China has a complex and diverse forest ecological environment, which breeds abundant forest macrofungi, including some edible, medicinal, and poisonous species. During the investigations of macrofungi in the Saihanba National Nature Reserve, North China, we collected abundant specimens of Agaricales and Polyporales within the Agaricomycetes. Based on the morphological characters and molecular evidence of DNA sequences including the internal transcribed spacer (ITS) regions, the large subunit of nuclear ribosomal RNA gene (nLSU), the small subunit of mitochondrial rRNA gene (mtSSU), the small subunit of nuclear ribosomal RNA gene (nuSSU), the largest subunit of RNA polymerase II (RPB1), the second largest subunit of RNA polymerase II gene (RPB2), the β-tubulin gene (TUB), and the translation elongation factor 1-α gene (TEF1), this study identifies ten species of Agaricales and Polyporales new to science, viz. Cyanosporus subpopuli, Gelatinofungus betulina, Lycoperdon pseudoperlatum, Macrocystidia hebeiensis, Mycena subbrunnea, M. subpura, M. variispora, M. violocea-ardesiaca, Picipes griseus, and Pleuroflammula hebeiensis. Detailed morphological descriptions, fruiting bodies, and microscopic structure diagrams of these ten novel species are provided.

Abstract Soil organic carbon (SOC) persistence is predominantly governed by mineral protection, consequently, soil mineral-associated (MAOC) and particulate organic carbon (POC) turnovers have different impacts on the vulnerability of SOC to climate change. Here, we generate the global MAOC and POC maps using 8341 observations and then infer the turnover times of MAOC and POC by a data-model integration approach. Global MAOC and POC storages are $${975}_{964}^{987}$$ 975 964 987 Pg C (mean with 5% and 95% quantiles) and $${330}_{323}^{337}$$ 330 323 337 Pg C, while global mean MAOC and POC turnover times are $${129}_{45}^{383}$$ 129 45 383 yr and $${23}_{5}^{82}$$ 23 5 82 yr in the top meter, respectively. Climate warming-induced acceleration of MAOC and POC decomposition is greater in subsoil than that in topsoil. Overall, the global atlas of MAOC and POC turnover, together with the global distributions of MAOC and POC stocks, provide a benchmark for Earth system models to diagnose SOC-climate change feedback.

Abstract The sensitivity of soil organic carbon (SOC) decomposition in seasonally frozen soils, such as alpine ecosystems, to climate warming is a major uncertainty in global carbon cycling. Here we measure soil CO2 emission during four years (2018–2021) from the whole-soil warming experiment (4 °C for the top 1 m) in an alpine grassland ecosystem. We find that whole-soil warming stimulates total and SOC-derived CO2 efflux by 26% and 37%, respectively, but has a minor effect on root-derived CO2 efflux. Moreover, experimental warming only promotes total soil CO2 efflux by 7-8% on average in the meta-analysis across all grasslands or alpine grasslands globally (none of these experiments were whole-soil warming). We show that whole-soil warming has a much stronger effect on soil carbon emission in the alpine grassland ecosystem than what was reported in previous warming experiments, most of which only heat surface soils.

Abstract With the increase in the number of cases of autoimmune encephalitis (AE), the cerebrospinal fluid (CSF) of people living with HIV (PLWH) showing abnormal behavior, cognitive impairment or abnormal movements should be actively screened for the antibody panel of AE. Early recognition and treatment can prevent severe seizures or coma and markedly improve the prognosis of patients. The first-line immunotherapy for AE includes intravenous methylprednisolone and immunoglobulin. However, whether long-time immunosuppressive maintenance therapy is needed is debated. For PLWH, immunosuppressive therapy and even steroids could be more challenging. Here, we review and summarize the clinical characteristics often reported cases and report one case from our center to improve the diagnosis and treatment of anti-N-methyl-d-aspartate receptor encephalitis in PLWH.

Abstract Introduction Elvitegravir/cobicistat/emtricitabine/tenofovir alafenamide (E/C/F/TAF) has been increasingly replaced by bictegravir/emtricitabine/tenofovir alafenamide (B/F/TAF) in the treatment of human immunodeficiency virus (HIV) owing to its more favorable pharmacokinetics and fewer drug–drug interactions. However, the effect of this switch on plasma lipids and lipidomic profiles remains poorly characterized. Methods HIV infected patients on an E/C/F/TAF regimen were recruited into the study and followed up every 12 weeks. Participants were divided into E/C/F/TAF and B/F/TAF groups depending on whether they were switched to B/F/TAF during follow-up. Clinical information and blood samples were collected at 0, 12, and 24 weeks, and lipidomic analysis was performed using liquid chromatography mass spectrometry. Results No significant differences were observed between the groups at baseline. At week 24, patients switched to B/F/TAF had lower triglyceride [mmol/L; 1.23 (0.62) versus 2.03 (0.75), P = 0.001] and very low-density lipoprotein cholesterol [mmol/L; 0.64 (0.26) versus 0.84 (0.32), P = 0.037) compared with patients who continued E/C/F/TAF therapy. Small decrease from baseline in Framingham general cardiovascular risk score (FRS) was observed in the B/F/TAF arm [week (W) 0: 2.59 (1.57) versus W24: 2.18 (1.01), P = 0.043]. Lipidomic analysis indicated that E/C/F/TAF treatment increased the levels of several diglycerides (DGs), triacylglycerols (TAGs), and lyso-phosphatidylcholines (LPCs), whereas switching to B/F/TAF led to increased sphingolipids and glycerophospholipids. After adjusting for demographic and clinical parameters, only DG (16:0/18:2), DG (18:2/22:6), DG (18:3/18:2), DG (20:5/18:2), TAG (18:3/18:2/21:5), TAG (20:5/18:2/22:6), and LPC (22:6) were found to be significantly associated with FRS (regression coefficient of 0.17–6.02, P

Glucosinolates (GSLs) are a group of nitrogen- and sulfur-containing secondary metabolites, synthesized primarily in members of the Brassicaceae family, that play an important role in food flavor, plant antimicrobial activity, resistance to insect attack, stress tolerance, and human anti-cancer effects. As a sulfur-containing compound, glutathione has a strong connection with GSLs biosynthesis as a sulfur donor or redox system, and exists in reduced (glutathione; GSH) and oxidized (glutathione disulfide; GSSG) forms. However, the mechanism of GSH regulating GSLs biosynthesis remainds unclear. Hence, the exogenous therapy to pakchoi under normal growth condition and sulfur deficiency condition were conducted in this work to explore the relevant mechanism. The results showed that exogenous application of buthionine sulfoximine, an inhibitor of GSH synthesis, decreased the transcript levels of GSLs synthesis-related genes and transcription factors, as well as sulfur assimilation-related genes under the normal growth condition. Application of exogenous GSH inhibited the expression of GSLs synthesis- and sulfur assimilation-related genes under the normal condition, while the GSLs biosynthesis and the sulfur assimilation pathway were activated by exogenous application of GSH when the content of GSH in vivo of plants decreased owing to sulfur deficiency. Moreover, exogenous application of GSSG increased the transcript levels of GSLs synthesis- and sulfur assimilation-related genes under the normal growth condition and under sulfur deficiency. The present work provides new insights into the molecular mechanisms of GSLs biosynthesis underlying glutathione regulation.

