Its main symptoms, occurring early during pathology development, are skin dehydration, redness, erythema, and telangiectasia. Given the lack of a resolutive cure, therapeutic approaches able to relieve the main symptoms are needed.Purpose: The aim of this research article is to evaluate the beneficial effect of a topical product (Serum BK46) on rosacea symptoms.Patients and Methods: A monocentric single-arm, non-blinded study was performed to assess the clinical effect of Serum BK46 in relieving the main symptoms of rosacea: skin dryness, increased trans epidermal water loss (TEWL), redness, and abnormal vascularization. Twenty patients with mild to moderate rosacea were enrolled in the study and asked to apply the product twice per day for 56 days. Skin moisturization, TEWL, and erythema index were instrumentally assessed at baseline and following 24 h and 14, 28 and 56 days of treatment. Clinical parameters, including redness and telangiectasia imperfection visibility, were evaluated on a 5-point scale by a specialized dermatologist at baseline and after 14, 28, and 56 days of treatment. Finally, the visibility of vessel diameter was evaluated at baseline and after 28 and 56 days of treatment.Results: Serum BK46 application restored skin hydration and prevented the loss of water by the skin. Long-term treatment with Serum BK46 significantly reduced skin redness, erythema index, and the visibility of telangiectasia imperfections and superficial vessels. The investigated product’s clinical effect was demonstrated by both instrumental and clinical evaluation. Furthermore, Serum BK46 was completely tolerated and no adverse effects were recorded.Conclusion: The moisturizing and skin barrier restoring action of Serum BK46 has been clearly proven in patients displaying mild to moderate rosacea; thus, this product is a good candidate for rosacea treatment.Keywords: rosacea, topical product, transepidermal water loss, skin redness, telangiectasia

Embryonic-type neuroectodermal tumor (ENT) is a somatic-type malignancy characterized by overgrowth of embryonic-type neuroectodermal tissue (EtNT). In germ cell tumors, EtNT is frequently intermingled with other components that may exhibit significant morphologic overlap [mature neuro-glial tissue (MNGT), nephroblastomatous tissues, and primitive endodermal-type glands]. Therefore, the quantification of EtNT (crucial for the diagnosis of ENT) can be challenging. In this study, we investigated the immunohistochemical profile of ENT, EtNT, and MNGT using a broad immunohistochemical panel. We found that SOX2 was the most sensitive marker for EtNT (100%), but it also stained MNGT (28.6%). GFAP and S100 were relatively sensitive (71.4%) and highly specific (GFAP 100%, S100 85.8%) for MNGT, whereas synaptophysin stained both. Combining our results with those of previous studies, we propose that a combination of SOX11, SOX2, GFAP, S100, AFP, villin, CDX2, PAX8, and nuclear WT1 may help to identify and quantify EtNT in germ cell tumors., (© 2024. This article presents a novel probabilistic forecasting method called ensemble conformalized quantile regression (EnCQR). EnCQR constructs distribution-free and approximately marginally valid prediction intervals (PIs), which are suitable for nonstationary and heteroscedastic time series data. EnCQR can be applied on top of a generic forecasting model, including deep learning architectures. EnCQR exploits a bootstrap ensemble estimator, which enables the use of conformal predictors for time series by removing the requirement of data exchangeability. The ensemble learners are implemented as generic machine learning algorithms performing quantile regression (QR), which allow the length of the PIs to adapt to local variability in the data. In the experiments, we predict time series characterized by a different amount of heteroscedasticity. The results demonstrate that EnCQR outperforms models based only on QR or conformal prediction (CP), and it provides sharper, more informative, and valid PIs.

Latin American and the Caribbean regions (LAC) harbor one of the most biodiverse areas of the world, the Neotropics. True bugs (Hemiptera: Heteroptera) are a diverse lineage of insects, with more than 45,000 species, particularly speciose in the Neotropical region. True bugs are fundamental in the dynamics of natural and modified ecosystems, with several species critical to agriculture and public health. We compiled Heteroptera research in LAC from 1998-2022 using bibliographic databases. Productivity, collaborative networks, and the main topics studied were analyzed. A total of 1,651 Heteroptera studies from LAC were found, with continuous growth being 2021 the most prolific. Four categories (Taxonomy of extant species, Faunistic inventories and new records, Pest species biology, and Community ecology) represent most of the published research. About 60 percent of the records evaluated correspond to five families (Pentatomidae, Reduviidae, Coreidae, Miridae, and Rhyparochromidae). We emphasize the need to keep working on Heteroptera taxonomy because it will allow further advances in other areas such as phylogenetic analyses, biogeography, ecology, and natural history, among others. The results of our analyses characterize the current state of heteropterology in the region, establishing a baseline for future studies and efforts to broaden the knowledge of the group.

Many recent works in the field of graph machine learning have introduced pooling operators to reduce the size of graphs. In this article, we present an operational framework to unify this vast and diverse literature by describing pooling operators as the combination of three functions: selection, reduction, and connection (SRC). We then introduce a taxonomy of pooling operators, based on some of their key characteristics and implementation differences under the SRC framework. Finally, we propose three criteria to evaluate the performance of pooling operators and use them to investigate the behavior of different operators on a variety of tasks.

The 2022 WHO classification of urinary and male genital tumors introduced several novel kidney entities exhibiting eosinophilic/oncocytic features with specific mutational backgrounds. Thus, molecular techniques, such as next-generation sequencing (NGS), became more commonly used for their evaluation. We studied 12 low-grade oncocytic tumors (LOT) of the kidney (from 11 patients), identified in a cohort of 210 eosinophilic/oncocytic renal tumors, diagnosed in our institution between October 2019 and May 2023, which represented 5.7% (12/210) of all eosinophilic/oncocytic renal tumors during this period. We reviewed their clinicopathologic, histologic, and immunohistochemical features, as well as their mutational profiles. We also reviewed the literature on NGS-derived data of LOT, by selecting papers in which LOT diagnosis was rendered according to the criteria proposed initially. Median age was 65 years (mean: 63.5; range 43-79) and median tumor size was 2.0 cm (mean: 2.2; range: 0.9-3.1). All tumors were positive for PAX8, CK7, and GATA3, and negative or focally positive for CD117/KIT. We found the following gene mutations: MTOR ((6/11), 54.5%)), TSC1 ((2/11), 18.2%)), and 1 had both NOTCH1 and NOTCH4 ((1/11), 9.1%)). Wild-type status was found in 2/11 (18.2%) patients and one tumor was not analyzable. A review of 8 previous studies that included 79 LOTs revealed frequent mutations in the genes that regulate the mammalian target of rapamycin (mTOR) pathway: MTOR (32/79 (40.5%)), TSC1 (21/79 (26.6%)), and TSC2 (9/79 (11.4%)). Other mutated genes included PIK3CA, NF2, and PTEN, not typically known to affect the mTOR pathway, but potentially acting as upstream and downstream effectors. Our study shows that LOT is increasingly diagnosed in routine practice when applying the appropriate diagnostic criteria. We also confirm that the mTOR pathway is strongly implicated in the pathogenesis of this tumor mainly through MTOR, TCS1, and TSC2 mutations, but other genes could also be involved in the pathway activation, especially in LOTs without "canonical" mutations., (© 2023. DNA Barcoding is an important tool for disciplines such as taxonomy, phylogenetics and phylogeography, with Barcode of Life Data System (BOLD) being the largest database of partial cytochrome c oxidase subunit I (COI) sequences. We provide the first extensive revision of the information available in this database for the insect order Thysanoptera, to assess: how many COI sequences are available; how representative these sequences are for the order; and the current potential of BOLD as a reference library for specimen identification and species delimitation. The COI database at BOLD currently represents only about 5% of the over 6400 valid thrips species, with a heavy bias towards a few species of economic importance. Clear Barcode gaps were observed for 24 out of 33 genera evaluated, but many outliers were also observed. We suggest that the COI sequences available in BOLD as a reference would not allow for accurate identifications in about 30% of Thysanoptera species in this database, which rises to 40% of taxa within Thripidae, the most sampled family within the order. Thus, we call for caution and a critical evaluation in using BOLD as a reference library for thrips Barcodes, and future efforts should focus on improving the data quality of this database.

