The piriform cortex (PCx) is essential for the adaptive processing of olfactory information. Neuromodulatory systems, including those utilizing serotonin, acetylcholine, noradrenaline, and dopamine, innervate and regulate neuronal activity in the PCx. Previous research has demonstrated the importance of acetylcholine, noradrenaline and serotonin in odor learning and memory. In contrast, the role of dopamine in the PCx remains under-explored. Here we examined how dopamine modulates the intrinsic electrical properties of identified classes of neurons in the PCx. We found that dopamine had no consistent effect on the intrinsic electrical properties of two types of glutamatergic neurons (semilunar and superficial pyramidal cells) or three types of GABAergic interneurons (horizontal, neurogliaform and somatastatin-expressing regular-spiking cells). However, dopamine had a striking effect on the intrinsic excitability of the parvalbumin-expressing fast-spiking (FS) class of GABAergic interneuron. Dopamine depolarized the resting potential, increased the input resistance and increased the firing frequency of FS cells. Co-application of dopamine with the D1-class dopamine receptor antagonist SCH 23390 blocked the effects of dopamine modulation on FS cells. Conversely, co-application of dopamine with the D2-class antagonist RS-(±)-sulpiride had no effect on dopamine modulation of these cells. Our results indicate that dopamine binds to D1-class dopamine receptors to increase the intrinsic excitability of FS cells. These findings suggest that dopamine has a highly targeted effect in the PCx and reveal how dopamine may modulate the balance between excitation and inhibition, with consequences for odor processing. Feedforward inhibitory circuits are key contributors to the complex interplay between excitation and inhibition in the brain. Little is known about the function of feedforward inhibition in the primary olfactory (piriform) cortex. Using in vivo two-photon-targeted patch clamping and calcium imaging in mice, we find that odors evoke strong excitation in two classes of interneurons - neurogliaform (NG) cells and horizontal (HZ) cells - that provide feedforward inhibition in layer 1 of the piriform cortex. NG cells fire much earlier than HZ cells following odor onset, a difference that can be attributed to the faster odor-driven excitatory synaptic drive that NG cells receive from the olfactory bulb. As a result, NG cells strongly but transiently inhibit odor-evoked excitation in layer 2 principal cells, whereas HZ cells provide more diffuse and prolonged feedforward inhibition. Neurons typically form daisy chains of synaptic connections with other neurons, but they can also form synapses with themselves. Although such self-synapses, or autapses, are comparatively rare in vivo , they are surprisingly common in dissociated neuronal cultures. At first glance, autapses in culture seem like a mere curiosity. However, by providing a simple model system in which a single recording electrode gives simultaneous access to the pre- and postsynaptic compartments, autaptic cultures have proven to be invaluable in facilitating important and elegant experiments in the area of synaptic neuroscience. Here, I provide detailed protocols for preparing and recording from autaptic cultures (also called micro-island or microdot cultures). Variations on the basic procedure are presented, as well as practical tips for optimizing the outcomes. I also illustrate the utility of autaptic cultures by reviewing the types of experiments that have used them over the past three decades. These examples serve to highlight the power and elegance of this simple model system, and will hopefully inspire new experiments for the interrogation of synaptic function., (Copyright © 2020 Bekkers.)

Key Points: The primary olfactory (or piriform) cortex is a promising model system for understanding how the cerebral cortex processes sensory information, although an investigation of the piriform cortex is hindered by a lack of detailed information about the intrinsic electrical properties of its component neurons. In the present study, we quantify the properties of voltage-dependent sodium currents and voltage- and calcium-dependent potassium currents in two important classes of excitatory neurons in the main input layer of the piriform cortex. We identify several classes of these currents and show that their properties are similar to those found in better-studied cortical regions. Our detailed quantitative descriptions of these currents will be valuable to computational neuroscientists who aim to build models that explain how the piriform cortex encodes odours., Abstract: The primary olfactory cortex (or piriform cortex, PC) is an anatomically simple palaeocortex that is increasingly used as a model system for investigating cortical sensory processing. However, little information is available on the intrinsic electrical conductances in neurons of the PC, hampering efforts to build realistic computational models of this cortex. In the present study, we used nucleated macropatches and whole-cell recordings to rigorously quantify the biophysical properties of voltage-gated sodium (Na V ), voltage-gated potassium (K V ) and calcium-activated potassium (K Ca ) conductances in two major classes of glutamatergic neurons in layer 2 of the PC, semilunar (SL) cells and superficial pyramidal (SP) cells. We found that SL and SP cells both express a fast-inactivating Na V current, two types of K V current (A-type and delayed rectifier-type) and three types of K Ca current (fast-, medium- and slow-afterhyperpolarization currents). The kinetic and voltage-dependent properties of the Na V and K V conductances were, with some exceptions, identical in SL and SP cells and similar to those found in neocortical pyramidal neurons. The K Ca conductances were also similar across the different types of neurons. Our results are summarized in a series of empirical equations that should prove useful to computational neuroscientists seeking to model the PC. The piriform cortex (PC), like other cortical regions, normally operates in a state of dynamic equilibrium between excitation and inhibition. Here we examined the roles played by pre- and postsynaptic GABA B receptors in maintaining this equilibrium in the PC. Using whole-cell recordings in brain slices from the anterior PC of mice, we found that synaptic activation of postsynaptic GABA B receptors hyperpolarized the two major classes of layer 2 principal neurons and reduced the intrinsic electrical excitability of these neurons. Presynaptic GABA B receptors are expressed on the terminals of associational (intracortical) glutamatergic axons in the PC. Heterosynaptic activation of these receptors reduced excitatory associational inputs onto principal cells. Presynaptic GABA B receptors are also expressed on the axons of GABAergic interneurons in the PC, and blockade of these autoreceptors enhanced inhibitory inputs onto principal cells. Hence, presynaptic GABA B autoreceptors produce disinhibition of principal cells. To study the functional consequences of GABA B activation in vivo , we used 2-photon calcium imaging to simultaneously monitor the activity of ~200 layer 2 neurons. Superfusion of the GABA B agonist baclofen reduced spontaneous random firing but also promoted synchronous epileptiform activity. These findings suggest that, while GABA B activation can dampen excitability by engaging pre- and postsynaptic GABA B heteroreceptors on glutamatergic neurons, it can also promote excitability by disinhibiting principal cells by activating presynaptic GABA B autoreceptors on interneurons. Thus, depending on the dynamic balance of hetero- and autoinhibition, GABA B receptors can function as variable modulators of circuit excitability in the PC.

