Upon drainage, peatlands subside and release stored carbon (C) into the atmosphere. However, little is known about the magnitude of the total, physical and oxidative subsidence (TS, PS and OS) over long historical periods and the past contribution of humans to the degradation observed in contemporary times. Lack of historical subsidence data may hinder contemporary restoration efforts, highlighting the need for insights into past degradation to inform current management practices. Therefore, the present study aims to assess the effect of human activities on the degradation of a raised bog in Denmark in terms of the TS, PS, OS and the C lost between 1880 and 2010 using legacy datasets. The results indicate that 84 % of the bog had been drained by 1997. The TS was 1.6 m on average with a rate of 1.8 cm y-1. A total of 117 million m3 of peat was lost in the 130-year period. The PS was most important shortly after the start of drainage while OS gained importance in the long term. Up to 540 t ha-1 of C-stock was lost at a rate of 6 t ha-1 y-1, translating to 22 t ha-1 y-1 in carbon dioxide equivalents. Therefore, the bog represents a C source in contemporary times, which necessitates targeted and positive management interventions that favour peat growth and C accumulation.

Draining peatlands for agriculture transforms them into significant carbon (C) sources. Restoring drained peatlands is increasingly recognized as a climate action strategy to reduce terrestrial greenhouse gas emissions. Restoration efforts often require accurate inputs, like peat thickness (PT), for C-stock estimation and monitoring; however, these are often lacking or available at suboptimal accuracy levels. In this study, apparent electrical conductivity (ECa) from proximal electromagnetic induction (EMI) surveys and topographic variables derived from a LiDAR-based digital elevation model were assessed as covariates for PT mapping of an agricultural bog, separately and combined, using the quantile random forest algorithm. Local models were trained separately for the large (308 ha) and small (42 ha) EMI surveyed areas, while global models combined data from both areas for a full site analysis. The subsurface was characterized based on resistivity variations in inverted towed transient electromagnetic (tTEM) data. The results indicated that combining topographic and ECa covariates yielded the best PT prediction accuracy for the global model, with a coefficient of determination of 0.61 and a normalized root mean square error (NRMSE) of 0.10. The best large area local model was less accurate than the former (NRMSE of 0.18), while the best small area local model (NRMSE of 0.11) was superior to the best global model. Models trained with only topographic or ECa covariates were the least accurate, especially for the ECa-only model. The tTEM results revealed a heterogenous site characterized by a thin, resistive peat layer overlying stratified postglacial deposits of clay, sand, and saline chalk. Our findings show that covariates characterizing surface and subsurface properties are essential for accurate PT mapping and can inform tailored land use planning and restoration initiatives for degraded peatlands.

The role of mitochondria spans from the regulation of the oxidative phosphorylation, cell metabolism and survival/death pathways to a more recently identified function in chronic inflammation. In stress situations, mitochondria release some pro-inflammatory mediators such as ATP, cardiolipin, reactive oxygen species (ROS) or mitochondrial DNA, that are believed to participate in chronic diseases and aging. These mitochondrial Damage-Associated Molecular Patterns (mito-DAMPs) can modulate specific receptors among which TLR9, NLRP3 and cGAS-STING, triggering immune cells activation and sterile inflammation. In order to counter the development of chronic diseases, a better understanding of the underlying mechanisms of low grade inflammation induced by mito-DAMPs is needed. In this context, monoamine oxidases (MAO), the mitochondrial enzymes that degrade catecholamines and serotonin, have recently emerged as potent regulators of chronic inflammation in obesity-related disorders, cardiac diseases, cancer, rheumatoid arthritis and pulmonary diseases. The role of these enzymes in inflammation embraces their action in both immune and non-immune cells, where they regulate monoamines levels and generate toxic ROS and aldehydes, as by-products of enzymatic reaction. Here, we discuss the more recent advances on the role and mechanisms of action of MAOs in chronic inflammatory diseases.

