Abstract Background Development of the cow-calf bond post-partum and passive immunity of calves from spring-calving beef × beef (B×B) and beef × dairy (B×D) cow genotypes was determined using primiparous and multiparous (Experiment 1), and primiparous and second-parity (Experiment 2) animals. In Experiment 1, calves either suckled colostrum naturally (‘natural-suckling’) (n = 126), or were fed colostrum, using an oesophageal-tube (‘artificially-fed’) (n = 26), from their dam within 1-h post-partum. In Experiment 2, all calves (n = 60) were artificially-fed colostrum from their dam. Prior to colostrum suckling/feeding, colostrum was sampled for IgG analysis. The cow-calf bond was assessed using CCTV recordings during the first 4-h post-partum. Calves were blood sampled at 48-h post-partum to determine IgG and total protein (TP) concentrations, and zinc sulphate turbidity (ZST) units. Results There was no difference (P > 0.05) in cow licking behaviours and calf standing and suckling behaviours between the genotypes, except in Experiment 2 where B×D calves had more attempts to suckle before suckling occurred (P ≤ 0.05) compared to B×B calves. In Experiment 1, multiparous cows licked their calves sooner (P ≤ 0.05) and for longer (P 0.05) between the genotypes in either Experiment. In Experiment 1, colostrum IgG concentrations were greater (P ≤ 0.05) in multiparous compared to primiparous cows and their calves had superior (P ≤ 0.05) passive immunity; no effect of parity was found in Experiment 2. Passive immunity did not differ (P > 0.05) between suckled and artificially-fed calves in Experiment 1. Conclusions Cow genotype had little effect on cow-calf behaviours, but under ‘natural-suckling’ conditions primiparous cows expressed maternal inexperience and their calves were less vigorous than multiparous cows. Colostrum IgG concentration and calf passive immunity measures were unaffected by genotype, but under ‘natural-suckling’ conditions calves from primiparous cows had lower passive immunity.

The components of milk from beef cows remain to be elucidated. This study examined the differences in the antimicrobial components of milk between dairy and beef cows. Quarter milk was collected from both Japanese Black (beef type) and Holstein (dairy type) cows to compare the concentrations of antimicrobial components. The concentration of lingual antimicrobial peptide (LAP) was higher, whereas that of the other antimicrobial components (lactoferrin, S100A7, and S100A8) was lower in beef cows than in dairy cows. Overall, these results indicate that the differences in antimicrobial components between beef and dairy cows may be associated with the difference in the prevalence of mastitis between them. A deep machine learning model, YOLOv8, was developed to detect cow estrus behaviour using video images. The model was trained using Google-Colab on 130 original images from a mobile phone camera based on original images from various scenes. The performance was compared between two models with two different annotation patterns: 1) the mounting cow model (M), involving drawing bounding boxes around cows showing signs of mounting other cows, and 2) the standing cow model (S), involving drawing bounding boxes around cows showing signs of standing estrus and cows showing signs of mounting other cows. The final trained weight of each model was used with the Python program to test the estrus behaviour video data set, using for the first time daytime and night-time videos recorded with an internet-protocol camera installed inside the cow stable area. Results revealed superior performance of the M model, exhibiting higher precision, recall, and F1- score across the YOLOv8n, YOLOv8s and YOLOv8m models when compared to the S model. Notably, the inference speed of the models ranged from 36 to 48 frames per sec., meeting the crucial requirements for fast and accurate detection of cow estrus events through video surveillance by internet-protocol cameras. A computer vision program was developed to detect cow estrus by analysing instances where a cow remained still for over 3 sec. while others attempted mounting. The system, which uses an internet-protocol camera, sends data remotely and allows farmers to monitor their cows online. Our objectives were to validate the accuracy of the published equations using more recent data and to propose alternative prediction models. Criteria for inclusion in the evaluation dataset included projects conducted or published since 2002, direct measurement of feed intake, adequate protein supply, and pen feeding (no metabolism crate data). After removing outliers, the dataset included 53 treatment means for nonlactating cows and 32 treatment means for lactating cows. Means for the nonlactating dataset were dry matter intake (DMI) = 13.2 ± 2.9 kg/d, shrunk body weight (SBW) = 578 ± 83.9 kg, body condition score = 5.7 ± 0.73, and Mcal net energy for maintenance (NEm)/kg of feed = 1.27 ± 0.15 Mcal/kg. The best-fit empirical model accounted for 68% of the variation in daily feed intake with standard error of the estimate Sy root mean squared error = 1.31. Abstract Objectives Animal movement is an important factor in the transmission of infectious diseases among livestock. A better understanding of animal movement characteristics provides a more reliable estimation of disease spread and promotes modeling studies. In Japan, all the cattle movement information is recorded in a national database called the Individual Cattle Identification Register (ICIR).” Our previous studies using this information demonstrated heterogeneity in the movement of dairy and beef cows according to location, season, and age. The present study describes the probability distributions of the movement of Japanese dairy and beef cows in the following month on a regional basis. Data description This publication contains four probability distribution datasets for the predicted locations of dairy and beef cows in Japan in the following month, which were developed using individual cattle movement information obtained from the ICIR. These datasets provide information on cattle movement in the following month on a regional basis, given properties such as birth region, location, time, and age.