Neurons in the neocortex exhibit spontaneous spiking activity in the absence of external stimuli, but the origin and functions of this activity remain uncertain. Here, we show that spontaneous spiking is also prominent in a sensory paleocortex, the primary olfactory (piriform) cortex of mice. In the absence of applied odors, piriform neurons exhibit spontaneous firing at mean rates that vary systematically among neuronal classes. This activity requires the participation of NMDA receptors and is entirely driven by bottom-up spontaneous input from the olfactory bulb. Odor stimulation produces two types of spatially dispersed, odor-distinctive patterns of responses in piriform cortex layer 2 principal cells: Approximately 15% of cells are excited by odor, and another approximately 15% have their spontaneous activity suppressed. Our results show that, by allowing odor-evoked suppression as well as excitation, the responsiveness of piriform neurons is at least twofold less sparse than currently believed. Hence, by enabling bidirectional changes in spiking around an elevated baseline, spontaneous activity in the piriform cortex extends the dynamic range of odor representation and enriches the coding space for the representation of complex olfactory stimuli.

Despite its comparatively simple trilaminar architecture, the primary olfactory (piriform) cortex of mammals is capable of performing sophisticated sensory processing, an ability that is thought to depend critically on its extensive associational (intracortical) excitatory circuits. Here, we used a novel transgenic mouse model and optogenetics to measure the connectivity of associational circuits that originate in semilunar (SL) cells in layer 2a of the anterior piriform cortex (aPC). We generated a mouse line (48L) in which channelrhodopsin-2 (ChR) could be selectively expressed in a subset of SL cells. Light-evoked excitatory postsynaptic currents (EPSCs) could be evoked in superficial pyramidal cells (17.4% of n = 86 neurons) and deep pyramidal cells (33.3%, n = 9) in the aPC, but never in ChR- SL cells (0%, n = 34). Thus, SL cells monosynaptically excite pyramidal cells, but not other SL cells. Light-evoked EPSCs were also selectively elicited in 3 classes of GABAergic interneurons in layer 3 of the aPC. Short-beaked echidnas (Tachyglossus aculeatus) are myrmecophages, or ant and termite insectivore specialists, and replicating their exact diet in captivity is problematic. Diets for captive animals often incorporate raw meat, eggs and cat food mixed together with water, and vitamin and mineral supplements. These diets have promoted a number of health problems in captive echidnas, such as gastritis, cystitis, gut impaction, obesity, and diarrhea. A manufactured diet was designed and three echidnas from two zoos were transitioned onto this diet to assess the acceptability and digestibility of this diet for echidnas. The new "test" diet was readily accepted by the echidnas with a 1 week transition period. Daily digestible energy intake was 280 kJ kg -0.75 d -1 , similar to another myrmecophagous species. Digestibility values were above 74% for all macronutrients. It was determined that this diet was an acceptable replacement for the previous diets and it was decided that the remaining echidnas at both institutions would be transitioned to the new diet. The diet will also be used for wild echidnas being rehabilitated in the zoo hospitals prior to release and commercially available within Australia. Further data are being collected to assess the use of this diet for seasonal weight management, transitioning hand-reared puggles and effects on gastrointestinal tract health. Neural circuits are typically maintained in a state of dynamic equilibrium by balanced synaptic excitation and inhibition. However, brain regions that are particularly susceptible to epilepsy may have evolved additional specialized mechanisms for inhibiting over-excitation. Here we identify one such possible mechanism in the cerebral cortex and hippocampus of mice. Recently it was reported that some types of GABAergic interneurons can slowly integrate excitatory inputs until eventually they fire persistently in the absence of the original stimulus. This property, called persistent firing or retroaxonal barrage firing (BF), is of unknown physiological importance. We show that two common types of interneurons in cortical regions, neurogliaform (NG) cells and fast-spiking (FS) cells, are unique in exhibiting BF in acute slices (~85 and ~23% success rate for induction, respectively). BF can also be induced in vivo, although the success rate for induction is lower (~60% in NG cells). In slices, BF could reliably be triggered by trains of excitatory synaptic input, as well as by exposure to proconvulsant bath solutions (elevated extracellular K(+), blockade of GABAA receptors). Using pair recordings in slices, we confirmed that barrage-firing NG cells can produce synaptic inhibition of nearby pyramidal neurons, and that this inhibition outlasts the original excitation. The ubiquity of NG and FS cells, together with their ability to fire persistently following excessive excitation, suggests that these interneurons may function as cortical sentinels, imposing an activity-dependent brake on undesirable neuronal hyperexcitability.

Increased understanding of the early stages of olfaction has lead to a renewed interest in the higher brain regions responsible for forming unified 'odor images' from the chemical components detected by the nose. The piriform cortex, which is one of the first cortical destinations of olfactory information in mammals, is a primitive paleocortex that is critical for the synthetic perception of odors. Here we review recent work that examines the cellular neurophysiology of the piriform cortex. Local inhibition by GABA-releasing neurons is important for the operation of sensory cortices, but the details of these inhibitory circuits remain unclear. We addressed this question in the olfactory system by making targeted recordings from identified classes of inhibitory and glutamatergic neurons in the piriform cortex (PC) of mice. First, we looked for feedforward synaptic inhibition provided by interneurons located in the outermost layer of the PC, layer Ia, which is the unique recipient of afferent fibers from the olfactory bulb. We found two types of feedforward inhibition: a fast-rising, spatially restricted kind that was generated by horizontal cells, and a slow-rising, more diffuse kind generated by neurogliaform cells. Both cell types targeted the distal apical dendrites of layer II principal neurons. Next, we studied feedback synaptic inhibition in isolation by making a tissue cut across layer I to selectively remove feedforward inhibitory connections. We identified a powerful type of feedback inhibition of layer II neurons, mostly generated by soma-targeting fast-spiking multipolar cells in layer III, which in turn were driven by feedforward excitation from layer II semilunar cells. Dynamic clamp simulation of feedback inhibition revealed differential effects of this inhibition on the two main types of layer II principal neurons. Thus, our results articulate the connectivity and functions of two important classes of inhibitory microcircuits in the PC. Feedforward and feedback inhibition generated by these circuits is likely to be required for the operation of this sensory paleocortex during the processing of olfactory information.

