The 3-30-300 rule offers benchmarks for cities to promote equitable nature access. It dictates that individuals should see three trees from their dwelling, have 30 % tree canopy in their neighborhood, and live within 300 m of a high-quality green space. Implementing this demands thorough measurement, monitoring, and evaluation methods, yet little guidance is currently available to pursue these actions. To overcome this gap, we employed an expert-based consensus approach to review the available ways to measure 3-30-300 as well as each measures strengths and weaknesses. We described seven relevant data and processes: vegetation indices, street level analyses, tree inventories, questionnaires, window view analyses, land cover maps, and green space maps. Based on the reviewed strengths and weaknesses of each measure, we presented a suitability matrix to link recommended measures with each component of the rule. These recommendations included surveys and window-view analyses for the 3 component, high-resolution land cover maps for the 30 component, and green space maps with network analyses for the 300 component. These methods, responsive to local situations and resources, not only implement the 3-30-300 rule but foster broader dialogue on local desires and requirements. Consequently, these techniques can guide strategic investments in urban greening for health, equity, biodiversity, and climate adaptation.

There is an urgent call for science and STEM teachers to incorporate practices for equity-centered environments, social justice-orientations, criticality and other practices that promote system change. Yet this demand occurs against the backdrop marginalization of teachers' from having a say in the planning, teaching, and assessments in their own classrooms as teachers are stripped of agency, authority, and autonomy. We argue that these two simultaneous trends in science and STEM education--the drive for equity-centered environments and the systemic devaluation of teachers' expertise--are incompatible. Realizing the goal of science classrooms that support equity-centered and social justice-oriented learning requires that teachers have equal voice in important decisions on matters that impact their day-to-day actions, which includes the power to determine goals and practices. This means dismantling some of the power structures in the educational system that disenfranchise teachers' from making decisions based on their own knowledge, and their own goals for learning, particularly goals for social-justice and equity. In this article we describe pedagogical frameworks for making transformational change in science classrooms and in wider society and show how each framework is dependent on equity for teachers. We end with implications for this shift--moving toward a system where teachers have equal power--that is an essential part of creating science classrooms as sites of equity, social justice and science learning.

In this paper, we present SCAMP -- Social Configuration Affordances for Museum Play -- an analytical framework we develop and use to highlight the relationship between designed affordances at interactive museum exhibits and different social playful behaviors they trigger and support. We do this through a selective case study analysis of Rainbow Agents, an interactive museum exhibit designed to support play across multiple social configurations. This variety of configurations is valuable for museum settings, as it helps museum visitors engage with each other according to their preferences and also enables the emergence of modes of collaboration and competition novel for learners in such contexts. Our SCAMP analysis of Rainbow Agents sheds light on design features which successfully support different forms of productive social play -- including short and long episodes of competitive, collaborative and parallel play, spanning play, teaching, and receiving interpersonal interactions. In particular, we pay attention to behaviors representing a variety of mentoring and learning opportunities -- in line with and extending the vision and goals of educational games' and science museum exhibits' designers and researchers.

Summary: Overfeeding animals beyond what they eat ad libitum causes rapid adipose tissue expansion, leading to an unusual form of obesity characterized by low immune cell accumulation in fat and sustained anorexia. To investigate how overfeeding affects adipose tissue, we studied the protein secretome of fat from equally obese overfed and ad libitum-fed mice. Fat from overfed animals secretes lower amounts of immune regulatory proteins. Unexpectedly, fat from overfed mice releases larger amounts of mitochondrial proteins. Microscopy identified mitochondria in the conditioned medium of cultured fat that were found not within extracellular vesicles but rather as free extracellular organelles. The protein profile of released mitochondria was distinct from the mitochondrial protein profile of the whole fat, suggesting that the metabolic stress of overfeeding leads to the release of a mitochondrial subset favoring de novo lipogenesis. These findings add to growing evidence that cells alter their energy profiles through the release of mitochondria.