VEXAS (vacuoles, E1 enzyme, X-linked, auto-inflammatory, somatic) syndrome is an inflammatory disorder with hematological and systemic features. A recent study demonstrated that the dermal infiltrate in neutrophilic dermatosis from VEXAS patients is derived from the pathological UBA1-mutated myeloid clone. Neutrophilic dermatosis is, however, only one of the various skin involvements observed in VEXAS syndrome. We analyzed 10 formalin-fixed paraffin-embedded skin biopsies from genetically confirmed VEXAS syndrome. UBA1 mutation was found in the biopsies related to neutrophilic dermatitis but in none of the other histological patterns (leukocytoclastic vasculitis and septal panniculitis). This could lead to a distinction between clonal and paraclonal cutaneous involvements in VEXAS syndrome, which could in turn improve therapeutic outcomes.

Abstract This study aimed to identify the risk factors for placenta accreta spectrum (PAS) in women who had at least one previous cesarean delivery and a placenta previa or low-lying. The PACCRETA prospective population-based study took place in 12 regional perinatal networks from 2013 through 2015. All women with one or more prior cesareans and a placenta previa or low lying were included. Placenta accreta spectrum (PAS) was diagnosed at delivery according to standardized clinical and histological criteria. Of the 520,114 deliveries, 396 fulfilled inclusion criteria; 108 were classified with PAS at delivery. Combining the number of prior cesareans and the placental location yielded a rate ranging from 5% for one prior cesarean combined with a posterior low-lying placenta to 63% for three or more prior cesareans combined with placenta previa. The factors independently associated with PAS disorders were BMI ≥ 30, previous uterine surgery, previous postpartum hemorrhage, a higher number of prior cesareans, and a placenta previa. Finally, in this high-risk population, the rate of PAS disorders varies greatly, not only with the number of prior cesareans but also with the exact placental location and some of the women's individual characteristics. Risk stratification is thus possible in this population.

This article documents the design and implementation of a culturally responsive critical media literacies curriculum centered around media representations of the Dakota Access Pipeline (DAPL). Students (grades 6-8) were invited to discuss media imagery relating to DAPL and to create memes reflecting their understandings. To situate this work, we articulate a framework that blends critical media literacies and culturally responsive and sustaining pedagogy. We analyze students' spoken and multimodal responses to a curriculum that purposefully foregrounded Native perspectives and digital media. Ultimately, we argue that students must be invited to leverage their epistemic privilege in responding to contemporary social issues.

We explore how valuing Black male students' literacies within academic contexts during multimodal writing can position students' ways of knowing at the center of their learning. This centering requires a repositioning of students' cultural literacies at the core of instruction. Using multiliteracies and Critical Discourse Analysis frameworks, we analyze and share excerpts from conversations with three Black adolescent high school seniors as they composed and reflected upon authoring digital autobiographies for an assignment in their Black Literature class. These reflections illuminate how the students drew on culturally salient texts to share elements of themselves with their peer and teacher audience.

Abstract Coupling the release of pituitary hormones to the developmental stage of the oocyte is essential for female fertility. It requires estrogen to restrain kisspeptin (KISS1)-neuron pulsatility in the arcuate hypothalamic nucleus, while also exerting a surge-like effect on KISS1-neuron activity in the AVPV hypothalamic nucleus. However, a mechanistic basis for this region-specific effect has remained elusive. Our genomic analysis in female mice demonstrate that some processes, such as restraint of KISS1-neuron activity in the arcuate nucleus, may be explained by region-specific estrogen receptor alpha (ERα) DNA binding at gene regulatory regions. Furthermore, we find that the Kiss1-locus is uniquely regulated in these hypothalamic nuclei, and that the nuclear receptor co-repressor NR0B1 (DAX1) restrains its transcription specifically in the arcuate nucleus. These studies provide mechanistic insight into how ERα may control the KISS1-neuron, and Kiss1 gene expression, to couple gonadotropin release to the developmental stage of the oocyte.