Aims: Yolk sac tumour postpubertal-type (YSTpt) shows a wide range of histological patterns and is challenging to diagnose. Recently, forkhead box transcription factor A2 (FoxA2) emerged as a driver of YSTpt formation and a promising marker for diagnosing YSTpt. However, FoxA2 has not been tested in the different patterns of YSTpt. This study aimed to assess the staining pattern of FoxA2 in te different patterns of YSTpt and other germ cell tumours of the testis (GCTT), comparing it with glypican-3 (GPC3) and α-fetoprotein (AFP)., Methods and Results: FOXA2, GPC3 and AFP immunohistochemistry was performed on 24 YSTpt (24 microcystic/reticular, 10 myxoid, two macrocystic, five glandular/alveolar, two endodermal sinus/perivascular, four solid, two polyembryoma/embryoid body and two polyvesicular vitelline) and 81 other GCTT. The percentage of positive cells (0, 1+, 2+, 3+) and the intensity (0, 1, 2, 3) were evaluated regardless of and within each YSTpt pattern. FoxA2 was positive in all YSTpt (24 of 24) and all but one (23 of 24) exhibited 2+/3+ stain, with higher intensity [median value (mv): 2.6] than AFP (1.8) and GPC3 (2.5). Both FoxA2 and GPC3 were positive in all microcystic/reticular (24 of 24), myxoid (10 of 10), macrocystic (two of two), endodermal sinus/perivascular (four of four) and polyembryoma/embryoid body (two of two) patterns. Nevertheless, only FoxA2 was positive in all glandular/alveolar (five of five), solid (four of four) and polyvesicular vitelline (two of two) patterns. The intensity of FoxA2 was higher than AFP and GPC3 in almost all YST patterns. In the other GCTT, FoxA2 was positive only in teratoma postpubertal-type (Tpt) [13 of 20 (65%)], with staining almost exclusively confined to the mature gastrointestinal/respiratory tract epithelium., Conclusions: FoxA2 is a highly sensitive and specific biomarker that supports the diagnosis of YSTpt. FoxA2 is superior to GPC3 and AFP, especially in rare and difficult-to-diagnose histological patterns of YSTpt, but mature glands of Tpt could represent a potential diagnostic pitfall., (© 2023 John Wiley & Sons Ltd.)

Background: In recent years, several studies focused on the process of reprogramming of seminoma (S) cells, which regulates the transition from pure S (P-S) to S component (S-C) of mixed germ cell tumors of the testis (GCTT) and finally to embryonal carcinoma (EC) and other nonseminomatous GCTT (NS-GCTT). The accepted pathogenetic model is driven and regulated by cells (macrophages, B- and T-lymphocytes) and molecules of the tumor microenvironment (TME). Herein, we tested a series of GCTT with double staining (DS) for CD68-PD-L1 to evaluate tumor-associated macrophages (TAMs) expressing programmed death-ligand 1 (PD-L1) [TAMs PD-L1(+)] and clarify if these cells may be involved in establishing the fate of GCTT., Methods: We collected 45 GCTT (comprising a total of 62 different components of GCTT). TAMs PD-L1(+) were evaluated with three different scoring systems [TAMs PD-L1(+)/mm 2 , TAMs PD-L1(+)/mm 2 H-score, TAMs PD-L1(+) %], and compared using pertinent statistic tests (Student's t-test and Mann-Whitney U test)., Results: We found that TAMs PD-L1(+) values were higher in S rather than EC (p = 0.001, p = 0.015, p = 0.022) and NS-GCTT (p < 0.001). P-S showed statistically significant differences in TAMs PD-L1(+) values compared to S-C (p < 0.001, p = 0.006, p = 0.015), but there were no differences between S-C and EC (p = 0.107, p = 0.408, p = 0.800). Finally, we found statistically significant differences also in TAMs PD-L1(+) values between EC and other NS-GCTT (p < 0.001)., Conclusions: TAMs PD-L1(+) levels gradually decrease during the reprogramming of S cells {P-S [(high values of TAMs PD-L1(+)] → S-C and EC [(intermediate values of TAMs PD-L1(+)] → other NS-GCTT [(low values of TAMs PD-L1(+)], supporting a complex pathogenetic model where the interactions between tumor cells and TME components [and specifically TAMs PD-L1(+)] play a key role in determining the fate of GCTT., Competing Interests: Declaration of Competing Interest All the authors present in his article declare no conflict of interests., (Copyright © 2023 Elsevier GmbH. Taxonomic researchers have used multiple sources of evidence to support species hypotheses and delimitations. Grammostola Simon (Mygalomorphae: Theraphosidae) comprises 20 valid species endemic to South America, six occurring in Brazil. The classical morphological approach based mainly on genitalia may be misleading in recognizing species in this genus. Thus, we used morphology, geographical distribution, genetic distance, and phylogeny to support the redescription of Grammostola pulchra from southern Brazil, a species described a century ago. We also diagnosed and illustrated the species. Males have a developed apical keel at the apex of the embolus; for the first time, this type of structure has been reported in a species of Grammostola . The molecular analyses using the partial sequence of Cytochrome c oxidase subunit I showed 7% of genetic distance ( p -distance) between G. pulchra and Grammostola anthracina . Distance and tree-based methods (ASAP and bPTP, respectively) assigned G. pulchra as a valid species. The gene-tree under Bayesian and Maximum-Likelihood recovered a similar topology, placing G. pulchra as closely related to Grammostola burzaquensis and G. anthracina . Morphological characters which could be important in the taxonomy of the genus are further discussed.

