Abstract

* Context.--In 2014, the College of American Pathologists developed an evidence-based guideline to address analytic validation of immunohistochemical assays. Fourteen recommendations were offered. Per the National Academy of Medicine standards for developing trustworthy guidelines, guidelines should be updated when new evidence suggests modifications. Objective.--To assess evidence published since the release of the original guideline and develop updated evidence-based recommendations. Design.--The College of American Pathologists convened an expert panel to perform a systematic review of the literature and update the original guideline recommendations using the Grading of Recommendations Assessment, Development and Evaluation approach. Results.--Two strong recommendations, 1 conditional recommendation, and 12 good practice statements are offered in this updated guideline. They address analytic validation or verification of predictive and nonpredictive assays, and recommended revalidation procedures following changes in assay conditions. Conclusions.--While many of the original guideline statements remain similar, new recommendations address analytic validation of assays with distinct scoring systems, such as programmed death receptor-1 and analytic verification of US Food and Drug Administration approved/ cleared assays; more specific guidance is offered for validating immunohistochemistry performed on cytology specimens. Abstract

High-grade neuroendocrine neoplasms are a rare disease entity and account for approximately 10% of all neuroendocrine neoplasms. Because of their rarity, there is an overall lack of prospectively collected data available to advise practitioners as to how best to manage these patients. As a result, best practices are largely based on expert opinion. Recently, a distinction was made between well-differentiated high-grade (G3) neuroendocrine tumors and poorly differentiated neuroendocrine carcinomas, and with this, pathologic details, appropriate imaging practices and treatment have become more complex. In an effort to provide practitioners with the best guidance for the management of patients with high-grade neuroendocrine neoplasms of the gastrointestinal tract, pancreas, and gynecologic system, the North American Neuroendocrine Tumor Society convened a panel of experts to develop a set of recommendations and a treatment algorithm that may be used by practitioners for the care of these patients. Here, we provide consensus recommendations from the panel on pathology, imaging practices, management of localized disease, management of metastatic disease and surveillance and draw key distinctions as to the approach that should be utilized in patients with well-differentiated G3 neuroendocrine tumors vs poorly differentiated neuroendocrine carcinomas.

Abstract

Abstract Earthquakes pose substantial threats to communities worldwide. Understanding how people respond to the fast-changing environment during earthquakes is crucial for reducing risks and saving lives. This study aims to study people’s protective action decision-making in earthquakes by leveraging explainable machine learning and video data. Specifically, this study first collected real-world CCTV footage and video postings from social media platforms, and then identified and annotated changes in the environment and people’s behavioral responses during the M7.1 2018 Anchorage earthquake. By using the fully annotated video data, we applied XGBoost, a widely-used machine learning method, to model and forecast people’s protective actions (e.g., drop and cover, hold on, and evacuate) during the earthquake. Then, explainable machine learning techniques were used to reveal the complex, nonlinear relationships between different factors and people’s choices of protective actions. Modeling results confirm that social and environmental cues played critical roles in affecting the probability of different protective actions. Certain factors, such as the earthquake shaking intensity and number of people shown in the environment, displayed evident nonlinear relationships with the probability of choosing to evacuate. These findings can help emergency managers and policymakers design more effective protective action recommendations during earthquakes.

Abstract

Classical novae are cataclysmic binary star systems in which the matter of a companion star is accreted on a white dwarf (WD). Accumulation of hydrogen in a layer eventually causes a thermonuclear explosion on the surface of the WD, brightening the WD to ~10^5 solar luminosities and triggering ejection of the accumulated matter.They provide extreme conditions required to accelerate particles, electrons or protons, to high energies. Here we present the detection of gamma rays by the MAGIC telescopes from the 2021 outburst of RS Ophiuchi (RS Oph), a recurrent nova with a red giant (RG) companion, that allowed us, for the first time, to accurately characterize the emission from a nova in the 60 GeV to 250 GeV energy range. The theoretical interpretation of the combined Fermi-LAT and MAGIC data suggests that protons are accelerated to hundreds of GeV in the nova shock. Abstract

