Abstract

Autologous T-cells expressing Chimeric Antigen Receptor (CAR) have shown great efficacy against hematologic malignancies, however, severe side effects limit their clinical use. Natural Killer (NK) cells are an attractive alternative to T-cells as carriers for CARs, since they could be used as an allogeneic treatment, and have no, or minimal, risk for Cytokine Release Syndrome (CRS). CAR-NK cells have recently shown promising effect against leukemia without serious side effects after adoptive transfer. Possible allogeneic application of CAR-NK cells could dramatically reduce the costs of such adoptive cell therapy, nevertheless sufficient ex vivo expansion of NK cells remains challenging. Transgenic expression of cytokines in CAR-NK cells was shown to partially improve pro-survival or proliferative properties (e.g., IL-15), however what is the optimal cytokine for CAR NK cells is not clear to date. Here, we compared 2nd generation CD19-targeted CAR (CAR.19) with a fourth generation CAR.19 coexpressing Il-27 (CAR.19-IL27) in promoting NK-92 cell line proliferation, activation, and cytotoxic activity against B-cell cancers. CAR constructs were inserted into lentiviral vectors and subsequently transduced into the NK-92 cell line. Effects of CAR-NK cells were assessed against CD19-expressing B-cell lines NALM-6 or Raji in vitro and in vivo in a murine model of B-cell neoplasia with Raji cells. Tumor burden was measured by bioluminescence and by the curve of survival. We have demonstrated that CAR.19-IL27 exhibits a superior expansion capacity in comparison to both NK-92wt and CAR.19 cells. Moreover, CAR.19-IL27 cells showed higher cytotoxicity towards NALM-6 and Raji compared to CAR.19 in vitro (**p < 0.001). Degranulation assay, assed by CD107a, showed that CAR.19-IL27 have higher expression of CD107a compared to CAR.19 cells (**p < 0.001), which indicates increased secretion of perforin and granzymes. Our systematic transcriptome analysis of activated NK-92 CAR variants shows that IL-27 may be involved in the PI3K/AKT, and MAPK signaling pathways, as genes related to proliferation from these pathways were increased, e.g., PDK1, RPS6, MYC, and PGK1. In vivo studies with a xenograft mouse model of B-cell neoplasia showed that CAR.19-IL27 reduced tumor burden compared to CAR.19-treated. Altogether, CD19-CAR NK cells co-expressing Il-27 showed potent and selective antitumor activity with the potential to improve clinical use of CAR NK cells.

Abstract

BackgroundThe use of CD20-depleting monoclonal antibodies has shown to improve the long-term outcome of patients with anti-leucine-rich glioma-inactivated protein 1 antibodies (anti-LGI1-Abs) encephalitis after first-line immunotherapy, but currently predictive markers of treatment response and disease activity are lacking.Case presentationA 75-year-old man presented cognitive impairment and faciobrachial dystonic seizures (FBDS), with mild abnormalities at electroencephalography (EEG), normal brain magnetic resonance and cerebrospinal fluid (CSF) analysis. Anti-LGI1-Abs were detected in serum and CSF, and corticosteroids and intravenous immunoglobulins were administered. Despite partial cognitive improvement, 18F-fluoridesoxyglucose-positron emission tomography (18F-FDG-PET) showed the persistence of temporo-mesial hypermetabolism, and FBDS were still detected by long-term monitoring video EEG (LTMV EEG). Rituximab was therefore administered with FBDS disappearance, further cognitive improvement, and resolution of 18F-FDG-PET temporo-mesial hypermetabolism.ConclusionsOur experience supports the use of 18F-FDG-PET and LTMVEEG as useful tools to measure disease activity, evaluate treatment response and guide therapeutic decisions in the long-term management of anti-LGI1-antibody encephalitis.

