The aim of this work was to test whether salinity is the driving factor for size and physiological differences in the dinoflagellate Prorocentrum cordatum. Three strains, isolated from 3 sites with different salinities (the central Baltic Sea, Sweden, salinity 8; Chesapeake Bay, Maryland, USA, salinity 16; and the Gulf of Trieste, northern Adriatic Sea, Italy, salinity 32), were included in this study. Following their molecular and morphological characterisation, specific growth rates and dimensions were examined for each strain cultivated at all 3 salinities (i.e. 8, 16 and 32). The Adriatic strain always showed the largest cells and the Baltic strain the smallest. Adriatic and American strains showed lower specific growth rates at salinity 8 and highest at salinity 16. The Baltic strain growth rates were similar at all salinities. As salinity 16 proved to be optimal for the strains, additional parameters were further compared at this salinity: cell volume, particulate organic carbon, particulate nitrogen, delayed fluorescence and in vivo chl a fluorescence. The Adriatic strain had the highest particulate organic carbon, particulate nitrogen, and delayed and in vivo fluorescence. Conversely, the Baltic strain had the lowest values. Although the strains had different specific growth rates with respect to salinity, this was not the main driving factor for the different sizes and photosynthetic activity among the strains. Thus, although there was low genetic variability among strains from the 3 geographical areas, intraspecific variability was suggested.

BACKGROUND: Microvascular dysfunction is involved in the development of various cerebral disorders. It may contribute to these disorders by disrupting white matter tracts and altering brain connectivity, but evidence is scarce. We investigated the association between multiple biomarkers of microvascular function and whole-brain white matter connectivity. METHODS AND RESULTS: Cross-sectional data from The Maastricht Study, a Dutch population-based cohort (n=4326; age, 59.4±8.6 years; 49.7% women). Measures of microvascular function included urinary albumin excretion, central retinal arteriolar and venular calibers, composite scores of flicker light-induced retinal arteriolar and venular dilation, and plasma biomarkers of endothelial dysfunction (intercellular adhesion molecule-1, vascular cell adhesion molecule-1, E-selectin, and von Willebrand factor). White matter connectivity was calculated from 3T diffusion magnetic resonance imaging to quantify the number (average node degree) and organization (characteristic path length, global efficiency, clustering coefficient, and local efficiency) of white matter connections. A higher plasma biomarkers of endothelial dysfunction composite score was associated with a longer characteristic path length (β per SD, 0.066 [95% CI, 0.017-0.114]) after adjustment for sociodemographic, lifestyle, and cardiovascular factors but not with any of the other white matter connectivity measures. After multiple comparison correction, this association was nonsignificant. None of the other microvascular function measures were associated with any of the connectivity measures. CONCLUSIONS: These findings suggest that microvascular dysfunction as measured by indirect markers is not associated with whole-brain white matter connectivity.

Abstract The presented work brings new knowledge in the field of spinning electrodes for AC‑electrospinning technology, which is used for producing nanofibrous structures using a solution of polyvinyl butyral. It presents new types of spinning weir‑electrodes and describes research on the influence of electrode design parameters on the stability of the spinning process and the productivity of nanofiber production. The multistage spinning electrode is presented in the ratio of stages one to five. Research is also focused on the effect of the parameters of the electric signal used as a source for the spinning electrode on spinning stability and productivity. Observed parameters were voltage level, frequency and shape such as sine wave, rectangle wave and modified sine wave. An analysis of the influence of the spinning conditions on the resulting nanofibrous structure was also performed by analyzing results gained by SEM; the defects were investigated mainly. The results of the research presented in the thesis open up new possibilities for follow-up research in the field of AC-electrospinning.

In 2022 and 2023, aquatic molluscs were surveyed in brown coal quarries ČSA and Vršany, northwestern Bohemia, where mining is ending and reclamation is in progress. Most water bodies were artificially created and relatively young, though some older habitats were present at the quarry edges. In the quarry ČSA, 20 species (17 gastropods, 3 bivalves) were recorded at 40 sites, while 13 species (10 gastropods, 3 bivalves) were found at 39 sites in Vršany. Molluscan communities were generally species-poor, particularly in deeper waters without shallows or with dense reed growth. Shallow, vegetated areas were more favorable. Physa acuta was the most frequent species. Three non-native species (Physa acuta, Potamopyrgus antipodarum, Ferrissia californica) were confirmed, and no protected or endangered species were found.