Introduction: CoronaVac, an inactivated vaccine developed by Sinovac Life Sciences, has been widely used for protection against Coronavirus Disease 2019 (COVID-19). This study investigates its effect on the HIV reservoir and T cell repertoires in people living with HIV (PLWHs). Methods: Blood samples were collected from fifteen PLWHs who were administered at least two doses of CoronaVac between April 2021 and February 2022. The levels of cell-associated HIV RNA (CA HIV RNA) and HIV DNA, as well as the T cell receptor (TCR) repertoire profiles, TCR clustering and TCRβ annotation, were studied. Results: A significant increase was observed in CA HIV RNA at 2 weeks (431.5 ± 164.2 copies/106 cells, P = 0.039) and 12 weeks (330.2 ± 105.9 copies/106 cells, P = 0.019) after the second dose, when compared to the baseline (0 weeks) (73.6 ± 23.7 copies/106 cells). Abstract Background Despite the relationship between Adverse childhood experiences (ACEs) and depressive symptoms, which has been well researched in general populations, little is known about homosexual and bisexual populations, especially lesbian and bisexual women in China. This study aims to investigate the prevalence of ACEs and depressive symptoms and to analyze the relationship between them among lesbian and bisexual women in China. Methods The eligible participants were aged 16 years or older who report their sexual orientation as homosexual or bisexual. The data was collected through anonymous questionnaires with the help of Lespark in Beijing from July 18 to December 29, 2018, and all participants had informed consent to this study. Univariate analysis and multiple linear regression analyses were performed to explore the relationship between ACEs and depressive symptoms among lesbian and bisexual women. All statistical analyses were conducted by the software of SPSS 22.0. Results Among 301 lesbian and bisexual women, 81.4% were lesbian, 18.4% were bisexual women, and the majority were 21–30 years. As for ACEs, 51.5% reported at least one ACE, in which emotional neglect (22.6%) and emotional abuse (22.3%) were common ACEs. As for depressive symptoms of lesbian and bisexual women, the detection rate was 56.1%. The multiple linear regression analyses showed that abuse (β = 2.95, 95%CI:1.07–4.83) and neglect (β = 3.21, 95%CI:1.09–5.31) were positively associated with depressive symptoms and lesbian and bisexual women with three (β = 4.11, 95%CI: 0.99–7.22) or more (β = 6.02, 95%CI: 3.23–8.78) ACEs suffered from more depressive symptoms than others. Conclusion Adverse childhood experiences (ACEs) and depressive symptoms were at high prevalence among lesbian and bisexual women in China. ACEs were associated with depressive symptoms, especially childhood abuse and neglect experiences that have a significant effect on lesbian and bisexual women mental health.

Abstract To investigate the occurrence and 90-day mortality of cancer patients following unplanned admission to the intensive care unit (ICU), as well as to develop a risk prediction model for their 90-day prognosis. We prospectively analyzed data from cancer patients who were admitted to the ICU without prior planning within the past 7 days, specifically between May 12, 2021, and July 12, 2021. The patients were grouped based on their 90-day survival status, and the aim was to identify the risk factors influencing their survival status. A total of 1488 cases were included in the study, with an average age of 63.2 ± 12.4 years. The most common reason for ICU admission was sepsis (n = 940, 63.2%). During their ICU stay, 29.7% of patients required vasoactive drug support (n = 442), 39.8% needed invasive mechanical ventilation support (n = 592), and 82 patients (5.5%) received renal replacement therapy. We conducted a multivariate COX proportional hazards model analysis, which revealed that BMI and a history of hypertension were protective factors. On the other hand, antitumor treatment within the 3 months prior to admission, transfer from the emergency department, general ward, or external hospital, high APACHE score, diagnosis of shock and respiratory failure, receiving invasive ventilation, and experiencing acute kidney injury (AKI) were identified as risk factors for poor prognosis within 90 days after ICU admission. The average length of stay in the ICU was 4 days, while the hospital stay duration was 18 days. A total of 415 patients died within 90 days after ICU admission, resulting in a mortality rate of 27.9%. We selected 8 indicators to construct the predictive model, which demonstrated good discrimination and calibration. The prognosis of cancer patients who are unplanned transferred to the ICU is generally poor. Assessing the risk factors and developing a risk prediction model for these patients can play a significant role in evaluating their prognosis.

In bone tissue engineering, scaffolds with excellent mechanical and bioactive properties play prominent roles in space maintaining and bone regeneration, attracting increasingly interests in clinical practice. In this study, strontium-incorporated β-tricalcium phosphate (β-TCP), named Sr-TCP, bioceramic triply periodic minimal surface (TPMS) structured scaffolds were successfully fabricated by digital light processing (DLP)-based 3D printing technique, achieving high porosity, enhanced strength, and excellent bioactivity. The Sr-TCP scaffolds were first characterized by element distribution, macrostructure and microstructure, and mechanical properties. Notably, the compressive strength of the scaffolds reached 1.44 MPa with porosity of 80%, bringing a great mechanical breakthrough to porous scaffolds. Furthermore, the Sr-TCP scaffolds also facilitated osteogenic differentiation of mouse osteoblastic cell line (MC3T3-E1) cells in both gene and protein aspects, verified by alkaline phosphatase (ALP) activity and polymerase chain reaction (PCR) assays. Overall, the 3D-printed Sr-TCP bioceramic TPMS structured scaffolds obtained high porosity, boosted strength, and superior bioactivity at the same time, serving as a promising approach for bone regeneration.