Abstract

This study investigates the influence of EU Registration, Evaluation, Authorisation and Restriction of Chemicals (REACH) legislation on the technological trajectory of the chemical industry. Focusing on the Substances of Very High Concern (SVHC) list, this study used a novel dataset of chemical patents and structures filed at the European Patent Office (EPO) over the period from 1990 to 2016. A staggered difference-in-differences approach with inverse probability weights was used to measure the impact of the SVHC list on the novelty and toxicity of chemical patents. The analysis revealed a substantial increase in the chemical novelty of patents after the introduction of the SVHC list. Abstract

Given the global decline in adherence to the Mediterranean Diet (MD), even within its native region, it is key to identify the factors influencing this trend to mitigate the negative health outcomes associated with westernized diets. To this end, 4025 individuals (49.6% women, 42.6 ± 14.2 y/o) from Greece, Italy, Morocco, Slovenia, and Tunisia remotely completed a series of measures assessing motives, attitudes, and psychosocial factors related to MD adherence, which was evaluated using the MEDAS questionnaire. The results suggested medium-to-low adherence across all countries, with the highest adherence in Italy and Morocco and the lowest in Slovenia. Structural equation modeling revealed that positive attitudes toward the healthiness of food were the strongest predictors of adherence, whereas picky eating was a significant negative predictor in all countries except Greece. Adherence to the MD was positively influenced by health motivations in Morocco and weight control in Slovenia and Greece, while sensory appeal negatively influenced adherence in Italy. Additionally, price and convenience were significant barriers in Tunisia and Greece, whereas a preference for local and seasonal foods promoted adherence in Morocco and Greece. Overall, our findings underscore the need for country-specific interventions and policies that address distinct local factors and motivations to ease favorable shifts in dietary patterns toward MD principles. Abstract The aim of this work was to assess if the commercially available Fluconazole drug products (Reference, Generic and Similar) would meet the biowaiver criteria from Food and Drug Administration (FDA) and Brazilian Agency for Health Surveillance (ANVISA) agencies. All formulations were evaluated considering the dissolution profile carried out in Simulated Gastric Fluid (SGF) pH 1.2, Acetate Buffer (AB) pH 4.5 and Simulated Intestinal Fluid (SIF) pH 6.8. The results demonstrated that all formulations fulfilled the 85% of drug dissolved at 30 min criterion in SGF pH 1.2. However, in AB pH 4.5 and SIF pH 6.8, some formulations, including the comparator, did not achieve this dissolution percentage. The discrepant dissolution profiles also failed the ƒ2 similarity factor analysis, since none of the formulations showed values between 50 and 100 in the three dissolution media. Comparative dissolution profiles were not similar, considering that the main issues concerning the dissolution were evidenced for the comparator product. Hence, a revision in the regulatory norms in order to establish criteria to switch the comparator could result in an increased application of drugs based on biowaiver criteria.

Abstract Background A proper restoration of hip biomechanics is fundamental to achieve satisfactory outcomes after total hip arthroplasty (THA). A global hip offset (GO) postoperatively reduction of more than 5 mm was known to impair hip functionality after THA. The equine head is an anatomically complex area, therefore advanced tomographic imaging techniques, such as computed tomography or magnetic resonance imaging (MRI), are often required for diagnosis and treatment planning. The purpose of this multicenter retrospective study was to describe MRI characteristics for a large sample of horses with head disorders. Horses imaged over a period of 13 years were recruited. Eighty-four horses met the inclusion criteria, having neurological (n = 65), sinonasal (n = 14), and soft tissue (n = 5) disorders. Magnetic resonance imaging accurately depicted the anatomy and allowed identification of the primary lesion and associated changes. There were good correlations between MRI findings and intraoperative or postmortem results. Magnetic resonance imaging showed the exact localization of the lesions, their size, and relation to surrounding structures. However, in the neurological group, there were 45 horses with no MRI abnormalities, 29 of which had a history of recurrent seizures, related to cryptogenic epilepsy. Abstract

A diet rich in phytonutrients from fruit and vegetables has been acknowledged to afford protection against a range of human diseases, but many of the most popular vegetables are low in phytonutrients. Wild relatives of crops may contain allelic variation for genes determining the concentrations of these beneficial phytonutrients, and therefore understanding the genetic basis of this variation is important for breeding efforts to enhance nutritional quality. In this study, lettuce recombinant inbred lines, generated from a cross between wild and cultivated lettuce (Lactuca serriola and Lactuca sativa, respectively), were analysed for antioxidant (AO) potential and important phytonutrients including carotenoids, chlorophyll and phenolic compounds. When grown in two environments, 96 quantitative trait loci (QTL) were identified for these nutritional traits: 4 for AO potential, 2 for carotenoid content, 3 for total chlorophyll content and 87 for individual phenolic compounds (two per compound on average). Most often, the L. serriola alleles conferred an increase in total AOs and metabolites. Candidate genes underlying these QTL were identified by BLASTn searches; in several cases, these had functions suggesting involvement in phytonutrient biosynthetic pathways. Analysis of a QTL on linkage group 3, which accounted for >30% of the variation in AO potential, revealed several candidate genes encoding multiple MYB transcription factors which regulate flavonoid biosynthesis and flavanone 3-hydroxylase, an enzyme involved in the biosynthesis of the flavonoids quercetin and kaempferol, which are known to have powerful AO activity. Follow-up quantitative RT-PCR of these candidates revealed that 5 out of 10 genes investigated were significantly differentially expressed between the wild and cultivated parents, providing further evidence of their potential involvement in determining the contrasting phenotypes. These results offer exciting opportunities to improve the nutritional content and health benefits of lettuce through marker-assisted breeding.

