An effective and fast vortex-assisted dispersive liquid–liquid extraction method was developed for the extraction of paraben in cosmetic samples and water samples. The paraben was determined and quantified using ultraviolet-visible (UV-Vis) spectrometry. A response surface methodology (RSM) based on the central composite design was used for the optimization of factors (composition of the extractant, volume of extractant, extraction time, centrifugation time, and centrifugation velocity) affecting the extraction efficiency of the procedure. The optimum parameters for vortexassisted dispersive liquid–liquid extraction (VA-DLLE) are: chloroform used as the extractant solvent, 5 ml volume of extractant, 3 min extraction time, 5 min centrifugation time, and 2400 rpm centrifugation velocity. The limit of detection (LOD) and the limit of quantification (LOQ) for paraben are 0.0476 and 0.1442 μg/mL, respectively. Spiked cosmetic samples have the extraction recoveries in the range of 81.2–96.8%, whereas spiked water sample extraction recoveries were in the range of 88.8–100.63%. Each sample was repeated (n = 2), with a relative standard deviation of <5.74% for cosmetic samples and <9.03% for water samples. Obesity poses significant public health concerns, including Type 2 diabetes mellitus, cardiovascular disease and stroke. Although deleterious metabolic profiles typically accompany individuals with obesity, various epidemiological studies have identified a subset of patients with a favourable metabolic phenotype referred to as the metabolically healthy obese. Metabolically healthy obesity is a novel concept that stratifies obese individuals according to their respective metabolic status. It has important implications for healthcare policies, particularly the efficient allocation of resources in the targeted treatment of obesity and prevention of metabolic ill‐health. However, conflicting evidence in the literature regarding its risk profile questions its clinical relevance. In addition, the lack of a unified definition of metabolically healthy obesity and agreement on its progression further impede its utility as a stratification strategy. This review aims to describe current concepts of metabolically healthy obesity within the literature, evidence of this metabolic phenotype, the clinical implications of patient stratification and limitations of the concept. Introduction: The present study aimed to assess the demographic, socioeconomic, medical and lifestyle factors associated with the progression of a threatened miscarriage to a complete miscarriage in the first trimester.Methods: A prospective cohort study was conducted on 157 women who presented with vaginal bleeding in the fifth to tenth week of gestation. Cox regression analysis was used to determine the risk factors for progression to a complete miscarriage within 16 weeks of gestation.Results: Of the 139 women included for data analysis, 36 (25.9%) had a miscarriage, mostly within two weeks of presentation. The results of our study showed that women aged ≥ 34 years were more likely to miscarry (hazard ratio [HR] = 1.95). Compared to women whose partner was 20-30 years of age, women whose partner was ≥ 41 years of age also had a higher likelihood of experiencing a miscarriage (HR = 8.33). However, the presence of nausea (HR = 0.33) and a high stress score (i.e. ≥ 17) on the Perceived Stress Scale (HR = 0.49) were associated with a reduced likelihood of miscarriage.Conclusion: Older pregnant women experiencing a threatened miscarriage should be counselled about their higher risk of miscarriage, especially if they have an older partner. Mitochondrial electron transport chain (ETC) complexes partition between free complexes and quaternary assemblies known as supercomplexes (SCs). However, the physiological requirement for SCs and the mechanisms regulating their formation remain controversial. Here, we show that genetic perturbations in mammalian ETC complex III (CIII) biogenesis stimulate the formation of a specialized extra-large SC (SC-XL) with a structure of I 2 +III 2 , resolved at 3.7 Å by cryoelectron microscopy (cryo-EM). SC-XL formation increases mitochondrial cristae density, reduces CIII reactive oxygen species (ROS), and sustains normal respiration despite a 70% reduction in CIII activity, effectively rescuing CIII deficiency. Consequently, inhibiting SC-XL formation in CIII mutants using the Uqcrc1 DEL:E258-D260 contact site mutation leads to respiratory decompensation. Lastly, SC-XL formation promotes fatty acid oxidation (FAO) and protects against ischemic heart failure in mice. Amygdala activation by emotional arousal during memory formation can prioritize events for long-term memory. Building upon our prior demonstration that brief electrical stimulation to the human amygdala reliably improved long-term recognition memory for images of neutral objects without eliciting an emotional response, our study aims to explore and describe individual differences and stimulation-related factors in amygdala-mediated memory modulation. Thirty-one patients undergoing intracranial monitoring for intractable epilepsy were shown neutral object images paired with direct amygdala stimulation during encoding with recognition memory tested immediately and one day later. Adding to our prior sample, we found an overall memory enhancement effect without subjective emotional arousal