The ability of neurons to process synaptic inputs depends critically on their passive electrical properties. The intracellular resistivity, R(i), is one of the parameters that determine passive properties, yet few experiments have explored how changes in R(i) might affect synaptic integration. In this work, I addressed this issue by using targeted dendritic occlusion to locally increase R(i) in cerebellar Purkinje cells and examining the consequences of this manipulation for the summation of synaptic inputs. To achieve dendritic occlusion, I used two glass micropipettes to gently pinch the dendritic trunk close to the soma. This pinching produced stereotypical changes in the responses to test pulses applied at the soma under voltage and current clamp. A simple model confirmed that these changes were due to increases in R(i) in the dendritic trunk. These localized increases in R(i) produced striking alterations in the shapes of postsynaptic potentials at the soma, increasing their amplitude and accelerating their decay kinetics. As a consequence, dendritic occlusion sharpened temporal precision during the summation of synaptic inputs. The primary olfactory (or piriform) cortex is a trilaminar paleocortex that is thought to construct unified "odor images" from the odor components identified by the olfactory bulb. How the piriform cortex (PC) accomplishes this sophisticated synthetic task, despite its relatively simple architecture, is unknown. Here we used in vitro patch-clamp recordings from acute slices of the anterior PC of mice to identify microcircuits involved in excitatory synaptic processing. Cluster analysis confirmed the presence of two prominent classes of glutamatergic principal cells in the main input layer (layer II) of the PC: semilunar (SL) cells and superficial pyramidal (SP) cells. SL cells received stronger afferent excitatory input from the olfactory bulb, on average, than did SP cells. This was due to the larger mean strength of single-fiber afferents onto SL cells. In contrast, SP cells received stronger associational (intracortical) excitatory inputs, most likely due to their more extensive dendritic trees within the associational layers. Tissue-cut experiments and dual recordings from SL and SP cells in disinhibited slices were consistent with the distinctive patterns of connectivity of these two cell classes. Our findings suggest that the anterior PC employs at least two layers of excitatory synaptic processing: one involving strong afferent inputs onto SL cells, and another involving strong intracortical inputs onto SP cells. This architecture may allow the PC to sequentially process olfactory information within segregated subcircuits.

The primary olfactory (or piriform) cortex is a trilaminar paleocortex that is seen increasingly as an attractive model system for the study of cortical sensory processing. Recent findings highlight the importance of γ-amino butyric acid (GABA)-releasing interneurons for the function of the piriform cortex (PC), yet little is known about the different types of interneurons in the PC. Here, we provide the first detailed functional characterization of the major classes of GABAergic interneurons in the anterior piriform cortex (aPC) and show how these classes differentially engage in phasic synaptic inhibition. By measuring the electrical properties of interneurons and combining this with information about their morphology, laminar location, and expression of molecular markers, we have identified 5 major classes in the aPC of the mouse. Each layer contains at least one class of interneuron that is tuned to fire either earlier or later in a train of stimuli resembling the input received by the PC in vivo during olfaction. This suggests that the different subtypes of interneuron are specialized for providing synaptic inhibition at different phases of the sniff cycle. Thus, our results suggest mechanisms by which classes of interneurons play specific roles in the processing performed by the PC in order to recognize odors.

The primary olfactory cortex (or piriform cortex, PC) is attracting increasing attention as a model system for the study of cortical sensory processing, yet little is known about inhibitory neurons in the PC. Here we provide the first systematic classification of GABA-releasing interneurons in the anterior PC of mice, based on the expression of molecular markers. Our experiments used GAD67-GFP transgenic mice, in which gamma-aminobutyric acid (GABA)-containing cells are labeled with green fluorescent protein (GFP). We first confirmed, using paired whole-cell recordings, that GFP(+) neurons in the anterior PC of GAD67-GFP mice are functionally GABAergic. Next, we performed immunolabeling of GFP(+) cells to quantify their expression of every possible pairwise combination of seven molecular markers: calbindin, calretinin, parvalbumin, cholecystokinin, neuropeptide Y, somatostatin, and vasoactive intestinal peptide. We found that six main categories of interneurons could be clearly distinguished in the anterior PC, based on the size and laminar location of their somata, intensity of GFP fluorescence, patterns of axonal projections, and expression of one or more of the seven markers. A number of rarer categories of interneurons could also be identified. These data provide a road map for further work that examines the functional properties of the six main classes of interneurons. An autapse is a synapse between a neuron and itself, a peculiar structure with an unclear function. A new study suggests that excitatory autapses contribute to a positive-feedback loop that maintains persistent electrical activity in neurons.

Synaptic transmission depends on the continued availability of neurotransmitter-filled synaptic vesicles (SVs) for triggered release from presynaptic boutons. Surprisingly, small boutons in the brain, that already contain comparatively few SVs, are thought to retain the majority of these SVs in a "reserve" pool that is not mobilized under physiological conditions. Why such a scarce synaptic resource is normally inaccessible has been a matter of debate. Here, we readdress this issue by developing an electrophysiological approach for counting SVs released from boutons formed by a single, isolated neuron on itself ("autapses"). We show that, after treatment with Bafilomycin A1 to prevent reloading of discharged SVs with glutamate, each SV is counted only once on first-time release. Hence, by integrating all autaptic currents as they run down over time, we can estimate the total number of SVs released by a single neuron. This total can be normalized to the number of boutons on the neuron, giving the mean number of SVs released per bouton. We estimate that up to approximately 130 vesicles can be released per bouton over approximately 10 min of stimulation at 0.2 Hz. This number of vesicles represents a substantial proportion of the total number of SVs (100-200) that have been counted in these boutons by using electron microscopy. Thus, mild electrical stimulation, when maintained for sufficient time, causes the eventual release of many of the SVs in a bouton, including those in the putative reserve pool. This result suggests that SVs are functionally homogeneous in that the majority can contribute to basal synaptic transmission.