Abstract Background The reversible maneuver that mimics the fluid challenge is a widely used test for evaluating volume responsiveness. However, passive leg raising (PLR) does have certain limitations. The aim of the study is to determine whether the supine transfer test could predict fluid responsiveness in adult patients with acute circulatory failure who do not have intra-abdominal hypertension, by measuring changes in cardiac index (CI). Methods Single-center, prospective clinical study in a 25-bed surgery intensive care unit at the Fudan University Shanghai Cancer Center. Thirty-four patients who presented with acute circulatory failure and were scheduled for fluid therapy. Every patient underwent supine transfer test and fluid challenge with 500 mL saline for 15–30 min. There were four sequential steps in the protocol: (1) baseline-1: a semi-recumbent position with the head of the bed raised to 45°; (2) supine transfer test: patients were transferred from the 45° semi-recumbent position to the strict supine position; (3) baseline-2: return to baseline-1 position; and (4) fluid challenge: administration of 500 mL saline for 15–30 min. Hemodynamic parameters were recorded at each step with arterial pulse contour analysis (ProAQT/Pulsioflex). A fluid responder was defined as an increase in CI ≥ 15% after fluid challenge. The receiver operating characteristic curve and gray zone were defined for CI. Results Seventeen patients were fluid challenge. The r value of the linear correlations was 0.73 between the supine transfer test- and fluid challenge-induced relative CI changes. The relative changes in CI induced by supine transfer in predicting fluid responsiveness had an area under the receiver operating characteristic curve of 0.88 (95% confidence interval 0.72–0.97) and predicted a fluid responder with 76.5% (95% confidence interval 50.1–93.2) sensitivity and 88.2% (95% confidence interval 63.6–98.5) specificity, at a best threshold of 5.5%. Nineteen (55%) patients were in the gray zone (CI ranging from -3 and 8 L/min/m2). Conclusion The supine transfer test can potentially assist in detecting fluid responsiveness in patients with acute circulatory failure without intra-abdominal hypertension. Nevertheless, the small threshold and the 55% gray zone were noteworthy limitation. Trial registration Predicting fluid responsiveness with supine transition test (ChiCTR2200058264). Abstract Our knowledge on the responses of permafrost ecosystems to climate warming is critical for assessing the direction and magnitude of permafrost carbon‐climate feedback. However, most of the previous experiments have only been able to warm the air and surface soil, with limited effects on the permafrost temperature. Consequently, it remains challenging to realistically simulate permafrost thawing in terms of increased active layer (a layer freezing and thawing seasonally above permafrost) thickness under climate warming scenarios. Here, we presented the experimental design and warming performance of a novel experiment, Simulate Warming at Mountain Permafrost (SWAMP), the first one to successfully simulate permafrost warming and the subsequent active layer deepening in a swamp meadow situated on the Tibetan Plateau. Infrared heating was employed as above‐ground warming to elevate the temperature of the air and surface soil, and heating rods were inserted vertically in the soil to provide below‐ground warming for transmitting heat to the deep active layer and even to permafrost deposits. In 3 m diameter warmed circular plots, the air and the entire soil profile (from surface soil to 120 cm) was effectively heated, with an increase of approximately 2°C in the upper 60 cm, which progressively weakened with soil depth. Warming increased soil moisture across the growing season by inducing an earlier thawing of the soil. Values varied from 1.8 ± 1.8 to 12.3 ± 2.3% according to the soil depth. Moreover, during the growing season, the warmed plots had greater thaw depths and a deeper active layer thickness of 12.6 ± 0.8 cm. In addition, soil thawing duration was prolonged by the warming, ranging from 22.8 ± 3.3 to 49.3 ± 4.5 days depending on the soil depth. The establishment of SWAMP provides a more realistic simulation of warming‐induced permafrost thaw, which can then be used to explore the effect of climate warming on permafrost ecosystems and the potential permafrost carbon‐climate feedback. Notably, our experiment is more advantageous for investigating how deep soil processes respond to climate warming and active layer deepening, compare with experiments which use passive warming techniques such as open top chambers (OTCs).

Abstract Whether bone marrow modulates systemic metabolism remains unknown. Our recent study suggested that myeloid-derived growth factor (MYDGF) improves insulin resistance. Here, we found that myeloid cell-specific MYDGF deficiency aggravated hepatic inflammation, lipogenesis, and steatosis, and show that myeloid cell-derived MYDGF restoration alleviated hepatic inflammation, lipogenesis, and steatosis. Additionally, recombinant MYDGF attenuated inflammation, lipogenesis, and fat deposition in primary mouse hepatocytes (PMHs). Importantly, inhibitor kappa B kinase beta/nuclear factor-kappa B (IKKβ/NF-κB) signaling is involved in protection of MYDGF on non-alcoholic fatty liver disease (NAFLD). These data revealed that myeloid cell-derived MYDGF alleviates NAFLD and inflammation in a manner involving IKKβ/NF-κB signaling, and serves as a factor involved in the crosstalk between the liver and bone marrow that regulates liver fat metabolism. Bone marrow functions as an endocrine organ and serves as a potential therapeutic target for metabolic disorders.

Brassicaceae vegetables are an important traditional daily food in China and around the world, which provide nutrients and phytochemicals that are beneficial for human health. Among them, Brassica and Raphanus are widely cultivated and eaten, have been evolved and/or bred for special characteristics during the long history of cultivation. Epidemiological studies suggest that the health benefits of Brassicaceae vegetables are mainly associated with glucosinolates (GSLs) and their hydrolytic products. In this review, we discuss the diversity of common consumed Brassicaceae vegetables and their GSL composition in edible parts. We also discuss the diversity factors affecting GSL content, and the diversity roles and functions of GSL. The information in this review provides guidance for consumers to select vegetables with a high GSL content, optimum edible stages, suitable edible methods, and provides a theoretical basis for crop molecular breeding and market development of GSL products.

Abstract Background Patients infected with HIV are at high risk of developing Epstein-Barr Virus (EBV)-related diseases. The genotype and viral biological behavior of EBV infection in patients with human immunodeficiency virus-1 (HIV) in China remain unclear. This study analyzed the characteristics of EBV in patients infected with HIV in southeastern China. Methods A total of 162 HIV-infected patients and 52 patients without HIV were enrolled in this study. EBV viral load in blood was determined by fluorescence quantitative PCR. EBV typing was performed using saliva according to polymorphisms in the EBNA3C region. EBV LMP-1 carboxy terminus (C-ter) was sequenced, and compared with the epidemic strains in the world. Results Among HIV infected patients, the EBV strain variant was mainly EBV-1, while EBV-2 had a higher viral load than EBV-1 (P = 0.001) and EBV-1/2 (P = 0.002). HIV infected patients had higher active virus replication. The EBV LMP-1 variants were mainly the China1 variant. HIV-infected patients had different nucleic acid positions of 30-bp deletion (del30) and had a higher incidence of high 33-bp tandem repeats (rep33) copies than non-HIV-infected patients. Abstract Background Hepatocellular carcinoma (HCC) is one of the most common malignant cancers in humans and has a high fatality rate. Despite pharmacological advances such as sorafenib and lenvatinib approval, responses are seen only in a limited fraction of HCCs, and the majority of HCC patients do not benefit from this treatment. In recent years, researchers have verified that the long noncoding RNAs (lncRNAs) impact the efficiency of lenvatinib and the prognosis of patients with HCC. Materials and methods This work obtained gene expression profile from an Arraystar lncRNA microarray. Expression of HOTAIRM1, Beclin-1, and p62 in HCC was characterized in clinical HCC tissues of 24 patients with HCC. Overexpression and knockdown experiments were performed in HCC cells to examine the effects of the HOTAIRM1 on lenvatinib sensitivity. The interactions between HOTAIRM1, miR-34a and Beclin-1 were predicted according to GSEA and CNC network. The effects of HOTAIRM1, autophagy and lenvatinib on tumor inhibit were validated in orthotopic tumor-bearing nude mouse model. Results Lenvatinib-resistant HCC cell lines were established using the concentration gradient method. Data from an Arraystar lncRNA microarray indicated that HOTAIRM1, a specific lncRNA located in an evolutionarily highly conserved HOX gene cluster, was differentially expressed between lenvatinib-resistant HCC cells and their parental cells. Expression of HOTAIRM1 and Beclin-1 in HCC was characterized in clinical HCC tissues of 24 patients who have different sensitivity to lenvatinib. Knocking down of HOTAIRM1 decreased the autophagy level in lenvatinib-resistant HCC cells and increased their sensitivity to lenvatinib, especially when combined with autophagy inhibitors both in vitro and in vivo. Further study indicated that knocking down HOTAIRM1 in lenvatinib-resistant cell lines increased the level of miR-34a and inhibited the expression of Beclin-1 in Huh7-R and HepG2-R cells. Investigation according to GSEA and CNC network, lncRNA and nearby coding gene and lncRNA-miRNA analyses demonstrated that the resistance of HCC to lenvatinib was affected by the HOTAIRM1-miR-34a-Beclin-1 regulatory axis. Conclusion HOTAIRM1 is an independent drug resistance factor which significantly associated with the efficacy of lenvatinib in HCC. HOTAIRM1 may downregulation of miR-34a and upregulation of Beclin-1, leading to activation of autophagy, thereby inducing lenvatinib resistance in HCC.