Background: The magnitude and knowledge gaps regarding antimicrobial use (AMU) and antimicrobial resistance (AMR) have not been summarized for the North American cow–calf production sector, although estimates of AMU and AMR are essential to AMR risk analysis. The objectives of this scoping review were to map AMU and AMR in the beef cow–calf sector in Canada and the United States, summarize published AMU/AMR predictors, and identify research gaps. Methods: An electronic search was conducted of four bibliographic databases and Google Scholar, augmented by a hand-search of captured studies. Results: Twenty-three of three-hundred and forty-three publications screened advanced to data extraction. Of these, 10 were conducted in the USA and 13 in Canada. Thirteen studied AMR and twelve studied AMU, with two reporting both. Of twelve captured AMU studies, nine presented counts of herd AMU by antimicrobial class or specific antimicrobial. Antimicrobial resistance in Escherichia coli (E. coli) was reported in nine studies. Risk factors for AMU include herd size, vaccine use, and start date of calving season. Conclusions: Overall, a small number of AMR studies were available for synthesis in primarily one population (cows) reporting E. coli AMR. Additional studies targeting reasons for AMU in calves, the impact of management procedures on AMU, potential environmental AMR sources, and AMR in respiratory pathogens and enteric organisms other than E. coli for pre-weaning calves are required to inform AMR risk mitigation strategies. Simple Summary: The decision to provide energy and protein sources to beef cows, offering low- or medium-quality forages, is critical to producers in terms of economics and animal production, particularly during gestation and lactation stages. While corn has been the predominant energy supplement, co-products of corn have increasingly been incorporated as energy supplements in forage-based diets because of lower cost. The objective of this study was to determine the effect of level of hominy feed supplementation on intake, digestibility, ruminal fermentation characteristics, and in situ dry matter disappearance of bermudagrass hay in lactating beef cows. Treatments were low hominy feed fed at 0.25% of body weight, medium hominy feed fed at 0.50% of body weight, low ground corn fed at 0.25% of body weight, medium ground corn fed at 0.50% of body weight, and no supplement (control). Hay dry-matter intake was not affected by supplementation and total dry-matter intake was greater with medium rates of supplementation. Supplementation did not affect ruminal parameters and did not affect dry matter disappearance of forages. Therefore, hominy feed can be used as an alternative feed to corn as an energy supplement without causing negative effects on measurements that are potential indicators of animal performance. Hominy feed (HF) has been evaluated in feedlot and dairy rations but has not been evaluated as a supplemental energy source for lactating beef cows. The objective of this study was to determine the effect of level of HF supplementation on intake, digestibility, ruminal fermentation characteristics, and in situ dry matter (DM) disappearance of bermudagrass hay. Five ruminally cannulated lactating beef cows (body weight (BW) = 596 kg, SE = 13.9) were used in an experiment with a 5 × 5 Latin square design. Treatments were a bermudagrass hay-basal diet with low HF (LH; 0.25% BW), medium HF (MH; 0.50% BW), low ground corn (LC; 0.25% BW), medium ground corn (MC; 0.50% BW) and no supplement (CON). Cows were housed individually, and supplements were offered at 0800 daily. Hay was offered to maintain 10% refusal. Periods were 16–d, with 10 d for adaptation. Ruminal fluid was sampled on d 14 of each period for measurement of pH, volatile fatty acids, and rumen ammonia-N. An in situ degradation experiment for bermudagrass hay was completed for each diet. Hay dry-matter intake (DMI % BW) was not affected (p = 0.14) by supplement, but total DMI (DMI % BW) was greater (p ≤ 0.05) in MH and MC compared to LH and CON. Digestible DMI % BW was greater (p = 0.05) in MH compared to LC, MC was intermediate, and LH and CON were lesser (p ≤ 0.01) than all other diets. Dry-matter fill, passage rate, and retention time did not differ by diet (p ≥ 0.31). A diet × time interaction was observed for ammonia-N (p = 0.0002), and propionate (p = 0.02) time effects were observed for other parameters, but no diet effects. Bermudagrass hay's potentially degradable fraction was greater (p ≤ 0.05) in LH than MH and CON, but effective DM degradability was not different (p = 0.39) among diets. Overall, no diets reduced hay intake or disappearance compared to CON; therefore, no negative associative effects were observed from any of the supplements at the levels offered in this study. At 11 ± 4 d of lactation, 30 multiparous Angus cows [body weight (BW) = 476 ± 17 kg, body condition score (BCS) = 4.5 ± 0.3) were stratified by BW and allotted in 6 pens (5 cows/pen). During 75 d of experiment, all pens received ad libitum low quality hay (Panicum