Background: Lymphovascular invasion (LVI) is a relevant prognostic factor in germ cell tumors of the testis (GCTT), and it is included in the pT stage. However, its detection on hematoxylin and eosin (H&E) slides is very challenging, and previous studies reported fair to moderate inter-observer agreement among dedicated uropathologists. In the present study, we tested H&E and a recently developed in-house double staining for OCT4/CD34 to detect LVI in GCTT., Methods: Nine authors [5 non-uropathologists and 4 uropathologists] independently evaluated 34 consecutive and retrospectively enrolled cases of GCTT. We assessed the inter-observer agreement (Fleiss's Kappa) with both H&E and OCT4/CD34. Besides, we compared the consensus diagnosis on both H&E and OCT4/CD34-stained sections with the original diagnosis to evaluate the pT re-staging (McNemar test) and identify the sources of disagreement., Results: The inter-observer agreement among uropathologists plus non-uropathologists was fair with both H&E (KF=0.398; p < 0.001) and OCT4/CD34 (KF=0.312; p < 0.001). OCT4/CD34 (KF=0.290; p < 0.001) slightly reduces the inter-observer agreement compared to H&E (KF=0.321; p < 0.001) for non-uropathologists; in contrast, OCT4/CD34 (KF=0.293; p < 0.001) significantly reduces the inter-observer agreement compared to H&E (KF=0.529; p < 0.001) for uropathologists, changing it from moderate to fair. Consensus diagnosis with H&E modified the LVI status of the original diagnosis in 8/34 (23.5 %) cases (p: 0.070), with pT re-staging in 2/34 (5.9 %) cases (p: 0.500). Consensus diagnosis with OCT4/CD34 modified the LVI status of the original diagnosis in 8/34 (23.5 %) cases (p: 0.289), with pT re-staging in 3/34 (8.8 %) cases (p: 0.250). The consensus diagnosis with OCT4/CD34 modified the consensus diagnosis with H&E in 8/34 (23.5 %) cases (p: 0.727), and these findings resulted in pT-restaging in 3/34 (8.8 %) cases (p: 0.500). The sources of disagreement among uropathologists were: H&E [artefactual clefts misinterpreted as LVI in 4/6 (66.7 %) cases and true foci of LVI misinterpreted as clusters of histiocytes within the vessels in 2/6 (33.3 %) cases], OCT4/CD34 [artefactual clefts misinterpreted as LVI in 2/8 (25 %) cases, true LVI misinterpreted as artefactual clefts in 2/8 (25 %) cases or floaters in 4/8 (50 %) cases]., Conclusions: OCT4/CD34 does not improve the inter-observer agreement for the assessment of LVI in OCT4(+) GCTT. Consensus diagnosis with H&E modifies the LVI status in a significant number of cases, resulting in changes of the pT stage in a relatively small subgroup. Consensus diagnosis with OCT4/CD34 provides little additional benefit since it cannot exclude mimickers of LVI such as floaters and artefactual clefts. The recently described SWI/SNF complex-deficient sinonasal carcinoma (SMARCB1 & SMARCA4) may exhibit a yolk sac-like morphology. Tumors with similar features (yolk sac-like histology combined with the immunohistochemical loss of SMARCB1/INI1 and/or SMARCA4/BRG1) have also been described in other sites, such as the female genital tract. In this study, we immunohistochemically assessed SMARCB1/INI1 and SMARCA4/BRG1 expression to evaluate if these proteins could be involved in the pathogenesis of testicular yolk sac tumors of postpubertal type (YSTpt). Specifically, we analyzed a retrospective case series comprising pure YSTpt and mixed germ cell tumors of the testis (GCTT) with YSTpt components. In the present study, no testicular YSTpt showed loss of SMARCB1/INI1 (0/24, 0%) or SMARCA4/BRG1 (0/24, 0%). However, testicular choriocarcinoma (CHC) and isolated syncytiotrophoblast cells (iSTCs) demonstrated abnormal staining patterns for SMARCA4/BRG1 [CHC: 4/4 (100%); iSTCs: 12/12 (100%), respectively], including focal or diffuse loss of expression in a subset of cases. Otoliths (ear-stones) in the inner ears of vertebrates containing visible year zones are used extensively to determine fish age. Analysis of otoliths is a time-consuming and difficult task that requires the education of human experts. Human age estimates are inconsistent, as several readings by the same human expert might result in different ages assigned to the same otolith, in addition to an inherent bias between readers. To improve efficiency and resolve inconsistent results in the age reading from otolith images by human experts, an automated procedure based on convolutional neural networks (CNNs), a class of deep learning models suitable for image processing, is investigated. We applied CNNs that perform image regression to estimate the age of Greenland halibut (Reinhardtius hippoglossoides) with good results for individual ages as well as the overall age distribution, with an average CV of about 10% relative to the read ages by experts. In addition, the density distribution of predicted ages resembles the density distribution of the ground truth. By using k*l-fold cross-validation, we test all available samples, and we show that the results are rather sensitive to the choice of test set. Finally, we apply explanation techniques to analyze the decision process of deep learning models. Background: In recent years, several studies investigated the complex process called "reprogramming" of seminoma (S) cells. The accepted pathogenetic model is a complex network including SOX2, SOX17, OCT3/4 and PRAME, which modulates the epigenetic transcription of numerous downstream genes and drives a divergent gene expression profile resulting in the transition from pure S (P-S) to S component (S-C) of mixed germ cell tumors of the testis (M-GCTT), and finally to embryonal carcinoma (EC). Herein, we tested a large cohort of GCTT with SOX2 and PRAME to evaluate their expression in the evolutionary steps of GCTT and verify if the modulation in the expression of these two molecules could be relevant for the fate of GCTT., Methods: We tested 43, 19 and 17 consecutive and retrospectively enrolled cases of GCTT, germ cell neoplasia in situ (GCNIS) and uninvolved background testes (UBT), respectively. SOX2 and PRAME expressions have been evaluated with H-score and compared by adopting the appropriate statistic tests (Student's t-test and Mann-Whitney U test)., Results: We found that SOX2 was more expressed by nonseminomatous-GCTT (NS-GCTT) (p < 0.001) and EC (p < 0.001) rather than S; by contrast, PRAME showed an opposite expression profile being expressed by S but not by NS-GCTT (p < 0.001) and EC (p < 0.001). S-C showed different expressions of SOX2 and PRAME compared to both P-S (p = 0.002 and <0.001, respectively) and EC (p < 0.001 and 0.042, respectively), with intermediate values between these latter two categories. GCNIS and UBT showed no expression of SOX2 (scattered positive Leydig cells) but high H-score levels of PRAME., Conclusions: SOX2 and PRAME are differentially expressed and specularly modulated during the "reprogramming" of S cells [P-S (high levels of PRAME, no expression/low levels of SOX2) → S-C (intermediate levels of PRAME, intermediate levels of SOX2) → EC (no expression/low levels of PRAME, high levels of SOX2)], therefore supporting a complex pathogenetic model where the interactions between these two molecules are crucial in determining the fate of GCTT., Competing Interests: Declaration of Competing Interest All the Authors present in this article declare NO CONFLICT OF INTERESTS., (Copyright © 2022. Euschistus Dallas is distributed throughout the Western Hemisphere and is composed of 67 species in three subgenera: Euschistomorphus Jensen-Haarup, Lycipta Stl, and the nominate subgenus. Euschistus (Euschistus) includes several economically important species. Due to the lack of a phylogenetic analysis for the subgenus Euschistus, its groupings are based primarily on male genitalia. In this paper, the following Nearctic taxa that were first grouped together by Stl (1872), namely E. ictericus (Linnaeus), E. variolarius (Palisot de Beauvois), E. servus servus (Say), E. servus euschistoides (Vollenhoven), and E. tristigmus tristigmus (Say), are reviewed and redescribed. Several other similar, and probably related, taxa are also included in this study: E. tristigmus luridus (Dallas), E. inflatus Van Duzee, and E. latimarginatus Zimmer. Morphological patterns pertaining to the internal and external male and female genitalia allowed us to formalize the ictericus group to include the above-mentioned species (except E. tristigmus). The morphological overlap between the subspecies and sympatric areas of distribution have led us to consider E. servus euschistoides as a junior synonym of E. servus servus, and to corroborate the synonymy first proposed by Uhler (1861) of E. tristigmus luridus under E. tristigmus tristigmus.