Context.--Therapy targeted at human epidermal growth factor receptor 2 (HER2; also known as ERBB2) was used initially for breast and gastroesophageal carcinoma and has more recently been adopted for endometrial serous carcinoma (ESC) and colorectal carcinoma (CRC). There is evidence that predictive biomarker testing algorithms for HER2 must be tumor type specific and that an algorithm validated for one tumor type cannot be applied to another. Objective.--To describe current laboratory practices for HER2 assessment in ESC and CRC. Design.--We surveyed laboratories participating in the 2021 College of American Pathologists (CAP) HER2 immunohistochemistry proficiency testing program. Results.--The survey was distributed to 1548 laboratories and returned by 1195, of which 83.5% (998) were in the United States. For ESC, 24.0% (287) of laboratories reported performing in-house testing for HER2 by immunohistochemical staining and/or in situ hybridization; of these, 44.3% (127) performed it reflexively on all cases of ESC. The most common criterion for evaluating HER2 was the American Society of Clinical Oncology/CAP 2018 guideline for breast carcinoma (69.0%; 194 of 281), whereas only 16.0% (45) of laboratories used guidelines specific to ESC. For CRC, 20.2% (239 of 1185) of laboratories performed in-house HER2 testing, and 82.0% of these (196) did the test only at the clinician's request. A plurality (49.4%; 115 of 233) used gastroesophageal cancer guidelines when scoring CRC, 30.0% (70) used the CRC scoring system from the HERACLES trial, and 16.3% (38) used the American Society of Clinical Oncology/CAP 2018 guideline for breast carcinoma. Conclusions.--Laboratories vary in their approach to HER2 testing in ESC and CRC. Most laboratories did not report using tumor type-specific recommendations for HER2 interpretation. Abstract

Abstract Background Coronavirus disease COVID-19 is a heterogeneous condition caused by SARS-CoV-2 infection. Generally, it is characterized by interstitial pneumonia that can lead to impaired gas-exchange, acute respiratory failure, and death, although a complex disorder of multi-organ dysfunction has also been described. The pathogenesis is complex, and a variable combination of factors has been described in critically ill patients. COVID-19 is a particular risk for older persons, particularly those with frailty and comorbidities. Blood bacterial DNA has been reported in both physiological and pathological conditions and has been associated with some haematological and laboratory parameters but, to date, no study has characterized it in hospitalized old COVID-19 patients The present study aimed to establish an association between blood bacterial DNA (BB-DNA) and clinical severity in old COVID-19 patients. Results BB-DNA levels were determined, by quantitative real-time PCRs targeting the 16S rRNA gene, in 149 hospitalized older patients (age range 65–99 years) with COVID-19. Clinical data, including symptoms and signs of infection, frailty status, and comorbidities, were assessed. BB-DNA was increased in deceased patients compared to discharged ones, and Cox regression analysis confirmed an association between BB-DNA and in-hospital mortality. Furthermore, BB-DNA was positively associated with the neutrophil count and negatively associated with plasma IFN-alpha. Additionally, BB-DNA was associated with diabetes. Conclusions The association of BB-DNA with mortality, immune-inflammatory parameters and diabetes in hospitalized COVID-19 patients suggests its potential role as a biomarker of unfavourable outcomes of the disease, thus it could be proposed as a novel prognostic marker in the assessment of acute COVID-19 disease.

Abstract

Milky Way dwarf spheroidal galaxies (dSphs) are among the best candidates to search for signals of dark matter annihilation with Imaging Atmospheric Cherenkov Telescopes, given their high mass-to-light ratios and the fact that they are free of astrophysical gamma-ray emitting sources. Since 2011, MAGIC has performed a multi-year observation program in search for Weakly Interacting Massive Particles (WIMPs) in dSphs. Results on the observations of Segue 1 and Ursa Major II dSphs have already been published and include some of the most stringent upper limits (ULs) on the velocity-averaged cross-section $\langle \sigma_{\mathrm{ann}} v \rangle$ of WIMP annihilation from observations of dSphs. In this work, we report on the analyses of 52.1 h of data of Draco dSph and 49.5 h of Coma Berenices dSph observed with the MAGIC telescopes in 2018 and in 2019 respectively. No hint of a signal has been detected from either of these targets and new constraints on the $\langle \sigma_{\mathrm{ann}} v \rangle$ of WIMP candidates have been derived. In order to improve the sensitivity of the search and reduce the effect of the systematic uncertainties due to the $J$-factor estimates, we have combined the data of all dSphs observed with the MAGIC telescopes. Using 354.3 h of dSphs good quality data, 95 % CL ULs on $\langle \sigma_{\mathrm{ann}} v \rangle$ have been obtained for 9 annihilation channels. For most of the channels, these results reach values of the order of $10^{-24} $cm$^3$/s at ${\sim}1$ TeV and are the most stringent limits obtained with the MAGIC telescopes so far., Comment: 17 pages, 5 figures

