Background: Clear cell renal cell carcinoma (ccRCC) is a highly heterogeneous cancer type that lacks early diagnostic markers and effective targets for immunotherapy. WD repeat-containing protein 72 (WDR72) mutations have been linked to renal dysfunction and renal tubular acidosis. However, the potential prognosis-predictive value of WDR72 in ccRCC remains unelucidated. Methods: The current research used The Cancer Genome Atlas (TCGA) data to explore the expression and prognosis-predictive value of WDR72 in various solid tumors, including ccRCC. The analysis was validated using the TCGA–ccRCC cohort and Human Protein Atlas. The biological processes and signaling pathways linked to the expression of WDR72 were identified by conducting Gene Ontology and gene set enrichment analysis. Cox regression analysis was utilized to measure the prognosis-predictive value of WDR72 in ccRCC. Moreover, the association of WDR72 expression levels with immune cell infiltration, methylation, m6A, tumor mutation burden, immune checkpoint molecules, and drug sensitivity was analyzed. Finally, the study was verified by conducting quantitative real-time polymerase chain reaction and cytological experiments in ccRCC. Results: A pan-cancer analysis noted that low WDR72 expression levels were strongly linked to poor overall survival and progression-free interval in individuals with ccRCC. The WDR72 levels were remarkably reduced in ccRCC tissues in comparison with the adjacent tissues, and WDR72 was identified as an independent predictor of patients with ccRCC. Pathway enrichment analysis showed a close relationship between genes related to WDR72 and fat metabolism. Low WDR72 expression in ccRCC was linked to higher immune cell infiltration, stronger tumor mutation burden, and higher immune checkpoint molecules. Furthermore, individuals with ccRCC and low WDR72 expression had lower IC50 values for most drugs. Conclusion: Low WDR72 expression is strongly linked to the progression, prognosis, and microenvironmental immune regulation of individuals with ccRCC. Moreover, it may hold potential as a valuable prognostic biological marker and therapeutic measure for ccRCC. The findings provide a basis for future research by exploring the role of WDR72 in ccRCC tumor development.

BackgroundUreteral Fibroepithelial Polyps (UFPs) are benign tumors that arise from the mesodermal layer. The disease is a rare cause of ureteropelvic junction obstruction (UPJO), particularly in children. Case presentationwe present a case of a child preoperatively diagnosed with UPJO and urogenic sepsis owing to horseshoe kidney using magnetic resonance imaging(MRI). During surgery, we found that the ureteral polyp rather than the horseshoe kidney was the cause of the PUJO.Thenus, the ureteral polyp was resected, and end-to-end anastomosis was performed with the renal pelvis and ureters. An F5 double J tube was indwelling and removed by ureteroscopy three months later. No special adverse events were found during 1-year follow-up. ConclusionsWhen the clinical and radiographic findings of the patient are incompatible with the common etiology, ureteral fibroepithelial polyps should be considered as a rare cause of PUJO.

Objective: To evaluate the attenuation value [Hounsfield unit (HU)] in non-contrast CT for distinguishing pyonephrosis from hydronephrosis in parents with upper urinary tract (UTS) stones and to identify factors predicting the development of sepsis in patients with pyonephrosis. Methods: Between October 2019 and October 2021, patients with hydronephrosis or pyonephrosis secondary to upper urinary tract stones (UTS) were retrospectively searched in our study. All patients with UTS were treated with either percutaneous nephrostomy (PCN) or surgical management (percutaneous nephrolithotomy / transurethral ureteroscope lithotripsy). We excluded patients treated with extracorporeal shock-wave lithotripsy or patients with bilateral upper urinary tract stones. Patients whose CT was not performed in our hospital or treated in another hospital were also excluded. Clinical data regarding basic information, clinical feature, co-morbidities, previous intervention, infection-related indicators, Calculi-related indicators, HU values of the renal pelvis, the thick wall of the renal pelvis on CT were collected. Univariate and multivariate logistic analyses were performed. ROC curves were drawn to predict pyonephrosis and sepsis. Results: Two-hundred-forty patients with UTS were included in our study, 191 patients had hydronephrosis without pyonephrosis (Group 1), and 49 patients had hydronephrosis with pyonephrosis (Group 2). The HU value of the renal collecting system in Group 2 (mean, 15.46; range, +1/+30) was significantly higher than that in Group 1 (mean, 5.5;5 range -6/+24) (P = 0.02); the ROC curve analysis revealed that the HU value (AUC = 0.613) of 9.5 was the best cut-off value to predict calculi-related obstructive pyonephrosis, with 71.4% sensitivity and 70.2% specificity. But it cannot predict the development of sepsis in patients with postoperative pyonephrosis (p = 0.11). Multivariate analysis showed that age (P = 0.041), sex (p = 0.036), herpes labialis (p = 0.018), and serum albumin (p = 0.027) were significant predictors for the development of sepsis in patients with postoperative pyonephrosis. Conclusions: The HU attenuation value of the renal collecting system can distinguish pyonephrosis from hydronephrosis, but it cannot predict sepsis in patients with pyonephrosis postoperative. Older, low-serum albumin female patients with renal pyonephrosis caused by UUT stones are more likely to develop urogenic sepsis.