Normal brain function depends on an interplay between glutamatergic and GABAergic synaptic transmission, yet questions remain about the biophysical differences between these two classes of synapse. By taking advantage of a simple culture system, we present here a detailed comparison of excitatory and inhibitory neurotransmission under identical conditions using the variance-mean (V-M) method of quantal analysis. First, we validate V-M analysis for glutamatergic autapses formed by isolated hippocampal pyramidal neurons in culture, confirming that the analysis accurately predicts the quantal amplitude (Q). We also show that V-M analysis is only weakly sensitive to intersite and intrasite quantal variance and to the known inhomogeneities in release probability (P(r)). Next, by repeating the experiments with GABAergic autapses, we confirm that V-M analysis provides an accurate account of inhibitory neurotransmission in this system. Mean P(r), provided by V-M analysis, shows a dependence on extracellular Ca(2+) concentration that is nearly identical for both excitatory and inhibitory autapses. Finally, the V-M method allows us to compare the locus of short-term synaptic plasticity at these connections. Glutamatergic autapses exhibit paired-pulse depression that depends mainly on changes in P(r), whereas depression at GABAergic autapses appears to depend primarily on changes in the number of release sites. We conclude that, apart from differences in the mechanisms of short-term plasticity, the basic quantal properties of excitatory and inhibitory connections in this hippocampal system are remarkably similar.

After oral administration of tobramycin, as part of selective decontamination of the digestive tract (SDD) in critically ill patients, absorption of tobramycin from the gut into the blood may take place. To quantify low concentrations of tobramycin in human plasma, we developed and validated a simple (sample pre-treatment consisting of protein precipitation with acetonitrile using 200microl plasma), rapid (runtime 3min using a Pathfinder MR reversed-phase column) and sensitive (concentration range of 0.05-1.0mg/l using MS/MS detection) method.

1. The piriform cortex (PC) is the largest subdivision of the olfactory cortex and the first cortical destination of olfactory information. Despite the relatively simple anatomy of the PC and its obvious appeal as a model system for the study of cortical sensory processing, there are many outstanding questions about its basic cell physiology. In the present article, we review what is known about GABAergic inhibitory interneurons in the PC. 2. The GABA-containing neurons in the PC are morphologically diverse, ranging from small neurogliaform cells to large multipolar forms. Some of these classes are distributed across all three main layers of the PC, whereas others have a more restricted laminar expression. 3. Distinct and overlapping populations of GABAergic basket cells in Layers II and III of the PC express different combinations of calcium-binding proteins and neuropeptides. Few Layer I interneurons express any of the molecular markers so far examined. 4. The intrinsic firing properties of one or two types of putative PC interneurons have been measured and inhibitory post-synaptic responses have been recorded in PC pyramidal cells following extracellular stimulation. However, little is known about the physiology of the subtypes of interneurons identified. 5. In view of the likely importance of PC interneurons in olfactory learning, olfactory coding and epileptogenesis, further investigation of their properties is likely to be highly informative.

Neurons typically function as transduction devices, converting patterns of synaptic inputs, received on the dendrites, into trains of output action potentials in the axon. This transduction process is surprisingly complex and has been proposed to involve a two-way dialogue between axosomatic and dendritic compartments that can generate mutually interacting regenerative responses. To manipulate this process, we have developed a new approach for rapid and reversible occlusion or amputation of the primary dendrites of individual neurons in brain slices. By applying these techniques to cerebellar Purkinje and layer 5 cortical pyramidal neurons, we show directly that both the active and passive properties of dendrites differentially affect firing in the axon depending on the strength of stimulation. For weak excitation, dendrites act as a passive electrical load, raising spike threshold and dampening axonal excitability. For strong excitation, dendrites contribute regenerative inward currents, which trigger burst firing and enhance neuronal excitability. These findings provide direct support for the idea that dendritic morphology and conductances act in concert to regulate the excitability of the neuron.

The piriform (or primary olfactory) cortex is a trilaminar structure that is the first cortical destination of olfactory information, receiving monosynaptic input from the olfactory bulb. Here, we show that the main input layer of the piriform cortex, layer II, is dominated by two classes of principal neurons, superficial pyramidal (SP) and semilunar (SL) cells, with strikingly different properties. Action potentials in SP cells are followed by a Ni2+-sensitive afterdepolarization that promotes burst firing, whereas SL cells fire nonbursting action potentials that are followed by a powerful afterhyperpolarization. Synaptic inputs from the olfactory bulb onto SP cells exhibit prominent paired-pulse facilitation, which is attributable to residual presynaptic Ca2+ and a low probability of neurotransmitter release. In contrast, the same inputs onto SL cells do not facilitate. These distinctive synaptic and firing properties cause SP and SL cells to respond differently to in vivo-like bursts of afferent stimulation: SP cells tend to fire bursts of output action potentials at a higher frequency than the input, whereas SL cells tend to fire at a lower frequency than the input. When connected together in the canonical circuit of the piriform cortex, SP and SL cells transform the pattern of synaptic inputs they receive from the olfactory bulb, dispersing the firing rate and latency of output action potentials to an extent that depends on the strength of the input. Thus, the presence of two types of principal cells in layer II of the piriform cortex may underlie coding strategies used for the representation of odors.

Galanin is a widely-distributed neuropeptide that acts as an endogenous anticonvulsant. We have recently generated a galanin receptor type 1 knockout mouse (Galr1(-/-)) that develops spontaneous seizures. Our aim here was to characterize the seizures by making electroencephalogram (EEG) recordings from this animal, and also to elucidate the cellular basis of its epileptic phenotype by studying the neurophysiology of CA1 pyramidal neurons in acute hippocampal slices. EEGs showed that major seizures had a partial onset with secondary generalization, and that paroxysms of spike-and-slow waves occurred and were associated with hypoactivity. The interictal EEG was also abnormal, with a marked excess of spike-and-slow waves. Slice experiments showed that resting potential, input resistance, intrinsic excitability, paired-pulse facilitation of excitatory and inhibitory postsynaptic currents (EPSCs and IPSCs), stimulus--response plots for EPSCs, and several properties of spontaneous miniature EPSCs and IPSCs were all unchanged in the mutant mouse compared with wildtype. However, the frequency of miniature IPSCs was significantly reduced in the mutants. These results suggest that impaired synaptic inhibition in the hippocampus may contribute to the local onset of seizures in the Galr1(-/-) mouse.