Carrimycin is a potential immune-regulating agent for sepsis in patients with tumors. In this study, we investigated its effects on inflammation and immune function in tumor patients with sepsis. In total, 120 participants were randomized to receive either carrimycin treatment (400 mg/day) (n = 62) or placebo (n = 58) for 7 days. The primary outcomes were immune-related indicators. Subsequently, patients were stratified into two subgroups (CD4 < 38.25% and CD8 < 25.195%). Ninety-nine participants were analyzed: 47 and 52 in the carrimycin and placebo groups, respectively. HLA-DR levels were rapidly increased in the carrimycin group; however, the placebo group initially experienced a decline in HLA-DR level at 1 day after administration. In the subgroup with CD4

The presence of biodegradable microplastics (MPs) has the potential to affect soil pH, and possibly accelerate or inhibit the loss of soil inorganic carbon (SIC) in calcareous soils. However, most researchers have focused on the release of biotic carbon dioxide (CO2) from soils following MP amendments, and few studies have investigated SIC-derived CO2. In this experiment, three typical biodegradable MPs were applied to three calcareous soils amended with 1 % 13C-labeled (99 % atom) carbonate, and the release of CO2 originating from SIC was quantified. The total CO2 emissions, soil pH, and microbial functional genes involved in soil nitrification and denitrification were also detected. Throughout the experiment, the contribution of 13C-labeled carbonate to total CO2 emissions ranged from 0.42 % to 3.31 %. The impact of biodegradable MPs on SIC-derived CO2 varied with incubation period. At the early stage (≤20 days), the amendment of three biodegradable MPs increased the abiotic CO2 in some cases, and the CO2 emissions from 13C-labeled SIC were positively correlated with the total CO2 originating from the decomposition of SOC and MPs. At the late stage (20–70 days), the presence of biodegradable MPs inhibited the release of CO2 from 13C-labeled carbonate in most treatments. Moreover, there were negative relationships of SIC-derived CO2 with soil pH and the amoA gene of ammonia-oxidizing archaea (AOA), but positive correlations of SIC-derived CO2 with amoA of ammonia oxidizing bacteria (AOB) and nirK and nirS genes encoding nitrate reductase in denitrification. Our results indicate that long-term exposure to biodegradable MPs probably regulates the release of H+ in the nitrification process by controlling AOB, and then controlling the dynamics of SIC in calcareous soils.

Abstract Long-term observations have shown that many plants and aboveground animals have changed their phenology patterns due to warmer temperatures over the past decades. However, empirical evidence for phenological shifts in alpine organisms, particularly belowground organisms, is scarce. Here, we investigate how the activities and phenology of plants, soil microbes, and soil fauna will respond to warming in an alpine meadow on the Tibetan Plateau, and whether their potential phenological changes will be synchronized. We experimentally simulate an increase in soil temperature by 2–4 °C according to future projections for this region. We find that warming promotes plant growth, soil microbial respiration, and soil fauna feeding by 8%, 57%, and 20%, respectively, but causes dissimilar changes in their phenology during the growing season. Specifically, warming advances soil faunal feeding activity in spring and delays it in autumn, while their peak activity does not change; whereas warming increases the peak activity of plant growth and soil microbial respiration but with only minor shifts in their phenology. Such phenological asynchrony in alpine organisms may alter ecosystem functioning and stability.

The structure and function of plant communities in alpine meadow ecosystems are potentially susceptible to climate warming. Here, we utilized a unique field manipulation experiment in an alpine meadow on the Qinghai-Tibetan Plateau and investigated the responses of plant species diversity, composition, biomass, and net primary productivity (NPP) at both community and functional group levels to whole-soil-profile warming (3–4 °C across 0–100 cm) during 2018–2021. Plant species diversity, biomass and NPP (both above- and belowground) at the community level showed remarkable resistance to warming. However, plant community composition gradually shifted over time. Over the whole experimental warming period, aboveground biomass of legumes significantly decreased by 45%. Conversely, warming significantly stimulated aboveground biomass of forbs by 84%, likely because of better growth and competitive advantages from the warming-induced stimulation of soil water and other variables. However, warming showed minor effects on aboveground biomass of grasses and sedges. Overall, we emphasize that experimental warming may significantly affect plant community composition in a short term by triggering adjustments in plant interspecific competition or survival strategies, which may cause potential changes in plant productivity over a more extended period and lead to changes in carbon source-sink dynamics in the alpine meadow ecosystem.

Abstract Background Patients with human immunodeficiency virus-associated cryptococcal meningitis (HIV-CM) have persistent intracranial inflammation despite negative cerebrospinal fluid (CSF) fungal cultures after optimal treatment for CM, which could be devastating for the central nervous system. However, a definitive treatment strategy for persistent intracranial inflammation despite optimal antifungal therapies is undefined. Methods We identified 14 HIV-CM patients with persistent intracranial inflammation and conducted a 24-week, prospective, interventional study. All participants received lenalidomide (25 mg, p.o.) on days 1 to 21 of a 28-day cycle. Follow-up lasted for 24 weeks with visits at baseline and weeks 4, 8, 12, and 24. The primary endpoint was the change in clinical manifestations, routine CSF parameters, and MRI findings after lenalidomide treatment. An exploratory analysis was made on changes in cytokine levels in CSF. Safety and efficacy analyses were undertaken in patients who received at least one dose of lenalidomide. Results Of 14 participants, 11 patients completed the 24 weeks of follow-up. Rapid clinical remission following lenalidomide therapy was observed. Clinical manifestations (fever, headache, altered mentation) were reversed fully by week-4 and remained stable during follow-up. A significant reduction in white blood cell (WBC) count in CSF was noted occurred at week-4 (P = 0.009). The median protein concentration in CSF decreased from 1.4 (0.7–3.2) g/L at baseline to 0.9 (0.6–1.4) at week-4 (P = 0.004). The median albumin concentration in CSF decreased from 79.2 (48.4–149.8) mg/L at baseline to 55.3 (38.3–89.0) mg/L at week-4 (P = 0.011). The WBC count, protein level, and albumin level in CSF remained stable and approached a normal range through week-24. There was no significant change in immunoglobulin-G, intracranial pressure (ICP), or chloride-ion concentration at each visit. Brain MRI demonstrated multiple lesions to be absorbed post-therapy. Levels of tumor necrosis factor-α granulocyte colony stimulating factor, interleukin (IL)-6, and IL-17A decreased significantly during 24-week follow-up. Two (14.3%) patients had mild skin rash, which resolved spontaneously. Lenalidomide-related serious adverse events were not observed. Conclusion Lenalidomide could improve persistent intracranial inflammation in HIV-CM patients significantly and was well tolerated without serious adverse events observed. And the additional randomized controlled study is required to further validate the finding.

