The Occidental Low- and High-Saccharin-Consuming rats (respectively, LoS and HiS) have been selectively bred for decades to study the relationship between taste and behaviors in and beyond the ingestive domain. Whether the saccharin phenotype is associated with behavioral periodicities tied to reproductive status is not known. Here we describe for the first time variation across the estrous cycle in chow intake and wheel running by LoS and HiS rats. This study also shed light on why rats, humans, and some other mammals eat less and become more active as fertility increases. Wheel running increases when eating is reduced through restricted chow access, more so in LoS rats than in HiS rats (Dess et al., 2000). If the decrease in food intake from diestrus through estrus causes the increase in running (Eat Less → Run More hypothesis, ELRM), then the running peak should follow the eating nadir and be greater in LoS rats. Bayesian cyclic regression showed that estrous cycles were shorter in LoS rats than in HiS rats; implications are discussed. Contrary to ELRM, the running peak did not follow the eating nadir, and cycle amplitude did not distinguish LoS rats from HiS rats. These results indirectly support the No Time To Eat hypothesis (Fessler, 2003), according to which the periovulatory eating nadir and running peak reflect fitness-enhancing consequences of shifts away from eating and toward mating as fertility increases.

Up to 80% of all bird species are socially monogamous. Divorce (switching partners) or pair disruption (due to the death of a partner) has been associated with decreased reproductive success, suggesting social monogamy is a strategy that may maximize fitness via coordination between partners. Previous studies have demonstrated the effects of divorce and pair disruption on immediate reproductive success. Here, we used a paired experimental design in the zebra finch (Taeniopygia guttata) to examine the hormonal mechanisms that modulate parental behavior and reproductive success in response to a partnership change (hereafter divorce). Specifically, we examined the effects of divorce on the avian stress hormone corticosterone (CORT) in both parents and nestlings, parental behaviors (incubation and nestling provisioning), prolactin (PRL), and reproductive success. We found that divorce resulted in delayed clutch initiation, reduced clutch mass, and an increase in nestling CORT response to a standardized stressor. These effects on reproductive investment and chick CORT response were not clearly determined by parental endocrine responses. Divorce had no effect on the level of parental CORT. PRL levels were highly correlated within a pair regardless of treatment, were negatively related to the investment that males made in incubation, and increased in experimental males as a result of pair disruption. This study demonstrates the fundamental impact which divorce has not only on reproduction, but also the physiological stress responses of offspring and suggests that in socially monogamous animals the maintenance of a stable partnership over time could be advantageous for long term fitness.

Common carp (Cyprinus carpio) are one of the world's most destructive vertebrate pests. In Australia, they dominate many aquatic ecosystems causing a severe threat to aquatic plants, invertebrates, water quality, native fish and social amenity. The Australian Government is considering release of cyprinid herpesvirus-3 (CyHV-3) as a control measure and consequently a robust, continental-scale estimate of the carp population and biomass is essential to inform planning and risk management. Here, we pioneer a novel model-based approach to provide the first estimate of carp density (no/ha) and biomass density (kg/ha) at river reach/waterbody, basin and continental scales. We built a spatial layer of rivers and waterbodies, classified aquatic habitats and calculated the area of each throughout the range of carp in Australia. We then developed a database of fishery-independent electrofishing catch-per-unit-effort (CPUE) for habitat types, containing catch information for 574,145 carp caught at 4831 sites. Eastern Australia accounted for 96% of carp biomass and 92% of the total available wetted habitat area (16,686 km2) was occupied. To correct these data for variable detection efficiencies, we used existing electrofishing data and undertook additional field experiments to establish relationships between relative and absolute abundances. We then scaled-up site-based estimates to habitat types to generate continental estimates. The number of carp was estimated at 199.2 M (95%Crl: 106 M to 357.6 M) for an ‘average’ hydrological scenario and 357.5 M (95%Crl: 178.9 M to 685.1 M) for a ‘wet’ hydrological scenario. In eastern Australia, these numbers correspond with biomasses of 205,774 t (95%Crl: 117,532–356,482 t) (average scenario) and 368,357 t (95%Crl: 184,234–705,630 t) (wet scenario). At a continental scale the total biomass was estimated at 215,456 t for an ‘average’ hydrological scenario. Perennial lowland rivers had the highest CPUE and greatest biomass density (up to 826 kg/ha) and the modelled biomass exceeded a density-impact threshold of 80–100 kg/ha in 54% of wetlands and 97% of stream area in large lowland rivers. The continental-scale biomass estimates provide a baseline for focusing national conservation strategies to reduce carp populations below thresholds needed to restore aquatic ecosystems at a range of spatial scales.

