1. Internal-tide vertical structure and steric sea surface height signature south of New Caledonia revealed by glider observations

Abstract

In this study, we exploit autonomous underwater glider data to infer internal-tide dynamics south of New Caledonia, an internal-tide-generation hot spot in the southwestern tropical Pacific. By fitting a sinusoidal function to vertical displacements at each depth using a least-squares method, we simultaneously estimate diurnal and semidiurnal tides. Our analysis reveals regions of enhanced tidal activity, strongly dominated by the semidiurnal tide. To validate our findings, we compare the glider observations to a regional numerical simulation that includes tidal forcing. This comparison assesses the simulation's realism in representing tidal dynamics and evaluates the glider's ability to infer internal-tide signals and their signature in sea surface height (SSH). The glider observations and a pseudo glider, simulated using hourly numerical model output with identical sampling, exhibit similar amplitude and phase characteristics along the glider track. Existing discrepancies are in large part explained by tidal incoherence induced by eddy–internal-tide interactions. We infer the semidiurnal internal-tide signature in steric SSH by the integration of vertical displacements. Within the upper 1000 m, the pseudo glider captures roughly 78 % of the steric SSH total variance explained by the full water column signal. This value increases to over 90 % when projecting the pseudo glider's vertical displacements onto climatological baroclinic modes and extrapolating to full depth. Notably, the steric SSH from glider observations aligns closely with empirical estimates derived from satellite altimetry, highlighting the internal tide's predominant coherent nature during the glider's sampling.

Abstract

The southwestern tropical Pacific exhibits a complex bathymetry and represents a hot spot of internal-tide generation. Based on a tailored high-resolution regional model, we investigate for the first time the internal-tide field around the New Caledonia islands through energy budgets that quantify the coherent internal-tide generation, propagation, and dissipation. A total of 15.27 GW is converted from the barotropic to the baroclinic M2 tide with the main conversion sites associated with the most prominent bathymetric structures such as continental slopes and narrow passages in the north (2.17 GW) and ridges and seamounts south of New Caledonia (3.92 GW). The bulk of baroclinic energy is generated in shallow waters around 500 m depth and on critical to supercritical slopes, highlighting the limitations of linear semi-analytical models in those areas. Despite the strongly dominant mode-1 generation, more than 50 % of the locally generated energy either dissipates in the near field close to the generation sites or loses coherence. The remaining baroclinic energy propagates within well-defined tidal beams with baroclinic energy fluxes of up to 30 kW m−1 toward the open ocean. The New Caledonia site represents a challenge for SWOT (Surface Water and Ocean Topography) observability of balanced motion in the presence of internal tides with sea surface height (SSH) signatures >6 cm at similar wavelengths. We show for our study region that a correction of SSH for the coherent internal tide potentially increases the observability of balanced motion from wavelengths >160 km to well below 100 km.

Abstract

Mesoscale eddies modulate the ocean's physical, chemical, and biological properties. In cyclonic eddies (CEs), nutrient upwelling can stimulate primary production by phytoplankton. Yet, how this locally enhanced autotrophic production affects heterotrophy and consequently the metabolic balance between the synthesis and the consumption of dissolved organic matter (DOM) remains largely unknown. To fill this gap, we investigated the horizontal and vertical variability in auto- and heterotrophic microbial activity (biomass production and respiration) within a CE that formed off Mauritania and along the ∼ 900 km zonal corridor between Mauritania and the Cape Verde islands in the Eastern Tropical North Atlantic (ETNA). Our results show how the physical disturbances caused by the CE affected the biomass distribution of phyto- and bacterioplankton and their metabolic activities. The injection of nutrients into the sunlit surface resulted in enhanced autotrophic pico- and nanoplankton abundance and generally increased autotrophic activity as indicated by chlorophyll a (Chl a) concentration, primary production (PP), and extracellular release rates. However, the detailed eddy survey also revealed an uneven distribution of these variables with, for example, the highest Chl a concentrations and PP rates occurring near and just beyond the CE's periphery. The heterotrophic bacterial activity was similarly variable. Optode-based community respiration (CR), bacterial respiration (BR) estimates, and bacterial biomass production (BP) largely followed the trends of PP and Chl a. Thus, a submesoscale spatial mosaic of heterotrophic bacterial abundance and activities occurred within the CE that was closely related to variability in autotrophic production. Consistent with this, we found a significant positive correlation between concentrations of semi-labile dissolved organic carbon (SL-DOC; here the sum of dissolved hydrolysable amino acids and dissolved combined carbohydrates) and BR estimates. Extracellular release of carbon as indicated by primary production of dissolved organic carbon (PPDOC) was variable with depth and laterally and not always sufficient to compensate the bacterial carbon demand (BCD: BR + BP), with PPDOC accounting for between 28 % and 110 % of the BCD. Bacterial growth efficiency (BGE: BP / BCD) ranged between 1.7 % and 18.2 %. We estimated the metabolic state to establish whether the CE was a source or a sink of organic carbon. We showed that the CE carried a strong autotrophic signal in the core (PP / CR > 1). Our results suggest that submesoscale (0–10 km) processes lead to highly variable metabolic activities in both photoautotrophic and heterotrophic microorganisms. Overall, we revealed that the CEs not only trap and transport coastal nutrients and organic carbon to the open ocean but also stimulate phytoplankton growth, generating freshly produced organic matter during westward propagation. This drives heterotrophic processes and may contribute to the previously observed net heterotrophy in open Atlantic surface waters.

