In recent decades, controversies have arisen identifying internationally acceptable cut points using fasting plasma glucose (FPG), 2-h post-load glucose (2-h PG), and/or HbA1c for defining intermediate hyperglycemia (prediabetes). Despite this, there has been a steadfast global consensus of the 2-h PG for defining dysglycemic states during the OGTT. This article reviews the history of the OGTT and recent advances in its application, including the glucose challenge test and mathematical modeling for determining the shape of the glucose curve. Pitfalls of the FPG, 2-h PG during the OGTT, and HbA1c are considered as well. Finally, the associations between the 30-minute and 1-hour plasma glucose (1-h PG) levels derived from the OGTT and incidence of diabetes and its complications will be reviewed. The considerable evidence base supports modifying current screening and diagnostic recommendations with the use of the 1‐h PG. Measurement of the 1‐h PG level could increase the likelihood of identifying high-risk individuals when the pancreatic ß-cell function is substantially more intact with the added practical advantage of potentially replacing the conventional 2‐h OGTT making it more acceptable in the clinical setting.Keywords: OGTT history, OGTT, glycated hemoglobin, diabetes, gestational diabetes, pathophysiology, 1-h post-load glucose, prediction, shape index

2. Essential oil of Siparuna guianensis as an alternative tool for improved lepidopteran control and resistance management practices

Abstract

Abstract Although the cultivation of transgenic plants expressing toxins of Bacillus thuringiensis (Bt) represents a successful pest management strategy, the rapid evolution of resistance to Bt plants in several lepidopteran pests has threatened the sustainability of this practice. By exhibiting a favorable safety profile and allowing integration with pest management initiatives, plant essential oils have become relevant pest control alternatives. Here, we assessed the potential of essential oils extracted from a Neotropical plant, Siparuna guianensis Aublet, for improving the control and resistance management of key lepidopteran pests (i.e., Spodoptera frugiperda and Anticarsia gemmatalis). The essential oil exhibited high toxicity against both lepidopteran pest species (including an S. frugiperda strain resistant to Cry1A.105 and Cry2Ab Bt toxins). This high insecticidal activity was associated with necrotic and apoptotic effects revealed by in vitro assays with lepidopteran (but not human) cell lines. Furthermore, deficits in reproduction (e.g., egg-laying deterrence and decreased egg viability), larval development (e.g., feeding inhibition) and locomotion (e.g., individual and grouped larvae walking activities) were recorded for lepidopterans sublethally exposed to the essential oil. Thus, by similarly and efficiently controlling lepidopteran strains susceptible and resistant to Bt toxins, the S. guianensis essential oil represents a promising management tool against key lepidopteran pests.

Therefore, effective methods for diagnosing prediabetes will be required to reduce the risk of progressing to diabetes and its complications. The current biomarkers, glycated hemoglobin (HbA1c), fructosamine, and glycated albumin have limitations including moderate sensitivity and specificity and are inaccurate in certain clinical conditions. Therefore, identification of additional biomarkers is being explored recogni­zing that any single biomarker will also likely have inherent limitations. Therefore, combining several biomarkers may more precisely identify those at high risk for developing prediabetes and subsequent progression to diabetes. This review describes recently identified biomarkers and their potential utility for addressing the burgeoning epidemic of dysglycemic disorders. Abstract

Heightened attention towards negative information is characteristic of depression. Evidence is emerging for a negative attentional bias in Autism spectrum disorder (ASD), perhaps driven by the high comorbidity between ASD and depression. We investigated whether ASD is characterised by a negative attentional bias and whether this can be explained by comorbid (sub) clinical depression. Participants (n = 116) with current (CD) or remitted depression (RD) and/or ASD, and 64 controls viewed positively and negatively valenced (non-)social pictures. Groups were compared on three components of visual attention using linear mixed models. Both CD individuals with and without ASD, but not remitted depressed and never-depressed ASD individuals showed a negative bias, suggesting that negative attentional bias might be a depressive state-specific marker for depression in ASD.

