We present the requirements, design, and evaluation of the cryogenic continuously rotating half-wave plate (CHWP) for the Simons Observatory (SO). SO is a cosmic microwave background polarization experiment at Parque Astronómico de Atacama in northern Chile that covers a wide range of angular scales using both small (⌀0.42 m) and large (⌀6 m) aperture telescopes. In particular, the small aperture telescopes (SATs) focus on large angular scales for primordial B-mode polarization. To this end, the SATs employ a CHWP to modulate the polarization of the incident light at 8 Hz, suppressing atmospheric 1/f noise and mitigating systematic uncertainties that would otherwise arise due to the differential response of detectors sensitive to orthogonal polarizations. The CHWP consists of a 505 mm diameter achromatic sapphire HWP and a cryogenic rotation mechanism, both of which are cooled down to ∼50 K to reduce detector thermal loading. Under normal operation, the HWP is suspended by a superconducting magnetic bearing and rotates with a constant 2 Hz frequency, controlled by an electromagnetic synchronous motor. We find that the number of superconductors and the number of magnets that make up the superconducting magnetic bearing are important design parameters, especially for the rotation mechanism's vibration performance. The rotation angle is detected through an angular encoder with a noise level of 0.07 μrad s. During a cooldown process, the rotor is held in place by a grip-and-release mechanism that serves as both an alignment device and a thermal path. In this paper, we provide an overview of the SO SAT CHWP: its requirements, hardware design, and laboratory performance.

Our aim is to describe a possible class of nonpathological spontaneous movements that has so far received little attention in the scientific literature. These movements arise spontaneously without an underlying pathology such as Huntington's, Parkinson's, cerebral palsy or spinal cord injury. The movements arise in many different contexts including therapeutic, social, religious, and solitary settings. Anecdotal evidence suggests that the movements are related to development and maintenance of form, being part of inherited autoregulatory behaviors and hence bringing an overlooked therapeutic potential. We describe contexts in which they occur, illustrate with case reports, and characterize the movements in terms of their various triggers, movement phenotypes, and conscious and subconscious influences that can occur at both the individual level as well as during collaborative movement relationships between patient and therapist. Background: Nunavut, part of Inuit Nunangat, is a geographically vast territory in northern Canada, with a population of over 38,000 people. Most (85%) of the population identify as Inuit. Nunavut has experienced a significant rise in heterosexual infectious syphilis cases since 2012. Management of communicable diseases, including syphilis, is challenging due to high staff turnover and long delays in specimen transport times. Social determinants of health are also an important contributor. The aim of this study is to describe the epidemiology and program elements for infectious syphilis from 2012-2020 and to highlight beneficial interventions., Methods: Syphilis is a notifiable disease in Nunavut with all cases reported to the Territorial Department of Health. Cases were staged by a medical consultant. Data were analyzed and released in public reports as part of the public health program., Results: From 2012 to 2020, 655 infectious syphilis cases were reported, with 53% of reported cases among females. Infection rates were highest in 20 to 39-year-olds. There was significant variability in reported cases over this time period by geographic region, with the majority of infectious cases reported from the Kivalliq region. Despite 48 reported cases in pregnancy, no confirmed congenital syphilis cases were identified. Program staff identified strengths of the response as well as ongoing needs, such as plain language resources available in multiple languages., Conclusion: Despite the logistical challenges with syphilis management in the territory, the overall outcomes have been positive, with no confirmed congenital cases identified. We attribute this to a coordinated effort by multiple partners including key actions by public health nurses and community health representatives., Competing Interests: Competing interests: None.