The Queensland fruit fly (Qfly; Bactrocera tryoni Froggatt) is a major horticultural pest species in Australia and considered a serious quarantine concern, both domestically and internationally. When a Qfly outbreak is declared in the main horticultural regions of southeast Australia, a 15 km radius suspension zone is established (area of 707 km2) around the epicentre of the outbreak. This suspension radius was originally adopted in the early 1990s. Here, we analysed trapping data in New South Wales (Australia) from 2002 to 2010 to evaluate the suitability of the 15 km suspension radius. There were 439 Qfly incursions recorded in Qfly-free zones, but only 9.9% of these incursions resulted in a declared outbreak. For these outbreaks, we conducted two distinct analyses. First, we modelled the distribution of infestation radii from the outbreaks and predicted the suspension distance using multiple levels of risk protection (e.g. probit 8.7, probit 9). Second, we fitted various probability distributions to individual fly trapping data and then estimated upper confidence limits for suspension distances, based on dispersal patterns of Qfly. Overall, the predicted suspension radii were substantially smaller than the current 15 km radius. Our results concur with other studies suggesting that Qfly are relatively poor colonisers. We propose that the current 15 km could be reduced substantially to 3 or 4 km.

Mitochondria are critical for life, yet their underlying evolutionary biology is poorly understood. In particular, little is known about interaction between two levels of evolution: between individuals and within individuals (competition between cells, mitochondria or mitochondrial DNA molecules). Rapid evolution is suspected to occur frequently in mitochondrial DNA, whose maternal inheritance predisposes advantageous mutations to sweep rapidly though populations. Rapid evolution is also predicted in response to changed selection regimes after species invasion or removal of pathogens or competitors. Here, using empirical and simulated data from a model invasive bird species, we provide the first demonstration of rapid selection on the mitochondrial genome within individuals in the wild. Further, we show differences in mitochondrial DNA copy number associated with competing genetic variants, which may provide a mechanism for selection. We provide evidence for three rarely documented phenomena: selection associated with mitochondrial DNA abundance, selection on the mitochondrial control region, and contemporary selection during invasion.

Early detection of Dacinae (Diptera: Tephritidae) incursions is vital for quick and effective management. A key method for early detection is the implementation of trapping networks in risk areas. Australia relies on the use of extensive trapping networks at, and surrounding, major ports and fruit production areas to identity any fruit fly incursions. Therefore, improvements to the trap efficiency of these networks may result in earlier detection of incursions and hence more rapid response times for management and/or eradication. The current Australian standard, the Lynfield trap, is expensive and does not stack readily for transport. Here, we test the effectiveness of a new trap type, the Probodelt cone trap, compared to the Lynfield trap. We ran three separate trials: an experimental trial in an inland orchard and two field trials using the Sydney trapping network. At the orchard site, Lynfield and Probodelt traps had similar capture rates (within 10%) for Bactrocera tryoni. For the Sydney trapping network, no significant differences were found in the number of species caught or trap rates for B. tryoni, Bactrocera cacuminata, Dacus absonifacies, and Dacus aequalis. Thus, the Probodelt cone trap was at least comparable to the Lynfield trap. Given the economic advantages of the Probodelt cone trap (e.g. reduced shipping and storage costs), the use of the Probodelt trap may be preferred over the Lynfield trap.

We employed patch use theory to evaluate how several environmental factors influence the foraging behaviour of two rodent species: Grammomys dolichurus and Acomys cahirinus. Foraging efficiency was determined by measuring the remaining food in artificial food patches (giving-up densities: GUDs) from two experiments. In the first experiment, we placed patches in different microhabitat types (cover vs open) and at varying distances from cover. This experiment was conducted during three moon stages (waxing, full, waning).We found that the rodents had higher GUDs (lower foraging efficiency) in the open microhabitat. The distance from nearest shelter had a marginally significant positive effect on GUDs. GUDs were higher in both microhabitat types during the waxing and full phases, but decreased sharply once the moon began to rise after sunset. These results are likely due to higher predation risk away from cover and in more illuminated environments. In the second experiment, we examined mouse responses to seeds impregnated with plant toxins. Seeds impregnated with oxalic acid were avoided by the rodents, while seeds soaked in tannic acid did not differ significantly from control seeds. Our results highlight important ecological factors affecting the foraging behaviour of these rodents.Key words: giving-up densities, foraging ecology, kopje, lunar.