Abstract

The impact of heat stress (HS) on production is intricately linked with feed intake. We investigated the effects of HS on intestines and diencephalic genes in Pekin ducks. One hundred and sixty adult ducks were allocated to two treatment rooms. The control room was maintained at 22°C and the HS room at 35°C for the first 10 h of the day then reduced to 29.5°C. After 3 weeks, 10 hens and 5 drakes were euthanized from each room and jejunum and ileum collected for histology. Brains were collected for gene expression analysis using qRT‐PCR. Intestinal morphology data were analyzed with two‐way ANOVA and diencephalic gene data were analyzed with Kruskal–Wallis test. There was an increase in villi width in the ileum (p = .0136) and jejunum (p = .0019) of HS hens compared to controls. HS drakes showed a higher crypt depth (CD) in the jejunum (p = .0198) compared to controls. There was an increase in crypt goblet cells (GC) count in the ileum (p = .0169) of HS drakes compared to HS hens. There was higher villi GC count (p = .07) in the jejunum of HS drakes compared to controls. There was an increase in the crypt GC density (p = .0054) in the ileum, not jejunum, of HS drakes compared to HS hens. Further, there were no differences in the proopiomelanocortin gene expression in either sex but there was an increase in the expression of neuropeptide Y (NPY) gene in HS hens (p = .031) only and a decrease in the corticotropin releasing hormone gene in the HS drakes (p = .037) compared to controls. These data show that there are sex differences in the effect of HS on gut morphology while the upregulation in NPY gene may suggest a role in mediating response to chronic HS. [ABSTRACT FROM AUTHOR]

14. On communicating extruded aluminium surface quality along the supply chain : a customer approach to sustainable surfaces

Today, far too many products are scrapped due to surface related issues, products with perfect function but with minor surface blemishes. The complaints are often offset by goodwill commitments from suppliers at great cost to them and delivery delays and lead time costs for customers. The reason is that the industry relies on several non-standardized classification systems for surface quality that are based on various combinations of and designations for surface defects, assessed by visual inspections at a defined distance to determine the severity of any detected surface deviations. These similar classification systems provide far too much scope for subjective and non-repeatable assessments causing communication problems between customer and producer at all stages in the supply chain. To challenge this situation, a common toolbox to communicate, describe and define surface quality should be developed, i.e. a standardisation of surface quality assessment including various effects and defects with a jointly established nomenclature and evaluation parameters. This work presents the first step of a research project bringing together 11 suppliers and OEMs along the supply chain, from the delivery of raw aluminium to finished alumina profiles included in consumer products. The final goal of the project is to develop an ‘objective classification of visual requirements’ on alumina profiles towards increased sustainability and decreased material wastage. Presented result is a common terminology with links to the process chain, surface defect geometry and visual appearance aiming at making the communication between producers and buyers of the aluminium profiles clearer and more unambiguous when it comes to specification and requirements of profile surfaces in each of the supply-chain links. Future work will add measurable parameters specifying surface quality.

15. Acculturation, identity, and intergroup relations of Mexican Americans and European Americans

Abstract

This thesis explores the dynamic interaction between identity and intergroup relations. More specifically, it focuses on how members from a particular ethnic group represent their self, their ingroup, and a salient outgroup, and how these representations are linked to attitudinal and behavioural statements toward that outgroup. The first four chapters of this work examine theoretical, conceptual, and methodological issues, and suggest possible improvements with respect to social psychological theory and research on ethnic identity and intergroup relations, especially as these relate to intergroup contacts and acculturation. While ethnicity is generally understood as a powerful explanatory variable - accounting for variations in attitudes, behaviours, and identities - it is argued here that within-group variations deserve equal attention, since these may reveal a considerable divergence in identities and experiences of intergroup relations despite the constraints imposed by ethnic group membership. Such bifocal considerations provide important insights into the dynamics of identity formation as well as cooperation and conflicts between groups. The subsequent three chapters apply these theoretical and methodological proposals empirically. More precisely, the empirical research explores the propositions with data collected from Mexican Americans and European Americans residing in the south-west of the US. Four types of evidence are used for the purposes of this thesis. Existing literature not only provides an account of the variations of identity and intergroup relations, but also exemplifies variations in framing and positioning in terms of identity and intergroup relations. Ethnographic data reveal additional insights into the structures and processes considered in this work. Third, interview data serve not only to formulate theory and hypotheses but also to refine the interpretation of the statistical results. Finally, questionnaire responses from 201 European Americans and 93 Mexican Americans permit testing and exploration of the theoretical and conceptual propositions.

16. Analysis of Base-Heating Environment During Ground Testing of a Lunar Lander Demonstrator

Abstract

A three-dimensional, unstructured-grid, pressure-based, reacting flow, computational fluid dynamics (CFD) and heat transfer methodology was employed to study the base-heating environment of a lunar lander demonstrator, during terrestrial ground testing. Two base-heating environments were investigated: lunar lander demonstrator sitting on pad, and lunar lander demonstrator hovering at a distance above ground. Two unique and quite different base-flow physics are found for these two cases. In the case of demonstrator sitting on pad, the computed heat fluxes are high and the base flow is unsteady, caused by a Coanda effect precursor that makes the fountain jet oscillate about the center of the base. For the second case, due to the higher elevation of the demonstrator and the layout of the nozzles, the Coanda effect forces the fountain jet to be attached to two of the nozzle plumes, resulting in much lower computed base heat fluxes.