Bladder dysfunction is widespread following traumatic spinal cord injury (TSCI). Early diagnosis of bladder dysfunction is crucial in preventing complications, determining prognosis, and planning rehabilitation. We aim to suggest the first clinical protocol specifically designed to evaluate and manage bladder dysfunction in TSCI patients during acute care. A retrospective cohort study was conducted on 101 patients admitted for an acute TSCI between C1 and T12. Following spinal surgery, presence of voluntary anal contraction (VAC) was used as a criterion for removal of indwelling catheter and initiating trial of void (TOV). Absence of bladder dysfunction was determined from three consecutive post-void bladder scan residuals ≤200 mL without incontinence. All patients were reassessed 3 months post-injury using the Spinal Cord Independence Measure (SCIM). A total of 74.3% were diagnosed with bladder dysfunction during acute care, while 57.4% had a motor-complete TSCI. Three months later, 94.7% of them reported impaired bladder function. None of the patients discharged from acute care after a functional bladder was diagnosed reported impaired bladder function at the 3-month follow-up. A total of 95.7% patients without VAC had persisting impaired bladder function at follow-up. The proposed protocol is specifically adapted to the dynamic nature of neurogenic bladder function following TSCI. The assessment of VAC into the protocol provides major insight on the potential for reaching adequate bladder function during the subacute phase. Conducting TOV using bladder scan residuals in patients with VAC is a non-invasive and easy method to discriminate between a functional and an impaired bladder following acute TSCI.

Synthetic derivatization of hormonally active brassinosteroids (BRs) can provide useful small molecule tools to probe BR signaling pathways, such as fluorescent analogs. However, most biologically active BRs are not suitable for direct chemical conjugation techniques because their derivatization typically requires extensive synthetic work and chemistry expertise. Here, we describe an operationally simple, two-step procedure to prepare and purify an Alexa Fluor 647-castasterone (AFCS) from commercially available materials. The reported strategy is also amenable to the introduction of various other amine-based labeling groups.

Long, multifunctional sequence-defined oligomers were obtained on solid support from a protecting-group-free two-step iterative protocol, based on the inherent reactivity of a readily available molecule containing an isocyanate and a thiolactone. Aminolysis of the latter entity with an amino alcohol liberates a thiol that reacts with an acrylate or acrylamide, present in the same medium. Subsequently, a new thiolactone can be reinstated by means of an α-isocyanato-γ-thiolactone. Different acrylic compounds were used to incorporate diverse functionalities in the oligomers, which were built up to the level of decamers. The reaction conditions were closely monitored in order to fine-tune the applied strategy as well as facilitate the translation to an automated protocol.

The Arabidopsis thaliana endogenous elicitor peptides (AtPeps) are released into the apoplast after cellular damage caused by pathogens or wounding to induce innate immunity by direct binding to the membrane-localized leucine-rich repeat receptor kinases, PEP RECEPTOR1 (PEPR1) and PEPR2. Although the PEPR-mediated signaling components and responses have been studied extensively, the contributions of the subcellular localization and dynamics of the active PEPRs remain largely unknown. We used live-cell imaging of the fluorescently labeled and bioactive pep1 to visualize the intracellular behavior of the PEPRs in the Arabidopsis root meristem. We found that AtPep1 decorated the plasma membrane (PM) in a receptor-dependent manner and cointernalized with PEPRs. Trafficking of the AtPep1-PEPR1 complexes to the vacuole required neither the trans-Golgi network/early endosome (TGN/EE)-localized vacuolar H(+)-ATPase activity nor the function of the brefeldin A-sensitive ADP-ribosylation factor-guanine exchange factors (ARF-GEFs). In addition, AtPep1 and different TGN/EE markers colocalized only rarely, implying that the intracellular route of this receptor-ligand pair is largely independent of the TGN/EE. Inducible overexpression of the Arabidopsis clathrin coat disassembly factor, Auxilin2, which inhibits clathrin-mediated endocytosis (CME), impaired the AtPep1-PEPR1 internalization and compromised AtPep1-mediated responses. The predictable 3D structure of double-stranded DNA renders it ideally suited as a template for the bottom-up design of functionalized nucleic acid-based active sites. We here explore the use of a 14mer DNA duplex as a scaffold for the precise and predictable positioning of catalytic functionalities. Given the ubiquitous participation of the histidine-based imidazole group in protein recognition and catalysis events, single histidine-like modified duplexes were investigated. Tethering histamine to the C5 of the thymine base via an amide bond, allows the flexible positioning of the imidazole function in the major groove. The mutual interactions between the imidazole and the duplex and its influence on the imidazolium pKaH are investigated by placing a single modified thymine at four different positions in the center of the 14mer double helix. Using NMR and unrestrained molecular dynamics, a structural motif involving the formation of a hydrogen bond between the imidazole and the Hoogsteen side of the guanine bases of two neighboring GC base pairs is established. The motif contributes to a stabilization against thermal melting of 6°C and is key in modulating the pKaH of the imidazolium group. Functionalized oligonucleotides have recently gained increased attention for incorporation in modified nucleic acid structures both for the design of aptamers with enhanced binding properties as well as the construction of catalytic DNA and RNA. As a shortcut alternative to the incorporation of multiple modified residues, each bearing one extra functional group, we present here a straightforward method for direct linking of functionalized amino acids to the nucleoside base, thus equipping the nucleoside with two extra functionalities at once. As a proof of principle, we have introduced three amino acids with functional groups frequently used as key-intermediates in DNA- and RNAzymes via an efficient and straightforward domino carboxamidation reaction.

