Phylogenetic relationships among 74 accessions belonging to six species of Phaseolus are investigated using variation in chloroplast DNA assessed according to a PCR-RFLP protocol. Three fragments of chloroplast DNA are amplified using universal primers, and then digested with 10 restriction enzymes. Thirty-six haplotypes are identified on the basis of the polymorphism in fragment number and size. Three main phylogenetic groups, strongly supported through bootstrap analysis, are identified: (1) accessions from Phaseolus lunatus and Phaseolus xolocotzii; (2) accessions from Phaseolus glabellus; (3) accessions from Phaseolus vulgaris, Phaseolus polyanthus and Phaseolus coccineus. Within the third group, accessions of Phaseolus coccineus are scattered along the phylogenetic tree, which provides some evidence that coccineus accessions are paraphyletic with respect to Phaseolus vulgaris and Phaseolus polyanthus. An analysis of molecular variance applied on four species show that they are significantly differentiated with 79% of molecular variance among species and 21% within species. The results agree with previous investigations on chloroplast DNA variation in the genus Phaseolus, and suggest that PCRRFLP methods, which are technically less labour-intensive than previous methods, are of great value for phylogenetic investigations at the generic level.

Background: Turkey is one of the major exporters of mandarins in the Mediterranean region. Seedlessness in citrus, which is one of the most desired fruit quality traits, especially in fresh mandarin export markets, can be obtained via triploidy as in many fruit species. Triploid plants can be recovered by 2x × 2x hybridizations in citrus, as well as 2x × 4x and 4x × 2x crosses. Hence, we aimed to develop local triploid hybrids by using the embryo rescue technique in five crosses using eight different citrus varieties in the present study., Methods and Results: Embryos isolated from abortive seeds derived by 135 days after pollinations were cultured on modified Murashige and Tucker (MT) medium by adding different levels of GA 3 to achieve a high germination rate. A population of 574 plants was developed as a result of embryo rescue. After screening the ploidy levels of this 574-plant population with the aid of flow cytometry, 4 triploids from 'Encore' × 'Murcott', 8 triploids from 'Fortune' × 'Willow leaf', 1 triploid from 'Kiyomi' × 'Murcott', and 1 triploid from 'Ortanique' × 'Murcott' hybridization were recovered. Triploid hybrid plants and related parents were analyzed with SSR markers heterozygotic for parental mandarin varieties. In addition, we evaluated stomatal characteristics of diploid and triploid hybrids obtained from different crosses. Stomatal traits of diploid and triploid hybrids in all crosses significantly differed except the stomata index., Conclusions: Genotyping of triploid plants confirmed using Simple Sequence Repeat (SSR) molecular markers and five SSRs were able to identify three alleles of triploid hybrids. Selected triploid mandarin hybrids have been grafted on several rootstocks for field trials and are in the process of yield and quality performances., (© 2022. This study examines the mechanistic processes governing multiphase flow of a water-cosolvent-NAPL system in saturated porous media. Laboratory batch and column flushing experiments were conducted to determine the equilibrium properties of pure NAPL and synthetically prepared NAPL mixtures as well as NAPL recovery mechanisms for different water-ethanol contents. The effect of contact time was investigated by considering different steady and intermittent flow velocities. A modified version of multiphase flow simulator (UTCHEM) was used to compare the multiphase model simulations with the column experiment results. The effect of employing different grid geometries (1D, 2D, 3D), heterogeneity and different initial NAPL saturation configurations was also examined in the model. It is shown that the change in velocity affects the mass transfer rate between phases as well as the ultimate NAPL recovery percentage. The experiments with low flow rate flushing of pure NAPL and the 3D UTCHEM simulations gave similar effluent concentrations and NAPL cumulative recoveries. Model simulations over-estimated NAPL recovery for high specific discharges and rate-limited mass transfer, suggesting a constant mass transfer coefficient for the entire flushing experiment may not be valid. When multi-component NAPLs are present, the dissolution rate of individual organic compounds (namely, toluene and benzene) into the ethanol-water flushing solution is found not to correlate with their equilibrium solubility values., (Copyright © 2012 Elsevier B.V. This paper examines the potential for enhanced NAPL recovery from the subsurface through the combined application of hot water and cosolvent flushing. Batch experiments were conducted to determine the effect of temperature on fluid properties and the multiphase behavior of the ethanol-water-toluene system and to assess the impact of temperature on the capillary, Bond and total trapping numbers and on flooding stability. Column flooding experiments were also conducted to evaluate toluene NAPL recovery efficiency for different ethanol contents and flushing solution temperatures. The ethanol content considered ranged from 20 to 100% by mass, while the flushing solution temperatures were varied from 10 to 40°C. It is shown that small variations in the system temperature can strongly influence the solubilization, mobilization and stability of the multiphase system, but that the impact of temperature on the enhanced NAPL recovery is also dependent on the ethanol content of the flushing solution. The impact of hot water on NAPL recovery was most pronounced at intermediate ethanol contents (40-60% by mass) where the increase in system temperature led to enhanced NAPL solubilization as well as NAPL mobilization. This study demonstrates that coupling of hot water with in situ cosolvent flooding is a potentially effective remedial alternative that can optimize NAPL recovery while reducing the amount of chemicals injected into the subsurface., (Copyright © 2010 Elsevier B.V.