Abstract

Drinking water enters buildings with a given microbiological community composition. Within premise plumbing systems, the drinking water is subject to very different conditions and temperatures. Whereas part of the water stays cold, another part is heated to provide hot water. In this study, drinking water samples were taken at different locations in four buildings that had central heating circles and that were equipped with ultrafiltration modules. The latter were intended to keep bacterial numbers low. When studying the increase in bacterial concentrations in these water samples using regrowth tests at different incubation temperatures, a temperature-dependence could be observed. Bacteria in cold water samples propagated best when incubated at 22°C, but often poorly at 36°C and not at all at 50°C. Bacteria in hot water samples showed the reverse behavior and grew best when incubated at 50°C, whereas growth at 22°C was poor or associated with a long growth lag. Water samples from distal taps in periphery locations used for retrieving both cold and hot water showed intermediate growth behaviors. Results suggest the existence of different temperature-adapted bacterial populations within domestic drinking water systems. The finding was supported by sequence data revealing distinct differences in the microbiomes between cold and hot water samples. Abundant bacterial groups in hot water included Deinococci, Kryptonia, Ignavibacteria, Nitrospiria, Gemmatimonadetes and different genera of Gammaproteobacteria. Stagnation of hot water at 50°C, 55°C, or 60°C furthermore shaped the microbiome in different ways indicating that small temperature differences can have a substantial impact on the bacterial communities.

Abstract

Dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) is used to quantify perfusion and vascular permeability. In most cases a bolus arrival time (BAT) delay exists between the arterial input function (AIF) and the contrast agent arrival in the tissue of interest which needs to be estimated. Existing methods for BAT estimation are tailored to tissue concentration curves which have a fast upslope to the peak as frequently observed in patient data. However, they may give poor results for curves that do not have this characteristic shape such as tissue concentration curves of small animals. In this paper, we propose a novel method for BAT estimation of signals that do not have a fast upslope to their peak. The model is based on splines which are able to adapt to a large variety of concentration curves. Furthermore, the method estimates BATs on a continuous time scale. All relevant model parameters are automatically determined by generalized cross validation. We use simulated concentration curves of small animal and patient settings to assess the accuracy and robustness of our approach. The proposed method outperforms a state-of-the-art method for small animal data and it gives competitive results for patient data. Finally, it is tested on in vivo acquired rat data where accuracy of BAT estimation was also improved upon the state-of-the-art method. The results indicate that the proposed method is suitable for accurate BAT estimation of DCE-MRI data, especially for small animals.

