Lipase is one of the promising biocatalyst in field of Enzymatic Biodiesel production. Screening of lipase producing bacteria and its media optimization facilitates enhanced lipase production. Considering the advantages of enzymatic biodiesel production using lipases the objective of present work was to isolate lipase producing bacteria and its media optimization. Bacterial strain were isolated from oil rich sample showing maximum zone of hydrolysis on TBA Media was selected and identified as Lysinibacillus macroides FS1 by 16SrDNA gene sequence analysis. Lipase production was carried out in production media and lipase activity of 3.1U/ ml was assayed by titrimetric method. Various media parameters were optimized and maximal lipase activity of 16.75U/ml were observed at 48 hr of incubation in optimized media with Honge oil as inducer, Galactose as carbon source , Ammonium Chloride and Beef extract as source of nitrogen, pH 7 and 37oC temperature with agitation speed of 120 rpm. Under optimized condition 5.4 fold increases in the lipase activity than innate activity was observed. 50.74% yield was observed with specific activity of 21.25U/mg after purification with 30% ammonium sulphate. Purified lipase was stable in presence of methanol at 37°C. Results obtained shows that Lipase from Lysinibacillus macroides FS1 was promising biocatalyst for Biodiesel production.

Biodiesel a fatty acid alkyl esteris one of the promising biofuel and a clean energy source as an alternative to petroleum-based diesel fuels. The Enzymatic transesterification reaction is influenced by many factors such as amount of biocatalyst, molar ratio of oil to alcohol, temperature, pH, rpm, time etc. Effective variables for transesterification may vary based on the type of feedstock and catalyst used, therefore it is essential to optimise the process suitable for each type of feedstock to achieve higher yield of biodiesel. The statistically-based Plackett-Burman experimental design was adopted in this study to identify effective factors for transesterification reaction of Pongamiapinnata seed oil using immobilized lipase. Transesterification is a well known chemical process of producing biodiesel from edible and non-edible oils, animal fats and cooked oils. Base catalytic transesterification of non-edible oil is a popular method for production of biodiesel. Biodiesel production from Karanja or Honge (Pongamia pinnata) seed oil is gaining momentum in India and other parts of world. The real time monitoring of transesterification reaction progress is essential step towards automating biodiesel production. Two planar inter digital capacitive sensors (IDCS) are fabricated for the purpose of monitoring the transesterification progress of Karanja oil and their performance are evaluated. The repeatability and sensitivity of fabricated sensor is reliable and acceptable. The sensor with associated signal conditioning circuit will be useful in automating the biodiesel production process and play vital role in energy efficiency and the quality of end products.

Bacterial blight in pomegranate is a devastating disease caused by bacterial pathogen Xanthomonas axonopodis pv. punicae (XAP), recording huge damage to pomegranate crop worldwide. Antibiotics and copper‐based chemicals are being used for the management of this blight, while in this present work, we investigated the effect of eugenol and clove oil either singly or in combination with copper oxychloride (COC) on the induction of plant defense responses and concomitant prevention of bacterial blight. Our results provided evidence that clove oil (0.2%–1%) and eugenol (0.1% and 0.2%) successfully inhibit the growth of XAP in paper disk diffusion assay. Strikingly under the greenhouse condition, clove oil (0.2%) as foliar application 24 h before XAP inoculation recorded the lowest disease severity of 7.34%, whereas eugenol (0.2%) recorded maximum disease severity of 14.56%. However, the combination of clove oil (0.2%) and copper oxychloride (0.3%) recorded the least disease severity of 2.38%. A similar trend was observed in field conditions. Prophylactic application of clove oil leads to enhanced nitrate reductase activity and nitric oxide production which was further enhanced in clove oil pre‐treated plants challenged with XAP. Strikingly, the total ROS and H2O2 levels were reduced in response to clove oil application. Clove oil also induced the systemic response by inducing expression levels of defense genes. The reduction of disease severity by clove oil and COC combination also reflected on total yield recording via large‐scale field experiments where maximum yield of 14.04 tonnes/acre was observed, whereas streptocycline application recorded 11.12 tonnes/acre. Application of COC and clove oil resulted in a high remunerative value of ₹ 1:5.6, compared to streptocycline (1:4.85) and control (1:1.85). The present study revealed that clove oil as a plant derivative and eugenol as a synthetic option can be effectively used for the successful management of bacterial blight in pomegranate.