Popular graph neural networks implement convolution operations on graphs based on polynomial spectral filters. In this paper, we propose a novel graph convolutional layer inspired by the auto-regressive moving average (ARMA) filter that, compared to polynomial ones, provides a more flexible frequency response, is more robust to noise, and better captures the global graph structure. We propose a graph neural network implementation of the ARMA filter with a recursive and distributed formulation, obtaining a convolutional layer that is efficient to train, localized in the node space, and can be transferred to new graphs at test time. We perform a spectral analysis to study the filtering effect of the proposed ARMA layer and report experiments on four downstream tasks: semi-supervised node classification, graph signal classification, graph classification, and graph regression. Results show that the proposed ARMA layer brings significant improvements over graph neural networks based on polynomial filters.

Background: The differential diagnosis between flat urothelial lesions [reactive urothelial atypia (RUA), atypia of unknown significance (AUS), urothelial dysplasia (UD) and carcinoma in situ (CIS)] has relevant prognostic and therapeutic implications. This crucial distinction could be very challenging but it is currently performed on hematoxylin and eosin (H&E) slides, with a great amount of partially discordant and/or not conclusive findings of the potential adjunctive role of immunohistochemistry. Herein, we tested double staining (DS) for p53/CK20 to verify if p53(+) cells, CK20(+) cells and double-positive cells (DPCs) are differentially expressed among these lesions and if p53/CK20 could be a useful tool in this diagnostic setting., Methods: We tested 50, 9, 36 and 29 consecutive and retrospectively enrolled cases of RUA, AUS, UD and CIS, respectively. p53(+) cells, CK20(+) cells and DPCs were evaluated and compared by adopting the appropriate statistic tests (Mann-Whitney U and Kruskal-Wallis tests)., Results: We found that p53(+) cells (p = 0.000), CK20(+) cells (p = 0.000) and DPCs (p = 0.000) showed statistically significant differences among the different flat urothelial lesions. Besides, when dichotomized, both CIS and RUA are easily differentiable from their histological mimickers adopting all these markers; by contrast, AUS and UD did not reach statistically significant differences able to differentiate them from each other [p53(+) cells, p = 0.123; CK20(+) cells, p = 0.567; DPCs, p = 0.409], except if compared to CIS [AUS VS CIS: p53(+) cells, p = 0.013; CK20(+) cells, p = 0.000; DPCs, p = 0.000; UD vs CIS: p53(+) cells, p = 0.000; CK20(+) cells, p = 0.000; DPCs, p = 0.000]., Conclusions: p53(+) cells, CK20(+) cells and DPCs are differently expressed by flat urothelial lesions and p53/CK20 could be a time- and money-saving tool for the appropriate management of these lesions if applied to a routine scenario., (Copyright © 2022 Elsevier GmbH. Image translation with convolutional autoencoders has recently been used as an approach to multimodal change detection (CD) in bitemporal satellite images. A main challenge is the alignment of the code spaces by reducing the contribution of change pixels to the learning of the translation function. Many existing approaches train the networks by exploiting supervised information of the change areas, which, however, is not always available. We propose to extract relational pixel information captured by domain-specific affinity matrices at the input and use this to enforce alignment of the code spaces and reduce the impact of change pixels on the learning objective. A change prior is derived in an unsupervised fashion from pixel pair affinities that are comparable across domains. To achieve code space alignment, we enforce pixels with similar affinity relations in the input domains to be correlated also in code space. We demonstrate the utility of this procedure in combination with cycle consistency. The proposed approach is compared with the state-of-the-art machine learning and deep learning algorithms. Experiments conducted on four real and representative datasets show the effectiveness of our methodology.

In graph neural networks (GNNs), pooling operators compute local summaries of input graphs to capture their global properties, and they are fundamental for building deep GNNs that learn hierarchical representations. In this work, we propose the Node Decimation Pooling (NDP), a pooling operator for GNNs that generates coarser graphs while preserving the overall graph topology. During training, the GNN learns new node representations and fits them to a pyramid of coarsened graphs, which is computed offline in a preprocessing stage. NDP consists of three steps. First, a node decimation procedure selects the nodes belonging to one side of the partition identified by a spectral algorithm that approximates the MAXCUT solution. Afterward, the selected nodes are connected with Kron reduction to form the coarsened graph. Finally, since the resulting graph is very dense, we apply a sparsification procedure that prunes the adjacency matrix of the coarsened graph to reduce the computational cost in the GNN. Notably, we show that it is possible to remove many edges without significantly altering the graph structure. Experimental results show that NDP is more efficient compared to state-of-the-art graph pooling operators while reaching, at the same time, competitive performance on a significant variety of graph classification tasks.

Stink bugs (Pentatomidae) are a speciose group of insects that feed mostly on plants. Many species are considered agricultural pests of economically important crops around the globe. Mitochondrial genomes are valuable for evolutionary and phylogenetic studies, but have been little explored for Pentatomidae. Here, we characterized the mitochondrial genomes of four pentatomid species (Diceraeus melacanthus, Euschistus heros, Piezodorus guildinii, and Stiretrus anchorago) and performed a comparative analysis for this family and its subfamilies. Stink bug mitogenomes disclosed a conserved gene order and content, although we detected two uncommon armless tRNAs in E. heros and D. melacanthus. Phylogenetic results indicate that Pentatominae is polyphyletic, showing that internal relationships of Pentatomidae should be further investigated. Stink bug mitochondrial genes are under strong purifying selection, except for ATP8 which showed signs of positive selection.