The sterile insect technique has been routinely used to eradicate fruit fly Bactrocera tryoni (Froggatt) incursions. This study considers whether fly quality in a mass-rearing facility can be improved by reducing irradiation doses, without sacrificing reproductive sterility. Pupae were exposed to one of five target irradiation dose ranges: 0, 40-45, 50-55, 60-65, and 70-75 Gy. Pupae were then assessed using routine quality control measures: flight ability, sex ratio, longevity under nutritional stress, emergence, and reproductive sterility. Irradiation did not have a significant effect on flight ability or sex ratio tests. Longevity under nutritional stress was significantly increased at 70-75 Gy, but no other doses differed from 0 Gy. Emergence was slightly reduced in the 50-55, 60-65, and 70-75 Gy treatments, but 40-45 Gy treatments did not differ from 0 Gy, though confounding temporal factors complicate interpretation. Reproductive sterility remained acceptable (> 99.5%) for all doses--40-45 Gy (99.78%), 50-55 Gy (100%), 60-65 Gy (100%), and 70-75 Gy (99.99%). We recommend that B. tryoni used in sterile insect technique releases be irradiated at a target dose of 50-55 Gy, providing improved quality and undiminished sterility in comparison with the current 70-75 Gy standard while also providing a substantial buffer against risk of under dosing.

In the context of reproduction, glucocorticoids (GCs) are generally considered to have negative effects. However, in well-studied model species, GCs fluctuate predictability across the estrous cycles, and short-term increases promote healthy ovarian function. Reproductive challenges have plagued captive elephant populations, which are not currently self-sustaining. Efforts to understand reproductive dysfunction in elephants have focused on the suppressive effects of cortisol, but the potential permissive or stimulatory effects of cortisol are unknown. In this study, we provide a detailed examination of cortisol patterns across the estrous cycle in Asian elephants (Elephas maximus). Time series analysis was used to analyze cortisol and progesterone data for a total of 73 cycles from eight females. We also compared cortisol profiles between females that successfully conceived and females that failed to conceive despite repeated mating attempts. Our results revealed that cortisol fluctuates predictably across the estrous cycle, with a peak during the second half of the follicular phase followed by low levels throughout the luteal phase. Furthermore, this pattern was significantly altered in nulliparous females; cortisol concentrations did not decline during the luteal phase to the same extent as in parous females. This study highlights the complexity of cortisol signaling and suggests future directions for understanding the role of cortisol in reproductive dysfunction.

BACKGROUND The Sterile Insect Technique (SIT) requires vast numbers of consistently high quality insects to be produced over long periods. Quality control (QC) procedures are critical to effective SIT, both providing quality assurance and warning of operational deficiencies. We here present a potential new QC assay for mass rearing of Queensland fruit flies (Bactrocera tryoni Froggatt) for SIT; locomotor activity monitoring. We investigated whether automated locomotor activity monitors (LAMs) that simply detect how often a fly passes an infrared sensor in a glass tube might provide similar insights but with much greater economy. RESULTS Activity levels were generally lower for females than for males, and declined over five days in the monitor for both sexes. Female activity levels were not affected by irradiation, but males irradiated at 60 or 70 Gy had reduced activity levels compared with unirradiated controls. We also found some evidence that mild heat shock of pupae results in adults with reduced activity. CONCLUSION LAM offers a convenient, effective and economical assay to probe such changes. © 2013 Society of Chemical Industry