Bacterial blight (BB) caused by Xanthomonas axonopodis pv. punicae (Xap) is the major scourge in pomegranate cultivation leading to an extensive yield loss up to 60–80%. Hence, identifying a novel resistance source for BB is very necessary for developing a suitable management strategy. Host range analysis and cross-inoculation studies revealed that Xap is specific to pomegranate and there are no alternative hosts to the pathogen. Screening of 149 accessions recorded the varied disease resistance levels with mean disease severity of 30.67%. Accession lines IC318735, IC318724, and IC318762 exhibited maximum disease tolerance by exhibiting the lowest disease severity of 4.91, 5.66, and 6.82%, respectively. Comparative expression analysis of defence genes in IC318724 and IC318735 recorded significant upregulation of phenylalanine ammonia-lyase (PAL), callose synthase-3 (CS3), chitinase, pathogenesis-related protein-1 (PR1), and pathogenesis-related protein-10 (PR10), indicating these genes might be actively involved in conferring disease tolerance. Abiotic elicitors were tested to induce systemic resistance in agronomically superior and widely adapted variety Bhagwa for managing BB of pomegranate. Among the various elicitors tested; proline (600 ppm), gamma-aminobutyric acid (600 ppm), chitosan (600 ppm), β-aminobutyric acid (200 ppm), laminarin (600 ppm), and eugenol (200 ppm) recorded maximum disease protection in prophylactic treatment with disease protection of 89.59, 88.59, 87.15, 86.08, 81.05, and 78.72%, respectively. Similar observations were recorded when these were applied as curative treatment. The present study will broaden our understanding of host–pathogen interactions during BB infection in pomegranate, also aid in developing ideal approach for developing effective disease management. Background Enterococcus faecium though commensal in the human gut, few strains provide a beneficial effect to humans as probiotics while few are responsible for the nosocomial infection. Comparative genomics of E. faecium can decipher the genomic differences responsible for probiotic, pathogenic and non-pathogenic properties. In this study, we compared E. faecium strain 17OM39 with a marketed probiotic, non-pathogenic non-probiotic (NPNP) and pathogenic strains. Results E. faecium 17OM39 was found to be closely related with marketed probiotic strain T110 based on core genome analysis. Strain 17OM39 was devoid of known vancomycin, tetracycline resistance and functional virulence genes. Moreover, E. faecium 17OM39 genome was found to be more stable due to the absence of frequently found transposable elements. Genes imparting beneficial functional properties were observed to be present in marketed probiotic T110 and 17OM39 strains. Genes associated with colonization and survival within gastrointestinal tract was also detected across all the strains. Conclusions Beyond shared genetic features; this study particularly identified genes that are responsible for imparting probiotic, non-pathogenic and pathogenic features to the strains of E. faecium. Higher genomic stability, absence of known virulence factors and antibiotic resistance genes and close genomic relatedness with marketed probiotics makes E. faecium 17OM39 a potential probiotic candidate. The work presented here demonstrates that comparative genome analyses can be applied to large numbers of genomes, to find potential probiotic candidates. The human gut microbiome plays a crucial role in human health and efforts need to be done for cultivation and characterisation of bacteria with potential health benefits. Here, we isolated a bacterium from a healthy Indian adult faeces and investigated its potential as probiotic. The cultured bacterial strain 17OM39 was identified as Enterococcus faecium by 16S rRNA gene sequencing. The strain 17OM39 exhibited tolerance to acidic pH, showed antimicrobial activity and displayed strong cell surface traits such as hydrophobicity and autoaggregation capacity. The strain was able to tolerate bile salts and showed bile salt hydrolytic (BSH) activity, exopolysaccharide production and adherence to human HT-29 cell line. Importantly, partial haemolytic activity was detected and the strain was susceptible to the human serum. Genomics investigation of strain 17OM39 revealed the presence of diverse genes encoding for proteolytic enzymes, stress response systems and the ability to produce essential amino acids, vitamins and antimicrobial compound Bacteriocin-A. No virulence factors and plasmids were found in this genome of the strain 17OM39. Collectively, these physiological and genomic features of 17OM39 confirm the potential of this strain as a candidate probiotic.

Incidences of infection and occurrence ofKocuria rhizophilain human gut are prominent but certainly no reports on the species ability to withstand human gastrointestinal dynamics.Kocuria rhizophilastrain D2 isolated from healthy human gut was comprehensively characterized. The functional analysis revealed the ability to produce various gastric enzymes and sensitive to major clinical antibiotics. It also exhibited tolerance to acidic pH and bile salts. Strain D2 displayed bile-salt hydrolytic (BSH) activity, strong cell surface traits such as hydrophobicity, auto-aggregation capacity and adherence to human HT-29 cell line. Prominently, it showed no hemolytic activity and was susceptible to the human serum. Exploration of the genome led to the discovery of the genes for the above said properties and has ability to produce various essential amino acids and vitamins. Further, comparative genomics have identified core, accessory and unique genetic features. The core genome has given insights into the phylogeny while the accessory and unique genes has led to the identification of niche specific genes. Bacteriophage, virulence factors and biofilm formation genes were absent with this species. Housing CRISPR and antibiotic resistance gene was strain specific. The integrated approach of functional, genomic and comparative analysis denotes the niche specific adaption to gut dynamics of strain D2. Moreover the study has comprehensively characterized genome sequence of each strain to know the genetic difference and intern recognize the effects of on phenotype and functionality complexity. The evolutionary relationship among strains along and adaptation strategies has been included in this study.SignificanceReports of Kocuria rhizophila isolation from various sources have been reported but the few disease outbreaks in humans and fishes have been prominent, but no supportive evidence about the survival ability of Kocuria spp. within human GIT. Here, we report the gut adaption potential of K. rhizophila strain D2 by functional and genomic analysis. Further; comparative genomics reveals this adaption to be strain specific (Gluten degradation). Genetic difference, evolutionary relationship and adaptation strategies have been including in this study.
