The State of Michoacan has a wide and rich architectural heritage which is represented by different vernacular typologies which employ different materials and techniques according to the local resources. However, this wide knowledge and cultural heritage are disappearing, with several monuments and buildings being abandoned due to new dynamics and social changes. The traditional techniques have been substituted for modern and industrial construction systems since they are associated with obsolete ways of life. Through international cooperation for development and research projects, many activities were carried out to analyze the durability of local materials and vernacular constructions of the state. The main ones included the architectural survey and inspection of these buildings, photographic and drone surveys, characterization in the laboratory and oral interviews with local users. During the field work, these interviews with the local population, the creation of multimedia content and the use of audiovisual methods had crucial importance in the documentation of this local knowledge and culture, being great methods for scientific communication and dissemination. The main product is the documentary Xirangua, which means ‘roots’ in the Purépecha language, the main indigenous ethnicity in the state. This documentary explores the traditional architecture and construction of Michoacan, showing the reality within the loss of this vernacular heritage and portraying the importance of its conservation through the perspective of the local population, users and artisans. The use of these audiovisual techniques has demonstrated to be a suitable option to promote and preserve cultural heritage and educate, include the local actors in the process, and make the general public aware of the current issues, with this being one of the first works which explores this approach.

Background. Multiple acyl-CoA dehydrogenase deficiency is an autosomal recessive disorder of the amino acid metabolism and fatty acid oxidation due to the deficiency of the electron transfer protein or electron transfer protein ubiquinone oxidoreductase. The clinical picture ranges from a severe neonatal lethal presentation to late myopathic forms responsive to riboflavin. Up to now, there is no effective treatment for the neonatal form, which exhibits severe metabolic acidosis, hyperammonemia, hypoketotic hypoglycemia, and rhabdomyolysis. We present the case of a child who has had a good long-term outcome after a typical neonatal onset, with a dramatic drop in ammonia levels during the initial metabolic decompensation crisis and adequate control even during intercurrent diseases thereafter with N-carbamylglutamate treatment.

Reactive oxygen species present a challenge for marine organisms releasing gametes into the water. Thiol-containing molecules protect cells against oxidative stress, and ovothiol (OSH), an antioxidant-reducing mercaptohistidine, has been described as especially relevant in the oocytes of marine invertebrates. Ovothiol synthase (ovoA), in charge of the first step in OSH synthesis, was sequenced in mussels, Mytilus galloprovincialis. Transcription levels of ovoA in mantle did not significantly change along the reproductive cycle. No alterations of ovoA transcription were observed after a laboratory copper (10 µg/L) exposure or in mussels captured in a highly polluted site. Conversely, the metabolomic analysis of the hydrophilic metabolite content in mantle clearly classified mussels according to their site of origin, especially at the most advanced stages of oogenesis. Quantification of OSH-A and -B and glutathione (GSH), revealed stable levels in mantle at early gametogenesis in the unpolluted sampling site, but a strong increase in female mantle previous to spawning in the polluted site. These increased concentrations under pollution suggest that OSH-A accumulates along oogenesis, independent of gene transcription regulation. The concerted accumulation of OSH-A and GSH suggests the building of a balanced cellular redox-system to scavenge ROS produced in the oocyte before and during fertilization.

A principios del siglo XX, debido a la excitación provocada por la aparición del automóvil, los garajes funcionaron como centro social en el que los usuarios pasaban largas horas de la jornada compartiendo sus experiencias con la nueva máquina mientras el coche se limpiaba, reparaba o comercializaba. A mediados del siglo XX, la gran demanda de espacio para almacenaje del coche, la mecanización del garaje y el avance tecnológico en el uso de materiales, trajeron consigo dos consecuencias que transformarían por completo el tipo y la trama urbana: un cambio de escala considerable y una especialización programática. En la actualidad estas estructuras están adaptándose a los nuevos cambios en la movilidad y empiezan nuevamente a albergar usos coexistentes y a servir como vínculos comunitarios. El presente artículo describe y analiza la evolución cíclica de la tipología del garaje a través de varios ejemplos que van marcando los cambios de uso y funcionalidad de estos hitos urbanos desde principios del siglo XX hasta nuestros días (Figura 1).