Abstract

The LFI radiometers use a pseudo-correlation design where the signal from the sky is continuously compared with a stable reference signal, provided by a cryogenic reference load system. The reference unit is composed by small pyramidal horns, one for each radiometer, 22 in total, facing small absorbing targets, made of a commercial resin ECCOSORB CR (TM), cooled to approximately 4.5 K. Horns and targets are separated by a small gap to allow thermal decoupling. Target and horn design is optimized for each of the LFI bands, centered at 70, 44 and 30 GHz. Pyramidal horns are either machined inside the radiometer 20K module or connected via external electro-formed bended waveguides. The requirement of high stability of the reference signal imposed a careful design for the radiometric and thermal properties of the loads. Materials used for the manufacturing have been characterized for thermal, RF and mechanical properties. Abstract

The Lugano classification for response assessment in lymphoma recommends the use of the 5-point-scale Deauville Score (DS) to assess response evaluation of end-of-treatment FDG-PET/CT (eotPET) in Hodgkin Lymphoma (HL); nevertheless, there is a paucity of data on its accuracy and reproducibility. We focus here on the cohort of advanced stage IIb-IV HL patients enrolled in the HD0607 clinical trial (NCT identifier 00795613) that having had a negative interim PET performed 6 cycles of ABVD (Doxorubicin, Vinblastine, Vincristine and Dacarbazine) and then performed an eotPET. Negative patients were randomized to radiotherapy and no further treatment while positive patients were treated based on local policies. eotPET was re-evaluated independently by two readers evaluated and progression free survival was analysed (PFS). eotPET of 254 patients were analysed. The median follow-up was 43 months. The best receiver operator characteristics cut-off values to distinguish positive and negative patients was 4. The area-under-the-curve was 0.81 (95%CI, 0.70–0.91). Three-years PFS was 0.95 (95% CI 0.90–0.97) in eotPET negative and 0.22 (95% CI 0.11–0.43) in eotPET positive. DS demonstrated a good reproducibility of positivity/negativity between the readers consensus and local site evaluation where the agreement occurred on 95.0% of patients. The present study demonstrates that eotPET is an accurate tool to predict treatment outcome in HL and confirms the appropriateness of the Lugano classification for eotPET evaluation.

Abstract

Consumer awareness of food nutritional properties has increased significantly. A healthy diet is now viewed as the first step towards the prevention of many age-related chronic diseases such as cancer and heart disease. In most cases oxidative damage is involved in the onset of these diseases, much of which can be prevented by a diet high in antioxidants. However, throughout hundreds of years of domestication, many crop species, such as lettuce, have been genetically manipulated with the purpose of increasing favourable traits. These selective processes have resulted in the unintentional decrease of the antioxidant nutritional content of species such as Lactuca sativa. This project aimed to use natural resources for the enhancement of lettuce nutritional properties through the introgression of genes from a close relative of L. sativa. Initially, the total antioxidant content of baby lettuce leaves was investigated, with particular focus on carotenoids, chlorophyll and phenolic compounds. Wild lettuce (L. serriola, acc. UC96US23) was found to produce higher levels of total antioxidants, chlorophylls and carotenoids than the lettuce cultivated variety, Salinas cv. Subsequently, these traits were analysed in a Recombinant Inbred Line mapping population obtained by crossing these two lettuce species. Considerable transgressive segregation was observed and this data was then used to map Quantitative Trait Loci (QTL) on the population, which showed an additive effect of both parents contributing to these beneficial traits. Subsequent analyses of key genes involved in the synthesis of carotenoids and phenolic compounds revealed the presence of single nucleotide polymorphisms. Understanding the processes involved in the synthesis of nutritional properties and identifying the genes underlying these traits is key to producing a better quality lettuce.