Mammalian central synapses commonly specialize in one fast neurotransmitter, matching the content of their presynaptic vesicles with the appropriate receptors in their postsynaptic membrane. Here, I show that hippocampal cultures contain autaptic glutamatergic synapses that contravene this rule: in addition to postsynaptic glutamate receptors, they also express clusters of functional postsynaptic GABA(A) receptors yet lack presynaptic GABA. Hence, these synapses are presynaptically silent with respect to GABA. They can be unsilenced by loading GABA into presynaptic vesicles by endocytosis, after which a postload IPSC appears. This IPSC is similar to native IPSCs recorded from GABAergic interneurons in the same cultures. Thus, these "mistargeted" GABA(A) receptors, which apparently lack a signal that confers synaptic specificity, function almost normally. After GABA loading, glutamatergic miniature postsynaptic currents acquire a slow tail that is mediated by GABA(A) receptors, showing that synaptic vesicles can accommodate both the usual concentration of native glutamate and a saturating concentration of loaded GABA. After brief Ca(2+)-dependent exocytosis, endocytosis of GABA can proceed in low-Ca(2+) external solution. The amplitude of the postload IPSC declines exponentially with repetitive stimulation as the endocytosed GABA passes through the presynaptic vesicle cycle and is depleted. Hence, by using GABA as an exogenous but physiological tracer, the properties of these presynaptically silent synapses can provide novel insights into the content and cycling of vesicles in presynaptic terminals.

According to a basic formulation of the quantal model, evoked synaptic currents are made up of a linear summation of uniquantal synaptic currents, which in turn are equivalent to the spontaneous miniature synaptic currents ('minis') that often persist when evoked neurotransmitter release is blocked. Here I describe a convolution method for calculating linear summations of the 'mini' amplitude distribution, which can then be fitted to the measured amplitude distribution for evoked synaptic currents. Provided certain conditions are satisfied, this method can give information about the statistics of neurotransmitter release even when clear quantal peaks are not apparent in the evoked amplitude distribution. The method is illustrated by an experiment in which the appropriate minis are identified with the asynchronous excitatory postsynaptic currents that follow synaptic stimulation when the cell is bathed in strontium. Finally, I discuss the assumptions behind the convolution method, and the conditions under which the properties of the minis are likely to be appropriate for an analysis of this type.

A key step in the release of neurotransmitter is the entry of Ca(2+) into the presynaptic terminal via voltage-activated Ca(2+) channels. N-type and P/Q-type Ca(2+) channels play a predominant role but, surprisingly, their distribution across presynaptic terminals lacks any apparent order. They form a patchwork: at some terminals only N-type channels contribute to transmitter release and in others only P/Q-type channels contribute, but in many terminals both sub-types are active. The physiological implications of this non-uniform distribution are starting to emerge. Recent studies reveal that G-protein-mediated depression of N-type channels is stronger than that of P/Q-type channels, whereas voltage-dependent relief of inhibition is more pronounced for P/Q-type channels. The patchwork distribution of Ca(2+) channel subtypes might therefore enable terminal-specific modulation of transmitter release, enhancing the power of synaptic processing.

Autapses--synapses made by a neuron onto itself--have often been observed anatomically in vivo, but their purpose has remained uncertain. A recent paper describes functional autapses in the cerebral cortex and gives clues to their physiological role.

Periglomerular cells in the olfactory bulb are the 'gatekeepers' of the olfactory system. A recent study shows that these cells inhibit themselves by releasing GABA from their own dendrites.

Many neurons transduce synaptic inputs into action potentials (APs) according to rules that reflect their intrinsic membrane properties. Voltage-gated potassium channels, being numerous and diverse constituents of neuronal membrane, are important participants in neuronal excitability and thus in synaptic integration. Here we address the role of dendrotoxin-sensitive "D-type" potassium channels in the excitability of large pyramidal neurons in layer 5 of the rat neocortex. Low concentrations of 4-aminopyridine or alpha-dendrotoxin (alpha-DTX) dramatically increased excitability: the firing threshold for action potentials was hyperpolarized by 4-8 mV, and the firing frequency during a 1-sec-long 500 pA somatic current step was doubled. In nucleated outside-out patches pulled from the soma, alpha-DTX reversibly blocked a slowly inactivating potassium current that comprised approximately 6% of the total. This current first turned on at voltages just hyperpolarized to the threshold for spiking and activated steeply with depolarization. By assaying alpha-DTX-sensitive current in outside-out patches pulled from the axon and primary apical dendrite, it was found that this current was concentrated near the soma. We conclude that alpha-DTX-sensitive channels are present on large layer 5 pyramidal neurons at relatively low density, but their strategic location close to the site of action potential initiation in the axon may ensure that they have a disproportionate effect on neuronal excitability. Modulation of this class of channel would generate a powerful upregulation or downregulation of neuronal output after the integration of synaptic inputs.

The pharmacological effects of opioid- and adenosine-receptor agonists on neural signalling were investigated by measuring drug actions on barium current flowing through calcium channels in acutely-dissociated neurons of the rat nucleus accumbens (NAc). Under whole-cell voltage clamp, opioids acted via mu, but not delta or kappa, receptors to partially inhibit barium current. Mean inhibition was 35+/-2% (+/-s.e.mean, n = 33) for methionine-enkephalin and 37+/-1% (n = 65) for the selective mu receptor agonist DAMGO, both measured at saturating agonist concentrations in neurons with diameter > or = 20 microm. EC(50) for DAMGO was 100 nM. Perfusion of naloxone reversed the current inhibition by DAMGO. Adenosine also partially inhibited barium current in these neurons. Mean inhibition was 28+/-2% (n = 29) for adenosine and 33+/-3% (n = 27) for the selective A1 receptor agonist N(6)CPA, both at saturating concentrations in neurons with diameter > or = 20 microm. EC(50) for N(6)CPA was 34 nM. Adenosine inhibition was reversed by perfusion of an A1 receptor antagonist, 8-cyclopentyl-1,3-dipropylxanthine, while the selective A2A receptor agonist, CGS 21680, had no effect. Inhibition by opioids and adenosine was mutually occlusive, suggesting a converging pathway onto calcium channels. These actions involved a G-protein-coupled mechanism, as demonstrated by the partial relief of inhibition by strong depolarization and by the application of N-ethylmaleimide or GTP-gamma-S. Inhibition of barium current by opioids had their greatest effect in large neurons, that is, in presumed interneurons. In contrast, opioid inhibition in neurons with diameter < or = 15 microm was 11+/-2% (n = 26) for methionine-enkephalin and 11+/-4% (n = 17) for DAMGO, both measured at saturating agonist concentrations. Adenosine inhibition in neurons with diameter < or = 15 microm was 22+/-5% (n = 9). These results implicate the interneurons as a locus for the modulation of the excitability of projection neurons in the NAc during the processes of addiction and withdrawal.