Nitrogen (N) deposition rates of terrestrial ecosystems have gradually declined but are still high in some areas. Previous studies have reported that N addition elicits diverse impacts on soil organic carbon (SOC) pools. SOC can be divided into different functional fractions, namely, particulate organic carbon (POC) and mineral-associated organic carbon (MAOC). The responses of these fractions to N addition should be elucidated to better understand the changes in SOC pools. Here, we conducted a N addition experiment (0, 40, and 80 kg N ha−1 yr−1) in a subtropical Castanopsis fabri forest to simulate N deposition. The surface (0−10 cm) SOC fractions, aboveground litter product, fine root (diameter < 2 mm) biomass, soil exchangeable cation content, and soil enzyme activity under different N addition treatments were measured. The results showed the following: (1) N addition showed a positive effect on POC and SOC contents but did not significantly affect MAOC content; (2) POC content was negatively correlated with pH and soil enzyme activity and positively correlated with aboveground litter product, suggesting that POC accumulation was influenced by aboveground litter input and microbial decomposition; (3) a close negative relationship was observed between exchangeable Al3+ and Ca2+ or K+ contents, indicating that there is likely to be a trade-off between the mineral sorption and desorption, thus resulting in an insignificant reaction of MAOC to N addition. Overall, the accumulation of SOC under short-term N addition was found to be primarily driven by POC, and the response of different SOC functional fractions to N addition was inconsistent. By incorporating these nuances into ecosystem models, it is possible to predict SOC dynamics more accurately in response to global change.

Since November 2021, Omicron has emerged as the dominant severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variant, and its sublineages continue to appear one after another, significantly reducing the effectiveness of existing therapeutic neutralizing antibodies (NAbs). It is urgent to develop effective NAbs against circulating Omicron variants. Here, we isolated receptor binding domain (RBD)-specific single memory B cells via flow cytometry from a COVID-19 convalescent. The antibody variable region genes of the heavy chain (VHs) and light chain (VLs) were amplified and cloned into expression vectors. After antibody expression, ELISA screening and neutralizing activity detection, we obtained an IGHV3-53-encoded RBD-targeting cross-neutralizing antibody D6, whose VL originated from the IGKV1-9*01 germlines. D6 could potently neutralize circulating Omicron variants (BA.1, BA.2, BA.4/5 and BF.7), with IC50 values of less than 0.04 μg/mL, and the neutralizing ability against XBB was reduced but still effective. The KD values of D6 binding with RBD of the prototype and BA.1 were both less than 1.0 × 10−12 M. The protein structure of the D6-RBD model indicates that D6 interacts with the RBD external subdomain and belongs to the RBD-1 community. The sufficient contact and deep interaction of D6 HCDR3 and LCDR3 with RBD may be the crucial reason for its cross-neutralizing activity. The sorting and analysis of mAb D6 will provide important information for the development of anti-COVID-19 reagents.

The Lower Cambrian Niutitang Formation in the Northern Guizhou harbors abundant organic-rich mud shale, constituting the most significant marine shale gas reservoir in Guizhou. In this article, the reservoir characteristics of Lower Cambrian Niutitang Formation in Northern Guizhou are analyzed in terms of lithology, mineralogy, organic geochemistry, pore structure, gas content and continuous thickness of shale, and the exploration potential of shale gas in this area is evaluated. The results indicate that the content of brittle minerals in the shale of well QX1 is 65.29% to 95.22% (average of 82.10%). The total organic carbon (TOC) content ranges from 2.06% to 12.10% (average of 5.64%). The organic matter maturity (Ro) within the range of 2.29–2.67%, and the kerogen type is identified as type I. The shale samples from the Niutitang Formation have high TOC content, suitable thermal maturity, and a favorable kerogen type, suggesting good gas generation potential. The results of scanning electron microscopy (SEM) show that intergranular pores, intragranular pores and microfractures are developed in the shale of well QX1, which can provide sufficient storage space for shale gas. The shale exhibits a continuous thickness of 105.66 m in the QX1 well, comprising a gas-bearing interval of 32.89 m at the top (with an effective continuous thickness of 18 m) and a hydrocarbon source rock layer of 75.78 m at the bottom. In comparison with other shale gas regions, Niutitang Formation shale in Northern Guizhou exhibits characteristics such as favorable gas generation conditions, greater storage conditions, excellent gas-bearing, strong frackability, and substantial continuous thickness, it has greater potential for shale gas exploration.

Soil dissolved organic matter (DOM) is highly sensitive to external interference. However, little attention has been paid on how soil DOM characteristics in the dry period affect the warming response of soil organic carbon (SOC) decomposition in riparian ecosystems under flooding conditions. The temperature sensitivity (Q10) of SOC decomposition was determined by incubating 99 riparian soils under flooding across 11 transects of the Three Gorges Reservoir. Soil DOM characteristics in the dry period were measured by ultraviolet–visible spectroscopy (i.e., a254, a350, SUVA254 and SR) and 3D-fluorescence spectroscopy (such as FI, β:α, BIX, and HIX). The fluorescence components of C1, C3, and C4 were determined by parallel factor analysis. SOC decomposition was sensitive to warming with a higher Q10 value for CH4 (2.34 ± 1.30) and lower for CO2 (1.37 ± 0.23) under flooding. The soil DOM in dry period was mainly autochthonous with FI > 1.9 and 90% of BIX > 0.7. The humification of soil DOM was relatively weak at ∼ 95% of HIX

Background: The significance of hepatitis B virus (HBV) in cerebrospinal fluid (CSF) is unclear. Methods: Synchronous serum and CSF samples were collected from 13 patients. HBV DNA, full-length genome, quasispecies, phylogenetic tree, compartmentalization and mutation of the reverse transcriptase (RT) region were performed based on PCR and sequencing methods. Results: HBV DNA was detected in the CSF of 3 antiviral-naïve individuals and 1 individual after successful antiviral therapy. Complete full-length HBV genomes were isolated from the CSF of 5 individuals, including 2 with undetectable serum HBV DNA. Ten individuals exhibited distinct CSF-serum quasispecies, 8 harbored independent CSF-serum genetic compartmentalization and phylogenetic trees, and 5 lamivudine/entecavir-associated resistance mutations only in the CSF. The frequencies of rtL180M and rtM204I/V mutations in both serum and CSF were higher in HIV-HBV-coinfected individuals than in the HBV-monoinfected ones (serum: rtL180M: 3.9% vs. 0, P = 0.004; rtM204I/V: 21.3% vs. 0, P

The response of soil organic carbon to climate warming may be soil depth-dependent, but remains unquantified in situ. Here the authors show that warming induces more proportional soil carbon losses in topsoil than in subsoil, particularly from high-latitudinal carbon-rich soils.

Background and purposeLower-grade gliomas (LGG) exhibit a wide range of metabolic pathway changes, and metabolic reprogramming can be largely seen as a result of oncogenic driving events. Glycolysis, an important pathway of tumor energy source, has been poorly studied in gliomas. The aim of this article is to analyze the relationship between glycolysis and lower-grade glioma development and prognosis in order to explore the heterogeneous relevance of glycolysis in lower-grade gliomas.Methods and resultsOur study searched the TCGA database and identified three glycolytic subtypes with significant prognostic differences by unsupervised clustering analysis of core glycolytic genes, named C1, C2, and C3. By analysis of clinical prognosis, somatic cell variation, and immune infiltration, we found that C3 had the best prognosis with molecular features of IDHmut-codel, followed by C1 with major molecular features of IDHmut-non-codel, G -CIMP high subtype, while C2 had the worst prognosis, mainly exhibiting IDHwt, G-CIMP low and mesenchymal-like subtypes with seven important CNV features, including CDKN2A/B deletion, chr7 gain and chr10 deletion, chr19/20 co-gain, EGFR amplification and PDGFRA/B deletion phenotypes were significantly increased, with the highest level of stemness and significant T-cell depletion features. Finally, to quantify the level of abnormal glycolysis and its impact on prognosis, we developed GlySig to reflect the glycolytic activity of LGG and integrated molecular features to construct nomogram that can be independently assessed to predict prognosis.ConclusionsOur study analyzed the tumor characteristics of different glycolytic states, and our findings explain and describe the heterogeneity of glycolytic metabolism within diffuse LGGs.