Males often have reduced immune function compared to females but the proximate mechanisms underlying this taxonomically widespread pattern are unclear. Because immune function is resource-dependent and sexes may have different nutritional requirements, we hypothesized that sexual dimorphism in immune function may arise from differential nutrient intake (acquisition hypothesis). To test this hypothesis, we examined patterns of phenoloxidase (PO) activity in relation to nutrient consumption in Queensland fruit flies (Q-flies). In the first experiment, flies were allowed to choose their preferred nutrient intake. Compared with males, female Q-flies had higher PO activity, consumed more calories, and preferred a higher protein:carbohydrate (P:C) diet, suggesting that differential acquisition could explain sex differences. In the second experiment, we restricted flies to one of 12 diets varying in protein and carbohydrate concentrations and mapped PO activity for each sex onto a nutritional landscape. Counter to our hypothesis, females had higher PO activity than males at any given level of nutrient intake. Both carbohydrate and protein intake affected PO activity in females but only protein affected PO activity in males. Our results indicate that sex differences in Q-fly immune function are not solely explained by sex differences in nutrient intake, although nutrition does contribute to the magnitude of these sex differences.

There is a long-standing interest in behavioural ecology, exploring the causes and correlates of consistent individual differences in mean behavioural traits (‘personality’) and the response to the environment (‘plasticity’). Recently, it has been observed that individuals also consistently differ in their residual intraindividual variability (rIIV). This variation will probably have broad biological and methodological implications to the study of trait variation in labile traits, such as behaviour and physiology, though we currently need studies to quantify variation in rIIV, using more standardized and powerful methodology. Focusing on activity rates in guppies ( Poecilia reticulata ), we provide a model example, from sampling design to data analysis, in how to quantify rIIV in labile traits. Building on the doubly hierarchical generalized linear model recently used to quantify individual differences in rIIV, we extend the model to evaluate the covariance between individual mean values and their rIIV. After accounting for time-related change in behaviour, our guppies substantially differed in rIIV, and it was the active individuals that tended to be more consistent (lower rIIV). We provide annotated data analysis code to implement these complex models, and discuss how to further generalize the model to evaluate covariances with other aspects of phenotypic variation.

The trade-off between lifespan and reproduction is commonly explained by differential allocation of limited resources. Recent research has shown that the ratio of protein to carbohydrate (P : C) of a fly's diet mediates the lifespan–reproduction trade-off, with higher P : C diets increasing egg production but decreasing lifespan. To test whether this P : C effect is because of changing allocation strategies (Y-model hypothesis) or detrimental effects of protein ingestion on lifespan (lethal protein hypothesis), we measured lifespan and egg production in Queensland fruit flies varying in reproductive status (mated, virgin and sterilized females, virgin males) that were fed one of 18 diets varying in protein and carbohydrate amounts. The Y-model predicts that for sterilized females and for males, which require little protein for reproduction, there will be no effect of P : C ratio on lifespan; the lethal protein hypothesis predicts that the effect of P : C ratio should be similar in all groups. In support of the lethal protein hypothesis, and counter to the Y-model, the P : C ratio of the ingested diets had similar effects for all groups. We conclude that the trade-off between lifespan and reproduction is mediated by the detrimental side-effects of protein ingestion on lifespan.

Burst speed, a common metric of locomotor performance, is typically measured on horizontal surfaces. Studies of locomotor performance in spiders have examined vertical and horizontal locomotion, but not the effects of intermediate inclines. We measured the effect of angle of incline, body size, relative leg length (variation not explained by body size) and relative body mass (variation not explained by body size or leg length, often interpreted as ‘condition’) on maximum running speed in two Australian spiders differing in habitat architecture and foraging strategy. Males of a ground-dwelling jumping spider, Jacksonoides queenslandica, and an orb-web spider, Nephila plumipes, were forced to run on raceways inclined at +0°, +30° and +60°. At the population level, maximum running speed decreased on inclined compared to level raceways for both species, but the effects of slope varied substantially between individuals. While speed was not influenced by body size or relative mass in either species, it was influenced by leg length in both. In J. queenslandica, relative leg length did not influence horizontal running speed but was positively related to speed on slopes, principally because relatively short-legged spiders were slowed whereas relatively long-legged spiders were not affected. In N. plumipes relative leg length was related negatively to running speed on horizontal surfaces but positively to running speed on slopes. Unlike J. queenslandica, N. plumipes was little affected by slopes of 30° but slowed markedly on slopes of 60°, irrespective of relative leg length. We suggest that performance advantages on inclines may promote relatively longer legs.

