8. Facile preparation of 3-substituted 2-quinazolinones via electrogenerated base

Abstract

A new series of 3,4-disubstituted quinazolin-2-ones, with potential T-type calcium channel antagonist activity, and new 4-methylene-quinazolin-2-ones, promising catalysts as N-heterocyclic olefins, have been prepared in good yield by a simple reaction between 2-aminobenzophenone, or 2-aminoacetophenone, and cyanomethyl anion electrogenerated by acetonitrile reduction at a graphite electrode, followed by the addition of different organic isocyanates and subsequent heterocyclization.

Abstract

Some aromatic 1,2-dicarbonyl compounds, i.e. 9,10-phenanthrenequinone, acenaphthenequinone and benzil, and their corresponding N-phenyl monoimines, have been reduced, using dry acetonitrile as the solvent, in the presence of sodium cyanide as a reducing agent. Comparative potentiostatic preparative-scale electrolysis is described.

Abstract

We report a simple method for the synthesis of alkyl-2-(2-imino-4-oxothiazolidin-5-ylidene) acetate derivatives3in good yields under mild conditions. The electrogenerated cyanomethyl base (EGB), obtained from electroreduction of acetonitrile-0.1 M TBABF4, assists the reaction between thiourea derivatives1and dialkyl acetylene dicarboxylate2. The expected products,3/4, and the structure obtained from X-ray diffraction confirm that the main products are the five-membered heterocycles3. Furthermore, a mechanism, to explain the reaction pathways, is proposed based on the thermodynamic and kinetic data obtained from quantum calculations. [ABSTRACT FROM PUBLISHER]

Abstract

The reaction of hexamethylphosphotriamide or methylphosphonic bis(dimethylamide) compounds with amidines derived from N-Benzimidazoyl imidates 1 leads to (1,2a)benzimidazolo-1,3,5,2-tiazaphosphorine-2-oxides 4 in good yields. If the condensation is realized at room temperature, N-phosphonic amidines 3 can be isolated as intermediates. The isolated compounds 2 , 3 , and 4 are identified by spectroscopic methods: IR, 1 H, 13 C, 31 P, NMR, and M.S. [ABSTRACT FROM AUTHOR]
