388 adolescents (1236 males and 1152 females) from four urban cities in R. Macedonia. Participants were measured with standard equipment and measurement technique according to the International Biological Program. Ten anthropometric parameters were measured to assess the somatotype according to Heat–Carter somatotyping method. The examinees filled out the questionnaire with the data on socioeconomic status, parent’s education and employment. 1.3. Results: There were significant differences for endomorph component in male adolescents between those with parents with high and low level of education. There were significant differences for mesomorph component in males related to parents educational level. Ectomorph component had a higher value in male adolescents when mother is with low educational level and when mother and father are unemployed. There were significant differences for endomorph component in female adolescents in relation to all socioeconomic factors except for employment status of the father; mesomorph and ectomorph component in females were related to parents educational level 1.4. Cconclusion:Our findings suggest that socioeconomic factors may be related to somatotype components in Macedonian adolescents

Abstract

NK-cell killing requires both the expression of activating receptor ligands and low MHC class I expression by target cells. Here we demonstrate that the expression of any of the murine ligands for the NK-cell activating receptor NKG2D results in a concomitant reduction in MHC class I expression. We show this both in tumor cell lines and in vivo. NK-cell lysis is enhanced by the decrease in MHC class I expression, suggesting the change is biologically relevant. These results demonstrate that NKG2D ligand expression on target cells not only allows for activating receptor recognition, but also actively reduces expression of the inhibitory ligand, MHC class I, leading to enhanced recognition and killing by NK cells.

Abstract

CD4-unhelped CD8(+) T cells are functionally defective T cells primed in the absence of CD4(+) T cell help. Given the co-stimulatory role of natural-killer group 2, member D protein (NKG2D) on CD8(+) T cells, we investigated its ability to rescue these immunologically impotent cells. We demonstrate that augmented co-stimulation through NKG2D during priming paradoxically rescues memory, but not effector, CD8(+) T cell responses. NKG2D-mediated rescue is characterized by reversal of elevated transcription factor T-box expressed in T cells (T-bet) expression and recovery of interleukin-2 and interferon-γ production and cytolytic responses. Rescue is abrogated in CD8(+) T cells lacking NKG2D. Augmented co-stimulation through NKG2D confers a high rate of survival to mice lacking CD4(+) T cells in a CD4-dependent influenza model and rescues HIV-specific CD8(+) T cell responses from CD4-deficient HIV-positive donors. These findings demonstrate that augmented co-stimulation through NKG2D is effective in rescuing CD4-unhelped CD8(+) T cells from their pathophysiological fate and may provide therapeutic benefits.

Abstract

SummaryThe mechanisms by which cytotoxic T lymphocytes (CTLs) enter and are retained in nonlymphoid tissue are not well characterized. With a transgenic mouse expressing the NKG2D ligand retinoic acid early transcript 1ε (RAE1ε) in β-islet cells of the pancreas, we found that RAE1 expression was sufficient to induce the recruitment of adoptively transferred CTLs to islets. This was dependent on NKG2D expression by the CTLs and independent of antigen recognition. Surprisingly, the recruitment of CTLs resulted in the subsequent recruitment of a large number of endogenous lymphocytes. Whereas transgenic mice did not develop diabetes, RAE1 expression was sufficient to induce insulitis in older, unmanipulated transgenic mice that was enhanced by viral infection and pancreatic inflammation. These results demonstrate that the expression of an NKG2D ligand in islets is sufficient to recruit CTLs regardless of their antigen specificity and to induce insulitis.

Abstract

Epithelial cells respond to physicochemical damage with up-regulation of major histocompatibility complex-like ligands that can activate the cytolytic potential of neighboring intraepithelial T cells by binding the activating receptor, NKG2D. The systemic implications of this lymphoid stress-surveillance response, however, are unknown. We found that antigens encountered at the same time as cutaneous epithelial stress induced strong primary and secondary systemic, T helper 2 (T(H)2)-associated atopic responses in mice. These responses required NKG2D-dependent communication between dysregulated epithelial cells and tissue-associated lymphoid cells. These data are germane to uncertainty over the afferent induction of T(H)2 responses and provide a molecular framework for considering atopy as an important component of the response to tissue damage and carcinogenesis.