Abstract

We report multiwavelength observations of the gravitationally lensed blazar QSO B0218+357 in 2016-2020. Optical, X-ray and GeV flares were detected. The contemporaneous MAGIC observations do not show significant very-high-energy (VHE, >= 100 GeV) gamma-ray emission. The lack of enhancement in radio emission measured by OVRO indicates the multi-zone nature of the emission from this object. We constrain the VHE duty cycle of the source to be < 16 2014-like flares per year (95% confidence). For the first time for this source, a broadband low-state SED is constructed with a deep exposure up to the VHE range. A flux upper limit on the low-state VHE gamma-ray emission of an order of magnitude below that of the 2014 flare is determined. The X-ray data are used to fit the column density of (8.10 +- 0.93 stat ) x 10^21 cm^-2 of the dust in the lensing galaxy. VLBI observations show a clear radio core and jet components in both lensed images, yet no significant movement of the components is seen. The radio measurements are used to model the source-lens-observer geometry and determine the magnifications and time delays for both components. The quiescent emission is modeled with the high-energy bump explained as a combination of synchrotron-self-Compton and external Compton emission from a region located outside of the broad line region. Abstract

We present a set of results obtained with an innovative eye-tracker based on magnetic dipole localization by means of an array of magnetoresistive sensors. The system tracks both head and eye movements with a high rate (100-200 Sa/s) and in real time. A simple setup is arranged to simulate head and eye motions and to test the tracker performance under realistic conditions. Multimedia material is provided to substantiate and exemplify the results. Abstract

Vaccine effectiveness (VE) studies provide real-world evidence to monitor vaccine performance and inform policy. The WHO Regional Office for the Eastern Mediterranean supported a regional study to assess the VE of COVID-19 vaccines against different clinical outcomes in four countries between June 2021 and August 2023. Health worker cohort studies were conducted in 2707 health workers in Egypt and Pakistan, of whom 171 experienced symptomatic laboratory-confirmed SARS-CoV-2 infection. Test-negative design case–control studies were conducted in Iran and Jordan in 4017 severe acute respiratory infection (SARI) patients (2347 controls and 1670 cases) during the Omicron variant dominant period. VE estimates were calculated for each study and pooled by study design for several vaccine types (BBIBP-CorV, AZD1222, BNT162b2, and mRNA-1273, among others). Among health workers, VE against symptomatic infection of a complete primary series could only be computed compared to partial vaccination, suggesting a benefit of providing an additional dose of mRNA vaccines (VE: 88.9%, 95%CI: 15.3–98.6%), while results were inconclusive for other vaccine products. Among SARI patients, VE against hospitalization of a complete primary series with any vaccine compared to non-vaccinated was 20.9% (95%CI: 4.5–34.5%). Effectiveness estimates for individual vaccines, booster doses, and secondary outcomes (intensive care unit admission and death) were inconclusive. Future VE studies will need to address challenges in both design and analysis when conducted late during a pandemic and will be able to utilize the strengthened capacities in countries.

Abstract

The results of gamma-ray observations of the binary system HESS J0632+057 collected during 450 hours over 15 years, between 2004 and 2019, are presented. Data taken with the atmospheric Cherenkov telescopes H.E.S.S., MAGIC, and VERITAS at energies above 350 GeV were used together with observations at X-ray energies obtained with Swift-XRT, Chandra, XMM-Newton, NuSTAR, and Suzaku. Some of these observations were accompanied by measurements of the H{\alpha} emission line. A significant detection of the modulation of the VHE gamma-ray fluxes with a period of 316.7+-4.4 days is reported, consistent with the period of 317.3+-0.7 days obtained with a refined analysis of X-ray data. The analysis of data of four orbital cycles with dense observational coverage reveals short timescale variability, with flux-decay timescales of less than 20 days at very high energies. Flux variations observed over the time scale of several years indicate orbit-to-orbit variability. The analysis confirms the previously reported correlation of X-ray and gamma-ray emission from the system at very high significance, but can not find any correlation of optical H{\alpha} parameters with X-ray or gamma-ray energy fluxes in simultaneous observations. The key finding is that the emission of HESS J0632+057 in the X-ray and gamma-ray energy bands is highly variable on different time scales. The ratio of gamma-ray to X-ray flux shows the equality or even dominance of the gamma-ray energy range. This wealth of new data is interpreted taking into account the insufficient knowledge of the ephemeris of the system, and discussed in the context of results reported on other gamma-ray binary systems., Comment: Accepted for publication by The Astrophysical Journal