Objective: We sought to assess the efficacy of a hydrolyzed collagen supplement (Lapi Gelatine, Empoli, Italy) for improving skin moisturization and elasticity as well as the appearance of wrinkles., Methods: A monocentric, randomized, controlled study was performed; the study included 52 volunteers who were divided into two groups. Twenty-six subjects took the collagen supplement and the other 26 participants received maltodextrin for a period of 56 days. Skin moisturization, skin elasticity, and wrinkle depth were instrumentally evaluated. Clinical parameters, including skin softness, skin firmness, skin smoothness, and the visibility of wrinkles were subjectively evaluated by a dermatologist. Finally, the gastrointestinal tolerability was evaluated during the period of treatment., Results: Improvements in skin moisturization, skin elasticity and wrinkle depth were measured in the participants taking the hydrolyzed collagen after 28 days. In all enrolled participants, the results of the analysis of variance showed a statistically significant effect of the interaction between product and time. The evolution over time of the investigated parameters was significantly different between the participants taking the hydrolyzed collagen and the participants taking the maltodextrin. Accurate taxonomic identifications and species delimitations are a fundamental problem in biology. The complex taxonomy of Nematoda is primarily based on morphology, which is often dubious. DNA barcoding emerged as a handy tool to identify specimens and assess diversity, but its applications in Nematoda are incipient. We evaluated cytochrome c oxidase subunit I ( cox1 ) efficiency as a DNA barcode for nematodes scrutinising 5241 sequences retrieved from BOLD and GenBank. The samples included genera with medical, agricultural, or ecological relevance: Anguillicola , Caenorhabditis , Heterodera , Meloidogyne , Onchocerca , Strongyloides , and Trichinella . We assessed cox1 performance through barcode gap and Probability of Correct Identification (PCI) analyses, and estimated species richness through Automatic Barcode Gap Discovery (ABGD). Each genus presented distinct gap ranges, mirroring the evolutionary diversity within Nematoda. Thus, to survey the diversity of the phylum, a careful definition of thresholds for lower taxonomic levels should be considered. PCIs were around 70% for both databases, highlighting operational biases and challenges in nematode taxonomy. ABGD inferred higher richness than the taxonomic labels informed by databases. The prevalence of specimen misidentifications and dubious species delimitations emphasise the value of integrative approaches to nematode taxonomy and systematics. Overall, cox1 is a relevant tool for integrative taxonomy of nematodes.

Purpose: Diagnosis of anterior prostate cancer (PCa) can be quite challenging, often leading to delay in treatment. mpMRI-guided biopsy (GB) has been introduced aiming to increase the number of diagnoses of clinically significant PCa with fewer cores. The aim of our study is to compare pathological findings of prostate biopsy, In-bore or Fusion technique, with histopathological evaluation of radical prostatectomy., Methods: We prospectively collected data from 90 consecutive patients who underwent either In-bore or Fusion biopsy following the detection of an index suspicious lesion at mpMRI in the anterior part of the prostatic gland. Bioptical pathological findings were compared with pathological findings reported after robot-assisted radical prostatectomy., Results: Patients who underwent In-bore GB had a higher rate of previous negative prostate biopsies (19% vs 44%, p = 0.02). Median number of bioptic cores taken (13 vs 2) and number of positive cores (3 vs 2) were significantly superior in the Fusion group compared to the In-bore group (p < 0.001 and p = 0.002, respectively), whilst clinical International Society of Urological Pathology (ISUP) grade was homogeneous within groups. The concordance between anterior lesions detected at biopsy and those reported in the histopathological finding of radical prostatectomy was very high, without statistically significant difference between groups., Conclusion: Both Fusion and In-bore GB are accurate in detecting anterior PCa, with enhanced precision detecting clinically significant tumours, as evidenced by pathologic examinations which confirmed the presence of index anterior PCa in > 50% of patients overall. Additional sextant biopsy is still required, especially among biopsy-näive patients, to avoid missing clinically significant PCa.

Classification of multivariate time series (MTS) has been tackled with a large variety of methodologies and applied to a wide range of scenarios. Reservoir computing (RC) provides efficient tools to generate a vectorial, fixed-size representation of the MTS that can be further processed by standard classifiers. Despite their unrivaled training speed, MTS classifiers based on a standard RC architecture fail to achieve the same accuracy of fully trainable neural networks. In this article, we introduce the reservoir model space, an unsupervised approach based on RC to learn vectorial representations of MTS. Each MTS is encoded within the parameters of a linear model trained to predict a low-dimensional embedding of the reservoir dynamics. Compared with other RC methods, our model space yields better representations and attains comparable computational performance due to an intermediate dimensionality reduction procedure. As a second contribution, we propose a modular RC framework for MTS classification, with an associated open-source Python library. The framework provides different modules to seamlessly implement advanced RC architectures. The architectures are compared with other MTS classifiers, including deep learning models and time series kernels. Results obtained on the benchmark and real-world MTS data sets show that RC classifiers are dramatically faster and, when implemented using our proposed representation, also achieve superior classification accuracy.

The Carpocorini are distributed worldwide, and it is one of the most speciose tribes within the Pentatomidae with 127 genera and more than 500 valid species. Recently, Adustonotus Bianchi was described to contain eight species formerly placed within Euschistus Dallas. Among them, Adustonotus grandis (Rolston) and Adustonotus latus (Dallas) are remarkable for their large size. Herein, the phylogenetic position of a new taxon is inferred by a total evidence analysis based on 85 morphological characters and four molecular markers. Adustonotus graziae sp. nov. is described, and is recovered in a polytomic lineage, including A. grandis and A. latus. These species share a solid combination of features that enable them to be separated from the other Adustonotus species (e.g., large size, the humeral angles spatulate and exceptionally produced, and the capsula seminalis shortened). Illustrations of external and internal genitalia, and a distributional map are provided.

Pentatomoidea is the third largest superfamily in Heteroptera. The internal systematics and classification of this superfamily have an intricate history. The paper by Grazia et al. (2008) is a milestone to the phylogenetic hypothesis of Pentatomoidea. Subsequent papers explored the limited conclusions and unanswered questions left by Grazia et al. (2008). We proposed to look at the body of knowledge produced since Grazia et al. (2008) and to compile the molecular data for Pentatomoidea deposited in Genbank to achieve three aims: (i) to evaluate the advances on the phylogenetic relationships of the Pentatomoidea; (ii) to produce a phylogenetic hypothesis based on molecular data deposited in Genbank; and (iii) to highlight the shortcomings and strengths of the available data. We retrieved sequences of four molecular markers (COI, 16S, 18S, and 28S) for 167 terminal taxa, including 149 pentatomoids. A concatenated matrix was analyzed under maximum likelihood (ML) and parsimony (MP). Both methods supported the monophyly of Pentatomoidea, and poorly resolved internal relationships among the families. Acanthosomatidae, Dinidordae, Pentatomidae, Scutelleridae, Thaumastellidae, and Urostylididae were monophyletic (under ML and MP), and also Plataspidae and Thyreocoridae (under ML). Tessaratomidae and Cydnidae were non-monophyletic under both methods. Our results were compared to the phylogenetic hypotheses proposed for Pentatomoidea. The analysis of the data available on the GenBank allowed us to affirm that many problems mentioned previously remain unsolved, even though the sampling of terminals has increased. In summary, the efforts in the last two decades to better understand the relationships within the Pentatomoidea have been insufficient to propose robust advances in phylogenetic hypothesis for the group. We discuss topics we understand are paramount to upcoming developments:1) better taxon sample; 2) collection management; 3) increased markers; and 4) morphology and anatomical ontology.

