2. Formative Studies of First Year of the Competency-Based Field Center for Teacher Education at Hamburg, New York.

Abstract

This report describes the first-year progress of the Fredonia/Hamburg Teacher Education Center and is concerned with aspects of the general format, program, and organization of the center. Chapter one defines and discusses the development of competency-based programs and describes the purpose of the Teacher Education Center at Fredonia/Hamburg. Chapter two discusses the rationale, organization, and planning of the center. Also described is the progression from traditional student teaching to the internship. Chapter three evaluates the teacher education center according to three major concerns: classroom instruction, organization, and professional development of the interns. Chapter four makes eight recommendations concerning the establishment of a program committee, the value base of the program, competencies and associated activities, and staff responsibilities. Chapter five is an epilogue, briefly commenting on the second year of the program. A 17-item bibliography and three appendixes are included. (PD)

3. The Value of Selected Measures of Personality Characteristics as Predictors of College Achievement.

Abstract

The aim of this investigation was to determine whether the most effective prediction of freshman college achievement could be obtained through using raw scores from the Dogmation Scale, Gough-Sanford Rigidity Scale, California F Scale, and the Canadian Opinionation Scale or through using a set of factor scores extracted from a combination of the above four as predictors of grade point average. The four measures were administered to 738 freshmen. A group of 140 cases were then selected, 50 for cross validation purposes and 90 to develop the necessary regression equations. Results showed that neither the factor scores nor the raw scores from the measures proved to be strong predictors of academic performance as represented by grade point average. From the results of this study it can be concluded that only the raw scores for the four psychological tests are significantly related to academic achievement of college freshmen. The three factors which were extracted may be of some interest in relationship to student activism. (KJ)

Abstract

The PREDICTS project—Projecting Responses of Ecological Diversity In Changing Terrestrial Systems (www.predicts.org.uk)—has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity.

Abstract

Biodiversity continues to decline in the face of increasing anthropogenic pressures such as habitat destruction, exploitation, pollution and introduction of alien species. Existing global databases of species’ threat status or population time series are dominated by charismatic species. The collation of datasets with broad taxonomic and biogeographic extents, and that support computation ofa range of biodiversity indicators, is necessary to enable better understanding of historical declines and to project – and avert – future declines. We describe and assess a new database of more than 1.6 million samples from 78 countries representing over 28,000 species, collated from existing spatial comparisons of local-scale biodiversity exposed to different intensities and types of anthropogenic pressures, from terrestrial sites around the world. The database containsmeasurements taken in 208 (of 814) ecoregions, 13 (of 14) biomes, 25 (of 35) biodiversity hotspots and 16 (of 17) megadiverse countries. The database contains more than 1% of the total number of all species described, and more than 1% of the described species within many taxonomic groups – including flowering plants, gymnosperms, birds, mammals, reptiles, amphibians, beetles, lepidopterans and hymenopterans. The dataset, which is still being added to, is therefore already considerably larger and more representative than those used by previous quantitative models of biodiversity trends and responses. The database is being assembled as part of the PREDICTS project (Projecting Responses of Ecological Diversity In Changing Terrestrial Systems – www.predicts.org.uk).