IntroductionThe novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic has been posing a severe threat to global public health. Although broadly neutralizing antibodies have been used to prevent or treat corona virus disease 2019 (COVID-19), new emerging variants have been proven resistant to these antibodies.MethodsIn this study, we isolated receptor binding domain (RBD)-specific memory B cells using single-cell sorting method from two COVID-19 convalescents and expressed the antibody to test their neutralizing activity against diverse SARS-CoV-2 variants. Then, we resolved antibody-RBD complex structures of potent RBD-specific neutralizing antibodies by X-ray diffraction method. Finally, we analyzed the whole antibody repertoires of the two donors and studied the evolutionary pathway of potent neutralizing antibodies.Results and discussionWe identified three potent RBD-specific neutralizing antibodies (1D7, 3G10 and 3C11) from two COVID-19 convalescents that neutralized authentic SARS-CoV-2 WH-1 and Delta variant, and one of them, 1D7, presented broadly neutralizing activity against WH-1, Beta, Gamma, Delta and Omicron authentic viruses. The resolved antibody-RBD complex structures of two antibodies, 3G10 and 3C11, indicate that both of them interact with the external subdomain of the RBD and that they belong to the RBD-1 and RBD-4 communities, respectively. From the antibody repertoire analysis, we found that the CDR3 frequencies of the light chain, which shared high degrees of amino acid identity with these three antibodies, were higher than those of the heavy chain. This research will contribute to the development of RBD-specific antibody-based drugs and immunogens against multiple variants.

The correlation between runoff and sediment challenges ecological preservation and sustainable development in the Yellow River Basin. An understanding of the key factors influencing variations in runoff and sediment transport in crucial river basins is essential for effective soil erosion management within the context of ecological and economic development. The Mann–Kendall test, Pettitt test, and Morlet wavelet analysis were employed in the Wuding River Basin to analyze the trends in runoff and sediment changes from 1960 to 2020. We explored the double cumulative curve method to assess the contribution rates of precipitation and human activities to the variability of runoff and sediment transport. We explored the primary factors driving the changes in runoff and sediment transport through random forest regression analysis. (1) From 1960 to 2020, annual precipitation in the Wuding River Basin increased minimally, while annual runoff and sediment transport decreased strongly with abrupt changes. Abrupt changes in annual runoff and sediment transport occurred in 1971 and 1979, respectively. (2) The relationship between runoff and sediment transport changed in approximately 1972 and 2000. The distribution of monthly runoff became more uniform during Periods II (1973–2001) and III (2002–2020) compared to that during the baseline period (1960–1972, Period I), while sediment transport became increasingly concentrated in the flood season. (3) During Period II, the contribution rates of climate and human activities to runoff and sediment transport were 11.94% and −14.5%, respectively, compared to the baseline period. During Period III, the contribution rates of climate and human activities to runoff and sediment transport were −11.9% and −17.7%, respectively. Human activities substantially reduced runoff and sediment, with greater impacts on sediment reduction. Climate weakly influenced basin sediment transport variations. (4) The normalized difference vegetation index (NDVI) and grassland area extent had the greatest impact on runoff, while the NDVI and forest area extent affected sediment transport.

β-glucan is the most abundant polysaccharide in the cell wall of Pneumocystis jirovecii, which has attracted extensive attention because of its unique immunobiological characteristics. β-glucan binds to various cell surface receptors, which produces an inflammatory response and accounts for its immune effects. A deeper comprehension of the processes by Pneumocystis β-glucan recognizes its receptors, activates related signaling pathways, and regulates immunity as required. Such understanding will provide a basis for developing new therapies against Pneumocystis. Herein, we briefly review the structural composition of β-glucans as a vital component of the Pneumocystis cell wall, the host immunity mediated by β-glucans after their recognition, and discuss opportunities for the development of new strategies to combat Pneumocystis.

Savolitinib, a small-molecule inhibitor of the receptor tyrosine kinase mesenchymal-epithelial transition (MET) factor, was approved for the treatment of non-small cell lung cancer (NSCLC) by the China National Medical Products Administration in June 2021. Its safety for NSCLC treatment has been confirmed in several prospective cohort studies. Herein, we report a rare case of shock, a serious adverse event, after treatment with savolitinib in an HIV-1–positive patient with advanced NSCLC. A 38-year-old man with an 8-year history of HIV-1 positivity was diagnosed with NSCLC 5 years ago; the lung cancer recurred after surgical resection. Despite chemotherapy, immunotherapy, and targeted therapy, tumor progression continued. He received savolitinib because of MET amplification. In the first 2 weeks of savolitinib use, he developed a mild rash on his trunk. In the following month, he was hospitalized for fever and circulatory shock thrice after taking savolitinib 400 mg. He had no urticaria or eosinophilia. During the three hospitalizations, he was negative for pathogens. His condition gradually improved after treatment with antibiotics, steroids, and vasopressors. Attention should be paid to the occurrence of septic shock-like presentations when using savolitinib in HIV-1 patients with NSCLC.

Abstract Background The impact of ventriculoperitoneal shunt on cerebrospinal fluid (CSF) biochemical profiles in HIV-associated cryptococcal meningitis (HCM) patients remains unclear. Methods Twenty-nine HCM patients who underwent ventriculoperitoneal shunt (the VPS group) and 57 HCM patients who did not undergo ventriculoperitoneal shunt (the non-VPS group) were enrolled in this propensity score matching analysis. Demographic characteristics, symptoms, CSF biochemical profiles, and adverse events were compared between the two groups. The Kaplan–Meier method was used to analyze the survival rate. Univariate and multivariate logistic regression analyses were performed to identify the risk factors for increased CSF protein levels. Results After 24 weeks of treatment, the intracranial pressure was significantly lower in the VPS group than in the non-VPS group (mmH2O; 155.0 [120.0–190.0] vs. 200.0 [142.5–290.0]; P = 0.025), and the rate of neuroimaging improvement was significantly higher in the VPS group (16/17 [94.1%] vs. 2/10 [20%]; P

This study has designed and developed a smart data glove based on five-channel flexible capacitive stretch sensors and a six-axis inertial measurement unit (IMU) to recognize 25 static hand gestures and ten dynamic hand gestures for amphibious communication. The five-channel flexible capacitive sensors are fabricated on a glove to capture finger motion data in order to recognize static hand gestures and integrated with six-axis IMU data to recognize dynamic gestures. This study also proposes a novel amphibious hierarchical gesture recognition (AHGR) model. This model can adaptively switch between large complex and lightweight gesture recognition models based on environmental changes to ensure gesture recognition accuracy and effectiveness. The large complex model is based on the proposed SqueezeNet-BiLSTM algorithm, specially designed for the land environment, which will use all the sensory data captured from the smart data glove to recognize dynamic gestures, achieving a recognition accuracy of 98.21%. The lightweight stochastic singular value decomposition (SVD)-optimized spectral clustering gesture recognition algorithm for underwater environments that will perform direct inference on the glove-end side can reach an accuracy of 98.35%. This study also proposes a domain separation network (DSN)-based gesture recognition transfer model that ensures a 94% recognition accuracy for new users and new glove devices.