Abstract

Natural killer cell recognition of “missing self” contributes meaningfully to control of mouse cytomegalovirus infection in vivo., Cytomegaloviruses (CMVs) are renowned for interfering with the immune system of their hosts. To sidestep antigen presentation and destruction by CD8+ T cells, these viruses reduce expression of major histocompatibility complex class I (MHC I) molecules. However, this process sensitizes the virus-infected cells to natural killer (NK) cell–mediated killing via the “missing self” axis. Mouse cytomegalovirus (MCMV) uses m152 and m06 encoded proteins to inhibit surface expression of MHC I molecules. In addition, it encodes another protein, m04, which forms complexes with MHC I and escorts them to the cell surface. This mechanism is believed to prevent NK cell activation and killing by restoring the “self” signature and allowing the engagement of inhibitory Ly49 receptors on NK cells. Here we show that MCMV lacking m04 was attenuated in an NK cell– and MHC I–dependent manner. NK cell–mediated control of the infection was dependent on the presence of NK cell subsets expressing different inhibitory Ly49 receptors. In addition to providing evidence for immunoevasion strategies used by CMVs to avoid NK cell control via the missing-self pathway, our study is the first to demonstrate that missing self–dependent NK cell activation is biologically relevant in the protection against viral infection in vivo.

Abstract

To assess the anthropometric parameters of growth and nutritional status in relation to socioeconomic status (SES) of Macedonian adolescents.The study included 546 adolescents from urban regions of the Republic of Macedonia, aged 14 to 15 years. Participants were measured with standard equipment and measurement technique according to the International Biological Programme. The following anthropometric indices were calculated: height-for-age (BH), weight-for-age (BW) and BMI-for-age (kg/m2). For measuring family wealth, the Family Affluence Scale (FAS) has been used. Adolescents were grouped into three SES groups.Age-specific differences were found for body height and weight in favour of 15-year-old males (p0.05). There were no significant differences in height and weight among different SES groups. Female participants were more overweight in the high SES group (13.2%) and middle SES group (10.8%), compared to those in the low SES group (6.7%). According to the obtained results in females, the prevalence of obese and underweight females is higher in those with low SES.Male adolescents were found to be taller and heavier than female adolescents. Both male and female adolescents were taller and heavier in high SES group than those in other SES groups. There were no SES differences in prevalence of underweight, overweight and obesity among Macedonian school adolescents living in urban areas, except among females, those in the low SES group had a significantly higher prevalence of obesity than did those in the MSES and HSES groups.

Abstract

The human palms and soles are textured with skin different to that of the other body surface. Instead of sebaceous glands or hairs, there are sweat glands opening into epidermal ridges. They are variously oriented, thus forming different patterns. The aim of the study was to assess some dermatoglyphic patterns, the TRC and the a-b ridge count in a sample of patients and a control group of healthy examinees.59 male and 50 female patients with schizophrenia and 60 healthy male and 50 female individuals as a control group were included in the study. Hand prints were taken using the method of Cummins and Midlo; pattern types were determined and classified using the Henry classification. An analysis of pattern frequency in schizophrenia compared to the normal controls was conducted.The patterns most frequently present are loops (ulnar) with greater incidence and whorls with smaller incidence in patients with schizophrenia compared to the healthy individuals. The number of arches is higher in male patients and lower in female patients. Complex patterns are rare. TRC in females has lower values found in patients. The a-b ridge count has significant difference in male and female patients; higher values in male and lower in female patients.There is some association between the frequency of certain dermatoglyphic characteristicss and schizophrenia. Our findings form a useful database for the dermatoglyphic marks present in patients with schizophrenia and healthy subjects of Macedonian nationality.