italicum; 54.7% TDN, 6.1% CP DM basis) and were randomly assigned to one of two treatments: 1) PS; cows received ad libitum 30% CP supplement including an intake limiter component (Controlmix, Nutralmix SRL, Argentina), or 2) NS; cows were not supplemented. After the experimental period, dams and their calves (PS: 7 males and 8 females; NS: 8 males and 7 females) were managed on improved pastures as a single group until weaning at 213 d of age. After weaning, heifer and steer calves were reared on improved pastures until 448 d of age, then placed into a feedlot with a finishing diet during 73 d until slaughter. Pens were considered the experimental unit and data were analyzed using the PROC MIXED of SAS, considering treatment and sex as a fixed effects and pen as a random effect. Overage dry matter intake (DMI) of hay was not affected by treatments (P > 0.10), and DMI of supplement in PS cows was 4.4 kgּ·cow-1ּ·d-1. Milk production was greater in PS than NS cows (PS: 5.3, NS: 3.6 kgּ·cow-1ּ·d-1ּ; P < 0.01). Progeny from PS cows increased BW at weaning (PS: 180, NS: 141 kg; P < 0.01), BW at initial feedlot phase (PS: 300, NS: 252 kg; P = 0.01) and final BW (PS: 415, NS: 354 kg; P = 0.01). After slaughter, progeny from PS cows tended to have in- creased dressing percentage (PS: 59.0, NS: 58.1%; P = 0.09), had an increased hot carcass weight (PS: 227, NS: 191 kg; P < 0.01), and increased Longissimus muscle (LM) area (PS: 68.5, NS: 60.1 cm2; P = 0.02), while rib fat thickness was not affected by treatments (PS: 7.8, NS: 7.6 mm; P = 0.68). There was no effect of protein supplementation on color (L*, a*, b*) and pH of LM at 24 h postmortem (P > 0.10). In addition, moisture, ether extract and shear force of LM after 4 d of aging were similar between treatments (P > 0.10). There was no treatment × sex interaction for any variables (P > 0.10). This study showed that protein supplementation during early lactation im- proved growth performance and carcass quality, but did not result in enhanced meat quality of progeny. Winter and spring precipitation are predicted to increase in the Midwest region of the United States, causing muddy conditions. In a previous experiment, Angus cows (8 per treatment) were paired based on initial body weight (BW) and one cow from each pair was randomly allocated to either the mud or control treatment. Though cows consumed the same amount of dry matter, cows in the mud treatment weighed 37.4 kg less than cows in the control treatment by day 269 of gestation. The objective of this experiment was to evaluate developmental programming effects of steers born to cows in the mud treatment (MUD; n = 7) or the control treatment (CON; n = 6). Steers were weighed at birth and then weekly from approximately 56 d of age until weaning and were subjected to a glucose tolerance test (GTT) and adrenocorticotropic hormone (ACTH) challenge after weaning. Steers were then placed in the feedlot for an 84-d growing phase and were weighed weekly and 12th rib back fat (BF) and ribeye area (REA) were imaged every 28 d using ultrasonography. Data were analyzed as a randomized complete block design with repeated measurements when appropriate (SAS 9.4). Although there was a 37.4 kg decrease in BW of cows by the end of gestation, there was no evidence of a pen treatment effect on calf birth weight (P = 0.60) or weaning weight (P = 0.99). Additionally, there was no evidence of a pen treatment × day effect for steer BW from birth to weaning (P = 0.67) or growing phase BW (P = 0.60). There was evidence of a treatment × day of growing phase effect (P = 0.02) for BF, such that CON steers had greater BF on day 28 of the growing phase; however, there was no evidence of a treatment × day effect for REA (P = 0.20). Furthermore, there was no evidence of a pen treatment effect for the growing phase average daily gain (P = 0.74), dry matter intake (P = 0.65), gain:feed (P = 0.48), plasma glucose concentration (P = 0.67) or plasma insulin concentration (P = 0.61) in response to the GTT, or plasma cortisol concentration in response to the ACTH challenge (P = 0.51). These results indicate that while mud increased net energy requirements for cows in the MUD treatment, there were no subsequent effects observed for steer BW, gain:feed, or response to glucose and ACTH during the growing phase. Changes in maternal nutrition during the periconceptional period can influence postnatal growth in cattle. This study aimed to identify the impact of supplementing beef cows with rumen-protected methionine (RP-Met) during the periconceptional period on their female progeny. In experiment 1, plasma methionine (Met) levels were analyzed in samples from 10 Angus crossbred, non-lactating beef cows. Cows were randomly assigned to receive 454 g of cottonseed meal with 15 g/d of RP-Met (RPM; Smartamine M, Adisseo) or not (CON) for 5 d and data were analyzed as a completely randomized design with repeated measures. A treatment-by-day interaction was observed (P < 0.001), where plasma Met concentrations increased in the RPM treatment yet remained basal in CON. In experiment 