6. How do eyespot resistance genes transferred into winter wheat breeding lines affect their yield?

Abstract

Eyespot can reduce yields, even up to 50%. There are four genetically characterized resistances in wheat varieties, controlled by: (1) the Pch1 gene, transferred from Aegilops ventricosa; (2) the Pch2 gene, originating from wheat variety Capelle Desprez; (3) the Pch3 gene, originating from Dasypyrum villosum; and (4) the Q.Pch.jic-5A gene, a quantitative trait locus (QTL) located on chromosome 5A of Capelle Desprez. However, those loci have drawbacks, such as linkage of Pch1 with deleterious traits and limited effectiveness of Pch2 against the disease. Here we present an initial study which aims to characterize wheat pre-registration breeding lines carrying 12 eyespot resistance genes, consider their resistance expression in inoculation tests and the influence of resistance genotypes on the yield. We selected four groups of breeding lines, carrying: (1) the Pch1 gene alone: one line; (2) the Pch2 gene alone: four lines; (3) the Q.Pch.jic-5A gene alone: one line; and (4) Pch1 + Q.Pch.jic-5A: three lines. For the first time, the effect of the combination of Pch1 and Q.Pch.jic-5A genes was compared with resistance conferred by Pch1 or Q.Pch.jic-5A alone. We found significant differences between infection scores evaluated in resistant lines carrying Pch1 and Q.Pch.jic-5A alone, while no differences in terms of the level of resistance expression were detected between Pch1 alone and Pch1 + Q.Pch.jic-5A, and between wheat lines carrying Pch1 and Pch2 alone. Moreover, we demonstrated that the Pch1 gene, together with an Ae. ventricosa segment, caused statistically significant yield losses, both as a single eyespot resistance source or in a combination with Q.Pch.jic-5A. Yield scores showed that wheat lines with Q.Pch.jic-5A had the highest yields, similar to the yielding potential of Pch2-bearing lines and control varieties.

19. Molecular identification of triticale introgression lines carrying leaf rust resistance genes transferred from Aegilops kotschyiBoiss. and Ae. tauschiiCoss

Abstract

Triticale (× TriticosecaleWittmack) is a commercial hybrid harboring wheat (Triticumsp.) and rye (Secale cerealeL.) genomes. The limited genetic diversity of this crop resulted in the collapse of fungal disease resistance. Leaf rust disease, caused by Puccinia triticinaEriks., is reported to reduce the triticale yield significantly (more than 30%). There is a need to enlarge the genetic variability of this crop including leaf resistance genes. The main aim of this research was to evaluate the leaf rust resistance of the offspring of translocation lines of triticale carrying chromatin of Ae. tauschiiand Ae. kotschyi. A reaction of seedlings of 200 plants of two triticale-Aegilopstranslocation lines (Bogo-2Dt.2R and Sekundo-2Sk.2R) was compared after inoculation with a natural mixture of P. triticinaraces, specific to triticale in controlled condition. Before inoculation, each plant was screened using molecular cytogenetics and molecular markers linked to leaf rust resistance genes. The presence of Aegilopschromosome segments was confirmed using genomic in situ hybridization (GISH). Lr39and Lr54leaf rust resistance genes were identified using Xgdm35and S14molecular markers, respectively. After inoculation, a significant improvement of resistance severity was observed in Sekundo-2Sk.2R in comparison with triticale cv. Sekundo plants. The resistance level of Bogo-2Dt.2R did not differ compared with triticale cv. Bogo plants. It was shown that Lr39gene did not increase the leaf rust resistance level of triticale cv. Bogo.

Abstract

Alien chromosome introgression has become a valuable tool to broaden the genetic variability of crop plants via chromosome engineering. This study details the procedure to obtain monosomic addition and monosomic substitution lines of the triticale carrying 2Sk chromosome from Aegilops kotchyi Boiss., which harbors Lr54 + Yr37 leaf and stripe rust-resistant gene loci, respectively. Initially, A. kotschyi × Secale cereale artificial amphiploids (2 n = 6 x = 42 chromosomes, UUSSRR) were crossed with triticale cv. "Sekundo" (2 n = 6 x = 42, AABBRR) in order to obtain fertile offspring. Cyto-molecular analyses of five subsequent backcrossing generations revealed that 2Sk chromosome was preferentially transmitted. This allowed for the selection of monosomic 2Sk addition (MA2Sk) lines of triticale. Finally, the 2Sk(2R) substitution plants were obtained by crossing MA2Sk with the nullisomic (N2R) plants of triticale. The presence of 2Sk chromosome in subsequent generations of plants was evaluated using SSR markers linked to Lr54 + Yr37 loci. Disease evaluation of the monosomic 2Sk(2R) substitution plants for the reaction to leaf and stripe rust infection were carried out under controlled conditions in a growth chamber. The results showed significant improvement of leaf rust resistance severity of monosomic substitution plants compared with control ("Sekundo"). In contrast, the introgression of the Lr54 + Yr37 loci did not lead to improvement of stripe rust resistance. In summary, the creation of monosomic addition and monosomic substitution lines of triticale is the starting point for the precise and guided transfer of Lr54 + Yr37 loci. 35. Toxigenic Fusarium species infecting wheat heads in Poland.

