Background: Shattercane [Sorghum bicolor (L.) Moench ssp. Arundinaceum (Desv.)] is a competitive weed in North America's corn, soybean, sorghum, and other agronomic crops. Control of shattercane with POST herbicides in corn became possible with the introduction of acetolactate synthase (ALS)-inhibiting herbicides in the 1980s, and their extensive use resulted in the evolution of ALS-inhibitors resistant shattercane., Results: Shattercane seeds were collected from 16 south-eastern and south-central Nebraska fields that were treated with primisulfuron for three consecutive years. Three resistant plants were found in greenhouse evaluations of more than 30,000 plants. Results from a greenhouse bioassay conducted to assess the response of each shattercane biotype to ALS-inhibiting herbicides showed a differential response to ALS inhibitors within and between chemical classes. Biotype P8-30 was resistant or partially resistant to all ALS-inhibiting herbicides applied and displayed a unique amino acid sequence substitution (Trp574 to Leu) relative to the other two resistant biotypes, P2-205 and P9-102. Whole plant dose-response studies confirmed a 4- to the 12-fold level of primisulfuron resistance in three shattercane biotypes compared with the known primisulfuron-susceptible shattercane biotype. The ALS gene was sequenced using primers designed from the corn ALS sequence to identify mutations in the ALS gene that confer resistance. A total of seven nucleotide substitutions were detected in the three herbicide-resistant biotypes P2-205, P8-30, and P9-102. These biotypes are being crossed to adapted sorghum lines (grain, sweet, and forage) to broaden germplasm with resistance to ALS-inhibiting herbicides., Conclusion: The discovery of these mutants should accelerate the development of sorghum genotypes that tolerate ALS-based herbicides, which provide additional choices for sorghum farmers to control weeds, especially grasses, in their fields., (© 2023. Autophagy is an evolutionarily conserved pathway in eukaryotes that delivers unwanted cytoplasmic materials to the lysosome/vacuole for degradation/recycling. Stimulated autophagy emerges as an integral part of plant immunity against intracellular pathogens. In this study, we used turnip mosaic virus (TuMV) as a model to investigate the involvement of autophagy in plant RNA virus infection. The small integral membrane protein 6K2 of TuMV, known as a marker of the virus replication site and an elicitor of the unfolded protein response (UPR), upregulates the selective autophagy receptor gene NBR1 in a UPR-dependent manner. NBR1 interacts with TuMV NIb, the RNA-dependent RNA polymerase of the virus replication complex (VRC), and the autophagy cargo receptor/adaptor protein ATG8f. The NIb/NBR1/ATG8f interaction complexes colocalise with the 6K2-stained VRC. Overexpression of NBR1 or ATG8f enhances TuMV replication, and deficiency of NBR1 or ATG8f inhibits virus infection. In addition, ATG8f interacts with the tonoplast-specific protein TIP1 and the NBR1/ATG8f-containing VRC is enclosed by the TIP1-labelled tonoplast. In TuMV-infected cells, numerous membrane-bound viral particles are evident in the vacuole. Ceramic membranes suffer from rapid permeability loss during filtration of organic matter due to their fouling propensity. To address this problem, iron oxide ultrafiltration membranes were coated with poly(sulfobetaine methacrylate) (polySBMA), a superhydrophilic zwitterionic polymer. The ceramic-organic hybrid membrane was characterized by scanning electron microscopy (SEM) and optical profilometry (OP). Membranes with and without polySBMA coating were subjected to fouling with bovine serum albumin solution. Hydraulic cleaning was significantly more effective for the coated membrane than for the non-coated one, as 56%, 66%, and 100% of the fouling was removed for the first, second, and third filtration cycle, respectively. Therefore, we can highlight the improved cleaning due to an increased fouling reversibility. Although some loss of polymer during operation was detected, it did not affect the improved behavior of the tested membranes.

