Abstract

It has been possible to obtain evidence to support a model for the local accumulation of gephyrin and GlyR under glycinergic nerve terminals. This model gives transmitter-induced activity a key role in receptor clustering. It was found that competitive GlyR antagonist strychnine and L-type Ca2+-channel blockers prevent the accumulation of GlyR and gephyrin at postsynaptic membrane areas in cultured rat spinal neurons. This is consistent with a model in which GlyR activation that leads to Ca2+ influx is needed for the clustering of gephyrin and GlyR at developing postsynaptic sites.

Abstract

The membrane protein syntaxin participates in several protein-protein interactions that have been implicated in neurotransmitter release. To probe the physiological importance of these interactions, we microinjected into the squid giant presynaptic terminal botulinum toxin C1, which cleaves syntaxin, and the H3 domain of syntaxin, which mediates binding to other proteins. Both reagents inhibited synaptic transmission yet did not affect the number or distribution of synaptic vesicles at the presynaptic active zone. Recombinant H3 domain inhibited the interactions between syntaxin and SNAP-25 that underlie the formation of stable SNARE complexes in vitro. These data support the notion that syntaxin-mediated SNARE complexes are necessary for docked synaptic vesicles to fuse.

Introduction of either peptides that mimic SNAP binding sites or recombinant SNAPs in squid giant synapses result in either inhibited or enhanced transmitter release, respectively. This reveals that a SNAP-mediated protein-protein interaction is necessary for transmitter release mechanism which involves constitutive membrane fusion.

Abstract

The formation of glycinergic-rich regions in the dendritic plasma membrane necessitates the presence of gephyrin antisense oligonucleotides which direct the aggregation of GlyR in spiral neurons. Affinity chromatography enables co-purification of the peripheral membrane protein gephyrin receptor (GlyR). The protein is present on the cytoplasmic side of glycinergic postsynaptic membranes and identified during ultrastructural studies. The molecular pathway underlying gephyrin-directed GlyR aggregation, which is yet to be identified, may involve specific binding of gephyrin to tubulin.

48. Lightning overvoltage protection of combined overhead line and underground cable distribution network

Abstract

This paper presents the analysis of lightning overvoltages in a mixed overhead line and underground cable distribution network supplying an industrial consumer. Different overvoltage protection scenarios were discussed, including the installation of surge arresters at both cable ends. Lightning overvoltages in 35 kV distribution network were calculated by using EMTP-ATP software. Open end of a cable was considered in simulations as the most unfavorable switching configuration. Lightning parameters derived from LINET lightning location system observations around 35 kV overhead line were analyzed and compared to the ones used in literature and standards. Lightning flash multiplicity was taken into account in order to determine the probability distribution of lightning current amplitude of the first and subsequent cloud-to-ground strokes. The energy absorbed by surge arresters was determined and an optimal overvoltage protection of the underground cable was proposed.