Voltage-gated potassium currents were studied in nucleated outside-out patches obtained from large layer 5 pyramidal neurons in acute slices of sensorimotor cortex from 13- to 15-day-old Wistar rats (22-25 C). Two main types of current were found, an A-current (IA) and a delayed rectifier current (IK), which were blocked by 4-aminopyridine (5 mM) and tetraethylammonium (30 mM), respectively. Recovery from inactivation was mono-exponential (for IA) or bi-exponential (for IK) and strongly voltage dependent. Both IA and IK could be almost fully inactivated by depolarising prepulses of sufficient duration. Steady-state inactivation curves were well fitted by the Boltzmann equation with half-maximal voltage (V ) and slope factor (k) values of -81.6 mV and -6.7 mV for IA, and -66.6 mV and -9.2 mV for IK. Peak activation curves were described by the Boltzmann equation with V and k values of -18.8 mV and 16.6 mV for IA, and -9.6 mV and 13.2 mV for IK. IA inactivated mono-exponentially during a depolarising test pulse, with a time constant ( approximately 7 ms) that was weakly dependent on membrane potential. IK inactivated bi-exponentially with time constants ( approximately 460 ms, approximately 4.2 s) that were also weakly voltage dependent. The time to peak of both IA and IK depended strongly on membrane potential. The kinetics of IA and IK were described by a Hodgkin-Huxley-style equation of the form mNh, where N was 3 for IA and 1 for IK. These results provide a basis for understanding the role of voltage-gated potassium currents in the firing properties of large layer 5 pyramidal neurons of the rat neocortex.

Voltage-gated potassium channels were studied in cell-attached and outside-out patches from the soma and primary apical dendrite of large layer 5 pyramidal neurons in acute slices of rat sensorimotor cortex (22-25 degrees C). Ensemble averages revealed that some patches contained only fast, I(A)-like channels, other contained only I(K)-like channels that did not inactivate or inactivated slowly, and the remainder contained mixtures of both types. I(A) and I(K) channels had mean unitary conductances of 8.5 and 20.3 pS, respectively, and had distinctive patterns of gating. Peak activation curves for ensemble-averaged currents were described by the Boltzmann equation with half-maximal voltage [V(1/2)] and slope factor (k) values of -24.5 mV and 16.9 mV for I(A) and -7.6 mV and 10.1 mV for I(K) (patches < 250 microm from the soma) or -22.9 mV and 16.2 mV for I(A) (patches > 250 microm from the soma). The steady-state inactivation curve for I(A) gave V(1/2) and k values of -72.3 mV and -5.9 mV (< 250 microm from the soma) or -83.1 mV and -6.5 mV (> 250 microm from the soma). These values were similar to the corresponding data for I(A) and I(K) in nucleated patches from the same cell. The amount of I(A) and I(K) present in patches depended weakly on distance along the primary apical dendrite from the soma. The amplitude of I(A) increased, on the average, by 2.3 pA per 100 microm, while the amplitude of I(K) decreased by 0.4 pA per 100 microm. I(A) and I(K) channels in dendritic cell-attached patches were activated by the passage of a back-propagating action potential past the tip of the patch electrode. These results show directly that these potassium channels participate in action potential repolarisation, and thus contribute to the process of synaptic integration in these neurons.

This work was designed to localize the Ca(2+)-activated K(+) channels underlying the slow afterhyperpolarization (sAHP) in hippocampal CA1 pyramidal cells. Cell-attached patches on the proximal 100 microm of the apical dendrite contained K(+) channels, but not sAHP channels, activated by backpropagating action potentials. Amputation of the apical dendrite approximately 30 microm from the soma, while simultaneously recording the sAHP whole cell current at the soma, depressed the sAHP amplitude by only approximately 30% compared with control. Somatic cell-attached and nucleated patches did not contain sAHP current. Amputation of the axon >/=20 microm from the soma had little effect on the amplitude of the sAHP recorded in cortical pyramidal cells. By this process of elimination, it is suggested that sAHP channels may be concentrated in the basal dendrites of CA1 pyramids.

1. The effects of GABA(B), opioid and alpha2 receptor activation on different subtypes of calcium channels in acutely-dissociated rat locus coeruleus (LC) neurones were investigated using whole-cell patch clamping. 2. Barium currents through calcium channels could be fractionated into four classes: L-type (nimodipine-sensitive), N-type (omega-conotoxin GVIA-sensitive), P/Q-type (omega-agatoxin IVA-sensitive) and R-type (remaining in the presence of all three blockers). The percentage of each was, respectively, 25+/-2, 34+/-1, 28+/-3 and 12+/-1% (mean+/-s.e.mean, n=4). 3. The GABA(B) receptor agonist, baclofen, and the opioid receptor agonist, enkephalin, partially inhibited the total barium current in a concentration-dependent manner with EC50 values of 2 and 0.3 microm , respectively. Maximal inhibition was 17+/-1% (n=38) for baclofen and 30+/-2% (n=20) for enkephalin. The alpha2-adrenoceptor agonist, UK14304 (10 microM), also inhibited barium current in these neurones (28+/-2%, n=11). The agonists did not shift the current-voltage relationship along the voltage axis. 4. Maximal baclofen inhibition of different calcium channel subtypes was 9+/-7% (L-type, n=4), 11+/-8% (N-type, n=4), 26+/-6% (P/Q-type, n=4), and 6+/-5% (R-type, n=5). The corresponding values for enkephalin inhibition were 5+/-9% (L-type), 30+/-11% (N-type), 37+/-9% (P/Q-type), and 17+/-8% (R-type). 5. In the presence of a saturating concentration of enkephalin, baclofen produced additional inhibition of the barium current. In contrast, in the presence of a saturating concentration of enkephalin, UK14304 produced no further inhibition of the barium current. 6. These results indicate that neuromodulation of calcium channels in LC neurones involves a complex pattern of overlapping and distinct second messenger pathways. Regulation of LC neuronal firing activity by the modulation of calcium channels may be important for LC-mediated behaviour such as alertness and vigilance.