The growing number of wildlife endocrinology studies have greatly enhanced our understanding of comparative endocrinology, and have also generated extensive longitudinal data for a vast number of species. However, the extensive graphical analysis required for these longitudinal datasets can be time consuming because there is often a need to create tens, if not hundreds, of graphs. Furthermore, routine methods for summarising hormone profiles, such as the iterative baseline approach and area under the curve (AUC), can be tedious and non-reproducible, especially for large number of individuals. We developed an R package, hormLong, which provides the basic functions to perform graphical and numerical analyses routinely used by wildlife endocrinologists. To encourage its use, hormLong has been developed such that no familiarity with R is necessary. Here, we provide a brief overview of the functions currently available and demonstrate their utility with previously published Asian elephant data. We hope that this package will promote reproducibility and encourage standardization of wildlife hormone data analysis.

Adult Queensland fruit flies, Bactrocera tryoni (Froggatt), require adequate post-teneral nutrition to complete reproductive development, to perform sexually and for maximum longevity. Recent research has focussed on nutritional requirements of adults released in sterile insect technique (SIT) programmes that are used to manage these pests. Several studies have suggested benefits of providing yeast hydrolysate (YH) in addition to sugar during the 24- to 48-h pre-release holding period. Current evidence suggests that provision of YH can induce faster development, increased mating probability, longer copulations, increased sperm storage by mates, higher levels of sexual inhibition in mated females and increased longevity. We here review research on adult B. tryoni nutritional requirements, assess the potential application of this information in the context of SIT programmes and highlight future research that will help to determine whether YH, or other supplements, should be included as pre-release treatment in B. tryoni SIT.

Group size and vigilance are tools that animals can use to mitigate predation risk, and many studies have reported a negative relationship between them. Vigilance studies often investigate the direct effect of group size on vigilance, but they ignore the effect of ecological factors on group size. As a consequence, these studies can overlook important indirect effects of ecological factors on vigilance via group size. We investigated how ecological factors affect vigilance behavior in rock hyraxes (Procavia capensis), both directly and indirectly via group size. First, we showed a direct negative relationship between group size and vigilance behavior by measuring vigilance behavior before and after a change in group size. Second, we conducted a path analysis that included group size and several ecological factors (distance from shelter, distance from center of kopje, vegetation cover, and time since start of foraging session). Similar to the 1st analysis, the path analysis identified a strong negative relationship between group size and vigilance behavior; however, the other variables had little effect on group size or vigilance behavior, or both.

The African unstriped ground squirrel (Xerus rutilus) is widely dispersed across various habitats in East Africa and hence encounters a diverse suite of predators and plant communities. It is not known how different habitats and plant characteristics affect the foraging behaviour of X. rutilus. We used giving-up densities (GUDs) as a measure of foraging efficiency to explore the foraging costs of environmental heterogeneity. To determine foraging efficiency across spatial scales, we established food patches in two microhabitats (open and cover), which were nested within three habitats (koppie, edge and bushland). When foraging in a cover microhabitat, foraging efficiency decreased away from the koppie, but when in the open microhabitat, foraging efficiency was lowest near the koppie edge. Second, to determine foraging efficiency with common plant toxins, we presented the squirrels with seeds soaked in either tannic acid, oxalic acid or distilled water (control). Foraging efficiency did not differ between tannic-treated and control seeds, but oxalic-treated seeds had higher GUDs. Overall, our results suggest that X. rutilus is a remarkably efficient forager across multiple axes of environmental heterogeneity, which may have intriguing consequences for the ecological community.

Understanding predator-prey dynamics requires an understanding of how prey assess predation risk. This study tested the effect of microhabitat, moon stages, and mammalian predator urines (Vulpes vulpes [Red Fox], Mustela vison [Mink], and Procyon lotor [Raccoon]) on the degree of predation risk perceived by Peromyscus leucopus (White-footed Mouse). Giving-up densities from artificial food patches were used to quantify perceived predation risk. White-footed Mice exhibited a strong preference for cover microhabitat and for the new moon stage. However, the mice did not significantly alter their foraging behavior in response to the predator urines compared to a water control. Additionally, mice foraged less on colder nights. The results suggest that mammalian predator urines may not provide reliable information on actual predation risk for the White-footed Mice and that the mice extensively use indirect cues to assess predation risk.