17. What Is a Dysfunctional School?

Abstract

Whether or not a school is dysfunctional depends largely on how dysfunctionality in schools is defined and measured. Dysfunctionality, as any construct, is subject to definition and interpretation, and it is thus always marked by perspectivism. But regardless of the definition games occasionally played by academics, some form of reality takes place beyond the constructivist horizon. Moreover, perspectivism and definition games may justify and even create adverse consequences within a school's lived reality. A definition focussing on Grade 12 pass rates, for example, tends to implicate teachers, while a definition focussing on infrastructure tends to exculpate teachers and incriminate government and administration. Accordingly, this article will not end but begin with the adage that a dysfunctional school is what one defines a dysfunctional school to be, and it will explore the conditions and consequences of conceptual variations.

Abstract

While mRNA vaccines are administrated worldwide in an effort to contain the COVID-19 pandemic, the heterogeneity of the humoral immune response they induce at the population scale remains unclear. Here, in a prospective, longitudinal, cohort-study, including 1245 hospital care workers and 146 nursing home residents scheduled for BNT162b2 vaccination, together covering adult ages from 19 to 99 years, we analyse seroconversion to SARS-CoV-2 spike protein and amount of spike-specific IgG, IgM and IgA before vaccination, and 3-5 weeks after each dose. We show that immunogenicity after a single vaccine dose is biased to IgG, heterogeneous and reduced with increasing age. The second vaccine dose normalizes IgG seroconversion in all age strata. These findings indicate two dose mRNA vaccines is required to reach population scale humoral immunity. The results advocate for the interval between the two doses not to be extended, and for serological monitoring of elderly and immunosuppressed vaccinees.

Abstract

Background: androgen receptor (AR) is critical to the initiation, growth, and progression of prostate cancer. Once activated, the AR binds to cis-regulatory enhancer elements on DNA that drive gene expression. Yet, there are 10-100x more binding sites than differentially expressed genes. It is unclear how or if these excess binding sites impact gene transcription. Results: to characterize the regulatory logic of AR-mediated transcription, we generated a locus-specific map of enhancer activity by functionally testing all common clinical AR binding sites with Self-Transcribing Active Regulatory Regions sequencing (STARRseq). Only 7% of AR binding sites displayed androgen-dependent enhancer activity. Instead, the vast majority of AR binding sites were either inactive or constitutively active enhancers. These annotations strongly correlated with enhancer-associated features of both in vitro cell lines and clinical prostate cancer samples. Evaluating the effect of each enhancer class on transcription, we found that AR-regulated enhancers frequently interact with promoters and form central chromosomal loops that are required for transcription. Somatic mutations of these critical AR-regulated enhancers often impact enhancer activity. Conclusions: using a functional map of AR enhancer activity, we demonstrated that AR-regulated enhancers act as a regulatory hub that increases interactions with other AR binding sites and gene promoters.

Abstract

Serological assays are valuable tools to study SARS-CoV-2 spread and, importantly, to identify individuals that were already infected and would be potentially immune to a virus reinfection. SARS-CoV-2 Spike protein and its receptor binding domain (RBD) are the antigens with higher potential to develop SARS-CoV-2 serological assays. Moreover, structural studies of these antigens are key to understand the molecular basis for Spike interaction with angiotensin converting enzyme 2 receptor, hopefully enabling the development of COVID-19 therapeutics. Thus, it is urgent that significant amounts of this protein became available at the highest quality. In this study, we produced Spike and RBD in two human derived cell hosts: HEK293-E6 and Expi293F™. We evaluated the impact of different and scalable bioprocessing approaches on Spike and RBD production yields and, more importantly, on these antigens' quality attributes. Using negative and positive sera collected from human donors, we show an excellent performance of the produced antigens, assessed in serologic enzyme-linked immunosorbent assay (ELISA) tests, as denoted by the high specificity and sensitivity of the test. We show robust Spike productions with final yields of approx. 2 mg/L of culture that were maintained independently of the production scale or cell culture strategy. To the best of our knowledge, the final yield of 90 mg/L of culture obtained for RBD production, was the highest reported to date. An in-depth characterization of SARS-CoV-2 Spike and RBD proteins was performed, namely the antigen's oligomeric state, glycosylation profiles, and thermal stability during storage. The correlation of these quality attributes with ELISA performance show equivalent reactivity to SARS-CoV-2 positive serum, for all Spike and RBD produced, and for all storage conditions tested. Abstract