We estimate the weekly excess all-cause mortality in Norway and Sweden, the years of life lost (YLL) attributed to COVID-19 in Sweden, and the significance of mortality displacement. We computed the expected mortality by taking into account the declining trend and the seasonality in mortality in the two countries over the past 20 years. From the excess mortality in Sweden in 2019/20, we estimated the YLL attributed to COVID-19 using the life expectancy in different age groups. We adjusted this estimate for possible displacement using an auto-regressive model for the year-to-year variations in excess mortality. We found that excess all-cause mortality over the epidemic year, July 2019 to July 2020, was 517 (95%CI = (12, 1074)) in Norway and 4329 [3331, 5325] in Sweden. There were 255 COVID-19 related deaths reported in Norway, and 5741 in Sweden, that year. During the epidemic period of 11 March-11 November, there were 6247 reported COVID-19 deaths and 5517 (4701, 6330) excess deaths in Sweden. We estimated that the number of YLL attributed to COVID-19 in Sweden was 45,850 [13,915, 80,276] without adjusting for mortality displacement and 43,073 (12,160, 85,451) after adjusting for the displacement accounted for by the auto-regressive model. In conclusion, we find good agreement between officially recorded COVID-19 related deaths and all-cause excess deaths in both countries during the first epidemic wave and no significant mortality displacement that can explain those deaths.

'We advise the authors to find a native English speaker to proofread the manuscript'. This is a standard feedback journals give to non-native English speakers. Journals are justifiably concerned with grammar but do not show the same rigour about another step crucial to biological research: specimen identification. Surveying the author guidelines of 100 journals, we found that only 6% of them request explicitly citation of the literature used in specimen identification. Authors hamper readers from contesting specimen identification whenever vouchers, identification methods, and taxon concepts are not provided. However, unclear taxonomic procedures violate the basic scientific principle of reproducibility. The scientific community must continuously look for practical alternatives to improve taxonomic identification and taxonomic verification. We argue that voucher pictures are an accessible, cheap and time-effective alternative to mitigate (not abolish) bad taxonomy by exposing preventable misidentifications. Voucher pictures allow scientists to judge specimen identification actively, based on available data. The popularization of high-quality image devices, photo-identification technologies and computer vision algorithms yield accurate scientific photo-documentation, improving taxonomic procedures. Taxonomy is timeless, transversal and essential to most scientific disciplines in biological sciences. It is time to demand rigour in taxonomic identifications.

The pentatomids (Hemiptera: Heteroptera) are the third most speciose family within the Heteroptera or the true bugs. The family occurs worldwide and comprises around five thousand valid species within 950 genera. Padaeus Stål belongs to a complex of other genera of Carpocorini Mulsant and Rey related to Euschistus Dallas. These genera present similarities in color, size, and shape, and share common features. However, among its four congeneric species, Padaeus bovillus Distant has been highlighted as an outlier by the posterior margins of the bucculae evanescent, while its congeneric species present posterior margins of the bucculae lobed. Thus, herein we redescribe P. bovillus and present a hypothesis regarding its phylogenetic placement within the Carpocorini. Furthermore, a new species similar to P. bovillus is described. Four molecular markers ( COI , CytB, 16S, and 28S) plus 86 morphological characters were used to infer the phylogeny under Maximum likelihood and Bayesian inference. For the descriptions, we measured 16 morphometric parameters and dissected the genitalic structures. We also include illustrations of the habitus, internal and external genitalic structures, and provide distribution maps. Mitripus seclusus sp. n. Bianchi, Krein, Rider, and Grazia is recovered as the sister species to Mitripus bovillus comb. n., and both within Mitripus Rolston. Among other shared characters, species of Mitripus have the femora unarmed, they have a macula near the apex of the radial vein, and the mandibular plates tapering apically. Mitripus bovillus comb. n. and Mitripus seclusus sp. n. have the posterior margin of the pygophore projecting as a spine, a unique pattern within the genus. According to our results, Mitripus including M. bovillus comb. n. and Mitripus seclusus sp. n. now includes five species.

As of November 2020, the number of COVID-19 cases was increasing rapidly in many countries. In Europe, the virus spread slowed considerably in the late spring due to strict lockdown, but a second wave of the pandemic grew throughout the fall. In this study, we first reconstruct the time evolution of the effective reproduction numbers R(t) for each country by integrating the equations of the classic Susceptible-Infectious-Recovered (SIR) model. We cluster countries based on the estimated R(t) through a suitable time series dissimilarity. The clustering result suggests that simple dynamical mechanisms determine how countries respond to changes in COVID-19 case counts. Inspired by these results, we extend the simple SIR model for disease spread to include a social response to explain the number X(t) of new confirmed daily cases. In particular, we characterize the social response with a first-order model that depends on three parameters ν1,ν2,ν3. The parameter ν1 describes the effect of relaxed intervention when the incidence rate is low; ν2 models the impact of interventions when incidence rate is high; ν3 represents the fatigue, i.e., the weakening of interventions as time passes. The proposed model reproduces typical evolving patterns of COVID-19 epidemic waves observed in many countries. Estimating the parameters ν1,ν2,ν3 and initial conditions, such as R0, for different countries helps to identify important dynamics in their social responses. One conclusion is that the leading cause of the strong second wave in Europe in the fall of 2020 was not the relaxation of interventions during the summer, but rather the failure to enforce interventions in the fall.

Introduction: The purpose of this study was to evaluate the impact of 3-dimensional (3D) digital reconstructions of renal models on the arterial clamping approach during partial nephrectomy (PN)., Patients and Methods: Fifty-seven patients with T1 renal mass, referred for PN, were prospectively enrolled in 2 groups: Group 1 (n = 32) with revision of both 2-dimensional (2D) computed tomography (CT) imaging and 3D virtual model before surgery; Group 2 (n = 25) with revision of 2D CT imaging. Segmentation of the 3D models from preoperative high-quality CT scan was achieved using D2P software. In a sub-analysis of patients treated with PN with the on-clamp approach (n = 36), the effective intraoperative level of arterial clamping was compared with the preoperative planning., Results: In the sub-group of patients referred to PN with the on-clamp approach, the intraoperative selective clamping was performed in 12 (57.1%) patients of Group 1 and in 2 (13.3%) cases of Group 2 (P = .01). The intraoperative management of the renal pedicle was done as preoperatively planned in 61.9% of patients in Group 1 and in 86.6% of cases in Group 2 (P = .1)., Conclusion: The 3D-guided plan of PN allows to perform selective clamping in higher proportion of patients compared with the standard 2D-guided approach without increasing intraoperative and postoperative complications., (Copyright © 2020 Elsevier Inc. Purpose: To perform an external validation of a recently published nomogram aimed to predict positive 68 Ga-PSMA-11 PET/CT in patients with biochemical recurrence (BCR) after radical prostatectomy (RP) by Rauscher et al. (Eur Urol 73(5):656-661, 2018)., Methods: Overall, 413 PCa patients with BCR after RP (two consecutive PSA ≥ 0.2 ng/ml) and PSA value between 0.2 and 1 ng/ml were included. A multivariable logistic regression model was produced to assess the predictors of positive 68 Ga-PSMA-11 PET/CT results. The performance characteristics of the model were assessed by quantifying the predictive accuracy, according to model calibration. Yuden's index was used to find the best nomogram's cut-off. Finally, decision curve analysis (DCA) was implemented to quantify the nomogram's clinical value., Results: In the external cohort, the overall detection rate of 68 Ga-PSMA-11 PET/CT was 44% vs. 64.7% in the original population. At multivariate analysis, PSA at 68 Ga-PSMA-11 PET/CT (OR: 7.06, p < 0.001) and ongoing ADT at time of 68 Ga-PSMA-11 PET/CT (OR: 2.07, p = 0.03) were the only independent predictors of PET/CT positivity. The predictive accuracy of nomogram was suboptimal and comparable to that reported in the original model (64% vs. 67%, respectively). The calibration plot indicated suboptimal concordance. The best nomogram's cut-off to predict positive 68 Ga-PSMA-11 PET/CT was 35% (AUC = 0.61). In DCA, the nomogram revealed clinical net benefit when the threshold probabilities of positive 68 Ga-PSMA-11 PET/CT is > 35%., Conclusion: We assessed similar suboptimal predictive accuracies in the external cohort compared to the original one. PSA and ongoing ADT were confirmed as positive predictors, and the most informative nomogram cut-off resulted 35%.