2, 114 cows were fed a roughage-based diet and randomized to receive 454 g/d of corn gluten supplemented with 15 g/d of RP-Met (RPM n = 56) or not (CON n = 58) from days -7 to 7 relative to timed-artificial insemination using sexed semen to obtain females. Amino acids were measured in plasma samples from days -8, 0, and 7 in cows. In the female progeny, body weight, withers height, body length, and heart girth were measured every 60 d from birth through weaning at an average age of 242 ± 5.8 d. Liver, adipose tissue, and longissimus dorsi muscle biopsies were collected at 187.88 ± 5.5 d of age and a subset of 20 random samples (CON = 10; RPM = 10) were selected for RNA-seq on each tissue. Data were analyzed using a generalized randomized block design with repeated measures. Methionine was increased (P < 0.01) in plasma from cows in the RPM treatment on days 0 and 7. After calving, 34 female calves (CON = 16; RPM = 18) remained in the study and no difference was observed in birth weights between treatments. Calves were taller at the withers for RPM than CON (P = 0.03; CON = 92 ± 1.0 cm; RPM = 95 ± 1 cm) but there were no effects of treatment on other measures of body size. A total of 30, 24, and 2 differentially expressed genes (DEGs; P < 0.01) were observed in liver, longissimus dorsi muscle, and adipose tissue respectively. In summary, feeding RP-Met to cows in the periconceptional period resulted in female calves that were taller than CON before weaning. There were DEGs in the tissue samples but no other changes in measurements associated with body size. Repeat breeding is condition of cows that have normal or near normal estrous cycles but fail to become pregnant after several insemination. In this study, we investigated the microbes in the cervical and vaginal mucus of beef cattle and dairy cows that experience repeat breeding and detected the pregnancy after the third or more artificial insemination. A total of 14 beef cattle and 6 dairy cows that experience repeat breeding in the cattle herd in the Yogyakarta region of Indonesia were used as samples in this study. Cervical and vaginal mucus samples were collected using plastic sheet when the cow was re-estrus. The samples were put into the 5 ml Brain Heart Infusion Broth for bacterial isolation and identification. Pregnancy examination was carried out on day 45 after artificial insemination using ultrasound method. Bacteria that could be isolated and identified from cervical mucus and vaginal mucus of beef cattle and dairy cows included Bacillus sp., Staphylococcus epidermidis, Staphylococcus aureus, and Pseudomonas sp. Some cows detected positive in pregnancy examination even though the bacteria were presence in the cervical and/or vaginal mucus. There was various composition of bacteria found in the cervical mucus and vaginal mucus, of beef cattle and dairy cows with repeat breeding in livestock groups in Yogyakarta. The presence of bacterial in the cervical and vaginal mucus during estrus was not always become the causes of failed pregnancy.

Animal movement is an important factor in the transmission of animal infectious diseases. A better understanding of movement patterns is therefore necessary for developing effective control measures against disease spread. In Japan, a cattle tracing system was established in 2003, following a bovine spongiform encephalopathy epidemic, and the information on all cattle movements has been stored in a national database maintained by the National Livestock Breeding Center. Using these data, we previously analyzed the movement of dairy cows, concluding that heterogeneities in cattle movement are associated with regional and seasonal factors. In the present study, we aimed to identify specific factors affecting the regional and seasonal movement patterns of beef cows in Japan. From April 2012 to March 2017, 797,553 farm-to-farm movement events were recorded. We analyzed movements by month and by cattle age and looked at the frequency of movement within and between seven regions spanning the national territory. Our results show that calf movement peaked at 9–10 months old; these movements were considered to be via the market and were frequent within and between regions. For inter-regional movements, Kyushu region was the top producer of calves for calf trading markets throughout Japan. With regard to intra-regional movements, round-trip movements for summer grazing were observed in May and October for cattle of various ages in the northern regions, especially Hokkaido and Tohoku. Moreover, the movements of Japanese Shorthorn breeds in Tohoku region exhibited consistent annual peaks in May and October/November, in accordance with their seasonal breeding practice. In the areas with high relative densities of dairy cows, such as Hokkaido, the shipping of newborn beef calves produced via embryo transfer to dairy cows was also observed. Overall, understanding the patterns of beef cow movement will help develop effective disease surveillance measures, such as pre-movement inspections focused on specific regions and types of movement.

