Abstract

In the last years, optical readout of Micromegas gaseous detectors has been achieved by implementing a Micromegas detector on a glass anode coupled to a CMOS camera. Effective X-ray radiography was demonstrated using integrated imaging approach. High granularity values have been reached for low-energy X-rays from radioactive sources and X-ray generators. Detector characterization with X-ray radiography has led to two applications: neutron imaging for non-destructive examination of highly gamma-ray emitting objects and beta imaging for the single cell activity tagging in the field of oncology drug studies. Abstract

The steadily increasing luminosity of the LHC requires an upgrade with high-rate and high-resolution detector technology for the inner end cap of the ATLAS muon spectrometer: the New Small Wheels (NSW). In order to achieve the goal of precision tracking at a hit rate of about 15 kHz/cm$^2$ at the inner radius of the NSW, large area Micromegas quadruplets with 100\,\microns spatial resolution per plane have been produced. % IRFU, from the CEA research center of Saclay, is responsible for the production and validation of LM1 Micromegas modules. The construction, production, qualification and validation of the largest Micromegas detectors ever built are reported here. Abstract

Optical readout of Micromegas gaseous detectors has been achieved by implementing a Micromegas detector on a glass substrate with a glass anode and a CMOS camera. Efficient X-ray radio-graphy has been demonstrated due to the integrated imaging approach inherent to optical readout. High granularity values have been reached for low-energy X-rays from radioactive sources and X-ray generators taking advantage of image sensors with several megapixel resolution. Detector characterization under X-ray radiography opens the way to different applications from beta imaging to neutron radiography. Here we will focus on one application: neutron imaging for non-destructive examination of highly gamma-ray emitting objects. This article reports the characterization of the detectors when exposed to a low activity neutron source. The response of the detector to thermal neutrons has been studied with different field configurations and gap thicknesses.

10. Propulsion Without Propellent Mass; a Time-Varying Electromagnetic Field Effect

Abstract

A propulsion generated without propellent mass and external forces using a well known physics is possible theoretically. To do that, it is proposed to use two specific time-varying and constrained distributions of electric charges which own a symmetry axis and are adequately separated in space. Total charge of each distribution is zero. With a relative temporal phase-shift between them, an asymmetry into the far fields momentum variation rate distribution along the symmetry axis can be induced. Consequently, an electromagnetic propulsive force is generated and applied over both charge distributions in a same direction along the axis-say. Magnetic part of this force is dissipative because closely related to energy lost by radiation. But its electric counterpart gives a conservative contribution to the propulsive force with no associated energy losses at zero velocity. Momentum conservation law for the entire system; the two charge distributions and fields, is respected because all far fields; radiation ones and relativistic correction one, are taken into account. Abstract

Abstract Myositis is characterised by muscle inflammation and weakness. Although generally thought to be driven by a systemic autoimmune response, increasing evidence suggests that intrinsic changes in the muscle might also contribute to the pathogenesis. Long non-coding RNAs (lncRNAs) are a family of novel genes that regulate gene transcription and translation. To determine the potential role of lncRNAs, we employed next generation sequencing to examine the transcriptome in muscle biopsies obtained from two histologically distinct patient populations, inclusion body myositis (IBM) and anti-Jo-1-associated myositis (Jo-1). 1287 mRNAs and 1068 mRNAs were differentially expressed in the muscle from Jo-1 and IBM patients, respectively. Pathway analysis showed the top canonical pathway in both Jo-1 and IBM was oxidative phosphorylation and mitochondrial dysfunction. We identified 731 known and 325 novel lncRNAs in the muscles biopsies. Comparison with controls showed 55 and 46 lncRNAs were differentially expressed in IBM and Jo-1 myositis, respectively. Of these, 16 lncRNAs were differentially expressed in both IBM and Jo-1 myositis and included upregulated H19, lncMyoD and MALAT1. Given that these are known to regulate muscle proliferation and differentiation, we speculate that changes in lncRNAs might contribute to the phenotypic changes in Jo-1 and IBM myositis.