Abstract

PSR J0218+4232 is one of the most energetic millisecond pulsars known and has long been considered as one of the best candidates for very high-energy (VHE; >100 GeV) gamma-ray emission. Using 11.5 years of Fermi Large Area Telescope (LAT) data between 100 MeV and 870 GeV, and ~90 hours of MAGIC observations in the 20 GeV to 20 TeV range, we have searched for the highest energy gamma-ray emission from PSR J0218+4232. Based on the analysis of the LAT data, we find evidence for pulsed emission above 25 GeV, but see no evidence for emission above 100 GeV (VHE) with MAGIC. We present the results of searches for gamma-ray emission, along with theoretical modeling, to interpret the lack of VHE emission. We conclude that, based on the experimental observations and theoretical modeling, it will remain extremely challenging to detect VHE emission from PSR J0218+4232 with the current generation of Imaging Atmospheric Cherenkov Telescopes (IACTs), and maybe even with future ones, such as the Cherenkov Telescope Array (CTA)., Comment: 19 pages, 9 figures, Accepted to ApJ

Abstract

We report here on the first multi-wavelength (MWL) campaign on the blazar TXS 1515-273, undertaken in 2019 and extending from radio to very-high-energy gamma rays (VHE). Up until now, this blazar had not been the subject of any detailed MWL observations. It has a rather hard photon index at GeV energies and was considered a candidate extreme high-synchrotronpeaked source. MAGIC observations resulted in the first-time detection of the source in VHE with a statistical significance of 7.6$\sigma$. The average integral VHE flux of the source is 6 $\pm$ 1% of the Crab nebula flux above 400 GeV. X-ray coverage was provided by Swift-XRT, XMMNewton, and NuSTAR. The long continuous X-ray observations were separated by $\sim$ 9 h, both showing clear hour scale flares. In the XMM-Newton data, both the rise and decay timescales are longer in the soft X-ray than in the hard X-ray band, indicating the presence of a particle cooling regime. The X-ray variability timescales were used to constrain the size of the emission region and the strength of the magnetic field. The data allowed us to determine the synchrotron peak frequency and classify the source as a flaring high, but not extreme, synchrotron peaked object. Considering the constraints and variability patterns from the X-ray data, we model the broad-band spectral energy distribution. We applied a simple one-zone model, which could not reproduce the radio emission and the shape of the optical emission, and a two-component leptonic model with two interacting components, enabling us to reproduce the emission from radio to VHE band.

Abstract

We present a detailed characterisation and theoretical interpretation of the broadband emission of the paradigmatic TeV blazar Mrk 421, with special focus on the multi-band flux correlations. The dataset has been collected through an extensive multiwavelength campaign organised between 2016 December and 2017 June. The instruments involved are MAGIC, FACT, Fermi-LAT, Swift, GASP-WEBT, OVRO, Medicina and Mets\"ahovi. Additionally, four deep exposures (several hours long) with simultaneous MAGIC and NuSTAR observations allowed a precise measurement of the falling segments of the two spectral components. The very-high-energy (VHE; E > 100 GeV) gamma rays and X-rays are positively correlated at zero time lag, but the strength and characteristics of the correlation change substantially across the various energy bands probed. The VHE versus X-ray fluxes follow different patterns, partly due to substantial changes in the Compton dominance during a few days without a simultaneous increase in the X-ray flux (i.e. orphan gamma-ray activity). Studying the broadband spectral energy distribution (SED) during the days including NuSTAR observations, we show that these changes can be explained within a one-zone leptonic model with a blob that increases its size over time. Our multi-band correlation study also hints at an anti-correlation between UV/optical and X-ray at a significance higher than 3 sigmas. A VHE flare observed on 2017 February 4 shows gamma-ray variability on multi-hour timescales, with a factor 10 increase in the TeV flux but only a moderate increase in the keV flux. The related broadband SED is better described by a two-zone leptonic scenario rather than by a one-zone scenario. Abstract