The potential for increasing the productivity in cow-calf operation has gained wide acceptance in the last decade among Brazilian farms. Our objectives were to assess the impacts of providing supplementation to pregnant beef cows during the dry grazing period on cow and calf performance. During a 2-yr period, Tabapuã cows (n = 36) were blocked by parity and body weight (BW), and randomly assigned to pastures at the beginning of the rainy season. Brachiaria pastures were assigned to receive i) no application of N and cows were supplemented with mineral mix (CON), ii) application of 165 kg of N ha-1. yr-1 and cows supplemented with a commercial product containing 40% of CP, macro and micro minerals, and vitamins (SUP), or iii) application of 165 kg of N ha-1. yr-1 and cows supplemented with grass silage (GS). For SUP system, one-half of the paddocks were deferred in late summer. For GS system, one-half of the paddocks were deferred in early summer and silage was made after 90 d of regrowth. During silage making, 21% of DDGs on a dry matter (DM) basis was mixed into grass. After silage harvesting, the area was differed again. At the beginning of the dry period, deferred areas had free access to animals for SUP and GS treatments. The commercial supplement and silage were fed for 140 d (from 116 ± 27 to 256 ± 27 d of gestation). The mineral mix was offered all year round. The PROC MIXED procedure of SAS was used to analyze all variables. The statistical model included supplementation program as a fixed effect. Block, paddock, and year were included as random variables. Initial BW, initial body condition score (BCS), and gestation period of cows were included as covariates. Initial BW and BCS, and final BW and BCS did not differ among treatments. The GS treatment provided the greatest average daily gain (ADG) during prepartum (0.49 kg/d) than CON and SUP strategies (an average of 0.37 kg/d). Whereas ADG during postpartum was not altered by treatments. Considering the full cycle (prepartum and postpartum), supplementation strategies did not affect ADG and BCS. No differences were obtained for birth weight, weaning weight, and ADG in calves. Overall, the supplementation programs adopted in this study did not influence individual gains in both cow and calf. The objective of this study was to evaluate whether oversupplying metabolizable protein (MP) during late gestation influences glucose and insulin concentrations, and insulin resistance (IR) in late gestation and early lactation. Crossbred Hereford, first-lactation heifers were individually fed diets to supply 133% (HMP, n = 11) or 100% (CON, n = 10) of their predicted MP requirements for 55 ± 4 d (mean ± SD) prior to calving. All heifers received a common lactation ration formulated to meet postpartum requirements (103% MP and 126% ME). After feed was withheld for 12 h, cattle underwent an intravenous glucose tolerance test (IVGTT) on days −6.7 ± 0.9 and 14.3 ± 0.4 by infusing a 50% dextrose solution (1.36 g glucose/kg BW0.75) through a jugular catheter with plasma collected at −10, 0 (immediately after infusion), 5, 10, 15, 20, 25, 30, 45, 60, 75, 90, and 120 min, respective to the infusion. Glucose and insulin concentrations were assessed. Insulin resistance indices (homeostasis model of insulin resistance [HOMA-IR], quantitative insulin sensitivity check index [QUICKI], revised quantitative insulin sensitivity check index [RQUICK], and RQUICKI incorporating serum beta-hydroxybutyrate concentrations [RQUICKIBHB]) were calculated from measurements of serum non-esterified fatty acids and beta-hydroxybutyrate and plasma glucose and insulin concentrations on days −34 ± 4, −15 ± 4, 7 ± 1, 28 ± 3, 70 ± 3, and 112 ± 3. Colostrum samples were collected within an hour of calving (prior to suckling) and analyzed for insulin concentration. Data were analyzed as a randomized block design using the PROC GLIMMIX of SAS, accounting for repeated measurements when necessary. Baseline (−10 min) plasma glucose and insulin concentrations were elevated (P ≤ 0.038) for HMP heifers during the antepartum IVGTT, but not (P ≥ 0.25) during the postpartum IVGTT. Plasma glucose and insulin concentrations throughout the antepartum or postpartum IVGTT did not differ (P ≥ 0.18) by prepartum treatment, nor did other glucose and insulin IVGTT parameters (i.e. max concentration and time to reach max concentration, nadir values, clearance rates and half-lives, area-under-the-curve, and insulin sensitivity index; P ≥ 0.20). Antepartum IVGTT IR indices indicated that HMP heifers were more (P ≤ 0.011) IR than their counterparts. Similarly, the prepartum HOMA-IR was greater (P = 0.033) for HMP heifers, suggesting increased IR. Postpartum IR indices did not (P ≥ 0.25) indicate that prepartum MP consumption impacted postpartum IR. Colostrum insulin concentration was increased (P = 0.004) by nearly 2-fold for HMP relative to CON heifers. These data demonstrate that prepartum MP overfeeding alters baseline glucose-insulin concentrations in late-pregnant beef heifers and increases colostrum insulin content without having carry-over effects on postpartum glucose-insulin concentrations and IR. The objective of this study was to examine the effects of recombinant bovine somatotropin (bST) administration on uterine hemodynamics and subsequent fetal programming in suckled cows during the first trimester of gestation. Crossbred beef cows (n = 152) were stratified by breed, days postpartum, parity, cyclicity status, and body condition score (BCS) before being assigned to either receive injections of bST every other week (BST; 500-mg/14 d) starting at fixed-time artificial insemination (TAI; d 0) until d 97 or to receive no bST (CTL). Blood samples were collected until d 97 for analysis of plasma concentrations of insulin-like growth factor 1 (IGF-1). Pregnancy was assessed via transrectal ultrasonography on d 41 and 173. A subset of pregnant cows (BST, n = 24; CTL, n = 28) were selected for assessment of uterine