Abstract

Thermal and chemical disinfection of technical water systems not only aim at minimizing the level of undesired microorganisms, but also at preventing excessive biofouling, clogging and interference with diverse technical processes. Typically, treatment has to be repeated in certain time intervals, as the duration of the effect is limited. The transient effect of disinfection was demonstrated in this study applying different treatments to water and biofilms including heat, chlorination, a combination of hydrogen peroxide and peracetic acid and monochloramine. Despite the diverse treatments, the reduction in live bacteria was followed by regrowth in all cases, underlining the universal validity of this phenomenon. The study shows that autochthonous bacteria can reach the concentrations given prior to treatment. The reason is seen in the nutrient concentration that has not changed and that forms the basis for regrowth. Nutrients are released by disinfection from lysed cells or are still fixed in dead biomass that is subsequently scavenged by necrotrophic growth. Treatment cycles therefore only provide a transient reduction of water microbiology if nutrients are not removed. When aiming at greater sustainability of the effect, biocidal treatment has to be equally concerned about nutrient removal by subsequent cleaning procedures as about killing efficiency. Abstract

The science guiding the EUREC4A campaign and its measurements is presented. EUREC4A comprised roughly 5 weeks of measurements in the downstream winter trades of the North Atlantic – eastward and southeastward of Barbados. Through its ability to characterize processes operating across a wide range of scales, EUREC4A marked a turning point in our ability to observationally study factors influencing clouds in the trades, how they will respond to warming, and their link to other components of the earth system, such as upper-ocean processes or the life cycle of particulate matter. This characterization was made possible by thousands (2500) of sondes distributed to measure circulations on meso- (200 km) and larger (500 km) scales, roughly 400 h of flight time by four heavily instrumented research aircraft; four global-class research vessels; an advanced ground-based cloud observatory; scores of autonomous observing platforms operating in the upper ocean (nearly 10 000 profiles), lower atmosphere (continuous profiling), and along the air–sea interface; a network of water stable isotopologue measurements; targeted tasking of satellite remote sensing; and modeling with a new generation of weather and climate models. In addition to providing an outline of the novel measurements and their composition into a unified and coordinated campaign, the six distinct scientific facets that EUREC4A explored – from North Brazil Current rings to turbulence-induced clustering of cloud droplets and its influence on warm-rain formation – are presented along with an overview of EUREC4A's outreach activities, environmental impact, and guidelines for scientific practice. 12. Longitudinal MRI study after carbon ion and photon irradiation: shorter latency time for myelopathy is not associated with differential morphological changes

Abstract

Abstract Background Radiation-induced myelopathy is a severe and irreversible complication that occurs after a long symptom-free latency time if the spinal cord was exposed to a significant irradiation dose during tumor treatment. As carbon ions are increasingly investigated for tumor treatment in clinical trials, their effect on normal tissue needs further investigation to assure safety of patient treatments. Magnetic resonance imaging (MRI)-visible morphological alterations could serve as predictive markers for medicinal interventions to avoid severe side effects. Thus, MRI-visible morphological alterations in the rat spinal cord after high dose photon and carbon ion irradiation and their latency times were investigated. Methods Rats whose spinal cords were irradiated with iso-effective high photon (n = 8) or carbon ion (n = 8) doses as well as sham-treated control animals (n = 6) underwent frequent MRI measurements until they developed radiation-induced myelopathy (paresis II). MR images were analyzed for morphological alterations and animals were regularly tested for neurological deficits. In addition, histological analysis was performed of animals suffering from paresis II compared to controls. Results For both beam modalities, first morphological alterations occurred outside the spinal cord (bone marrow conversion, contrast agent accumulation in the musculature ventral and dorsal to the spinal cord) followed by morphological alterations inside the spinal cord (edema, syrinx, contrast agent accumulation) and eventually neurological alterations (paresis I and II). Latency times were significantly shorter after carbon ions as compared to photon irradiation. Conclusions Irradiation of the rat spinal cord with photon or carbon ion doses that lead to 100% myelopathy induced a comparable fixed sequence of MRI-visible morphological alterations and neurological distortions. However, at least in the animal model used in this study, the observed MRI-visible morphological alterations in the spinal cord are not suited as predictive markers to identify animals that will develop myelopathy as the time between MRI-visible alterations and the occurrence of myelopathy is too short to intervene with protective or mitigative drugs.