at the one-day delay, but not at the immediate delay, for previously stimulated objects compared to not stimulated objects. Importantly, we observed a larger variation in performance across this larger sample than our initial sample, including some participants who showed a memory impairment for stimulated objects. Of the explored individual differences, the factor that most accounted for variability in memory modulation was each participant's pre-operative memory performance. Worse memory performance on standardized neuropsychological tests was associated with a stronger susceptibility to memory modulation in a positive or negative direction. Sex differences and the frequency of interictal epileptiform discharges (IEDs) during testing also accounted for some variance in amygdala-mediated memory modulation. Given the potential and challenges of this memory modulation approach, we discuss additional individual and stimulation factors that we hope will differentiate between memory enhancement and impairment to further optimize the potential of amygdala-mediated memory enhancement for therapeutic interventions., Competing Interests: Declarations. Objective: The current study sought to characterize the relationship of the N400 (N4) effect event-related potential to Alzheimer's disease (AD) biomarkers and broader cognition in older adults on the late-life cognitive continuum., Method: Participants who were cognitively intact (n = 43), or had amnestic mild cognitive impairment (MCI; n = 19), or mild AD (n = 12), completed a word-pair judgement task during concurrent EEG recording to elicit the N400. The Repeatable Battery for the Assessment of Neuropsychological Status (RBANS) and biomarker data (PET-imaged beta-amyloid (aβ) deposition, apolipoprotein-E ε4 (APOE4) allele status, hippocampal volumes) were collected as part of a larger study., Results: The AD group had slower response times and poorer accuracy on the word-pair judgement task than the intact group. The N4 effect was smaller and occurred later in AD relative to intact participants. MCI participants' values were intermediate. N4 effect amplitudes were not associated with RBANS scores but were positively associated with aβ deposition. Conversely, poorer performance across most RBANS Indexes and the Total score was associated with longer N4 latencies. There was also a negative association between hippocampal volumes and the N4 latency and a positive association between aβ deposition and latency. Callous-unemotional (CU) traits are associated with severe and persistent juvenile offending. CU traits are also associated with dampened emotional arousal, which suggests that fundamental frequency (f 0 ), a measure of vocally-encoded emotional arousal, may serve as an accessible psychophysiological marker of CU traits in youth. This study investigated the associations between f 0 range measured during an emotionally evocative task, CU traits, and emotion dysregulation in a mixed-gender sample of 168 justice-involved youth. For boys, after controlling for covariates, wider f 0 range-indicating greater emotional arousal-was negatively associated with CU traits and positively associated with emotion dysregulation. For girls, no significant associations with f 0 range emerged; however, CU traits were positively associated with emotion dysregulation. Findings suggest that f 0 range may serve as a valid indicator of CU traits in JJ-involved boys, and that detained boys and girls with high CU traits are characterized by different profiles of emotion dysregulation., (© 2022. Background: In pathology and other specialties of diagnostic medicine, longitudinal studies and competency assessments often involve physicians interpreting the same images multiple times. In these designs, a washout period is used to reduce the chances that later interpretations are influenced by prior exposure., Objective/s: The present study examines whether a washout period between 9-39 months is sufficient to prevent three effects of prior exposure when pathologists review digital breast tissue biopsies and render diagnostic decisions: faster case review durations, higher confidence, and lower perceived difficulty., Methods: In a longitudinal breast pathology study, 48 resident pathologists reviewed a mix of five novel and five repeated digital whole slide images during Phase 2, occurring 9-39 months after an initial Phase 1 review. Importantly, cases that were repeated for some participants in Phase 2 were novel for other participants in Phase 2. We statistically tested for differences in participants' case review duration, self-reported confidence, and self-reported difficulty in Phase 2 based on whether the case was novel or repeated., Results: No statistically significant difference in review time, confidence, or difficulty as a function of whether the case was repeated or novel in a Phase 2 review occurring 9-39 months after initial viewing; this same result was found in a subset of participants with a shorter (9-14-month) washout., Conclusion: These results provide evidence to support the efficacy of at least a 9-month washout period in the design of longitudinal medical imaging and informatics studies to ensure no detectable effect of initial exposure on participant's subsequent case review., Competing Interests: Competing interests: The authors declare that they have no competing interests. more...

