Στην εργασία αυτή μελετήθηκε η δυνατότητα ενσωμάτωσης της γονιδιακής κατασκευής Ρ77 των βοοειδών σε ζυγωτά και έμβρυα δυο κυττάρων προβάτων φυλής Χίου. Η γονιδιακή κατασκευή Ρ77 των βοοειδών περιλαμβάνει 2,9 kb από τον προαγωγέα του γονιδίου της α8 1 - καζεΐνης που συγχωνεύθηκαν με το γονίδιο της χυμοζίνης ανάμεσα στο 2 και 9 εξονιο. Η έγχυση της γονιδιακής κατασκευής έγινε με μικροχειριστήριο υπό παρατήρηση σε ανάστροφο μικροσκόπιο σε έναν από τους δυο προπυρήνες του ζυγωτοΰ ή στον πυρήνα του ενός βλαστομεριδίου του εμβρύου. Σε κάθε πυρήνα έγινε μικροέγχυση διαλύματος 1 pi, το οποίο περιείχε περίπου 1000 αντίγραφα της αλληλουχίας DNA. Χρησιμοποιήθηκαν συνολικά 193 ζυγωτά και 87 έμβρυα δυο κυττάρων, τα οποία συλλέχθηκαν από 49 προβατίνες δότες φυλής Χίου. Τα 175 ζυγωτά και τα 76 έμβρυα δυο κυττάρων, στα οποία η μικροέγχυση διενεργήθηκε με επιτυχία, μεταφέρθηκαν με λαπαροσκοπικές τεχνικές στους ωαγωγοΰς 68 προβατίνων δεκτών μετά από in vitro καλλιέργεια 1-5 ωρών. Οι 22 (32,4%) από τις προβατίνες δέκτες γέννησαν 29 αρνιά που αντιστοιχούν στο 11,6% του συνόλου των ζυγωτών - εμβρύων που μεταφέρθηκαν. Η ανάλυση της γενετικής σύστασης των αρνιών που γεννήθηκαν έγινε με τεχνικές PCR σε δείγματα δέρματος. Σε ένα θηλυκό αρνί διαπιστώθηκε ενσωμάτωση της γονιδιακής κατασκευής Ρ77 των βοοειδών στο γονιδίωμά του. Η ανάπτυξη και ενήβωση του διαγονιδιακοΰ αρνιού είναι φυσιολογική, βρίσκεται δε σε κατάσταση εγκυμοσύνης, η οποία εξελίσσεται φυσιολογικά. Είναι το πρώτο διαγονιδιακό παραγωγικό ζώο που γεννήθηκε στην Ελλάδα., The feasibility of integration of the bovine P77 construct into zygotes and two cell embryos of Chios sheep breed was examined. The P77 gene construct comprised bovine as l casein promoter regulatory sequence fused to the fragment between exons 2 and 9 of the bovine chymosin gene. One of the two pronuclei of the zygote or a nucleus of a blastomere of a two cell embryo was microinjected with 1 pi of DNA solution containing approximately 1000 copies of the gene construct. In total 193 zygotes and 87 two cell embryos collected from 49 donor ewes were used. After 1 - 5 h culture microinjected zygotes (n=175) and two cell embryos (n=76) were transferred by a laparoscopic technique to the oviducts of 68 recipients. 22 recipients (32.4%) gave birth to 29 lambs which correspond to 11.6% of the total number of zygotes - embryos transferred. PCR analysis performed in skin samples of these lambs revealed that in one ewe lamb the P77 construct has been integrated in its genome. The growth and puberty of this lamb was physiological and is currently pregnant. This is the first transgenic farm animal born in Greece.