Endocytosis and endosomal trafficking play essential roles in diverse biological processes including responses to pathogen attack. It is well established that animal viruses enter host cells through receptor-mediated endocytosis for infection. However, the role of endocytosis in plant virus infection still largely remains unknown. Plant dynamin-related proteins 1 (DRP1) and 2 (DRP2) are the large, multidomain GTPases that participate together in endocytosis. Recently, we have discovered that DRP2 is co-opted by Turnip mosaic virus (TuMV) for infection in plants. We report here that DRP1 is also required for TuMV infection. We show that overexpression of DRP1 from Arabidopsis thaliana (AtDRP1A) promotes TuMV infection, and AtDRP1A interacts with several viral proteins including VPg and cylindrical inclusion (CI), which are the essential components of the virus replication complex (VRC). AtDRP1A colocalizes with the VRC in TuMV-infected cells. Transient expression of a dominant negative (DN) mutant of DRP1A disrupts DRP1-dependent endocytosis and supresses TuMV replication. As adaptor protein (AP) complexes mediate cargo selection for endocytosis, we further investigated the requirement of AP in TuMV infection. Our data suggest that the medium unit of the AP2 complex (AP2β) is responsible for recognizing the viral proteins as cargoes for endocytosis, and knockout of AP2β impairs intracellular endosomal trafficking of VPg and CI and inhibits TuMV replication. Virus-based expression systems have been widely exploited for the production of recombinant proteins in plants during the last thirty years. Advances in technology have boosted scale-up manufacturing of plant-made pharmaceuticals to high levels, via the complementation of transient expression and viral vectors. This combination allows proteins of interest to be produced in plants within a matter of days and thus, is well suited for the development of plant-made vaccines or therapeutics against emerging infectious diseases and potential bioterrorism agents. Several plant-based products are currently in varying stages of clinical development. To investigate the viability of virus-based expression systems for plant-made vaccines against porcine reproductive and respiratory syndrome virus (PRRSV), the most devastating threat to the pork industry in Canada, we cloned the full-length genome of a cucumber green mottle mosaic virus (CGMMV) isolate and developed a CGMMV-based expression vector. We further employed this vector to express the neutralizing epitope (NE) of PRRSV glycoprotein 5 (GP5) in cucumber leaves via agroinfiltration. The coding region of the GP5 NE was inserted downstream of the open reading frame for coat protein (CP) and expressed by a readthrough mechanism. The chimeric virus particles were stable and the expression levels reached as high as 35.84 mg/kg of cucumber leaf fresh weight. This study offers a promising solution to the production of a low cost, versatile and robust vaccine for oral administration against PRRSV through a chimeric virus particle display system., (Crown Copyright © 2019. Positive-sense RNA viruses have a small genome with very limited coding capacity and are highly dependent on host components to fulfill their life cycle. Recent studies have suggested that DEAD-box RNA helicases play vital roles in many aspects of RNA metabolism. To explore the possible role of the RNA helicases in viral infection, we used the Turnip mosaic virus (TuMV)-Arabidopsis pathosystem. The Arabidopsis genome encodes more than 100 putative RNA helicases (AtRH). Over 41 Arabidopsis T-DNA insertion mutants carrying genetic lesions in the corresponding 26 AtRH genes were screened for their requirement in TuMV infection. TuMV infection assays revealed that virus accumulation significantly decreased in the Arabidopsis mutants of three genes, AtRH9, AtRH26, and PRH75. In the present work, AtRH9 was further characterized. Yeast two-hybrid and bimolecular fluorescence complementation (BiFC) assays showed that AtRH9 interacted with the TuMV NIb protein, the viral RNA-dependent RNA polymerase. Moreover, the subcellular distribution of AtRH9 was altered in the virus-infected cells, and AtRH9 was recruited to the viral replication complex. These results suggest that Arabidopsis AtRH9 is an important component of the TuMV replication complex, possibly recruited via its interaction with NIb.

This study evaluated the performance of an algal membrane bioreactor (A-MBR) for secondary wastewater effluent polishing and determined the membrane fouling behavior and dominance of algae in the A-MBR. The continuous flow A-MBR (effective volume = 7.2 L) was operated with low biomass wastage for more than 180 days, resulting in an average algal mixed liquor suspended solid concentration of 4922 mg/L. At the influent concentrations of 43 mg/L COD, 1.6 mg/L total phosphorus (TP), and 11.8 mg/L total nitrogen (TN), the effluent COD, TP and TN concentrations were 26 ± 6 mg/L, 0.7 ± 0.3 mg/L, and 9.6 ± 1.2 mg/L, respectively. High-density algae cultivation facilitated P adsorption and chemical precipitation. However, the TN removal efficiency was only 14% because of low biomass wastage. Although bacteria represented less than 2% of the total biomass in the A-MBR, bacterial growth in the secondary wastewater effluent accelerated membrane fouling.

To realize a fast and selective capture of nano drug vehicles (NDVs) by malignant tumors, an instant affinity switchable NDV based on pH-sensitive surface protonation was prepared. This NDV is prepared from a zwitterionic polymer system with an ultra resistant poly(carboxybetaine) (pCB) shell and sulfo groups near a hydrophobic doxorubicin (DOX) loaded core. The results show that this new NDV system had an instant and sensitive affinity switch from strong resistance in physiological pH to a high affinity to tumor cell membranes in the slightly acidic extracellular pH of malignant tumors depending on if the net charge of the NDV is reversed or not. The pH of this affinity switch can be controlled by the amount of the strong acidic sulfo groups that act as the zeta potential adjustor of the NDV near the inner hydrophobic core. As a result, this CB-based NDV was captured by solid tumor tissue efficiently and thus inhibited tumor growth much more effectively than the PEGylated NDV system, while eliminating unwanted side effects in other healthy tissues. Abstract: A performance comparison between the conventional anaerobic/anoxic/oxic (A2/O) process and the anoxic/anaerobic/oxic (reverse A2/O) process was conducted in lab-scale at the hydraulic retention time of 0.75 d and solids retention time of 10 d for more than 200 days. Both processes demonstrated excellent removal efficiency for organic matter (> 96%) and total nitrogen (> 85%), with no significant difference in microbial activities, sludge phosphorus (P) content, sludge settling property, and nitrifying community (dominated by Nitrosomonas and Nitrospira). However, there was significant difference in P removal, with the total P removal efficiencies in the reverse A2/O and A2/O systems of 76 ± 6% and 70 ± 6%, respectively. Placing an anoxic stage before the anaerobic stage in the reverse A2/O process resulted in a lower oxidation-reduction potential (ORP = -268 ± 45 mV) of the anaerobic zone, which contributed to higher P uptake by bacteria under subsequent aerobic respiration.