The Ranská jezírka are system of water bodies located in a forest complex on the western edge of the Žďárské vrchy PLA. These water bodies were created by flooding of depressions after surface mining of iron limonite ores. Currently, there are several larger and a number of smaller oligotrophic to mesotrophic pools covered with various types of littoral vegetation. Aquatic molluscs of this isolated and biologically interesting site were studied in 2023. In total, 15 species (6 gastropods and 9 bivalves) were found at 7 sites. Molluscan communities in water bodies with poorer vegetation were species-poor while water bodies more overgrown with littoral vegetation were inhabited by relatively species-richer communities. The rare and endangered bivalve Sphaerium nucleus was found. No non-native species were found.

The reliability of organic molecular crystal structure prediction has improved tremendously in recent years. Crystal structure predictions for small, mostly rigid molecules are quickly becoming routine. Structure predictions for larger, highly flexible molecules are more challenging, but their crystal structures can also now be predicted with increasing rates of success. These advances are ushering in a new era where crystal structure prediction drives the experimental discovery of new solid forms. After briefly discussing the computational methods that enable successful crystal structure prediction, this perspective presents case studies from the literature that demonstrate how state-of-the-art crystal structure prediction can transform how scientists approach problems involving the organic solid state. Applications to pharmaceuticals, porous organic materials, photomechanical crystals, organic semi-conductors, and nuclear magnetic resonance crystallography are included. Finally, efforts to improve our understanding of which predicted crystal structures can actually be produced experimentally and other outstanding challenges are discussed.

Counterfactuals are alternative outcomes to past events. Curiosity for the counterfactual acts as an important driver of learning under uncertainty and helps to improve on past behaviors by informing future choices. Humans have demonstrated the desire to collect counterfactual information when it is perceived as useful, during times of uncertainty, and even when it comes at a cost. The comparative literature on counterfactual use and curiosity is small but suggests that rhesus macaques may possess similar tendencies to humans. The goal of the current study was to add a new species, capuchin monkeys (Sapajus apella), to the comparative counterfactual literature and to test capuchins and humans on nearly identical tasks. This study involved a three-choice gambling task that consisted of one hidden and two visible reward values. Visible reward values were always equivalent but had differently colored borders to indicate whether participants would view the counterfactual information associated with the gamble or not on trials when they chose not to gamble. Only humans showed evidence of counterfactual curiosity and collected the information at levels different than chance. Humans also collected counterfactual information significantly more than capuchins, indicating a species difference. However, it is not clear whether this difference is simply due to the capuchins’ lack of curiosity surrounding this particular task or whether there is a broader species difference for seeking counterfactuals resulting from different evolutionary histories of these species.

The effect of feed push-up has already been proven in several studies, so this topic can be considered one of the main points that helps improve the health status of dairy cattle. This study aimed to determine how the frequency of feed push-ups influences the health status of udder (mastitis), somatic cell counts, and reproduction. The effect of feed push-up on mastitis, the somatic cell counts, and the conception of dairy cows was evaluated. The feed was pushed-up at a frequency of 2x, 3x, 4x, 5x, and 6x a day for one calendar month. The effect on the number of dairy cows treated with mastitis was insignificant (P ≥ 0.05). The lowest number of cows with mastitis was found when feed was pushed-up five times daily. The effect on the somatic cell counts was insignificantly, too (P ≥ 0.05). The worst milk quality was found in the experimental group, which had a frequency of push-up 5x/day. However, it has been shown that the frequency of food push-up positively affected the conception rate in dairy cows (P < 0.001).

This paper presents important faunistic records conducted in the Czech and Slovak Republics during 2023. We also include records generated before 2023, which have yet to be published, mainly because their correct identification was unavailable earlier. In a separate section we present records of unintentionally introduced species, reported for the first time for the countries from greenhouses (Dryachloa dauca and Guppya gundlachii). The first outdoor colony of Melanoides tuberculata was found in a stream artificially heated by wastewater from the Dukovany nuclear power plant and Planorbella duryi found in a brook with thermal water flowing from the Sliač spa. Records of native species include findings of protected and rare species (e.g., Aplexa hypnorum, Ladislavella occulta, Daudebardia brevipes, and Theodoxus danubialis), but also still poorly known species (Ampullaceana lagotis and Pyramidula saxatilis). During 2023, many new records were made for non-native and currently spreading species as for example Clathrocaspia knipowitschii, Corbicula fluminea, Hygromia cinctella, Krynickillus melanocephalus, Sinanodonta woodiana, and Tandonia kusceri.