Abstract

Abstract. The science guiding the EUREC4A campaign and its measurements is presented. EUREC4A comprised roughly 5 weeks of measurements in the downstream winter trades of the North Atlantic – eastward and southeastward of Barbados. Through its ability to characterize processes operating across a wide range of scales, EUREC4A marked a turning point in our ability to observationally study factors influencing clouds in the trades, how they will respond to warming, and their link to other components of the earth system, such as upper-ocean processes or the life cycle of particulate matter. This characterization was made possible by thousands (2500) of sondes distributed to measure circulations on meso- (200 km) and larger (500 km) scales, roughly 400 h of flight time by four heavily instrumented research aircraft; four global-class research vessels; an advanced ground-based cloud observatory; scores of autonomous observing platforms operating in the upper ocean (nearly 10 000 profiles), lower atmosphere (continuous profiling), and along the air–sea interface; a network of water stable isotopologue measurements; targeted tasking of satellite remote sensing; and modeling with a new generation of weather and climate models. In addition to providing an outline of the novel measurements and their composition into a unified and coordinated campaign, the six distinct scientific facets that EUREC4A explored – from North Brazil Current rings to turbulence-induced clustering of cloud droplets and its influence on warm-rain formation – are presented along with an overview of EUREC4A's outreach activities, environmental impact, and guidelines for scientific practice. 23. Regional modeling of internal tide dynamics around New Caledonia: energetics and sea surface height signature

Abstract

The Southwestern Tropical Pacific exhibits a complex bathymetry and represents a hot spot of internal tide generation. Based on a tailored high-resolution regional model, we investigate for the first time the internal tide field around the New Caledonia islands through energy budgets that quantify the internal tide generation, propagation, and dissipation. A total of 15.97 GW is converted from the barotropic to the baroclinic tide with the main conversion sites associated with the most prominent bathymetric structures such as continental slopes and narrow passages in the north (2.17 GW) and ridges and seamounts south of New Caledonia (3.92 GW). The bulk of baroclinic energy is generated in shallow waters around 500 m depth and on critical to supercritical slopes highlighting the limitations of linear semi-analytical models in those areas. Despite the strongly dominant mode-1 generation, more than 50 % of the locally generated energy dissipates in the near-field close to the generation sites. The remaining energy propagates within well-defined tidal beams with baroclinic energy fluxes of up to 30 kW m−1 toward the open ocean, strongly dominated by mode-1. The energetic mesoscale eddy activity in the region appears to be the main source of tidal incoherence. Locally, mesoscale eddy-driven stratification changes induce variations of the conversion term. In the far-field, incoherence of the energy flux arises through the interaction of the tidal beam with the eddying background flow. The New Caledonia site represents a challenge for SWOT (Surface Water Ocean Topography) observability of meso- and submesoscale dynamics in the presence of internal tides with sea surface height signatures > 6 cm. We show that a correction of the coherent baroclinic tide may improve the observability range by shifting the transition scale between balanced and unbalanced flow in winter from 180 km to 50–80 km. In contrast, in summer observability increases only marginally due to the seasonally amplified signature of the incoherent tide at scales below 100 km.

Abstract

Background and purpose Radiation-induced myelopathy, an irreversible complication occurring after a long symptom-free latency time, is preceded by a fixed sequence of magnetic resonance- (MR-) visible morphological alterations. Vascular degradation is assumed the main reason for radiation-induced myelopathy. We used dynamic contrast-enhanced (DCE-) MRI to identify different vascular changes after photon and carbon ion irradiation, which precede or coincide with morphological changes. Materials and methods The cervical spinal cord of rats was irradiated with iso-effective photon or carbon (12C-)ion doses. Afterwards, animals underwent frequent DCE-MR imaging until they developed symptomatic radiation-induced myelopathy (paresis II). Measurements were performed at certain time points: 1 month, 2 months, 3 months, 4 months, and 6 months after irradiation, and when animals showed morphological (such as edema/syrinx/contrast agent (CA) accumulation) or neurological alterations (such as, paresis I, and paresis II). DCE-MRI data was analyzed using the extended Toft’s model. Results Fit quality improved with gradual disintegration of the blood spinal cord barrier (BSCB) towards paresis II. Vascular permeability increased three months after photon irradiation, and rapidly escalated after animals showed MR-visible morphological changes until paresis II. After 12C-ion irradiation, vascular permeability increased when animals showed morphological alterations and increased further until animals had paresis II. The volume transfer constant and the plasma volume showed no significant changes. Conclusion Only after photon irradiation, DCE-MRI provides a temporal advantage in detecting early physiological signs in radiation-induced myelopathy compared to morphological MRI. As a generally lower level of vascular permeability after 12C-ions led to an earlier development of paresis as compared to photons, we conclude that other mechanisms dominate the development of paresis II.