Abstract

Toxigenic Fusarium species are common pathogens of wheat and other cereals worldwide. In total, 449 wheat heads from six localities in Poland, heavily infected with Fusarium during 2009 season, were examined for Fusarium species identification. F. culmorum was the most common species (72.1% on average) with F. graminearum and F. avenaceum the next most commonly observed, but much less frequent (13.4 and 12.5% respectively). F. cerealis was found in 1.8% of all samples, and F. tricinctum was found only in one sample (0.2%). Subsequent quantification of the three major mycotoxins (deoxynivalenol, zearalenone and moniliformin) in grain and chaff fractions with respect to associated prevailing pathogen species uncovered the following patterns. Moniliformin (MON) was found in low amounts in all samples with F. avenaceum present. In contrast, deoxynivalenol (DON) and zearalenone (ZEA) were the contaminants of F. culmorum- and F. graminearum-infected heads. The highest concentration of DON was recorded in grain sample collected in Radzików (77 µg g −1). High temperatures in Central Poland during July and August accompanied with high rainfall in July were responsible for this high DON accumulation. Abstract

This paper shows the results of effective uses of a molecular cytogenetics toolbox and molecular marker to transfer leaf rust resistance genes from Aegilops tauschii× Secale cereale(DDRR, 2n = 4x = 28) amphiploid forms to triticale cv. Bogo (AABBRR, 2n = 6x = 42). The molecular markers of resistance genes and in situ hybridization analysis of mitotic metaphase of root meristems confirmed the stable inheritance of chromosome 3D segments carrying Lr32from the BC2F2to the BC2F5generation of (Ae. tauschii× S. cereale) × triticale hybrids. The chromosome pairing analysis during metaphase I of meiosis of BC2F4and BC2F5hybrids showed increasing regular bivalent formation of 3D chromosome pairs and decreasing number of univalents in subsequent generations. The results indicate that using amphiploid forms as a bridge between wild and cultivated forms can be a successful technology to transfer the D-genome chromatin carrying leaf rust resistance genes into triticale.

Abstract

The aim of this study was to search the breeding collection of hexaploid wheat in terms of eyespot resistance identification. Eyespot is one of the common fungal stem-base disease of wheat (Triticum aestivum L.) and other cereals in temperate regions. The causal agents of eyespot are Oculimacula acuformis (syn. Tapesia acuformis; anamorph: Helgardia acuformis syn. Pseudocercosporella herpotrichoides var. acuformis) and O. yallundae (syn. T. yallundae; anamorph: H. yallundae syn. P. h. var. herpotrichoides). Eye-shaped, elliptic lesions on the lower portion are the symptoms of this disease, which can weaken stem bases and cause them to bend or break. Crops from infected plants are slighter with reduced quality and yield of even up to 50%. There are three eyespot resistance genes (Pch1, Pch2 and QTL on chromosome 5A), which have been characterized in hybrid forms of wheat. Pch1 located on chromosome 7DL is the most important and most effective resistance gene and was introduced into wheat from the wild grass Aegilops ventricosa. However, Pch1 locus is associated with lower yield in the absence of the disease, which is a limiting factor considering elite varieties breeding. On the other side, both Pch2 and Pch-QTL-5A originate from the French wheat cultivar Cappelle Desprez, but confer only a moderate level of resistance and are unlikely to prevent yield loss under high disease pressure. The main aims of this study were (1) to screen the collection of 150 winter wheat breeding lines in order to identify genotypes with Pch1, Pch2 and Q.Pch.jic-5A loci, (2) to analyze the resistance expression in those lines using inoculation tests at BBCH10-12 (seedling) and BBCH31-32 growth stage and (3) to compare the yield components with the results of markers identification and inoculation tests in order to evaluate the influence of the genotype on technological values of breeding lines. We have identified 7 genotypes carrying cumulated two or three Pch genes. The results of inoculation tests confirm the resistance expression in those genotypes. [ABSTRACT FROM AUTHOR]

38. Development and Cytomolecular Identification of Monosomic Alien Addition and Substitution Lines of Triticale (× Triticosecale Wittmack) With 2S k Chromosome Conferring Leaf Rust Resistance Derived From Aegilops kotschyi Boiss.

Abstract

Alien chromosome introgression has become a valuable tool to broaden the genetic variability of crop plants via chromosome engineering. This study details the procedure to obtain monosomic addition and monosomic substitution lines of the triticale carrying 2S k chromosome from Aegilops kotchyi Boiss., which harbors Lr54 + Yr37 leaf and stripe rust-resistant gene loci, respectively. Initially, A. kotschyi × Secale cereale artificial amphiploids (2 n = 6 x = 42 chromosomes, UUSSRR) were crossed with triticale cv. "Sekundo" (2 n = 6 x = 42, AABBRR) in order to obtain fertile offspring. Cyto-molecular analyses of five subsequent backcrossing generations revealed that 2S k chromosome was preferentially transmitted. This allowed for the selection of monosomic 2S k addition (MA2S k ) lines of triticale. Finally, the 2S k (2R) substitution plants were obtained by crossing MA2S k with the nullisomic (N2R) plants of triticale. The presence of 2S k chromosome in subsequent generations of plants was evaluated using SSR markers linked to Lr54 + Yr37 loci. Disease evaluation of the monosomic 2S k (2R) substitution plants for the reaction to leaf and stripe rust infection were carried out under controlled conditions in a growth chamber. The results showed significant improvement of leaf rust resistance severity of monosomic substitution plants compared with control ("Sekundo"). In contrast, the introgression of the Lr54 + Yr37 loci did not lead to improvement of stripe rust resistance. In summary, the creation of monosomic addition and monosomic substitution lines of triticale is the starting point for the precise and guided transfer of Lr54 + Yr37 loci.