The defensive slime of hagfishes contains thousands of intermediate filament protein threads that are manufactured within specialized gland thread cells. The material properties of these threads rival those of spider dragline silks, which makes them an ideal model for biomimetic efforts to produce sustainable protein materials, yet how the thread is produced and organized within the cell is not well understood. Here we show how changes in nuclear morphology, size and position can explain the three-dimensional pattern of thread coiling in gland thread cells, and how the ultrastructure of the thread changes as very young thread cells develop into large cells with fully mature coiled threads. Our model provides an explanation for the complex process of thread assembly and organization that has fascinated and perplexed biologists for over a century, and provides valuable insights for the quest to manufacture high-performance biomimetic protein materials.

An algae-based membrane bioreactor (A-MBR) was evaluated for high-density algae cultivation and phosphorus (P) removal. The A-MBR was seeded with Chlorella emersonii and operated at a hydraulic retention time of 1day with minimal biomass wastage for about 150days. The algae concentration increased from initially 385mg/L (or 315mg biomass COD/L) to a final of 4840mg/L (or 1664mg COD/L), yielding an average solids (algae biomass+minerals) production rate of 32.5gm(-3)d(-1) or 6.2gm(-2)d(-1). The A-MBR was able to remove 66±9% of the total P from the water while the algal biomass had an average of 7.5±0.2% extracellular P and 0.4% of intracellular P. The results suggest that algae-induced phosphate precipitation by algae is key to P removal and high-density algae cultivation produces P-rich algal biomass with excellent settling properties., (Copyright © 2013 Elsevier Ltd. Polyampholyte polymers are a subclass of zwitterionic materials that are composed of mixtures of charged monomer subunits. These materials have properties that mimic those of the more widely characterized zwitterionic polymers as long as their polymer chemistry has been well controlled. In particular, overall neutral polyampholyte polymers have been shown to have resistance to nonspecific protein adsorption and protein conjugation capability. This has been demonstrated for both polymer brush and hydrogel-based platforms. In this review, our recent work demonstrating these characteristics will be summarized and placed into the broader context of zwitterionic materials.

Melamine is a nitrogen-rich (67% nitrogen by mass) heterocyclic aromatic compound that could significantly increase effluent total nitrogen concentrations. In this study, we investigated the degradation of melamine and its impact on activated sludge operations by employing two common activated sludge processes, namely the Modified Ludzack-Ettinger (MLE) process and the continuous stirred tank reactor (CSTR) process. Melamine was dosed continuously from day 125 in both activated sludge treatment systems at an influent concentration of 3 mg/L for about 100 days. Even after such a long period of sludge adaptation, melamine appeared not to be easily biodegradable. The average melamine removal efficiencies in the CSTR and MLE systems were 14 ± 10% and 20 ± 15%, respectively. There was no significant difference in melamine removal between the two different activated sludge processes. The long-term input of melamine resulted in a decrease in the nitrifying bacterial activities (by 82 ± 8%) and population in both systems. Short-term microtiter assay results also showed that melamine reduced activated sludge growth by 80% when supplied at a concentration of 75.6 mg/L. These results suggest that sludge adaptation plays a minimal role in melamine degradation, as the enzymes responsible for hydrolytic deamination of melamine in activated sludge are not easily induced. The insignificant biodegradation of melamine is also attributed to bacterial growth inhibition under long-term dosing conditions with melamine, resulting in a significant decrease in effluent water quality., (Copyright © 2013 Elsevier Ltd. Interpenetrating polymer networks (IPNs) were prepared by the modification of a segmented polyurethane (SPU) with a cross-linked sulfobetaine methacrylate (SBMA) polymer. The IPN films that were prepared can effectively resist nonspecific protein adsorption when the distribution of SBMA units within the SPU film is well controlled, and they retain high mechanical strengths inherent from the base SPU films. Furthermore, the zwitterionic and biomimetic nature of sulfobetaine and the ease of SBMA preparation make SBMA-based materials very attractive for a wide range of applications. It is challenging to control the diffusion of highly polar SBMA into the hydrophobic network of SPU. In this study, various parameters governing the formation of IPNs containing SBMA were studied. The chemical composition depth profile of the IPN films was determined by confocal Raman microscopy. The morphology and thickness of these IPN films were examined by atomic force microscopy and scanning electron microscopy. The amount of adsorbed proteins on the IPN films was determined by an enzyme-linked immunosorbent assay. Results show that the amount of adsorbed proteins on the IPN films depends on the incubation conditions, including solvent polarity, incubation time, SBMA monomer ratio, and incubation concentration. It appears that the IPN films prepared in a mixed solvent of higher polarity with long incubation time lead to very low protein adsorption. This study not only introduces a new IPN system containing SBMA, but also provides a fundamental understanding of various parameters governing the formation of IPNs.