25. Eddy-enhanced primary production sustains heterotrophic microbial activities in the Eastern Tropical North Atlantic

Abstract

Mesoscale eddies modulate the ocean's physical, chemical, and biological properties. In cyclonic eddies (CEs), nutrient upwelling can stimulate primary production by phytoplankton. Yet, how this locally enhanced autotrophic production affects heterotrophy and consequently the metabolic balance between the synthesis and the consumption of dissolved organic matter (DOM) remains largely unknown. To fill this gap, we investigated the horizontal and vertical variability in auto- and heterotrophic microbial activity (biomass production and respiration) within a CE that formed off Mauritania and along the ∼ 900 km zonal corridor between Mauritania and the Cape Verde islands in the Eastern Tropical North Atlantic (ETNA). Our results show how the physical disturbances caused by the CE affected the biomass distribution of phyto- and bacterioplankton and their metabolic activities. The injection of nutrients into the sunlit surface resulted in enhanced autotrophic pico- and nanoplankton abundance and generally increased autotrophic activity as indicated by chlorophyll a (Chl a) concentration, primary production (PP), and extracellular release rates. However, the detailed eddy survey also revealed an uneven distribution of these variables with, for example, the highest Chl a concentrations and PP rates occurring near and just beyond the CE's periphery. The heterotrophic bacterial activity was similarly variable. Optode-based community respiration (CR), bacterial respiration (BR) estimates, and bacterial biomass production (BP) largely followed the trends of PP and Chl a. Thus, a submesoscale spatial mosaic of heterotrophic bacterial abundance and activities occurred within the CE that was closely related to variability in autotrophic production. Consistent with this, we found a significant positive correlation between concentrations of semi-labile dissolved organic carbon (SL-DOC; here the sum of dissolved hydrolysable amino acids and dissolved combined carbohydrates) and BR estimates. Extracellular release of carbon as indicated by primary production of dissolved organic carbon (PPDOC) was variable with depth and laterally and not always sufficient to compensate the bacterial carbon demand (BCD: BR + BP), with PPDOC accounting for between 28 % and 110 % of the BCD. Bacterial growth efficiency (BGE: BP / BCD) ranged between 1.7 % and 18.2 %. We estimated the metabolic state to establish whether the CE was a source or a sink of organic carbon. We showed that the CE carried a strong autotrophic signal in the core (PP / CR > 1). Our results suggest that submesoscale (0–10 km) processes lead to highly variable metabolic activities in both photoautotrophic and heterotrophic microorganisms. Overall, we revealed that the CEs not only trap and transport coastal nutrients and organic carbon to the open ocean but also stimulate phytoplankton growth, generating freshly produced organic matter during westward propagation. This drives heterotrophic processes and may contribute to the previously observed net heterotrophy in open Atlantic surface waters.

Abstract

The science guiding the EUREC4A campaign and its measurements is presented. EUREC4A comprised roughly 5 weeks of measurements in the downstream winter trades of the North Atlantic - eastward and southeastward of Barbados. Through its ability to characterize processes operating across a wide range of scales, EUREC4A marked a turning point in our ability to observationally study factors influencing clouds in the trades, how they will respond to warming, and their link to other components of the earth system, such as upper-ocean processes or the life cycle of particulate matter. This characterization was made possible by thousands (2500) of sondes distributed to measure circulations on meso- (200 km) and larger (500 km) scales, roughly 400 h of flight time by four heavily instrumented research aircraft; four global-class research vessels; an advanced ground-based cloud observatory; scores of autonomous observing platforms operating in the upper ocean (nearly 10 000 profiles), lower atmosphere (continuous profiling), and along the air-sea interface; a network of water stable isotopologue measurements; targeted tasking of satellite remote sensing; and modeling with a new generation of weather and climate models. In addition to providing an outline of the novel measurements and their composition into a unified and coordinated campaign, the six distinct scientific facets that EUREC4A explored - from North Brazil Current rings to turbulence-induced clustering of cloud droplets and its influence on warm-rain formation - are presented along with an overview of EUREC4A's outreach activities, environmental impact, and guidelines for scientific practice. Abstract