A rapid and efficient total synthesis is reported for the cyclic lipodepsipeptide pseudodesmin A. This member of the Pseudomonas viscosin group is active against Gram-positive bacteria and features self-assembling properties. A conserved serine residue within the lactone macrocycle is exploited for initial immobilization on 2-chlorotrityl chloride resin through ether formation with the side-chain alcohol. Subsequent elongation proceeds through Fmoc solid-phase peptide synthesis, including automated incorporation of the enantioselectively synthesized (R)-3-hydroxydecanoic acid lipid tail. Following esterification to generate the incipient lactone bond, the macrocycle is formed by on-resin head-to-tail macrolactamization and cleaved from the resin to give the desired compound in good purity. The short and efficient synthesis route allows rapid generation of analogues by facile variation of both the peptide and lipid moieties with good control of epimerization while maximizing automation. Synthesis of the pseudodesmin A enantiomer yields identical self-assembly and biological activity to that observed for the natural compound, showing that activity is not mediated by chiral interactions. A D-Asn8 analogue developed en route retains self-assembly, but loses activity. The synthesis strategy should be generally applicable for the rapid generation of analogues from various cyclic lipodepsipeptide groups, allowing an investigation of their self-assembling properties and structure-activity relationships., (© 2014 WILEY-VCH Verlag GmbH & Co. Receptor-mediated endocytosis is an integral part of signal transduction as it mediates signal attenuation and provides spatial and temporal dimensions to signaling events. One of the best-studied leucine-rich repeat receptor-like kinases in plants, BRASSINOSTEROID INSENSITIVE 1 (BRI1), perceives its ligand, the brassinosteroid (BR) hormone, at the cell surface and is constitutively endocytosed. However, the importance of endocytosis for BR signaling remains unclear. Here we developed a bioactive, fluorescent BR analog, Alexa Fluor 647-castasterone (AFCS), and visualized the endocytosis of BRI1-AFCS complexes in living Arabidopsis thaliana cells. Impairment of endocytosis dependent on clathrin and the guanine nucleotide exchange factor for ARF GTPases (ARF-GEF) GNOM enhanced BR signaling by retaining active BRI1-ligand complexes at the plasma membrane. Increasing the trans-Golgi network/early endosome pool of BRI1-BR complexes did not affect BR signaling. Our findings provide what is to our knowledge the first visualization of receptor-ligand complexes in plants and reveal clathrin- and ARF-GEF-dependent endocytic regulation of BR signaling from the plasma membrane.

Muscle hexokinase was ascertained in the gastrocnemius muscles of 160 male chicks. Observations were made in four age groups and two diets. No differences in enzyme activity were observed which could be attributed to the feeding of either the high-carbohydrate or high-fat diet. Hexokinase activity declined significantly between 6 and 12 days of age in the chicks fed the respective diets.

