Garlic cultivars are sexually sterile under standard growth conditions, with direct implications for commercial production costs as well as breeding programs. Garlic is propagated commercially via bulblets, which facilitates disease transmission and virus load accumulation over vegetative generations. Tissue culture produces virus-free clones that are more productive, while keeping the desired traits of the cultivar. Consequently, this technique allows studies of garlic genetics as well as guarantees genetic conservation of varieties. We aimed at analyzing the in vitro regeneration of eight marketable cultivars of garlic using root segments as explants. For each genotype, bulblet-derived explants were isolated and introduced into MS medium supplemented with 2,4-D and 2-iP. Calli were transferred to MS medium supplemented with 8.8 mM BAP and 0.1 mM NAA (regeneration medium A), or with 4.6 mM kinetin alone (regeneration medium B). The calli were then evaluated for regeneration frequency after sixty days of in vitro cultivation. The noble cultivar 'Jonas' presented the highest rates of plant regeneration among the cultivars tested. The medium A, which contained auxin and cytokinin, induced the highest regeneration rates of all cultivars. The process described herein is simple, reproducible and can potentially be used as a tool in molecular breeding strategies for other marketable cultivars and genotypes of garlic. Cultivares de alho são sexualmente estéreis sob condições padrão de cultivo, com implicações diretas nos custos de produção comercial, bem como em programas de melhoramento. O alho é comercialmente propagado por meio de bulbilhos, o que facilita a transmissão de doenças e leva ao acúmulo de cargas virais ao longo das gerações. A cultura de tecidos produz clones livres de vírus que são mais produtivos, mantendo as características desejadas da cultivar. Consequentemente, esta técnica permite estudar a genética do alho, bem como garantir a conservação genética das variedades. Nosso objetivo foi analisar a regeneração in vitro de oito cultivares comerciais de alho usando segmentos de raiz como explante. Para cada genótipo, explantes derivados de bulbilho foram isolados e introduzidos em meio MS suplementado com 2,4-D e 2-iP. Calos foram transferidos para meio MS suplementado com 8,8 mM BAP e 0,1 mM NAA (meio de regeneração A), ou com 4,6 mM cinetina somente (meio de regeneração B). Os calos foram avaliados quanto à frequência de regeneração após sessenta dias de cultivo in vitro. A cultivar nobre 'Jonas' apresentou as maiores taxas de regeneração entre as cultivares testadas. O meio de cultura A, o qual continha auxina e citocinina, induziu as maiores taxas de regeneração em todas as cultivares. O processo aqui descrito é simples, reprodutível e pode ser usado potencialmente como uma ferramenta em estratégias de melhoramento para outras cultivares comerciais e genótipos de alho.

A floração é um processo vital durante o ciclo de vida das plantas e é marcado pela conversão do meristema apical vegetativo em reprodutivo devido a interações de fatores internos e externos à planta. Apesar de amplo conhecimento ter sido gerado nessa área, ainda se conhece muito pouco sobre o processo de floração e frutificação em tomateiro. Pesquisas anteriores realizadas pelo nosso grupo identificaram o cDNA homólogo a Proteína Reprimida por Auxina (ARP) em bibliotecas subtrativas reprodutivas de tomateiro. Existem poucos dados sobre a proteína ARP na literatura, há relatos de que o gene ARP está relacionado com a maturação do fruto em morango, dormência da gema lateral em ervilha e maturação do pólen em tabaco, mas a função da proteína ARP ainda não está clara. Portanto, o intuito deste trabalho foi compreender o papel da proteína ARP no desenvolvimento vegetativo e nos processos de floração e frutificação por meio da perda e/ou do ganho de função deste cDNA em plantas transgênicas de tomateiro contendo o cassete de superexpressão em orientação senso e antissenso para este cDNA. Dessa forma, foram observadas algumas alterações fenotípicas e estruturais nas plantas transgênicas (35S::SlARP antissenso e senso), como floração e frutificação precoce verificada nas plantas 35S::SlARP antissenso nas gerações T1 a T4, em relação às controles (transformada com o plasmídeo vazio e não transformada). Na análise histológica, notaram-se óvulos maduros nas flores das plantas 35S::SlARP antissenso (gerações T2 a T4), enquanto que as flores das controles não apresentaram óvulos maduros no mesmo período de tempo. Outra modificação observada foi o número superior de gemas laterais desenvolvidas nas plantas 35S::SlARP antissenso nas gerações T3 e T4 quando comparado às plantas 35S::SlARP senso e às plantas controles (transformada com o plasmídeo vazio e não transformada) . Essas produziram ramos com folhas, flores e frutos. Assim como alterações estruturais nos pecíolos das plantas 35S::SlARP antissenso, incluindo uma aparente quantidade maior de elementos de vaso (xilema e floema) do que os pecíolos as plantas 35S::SlARP senso e mutantes relacionados à sinalização da auxina (dgt e entire) e plantas controles. Portanto, estes dados sugerem que a proteína ARP possa estar envolvida na floração, frutificação e na dormência das gemas axilares em tomateiro. Flowering is a vital process during the plant life cycle and it is characterized by the conversion from the vegetative apical meristem into reproductive meristem due to internal and external factors. Despite the considerable knowledge has been produced in this field, not much is known about the flowering and fruiting process in tomato. Furthermore, previous research from our group has identified a cDNA with homology to AUXIN REPRESSED PROTEIN (ARP) in reproductive subtractive libraries from tomato. In the literature, there are few reports where the ARP gene has been associated to fruit ripening in strawberry, dormancy in pea and pollen maturation in tobacco plants, but it is not clear the ARP protein function yet. Therefore, the aim of this work was to understand the role of ARP protein in vegetative development, flowering and fruit set processes through the loss and/or gain of function using this cDNA in transgenic tomato plants with the overexpression cassette constructs in sense and antisense cDNA orientation. Thus, it was observed some phenotypic and structural modifications in transgenic plants (35S::SlARP antisense and sense) such as early flowering and fruiting that was observed in 35S::SlARP antisense plants from the T1 to T4 progeny when compared to controls plants (with the empty vector and wild type plant). In the histological analysis, it was observed mature ovule in 35S::SlARP antisense flowers (progeny T2 to T4), while for the controls plant it wasn’t observed a mature ovule at the same period of time. The other modification observed was a higher number of lateral buds developed in 35S::SlARP antisense plants (progeny T3 and T4) when compared to 35S::SlARP sense and controls plants. These plants produced branches with leaves, flowers and fruits. Besides, it has been observed some structural changes in the petioles from 35S::SlARP antisense plants, including a high number of vessel elements (xylem and phloem) when compared to 35S::SlARP sense, and to dgt and entire mutants (related to auxin signaling) and controls plants (with empty vector and wild type plant). Therefore, these data suggest that ARP protein may be involved in flowering, fruit set and dormancy of axillary buds in tomato.