AimsTo characterize the discourse trends and level of argumentation of girls and boys by analysing the differences in the conceptions and systems of conceptions of children and adolescents aged between 6 and 18.Materials and methodsThis is a multi-centre qualitative study in the human and social sciences, based on data collected in two different phases using two different tools: e.Photoexpression©, which deals with health in general, and Photonarration, which deals with cancer. The aim of this open, exploratory method, which uses photographs, is to gather data on the experiences and knowledge specific to each child and adolescent. The informative value of data from qualitative collection tools, combined with a mixed analysis methodology, enabled us to characterize the differences in perceptions of health determinants and cancer between girls and boys aged between 6 and 18.Results4,174 productions were collected from 1,068 children aged 6 to 18, identifying 30 determinants of health and cancer. For all of these results, there were significant gender differences from a very early age: boys focused on leisure activities and physical activity, while girls took a more global view, focusing on the environment, food, emotional aspects of social relationships, hygiene, care, prevention, etc. As they got older, the focus shifted to health determinants. As we get older, we see a change in discourse trends and in the level of argumentation, with girls becoming richer and boys poorer.Discussion and outlookThe trends in girls’ and boys’ discourse on what, in their view, determines health demonstrate the interest and relevance of adapting the prevention methods used as closely as possible to the conceptions of children and adolescents. The differences observed between boys and girls are a crucial lever that takes into account the specific characteristics of a population. They offer the possibility of taking more effective action, both in the context of interventions aimed at teenagers and in support of decision-making in the context of prevention policies.

This paper explores enabling conditions for scaling high-quality project-based learning (PBL) to understand factors that influence how PBL spreads, whether and how it can be sustained and the extent to which it informs meaningful change in schools. We report on a year-long collaboration across three research projects. Each project team analyzed qualitative data from their individual project and then aggregated data across projects to understand similarities and variations in conditions that support the long-term implementation goals of PBL. We used systems mapping as a methodological tool and a case study approach to test and refine the map. We focus on two enabling conditions for PBL that emerged across all contexts: teacher agency and productive disciplinary engagement (PDE). Teachers reported having agency and described making instructional decisions and adapting PBL to support students' needs. PDE motivated teachers to deepen PBL practices. While the studied collaboratory is not the first to pursue shared goals, to our knowledge it is the first to produce research that aggregates knowledge and data across projects. While scaling innovations in schools is complex, the results suggest that certain conditions enable PBL to be implemented with greater depth and can be generalized across contexts. We discuss the implications of this approach for researchers, stakeholders, and practitioners.

Sorg er beskrevet som en emosjonell respons relatert til ulike helseutfordringer. Hensikten med studien var å undersøke hvilke erfaringer enker og enkemenn har med ulike helseutfordringer relatert til sorg og tap og hva som ble opplevd som hjelpsomt. Studien anvendte et eksplorativt design og kvalitativ metode. Et hovedtema, tre tema og to undertema framkom fra analysen. Hovedtema viste behov for selvbestemmelse. I det første tema framkom at både enker og enkemann helst ville klare seg selv og ikke bry andre med sine fysiske helseproblem. Det andre tema viste at tilbaketrekking ble anvendt som en overlevelsesstrategi, mens det tredje tema avdekket at kontakt med andre ga en helsegevinst. Sorgen hadde ingen tidsavgrensning, og fulgte personene gjennom livsløpet. Sosiale aspekter hadde innvirkning på hva som ble opplevd som hjelpsomt.

Modeling microbial interactions as sparse and reproducible networks is a major challenge in microbial ecology. Direct interactions between the microbial species of a biome can help to understand the mechanisms through which microbial communities influence the system. Most state-of-the art methods reconstruct networks from abundance data using Gaussian Graphical Models, for which several statistically grounded and computationnally efficient inference approaches are available. However, the multiplicity of existing methods, when applied to the same dataset, generates very different networks. In this article, we present OneNet, a consensus network inference method that combines seven methods based on stability selection. This resampling procedure is used to tune a regularization parameter by computing how often edges are selected in the networks. We modified the stability selection framework to use edge selection frequencies directly and combine them in the inferred network to ensure that only reproducible edges are included in the consensus. We demonstrated on synthetic data that our method generally led to slightly sparser networks while achieving much higher precision than any single method. We further applied the method to gut microbiome data from liver-cirrothic patients and demonstrated that the resulting network exhibited a microbial guild that was meaningful in terms of human health.