Abstract Low-temperature thermochronology is a powerful tool for constraining the thermal evolution of rocks and minerals in relation to a breadth of tectonic, geodynamic, landscape evolution, and natural resource formation processes through deep time. However, complexities inherent to these analytical techniques can make interpreting the significance of results challenging, requiring them to be placed in their geological context in 4-dimensions (3D + time). We present a novel tool for the geospatial archival, analysis and dissemination of fission-track and (U-Th)/He data, built as an extension to the open-access AusGeochem platform ( https://ausgeochem.auscope.org.au ) and freely accessible to scientists from around the world. To demonstrate the power of the platform, three regional datasets from Kenya, Australia and the Red Sea are placed in their 4D geological, geochemical, and geographic contexts, revealing insights into the tectono-thermal evolutions of these areas. Beyond facilitating data interpretation, the archival of fission track and (U-Th)/He (meta-)data in relational schemas unlocks future potential for greater integration of thermochronology and numerical geoscience techniques. The power of formatting data to interface with external tools is demonstrated through the integration of GPlates Web Service with AusGeochem, enabling thermochronology data to be readily viewed in their paleogeographic context through deep time from within the platform.

Pristine peatlands being excellent storage for terrestrial Carbon (C) play a crucial role in regulating climate and water and provide several important ecosystem services. However, peatlands have been heavily altered (e.g., by draining the water table), increasing greenhouse gas (GHG) emissions. Restoring peatlands requires a comprehensive characterization, including knowledge of peat depth (PD; m). Traditionally, this requires the physical insertion of a push probe, which is time-consuming and labor-intensive. It has been shown that non-invasive proximal sensing techniques such as electromagnetic induction and ground penetrating radar can add value to PD data. In this research, we want to assess the potential of proximally sensed gamma-ray (γ-ray) spectrometry (i.e., potassium [K], thorium [Th], uranium [U], and the count rate [CR]) and terrain attributes data (i.e., elevation, slope, SAGAWI, and MRVBF) to map PD either alone or in combination across a small (10 ha) peatland area in ØBakker, Denmark. Here, the PD varies from 0.1 m in the south to 7.3 m in the north. We use various prediction models including ordinary kriging (OK) of PD, linear regression (LR), multiple LR (MLR), LR kriging (LRK), MLR kriging (MLRK) and empirical Bayesian kriging regression (EBKR). We also determine the minimum calibration sample size required by decreasing sample size in decrements (i.e., n = 100, 90, 80,…, 30). We compare these approaches using prediction agreement (Lin’s concordance correlation coefficient; LCCC) and accuracy (root mean square error; RMSE). The results show that OK using maximum calibration size (n = 108) had near perfect agreement (0.97) and accuracy (0.59 m), compared to LR (ln CR; 0.65 and 0.78 m, respectively) and MLR (ln K, Th, CR and elevation; 0.85 and 0.63 m). Improvements are achieved by adding residuals; LRK (0.95 and 0.71 m) and MLRK (0.96 and 0.51 m). The best results were obtained using EBKR (0.97 and 0.63 m) given all predictions were positive and no significant change in agreement and standard errors with the decrement of calibration sample size (e.g., n = 30). The results have implications towards C stocks assessment and improved land use planning to control GHG emissions and slow down global warming.

Non-communicable diseases (NCDs), including coronary heart disease, stroke, hypertension, type 2 diabetes, dementia, depression and cancers, are on the rise worldwide and are often associated with a lack of physical activity (PA). Globally, the levels of PA among individuals are below WHO recommendations. A lack of PA can increase morbidity and mortality, worsen the quality of life and increase the economic burden on individuals and society. In response to this trend, numerous organisations came together under one umbrella in Hamburg, Germany, in April 2021 and signed the ‘Hamburg Declaration’. This represented an international commitment to take all necessary actions to increase PA and improve the health of individuals to entire communities. Individuals and organisations are working together as the ‘Global Alliance for the Promotion of Physical Activity’ to drive long-term individual and population-wide behaviour change by collaborating with all stakeholders in the community: active hospitals, physical activity specialists, community services and healthcare providers, all achieving sustainable health goals for their patients/clients. The ‘Hamburg Declaration’ calls on national and international policymakers to take concrete action to promote daily PA and exercise at a population level and in healthcare settings.