Nitrification, the oxidation of ammonia via nitrite to nitrate, has always been considered to be a two-step process catalysed by chemolithoautotrophic microorganisms oxidizing either ammonia or nitrite. No known nitrifier carries out both steps, although complete nitrification should be energetically advantageous. This functional separation has puzzled microbiologists for a century. Here we report on the discovery and cultivation of a completely nitrifying bacterium from the genus Nitrospira, a globally distributed group of nitrite oxidizers. The genome of this chemolithoautotrophic organism encodes the pathways both for ammonia and nitrite oxidation, which are concomitantly expressed during growth by ammonia oxidation to nitrate. Genes affiliated with the phylogenetically distinct ammonia monooxygenase and hydroxylamine dehydrogenase genes of Nitrospira are present in many environments and were retrieved on Nitrospiracontigs in new metagenomes from engineered systems. Abstract

Mesoscale eddies play essential roles in modulating the ocean's physical, chemical, and biological properties. In cyclonic eddies (CE) nutrient upwelling can stimulate primary production by phytoplankton. Yet, how this locally enhanced autotrophic production affects heterotrophic bacterial activities (biomass production and respiration) and consequently the metabolic balance between the synthesis and the consumption of dissolved organic matter (DOM) remains largely unknown. To address this gap, we investigated the horizontal and vertical variability of phytoplankton and heterotrophic bacterial activity along ~900 km zonal corridor between the coast of Mauretania and the Cape Verde Islands in the eastern tropical North Atlantic (ETNA). We additionally collected samples from a CE along this transect at high spatial resolution. Our results show cascading effects of physical disturbances induced by a CE on phyto- and bacterioplankton biomass and metabolic activities. Specifically, the injection of nutrients into the sunlit surface resulted in enhanced autotrophic plankton abundance and activity as indicated by Chlorophyll a (Chl-a) concentration, DOM exudation, and primary productivity (PP). However, the detailed eddy survey revealed an uneven distribution of these parameters with, for example, the highest Chl-a concentrations and PP rates near and just beyond the CE’s periphery. The heterotrophic bacterial activity was similarly variable. Optode-based bacterial respiration (BR) and biomass production (BP) largely followed the trends of PP and Chl-a. Thus, a submesoscale spatial mosaic of heterotrophic bacterial abundance and activities occurred within the CE studied here that was closely related to variability in autotrophic production. Abstract

Mesoscale eddies modulate the ocean's physical, chemical, and biological properties. In cyclonic eddies (CEs), nutrient upwelling can stimulate primary production by phytoplankton. Yet, how this locally enhanced autotrophic production affects heterotrophy and consequently the metabolic balance between the synthesis and the consumption of dissolved organic matter (DOM) remains largely unknown. To fill this gap, we investigated the horizontal and vertical variability in auto- and heterotrophic microbial activity (biomass production and respiration) within a CE that formed off Mauritania and along the ∼ 900 km zonal corridor between Mauritania and the Cape Verde islands in the Eastern Tropical North Atlantic (ETNA). Our results show how the physical disturbances caused by the CE affected the biomass distribution of phyto- and bacterioplankton and their metabolic activities. The injection of nutrients into the sunlit surface resulted in enhanced autotrophic pico- and nanoplankton abundance and generally increased autotrophic activity as indicated by chlorophyll a (Chl a) concentration, primary production (PP), and extracellular release rates. However, the detailed eddy survey also revealed an uneven distribution of these variables with, for example, the highest Chl a concentrations and PP rates occurring near and just beyond the CE's periphery. The heterotrophic bacterial activity was similarly variable. Optode-based community respiration (CR), bacterial respiration (BR) estimates, and bacterial biomass production (BP) largely followed the trends of PP and Chl a. Thus, a submesoscale spatial mosaic of heterotrophic bacterial abundance and activities occurred within the CE that was closely related to variability in autotrophic production. Abstract

