12. A Missense Mutation in the Extracellular Domain of alphaENaC Causes Liddle Syndrome

Item does not contain fulltext, Liddle syndrome is an autosomal dominant form of hypokalemic hypertension due to mutations in the beta- or gamma-subunit of the epithelial sodium channel (ENaC). Here, we describe a family with Liddle syndrome due to a mutation in alphaENaC. The proband was referred because of resistant hypokalemic hypertension, suppressed renin and aldosterone, and no mutations in the genes encoding beta- or gammaENaC. Exome sequencing revealed a heterozygous, nonconservative T>C single-nucleotide mutation in alphaENaC that substituted Cys479 with Arg (C479R). C479 is a highly conserved residue in the extracellular domain of ENaC and likely involved in a disulfide bridge with the partner cysteine C394. In oocytes, the C479R and C394S mutations resulted in similar twofold increases in amiloride-sensitive ENaC current. Quantification of mature cleaved alphaENaC in membrane fractions showed that the number of channels did not increase with these mutations. Trypsin, which increases open probability of the channel by proteolytic cleavage, resulted in significantly higher currents in the wild type than in C479R or C394S mutants. In summary, a mutation in the extracellular domain of alphaENaC causes Liddle syndrome by increasing intrinsic channel activity. This mechanism differs from that of the beta- and gamma-mutations, which result in an increase in channel density at the cell surface. This mutation may explain other cases of patients with resistant hypertension and also provides novel insight into ENaC activation, which is relevant for kidney sodium reabsorption and salt-sensitive hypertension.

16. Sodium phenylacetate enhances the inhibitory effect of dextran derivative on breast cancer cell growth in vitro and in nude mice.

Abstract

Sodium phenylacetate (NaPa) and carboxymethyl benzylamide dextran derivative (CMDB[SUBLS4]) are able to inhibit growth of breast tumour cells. In this study, we explored whether the combination of NaPa and CMDB[SUBLS4] may enhance their respective inhibitory effects on the MCF-7ras cell growth in vitro and in vivo. NaPa inhibited MCF-7ras cell proliferation by reducing the DNA replication concomitantly with a recruitment of cells in G0/G1 phase and by inducing apoptosis in a dose- and time-dependent manner. The addition of CMDB[SUBLS4] potentiated the NaPa antiproliferative effect in the manner dependent on the ratio of CMDB[SUBLS4] and NaPa concentrations. In nude mice, CMDB[SUBLS4] (150 mg kg[SUP-1]) or NaPa (40 mg kg[SUP-1]) administrated twice a week, for 7 weeks inhibited MCF-7ras xenograft growth by 40% and 60%, respectively. The treatment by both, CMDB[SUBLS4] and NaPa, decreased tumour growth by 83% without any toxicity. To better understand the mechanism of NaPa and CMDB[SUBLS4] action we assessed their effect on mitogenic activity of MCF-7ras conditioned medium (CM) on BALBC/3T3 fibroblasts. CMDB[SUBLS4] added to the CM, inhibited its mitogenic activity whereas NaPa had an anti-mitogenic effect when CM was prepared from MCF-7ras cells pretreated with NaPa. Thus, the antiproliferative effects of NaPa and CMDB[SUBLS4] involve 2 different mechanisms explaining, at least in part, the possible synergism between them. Overall, this study points to the potential use of a combination of dextran derivatives with NaPa to inhibit the breast tumour growth. [ABSTRACT FROM AUTHOR]

Abstract

The power electronic converters have a heavily influence in our life-style, since they are today used everywhere in-home applications, office, medicine, industry, and energy transmission machines and processes. The power electronic systems are based on fast-switching power semiconductors, such as diodes, mosfets, igbts, etc. These power devices represent the fragile part in a power electronic converter. Thus, to ensure safety and reliability of the power conversion system, a fault detection method must be provided. When a failure occurs, the fault detection and protection system become the most important function of the converter control unit. If the fault management is not adequate, the entire converter can be totally destroyed, causing catastrophic effects on the entire power conversion system. Accordingly, fault detection, classification, and diagnoses for all power electronic converters, including three phase rectifiers, three phase inverters, DC-DC converters and AC-AC converters, have to be investigated. In this paper, the fault analysis of the 3-phase 5-level E-Type Converter (3Φ5L E-Type Converter) is addressed. The 3Φ5L E-Type Converter is proposed for grid connected applications. All possible converter semiconductor switches and diodes faults are theoretically analyzed. The fault effects, the detectability and the safety shutdown actions are identified and summarized. The analysis is covered by simulation results performed in MATLAB/Simulink. The control algorithm has been implemented in FPGA using LabVIEW environmental. The experimental tests have been performed on the 20kVA prototype multilevel converter in order to validate the proposed analysis.

Abstract

A newly conceived symmetrical three-phase E- Type (3 $\Phi$7L E-Type) Inverter is proposed and analyzed in this paper. Converter topology is based on the single leg T- Type Inverter. The proposed topology shows seven voltage levels at the output providing better performance in terms of current and voltage stress of the power semiconductors and efficiency. The theoretical investigation has been carried out with reference to photovoltaic (PV) applications. A suitable thermal model of the power semiconductors has been developed in Plexim/PLECS environment. Simulation results of the 3 $\Phi$7L E-Type Inverter are performed in Matlab/Simulink environment. Furthermore, the prototype of the proposed converter has been carefully tested in order to validate the theoretical analysis.