Ulcerative colitis (UC), Crohn’s disease (CD), and celiac disease are prevalent intestinal inflammatory disorders with nonsatisfactory therapeutic interventions. Analyzing patient data-driven cohorts can highlight disease pathways and new targets for interventions. Long noncoding RNAs (lncRNAs) are attractive candidates, since they are readily targetable by RNA therapeutics, show relative cell-specific expression, and play key cellular functions. Uniformly analyzing gut mucosal transcriptomics from 696 subjects, we have highlighted lncRNA expression along the gastrointestinal (GI) tract, demonstrating that, in control samples, lncRNAs have a more location-specific expression in comparison with protein-coding genes. We defined dysregulation of lncRNAs in treatment-naive UC, CD, and celiac diseases using independent test and validation cohorts. Using the Predicting Response to Standardized Pediatric Colitis Therapy (PROTECT) inception UC cohort, we defined and prioritized lncRNA linked with UC severity and prospective outcomes, and we highlighted lncRNAs linked with gut microbes previously implicated in mucosal homeostasis. HNF1A-AS1 lncRNA was reduced in all 3 conditions and was further reduced in more severe UC form. Similarly, the reduction of HNF1A-AS1 ortholog in mice gut epithelia showed higher sensitivity to dextran sodium sulfate–induced colitis, which was coupled with alteration in the gut microbial community. These analyses highlight prioritized dysregulated lncRNAs that can guide future preclinical studies for testing them as potential targets.

Acid sulfate soils can cause environmental damage and geotechnical problems when drained or exposed to oxidizing conditions. This makes them one of the most harmful soils found in nature. In order to reduce possible damage derived from this type of soil, it is fundamental to create occurrence maps showing their localization. Nowadays, occurrence maps can be created using machine learning techniques. The accuracy of these maps depends on two factors: the dataset and the machine learning method. Previously, different machine learning methods were evaluated for acid sulfate soil mapping. To improve the precision of the acid sulfate soil probability maps, in this qualitative modeling study we have added more environmental covariates (17 in total). Since a greater number of covariates does not necessarily imply an improvement in the prediction, we have selected the most relevant environmental covariates for the classification and prediction of acid sulfate soils. For this, we have applied eleven different variable selection methods. The predictive abilities of each group of selected variables have been analyzed using Random Forest and Gradient Boosting. We show that the selection of each environmental covariate as well as the relationship between them are extremely important for an accurate prediction of acid sulfate soils. Among the variable selection methods analyzed, Random Forest stands out, as it is the one that has best selected the relevant covariates for the classification of these soils. Furthermore, the combination of two variable selection methods can improve the prediction of the model. Contrary to the general belief, a low correlation between the covariates does not guarantee a good performance of the model. In general, Random Forest has given better results in the prediction than Gradient Boosting. From the best results obtained, an acid sulfate soils occurrence map has been created. Compared with previous studies in the same area, variable selection has improved the accuracy by 15%–17% for the models based on Random Forest. The present study confirms the importance of variable selection for the prediction of acid sulfate soils.