The Southwestern Tropical Pacific exhibits a complex bathymetry and represents a hot spot of internal tide generation. Based on a tailored high-resolution regional model, we investigate for the first time the internal tide field around the New Caledonia islands through energy budgets that quantify the internal tide generation, propagation, and dissipation. A total of 15.97 GW is converted from the barotropic to the baroclinic tide with the main conversion sites associated with the most prominent bathymetric structures such as continental slopes and narrow passages in the north (2.17 GW) and ridges and seamounts south of New Caledonia (3.92 GW). The bulk of baroclinic energy is generated in shallow waters around 500 m depth and on critical to supercritical slopes highlighting the limitations of linear semi-analytical models in those areas. Despite the strongly dominant mode-1 generation, more than 50 % of the locally generated energy dissipates in the near-field close to the generation sites. The remaining energy propagates within well-defined tidal beams with baroclinic energy fluxes of up to 30 kW m-1 toward the open ocean, strongly dominated by mode-1. The energetic mesoscale eddy activity in the region appears to be the main source of tidal incoherence. Locally, mesoscale eddy-driven stratification changes induce variations of the conversion term. In the far-field, incoherence of the energy flux arises through the interaction of the tidal beam with the eddying background flow. The New Caledonia site represents a challenge for SWOT (Surface Water Ocean Topography) observability of meso- and submesoscale dynamics in the presence of internal tides with sea surface height signatures > 6 cm. We show that a correction of the coherent baroclinic tide may improve the observability range by shifting the transition scale between balanced and unbalanced flow in winter from 180 km to 50-80 km. In contrast, in summer observability increases only marginally due to the seasonally amplified signature of the incoherent tide at scales below 100 km. [ABSTRACT FROM AUTHOR]

Bacterial growth efficiency (BP/(BR+BP)) was variable (1.4–10.5 %) within the CE and carbon exudation was not always sufficient to compensate the bacterial carbon demand (BR+BP; 28.3–114.5 %). We have additionally estimated the metabolic state in our samples, which showed that the CE carried a strong autotrophic signal (PP/(BR+BP) > 1). Overall, our results show that submesoscale (0–10 km) processes lead to highly variable metabolic activities of both phototrophic and heterotrophic microbes, which has implications for biogeochemical models estimating oceanic carbon fluxes. Additionally, we revealed that the CE not only traps and transports coastal nutrients and carbon to the open ocean but also stimulates phytoplankton growth generating freshly produced organic matter during westward propagation. This organic matter may fuel heterotrophic processes in the open ocean and may help to explain the often-observed net heterotrophic metabolic state of these environments.

Diagnostic injections (blocks) are a valuable tool in the management of chronic noncancer pain. By precise blockade of specific neural structures and observation of pain responses, pain mechanisms can be accurately defined. With such information, therapeutic procedures targeting neural structures are possible. Fibromyalgia is a disorder of pain processing with characteristic symptoms. The 2010 American College of Rheumatologists fibromyalgia diagnostic criteria evaluates these symptoms in a scoring system, allowing more objectivity in the diagnosis. We hypothesize that patients with fibromyalgia phenotype fulfilling the 2010 American College of Rheumatologists criteria may respond to diagnostic blocks differently when compared to patients without fibromyalgia phenotype.This study was designed to establish whether diagnosis or suspected diagnosis of fibromyalgia should influence the decision to perform diagnostic blocks for chronic non-cancer pain.A prospective observational research study was performed at our institution. IRAS project ID: 231514.Tertiary pain clinic in the UK.Patients were selected to receive diagnostic block by usual clinical assessment after which they were asked to consent to take part in the study. All participating patients completed the 2010 American College of Rheumatologists fibromyalgia diagnostic questionnaire prior to the diagnostic block. Patients were divided into 2 groups A and B based on the outcome of block - primary outcome. Group A experienced a 70% or greater improvement in pain severity following the block for the anticipated duration of action of the local anesthetic, Group B experienced a less than 70% reduction in pain. Statistical analysis between groups A and B was conducted by comparing categorical data, described as percentages, with the chi squared test. Ordinal variables such as Widespread pain index and Symptom severity score are presented as median and analyzed with Mann-Whitney test.Seventy-seven patients were included in the study. Two patients were lost to follow-up. Of the 75 remaining patients, 44 received lumbar medial branch blocks, 19 genicular nerve blocks, 3 blocks to nerves supplying the sacroiliac joint, one suprascapular nerve block, and 6 cervical and 2 thoracic medial branch blocks. Group A contained 38 patients and group B contained 37 patients. There was no statistically significant difference in the prevalence of fibromyalgia screening questionnaire positive patients between groups A (13 out of 38 patients) and B (13 out of 37 patients), P = 0.93. There was no statistically significant difference in the prevalence of fibromyalgia screening questionnaire positive patients in subgroups undergoing the same type of diagnostic block (spinal pain and knee pain).Selection of patients prior to inclusion in the study may introduce bias. Patients were selected by individual treating clinicians using usual clinical practice; however, the exact selection criteria were not standardized.We conclude that after physician selection, the presence of fibromyalgia phenotype does not influence the outcome from diagnostic block. It is likely therefore that fibromyalgia phenotype should not influence the decision to perform diagnostic blocks if indicated based on assessment by an experienced pain physician.