Abstract

There is accumulating evidence to indicate that long non-coding RNAs (lncRNAs) are important regulators of the inflammatory response. In this report, we have employed next generation sequencing to identify 14 lncRNAs that are differentially expressed in human lung fibroblasts following the induction of inflammation using interleukin-1β (IL-1β). Knockdown of the two most highly expressed lncRNAs, IL7AS, and MIR3142HG, showed that IL7AS negatively regulated IL-6 release whilst MIR3142HG was a positive regulator of IL-8 and CCL2 release. Parallel studies in fibroblasts derived from patients with idiopathic pulmonary fibrosis showed similar increases in IL7AS levels, that also negatively regulate IL-6 release. In contrast, IL-1β-induced MIR3142HG expression, and its metabolism to miR-146a, was reduced by 4- and 9-fold in IPF fibroblasts, respectively. This correlated with a reduced expression of inflammatory mediators whilst MIR3142HG knockdown showed no effect upon IL-8 and CCL2 release. Pharmacological studies showed that IL-1β-induced IL7AS and MIR3142HG production and release of IL-6, IL-8, and CCL2 in both control and IPF fibroblasts were mediated via an NF-κB-mediated pathway. In summary, we have cataloged those lncRNAs that are differentially expressed following IL-1β-activation of human lung fibroblasts, shown that IL7AS and MIR3142HG regulate the inflammatory response and demonstrated that the reduced inflammatory response in IPF fibroblast is correlated with attenuated expression of MIR3142HG/miR-146a.

19. Catalog of Differentially Expressed Long Non-Coding RNA following Activation of Human and Mouse Innate Immune Response

Abstract

Despite increasing evidence to indicate that long non-coding RNAs (lncRNAs) are novel regulators of immunity, there has been no systematic attempt to identify and characterize the lncRNAs whose expression is changed following the induction of the innate immune response. To address this issue, we have employed next-generation sequencing data to determine the changes in the lncRNA profile in four human (monocytes, macrophages, epithelium, and chondrocytes) and four mouse cell types (RAW 264.7 macrophages, bone marrow-derived macrophages, peritoneal macrophages, and splenic dendritic cells) following exposure to the pro-inflammatory mediators, lipopolysaccharides (LPS), or interleukin-1β. We show differential expression of 204 human and 210 mouse lncRNAs, with positional analysis demonstrating correlation with immune-related genes. These lncRNAs are predominantly cell-type specific, composed of large regions of repeat sequences, and show poor evolutionary conservation. Comparison within the human and mouse sequences showed less than 1% sequence conservation, although we identified multiple conserved motifs. Of the 204 human lncRNAs, 21 overlapped with syntenic mouse lncRNAs, of which five were differentially expressed in both species. Among these syntenic lncRNA was IL7-AS (antisense), which was induced in multiple cell types and shown to regulate the production of the pro-inflammatory mediator interleukin-6 in both human and mouse cells. In summary, we have identified and characterized those lncRNAs that are differentially expressed following activation of the human and mouse innate immune responses and believe that these catalogs will provide the foundation for the future analysis of the role of lncRNAs in immune and inflammatory responses.

Abstract

Alectra vogelii Benth. is the second most important parasitic weed in cowpea [Vigna unguiculata (L.) Walp.] production in Burkina Faso. Several resistant varieties to this weed have been identified in the country among which are B301 and KVx414-22-2. The inheritance and allelic relationships of the resistance genes in the two varieties have not been studied with A. vogelii strains in Burkina Faso. The objective of this study was to determine the inheritance and allelic relationship of the resistance genes in B301 and KVx414-22-2. To determine the inheritance of the genes for resistance, the resistant varieties (B301 and KVx414-22-2) were each crossed to a susceptible variety IT82D-849 to generate F1 and F2 populations. For the allelic relationship study the two resistant genotypes were crossed among themselves to generate F1 and F2 offspring. The parents and their F1 and F2 progenies were screened in artificially infested pots with Alectra seed in a screen house at Kamboinsé Research Station in Burkina Faso. Resistance/susceptibility of genotypes was assessed by recording the number of emerged Alectra shoots. The data were subjected to the Chi-Square goodness-of-fit test for one, two and three genes segregation ratios. The results revealed that two independent dominant genes confer resistance in the variety B301 and a single dominant gene confers resistance in variety KVx414-22-2. The single dominant gene in KVx414-22-2 is non-allelic to the two genes in B301. The two resistance genes in variety B301 have already been named Rav1 and Rav2 whilst Rav3 is the name of the resistance gene in variety IT81D-994. Therefore, we propose the symbol Rav4 as the name for the resistance gene in variety KVx414-22-2.

Pharmacological studies showed that IL-1β-induced IL7AS and MIR3142HG production and release of IL-6, IL-8, and CCL2 in both control and IPF fibroblasts were mediated via an NF-κB-mediated pathway. In summary, we have cataloged those lncRNAs that are differentially expressed following IL-1β-activation of human lung fibroblasts, shown that IL7AS and MIR3142HG regulate the inflammatory response and demonstrated that the reduced inflammatory response in IPF fibroblast is correlated with attenuated expression of MIR3142HG/miR-146a.