Introduction: Odontogenic origin is a rare cause (1.3–5%) of cases of orbital cellulitis, but it can lead to very important morbidity such as blindness or thrombosis of the cavernous sinus. It is therefore important to know how to recognize it. Methods: A literature review was performed. The parameters analyzed included age, sex, ethnicity, clinical presentation, imaging to determine orbital involvement, etiology, microbiology, treatment (medical and/or surgical), and final outcome of each case. Chandler's classification was used to classify the different types of cellulitis. Results: Thirty-five cases of odontogenic orbital cellulitis have been described in literature from 1980 to 2022. In 42.9% of cases, the cellulitis corresponded to an intra-orbital abscess (Chandler stage IV). Thrombosis of the cavernous sinus (stage V) was detected in 5.7% of cases. Periorbital edema (100%), ocular or facial pain (82.9%) and limitation of eye movements (82.9%) were the three most common ophthalmological signs. The anamnesis revealed an element pointing to a dental origin in 97.1% of the cases, the two most frequent being a dental avulsion (20%) or an endodontic treatment (14.3%), in days or weeks preceding the onset of symptoms. Imaging was performed on admission in 94.3% of cases. Regarding the most frequently encountered germs, commensal streptococcus of the oral cavity or anaerobic bacteria were found in 25.7% of cases, and coagulase-negative staphylococcus in 22.9% of cases. In 94.3% of cases, broad-spectrum intravenous antibiotic therapy was initiated as soon as the diagnosis was made. The common feature was the use of metronidazole in 51.4% of cases, combined with a third-generation cephalosporin (11.4%) or amoxicillin-clavulanic acid (8.6%). Orbital drainage was necessary in 71.4% of cases to allow resolution of symptoms, associated with drainage of the maxillary sinus in 45.7% of cases. Finally, the treatment allowed a recovery without sequelae in 80% of cases. Discussion: In case of suspected orbital cellulitis, imaging is crucial to confirm the diagnosis, the type of cellulitis and plan the appropriate surgical treatment. The first step of treatment will be the quick start of a broad spectrum intravenous antibiotic therapy, targeting aerobic and anaerobic bacteria. However, it seems imperative to associate a surgical treatment consisting in a first step of an oral drainage and an elimination of the oral infectious source, as well as an orbital drainage whose approach will have been determined by the imaging. Conclusion: Orbital cellulitis is a rare complication of oral cavity infections, but it must be recognized and treated in time to avoid serious morbidity. An early medical and surgical treatment will usually allow good results and a healing process without sequelae.

The process of teaching and learning handwriting is complex and laborious. A digital learning environment in the form of a tablet app could enhance teachers' knowledge of writer characteristics and their management of classroom diversity. This study aims to identify the needs of cursive writing teachers from a survey of 164 kindergarten teachers in France teaching writing skills for children aged 5 to 6 years. The results provide insights into teacher needs and, therefore, can be used to identify the acceptability criteria of a tool designed to scaffold the teaching of writing gestures. The results of needs analyses have been interpreted in the light of new scientific findings on reading and writing skills, and there is a discussion on the potential of such a digital learning environment for the classroom, particularly during the training and skills assessment phases. Questionnaire answers show that the teachers interviewed rarely use digital tools in the classroom and this prompts reflection on the value and meaning that preschool children give to the digital trace.

Abstract Background Non-coding genetic variants that influence gene transcription in pancreatic islets play a major role in the susceptibility to type 2 diabetes (T2D), and likely also contribute to type 1 diabetes (T1D) risk. For many loci, however, the mechanisms through which non-coding variants influence diabetes susceptibility are unknown. Results We examine splicing QTLs (sQTLs) in pancreatic islets from 399 human donors and observe that common genetic variation has a widespread influence on the splicing of genes with established roles in islet biology and diabetes. In parallel, we profile expression QTLs (eQTLs) and use transcriptome-wide association as well as genetic co-localization studies to assign islet sQTLs or eQTLs to T2D and T1D susceptibility signals, many of which lack candidate effector genes. This analysis reveals biologically plausible mechanisms, including the association of T2D with an sQTL that creates a nonsense isoform in ERO1B, a regulator of ER-stress and proinsulin biosynthesis. The expanded list of T2D risk effector genes reveals overrepresented pathways, including regulators of G-protein-mediated cAMP production. The analysis of sQTLs also reveals candidate effector genes for T1D susceptibility such as DCLRE1B, a senescence regulator, and lncRNA MEG3. Conclusions These data expose widespread effects of common genetic variants on RNA splicing in pancreatic islets. The results support a role for splicing variation in diabetes susceptibility, and offer a new set of genetic targets with potential therapeutic benefit.