Abstract Marine Porifera host diverse microbial communities, which influence host metabolism and fitness. However, functional relationships between sponge microbiomes and metabolic signatures are poorly understood. We integrate microbiome characterization, metabolomics and microbial predicted functions of four coexisting Mediterranean sponges –Petrosia ficiformis, Chondrosia reniformis, Crambe crambe and Chondrilla nucula. Microscopy observations reveal anatomical differences in microbial densities. Microbiomes exhibit strong species-specific trends. C. crambe shares many rare amplicon sequence variants (ASV) with the surrounding seawater. This suggests important inputs of microbial diversity acquired by selective horizontal acquisition. Phylum Cyanobacteria is mainly represented in C. nucula and C. crambe. According to putative functions, the microbiome of P. ficiformis and C. reniformis are functionally heterotrophic, while C. crambe and C. nucula are autotrophic. The four species display distinct metabolic profiles at single compound level. However, at molecular class level they share a “core metabolome”. Concurrently, we find global microbiome-metabolome association when considering all four sponge species. Within each species still, sets of microbe/metabolites are identified driving multi-omics congruence. Our findings suggest that diverse microbial players and metabolic profiles may promote niche diversification, but also, analogous phenotypic patterns of “symbiont evolutionary convergence” in sponge assemblages where holobionts co-exist in the same area.

Online learning anxiety might be related to how university students perceived their learning and performance during the COVID-19 pandemic, and in course modules with mathematical content, math anxiety may also have come into play. The aim of this study was to examine the relationships between these types of anxiety, as well as to explore other possible factors associated with students’ experience of sudden transition to online learning in a module with high mathematical content. Participants were 125 psychology undergraduates enrolled in a Research Design module. The main results showed that students reporting greater online learning anxiety found online materials less useful and suffered from greater math anxiety; their self-perceived performance was also worse. In the year following lockdown, online learning anxiety was lower and the perceived usefulness of online material was greater than during lockdown. Finally, analysis of the association between psychological state in the use of e-learning tools during COVID-19 and online learning anxiety revealed that online learning readiness played a mediating role in this relationship. The paper concludes with a series of recommendations regarding how universities might better prepare for future unforeseen disruptions to face-to-face teaching. In particular, there is a need to improve students’ readiness for online learning and to ensure that online materials are perceived as useful.

Abstract SARS-CoV-2 reinfections have been frequent, even among those vaccinated. The aim of this study is to know if hybrid immunity (infection + vaccination) is affected by the moment of vaccination and number of doses received. We conducted a retrospective study in 746 patients with a history of COVID-19 reinfection and recovered the dates of infection and reinfection and vaccination status (date and number of doses). To assess differences in the time to reinfection(tRI) between unvaccinated, vaccinated before 6 months, and later; and comparing one, two or three doses (incomplete, complete and booster regime) we performed the log-rank test of the cumulative incidence calculated as 1 minus the Kaplan–Meier estimator. Coastal environments are complex systems that are influenced by a combination of natural processes and human activities. Scientific interest in the effects of coastal erosion/accretion and climatic change-related processes has greatly increased in recent decades due to the growing human development along coastal areas. This paper investigates the state and evolution of beach–dune systems for the 1977–2001 and 2001–2019 periods of the Mediterranean coast of Andalusia (Spain) using two different proxies: the dune toe line, which was used to track foredunes evolution, and the high-water line, which was used to assess shoreline evolution. Results showed a general erosional behavior of the studied beach–dune systems and identified cases where the main trend was altered through human interventions. During the 1977–2001 period, foredunes essentially showed erosion (54%), accretion (24%), and stability (22%) and shorelines showed accretion (40%) and erosion and stability (34% each). During the 2001–2019 period, foredunes essentially showed erosion (42%), stability (30%), and accretion (28%), and shorelines showed erosion (40%), accretion (34%), and stability (26%). Combining the evolution classes of each proxy (dune toe/shoreline) allows the behavior of both shoreline proxies to be assessed together and provides insights additional to those derived from the use of a single proxy. In this regard, Erosion/erosion (EE) and Accretion/accretion (AA) were the most frequent behaviors in the first and second periods. The results obtained provide additional insights on the nature and drivers of coastal change that aid local coastal managers and administrations in understanding erosion processes. The method can be applied to other areas around the world where a similar database is available.