Vaccination has proven to be highly effective against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), but the long-term immunogenicity and the functional preserved immune responses of vaccines are needed to inform evolving evidence-based guidelines for boosting schedules. We enrolled 205 healthcare workers into a cohort study; all had received three doses of BBIBP-CorV (China Sinopharm Bio-Beijing Company, Beijing, China) inactivated vaccine. We assessed SARS-CoV-2 specific binding antibodies, neutralizing antibodies, and peripheral T and B cell responses. We demonstrated that more robust antibody responses to SARS-CoV-2 were elicited by booster immunization compared with primary vaccination. Neutralizing antibody titers to SARS-CoV-2 Omicron BA.1 were also efficiently elevated post-homologous vaccine booster despite being in a lower titer compared with the prototype stain. In addition to S-specific humoral and cellular immunity, BBIBP-CorV also induced N-specific antibody and effector T cell responses. The third-dose vaccination led to further expansion of critical polyfunctional T cell responses, likely an essential element for vaccine protection. In particular, a functional role for Tfh cell subsets in immunity was suggested by the correlation between both CD4+ Tfh and CD8+ Tfh with total antibody, IgG, B cell responses, and neutralizing antibodies. Our study details the humoral and cellular responses generated by the BBIBP-CorV booster vaccination in a seven-month follow-up study. There is a clear immunologic boosting value of homologous inactivated SARS-CoV-2 vaccine boosters, a consideration for future vaccine strategies.

Turnip (Brassica rapa subsp. rapa) is a cruciferous plant cultivated worldwide that serves as a source of nutrients and bioactive compounds. Most turnip studies have focused on a few compounds or on part of the plant. The establishment of a complete chemical profile of different plant parts would facilitate its use for nutritional and medicinal purposes. In the current study, mineral elements, soluble sugars, free amino acids (FAA), total phenols (TP), total flavonoids (TF), and glucosinolates (GS) were quantified in the leaves, stems, and roots. Results were compared for 20 strains of turnip. The outcomes showed significant differences between parts of the plant and strains. The leaves exhibited the highest TF, TP, indispensable FAA, and microelement levels, and they showed a higher GS. Moreover, the stems had a high content of GS and macroelements. Furthermore, the roots showed high levels of free sugars and total FAA. The findings of this work provide the basis for utilizing each part of the turnip plant based on its chemical composition.

Background: The loss of soil organic carbon (SOC) following conversion of natural forests to managed plantations has been widely reported. However, how different SOC fractions and microbial necromass C (MNC) respond to forest management practices remains unclear. Methods: We sampled 0–10 ​cm mineral soil from three different management plantations and one protected forest in Guangxi, Southern China, to explore how forest management practices affect SOC through changing mineral-associated C (MAOC) and particulate organic C (POC), as well as fungal and bacterial necromass C. Results: Compared with the protected forest, SOC and POC in the abandoned, mixed and Eucalyptus plantations significantly decreased, but MAOC showed no significant change, indicating that the loss of SOC was mainly from decreased POC under forest management. Forest management also significantly reduced root biomass, soil extractable organic C, MNC, and total microbial biomass (measured by phospholipid fatty acid), but increased fungi-to-bacteria ratio (F:B) and soil peroxidase activity. Moreover, POC was positively correlated with root biomass, total microbial biomass and MNC, and negatively with F:B and peroxidase activity. These results suggested that root input and microbial properties together regulated soil POC dynamics during forest management. Conclusion: Overall, this study indicates that forest management intervention significantly decreases SOC by reducing POC in Guangxi, Southern China, and suggests that forest protection can help to sequester more soil C in forest ecosystems.

Abstract Oncolytic viruses have found a good place in the treatment of cancer. Administering oncolytic viruses directly or by applying genetic changes can be effective in cancer treatment through the lysis of tumor cells and, in some cases, by inducing immune system responses. Moreover, oncolytic viruses induce antitumor immune responses via releasing tumor antigens in the tumor microenvironment (TME) and affect tumor cell growth and metabolism. Despite the success of virotherapy in cancer therapies, there are several challenges and limitations, such as immunosuppressive TME, lack of effective penetration into tumor tissue, low efficiency in hypoxia, antiviral immune responses, and off‐targeting. Evidence suggests that oncolytic viruses combined with cancer immunotherapy‐based methods such as immune checkpoint inhibitors and adoptive cell therapies can effectively overcome these challenges. This review summarizes the latest data on the use of oncolytic viruses for the treatment of cancer and the challenges of this method. Additionally, the effectiveness of mono, dual, and triple therapies using oncolytic viruses and other anticancer agents has been discussed based on the latest findings.

Soil microbial resource limitation is an important indicator of the ecosystem function and process, especially the response mechanism of the ecosystem under global change. Known as an oasis in the degraded karst landscape, the microbial resource limitations of the karst tiankeng ecosystem are still unclear. In this study, the soil extracellular enzyme activities (EEAs) and stoichiometry (EES) related to carbon (C), nitrogen (N), and phosphorus (P) acquisition were assayed in karst tiankeng. Soil samples were collected from the sunny slope of tiankeng (TSU), the shady slope of tiankeng (TSH), the base of tiankeng (TB), and outside tiankeng (OT). The results showed that the soil enzymatic acquisition of C: N: P in karst tiankeng was deviated by 1: 1: 1, indicating that the stoichiometry of soil enzymes in karst tiankeng was in an unstable state. Soil microbial communities in karst tiankeng were limited by soil C and P, which were mainly achieved by altering soil nutrient stoichiometry. At the TSH site, soil microbes were minimally limited by C and P. The P-limitation was more evident outside karst tiankeng than inside karst tiankeng. These results showed that the unique habitat of karst tiankeng can alleviate microbial community P-limitation in karst land. This study highlights the characteristics of karst tiankeng ecosystems and their role in maintaining microbial survival and ecological function in karst fragile ecosystems.

Abstract Epidemiological studies have demonstrated that the genetic factors partly influence the development of same-sex sexual behavior, but most genetic studies have focused on people of primarily European ancestry, potentially missing important biological insights. Here, we performed a two-stage genome-wide association study (GWAS) with a total sample of 1478 homosexual males and 3313 heterosexual males in Han Chinese populations and identified two genetic loci (rs17320865, Xq27.3, FMR1NB, P meta = 8.36 × 10−8, OR = 1.29; rs7259428, 19q12, ZNF536, P meta = 7.58 × 10−8, OR = 0.75) showing consistent association with male sexual orientation. A fixed-effect meta-analysis including individuals of Han Chinese (n = 4791) and European ancestries (n = 408,995) revealed 3 genome-wide significant loci of same-sex sexual behavior (rs9677294, 2p22.1, SLC8A1, P meta = 1.95 × 10−8; rs2414487, 15q21.3, LOC145783, P meta = 4.53 × 10−9; rs2106525, 7q31.1, MDFIC, P meta = 6.24 × 10−9). These findings may provide new insights into the genetic basis of male sexual orientation from a wider population scope. Furthermore, we defined the average ZNF536-immunoreactivity (ZNF536-ir) concentration in the suprachiasmatic nucleus (SCN) as lower in homosexual individuals than in heterosexual individuals (0.011 ± 0.001 vs 0.021 ± 0.004, P = 0.013) in a postmortem study. In addition, compared with heterosexuals, the percentage of ZNF536 stained area in the SCN was also smaller in the homosexuals (0.075 ± 0.040 vs 0.137 ± 0.103, P = 0.043). More homosexual preference was observed in FMR1NB-knockout mice and we also found significant differences in the expression of serotonin, dopamine, and inflammation pathways that were reported to be related to sexual orientation when comparing CRISPR-mediated FMR1NB knockout mice to matched wild-type target C57 male mice.