L’auteur présente et analyse la pensée de Tenzin Gyatso, Sa Sainteté le quatorzième dalaï-lama, concernant la possibilité d’établir un dialogue fructueux entre le bouddhisme et la science moderne. Pour ce faire, il se base sur une oeuvre particulière du dalaï-lama : Tout l’Univers dans un atome. L’auteur présente ainsi l’oeuvre à l’étude, son intention, ainsi que le contexte dans lequel elle s’insère. Il divise ensuite la réflexion du dalaï-lama en trois grands axes. D’abord, l’auteur explore les points de convergence, relevés par Sa Sainteté, entre la science moderne et le bouddhisme. Ensuite, il s’intéresse à leurs points de divergence, ainsi qu’à leur possible complémentarité, en présentant par le fait même la critique que le dalaï-lama adresse aux scientifiques. He then divides His Holiness’s reflection in three axes. First, the author explores the converging views of modern science and Buddhism, as underlined by the Dalai Lama. He then takes an interest in their points of divergence and their possible complementarity, while discussing the criticisms that His Holiness addresses to scientists. The author concludes with the Dalai Lama’s ethical concerns about science, its technological productions and the world view that it tends to convey. International audience; Introduction and Aims : :. People who inject drugs (PWID) are exposed to associated viral, bacterial and fungal risks. These risks can be reduced by filtration. Large disparities in the quality of filtration exist between the various available filters. This paper compares both performance and user acceptability of three filters for drug injection (cotton filters, Sterifilt® and wheel filters) by combining epidemiological and bacteriological analyses. Design and Methods. A cross-sectional epidemiological study (ANRS-Coquelicot) using time–location sampling combined with the generalised weight sampling method was conducted among 985 PWID in France. Two filtration-based bacteriological studies of 0.20- and 0.45-μm wheel filters, Sterifilt filters and cotton filters were also conducted. Results. The bacteriological study highlighted the value of using wheel filters with a porosity of less than 0.5 μm, as they limit the risk of bacterial and fungal infection. The results of this study clearly highlight a distinction between the efficiency of Sterifilt and wheel filters, the latter being more effective. Our epidemiological study highlighted that the use of cotton filters is widespread and routine, but is the subject of much criticism among PWID. Sterifilt is not widely used, and its adoption is slow. Finally, the wheel filter remains a largely untested tool. Discussion and Conclusions. Low product retention and ease of use are the two most important factors for filters for PWID. Bacterial and fungal risk filtration is less important. Abstract

Summary Background Partial nephrectomy (PN) is recommended as first-line treatment for cT1 stage kidney tumors because of a better renal function and probably a better overall survival than radical nephrectomy (RN). For larger tumors, PN has a controversial position due to lack of evidence showing good cancer control. The aim of this study was to compare the results of PN and RN in cT2a stage on overall survival and oncological results. Method A retrospective international multicenter study was conducted in the frame of the French kidney cancer research network (UroCCR). We considered all patients aged ≥ 18 years who underwent surgical treatment for localized renal cell carcinoma (RCC) stage cT2a (7.1–10 cm) between 2000 and 2014. Cox and Fine-Gray models were performed to analyze overall survival (OS), cancer specific survival (CSS) and cancer-free survival (CFS). Comparison between PN and RN was realized after an adjustment by propensity score considering predefined confounding factors: age, sex, tumor size, pT stage of the TNM classification, histological type, ISUP grade, ASA score. Results A total of 267 patients were included. OS at 3 and 5 years was 93.6% and 78.7% after PN and 88.0% and 76.2% after RN, respectively. CSS at 3 and 5 years was 95.4% and 80.2% after PN and 91.0% and 85.0% after RN. No significant difference between groups was found after propensity score adjustment for OS (HR 0.87, 95% CI: 0.37–2.05, P = 0.75), CSS (HR 0.52, 95% CI: 0.18–1.54, P = 0.24) and CFS (HR 1.02, 95% CI: 0.50–2.09, P = 0.96). Conclusion PN seems equivalent to RN for OS, CSS and CFS in cT2a stage kidney tumors. The risk of recurrence is probably more related to prognostic factors than the surgical technique. The decision to perform a PN should depend on technical feasibility rather than tumor size, both to imperative and elective situation. Level of evidence 4.

Abstract

The human genome is widely transcribed outside of protein-coding genes, producing thousands of noncoding RNAs from different subfamilies including enhancer RNAs. Functional studies to determine the role of individual genes are challenging with noncoding RNAs appearing to be more difficult to knockdown than mRNAs. One factor that may have hindered progress is that the majority of noncoding RNAs are thought to be located within the nucleus, where the efficiency of traditional RNA interference techniques is debatable. Here we present an alternative RNA interference technique utilizing Locked Nucleic Acids, which is able to efficiently knockdown noncoding RNAs irrespective of intracellular location.