Abstract

In this study, we investigate the mesoscale flow field and how it enables energy to propagate vertically in form of near-inertial waves. As part of the EURAC4A-OA campaign the research vessels RV Maria S. Merian and NO L’Atalante simultaneously surveyed mesoscale eddy fronts in the western tropical North Atlantic. From velocity profile data, measured by a shipboard Acoustic Doppler Current Profiler (sADCP), we reconstruct eddies in the upper 1000m of the surveyed area, by fitting a Rankine Vortex model. The model derives an idealized velocity structure of the eddy as well as the location of its centre. Multiple occurrences of stacked eddies are identified and often surrounded by current shear structures associated with near-inertial waves. Using data from ship sections, where both research vessels operated less than 1nm apart, the vertical component of the relative vorticity (zeta) is calculated using different methods (single ship, two ships)[Shcherbina et al. 2013]. It is found that in particular zeta outside of the eddy cores is sensitive to the way the vorticity is calculated and may even change sign. Furthermore, the resulting zeta sections and its impact on the ability of near-inertial waves propagating vertically below the mixed layer is discussed.

Abstract

The science guiding the EUREC4A campaign and its measurements is presented. EUREC4A comprised roughly 5 weeks of measurements in the downstream winter trades of the North Atlantic – eastward and southeastward of Barbados. Through its ability to characterize processes operating across a wide range of scales, EUREC4A marked a turning point in our ability to observationally study factors influencing clouds in the trades, how they will respond to warming, and their link to other components of the earth system, such as upper-ocean processes or the life cycle of particulate matter. This characterization was made possible by thousands (2500) of sondes distributed to measure circulations on meso- (200 km) and larger (500 km) scales, roughly 400 h of flight time by four heavily instrumented research aircraft; four global-class research vessels; an advanced ground-based cloud observatory; scores of autonomous observing platforms operating in the upper ocean (nearly 10 000 profiles), lower atmosphere (continuous profiling), and along the air–sea interface; a network of water stable isotopologue measurements; targeted tasking of satellite remote sensing; and modeling with a new generation of weather and climate models. In addition to providing an outline of the novel measurements and their composition into a unified and coordinated campaign, the six distinct scientific facets that EUREC4A explored – from North Brazil Current rings to turbulence-induced clustering of cloud droplets and its influence on warm-rain formation – are presented along with an overview of EUREC4A's outreach activities, environmental impact, and guidelines for scientific practice. Abstract

During the collaborative project "Role of Eddies in the Carbon Pump of Eastern Boundary Upwelling Systems" (REEBUS), that took place in the eastern tropical North Atlantic in 2019, three cyclonic mesoscale eddies were intensely surveyed by ship and autonomous systems. The three eddies were located at different distances to the coast, the most intense of them (vorticity about 0.5 times f) was found in lee of the Cabo Verdian island of Fogo. Here we present the reconstruction of the 3-D structure for the three eddies from ship ADCP and hydrographic sections. Divergence estimates suggest the existence of a downwelling cell in the center of all three eddies. This cell extends from below the thermocline down to some hundred meters, at a diameter of about 10 nautical miles. Surface signatures of the eddies indicate elliptic and oscillating behavior which is further investigated using the interior ocean data. In this work we also explore the limitations of section-based ocean mesoscale eddy reconstructions.