Genital coevolution is a pervasive phenomenon as changes in one sex tend to impose fitness consequences on the other, generating sexual conflict. Sexual conflict is often thought to cause stronger selection on males due to the Darwin-Bateman's anisogamy paradigm. However, recent studies have demonstrated that female genitalia may be equally elaborated and perform diverse extra-copulatory functions. These characteristics suggest that female genitals can also be primary targets of selection, especially where natural selection acts on female-exclusive functions such as oviposition. Here, we test this hypothesis in a statistical phylogenetic framework across the whole beetle (Coleoptera) phylogeny, investigating whether coevolution of specific genital traits may be triggered by changes in females. We focus on traits of the proctiger, which composes part of the male terminalia and the female ovipositor. Our results present a comprehensive case of male-female genital coevolution and provide solid statistical evidence for a female-initiated coevolutionary process where the vast majority of evolutionary transitions in males have occurred only after changes in females. We corroborate the hypothesis that female traits may change independently and elicit counter-adaptations in males. Furthermore, by showing a consistent pattern across the phylogeny of the most diverse group of animals, our results suggest that this female-driven dynamics may persist through long time scales., (© 2020 European Society For Evolutionary Biology. Sibaria Stål is a genus of Pentatomidae occurring exclusively in the Neotropical region. Since its description, little information has been added to the knowledge of the taxon. The hypothesis of monophyly of Sibaria was recently supported by a phylogenetic analysis based on morphological and molecular data. The purpose of this study is to provide a review of Sibaria, redescribe the valid species, and describe a new species. The genus includes four species: S. andicola Breddin, S. armata (Dallas), S. englemani Rolston, and S. amazonica sp. nov.. Diagnoses, macrophotographs (habitus, external and internal genitalia), an identification key, and distribution records of the species are also provided.

Objectives: Transrectal ultrasound-guided biopsy (TRUS-GB) is the current reference standard procedure for diagnosis of prostate cancer (PCa) but this procedure has limitations related to the low detection rate (DR) described in the literature. The aim of the study was to evaluate the DR efficiency, and complication rate in a pure "in-bore" magnetic resonance imaging-guided biopsy (MRI-GB) series according to the Prostate Imaging Reporting and Data System, version 2 (PI-RADS v2)., Materials and Methods: From July 2015 to April 2018, a series of 142 consecutive patients undergoing MRI-GB were prospectively enrolled. According to the European Society of Urogenital Radiology guidelines, the presence of clinically significant PCa (csPCa) on multiparametric magnetic resonance imaging was defined as equivocal, likely, or highly likely according to a PI-RADS v2, score of 3, 4, or 5, respectively., Results: Of 142 patients, 76 (53.5%) were biopsy naive and 66 (46.5%) had ≤ 1 previous negative set of random TRUS-GB findings. The MRI-GB findings were positive in 75 of 142 patients with a DR of 52.8%. Of the 76 patients with ≤ 1 previous set of TRUS-GB, 43 had PCa found by MRI-GB, with a DR of 57.3%. The DR in the 66 biopsy-naive patients was 48% (32/66). Of the 75 patients with positive biopsy findings, 54 (80.5%) were found to have csPCa on histological examination. Of these 54 patients, 28 had an International Society of Urological Pathology grade 2; 5 had grade 3, 19 had grade 4, and 2 had grade 5. Considering the anatomic distribution of the index lesions using the PI-RADS v2 scheme, the probability of PCa was greater for lesions located in the peripheral zone (55 of 75, 73.3%) than for those in the central zone (20 of 75, 26.7%)., Conclusions: Our study conducted on 142 patients confirmed the greater DR of csPCa by MRI-GB, with a very low number of cores needed and a negligible incidence of complications, especially in patients with a previous negative biopsy. We address and discuss some of the many flaws exhibited by Costa et al. (2019) which tried to explain the twig-like camouflage of Cladomorphus phyllinus. Given the lack of both empirical and theoretical underpinnings in Costa et al. (2019), we call into question the validity of their conclusions, in particular, that horizontal gene transfer is a causal mechanism for the camouflage in C. phyllinus.

The genus Euschistus Dallas includes 67 species restricted to the New World, and several species are registered on cultivated plants in the Nearctic and Neotropical regions. In South America, most Euschistus species are completely overlooked due to the lack of information to allow accurate identification. Here, we redescribed Euschistus taurulus Berg, including for the first time, characterization of the internal and external genitalia of both sexes. We also report original information on bionomics, review and update information on geographical distribution and host plants records. Additionally, we provide DNA barcoding sequences for E. taurulus and three other morphologically similar key-agriculture pest species in South America: Euschistus heros (Fabricius), Dichelops melacanthus (Dallas), and Dichelops furcatus (Fabricius). We discuss means for correct identification of E. taurulus and its phylogenetic position within Euschistus and other similar stink bugs; the potential economic importance of the E. taurulus is also addressed.