Two experiments were conducted to investigate the relationship of energy source, age and glucose-6-phosphatase activity in the chick. In the first experiment liver glycogen and glucose-6-phosphatase were assayed in chicks of four age groups being fed either a high-fat or high-carbohydrate diet. It was observed that the feeding of the high-fat diet resulted in higher glucose-6-phosphatase activity and lower glycogen levels than did the feeding of the carbohydrate diet. Age was found to significantly influence enzyme activity. Both glucose-6-phosphatase and liver glycogen were found to be negatively correlated in the fat fed chicks. In the second experiment chicks were fed either a high-fat or high-carbohydrate diet for 28 days. Liver glucose-6-phosphatase activity was determined in representative samples of chicks from each group to establish basal levels. The diets were then reversed for a three-day period, after which the chicks were returned to their initial diets. Glucose-6-phosphatase was analysed at each step. Variation in enzyme activity in the chicks initially fed the high-fat diet could be attributed to differences in body weight. However, those chicks originally fed the carbohydrate diet did exhibit a true metabolic adaption when fed the high-fat diet.

Two experiments were conducted to investigate the response of two populations of chickens, divergently selected for high and low oxygen consumption, to varying protein levels. Protein levels used for Experiments 1 and 2 were 16, 18, 20, and 22% and 20, 22, 24, and 26%, respectively. The birds were weighed initially and weekly to 4 weeks of age. Feed consumption was ad libitum with amounts consumed recorded weekly. Gross feed efficiency was computed per pen as the difference between initial and weight at 4 weeks divided by feed consumed adjusted for spillage. The results from these experiments indicate that at low protein levels both lines were equally efficient, but, as the level of protein was increased, the low line was increasingly more efficient while the high line became less efficient. Gain for the low O2 birds was greater than the high O2 birds in all cases with both lines increasing in gains equally with increased protein levels.

Blood glucose levels and skeletal muscle glycogen concentrations were observed in 160 male chicks. Observations were made in four age groups and two diets. Carbohydrate diets resulted in higher blood glucose levels and glycogen concentrations than did fat diets. Age was found to significantly influence glycogen concentration in both diets. The linear regression of skeletal glycogen on blood glucose levels was significant in carbohydrate fed chicks.

This experiment involving 288 Coturnix quail was conducted to determine the effects of various fiber sources (alfalfa, wheat bran, dried brewer's grain, cellulose, and pectin) on serum, liver and egg yolk cholesterol, and egg production. The fiber sources were added at a rate to provide 6.25% fiber to a corn-soybean meal diet and fed for a 28-day experimental period. Serum, liver, and egg cholesterol levels were measured as pen composite samples at the end of the experimental period. No difference was exhibited in egg yolk cholesterol among any of the groups (P greater than .10). Liver and serum cholesterol levels were elevated in the birds fed the pectin and wheat brain diets as compared with that in birds fed the other fiber source; also, metabolizable energy intakes and eggs per hen day (EHD) were decreased for both groups. When data were adjusted for EHD by covariance analysis, the treatment differences no longer appeared (P greater than .05). These results indicate that 1) there is a basal quantity of cholesterol deposited in the egg on which fiber intake, energy consumed, or egg production have very little effect and that 2) there is an inverse relationship between serum and tissue cholesterol levels and the total quantity of cholesterol excreted via the egg.

Two experiments were conducted to investigate the effects of varying levels of pantothenic acid on the efficiency of energy utilization and changes in body composition of the growing chick. The results indicate that a pantothenic acid deficiency does not interfere with the chick's ability to obtain metabolizable energy from the diet. The data do show that the efficiency of energy utilization and body composition were markedly affected when chicks were fed diets deficient in pantothenic acid. In both experiments, protein, fat and energy stores were significantly reduced when pantothenic acid-deficient diets were fed. Increased estimates of heat increment per gram of diet were observed when levels of pantothenic acid below the N.R.C. (1971) recommended level of 10 mg. per kg of diet were fed.