Attenuated measles virus (MV) exerts its oncolytic activity in malignant pleural mesothelioma (MPM) cells that lack type-I interferon (IFN-I) production or responsiveness. However, other cells in the tumor microenvironment (TME), such as myeloid cells, possess functional antiviral pathways. In this study, we aimed to characterize the interplay between MV and the myeloid cells in human MPM. We cocultured MPM cell lines with monocytes or macrophages and infected them with MV. We analyzed the transcriptome of each cell type and studied their secretion and phenotypes by high-dimensional flow cytometry. We also measured transgene expression using an MV encoding GFP (MV-GFP). We show that MPM cells drive the differentiation of monocytes into M2-like macrophages. These macrophages inhibit GFP expression in tumor cells harboring a defect in IFN-I production and a functional signaling downstream of the IFN-I receptor, while having minimal effects on GFP expression in tumor cells with defect of responsiveness to IFN-I. Interestingly, inhibition of the IFN-I signaling by ruxolitinib restores GFP expression in tumor cells. Upon MV infection, cocultured macrophages express antiviral pro-inflammatory genes and induce the expression of IFN-stimulated genes in tumor cells. MV also increases the expression of HLA and costimulatory molecules on macrophages and their phagocytic activity. Finally, MV induces the secretion of inflammatory cytokines, especially IFN-I, and PD-L1 expression in tumor cells and macrophages. These results show that macrophages reduce viral proteins expression in some MPM cell lines through their IFN-I production and generate a pro-inflammatory interplay that may stimulate the patient’s anti-tumor immune response.

Lack of appropriate early diagnostic tools for drug-resistant tuberculosis (DR-TB) and their incomplete drug susceptibility testing (DST) profiling is concerning for TB disease control. Existing methods, such as phenotypic DST (pDST), are time-consuming, while Xpert MTB/RIF (Xpert) and line probe assay (LPA) are limited to detecting resistance to few drugs. Targeted next-generation sequencing (tNGS) has been recently approved by WHO as an alternative approach for rapid and comprehensive DST. We aimed to investigate the performance and feasibility of tNGS for detecting DR-TB directly from clinical samples in Bangladesh. pDST, LPA and tNGS were performed among 264 sputum samples, either rifampicin-resistant (RR) or rifampicin-sensitive (RS) TB cases confirmed by Xpert assay. Resistotypes of tNGS were compared with pDST, LPA and composite reference standard (CRS, resistant if either pDST or LPA showed a resistant result). tNGS results revealed higher sensitivities for rifampicin (RIF) (99.3%), isoniazid (INH) (96.3%), fluoroquinolones (FQs) (94.4%), and aminoglycosides (AMGs) (100%) but comparatively lower for ethambutol (76.6%), streptomycin (68.7%), ethionamide (56.0%) and pyrazinamide (50.7%) when compared with pDST. The sensitivities of tNGS for INH, RIF, FQs and AMGs were 93.0%, 96.6%, 90.9%, and 100%, respectively and the specificities ranged from 91.3 to 100% when compared with CRS. This proof of concept study, conducted in a high-burden setting demonstrated that tNGS is a valuable tool for identifying DR-TB directly from the clinical specimens. Its feasibility in our laboratory suggests potential implementation and moving tNGS from research settings into clinical settings.

In this design case, we describe The Handwavey Game (Handwavey)--a tabletop, cooperative role-playing game created to study how people can come to converge on novel physical gestures around meaning. In Handwavey, players are novice wizards who cast spells through signaling abstract images with hand movements: success is rewarded in-game and failure has humorous in-game consequences. This case walks through the path of exploration and development from the starting point of a research question to the development of game mechanics and concludes with a set of design recommendations for people interested in designing novel games with specific research or learning meta-goals.

Abstract This study evaluates the coded aperture imaging method for pitch angle observations of magnetospheric energetic electrons in the solar, Earth, and planetary space environments. We present a review of key previous energetic electron instruments with pitch angle‐resolved observations across a range of electron energies. We describe the coded aperture imaging method, typically used for high angular resolution X‐ray and gamma ray observations, and evaluate design parameters in the context of energetic electron observations. We present the results of simulations of energetic electrons in Geant4 and evaluate the method's ability to resolve sources with high angular and temporal resolution. We also evaluate the impact of secondary radiation produced from electron interactions in the tungsten coded aperture, as well as the impact of artifacts from the decoding process. With these simulated results, we identify key areas in magnetospheric science that would benefit from high angular resolution observations of energetic electrons. We find that coded aperture imaging may be well‐suited for high‐resolution observations of intense localized structures, such as low energy (tens of eV to several keV) field‐aligned electron beams or the electron strahl wind.