Dravet syndrome (DS), an intractable childhood epileptic encephalopathy with a high fatality rate, is typically caused by loss-of-function mutations in one allele of SCN1A, which encodes NaV1.1, a 250-kDa voltage-gated sodium channel. In contrast to other epilepsies, pharmaceutical treatment for DS is limited. Here, we demonstrate that viral vector–mediated delivery of a codon-modified SCN1A open reading frame into the brain improves DS comorbidities in juvenile and adolescent DS mice (Scn1aA1783V/WT). Notably, bilateral vector injections into the hippocampus and/or the thalamus of DS mice increased survival, reduced the occurrence of epileptic spikes, provided protection from thermally induced seizures, corrected background electrocorticographic activity and behavioral deficits, and restored hippocampal inhibition. Together, our results provide a proof of concept for the potential of SCN1A delivery as a therapeutic approach for infants and adolescents with DS-associated comorbidities.

Here we report the synthesis of series of bipyridine-based porous polymers as macroligands for molecular copper complexes. The resulting Cu@N-POP solids efficiently catalyze the dehydrogenative N–N coupling of di-p-tolylamine into the corresponding hydrazine using oxygen as the oxidant. The heterogenization of the Cu-based catalyst within porous macroligands improved the catalytic activity with regard to previously reported Cu-based molecular homogeneous systems. Increasing the availability of the bipyridine sites to Cu coordination led to enhanced catalytic performances. [ABSTRACT FROM AUTHOR]

Purpose: This study aims to investigate the ways affective intensities arise in the intra-actions within an assemblage (three Black girls, objects such as computers and hoodies, institutionalized discourse associated with race and successful participation in schools) as the girls create multimodal responses to literature. This paper shows how the intra-actions among the girls and material objects produce affective intensities or new ways of being and becoming through which youth reauthor themselves as central and peripheral participants. Design/methodology/approach: The authors present an illustrative case of the ways girls' embodied literacy identities emerge when Jillian, Isa, and Rhianna intra-act with materials in an assemblage that includes their material-discursive positionings through qualitative and multimodal interaction analysis. Findings: The analysis describes the ways the girls agentively participate through play, composing and moments of becoming (fluid subjectivities) that include emotive acts such as acts of solidarity, loving connectedness and possible frustration that inform who counts and who can be successful in the classroom. Research limitations/implications: This single case study gives a descriptive, in-depth analysis of the ways affective intensities emerge as three girls respond to literature to understand their embodied and discursive practices within the composing process. Originality/value: To fully understand agency and the students' emergent subjectivities, the authors combine embodiment and material-discursive analysis to understand affective intensities that evolve during three Black girls' composing processes and the ways the girls' subjectivities shift within the intra-actions.

Soil provides essential ecosystem services sustaining and improving human life, but mapping soil functions is an ongoing challenge. Denmark has a long history of carrying out soil assessments − originally in order to determine tax revenues for the king, and, more recently, for aiding policymakers and farmers. This knowledge has supported the development of intensive agricultural systems while maintaining the provision of ecosystem services (e.g., clean water). Getting an overview of historical soil surveys and pedological mapping approaches can generate useful information for mapping soil, identifying gaps and proposing directions for future research. In this review, we explore the evolution of soil and environmental inventories, the historical development of soil mapping methods, and how these factors contributed to a better spatial understanding of soil functions. Specifically, we discuss soil functions related to water regulation (e.g., drainage, groundwater and water surface interactions, water table), water filtering (e.g., nitrogen leaching), carbon sequestration (e.g., peatlands), agricultural production (e.g., land suitability, wheat yields), and threats related to soil degradation (e.g., soil erosion). Denmark has benefitted from a government-coordinated approach, promoting detailed and systematic national soil surveys and environmental monitoring programmes. The large databases produced in the surveys formed the basis for mapping several soil properties and functions at increasingly high resolutions over the last many years based on developments in machine learning. In contrast to methodological advances in soil mapping and relevant contributions to pedometric research, we identified a lack of spatial information on soil biodiversity. Detailed spatial information about soil functions is essential to address global issues, such as climate change, food security and water security, and the experience of mapping soil functions in Denmark can be a source of inspiration to other parts of the world.