BackgroundHemophagocytic lymphohistiocytosis (HLH) is a fatal immunological syndrome resulting from excessive production of inflammatory cytokines. The conventional therapies for HLH, which are based on cytotoxic agents, are not always efficacious and safe, especially in patients with severe immunodeficiency. Ruxolitinib, a strong inhibitor of Janus kinase (JAK) 1/2, has already been evaluated as salvage and first-line therapy for HLH. Despite its promising efficacy and tolerability in the treatment of secondary HLH, the efficacy and safety of ruxolitinib in HLH patients with HIV infection remain to be investigated.Case presentationTwo men (ages: 45 and 58 years) both presented at our hospital with a high fever. They were found to be HIV-positive with severe immunodeficiency and opportunistic infections. Their laboratory tests showed severe pancytopenia, hypofibrinogenemia, hypertriglyceridemia, and increased levels of inflammatory factors and ferritin. Hemophagocytosis was found in the bone marrow, and abdominal computed tomography or ultrasonography showed splenomegaly. Both patients were diagnosed with infection-induced HLH due to severe immunodeficiency. Given they were both highly immunocompromised, we chose ruxolitinib as a first-line treatment alternative to cytotoxic chemotherapy. Rapid remission of clinical symptoms and normalization of laboratory parameters were achieved after ruxolitinib therapy. Neither patient had any associated adverse drug reactions or other laboratory abnormalities. Both patients were eventually discharged and ruxolitinib was discontinued as their disease alleviated, and they did not show signs of relapse during the 3- and 5-month of follow-up examinations.ConclusionWe described two cases of AIDS-related secondary HLH treated with ruxolitinib. Our cases highlight the feasibility of using ruxolitinib as a first-line therapy in patients with HIV infection and secondary HLH. Nevertheless, the safety and efficacy of this novel treatment need to be evaluated in large clinical trials in the future.

Plant residue input plays a vital role in cropland carbon (C) balance, which can be greatly affected by climate change. Our knowledge of the effect of plant residue on soil respiration is crucial for evaluating C exchange between the atmosphere and terrestrial ecosystems, but large uncertainties remain in the effect of plant input on the temperature sensitivity (Q10) of soil organic C (SOC) decomposition in cropland ecosystems. Here, soils were sampled from two bare fallow plots (including a Mollisol and an Alfisol with different climate and land-use history) and their adjacent old field plots, and were incubated at 10 °C and 20 °C for 815 days. The ‘equal-time’, ‘equal-C’, ‘one-pool model’ and ‘two-pool model’ methods were used to evaluate the Q10 of SOC decomposition. Results indicated that in the early stage of incubation, exclusion of plant input increased the Q10 (Alfisol) or had minor effect on the Q10 (Mollisol), which may be related to the interactions between substrate quality decrease (increase of chemical stability) and clay mineral suppression on microbial decomposition. However, in the later stage of incubation, plant residue removal decreased the Q10 values in both soils possibly due to the limitation of substrate availability on microbial decomposition. Overall, the role of plant input in the temperature sensitivity of SOC decomposition should be considered when predicting SOC stocks in a future warmer world.

IntroductionImmune therapy has ushered in a new era of tumor treatment, at the expense of immune-related adverse events, including rare but fatal adverse cardiovascular events, such as myocarditis. Steroids remain the cornerstone of therapy for immune-related myocarditis, with no clear consensus on additional immunosuppressive treatment for steroid-refractory cases yet.Case reportHere, we report a patient with stage IV nasopharyngeal carcinoma who developed immune-related myocarditis in the fourth course of therapy with immune checkpoint inhibitors. The patient presented with precordial discomfort with elevation of cardiac enzymes and interleukin-6, atypical electrocardiographic abnormalities, and reduced left ventricular ejection fraction. Coronary computed tomography angiography excluded the possibility of acute coronary syndrome. The therapy with tofacitinib targeting the Janus kinase-signal transducer and activator of transcription signal pathway was successfully conducted, since there was no significant improvement in troponin under high-dose steroid and intravenous immunoglobulin treatment. The patient recovered without major adverse cardiac events during hospitalization.DiscussionThe safety and efficacy of tofacitinib in a patient with steroid-refractory immune-related myocarditis were investigated, hoping to provide a basis for prospective therapeutic strategies. Tofacitinib led to remarkable remissions in primary autoimmune disease by blocking the inflammatory cascade, indicating its potential therapeutic use in immune-related adverse events.

In a locale-scale mountainous forest, changes in slope aspect can cause microclimate heterogeneity, which can impact both soil environment and plant community, and influence the soil microbial communities as well. However, the relationship between the aboveground plant community and belowground soil microbial communities and the change in slope aspect is not well understood. A case study was carried out in a subalpine coniferous forest in western China to investigate the above- and belowground relationship of three slope aspects, the north-facing slope, the flat site and the south-facing slope. The plant community attributes were evaluated by the community survey, considering species diversity and the plant total basal area of trees and shrubs to represent the plant productivity. Soil bacteria were determined based on 16S rRNA gene and soil fungi on a nuclear ribosomal internal transcribed spacer (ITS) using high-throughput Illumina sequencing. The results showed that slope aspects significantly affect the aboveground plant productivity and diversity, and the community composition and structure of both aboveground plants and soil bacteria and soil fungi. Soil bacteria and soil fungi correlated differently with aboveground plant community changes in the slope aspects in terms of diversity and community composition and structure. A structural equation model (SEM) revealed that slope aspects caused by aboveground plant productivity changes correlated significantly with the soil fungal community composition and structure, while the soil bacterial community was rather weakly correlated with the plant community, and its changes in community composition and structure were mainly affected by the soil properties and soil fungal community. Further studies considering plant functional traits, soil microbial functional groups, and seasonal changes may reveal a deeper correlation between the aboveground plants and belowground soil microbials at the local scale in subalpine coniferous forests.

Compositional, functional, and nutritional properties are important for the use-value assessments of wild and cultivated edible plants. The aim of this study was to compare the nutritional composition, bioactive compounds, volatile compounds, and potential biological activities of cultivated and wild Zingiber striolatum. Various substances, such as soluble sugars, mineral elements, vitamins, total phenolics, total flavonoids, and volatiles, were measured and analyzed using UV spectrophotometry, ICP-OES, HPLC, and GC-MS methods. The antioxidant capacity of a methanol extract of Z. striolatum, as well as the hypoglycemic abilities of its ethanol and water extracts, were tested. The results showed that the contents of soluble sugar, soluble protein, and total saponin in the cultivated samples were higher, while the wild samples contained higher amounts of K, Na, Se, vitamin C, and total amino acids. The cultivated Z. striolatum also showed a higher antioxidant potential, while the wild Z. striolatum exhibited a better hypoglycemic activity. Thirty-three volatile compounds were identified using GC-MS in two plants, with esters and hydrocarbons being the main volatile compounds. This study demonstrated that both cultivated and wild Z. striolatum have a good nutritional value and biological activity, and can be used as a source of nutritional supplementation or even in medication.