Στην εργασία αυτή διερευνήθηκε η δυνατότητα αποτελεσματικής εφαρμογής μιας απλοποιημένης μεθόδου πρόκλησης πολλαπλής ωοθυλακιορρηξίας (ΠΩ) και συλλογής ωαρίων - ζυγωχών - εμβρύων (Ω-Ζ-Ε) κουνελιών και έγινε συσχέτιση χου αριθμού και της ποιότητάς τους με το χρώμα του βλεννογόνου χου αιδοίου. Για το σκοπό αυτό χρησιμοποιήθηκαν 35 θηλυκά κουνέλια φυλής New Zealand και 5 αρσενικά φυλής California. Η πρόκληση Π Ω έγινε με μία ενδομυϊκή έγχυση 20 IU PMSG /Kg Σ Β. Η Τεχνηχή Σπερματέγχυση (ΤΣ) πραγματοποιήθηκε 3 ημέρες αργότερα ταυτόχρονα με μία ενδοφλέβια έγχυση 120 IU hCG/ζώο. Τα θηλυκά κουνέλια θανατώθηκαν 19h μετά την ΤΣ και τα Ω-Ζ-Ε συλλέχθηκαν με έκπλυση των ωαγωγών. Το 54,29 % των θηλυκών κουνελιών εκδήλωσαν Π Ω (ομάδα Α), χο 40,00 % δεν είχαν την αναμενόμενη (< 10 ωχρά σωμάτια) αντίδραση (ομάδα Β) και το 5,71 % δεν είχαν καμία αντίδραση (ομάδα Γ). Ο μέσος αριθμός των Ω-Ζ-Ε που συλλέχθηκαν από τα θηλυκά κουνέλια της ομάδας Α ήταν 18,53 ± 10,77 και από εκείνα της ομάδας Β ήταν 6,69 ± 5,49 (Ρ, In the present study, the feasibility of the effective use of a simplified method for multiple ovulation and for the collection of oocytes-zygotes-embryos (O-Z-E) was investigated. The relationship between the number and quality of O-Z-E and the color of the mucosa of the vulva was also studied. Thirty five New Zealand does and 5 California bucks were used. Does were given one dose of 20 IU/Kg BW of PMSG i.m. Artificial insemination (AI) and administration of a dose of 120 IU/animal of hCG i.v. were carried out 3 days later. Does were sacrificed 19h after AI and the O-Z-E were collected by flushing of the oviducts. 54,24% of the does presented multiple ovulation (Group A), 40,00% did not present the expected (< 10 corpora lutea) reaction (group B) and 5,71% presented no reaction at all to hormone administration. This study examined the effect of the presence of single or multiple embryos on the transcriptome of the bovine oviduct. In experiment 1, cyclic (nonbred, n = 6) and pregnant (artificially inseminated, n = 11) heifers were slaughtered on Day 3 after estrus, and the ampulla and isthmic regions of the oviduct ipsilateral to the corpus luteum were separately flushed. Oviductal epithelial cells from the isthmus region, in which all oocytes/embryos were located, were snap-frozen for microarray analysis. In experiment 2, heifers were divided into cyclic (nonbred, n = 6) or pregnant (multiple embryo transfer, n = 10) groups. In vitro-produced presumptive zygotes were transferred endoscopically to the ipsilateral oviduct on Day 1.5 postestrus (n = 50 zygotes/heifer). Heifers were slaughtered on Day 3, and oviductal isthmus epithelial cells were recovered for RNA sequencing. Microarray analysis in experiment 1 failed to detect any difference in the transcriptome of the oviductal isthmus induced by the presence of a single embryo. In experiment 2, following multiple embryo transfer, RNA sequencing revealed 278 differentially expressed genes, of which 123 were upregulated and 155 were down-regulated in pregnant heifers. Most of the down-regulated genes were related to immune function. In conclusion, the presence of multiple embryos in the oviduct resulted in the detection of differentially expressed genes in the oviductal isthmus; failure to detect changes in the oviduct transcriptome in the presence of a single embryo may be due to the effect being local and undetectable under the conditions of this study. © 2015 by the Society for the Study of Reproduction, Inc.