Ab initio predictions of chemical shifts and electric field gradient (EFG) tensor components are frequently used to help interpret solid-state nuclear magnetic resonance (NMR) experiments. Typically, these predictions employ density functional theory (DFT) with generalized gradient approximation (GGA) functionals, though hybrid functionals have been shown to improve accuracy relative to experiment. Here, the performance of a dozen models beyond the GGA approximation are examined for the prediction of solid-state NMR observables, including meta-GGA, hybrid, and double-hybrid density functionals and second-order Møller-Plesset perturbation theory (MP2). These models are tested on organic molecular crystal data sets containing 169 experimental 13C and 15N chemical shifts and 114 17O and 14N EFG tensor components. To make these calculations affordable, gauge-including projector augmented wave (GIPAW) Perdew-Burke-Ernzerhof (PBE) calculations with periodic boundary conditions are combined with a local intramolecular correction computed at the higher level of theory. Within the context of typical NMR property calculations performed on a static, DFT-optimized crystal structure, the benchmarking finds that the double-hybrid DFT functionals produce errors versus experiment that are no smaller than those of hybrid functionals in the best cases, and they can be larger. MP2 errors versus experiment are even bigger. Overall, no practical advantages are found for using any of the tested double-hybrid functionals or MP2 to predict experimental solid-state NMR chemical shifts and EFG tensor components for routine organic crystals, especially given the higher computational cost of those methods. This finding likely reflects error cancellation benefiting the hybrid functionals. Improving the accuracy of the predicted chemical shifts and EFG tensors relative to experiment would probably require more robust treatments of the crystal structures, their dynamics, and other factors.

Statin treatment may increase the risk of diabetes; there is insufficient data on how statins affect glucose regulation and glycemic control and the effects of statins on liver enzymes related to carbohydrate metabolism have not been fully studied. Therefore, we aimed to compare the effects of the statin derivatives, pravastatin, and rosuvastatin, on carbohydrate metabolism in an experimental diabetic rat model. Female Wistar albino rats were used and diabetes was induced by intraperitoneal injection of streptozotocin. Thereafter, 10 and 20 mg kg−1 day−1 doses of both pravastatin and rosuvastatin were administered by oral gavage to the diabetic rats for 8 weeks. At the end of the experiment, body masses, the levels of fasting blood glucose, serum insulin, insulin resistance (HOMA-IR), liver glycogen, and liver enzymes related to carbohydrate metabolism were measured. Both doses of pravastatin significantly in creased the body mass in diabetic rats, however, rosuvastatin, especially at the dose of 20 mg kg−1 day−1 reduced the body mass signi ficantly. Pravastatin, especially at a dose of 20 mg kg−1 day−1, caused significant increases in liver glycogen synthase and glucose 6-phosphate dehydrogenase levels but significant decreases in the levels of glycogen phosphorylase, lactate dehydrogenase, and glucose-6-phosphatase. Hence, pravastatin partially ameliorated the adverse changes in liver enzymes caused by diabetes and, especially at the dose of 20 mg kg−1 day−1, reduced the fasting blood glucose level and increased the liver glycogen content. However, rosuvastatin, especially at the dose of 20 mg kg−1 day−1, significantly reduced the liver glycogen synthase and pyruvate kinase levels, but increased the glycogen phosphorylase level in diabetic rats. Rosuvastatin, 20 mg kg−1 day−1 dose, caused significant decreases in the body mass and the liver glycogen content of diabetic rats. It can be concluded that pravastatin, especially at the dose of 20 mg kg−1 day−1 is more effective in ameliorating the negative effects of diabetes by modulating carbohydrate metabolism.

