1. Correlation of Hsa circ0016599 expression level with the efficacy of neoadjuvant chemotherapy in patients with triple-negative breast cancer

Abstract

"Objective To analyze the expression of circular ribonucleic acid (circRNA) Hsa circ0016599 in patients with triple-negative breast cancer (TNBC) and its effect and predictive value for the efficacy of neoadjuvant chemotherapy (NACT). Methods A total of 124 patients with TNBC who received NACT treatment before operation in Cangzhou Peoples Hospital from April 2022 to February 2024 were selected retrospectively as the research objects. The expression of Hsa circ0016599 was measured in the resected cancer tissues and adjacent tissues, and they were divided into low expression groups (≤15.12, n=59) and high expression group (＞15.12, n=65)according to the median relative expression value of Hsa circ0016599 (2-ΔΔCt=15.12). The expression levels of Hsa circ0016599 in cancer tissues and adjacent tissues of 124 TNBC patients and the total effective rates of NACT in low expression group and high expression group were compared. The risk factors affecting the efficacy of NACT in TNBC patients were screened to construct a nomogram model. Results In 124 TNBC patients, the expression level of Hsa circ0016599 in cancer tissues was higher than that in adjacent tissues (19.76±4.31 vs 14.15±2.74, t=12.232, P＜0.01). Compared with the low expression group, the high expression group had a higher proportion of patients with regional lymph node metastasis, vascular tumor thrombus, and nerve invasion (P＜0.05), also had a lower total effective rate of NACT (35.38% vs 62.71%, χ2=9.248, P＜0.01）. High expression of Hsa circ0016599, presence of regional lymph node metastasis, vascular tumor thrombus, and nerve invasion were all risk factors affecting NACT efficacy in TNBC patients (OR=1.264, 1.239, 1.257, 1.247, P＜0.05). The calibration curve of Hsa circ0016599 expression level prediction model showed a good predictive value for NACT efficacy in TNBC patients. Conclusion Hsa circ0016599 is highly expressed in the cancer tissues of TNBC patients, and the total NACT effective rate of patients with high expression of Hsa circ0016599 is relatively low, indicating that Hsa circ0016599 has a certain predictive value for NACT efficacy."

Abstract

Trifluoromethyl (CF3) and other fluoroalkyl groups are of great significance in the fields of pharmaceutical chemistry and agricultural chemicals. Fluoroalkyl acids, especially trifluoroacetic acid (TFA) is considered the most ideal fluoroalkylation reagent due to its low cost and easy availability. However, the extremely high oxidation potential requirement of TFA limits its wide application. In recent years, since visible-light-induced fluoroalkylation through the ligand-to-metal charge transfer (LMCT) process can overcome the above limitations, it has become an effective synthetic tool for the construction of fluorinated compounds with complex molecules and structures. In this review, according to the classification of different metal catalysts, we summarize the trifluoromethylation and fluoroalkylation of olefins, heteroaromatics, and terminal alkynes in different metal catalytic systems and their corresponding reaction mechanisms. The photocatalytic fluoroalkylation via LMCT is believed to expedite the development of fluoro-containing drugs, and more novel fluoroalkylation methologies using this strategy will be disclosed. Abstract

Different types of natural K+ channels share similar core modules and cation permeability characteristics. In this study, we have developed novel artificial K+ channels by rebuilding the core modules of natural K+ channels in artificial systems. All the channels displayed high selectivity for K+ over Na+ and exhibited a selectivity sequence of K+≈Rb+ during the transport process, which is highly consistent with the cation permeability characteristics of natural K+ channels. More importantly, these artificial channels could be efficiently inserted into cell membranes and mediate the transmembrane transport of K+, disrupting the cellular K+ homeostasis and eventually triggering the apoptosis of cells. These findings demonstrate that, by rebuilding the core modules of natural K+ channels in artificial systems, the structures, transport behaviors, and physiological functions of natural K+ channels can be mimicked in synthetic channels. Abstract

Returning crop straw into the soil is an important practice to balance biogenic and bioavailable silicon (Si) pool in paddy, which is crucial for the healthy growth of rice. However, owing to little knowledge about soil microbial communities responsible for straw degradation, how straw return affects Si bioavailability, its uptake, and rice yield remains elusive. Herein, we investigate the change of soil Si fractions and microbial community in a 39-year-old paddy field amended by a long-term straw return. Results show that rice straw return significantly increased soil bioavailable Si and rice yield from 29.9% to 61.6% and from 14.5% to 23.6%, respectively, when compared to NPK fertilization alone. Straw return significantly altered soil microbial community abundance. Acidobacteria was positively and significantly related to amorphous Si, while Rokubacteria at phylum level, Deltaproteobacteria, and Holophagae at class level was negatively and significantly related to organic matter adsorbed and Fe/Mn-oxide-combined Si in soils. Redundancy analysis of their correlations further demonstrated that Si status significantly explained 12% of soil bacterial community variation. These findings suggest that soil bacteria community and diversity interact with Si mobility by altering its transformation, thus resulting in the balance of various nutrient sources to drive biological Si cycle in agroecosystem.

46. Supplying silicon alters microbial community and reduces soil cadmium bioavailability to promote health wheat growth and yield

Abstract

Soil amendments of black bone (BB), biochar (BC), silicon fertilizer (SI), and leaf fertilizer (LF) play vital roles in decreasing cadmium (Cd) availability, thereby supporting healthy plant growth and food security in agroecosystems. However, the effect of their additions on soil microbial community and the resulting soil Cd bioavailability, plant Cd uptake and health growth are still unknown. Therefore, in this study, BB, BC, SI, and LF were selected to evaluate Cd amelioration in wheat grown in Cd-contaminated soils. The results showed that relative to the control, all amendments significantly decreased both soil Cd bioavailability and its uptake in plant tissues, promoting healthy wheat growth and yield. This induced-decrease effect in seeds was the most obvious, wherein the effect was the highest in SI (52.54%), followed by LF (43.31%), and lowest in BC (35.24%) and BB (31.98%). Moreover, the induced decrease in soil Cd bioavailability was the highest in SI (29.56%), followed by BC (28.85%), lowest in LF (17.55%), and BB (15.30%). The significant effect in SI likely resulted from a significant increase in both the soil bioavailable Si and microbial community (Acidobacteria and Thaumarchaeota), which significantly decreased soil Cd bioavailability towards plant roots. In particular, a co-occurrence network analysis indicated that soil microbes played a substantial role in wheat yield under Si amendment. Therefore, supplying Si alters the soil microbial community, positively and significantly interacting with soil bioavailable Si and decreasing Cd bioavailability in soils, thereby sustaining healthy crop development and food quality.