Inherited thrombophilia (IT) has been implicated as a potential causal factor of adverse pregnancy outcomes (APOs), including recurrent miscarriage with and without the presence of antiphospholipid syndrome (APS). The aim of this study was to assess the prevalence and impact of IT on fetal–maternal outcomes and thrombotic risk in women within the spectrum of obstetric APS. Three hundred and twenty-eight women with APS-related obstetric morbidity ever pregnant were included. Of these, 74 met the APS classification criteria, 169 were non-criteria (NC)-APS, and 85 were seronegative (SN)-APS. Patients with other autoimmune diseases were excluded. APOs included early pregnancy loss, fetal death, preeclampsia, abruptio placentae, and preterm birth. Successful pregnancy was defined as the achievement of a live newborn. A literature search was also performed. The mean age of the overall group was 33.9 ± 5.3 years, and the patients were followed up for 35 (11–79) months. During the study period, there were 1332 pregnancies. Nearly 14% of the patients had an associated IT. IT patients more frequently received the standard-of-care (SoC) therapy. The presence of IT was not associated with worse maternal–fetal outcomes in patients treated with SoC treatment. Overall, IT patients had a lower frequency of newborns without treatment, especially those without definite APS. In addition, IT did not increase the risk of thrombosis during pregnancy or the postpartum period. A detailed analysis of the literature review identified only four publications related to our study and did not show conclusive evidence of the impact of IT on patients with obstetric APS. The group of women with APS-related obstetric morbidity and IT who did not receive treatment, especially those without definite APS, had a worse prognosis in terms of a live birth. However, with SoC therapy, the prognosis is similar in those patients without IT. The association of IT with APS does not seem to predispose to the development of thrombosis during pregnancy and/or the postpartum period.

A collaborative trial involving 16 participants from nine European countries was conducted within the NORMAN network in efforts to harmonise suspect and non-target screening of environmental contaminants in whole fish samples of bream (Abramis brama). Participants were provided with freeze-dried, homogenised fish samples from a contaminated and a reference site, extracts (spiked and non-spiked) and reference sample preparation protocols for liquid chromatography (LC) and gas chromatography (GC) coupled to high resolution mass spectrometry (HRMS). Participants extracted fish samples using their in-house sample preparation method and/or the protocol provided. Participants correctly identified 9–69 % of spiked compounds using LC-HRMS and 20–60 % of spiked compounds using GC-HRMS. From the contaminated site, suspect screening with participants’ own suspect lists led to putative identification of on average ∼145 and ∼20 unique features per participant using LC-HRMS and GC-HRMS, respectively, while non-target screening identified on average ∼42 and ∼56 unique features per participant using LC-HRMS and GC-HRMS, respectively. Within the same sub-group of sample preparation method, only a few features were identified by at least two participants in suspect screening (16 features using LC-HRMS, 0 features using GC-HRMS) and non-target screening (0 features using LC-HRMS, 2 features using GC-HRMS). The compounds identified had log octanol/water partition coefficient (KOW) values from −9.9 to 16 and mass-to-charge ratios (m/z) of 68 to 761 (LC-HRMS and GC-HRMS). A significant linear trend was found between log KOW and m/z for the GC-HRMS data. Overall, these findings indicate that differences in screening results are mainly due to the data analysis workflows used by different participants. Further work is needed to harmonise the results obtained when applying suspect and non-target screening approaches to environmental biota samples.

Slaked lime is traditionally a suitable material for use in construction activities in historic and traditional buildings whose constitutive characteristics demand compatible and appropriate material solutions. Therefore, knowing the physical and mechanical characterization of lime pastes and mortars for use in restoration is considered an important step in the process of scientific development of a material that, in the Mexican case, has lacked scientific rigor in its production, use and commercialization. With this in mind, the present article aims to investigate the characteristics of lime pastes and mortars that have been used in the traditional way and the new limes that exist in the market, offering suitable options for restoration, for which an integrated methodology was used consisting of physical tests such as surface area, electrical resistivity, pulse velocity, colorimetry, and mechanical tests such as compressive strength, giving as a result that “traditional slaked” lime is more likely to deteriorate than powdered lime, which leads us to conclude that powdered hydrated lime has optimal characteristics for restoration activities.

This study compared the effects of computer-mediated (CM) versus pen-and-paper (P&P) writing on written accuracy and feedback processing in tasks written and rewritten collaboratively following a pedagogical treatment in two intact authentic classrooms. The study involved 32 secondary education low-proficiency English-as-a-foreign-language (EFL) learners writing two descriptive texts collaboratively and receiving in-class training in the identification and correction of grammatical, lexical, and mechanical errors. Participants were provided with unfocused direct error correction (EC). Error logs were used to facilitate noticing of teacher corrections (i.e., feedback processing). Dyads were required to rewrite their texts for evidence of feedback uptake. Results indicate that writing collaboratively on the computer with the availability of the Internet contributes to increased grammatical and lexical accuracy. No differences were found between writing environments regarding feedback processing or accuracy of rewritten texts.