arterial blood flow (BF), pulsatility index, and resistance index (RI) of the uterine arteries ipsilateral and contralateral to the conceptus via color Doppler ultrasonography on d 97 and 233 of gestation. No differences (P = 0.99) were detected in body weight (BW) or BCS of dams; however, plasma concentrations of IGF-1 were greater (P < 0.001) in BST-treated cows. Color Doppler ultrasonography parameters differed whereby a treatment × day interaction (P = 0.007) was detected for RI on d 97 (P = 0.048); however, on d 233, RI did not differ (P > 0.10) but ipsilateral BF for BST-treated cows was greater (P = 0.0319) than controls. Mean heart girth diameter, crown-to-rump length, and neonate BW at 7 ± 5 d of calf age did not differ (P > 0.10). Liver tissue samples from each calf were collected for analysis of mRNA expression of target insulin-like growth factor system ligands. There was no difference in gene expression of hepatic IGF-1 between treatments (P = 0.99). A treatment × sex interaction was determined, where BST heifers had increased mRNA expression of IGFR1 compared to BST bulls (P = 0.03). Bi-weekly administration of bST until d 97 of pregnancy increased plasma concentrations of IGF-1, altered uterine hemodynamics in dams, induced sex-specific changes in liver gene expression of the offspring but failed to alter calf morphometries or calf performance until weaning.

Our objectives were to determine the effects of dam body condition score (BCS), age of dam, and calf sex on placental size and the relationships between dam body weight (BW) and calf size with placental size. Expelled placentas and calf size at birth were collected from crossbred beef heifers and cows during four experiments (n = 22 to 39/experiment). Placentas deemed complete by visual inspection were dissected; dry weights were determined for cotyledonary and intercotyledonary tissues. Mixed linear models were used to individually determine main effects of peripartum BCS category [Thin (<5), Moderate (=5), or Fleshy (≥6)], age of dam category [Primiparous (2 yr), Young (3–4 yr), or Mature (≥5 yr)], and calf sex on placental measures. Correlations were determined for placental characteristics with prepartum dam BW, gestation length, and calf size. Thin BCS dams had lower (P ≤ 0.05) cotyledonary, total placental, and average cotyledon weights and greater placental efficiency (calf birth BW/placental weight) than moderate and fleshy dams. Intercotyledonary weight was lower (P < 0.01) in thin BCS dams compared with fleshy dams. Thin and moderate BCS dams had smaller (P ≤ 0.04) calf birth BW than fleshy dams. Primiparous dams had lower (P ≤ 0.05) total placental and average cotyledon weights than young and mature dams, yet calf birth BW was unaffected (P = 0.17). Male calves were heavier (P = 0.01) than females, yet there were no differences (P ≥ 0.59) in placental weights. Calf birth BW and heart girth had moderate positive correlations (P < 0.01) and shoulder to rump length and abdominal girth had weak positive correlations (P < 0.01) with all placental weights. Dam prepartum BW and calf flank girth had moderate positive correlations (P < 0.01) with total placental weights and weak positive correlations (P < 0.01) with cotyledonary and average cotyledon weights. Intercotyledonary weight had moderate positive correlations (P < 0.01) with gestation length and calf flank girth and a weak positive correlation (P < 0.01) with dam prepartum BW. Gestation length had a weak positive correlation (P = 0.02) with total placental weight. Number of cotyledons was not correlated (P ≥ 0.28) with any dam or offspring characteristics. In conclusion, these data suggest that both maternal age and BCS affected placental size. The activity of the immune system in the reproductive tract has been proven to be crucial in the response to uterine diseases, normal reproductive functions, and tolerance to the allogeneic fetus during pregnancy. The objectives of the current study were to (1) evaluate uterine and vaginal cytokine concentrations in postpartum cows undergoing estrus synchronization followed by timed artificial insemination (TAI) and (2) correlate bacterial communities with cytokine concentrations. Postpartum Angus cows (n = 20) were subjected to a 7-Day Co-Synch protocol with pre-synchronization beginning 21 days prior (d −21) to TAI (d 0). Uterine and vaginal flushes were collected on d −21 and −2. Pregnancy was determined by transrectal ultrasound on d 30. Cytokines include interleukin (IL)-1b, IL-6, IL-10, transforming growth factor beta (TGF-β), and immunoglobin A (IgA) and concentrations were determined by commercial ELISA kits. No differences by day or pregnancy status in cytokine concentrations were detected in vaginal samples. No differences by day or pregnancy status in IgA, IL-10, or IL-1b concentrations were detected in uterine samples. Overall TGF-β concentrations in the uterus were greater in resulting pregnant than non-pregnant cows (44.0 ± 13.4 pg/mL vs. 14.7 ± 4.9 pg/mL; P = 0.047). Uterine TGF-β was correlated with the relative abundance of genera Treponema (r = −0.668; P = 0.049) in resulting non-pregnant cows on d −21 and with the relative abundance of genera Ureaplasma (r = 0.901; P = 0.0004) in resulting pregnant cows on d −2. In resulting pregnant animals, a tendency for a strong correlation was detected between d −2 progesterone concentrations and uterine TGF-β concentrations (r = 0.591, P = 0.07). Overall IL-6 concentrations in the uterus were greater in resulting non-pregnant than pregnant cows (198.7 ± 21.8 pg/mL vs. 144.3 ± 16.1 pg/mL; P = 0.045). A correlation was also detected between uterine IL-6 concentrations and the relative abundance of genera Butyrivibrio (r = 0.742; P = 0.022) in resulting non-pregnant cows on d −21. These results suggest possible relationships between different bacterial communities and cytokine concentrations within the uterus of beef cattle prior to TAI that may ultimately affect fertility outcomes.