Abstract

Context: Oculopharyngeal muscular dystrophy (OPMD) is a rare myopathy caused by polyalanine triplet repeat expansion in the gene for poly(A) binding protein 2 (PABP2) and is found in isolated cohorts throughout the world. We have observed numerous cases of OPMD in New Mexico.Objective: To characterize the clinical, genetic, and demographic features of the OPMD population in New Mexico.Design, Setting, and Participants: Cohort study with analysis of outpatient clinic medical records from 1965 to 2001 at the University of New Mexico Hospital and the New Mexico VA Health Care System in Albuquerque, which serve the entire state.Main Outcome Measures: Clinical phenotype, supplemented with genetic confirmation (n = 10 patients) and in-depth clinical evaluations (n = 49 patients).Results: We identified 216 cases of OPMD (99 women and 117 men) from 39 kindreds of New Mexicans spanning up to 4 generations. All patients were Hispanic, and the majority of probands came from northern New Mexico. In patients who had both ocular and pharyngeal muscle weakness, ptosis was just as likely to occur before or concurrent with dysphagia. Proximal limb muscle weakness and gait abnormalities were common and occurred later than ocular or pharyngeal weakness. The clinical expression of OPMD caused marked debility, although life-table analysis showed no decrease in life expectancy compared with unaffected family members (P =.81). Ten individuals from different kindreds were found to have an identical polyalanine triplet repeat expansion ([GCG](9)) in the PABP2 gene.Conclusions: Individuals in this cohort had clinical and genetic characteristics of classic OPMD. Longevity was not affected, but patients experienced considerable morbidity. The origin of the PABP2 mutation in New Mexican OPMD patients is unclear, although the geographic and genetic isolation of northern New Mexicans with a long ancestry in this region may have contributed to the development of this cohort. This disease cohort represents a large and previously unrecognized health care issue in the state of New Mexico and should serve to raise the awareness of this disorder among clinicians who treat Hispanics in the Southwest and throughout the United States. Abstract

Components of the pRb/p16/cyclin D1/CDK4 pathway are frequent targets in numerous tumour types, including those of pituitary origin. However, previous studies of pituitary tumours have examined individual components of this pathway. Therefore, to determine their overall contribution we have simultaneously examined the immunohistochemical status of pRb, p16 and cyclin D1 and analysed the CDK4 gene for a characterized activating mutation. Of the total pituitary tumour cohort (29 clinically non-functioning adenomas and 16 somatotrophinomas) abnormal expression of either pRb, p16 or cyclin D1 was observed in 36 of 45 (80%) tumours and was significantly (P = 0.005) associated with non-functioning tumours (27/29; 93%) compared with somatotrophinomas (9/16, 56%). Loss of either pRb or p16 expression was mutually exclusive in 23 of 45 (51%) tumours, whilst concomitant loss of pRb and p16 expression was observed in five tumours. Cyclin D1 overexpression was observed in 22 of 45 (49%) tumours, however, there was no significant association between overexpression of cyclin D1 and the expression status of either pRb or p16. In addition, no activating mutations within codon 24 of the CDK4 gene were detected. This study provides evidence for the first time that components of the pRb/p16/cyclin D1/CDK4 pathway, either alone or in combination, are frequently deregulated in human pituitary tumours, suggesting that this pathway may be a useful target in drug or gene therapeutic approaches.

41. Nucleic Acid Homologies Among Species of Saccharomyces

Abstract

Evolutionary divergence among species of the yeast genus Saccharomyceswas estimated from measurements of deoxyribonucleic acid (DNA)/DNA and ribosomal ribonucleic acid (RNA)/DNA homology. Much diversity was found in the DNA base sequences with several species showing little or no homology to the three reference species, S. cerevisiae, S. lactis, and S. fragilis. These three reference species also showed little or no homology to each other. On the other hand the diversity among ribosomal RNA base sequences was small since most species showed a high degree of homology to the reference species. The arrangement of species based on ribosomal RNA homologies agrees in most cases with current taxonomic groupings. A yeast hybrid (S. fragilis× S. lactis) was shown to contain two nonhomologous genomes. A minimum genome size of 9.2 × 109daltons for S. cerevisiaewas calculated from the rate of DNA renaturation.

Abstract

The behaviour of positive streamers in fields near and above the stability field has been studied in quasi-uniform fields of up to 18cm in extent with the aim of producing a model of the behaviour so that predictions can be made for much greater distances. In particular the evolution in air of free charge produced by the propagation has been monitored and the effect of pressure and propagation distance on this evolution studied. A frequency Gamma or coefficient gamma has been defined and measured which characterises the growth of free charge. The Schockley-Ramo theorem has been used to analyse the data which not only provides values for other relevant parameters such as attachment coefficient and streamer velocity but also provides a novel way of observing prebreakdown streamer modes. A model has been constructed which adequately accounts for the observed behaviour in small gaps but which predicts unrealistically large amounts of free charge in large gaps (1m). This suggests that some other mechanism might predominate in this case; an increase in electron density leading to channel thermalisation is suggested as a possibility.