Rapid urbanisation causes a rise in the need for infrastructure, which in turn fuels the creation of additional concrete and further increases cement supplies. Activation of illite-based clay mineral and usage in concrete production is one of the sustainable ways to address the cement industry anthropogenic issues. This study evaluates the durability properties of water transport (water absorption, and capillary water absorption), and resistance to aggressive environments (5% solutions of hydrochloric acid, HCl; sodium sulphate, Na2SO4; and calcium chloride, CaCl2) of meta-illite calcined clay (MCC)-based high-performance concrete (HPC). For this purpose, concrete was produced with 5, 10, 15, 20, 25 and 30% MCC content in partial substitution of CEM II. Results from the water absorption tests indicate an average percentage value of 3.57%, 3.35% and 2.52% for all the observed mixes at 28, 56 and 90 days, respectively, with MCCC-10 HPC having an average best value of 2.23% across the curing ages. On all observed days, the 5 to 15% cement replacements had very close average water sorptivity value of 0.125 ± 0.001 mm/min0.5 with the control mix (0.113 ± 0.011 mm/min0.5). The aggressive environments exposure findings of the hardened MCC-based HPC specimens of 10 to 20% recorded an approximately 15% compressive strength loss in HCl, Na2SO4 and CaCl2 solutions over the 90 days of curing. In all, the HPC mixes of 5 to 15% MCC content obtained an average durability performance factor of 89%. As a result, these findings imply that MCC can replace cement in up to 15% of HPC production.

This study aimed to develop a rheometer prototype and define a procedure for adjusting the automated control of the fresh state of concrete. Sixteen batches were produced, and their fresh behaviour was measured at different testing times by applying the Abrams cone and flow curve test (FCT) as the rheological protocol. During this test, the yield stress and plastic viscosity of the concretes were measured in relative units. The rheometer prototype was used to define a new protocol to select the most suitable rheometer impeller arrangement and optimal FCT configuration. This protocol considers the torque at the end of the breakdown period, torque reduction during the breakdown period, segregation, and negative values of the yield stress in relative units. This protocol also enabled an iterative adjustment procedure, facilitating the use of a rheometer for the automated control of the homogeneity and behaviour of fresh concrete, as well as real-time decision making.

ABSTRACT Cefotaximase-Munich (CTX-M) extended-spectrum beta-lactamase (ESBL) enzymes produced by Enterobacteriaceae confer resistance to clinically relevant third-generation cephalosporins. CTX-M group 1 variants, CTX-M-1 and CTX-M-15, are the leading ESBL-producing Enterobacteriaceae associated with animal and human infection, respectively, and are an increasing antimicrobial resistance (AMR) global health concern. The blaCTX-M-1 and blaCTX-M-15 genes encoding these variants have an approximate nucleotide sequence similarity of 98.7%, making effective differential diagnostic monitoring difficult. Loop-primer endonuclease cleavage loop-mediated isothermal amplification (LEC-LAMP) enables rapid real-time multiplex pathogen detection with single-base specificity and portable on-site testing. We have developed an internally controlled multiplex CTX-M-1/15 LEC-LAMP assay for the differential detection of blaCTX-M-1 and blaCTX-M-15. Assay analytical specificity was established using a panel of human, animal, and environmental Escherichia coli isolates positive for blaCTX-M-1 (n = 18), blaCTX-M-15 (n = 35), and other closely related blaCTX-Ms (n = 38) from Ireland, Germany, and Portugal, with analytical sensitivity determined using probit regression analysis. Animal fecal sample testing using the CTX-M-1/15 LEC-LAMP assay in combination with a rapid DNA extraction protocol was carried out on porcine fecal samples previously confirmed to be PCR-positive for E. coli blaCTX-M. Portable instrumentation was used to further analyze each fecal sample and demonstrate the on-site testing capabilities of the LEC-LAMP assay with the rapid DNA extraction protocol. The CTX-M-1/15 LEC-LAMP assay demonstrated complete analytical specificity for the differential detection of both variants with sensitive low-level detection of 8.5 and 9.8 copies per reaction for blaCTX-M-1 and blaCTX-M-15, respectively, and E. coli blaCTX-M-1 was identified in all blaCTX-M positive porcine fecal samples tested. IMPORTANCE CTX-M ESBL-producing E. coli is an increasing AMR public health issue with the transmission between animals and humans via zoonotic pathogens now a major area of interest. Accurate and timely identification of ESBL-expressing E. coli CTX-M variants is essential for disease monitoring, targeted antibiotic treatment and infection control. This study details the first report of portable diagnostics technology for the rapid differential detection of CTX-M AMR markers blaCTX-M-1 and blaCTX-M-15, facilitating improved identification and surveillance of these closely related variants. Further application of this portable internally controlled multiplex CTX-M-1/15 LEC-LAMP assay will provide new information on the transmission and prevalence of these CTX-M ESBL alleles. Furthermore, this transferable diagnostic technology can be applied to other new and emerging relevant AMR markers of interest providing more efficient and specific portable pathogen detection for improved epidemiological surveillance.