1. Excitatory postsynaptic currents (EPSCs) were recorded from granule cells of the dentate gyrus in acute slices of 17- to 21-day-old rats (22-25 C) using tissue cuts and minimal extracellular stimulation to selectively activate a small number of synaptic contacts. 2. Adding millimolar Sr2+ to the external solution produced asynchronous EPSCs (aEPSCs) lasting for several hundred milliseconds after the stimulus. Minimally stimulated aEPSCs resembled miniature EPSCs (mEPSCs) recorded in the same cell but differed from them in ways expected from the greater range of dendritic filtering experienced by mEPSCs. aEPSCs had the same stimulus threshold as the synchronous EPSCs (sEPSCs) that followed the stimulus with a brief latency. aEPSCs following stimulation of distal inputs had a slower mean rise time than those following stimulation of proximal inputs. These results suggest that aEPSCs arose from the same synapses that generated sEPSCs. 3. Proximally elicited aEPSCs had a mean amplitude of 6.7 +/- 2.2 pA (+/- s.d., n = 23 cells) at -70 mV and an amplitude coefficient of variation of 0. 46 +/- 0.08. 4. The amplitude distributions of sEPSCs never exhibited distinct peaks. 5. Monte Carlo modelling of the shapes of aEPSC amplitude distributions indicated that our data were best explained by an intrasite model of quantal variance. 6. It is concluded that Sr2+-evoked aEPSCs are uniquantal events arising at synaptic terminals that were recently invaded by an action potential, and so provide direct information about the quantal amplitude and quantal variance at those terminals. The large quantal variance obscures quantization of the amplitudes of evoked sEPSCs at this class of excitatory synapse.

The relationship between extracellular Ca2+ concentration and EPSC amplitude was investigated at excitatory autapses on cultured hippocampal neurons. This relationship was steeply nonlinear, implicating the cooperative involvement of several Ca2+ ions in the release of each vesicle of transmitter. The cooperativity was estimated to be 3.1 using a power function fit and 3.3 using a Hill equation fit. However, simulations suggest that these values underestimate the true cooperativity. The role of different Ca2+ channel subtypes in shaping the Ca2+ dose-response relationship was studied using the selective Ca2+ channel blockers omega-agatoxin GIVA (omega-Aga), which blocks P/Q-type channels, and omega-conotoxin GVIA (omega-CTx), which blocks N-type channels. Both blockers broadened the dose-response relationship, and the Hill coefficient was reduced to 2.5 by omega-Aga and to 2.6 by omega-CTx. This broadening is consistent with a nonuniform distribution of Ca2+ channel subtypes across presynaptic terminals. The similar Hill coefficients in omega-Aga or omega-CTx suggest that there was no difference in the degree of cooperativity for transmitter release mediated via N- or P/Q-type Ca2+ channels. A model of the role of calcium in transmitter release is developed. It is based on a modified Dodge-Rahamimoff equation that includes a nonlinear relationship between extracellular and intracellular Ca2+ concentration, has a cooperativity of 4, and incorporates a nonuniform distribution of Ca2+ channel subtypes across presynaptic terminals. The model predictions are consistent with all of the results reported in this study.

Recent evidence suggests that 'autapses' - synapses made by a neuron with itself - are much more common in the brain than previously thought, leading to the possibility that they are more than just curiosities and may have a real physiological role.

Spontaneous synaptic events can be difficult to detect when their amplitudes are close to the background noise level. Here we report a sensitive new technique for automatic detection of small asynchronous events. A waveform with the time course of a typical synaptic event (a template) is slid along the current or voltage trace and optimally scaled to fit the data at each position. A detection criterion is calculated based on the optimum scaling factor and the quality of the fit. An event is detected when this criterion crosses a threshold level. The algorithm automatically compensates for changes in recording noise. The sensitivity and selectivity of the method were tested using real and simulated data, and the influence of the template parameter settings was investigated. Its performance was comparable to that obtained by visual event detection, and it was more sensitive than previously described threshold detection techniques. Under typical recording conditions, all fast synaptic events with amplitudes of at least three times the noise standard deviation (3 sigma) could be detected, as could 75% of events with amplitudes of 2 sigma. The scaled template technique is implemented within a commercial data analysis application and can be applied to many standard electrophysiological data file formats.

Several subtypes of Ca2+ channel support the release of glutamate at excitatory synapses. We investigated the pattern of colocalization of these subtypes on presynaptic terminals in hippocampal cultures. N-type (conotoxin GVIA-sensitive) or P/Q-type (agatoxin IVA-sensitive) Ca2+ channels were blocked selectively, and the reduction in transmitter release probability (Pr) was measured with MK-801. The antagonists completely blocked release at some terminals, reduced Pr at others, and failed to affect the remainder. In contrast, nonselective reduction of presynaptic Ca2+ influx by adding Cd2+ or lowering external Ca2+ reduced Pr uniformly at all terminals. We conclude from these results that the mixture of N-type and P/Q-type channels varies markedly between terminals on the same afferent. The distribution of Ca2+ channel subtypes was the same for high and low Pr terminals. Given that Ca2+ channel subtypes are affected differentially by neuromodulators, these findings lead to the possibility of terminal-specific modulation of synaptic function.

Trains of action potentials in hippocampal pyramidal neurons are followed by a prolonged afterhyperpolarization (AHP) lasting several seconds, which is attributable to the activation of a slow calcium-activated potassium current ((sI)AHP). Here we examine the location of (sI)AHP on CA1 pyramidal neurons by comparing it with two GABAergic inhibitory postsynaptic currents (IPSCs) with known somatic and dendritic locations. Whole-cell patch-clamp recordings were made for CA1 pyramidal neurons in acute hippocampal slices. Stepping the membrane potential at the peak of (sI)AHP produced a relaxation ("switchoff") of the AHP current with a time constant of 7.4 +/- 0.4 msec (mean +/- SEM). The switchoff time constants for somatic and dendritic GABAA IPSCs were 3.5 +/- 0.5 msec and 8.8 +/- 0.3 msec, respectively. This data, together with cable modeling, indicates that active (sI)AHP channels are distributed over the proximal dendrites within approximately 200 micrometers of the soma. Excitatory postsynaptic potentials (EPSPs) evoked in stratum (s.) radiatum had their amplitudes shunted more by the AHP than did EPSPs evoked in s. oriens, suggesting that active AHP channels are restricted to the apical dendritic tree. Blockade of the AHP during a tetanus, which in control conditions elicited a decremental short-term potentiation (STP), converted STP to long-term potentiation (LTP). Thus, activation of the AHP increases the threshold for induction of LTP. These results suggest that in addition to its established role in spike frequency adaptation, the AHP works as an adjustable gain control, variably hyperpolarizing and shunting synaptic potentials arising in the apical dendrites.