A driving factor in designing interactive museum exhibits to support simultaneous users is that visitors learn from one another via observation and conversation. Researchers typically analyze such collaborative interactions among museumgoers through manual coding of live- or video-recorded exhibit use. We sought to determine how log data from an interactive multi-user exhibit could indicate patterns in visitor interactions that could shed light on informal collaborative learning. We characterized patterns from log data generated by an interactive tangible tabletop exhibit using factors like "pace of activity" and the timing of "success events." Here we describe processes for parsing and visualizing log data and explore what these processes revealed about individual and group interactions with interactive museum exhibits. Using clustering techniques to categorize museumgoer behavior and heat maps to visualize patterns in the log data, we found distinct trends in how users solved the exhibit. Some players seemed more reflective, while others seemed more achievement oriented. We also found that the most productive sessions occurred when players occupied all four areas of the table, suggesting that the activity design had the desired outcome of promoting collaborative activity.

Educating nurses in developing countries is challenging due to limited resources. Nurses International advances nursing education through the use of Open Educational Resources, which provide a unique model to teach and mentor nurse educators globally. Using current resources and materials that are free online and teacher-facing, an innovative program has been established that can be employed to support development of and scale education for the healthcare workforce, through teaching the teachers and enhancing the education of nurses.

Marine sediments can be used to reconstruct the evolution of seawater [SO42-] and δ34S over time, two key parameters that contribute to refine our understanding of the sulfur cycle and thus of Earth's redox state. δ34S evolution can be measured from carbonates, barites and sulfate evaporites. [SO42-] variations can be reconstructed using fluid inclusions in halites, a method that only allows a low-resolution record. Reconstruction of the past sulfur cycle could be improved if carbonates allowed the tracking of both seawater δ34S and [SO42-] variations in a sole, continuous sedimentary repository. However, most primary carbonates formed in the ocean are biogenic, and organisms tend to overprint the geochemical signatures of their carbonates through a combination of processes often collectively referred to as vital effects. Hence, calibrations are needed to allow seawater δ34S and [SO42-] reconstructions based on biogenic carbonates. Because foraminifera are important marine calcifiers, we opted to focus on calcite synthesized by individuals of rosalinid benthic foraminifera cultured in the laboratory under controlled conditions, with varying seawater [SO42-] (ranging from 0 to 180 mM). Our experimental design allowed us to obtain foraminiferal asexual reproduction over several generations. We measured bulk carbonate-associated sulfate (CAS) content and sulfur isotopic composition (δ34SCAS) on samples of tens to hundreds of specimens from a selection of culture media, where [SO42-] varied from 5 to 60 mM. Increasing or decreasing [SO42-] with respect to modern-day seawater concentration (28 mM) impacted foraminiferal population size dynamics and the total amount of bioprecipitated carbonate. Foraminiferal CAS concentration increased proportionally with [SO42-] concentration from 5 mM up to 28 mM and then showed a plateau from 28 to 60 mM. The existence of a threshold at 28 mM is interpreted as the result of a control on the precipitation fluid chemistry that foraminifera exert on the carbonate precipitation loci. However, at high seawater sulfate concentrations (> 40 mM) the formation of sulfate complexes with other cations may partially contribute to the non-linearity of the CAS concentration in foraminiferal tests at high increases in [SO42-]. Yet, despite the significant effect of seawater [SO42-] on foraminiferal reproduction and on CAS incorporation, the isotopic fractionation between CAS and seawater remains stable through varying seawater [SO42-]. Altogether, these results illustrate that CAS in biogenic calcite could constitute a good proxy for both seawater [SO42-] and δ34S and suggests that sulfate likely plays a role in foraminiferal biomineralization and biological activity.