Caloric restriction (CR) has been widely accepted as a mechanism explaining increased lifespan (LS) in organisms subjected to dietary restriction (DR), but recent studies investigating the role of nutrients have challenged the role of CR in extending longevity. Fuelling this debate is the difficulty in experimentally disentangling CR and nutrient effects due to compensatory feeding (CF) behaviour. We quantified CF by measuring the volume of solution imbibed and determined how calories and nutrients influenced LS and fecundity in unmated females of the Queensland fruit fly, Bactocera tryoni (Diptera: Tephritidae). We restricted flies to one of 28 diets varying in carbohydrate:protein (C:P) ratios and concentrations. On imbalanced diets, flies overcame dietary dilutions, consuming similar caloric intakes for most dilutions. The response surface for LS revealed that increasing C:P ratio while keeping calories constant extended LS, with the maximum LS along C:P ratio of 21:1. In general, LS was reduced as caloric intake decreased. Lifetime egg production was maximized at a C:P ratio of 3:1. When given a choice of separate sucrose and yeast solutions, each at one of five concentrations (yielding 25 choice treatments), flies regulated their nutrient intake to match C:P ratio of 3:1. Our results (i) demonstrate that CF can overcome dietary dilutions; (ii) reveal difficulties with methods presenting fixed amounts of liquid diet; (iii) illustrate the need to measure intake to account for CF in DR studies and (iv) highlight nutrients rather than CR as a dominant influence on LS.

Marsupial research, conservation, and management can benefit greatly from knowledge about glucocorticoid (GC) secretion patterns because GCs influence numerous aspects of physiology and play a crucial role in regulating an animal's response to stressors. Faecal glucocorticoid metabolites (FGM) offer a non-invasive tool for tracking changes in GCs over time. To date, there are relatively few validated assays for marsupials compared with other taxa, and those that have been published generally test only one assay. However, different assays can yield very different signals of adrenal activity. The goal of this study was to compare the performance of five different enzyme immunoassays (EIAs) for monitoring adrenocortical activity via FGM in 13 marsupial species. We monitored FGM response to two types of events: biological stressors (e.g., transport, novel environment) and pharmacological stimulation (ACTH injection). For each individual animal and assay, FGM peaks were identified using the iterative baseline approach. Performance of the EIAs for each species was evaluated by determining (1) the percent of individuals with a detectable peak 0.125–4.5 days post-event, and (2) the biological sensitivity of the assay as measured by strength of the post-event response relative to baseline variability (Z-score). Assays were defined as successful if they detected a peak in at least 50% of the individuals and the mean species response had a Z ⩾ 2. By this criterion, at least one assay was successful in 10 of the 13 species, but the best-performing assay varied among species, even those species that were closely related. Furthermore, the ability to confidently assess assay performance was influenced by the experimental protocols used. We discuss the implications of our findings for biological validation studies.

Animals that have a long pre-reproductive adult stage often employ mechanisms that minimize aging over this period in order to preserve reproductive lifespan. In a remarkable exception, one tephritid fruit fly exhibits substantial pre-reproductive aging but then mitigates this aging during a diet-dependent transition to the reproductive stage, after which life expectancy matches that of newly emerged flies. Here, we ascertain the role of nutrients, sexual maturation and mating in mitigation of previous aging in female Queensland fruit flies. Flies were provided one of three diets: ‘sugar’, ‘essential’, or ‘yeast-sugar’. Essential diet contained sugar and micronutrients found in yeast but lacked maturation-enabling protein. At days 20 and 30, a subset of flies on the sugar diet were switched to essential or yeast-sugar diet, and some yeast-sugar fed flies were mated 10 days later. Complete mitigation of actuarial aging was only observed in flies that were switched to a yeast-sugar diet and mated, indicating that mating is key. Identifying the physiological processes associated with mating promise novel insights into repair mechanisms for aging.