Abstract Background Point-of-care testing (POCT) devices are diagnostic tools that can provide quick and accurate results within minutes, making them suitable for diagnosing non-communicable diseases (NCDs). However, these devices are not widely implemented in healthcare systems and for this reason is relevant to understand the implementation process. Aim To describe the process and define a strategy to implement a multiparameter POCT device for diagnosing and managing NCDs in one region of Peru. Methods A descriptive and non-experimental study, using the participatory methodologies of co-creation process. It was conducted in one region of Peru (Tumbes) to design an intervention for implementing a multiparameter POCT device. Two co-creation sessions were conducted involving five groups: community members, primary healthcare workers, these groups in both rural and urban settings, and regional decision-makers. These sessions included activities to understand patient journeys in receiving care for NCDs, identify facilitators and barriers to POCT devices usage, and define an implementation strategy for POCT devices in both rural and urban settings of Tumbes. The research team analysed the data and summarized key topics for discussion after each session. Results A total of 78 participants were enrolled across the five groups. Among community members: 22.2% had only diabetes, 24.1% had only hypertension, and 18.5% had both diagnoses. In the patient journey, community members mentioned that it took at least three days to receive a diagnosis and treatment for an NCD. Most of the participants agreed that the POCT devices would be beneficial for their communities, but they also identified some concerns. The strategy for POCT devices implementation included healthcare workers training, POCT devices must be placed in the laboratory area and must be able to perform tests for glucose, glycated haemoglobin, cholesterol, and creatinine. Advertising about POCT devices should be displayed at the healthcare centres and the municipality using billboards and flyers. Conclusions The co-creation process was useful to develop strategies for the implementation of multiparameter POCT devices for NCDs, involving the participation of different groups of stakeholders guided by moderators in both, rural and urban, settings in Peru.

ABSTRACT: In recent decades, interest in non-traditionally colored eggs has increased. For breeders, this market interest means breeding lines of laying hens that lay eggs of varied colors, such as the blue-green eggshells (Dominant Greenshell) in this study. This study presents the results of genotyping the polymorphism of the O locus responsible for shell coloration and photometric measurement of eggshell color based on the CIELAb system, which was carried out on the unique Czech breeding population Dominant Greenshell. The aim was to use a combination of phenotyping using the CIELab System method and genotyping of the O locus using the end-point PCR approach with the main focus on the accuracy of distinguishing shell color genotypes, streamlining the selection of dominant homozygotes in the O locus, optimizing this technology for the most efficient and cost-effective selection procedure in practical hen breeding. The optometric method was able to reliably distinguish only dominant and recessive phenotypes and eliminate from the population only undesirable recessive homozygotes with a white colored shell. The parameter a* (redness/greenness) from the CIELab color space turned out to be absolutely key for distinguishing dominant and recessive phenotypes. Using the CART methodology, a classification tree built on discriminating optometric characteristics a-blunt was obtained, however, for the group of desirable O/O homozygotes, the selection approach would result in incorrect genotyping of 31% of individuals. Therefore, a combined approach based on rapid and simple elimination of recessive homozygotes using phenotyping (CIELab photometric measurement) and molecular identification of the EAV-HP insertion in the SLCO1B3 gene in dominant phenotypes, regardless of color intensity affected by laying time/order, and allowing reliable elimination, has proven to be the most effective method to distinguish heterozygotes from the breeding population. The combination of optometric and molecular selection methods then leads to more efficient selection, reduction of overall selection costs. This process led to the stabilization of the breeding population within one generation and the achievement of a pure homozygous line with regard to eggshell color.

Photomechanical molecular crystals have garnered attention for their ability to transform light into mechanical work, but difficulties in characterizing the structural changes and mechanical responses experimentally have hindered the development of practical organic crystal engines. This study proposes a new computational framework for predicting the solid-state crystal-to-crystal photochemical transformations entirely from first principles, and it establishes a photomechanical engine cycle that quantifies the anisotropic mechanical performance resulting from the transformation. The approach relies on crystal structure prediction, solid-state topochemical principles, and high-quality electronic structure methods. After validating the framework on the well-studied [4 + 4] cycloadditions in 9-methyl anthracene and 9-tert-butyl anthracene ester, the experimentally-unknown solid-state transformation of 9-carboxylic acid anthracene is predicted for the first time. The results illustrate how the mechanical work is done by relaxation of the crystal lattice to accommodate the photoproduct, rather than by the photochemistry itself. The large ∼107 J m-3 work densities computed for all three systems highlight the promise of photomechanical crystal engines. This study demonstrates the importance of crystal packing in determining molecular crystal engine performance and provides tools and insights to design improved materials in silico.