The past decade has witnessed a revolution in cancer treatment, shifting from conventional drugs (chemotherapies) towards targeted molecular therapies and immune-based therapies, in particular immune-checkpoint inhibitors (ICIs). These immunotherapies release the host’s immune system against the tumor and have shown unprecedented durable remission for patients with cancers that were thought incurable, such as metastatic melanoma, metastatic renal cell carcinoma (RCC), microsatellite instability (MSI) high colorectal cancer and late stages of non-small cell lung cancer (NSCLC). However, about 80% of the patients fail to respond to these immunotherapies and are therefore left with other less effective and potentially toxic treatments. Identifying and understanding the mechanisms that enable cancerous cells to adapt to and eventually overcome therapy can help circumvent resistance and improve treatment. In this review, we describe the recent discoveries on the onco-immunological processes which govern the tumor microenvironment and their impact on the resistance to PD-1/PD-L1 checkpoint blockade.

Abstract Energetic electron precipitation (EEP) from the radiation belts into Earth's atmosphere leads to several profound effects (e.g., enhancement of ionospheric conductivity, possible acceleration of ozone destruction processes). An accurate quantification of the energy input and ionization due to EEP is still lacking due to instrument limitations of low‐Earth‐orbit satellites capable of detecting EEP. The deployment of the Electron Losses and Fields InvestigatioN (ELFIN) CubeSats marks a new era of observations of EEP with an improved pitch‐angle (0°–180°) and energy (50 keV–6 MeV) resolution. Here, we focus on the EEP recorded by ELFIN coincident with electromagnetic ion cyclotron (EMIC) waves, which play a major role in radiation belt electron losses. The EMIC‐driven EEP (∼200 keV–∼2 MeV) exhibits a pitch‐angle distribution (PAD) that flattens with increasing energy, indicating more efficient high‐energy precipitation. Leveraging the combination of unique electron measurements from ELFIN and a comprehensive ionization model known as Boulder Electron Radiation to Ionization (BERI), we quantify the energy input of EMIC‐driven precipitation (on average, ∼3.3 × 10−2 erg/cm2/s), identify its location (any longitude, 50°–70° latitude), and provide the expected range of ion‐electron production rate (on average, 100–200 pairs/cm3/s), peaking in the mesosphere—a region often overlooked. Our findings are crucial for improving our understanding of the magnetosphere‐ionosphere‐atmosphere system as they accurately specify the contribution of EMIC‐driven EEP, which serves as a crucial input to state‐of‐the‐art atmospheric models (e.g., WACCM) to quantify the accurate impact of EMIC waves on both the atmospheric chemistry and dynamics.

Background For high bleeding-risk patients (HBR) undergoing percutaneous coronary intervention (PCI), the LEADERS FREE (LF) and LEADERS FREE II (LF II) trials established the safety and efficacy of a stainless steel polymer-free biolimus-coated stent (SS-BCS) with 30 days of dual antiplatelet treatment (DAPT). The LEADERS FREE III (LF III) trial investigated clinical outcomes after PCI with the next-generation cobalt-chromium thin-strut polymer-free biolimus-coated stent (CoCr-BCS) in HBR patients.Aims To report the final 3-year results of the LF III trial and compare them to LF II.Methods LF III was a prospective, multicentre, open-label single-arm study to evaluate the safety and efficacy of the CoCr-BCS stent. The primary safety endpoint was the composite of cardiac death (CD), myocardial infarction(MI) or definite/probable stent thrombosis (ST). The primary efficacy endpoint was clinically driven target lesion revascularisation (cd-TLR). We performed a propensity-matched comparison to the 3-year outcomes of LF II.Results After 3 years, CD/MI/ST had occurred in 57 patients (15%, 95% CI 11.8% to 19%) and cd-TLR in 23 (6.2%, 95% CI 4.1% to 9.2%) patients. In a propensity-matched comparison of patients treated with the CoCr-BCS versus the SS-BCS, there were similar rates of CD (6.6% vs 7.8%, p=0.50), MI (7.1% vs 8.3%, p=0.47) and definite/probable ST (1.1% vs 2%, HR 0.56, 95% CI 0.16 to 1.93, p=0.35). The rates of cd-TLR were 5.3% with CoCr-BCS versus 9.8% with SS-BCS (HR 0.54, 95% CI 0.31 to 0.96, p=0.03).Conclusion LF III confirms the long-term safety and efficacy of the CoCr-BCS in HBR patients treated with 1 month of DAPT.Trial registration number NCT02843633, NCT03118895.