The state of biodiversity in the world is critical where natural grassland is one of the habitat types showing the strongest deteriorating trend of biodiversity loss. At European Union level, 75 % of all grassland habitats have poor or bad status, with cessation of farming and subsequent overgrowth posing the greatest threat. To achieve favorable conservation status of natural grasslands, the area given over to grazing livestock or hay mowing needs to increase in many regions. The aim of this study was to calculate the number of cattle required to manage various types of unfertilized grazing land, and associated enteric methane emissions from these cattle, using Sweden as the study area. Four different scenarios with various categories of grazing cattle were evaluated: beef suckler cows and their offspring or dairy and beef cows in the current ratio and their offspring, combined with all male calves being raised as grazing steers or with males being raised as grazing steers in the current ratio and other males being raised as indoor bulls. Potential important factors that could reduce the number of additional cattle required were considered. The scenario requiring the smallest number of animals, beef cows and their grazing offspring, gave the lowest methane emissions, 16 kg/ha/year for low-yielding grazing land, such as wooded pasture, and 117 kg/ha/year for high-yielding grazing land, such as wet meadows. Taking all cattle reducing factors into account (mowing parts of the area, lower predicted stocking rate, use of all existing male cattle) significantly reduced the required cattle stock to one-third, corresponding to methane emissions of 5 and 34 kg/ha/year for low and high-yielding grazing land, respectively. In Sweden, 2.2 million ha of grazing land need to be restored to favorable conservation status, which would require 150 000–510 000 new beef cows and their grazing offspring would be needed, producing 33 000 to 109 000 tonnes of methane, compared to the 105 000 tonnes emitted by the present cattle stock (around 300 000 dairy cows and 210 000 beef cows). The components of milk from beef cows remain to be elucidated. This study examined the differences in the antimicrobial components of milk between dairy and beef cows. Quarter milk was collected from both Japanese Black (beef type) and Holstein (dairy type) cows to compare the concentrations of antimicrobial components. The concentration of lingual antimicrobial peptide (LAP) was higher, whereas that of the other antimicrobial components (lactoferrin, S100A7, and S100A8) was lower in beef cows than in dairy cows. Overall, these results indicate that the differences in antimicrobial components between beef and dairy cows may be associated with the difference in the prevalence of mastitis between them.

Objectives were to investigate the effects of supplementation with corn dried distiller's grains plus solubles (DDGS) to late gestating beef cows on arterial blood flow to the mammary glands during late gestation and early lactation; colostrum and milk production; dystocia and immunity; and calf BW. Cows were fed a control (CON ; n = 15; 5.1% CP; 36.2% ADF) diet consisting of 90% corn stover and 10% corn silage on a dry basis offered ad libitum or CON diet with supplementation of DDGS (0.30% of BW; SUP n = 12). Mammary gland blood flow was assessed on day 245 of gestation. At parturition, maternal and calving parameters were assessed; colostrum and jugular blood was sampled; and dams were weighed. Mammary gland blood flow and milk production was measured on day 44 of lactation. Calves were weighed fortnightly for 8 wk and at weaning. Colostrum production tended to be greater in SUP dams than in CON dams (837 vs. 614 ± 95 g, P = 0.10). Calves of SUP dams were heavier at birth and 24 h (0 h, 43.2 vs. 39.8 ± 1.0 kg, P = 0.02; 24 h, 44.0 vs. 40.4 ± 1.1 kg, P = 0.02). At birth and 24 h, blood pCO2 was greater in calves born to SUP dams (6.82 vs. 6.00 ± 0.41 kPa, P = 0.04). Serum IgG did not differ (P = 0.21) at 24 h. Ipsilateral mammary gland blood flow of SUP cows was greater than CON cows (2.76 vs. 1.76 ± 0.30 L/min; P = 0.03); however, when summed with contralateral, total blood flow was similar (P = 0.33). Hemodynamic measures on day 44 of lactation were similar (P ≥ 0.32). Milk production tended to be increased (13.5 vs. 10.2 ± 1.2 kg/d, P = 0.07) in SUP vs. CON cows. Despite similar BW through 56 d, calves from SUP cows were heavier (P = 0.04) at weaning (309.7 vs. 292.0 ± 6.0 kg). In conclusion, we accept our hypothesis that DDGS supplementation during gestation influenced mammary blood flow, milk production and calf weights. These findings implicate maternal nutrition's leverage on both nutrient and passive immunity delivery to the calf early in life as well as potential advantages on long-term performance. The objective of this experiment was to evaluate effects of extended-release eprinomectin on fescue toxicosis and impacts on