Digital investigations of stealthy attacks on Android devices pose particular challenges to incident responders. Whereas consequential late detection demands accurate and comprehensive forensic timelines to reconstruct all malicious activities, reduced forensic footprints with minimal malware involvement, such as when Living-Off-the-Land (LOtL) tactics are adopted, leave investigators little evidence to work with. Volatile memory forensics can be an effective approach since app execution of any form is always bound to leave a trail of evidence in memory, even if perhaps ephemeral. Just-in-Time Memory Forensics (JIT-MF) is a recently proposed technique that describes a framework to process memory forensics on existing stock Android devices, without compromising their security by requiring them to be rooted. Within this framework, JIT-MF drivers are designed to promptly dump in-memory evidence related to app usage or misuse. In this work, we primarily introduce a conceptualized presentation of JIT-MF drivers. Abstract

The Mediterranean island of Sardinia shows the highest suicide rate in Italy among men (23.07 x 10.sup.5 per year vs the average national rate of 13.80 x 10.sup.5 per year). In contrast, the suicide rate among Sardinian women is one of the lowest across all the Italian regions. The aim of this epidemiological study was to investigate the distribution of suicide mortality in Sardinia in relation with environmental and social factors using geospatial analysis. Data available from the total 377 Sardinian municipalities, including suicidal mortality and several socio-economic indicators, were retrieved and analysed. Ordinary least squares regression was used to investigate the association between suicide mortality and the selected indicators. Suicides among men reached a peak in the Southern-Eastern area of the island. Overall, the multivariable analysis showed that suicidal mortality was significantly and positively related with the altitude (0.243, P = 0.001), the occupational hardship index (0.230, P = 0.016), and the social services hardship index (0.149, P < 0.0001), and negatively with pro-capita income (â0.122, P = 0.012). After adjusting for all covariates, the index of multiple deprivation, the environmental and the educational index respectively, remained the only significant predictors. Abstract

We engaged in a participatory modeling approach with health sector stakeholders in Jordan to support government decision-making regarding implementing public health measures to mitigate COVID-19 disease burden. We considered the effect of 4 physical distancing strategies on reducing COVID-19 transmission and mortality in Jordan during March 2020–January 2021: no physical distancing; intermittent physical distancing where all but essential services are closed once a week; intermittent physical distancing where all but essential services are closed twice a week; and a permanent physical distancing intervention. Modeling showed that the fourth strategy would be most effective in reducing cases and deaths; however, this approach was only marginally beneficial to reducing COVID-19 disease compared with an intermittently enforced physical distancing intervention. Scenario-based model influenced policy-making and the evolution of the pandemic in Jordan confirmed the forecasting provided by the modeling exercise and helped confirm the effectiveness of the policy adopted by the government of Jordan.

Abstract

With the widespread phenomenon of antibiotic resistance and the diffusion of multiple drug-resistant bacterial strains, enormous efforts are being conducted to identify suitable alternative agents against pathogenic microorganisms. Since an association between biofilm formation and antibiotic resistance phenotype has been observed, a promising strategy pursued in recent years focuses on controlling and preventing this formation by targeting and inhibiting the Quorum Sensing (QS) system, whose central role in biofilm has been extensively demonstrated. Therefore, the research and development of Quorum Quenching (QQ) compounds, which inhibit QS, has gradually attracted the attention of researchers and has become a new strategy for controlling harmful microorganisms. Among these, a number of both natural and synthetic compounds have been progressively identified as able to interrupt the intercellular communication within a microbial community and the adhesion to a surface, thus disintegrating mature/preformed biofilms. This review describes the role played by QS in the formation of bacterial biofilms and then focuses on the mechanisms of different natural and synthetic QS inhibitors (QSIs) exhibiting promising antibiofilm ability against Gram-positive and Gram-negative bacterial pathogens and on their applications as biocontrol strategies in various fields.

Abstract

Functional copy-number alterations (fCNAs) are DNA copy-number changes with concordant differential gene expression. These are less likely to be bystander genetic lesions and could serve as robust and reproducible tumor biomarkers. To identify candidate fCNAs in neuroendocrine tumors (NETs), we integrated chromosomal microarray (CMA) and RNA-seq differential gene-expression data from 31 pancreatic (pNETs) and 33 small-bowel neuroendocrine tumors (sbNETs). Tumors were resected from 47 early-disease-progression (24 months) patients. Candidate fCNAs that accurately differentiated these groups in this discovery cohort were then replicated using fluorescence in situ hybridization (FISH) on formalin-fixed, paraffin-embedded (FFPE) tissues in a larger validation cohort of 60 pNETs and 82 sbNETs (52 early- and 65 late-disease-progression samples). Logistic regression analysis revealed the predictive ability of these biomarkers, as well as the assay-performance metrics of sensitivity, specificity, and area under the curve. Our results indicate that copy-number changes at chromosomal loci 4p16.3, 7q31.2, 9p21.3, 17q12, 18q21.2, and 19q12 may be used as diagnostic and prognostic NET biomarkers. This involves a rapid, cost-effective approach to determine the primary tumor site for patients with metastatic liver NETs and to guide risk-stratified therapeutic decisions.