Despite recent applications of a developmental psychopathology perspective to the perinatal period, these conceptualizations have largely ignored the role that childbirth plays in the perinatal transition. Thus, we present a conceptual model of childbirth as a bridge between prenatal and postnatal health. We argue that biopsychosocial factors during pregnancy influence postnatal health trajectories both directly and indirectly through childbirth experiences, and we focus our review on those indirect effects. In order to frame our model within a developmental psychopathology lens, we first describe "typical" biopsychosocial aspects of pregnancy and childbirth. Then, we explore ways in which these processes may deviate from the norm to result in adverse or traumatic childbirth experiences. We briefly describe early postnatal health trajectories that may follow from these birth experiences, including those which are adaptive despite traumatic childbirth, and we conclude with implications for research and clinical practice. We intend for our model to illuminate the importance of including childbirth in multilevel perinatal research. FMRI studies of autobiographical memory (AM) retrieval typically ask subjects to retrieve memories silently to avoid speech-related motion artifacts. Recently, some fMRI studies have started to use overt (spoken) retrieval to probe moment-to-moment retrieved content. However, the extent to which the overt retrieval method alters fMRI activations during retrieval is unknown. Here we examined this question by eliciting unrehearsed AMs during fMRI scanning either overtly or silently, in the same subjects, in different runs. Differences between retrieval modality (silent vs. narrated) included greater activation for silent retrieval in the anterior hippocampus, left angular gyrus, PCC, and superior PFC, and greater activation for narrated retrieval in speech production regions, posterior hippocampus, and the DLPFC. To probe temporal dynamics, we divided each retrieval period into an initial search phase and a later elaboration phase. The activations during the search and elaboration phases were broadly similar regardless of modality, and these activations were in line with previous fMRI studies of AM temporal dynamics employing silent retrieval. For both retrieval modalities, search activated the hippocampus, mPFC, ACC, and PCC, and elaboration activated the left DLPFC and middle temporal gyri. To examine content-specific reactivation during retrieval, the timecourse of narrated memory content was transcribed and modeled. We observed dynamic activation associated with object content in the lateral occipital complex, and activation associated with scene content in the retrosplenial cortex. The current findings show that both silent and narrated AMs activate a broadly similar memory network, with some key differences, and add to current knowledge regarding the content-specific dynamics of AM retrieval. Onset of non-suicidal self-injury (NSSI) is most frequent during adolescence. Etiological models indicate that abnormal affective reactivity and regulation within interpersonal contexts is related to heightened NSSI risk. The current study examined the effects of maternal emotional support on adolescent sympathetic nervous system (SNS) activity and observed anger during a conflict discussion among 56 mother-daughter dyads consisting of healthy adolescents and adolescents with a history of self-injury. During the conflict discussion task, maternal emotional support and adolescent anger were coded from behavior, and cardiovascular pre-ejection period was used to index SNS responding. Results demonstrated that maternal emotional support was negatively associated with adolescent anger and SNS activity during the conflict. However, these associations were not significant among adolescents with heightened NSSI history. Maternal emotional support may serve as an interpersonal mechanism for adolescent physiological and behavioral regulation, yet may function differently among adolescents with more frequent NSSI., (© 2022. In 2018, half of refugees worldwide were under the age of 18. Although often resilient, refugee youth experience chronic and pervasive traumatic events prior to and during migration, and are at risk for continued victimization and stressors while in their host country. In particular, refugee youth encounter ongoing discrimination during resettlement, a primary stressor that reduces their wellbeing and adaptation. Given the relevance of discrimination for refugee youth, the goal of the current review is to increase our understanding of the dimensions of discrimination faced by refugee youth and the risk and resilience processes that may be involved in these negative experiences. Thus, this review presents the existing theoretical and empirical research on this topic and creates a model that organizes these findings. Nucleotide-binding oligomerization domain (NBD), leucine-rich repeat (LRR) containing protein family (NLRs) are intracellular pattern recognition receptors that mediate innate immunity against infections. The endothelium is the first line of defense against blood-borne pathogens, but it is unclear which NLRs control endothelial cell (EC) intrinsic immunity. Here, we demonstrate that human ECs simultaneously activate NLRP1 and CARD8 inflammasomes in response to DPP8/9 inhibitor Val-boro-Pro (VbP). Enterovirus Coxsackie virus B3 (CVB3)-the most common cause of viral myocarditis-predominantly activates CARD8 in ECs in a manner that requires viral 2A and 3C protease cleavage at CARD8 p.G38 and proteasome function. Genetic deletion of CARD8 in ECs and human embryonic stem cell-derived cardiomyocytes (HCMs) attenuates CVB3-induced pyroptosis, inflammation, and viral propagation. Furthermore, using a stratified endothelial-cardiomyocyte co-culture system, we demonstrate that deleting CARD8 in ECs reduces CVB3 infection of the underlying cardiomyocytes.