Objective: One-hour plasma glucose (1-h PG) during the oral glucose tolerance test (OGTT) is an accurate predictor of type 2 diabetes. We performed a meta-analysis to determine the optimum cutoff of 1-h PG for detection of type 2 diabetes using 2-h PG as the gold standard. Research design and methods: We included 15 studies with 35,551 participants from multiple ethnic groups (53.8% Caucasian) and 2,705 newly detected cases of diabetes based on 2-h PG during OGTT. We excluded cases identified only by elevated fasting plasma glucose and/or HbA1c. We determined the optimal 1-h PG threshold and its accuracy at this cutoff for detection of diabetes (2-h PG ≥11.1 mmol/L) using a mixed linear effects regression model with different weights to sensitivity/specificity (2/3, 1/2, and 1/3). Results: Three cutoffs of 1-h PG, at 10.6 mmol/L, 11.6 mmol/L, and 12.5 mmol/L, had sensitivities of 0.95, 0.92, and 0.87 and specificities of 0.86, 0.91, and 0.94 at weights 2/3, 1/2, and 1/3, respectively. The cutoff of 11.6 mmol/L (95% CI 10.6, 12.6) had a sensitivity of 0.92 (0.87, 0.95), specificity of 0.91 (0.88, 0.93), area under the curve 0.939 (95% confidence region for sensitivity at a given specificity: 0.904, 0.946), and a positive predictive value of 45%. Conclusions: The 1-h PG of ≥11.6 mmol/L during OGTT has a good sensitivity and specificity for detecting type 2 diabetes. Prescreening with a diabetes-specific risk calculator to identify high-risk individuals is suggested to decrease the proportion of false-positive cases. Studies including other ethnic groups and assessing complication risk are warranted.

It is unclear how or if these excess binding sites impact gene transcription. Results: to characterize the regulatory logic of AR-mediated transcription, we generated a locus-specific map of enhancer activity by functionally testing all common clinical AR binding sites with Self-Transcribing Active Regulatory Regions sequencing (STARRseq). Only 7% of AR binding sites displayed androgen-dependent enhancer activity. Instead, the vast majority of AR binding sites were either inactive or constitutively active enhancers. These annotations strongly correlated with enhancer-associated features of both in vitro cell lines and clinical prostate cancer samples. Evaluating the effect of each enhancer class on transcription, we found that AR-regulated enhancers frequently interact with promoters and form central chromosomal loops that are required for transcription. Somatic mutations of these critical AR-regulated enhancers often impact enhancer activity. Abstract

This chapter outlines the basic assumptions of the mediated social communication (MSC) as well as its main differences, compared to a traditional understanding of mass communication. It summarises distinctive features of the MSC model and specifies its potential contribution to mass communication theory and research. In the mediation process, journalism provides highly selected and transformed messages which are disseminated to recipients by means of mass media. The main features of mass communication were often derived from comparison with face-to-face communication. Importantly, the impartiality concept is embedded in the MSC approach and thereby in a broader theoretical framework. The MSC approach offers a theoretical conception of the many-to-many structure of mass communication, including the analytical distinction between mediation and communication processes. The MSC approach, on the contrary, is strongly grounded in social theory and historical research and takes public assemblies as a starting point for theoretical reflection.

Abstract

A material and surface selection within the car industry is usually based on a comprehensive study based on sensation and perception, focusing on particular perceived qualities and impressions (emotional functions) through visual and tactile interaction of plastic surfaces. On top of that 'emotional function', the 'technical function' such as surface roughness or gloss for a certain matter in symbiosis, will result in a number of material and surface proposals. The range of materials fitting into the window of these requirements varies depending on the industry's ability to hit the target specified in regard to the 'emotional functions', usually defined by the designer's intention. Thus, to be able to get a deeper understanding of how to frame the 'emotional functions' and link them to the 'technical functions', two user studies were made in this paper. The user studies were made with two different designs however with the same main research target to be able to understand the varieties of these two. The aim of doing so was to be able to find the user study design that was the most time efficient and providing the most significant data linked to the 'technical functions' and process control/traceability. However also finding the user study design with the least strains for the participating users in regard to uninterrupted brain activity. By designing the user study in relevant subsets in a certain sequence, user study design nr 2 has proven to be more time efficient and provide more data regarding the soft metrology. Future work will focus on deeper knowledge about how these surfaces different material- and surface properties correlate to the participants responses regarding perceived quality of a car interior design. The development of a new non-contact measurement is also discussed, enabling the possibility to complement and improve the hard metrology set up.