Abstract

NKG2D is one of the most intensively studied immune receptors of the past decade. Its unique binding and signaling properties, expression pattern, and functions have been attracting much interest within the field due to its potent antiviral and anti-tumor properties. As an activating receptor, NKG2D is expressed on cells of the innate and adaptive immune system. It recognizes stress-induced MHC class I-like ligands and acts as a molecular sensor for cells jeopardized by viral infections or DNA damage. Although the activating functions of NKG2D have been well documented, recent analysis of NKG2D-deficient mice suggests that this receptor may have a regulatory role during NK cell development. In this review, we will revisit known aspects of NKG2D functions and present new insights in the proposed influence of this molecule on hematopoietic differentiation.

Abstract

It is uncertain whether NK cells modulate T cell memory differentiation. By using a genetic model that allows the selective depletion of NK cells, we show in this study that NK cells shape CD8+ T cell fate by killing recently activated CD8+ T cells in an NKG2D- and perforin-dependent manner. In the absence of NK cells, the differentiation of CD8+ T cells is strongly biased toward a central memory T cell phenotype. Although, on a per-cell basis, memory CD8+ T cells generated in the presence or the absence of NK cells have similar functional features and recall capabilities, NK cell deletion resulted in a significantly higher number of memory Ag-specific CD8+ T cells, leading to more effective control of tumors carrying model Ags. The enhanced memory responses induced by the transient deletion of NK cells may provide a rational basis for the design of new vaccination strategies.

Abstract

SummaryNKG2D is a potent activating receptor on natural killer (NK) cells and acts as a molecular sensor for stressed cells expressing NKG2D ligands such as infected or tumor-transformed cells. Although NKG2D is expressed on NK cell precursors, its role in NK cell development is not known. We have generated NKG2D-deficient mice by targeting the Klrk1 locus. Here we provide evidence for an important regulatory role of NKG2D in the development of NK cells. The absence of NKG2D caused faster division of NK cells, perturbation in size of some NK cell subpopulations, and their augmented sensitivity to apoptosis. As expected, Klrk1−/− NK cells are less responsive to tumor targets expressing NKG2D ligands. Klrk1−/− mice, however, showed an enhanced NK cell-mediated resistance to mouse cytomegalovirus infection as a consequence of NK cell dysregulation. Altogether, these findings provide evidence for regulatory function of NKG2D in NK cell physiology.

Abstract

Enantioseparation ability and enantiopreference of chiral stationary phases CSP 1–CSP 3, containing a terminal N-3,5-dinitrobenzoyl (N-DNB) unit, and CSP 4, containing a terminal N-benzoyl (N-B) unit, are studied. Separation factors (α) for the two sets of test racemates (TR) that structurally match the chiral selector of these CSPs have been determined. The first set consists of seven N-DNB α-amino acid isopropylesters (TR 1A–TR 7A), and the second one of their N-B analogues (TR 1–TR 7). The best enantioseparation (αaverage 1.27) is obtained when π-acceptor DNB unit is present in both TR and CSP. One π-acceptor unit, either in the analyte or in CSP, suffices for efficient enantioseparation (αaverage 1.19). Interaction between π-neutral units in the CSP and test racemate does not afford effective enantioseparation (αaverage 1.03). Using (S)-enantiomers of all TRs as standards, CD detection has revealed regular preference of the CSPs for the enantiomers containing amino acid amide of the same absolute configuration. The possible origin of such enantiopreference is discussed.

Abstract

In the absence of CD4+ T cell help, primed CD8+ T cells mount defective responses. However, the description of priming cues capable of rescuing these defective functions is lacking. Here, we show that concurrent delivery of DNA plasmids encoding antigen and NKG2D ligand, Rae-1ϵ, to antigen presenting cells rescues CD4-unhelped CD8+ T cell memory recall but not effector phase responses. NKG2D-mediated rescue of CD4-unhelped CD8+ T cell memory recall responses is characterized by reversal of elevated T-bet expression, recovery of granzyme B, IL-2, and IFNγ production and recovery of CTL function. NKG2D-mediated rescue requires NKG2D expression on CD8+ T cells and is dependent on IL-15. These findings have the potential to address limitations of current CD8+ T cell regiments used in the absence of CD4 help against intracellular pathogens and cancer, by generating effective CD8+ T cell memory recall responses.