27 28 Sexual dimorphism in immunocompetence, with males having lower immune 29 function, is a prevalent pattern in nature. The main evolutionary explanation for this 30 pattern is that males preferentially allocate resources away from immune function and 31 towards reproductive effort to increase their competitiveness for limited females. 32 However, the role of differential predation risk between the sexes has not been 33 considered, despite predation risk being a major driver of life history strategies and 34 male sexual traits often having associated predation costs. It is unclear whether 35 increased predation risk should increase or decrease investment in immune function, 36 as males have been shown to utilize both behavioural (e.g. decrease foraging activity) 37 and/or life-history (e.g. decrease investment in sexual trait) defense strategies to 38 manage predation risk. Here, we modelled optimal resource acquisition and 39 allocation towards immune function under differential predation risk with multiple 40 defense strategies. If males have limited defense strategies, increasing predation risk 41 caused males to trade-off immune function for reproductive effort, leading to reduced 42 immunocompetence. In contrast, if males can only decrease predation risk through 43 reduction of reproductive effort (e.g. decrease colouration or calling rates), then 44 increasing predation risk causes immune function to increase. If males can utilize 45 multiple defense strategies and sexual selection is low, then males maintain a constant 46 immune function as predation risk increases. Sexual selection robustly resulted in 47 decreased immunocompetence. Overall, our results suggest that predation plays an 48 important role in the evolution of sexual dimorphism in immunocompetence, but 49 predicting its effect requires understanding the integrated defense strategies available. 50 51 52 53

Viral RNAs are detected commonly in serum by hybridization and polymerase chain reaction (PCR) methods for both clinical and research purposes. Two methods of extracting viral RNA were evaluated prior to amplification by PCR. One was a conventional phenol-chloroform method and the other used a standardized, manufactured kit. The efficiency of extraction was tested by semi-quantitative amplification of hepatitis C viral and GB virus-C/hepatitis G viral RNAs. The standarized commercial method, although more time efficient, resulted in an approximately ten fold less sensitivity. Thus, in situations where maximum sensitivity is needed, the more labor intenstive phenol choloroform method is recommended.

The Sterile Insect Technique (SIT) requires vast numbers of consistently high quality insects to be produced over long periods. Quality control (QC) procedures are critical to effective SIT, both providing quality assurance and warning of operational deficiencies. We here present a potential new QC assay for mass rearing of Queensland fruit flies (Bactrocera tryoni Froggatt) for SIT; locomotor activity monitoring. We investigated whether automated locomotor activity monitors (LAMs) that simply detect how often a fly passes an infrared sensor in a glass tube might provide similar insights but with much greater economy.Activity levels were generally lower for females than for males, and declined over five days in the monitor for both sexes. Female activity levels were not affected by irradiation, but males irradiated at 60 or 70 Gy had reduced activity levels compared with unirradiated controls. We also found some evidence that mild heat shock of pupae results in adults with reduced activity.LAM offers a convenient, effective and economical assay to probe such changes.

SUMMARYFeeding behaviour is an expression of an animal’s underlying nutritional strategy. The study of feeding decisions can hence delineate nutritional strategies. Studies of Drosophila melanogaster feeding behaviour have yielded conflicting accounts, and little is known about how nutrients affect feeding patterns in this important model species. Here, we conducted two experiments to characterize nutrient prioritization and regulation. In a choice experiment, we allowed female flies to self-regulate their intake of yeast, sucrose and water by supplying individual flies with three microcapillary tubes: one containing only yeast of varying concentrations, another with just sucrose of varying concentrations, and the last with just water. Flies tightly regulated yeast and sucrose to a constant ratio at the expense of excess water intake, indicating that flies prioritize macronutrient regulation over excess water consumption. To determine the relative importance of yeast and sucrose, in a no-choice experiment, we provided flies with two microcapillary tubes: the first with one of the 28 diets varying in yeast and sucrose content and the other with only water. Flies increased total water intake in relation to yeast consumption but not sucrose consumption. Additionally, flies increased diet intake as diet concentration decreased and as the ratio of sugar to yeast equalized. Using a geometric scaling approach, we found that the patterns of diet intake can be explained by flies prioritizing protein and carbohydrates equally and by the lack of substitutability between the nutrients. We conclude by illustrating how our results harmonize conflicting results in the literature once viewed in a two-dimensional diet landscape.