Abstract

The spread of antibiotic-resistant pathogens has prompted the development of novel approaches to identify molecules that synergize with antibiotics to enhance their efficacy. This study aimed to investigate the effects of ten Essential Oils (EOs) on the activity of nine antibiotics in influencing growth and biofilm formation in Escherichia coli, Pseudomonas aeruginosa, and Enterococcus faecalis. The effects of the EOs alone and in combination with antibiotics on both bacterial growth and biofilm formation were analyzed by measuring the MIC values through the broth microdilution method and the crystal violet assay, respectively. All EOs inhibited the growth of E. coli (1.25 ≤ MIC ≤ 5 mg/mL) while the growth of P. aeruginosa and E. faecalis was only affected by EOs from Origanum vulgare, (MIC = 5 mg/mL) and O. vulgare (MIC = 1.25 mg/mL) and Salvia rosmarinus (MIC = 5 mg/mL), respectively. In E. coli, most EOs induced a four- to sixteen-fold reduction in the MIC values of ampicillin, ciprofloxacin, ceftriaxone, gentamicin, and streptomycin, while in E. faecalis such a reduction is observed in combinations of ciprofloxacin with C. nepeta, C. bergamia, C. limon, C. reticulata, and F. vulgare, of gentamicin with O. vulgare, and of tetracycline with C. limon and O. vulgare. A smaller effect was observed in P. aeruginosa, in which only C. bergamia reduced the concentration of tetracycline four-fold. EO-antibiotic combinations also inhibit the biofilm formation. More precisely, all EOs with ciprofloxacin in E. coli, tetracycline in P. aeruginosa, and gentamicin in E. faecalis showed the highest percentage of inhibition. Combinations induce up- and down-methylation of cytosines and adenines compared to EO or antibiotics alone. The study provides evidence about the role of EOs in enhancing the action of antibiotics by influencing key processes involved in resistance mechanisms such as biofilm formation and epigenetic changes. Synergistic interactions should be effectively considered in dealing with pathogenic microorganisms.

Abstract

Context. QSO B1420+326 is a blazar classified as a Flat Spectrum Radio Quasar (FSRQ). In the beginning of 2020 it underwent an enhanced flux state. An extensive multiwavelength campaign allowed us to trace the evolution of the flare. Aims. We search for VHE gamma-ray emission from QSO B1420+326 during this flaring state. We aim to characterize and model the broadband emission of the source over different phases of the flare. Methods. The source was observed with a number of instruments in radio, near infrared, optical (including polarimetry and spectroscopy), ultra-violet, X-ray and gamma-ray bands. We use dedicated optical spectroscopy results to estimate the accretion disk and the dust torus luminosity. We perform spectral energy distribution modeling in the framework of combined Synchrotron-Self-Compton and External Compton scenario in which the electron energy distribution is partially determined from acceleration and cooling processes. Results. During the enhanced state the flux of both SED components drastically increased and the peaks were shifted to higher energies. Follow up observations with the MAGIC telescopes led to the detection of very-high-energy gamma-ray emission from this source, making it one of only a handful of FSRQs known in this energy range. Modeling allows us to constrain the evolution of the magnetic field and electron energy distribution in the emission region. The gamma-ray flare was accompanied by a rotation of the optical polarization vector during a low polarization state. Also, a new, superluminal radio knot contemporaneously appeared in the radio image of the jet. Abstract