The HU value of the renal collecting system in Group 2 (mean, 15.46; range, +1/+30) was significantly higher than that in Group 1 (mean, 5.5; 5 range -6/+24) (P = .02); the receiver operative characteristic curve analysis revealed that the best cut-off value of 9.5 could predict the presence of pyonephrosis, with 71.4% sensitivity and 70.2% specificity (area under the curve = 0.613; 95% CI: 0.514-0.713). In this study, we found the HU attenuation value of the renal collecting system can be used to distinguish pyonephrosis from hydronephrosis in patients with UTS.

Accurate modeling of the instantaneous uncut chip thickness is one of the key issues in milling mechanics. Although most current cutting force models take into account the effects of tool deflection and tool run-out, they only consider single-edge continuous milling processes and do not consider processes where multiple edges are simultaneously engaged in milling. Considering the tool runout and tool deflection that affect the end milling process, a novel uncut chip thickness algorithm based on two-flute continuous milling is proposed in this paper, and an analytical force model for actual milling is established. There is no need to use iterative methods, and the predictive power can be obtained by solving a linear system of equations, so the computational cost is greatly reduced. At the same time, stainless steel 316 was processed, and the experimental results showed that the predicted force was in good agreement with the measured force.

The origin of amplitude synchronization (AS), or amplitude envelope synchronization, as a peculiar form of strong correlation between amplitudes of oscillators is studied by using a model of coupled Landau-Stuart periodic oscillators. We find that the AS extensively occurs within the traditional phase drift region, and the amplitude correlation does not change with variation of the coupling strength but is dampened with increase of the frequency mismatch. The AS appears only at weak couplings and before the occurrence of phase synchronization (PS), and the oscillator amplitude is modulated by its phase. This study could build a solid foundation for AS, which has not drawn much attention in the nonlinear dynamics field before, providing a clear physical picture for synchronization including not only PS, but also AS, and arousing general interest in many interdisciplinary fields, such as neuronal systems, laser dynamics, nanomechanical resonators, and power systems, etc., where phase and amplitude are always mutually influenced and both are important.

We used an animal model of experimental varicocele to investigate the effects on the ipsilateral testis during the progression of experimental varicocele in rat.Experimental varicocele was induced by partial ligation of the left renal vein in rats. Rats in each control group were subjected to a sham operation. After different periods of varicocele creation (6, 12, and 18 weeks), each animal's left testis was extirpated, and Johnsen's score, ultrastructure of seminiferous tubules, intratesticular testosterone concentration, and the germ cell apoptotic index of each left testis were compared between the experimental and control groups.Johnsen's score and intratesticular testosterone concentrations in each experimental varicocele group were significantly lower than those in the corresponding control groups (P0.05) and showed significant declines as the duration of varicocele gradually increased; the apoptotic index showed an opposite tendency. Ultrastructural injury of seminiferous tubules gradually became severe as the duration of varicocele gradually increased.Experimental varicocele caused progressive impairment of the ipsilateral testis; apoptosis mediated the pathophysiological processes of dyszoospermia.