Insect lifespan is often closely linked to diet, and diet manipulations have been central to studies of ageing. Recent research has found that lifespan for some flies is maximised on a very low yeast diet, but once all yeast is removed, lifespan drops precipitously. Although effects of yeast availability on lifespan are commonly interpreted in terms of protein, yeast is a complex mix of nutrients and provides a rich source of vitamins, minerals and sterols. Elucidating which components of yeast are involved in this lifespan drop provides insights into more specific nutritional requirements and also provides a test for the commonplace interpretation of yeast in terms of protein. To this end, we fed Queensland fruit flies (Bactrocera tryoni) one of eight experimental diets that differed in the nutrient group(s) found in yeast that were added to sucrose: none, vitamins, minerals, amino acids, cholesterol, vitamin + mineral + cholesterol (VMC), vitamin + mineral + cholesterol + amino acids (VMCA), and yeast. We measured survival rates and egg production in single sex and mixed sex cages, as well as nutrient intake of individual flies. We found that the addition of minerals increased lifespan of both male and female flies housed in single sex cages by decreasing baseline mortality. The addition of just amino acids decreased lifespan in female flies; however, when combined with other nutrient groups found in yeast, amino acids increased lifespan by decreasing both baseline mortality and age-specific mortality. Flies on the yeast and VMCA diets were the only ones to show significant egg production. We conclude that the drop in lifespan observed when all yeast is removed is explained by missing micronutrients (vitamins, minerals and cholesterol) as well as the absence of protein in females, whereas minerals alone can explain the pattern for males. These results indicate a need for caution when interpreting effects of dietary yeast as effects of protein.

Dietary restriction extends life span across a vast diversity of taxa, but significant challenges remain in elucidating the underlying mechanisms. Distinguishing between caloric and nutrient effects is an essential step. Recent studies with Drosophila and tephritid fruit flies have reported increased life span as dietary yeast-to-sugar ratios decreased and these effects have been attributed to changes in protein-to-carbohydrate (P:C) ratios of the diets rather than calories. However, yeast is a complex mix of macronutrients and micronutrients, and hence changes in yeast content of the diet necessarily alters other nutrients in lockstep. To explicitly test whether studies using yeast are justified in attributing results to diet protein content rather than correlated nutrients, we developed a chemically defined diet allowing manipulation of just the ratio of protein (free amino acids) to carbohydrate (sucrose) levels of diets while holding other nutrients constant. Mated, female Queensland fruit flies (Q-flies) were fed 1 of 18 diets varying in P:C ratios and diet concentration. Diet consumption, egg production, and life span were recorded for each fly. In close concordance with recent studies using yeast diets, flies had increased life span as P:C ratios decreased, and caloric restriction did not extend life span. Similarly, egg production was maximized on high P:C ratios, but lifetime egg production was maximized on intermediate P:C ratios, indicating a life history trade-off between life span and egg production rate. Finally, Q-flies adjusted their diet intake in response to P:C ratios and diet concentration. Our results substantiate recent claims that P:C ratios significantly modulate life span in flies.

Numbers of wild Tasmanian devils are declining as a result of the fatal, transmissible Devil Facial Tumor Disease. A captive insurance population program has been initiated but current captive breeding rates are sub-optimal and therefore the goal of this project was to increase our understanding of the estrous cycle of the devil and elucidate potential causes of failed male-female pairings. Temporal patterns of fecal progestagen and corticosterone metabolite concentrations were examined for females (n=41) in three categories of reproductive status (successful: viable young, n=20 estrous cycles; unsuccessful: paired with a male but no young confirmed, n=44 estrous cycles; non-mated: no access to a male during estrus, n=8 estrous cycles) but substantial differences were not found. Females were more likely to produce pouch young if pairing with the male extended into late proestrus (P0.05), thereby decreasing the time between pairing and presumed ovulation. The interval between the end of proestrous elevation in progestagen metabolite concentrations and the beginning of the luteal phase was 7.6±2.3 days in successful females. The length of the luteal phase in successful females was 12.5±1.4 days which was not different from unsuccessful or non-mated females (P0.05). Unsuccessful females had 1-3 estrous cycles within a single year. Successful females were predominantly wild-caught (17/19, 90%) and most produced young following the first estrous cycle of the season (18/20, 90%). Unsuccessful females were predominantly captive born (20/27, 74%) in this study. It is possible that a proportion of females that do not produce pouch young achieve conception but the timing of reproductive failure continues to be elusive in this species.