A promising direction in deep learning research consists in learning representations and simultaneously discovering cluster structure in unlabeled data by optimizing a discriminative loss function. As opposed to supervised deep learning, this line of research is in its infancy, and how to design and optimize suitable loss functions to train deep neural networks for clustering is still an open question. Our contribution to this emerging field is a new deep clustering network that leverages the discriminative power of information-theoretic divergence measures, which have been shown to be effective in traditional clustering. We propose a novel loss function that incorporates geometric regularization constraints, thus avoiding degenerate structures of the resulting clustering partition. Experiments on synthetic benchmarks and real datasets show that the proposed network achieves competitive performance with respect to other state-of-the-art methods, scales well to large datasets, and does not require pre-training steps., (Copyright © 2019 Elsevier Ltd. Canola (Brassicaceae: Brassica spp.) is an important feedstock for biodiesel production and a potential ingredient for use in the food industry. In different continents, various arthropod pests damage canola plants. Stink bugs (Heteroptera: Pentatomidae) are present in all zoogeographical regions, and many species are recognized by their economic importance as crop pests. Our aim was to describe the composition, structure, and diversity of the assemblage of stink bugs sampled on canola in southern Brazil. A total of 878 pentatomids were captured, belonging to 27 species. The dominant species were Euschistus heros (F.) (n = 439), Dichelops furcatus (F.) (n = 160), and Nezara viridula L. (n = 79). The species richness estimators indicated the samples correspond from 79.7 to 93.1% of the richness estimated. Comparing canola to other monocultures, the richness of Pentatomidae was much superior. Otherwise, when compared to studies conducted in native vegetation and urban fragments, the richness is similar, and sometimes higher. This relatively high number of associations brings out the vulnerability of the canola fields according to the expansion of its cultivated area. Nine species of Pentatomidae are recorded on canola for the first time in Brazil. The most abundant species reported here are often stressed as stink bugs of economic importance in agro-ecosystems. The economic importance of each species varies greatly depending on the plant attacked. Detailed studies are needed to evaluate the damage caused to canola by stink bugs.

Understanding the statistics of fluctuation driven flows in the boundary layer of magnetically confined plasmas is desired to accurately model the lifetime of the vacuum vessel components. Mirror Langmuir probes (MLPs) are a novel diagnostic that uniquely allow us to sample the plasma parameters on a time scale shorter than the characteristic time scale of their fluctuations. Sudden large-amplitude fluctuations in the plasma degrade the precision and accuracy of the plasma parameters reported by MLPs for cases in which the probe bias range is of insufficient amplitude. While some data samples can readily be classified as valid and invalid, we find that such a classification may be ambiguous for up to 40% of data sampled for the plasma parameters and bias voltages considered in this study. In this contribution, we employ an autoencoder (AE) to learn a low-dimensional representation of valid data samples. By definition, the coordinates in this space are the features that mostly characterize valid data. Ambiguous data samples are classified in this space using standard classifiers for vectorial data. In this way, we avoid defining complicated threshold rules to identify outliers, which require strong assumptions and introduce biases in the analysis. By removing the outliers that are identified in the latent low-dimensional space of the AE, we find that the average conductive and convective radial heat fluxes are between approximately 5% and 15% lower as when removing outliers identified by threshold values. For contributions to the radial heat flux due to triple correlations, the difference is up to 40%.

Introduction: Purpose of the study was to investigate the correlation of a preoperative multiparametric magnetic resonance imaging of the prostate (mpMRI) in patients with a suspicion of prostate cancer and eligible for Holmium Laser Enucleation of the Prostate (HoLEP)., Material and Methods: Data of 228 patients who had undergone HoLEP was selected and retrospectively analyzed from a multicentric database. All patients presented with a raised serum PSA and/or an abnormal digital rectal examination (DRE). Prostate cancer (PCa) was excluded either with a negative mpMRI (group 'NEGATIVE MRI' n = 113) or a standard biopsy (group 'NO MRI' n = 115). Preoperative characteristic surgical and histological outcomes were confronted. A univariate and multivariate logistic regression model was performed to investigate independent predictors of incidental Prostate Cancer (iPCa)., Results: Both groups presented with no statistical differences in preoperative characteristics besides previous acute urinary retention rates and post-voided residual volume, found to be higher (27.8% vs. 14.2% and median 120cc vs. 80cc) in NO MRI and NEGATIVE MRI respectively.No differences were registered in surgical time, removed tissue, catheterization time, hospital stay and complications rate.Statistically lower rate of iPCa (p = 0.03) was detected in the NEGATIVE MRI group (6.2%) in comparison with NO MRI group (14.8%). In multivariate logistic regression only presence of a preoperative negative mpMRI correlated (p = 0.04) as an independent predictive factor (OR 2.63; 95% CI: 1.02-6.75)., Conclusions: A negative mpMRI might be a useful tool to be included in a novel preoperative assessment to patients eligible for HoLEP with a suspicion of PCa in order to avoid an incidental PCa., Competing Interests: The authors declare no conflicts of interest.

Introduction: The aim of this study was to assess surgical and functional outcomes of 17 consecutive patients undergoing robot- assisted radical cystectomy (RARC) with palliative intent in a monocentric single surgeon series., Material and Methods: We collected data from 17 consecutive patients who underwent RARC with palliative intent performed by a single surgeon at our institution. Patients undergoing palliative RARC were those with advanced bladder cancer (BC) or advanced comorbidities. Clinical, surgical and functional outcomes were prospectively collected. Patients completed a specific questionnaire (Functional Assessment of Cancer Therapy-Bladder Cancer, FACT-BL) before and after surgery to assess the role of palliative RARC in terms of quality of life improvement., Results: Median age at surgery was 78 years, with median Charlson Comorbidity Index (CCI) and age-adjusted CCI of 3 and 7, respectively. Clinical stage was T2, T3 or T4 in 7, 8 and 2 patients respectively, with 52.9% and 29.4% with cN+ and cM+ disease. Median estimated blood loss was 200 ml, with 1 patient requiring intra-operative blood transfusion. Median hospital stay was 7 days. A total of 3 and 2 patients were re-hospitalized during the first 30 and 30-90 post-operative days, respectively. One major Clavien grade complication was recorded.At median follow-up of 8 months, 9 and 2 patients succumbed due to tumor progression and other causes. Pre-operative and post-operative FACT-BL scores improved significantly in each domain., Conclusions: A RARC is a safe and feasible technique which could be offered as part of palliative care in patients with advanced BC or comorbidities. Objectives: To assess the predictive accuracy and the clinical value of a recent nomogram predicting cancer-specific mortality-free survival after surgery in pN1 prostate cancer patients through an external validation., Methods: We evaluated 518 prostate cancer patients treated with radical prostatectomy and pelvic lymph node dissection with evidence of nodal metastases at final pathology, at 10 tertiary centers. External validation was carried out using regression coefficients of the previously published nomogram. The performance characteristics of the model were assessed by quantifying predictive accuracy, according to the area under the curve in the receiver operating characteristic curve and model calibration. Furthermore, we systematically analyzed the specificity, sensitivity, positive predictive value and negative predictive value for each nomogram-derived probability cut-off. Finally, we implemented decision curve analysis, in order to quantify the nomogram's clinical value in routine practice., Results: External validation showed inferior predictive accuracy as referred to in the internal validation (65.8% vs 83.3%, respectively). The discrimination (area under the curve) of the multivariable model was 66.7% (95% CI 60.1-73.0%) by testing with receiver operating characteristic curve analysis. The calibration plot showed an overestimation throughout the range of predicted cancer-specific mortality-free survival rates probabilities. However, in decision curve analysis, the nomogram's use showed a net benefit when compared with the scenarios of treating all patients or none., Conclusions: In an external setting, the nomogram showed inferior predictive accuracy and suboptimal calibration characteristics as compared to that reported in the original population. However, decision curve analysis showed a clinical net benefit, suggesting a clinical implication to correctly manage pN1 prostate cancer patients after surgery., (© 2018 The Japanese Urological Association.)