The flat spectrum radio quasar (FSRQ) PKS 1510-089 is known for its complex multiwavelength behavior, and is one of only a few FSRQs detected at very high energy (VHE, $E>100\,$GeV) $\gamma$-rays. VHE $\gamma$-ray observations with H.E.S.S. and MAGIC during late May and early June 2016 resulted in the detection of an unprecedented flare, which reveals for the first time VHE $\gamma$-ray intranight variability in this source. While a common variability timescale of $1.5\,$hr is found, there is a significant deviation near the end of the flare with a timescale of $\sim 20\,$min marking the cessation of the event. The peak flux is nearly two orders of magnitude above the low-level emission. For the first time, curvature is detected in the VHE $\gamma$-ray spectrum of PKS 1510-089, which is fully explained through absorption by the extragalactic background light. Optical R-band observations with ATOM reveal a counterpart of the $\gamma$-ray flare, even though the detailed flux evolution differs from the VHE ightcurve. Interestingly, a steep flux decrease is observed at the same time as the cessation of the VHE flare. In the high energy (HE, $E>100\,$MeV) $\gamma$-ray band only a moderate flux increase is observed with Fermi-LAT, while the HE $\gamma$-ray spectrum significantly hardens up to a photon index of 1.6. A search for broad-line region (BLR) absorption features in the $\gamma$-ray spectrum indicates that the emission region is located outside of the BLR. Radio VLBI observations reveal a fast moving knot interacting with a standing jet feature around the time of the flare. As the standing feature is located $\sim 50\,$pc from the black hole, the emission region of the flare may have been located at a significant distance from the black hole. If this correlation is indeed true, VHE $\gamma$ rays have been produced far down the jet where turbulent plasma crosses a standing shock., Comment: 25 pages, 17 figures. Abstract

The coincident detection of GW170817 in gravitational waves and electromagnetic radiation spanning the radio to MeV gamma-ray bands provided the first direct evidence that short gamma-ray bursts (GRBs) can originate from binary neutron star (BNS) mergers. On the other hand, the properties of short GRBs in high-energy gamma rays are still poorly constrained, with only $\sim$20 events detected in the GeV band, and none in the TeV band. GRB~160821B is one of the nearest short GRBs known at $z=0.162$. Recent analyses of the multiwavelength observational data of its afterglow emission revealed an optical-infrared kilonova component, characteristic of heavy-element nucleosynthesis in a BNS merger. Aiming to better clarify the nature of short GRBs, this burst was automatically followed up with the MAGIC telescopes, starting from 24 seconds after the burst trigger. Evidence of a gamma-ray signal is found above $\sim$0.5 TeV at a significance of $\sim3\,\sigma$ during observations that lasted until 4 hours after the burst. Assuming that the observed excess events correspond to gamma-ray emission from GRB 160821B, in conjunction with data at other wavelengths, we investigate its origin in the framework of GRB afterglow models. The simplest interpretation with one-zone models of synchrotron-self-Compton emission from the external forward shock has difficulty accounting for the putative TeV flux. Alternative scenarios are discussed where the TeV emission can be relatively enhanced. Abstract

We report a characterization of the multi-band flux variability and correlations of the nearby (z=0.031) blazar Markarian 421 (Mrk 421) using data from Mets\"{a}hovi, Swift, Fermi-LAT, MAGIC, FACT and other collaborations and instruments from November 2014 till June 2016. Mrk 421 did not show any prominent flaring activity, but exhibited periods of historically low activity above 1 TeV (F$_{>1\mathrm{TeV}}<$ 1.7$\times$10$^{-12}$ ph cm$^{-2}$ s$^{-1}$) and in the 2-10 keV (X-ray) band (F$_{2-10 \mathrm{keV}}<$3.6$\times$10$^{-11}$ erg cm$^{-2}$ s$^{-1}$), during which the Swift-BAT data suggests an additional spectral component beyond the regular synchrotron emission. The highest flux variability occurs in X-rays and very-high-energy (E$>$0.1 TeV) $\gamma$-rays, which, despite the low activity, show a significant positive correlation with no time lag. The HR$_\mathrm{keV}$ and HR$_\mathrm{TeV}$ show the harder-when-brighter trend observed in many blazars, but the trend flattens at the highest fluxes, which suggests a change in the processes dominating the blazar variability. Enlarging our data set with data from years 2007 to 2014, we measured a positive correlation between the optical and the GeV emission over a range of about 60 days centered at time lag zero, and a positive correlation between the optical/GeV and the radio emission over a range of about 60 days centered at a time lag of $43^{+9}_{-6}$ days.This observation is consistent with the radio-bright zone being located about 0.2 parsec downstream from the optical/GeV emission regions of the jet. The flux distributions are better described with a LogNormal function in most of the energy bands probed, indicating that the variability in Mrk 421 is likely produced by a multiplicative process., Comment: 45 pages, 30 figures, 6 tables, accepted for publication in MNRAS