The effect of histamine on N-methyl-D-aspartate currents was investigated in pyramidal neurons in the CA1 region of acute hippocampal slices from juvenile rats. The objective was to compare histamine effects in the slice with those previously reported in acutely dissociated and cultured hippocampal neurons. Micromolar concentrations of histamine had no effect on N-methyl-D-aspartate mediated excitatory postsynaptic currents in the slice, in contrast to the large enhancement seen in culture under identical conditions. However, millimolar concentrations of histamine blocked these currents both in the slice and in culture. Possible reasons for the lack of enhancement in the slice were explored as follows. (1) Histamine could not penetrate the slice or was already present at high concentrations inside the slice. This was tested by recording N-methyl-D-aspartate currents elicited in outside-out patches pulled from the somas of CA1 slice neurons. Histamine still had no effect in patches, whereas the corresponding experiment for cultured neurons showed robust enhancement. (2) Slices release an endogenous ligand that binds with high affinity to the histamine site on the N-methyl-D-aspartate receptor, blocking its activation. This was tested by superfusing cultures with supernatant from homogenized slice tissue. Histamine enhancement was maintained in these cultures. (3) CA1 slices and cultures express different N-methyl-D-aspartate receptor subtypes. The reverse transcription-polymerase chain reaction technique was used to examine the expression of messenger RNA encoding N-methyl-D-aspartate receptor subunits in the two systems. No difference was found in the whole-tissue expression of messenger RNA for the NR2A, 2B or 2C subunits or for the eight known splice variants of the NR1 subunit. It is hypothesized that the differential enhancing effect of histamine in slices and culture involves posttranslational modifications or other factors that modulate the N-methyl-D-aspartate receptor/ion channel according to its environment.

1. The passive cable properties of rat hippocampal neurons in dissociated culture were studied using focal application of hypertonic solution to locally elicit miniature excitatory postsynaptic currents (mEPSCs) on the soma and dendrites. Neurons were filled with Lucifer yellow and portions of their dendritic trees were measured. 2. The average mEPSC measured at the soma appeared smaller and slower as the site of sucrose application was made more distal. Normalizing to a 1-micron diam dendrite, the mean mEPSC peak amplitude and charge was reduced e-fold in 170 and 1,000 microns, respectively, and the mean mEPSC decay time constant was increased e-fold in 150 microns. However, for any particular sucrose site, individual mEPSCs varied widely in their amplitudes and time courses. Plots of individual peak amplitudes versus half-width or rise time showed much overlap for mEPSCs originating from sites as much as 100 microns apart. This suggests that use of such plots to estimate the electrotonic location of synaptic currents is highly prone to error. 3. Averaged mEPSCs recorded when applying sucrose at the soma were poorly fitted by an alpha function but were well-described by an equation of the form mxh, where m incorporates a rise-time constant tau 1 and h a decay time constant tau 2. Averaged fits to mean mEPSCs elicited at the somas of five cells gave (mean +/- SE): peak conductance = 832 +/- 126 pS, tau 1 = 0.29 +/- 0.06 ms, tau 2 = 3.03 +/- 0.24 ms, x = 4.7 +/- 0.7. 4. For three cells, the entire dendritic branch to which sucrose was applied was measured and used to construct a passive cable model. The specific membrane resistance (Rm) and intracellular resistivity (Ri) were varied systematically in the model (assuming membrane capacitance Cm = 1 microF/cm2) to search for the best agreement between the mean mEPSCs and the model. Optimal Rm was found to lie in the range 20-30 k omega cm2, Ri in the range 100-200 omega cm. 5. These results confirm those obtained by other methods and emphasize the considerable cable filtering of fast electrical events in cultured hippocampal neurons.

1. We have studied the statistical properties of excitatory post-synaptic currents (EPSCs) measured at small numbers of synaptic contacts between pairs of hippocampal neurons maintained in dissociated cell culture. Synaptic transmission at few synapses was enabled by microperfusion of a small region of the postsynaptic cell with Ca-containing solution, while blocking transmission at all other synaptic boutons by bathing them in low-Ca solution. Frequency histograms of the amplitudes of EPSCs recorded in this way showed no clear quantization. Numbers of active synapses, estimated immunohistochemically with the use of light microscopy, ranged from 4 to 14 in different experiments. 2. Miniature EPSCs (mEPSCs), originating in the same small population of synapses as produced the evoked EPSCs, were elicited by microperfusion of bath solution made hypertonic by the addition of sucrose. These "sucrose-evoked" mEPSCs appeared to be identical to "spontaneous" mEPSCs in every respect except control over their frequency and site of origin. Sucrose-evoked mEPSCs originating in few synapses still exhibited a broad amplitude distribution. Thus, if mEPSCs constitute the postsynaptic response to a single quantum of neurotransmitter (the "quantal amplitude"), their broad amplitude distribution would tend to obliterate evidence of quantization in evoked EPSC amplitudes, even if evoked release was, indeed, quantal. 3. This idea, which is a corollary of the Katz model of quantal transmission, was tested quantitatively by assuming 1) neurotransmitter release obeys uniform binomial statistics, and 2) the quantal amplitude has a distribution given by the observed distribution of sucrose-evoked mEPSCs. The expected distribution, calculated on the basis of these two assumptions, was fitted to the observed distribution of evoked EPSC amplitudes by varying two free parameters, the binomial parameters N and p. In five cells out of six that were fully analyzed, the Poisson limit of the binomial model (N large, p small) provided a very good fit to the data. This and other evidence suggests that the release probability at a single presynaptic terminal is low. In two out of the six cells, the binomial model, with N constrained to the histochemically determined bouton count, yielded acceptable fits; for the remaining cells the constrained binomial model could be rejected. 4. It is concluded that the Katz model of quantized release of neurotransmitter gives an adequate description of excitatory synaptic transmission in hippocampal cultures, when one assumes the broad distribution of mEPSC amplitudes reflects the distribution of the postsynaptic effect of a single quantum of transmitter.