performance and reproduction in fall-calving beef cows. Fall-calving Angus × Simmental multiparous cows [ n = 335; age = 5.8 ± 2.1 yr; 586.5 ± 6.0 kg body weight (BW); 5.48 ± 0.05 body condition score (BCS)] were stratified by BW, age, and BCS and randomly assigned to one of three treatments. Treatments included a spring injection of extended-release eprinomectin (SERE) on day 0, a fall injection of extended-release eprinomectin injection (FERE) on day 84, and a saline control (CON). All treatments were administered at a rate of 1 mL/50 kg BW. Prior to the experiment, all cows were treated with oral fenbendazole to minimize parasite load. Cows grazed endophyte-infected tall fescue. Hair coat score (HCS), BW, and BCS were recorded on all cattle. Fecal egg count (FEC), respiration rate (RR), horn fly and tick count, hematocrit (% packed cell volume, PCV), and serum prolactin were analyzed on a subset of cows (35/treatment). On day 194, cows were artificially inseminated (AI) and 11 d following AI were exposed to bulls for 51 d. Milk production was estimated on day 210 on a subset of 85 cow–calf pairs (28–29/treatment). There was a tendency for a treatment × time interaction (P = 0.07) for FEC likely driven by an increase in FEC of the CON cattle at day 126 compared to SERE and FERE. There was a tendency for a treatment × time interaction (P = 0.06) for cow BW, largely driven by time differences; however, there was no effect of treatment (P = 0.84) on BW. There was no difference (P ≥ 0.13) in cow PCV, fly and tick count, BCS, HCS, RR, and serum prolactin throughout the experiment. Additionally, there was no difference (P ≥ 0.46) in Julian calving date, calf birth BW, or milk production between treatments. Interestingly, heifer calves born to FERE dams tended to have greater (P = 0.06) weaning BW compared to heifer calves born to CON dams. In addition, there was no difference (P ≥ 0.17) in heat patch scores, AI conception rates, or overall pregnancy rates between treatments. Extended-release eprinomectin did not impact cow growth performance, reproductive performance or fescue toxicity symptoms when grazing endophyte-infected tall fescue; however, calf weaning BW tended to be improved. Beef cow herds are expected to be metabolically and reproductively efficient in varied and ever changing environmental conditions. Therefore, selection and management of grazing beef cows provides unique and diverse challenges in achieving optimal production efficiency for any environment. Beef cows face dynamic and highly variable nutritional environments that periodically are inadequate in meeting nutrient and energy requirements. Nutritional management during high metabolically stressed and key physiological states can lead to increased or decreased metabolic efficiency. Conversely, cow metabolic efficiency may be reduced in many production systems due to surplus nutritional inputs and reduced exposure to environmental stressors. Alternatively, metabolically potent supplementation strategies targeting enhanced energy metabolism and endocrine mechanisms would increase beef cow metabolic and economic efficiency. Metabolic efficient beef cows adapt to environmental changes by adjusting their metabolic energy utilization in order to match current environmental conditions and remain reproductively competent. This mechanism involves adaptive processes that drive adjustments in nutrient partitioning and energy utilization efficiency. However, the variation in metabolic and reproductive efficiency among beef cows within cow/calf production systems is substantial, suggesting a lack of complete integration of nutrition, genetics, and reproduction with environmental constraints and conditions. Better integration and understanding of the interactions may lead to advancements in metabolic efficiency of the cowherd. Metabolic flexibility is recognized as an important trait in dairy production but has received little attention thus far in beef cattle. Overall, management and supplementation strategies in cow/calf systems from a mechanistic, targeted nutritional approach during key physiological periods would hasten improvements in metabolic efficiency. In the dairy industry, excess dietary CP is consistently correlated with decreased conception rates. However, amount of excess CP effects on reproductive function in beef cattle is largely undefined. The objective of this experiment was to determine the effects of excess metabolizable protein (MP) supplementation from a moderately abundant rumen undegradable protein (RUP) source (corn gluten meal: 62% RUP) on ovarian function and circulating amino acid (AA) concentrations in beef cows consuming low quality forage. Non-pregnant, non-lactating beef cows (n=16) were allocated by age, BW and body condition score (BCS) to 1 of 2 isocaloric supplements designed to maintain BW for 60 days. Cows had ad libitum access to corn stalks and were individually offered a corn gluten meal-based supplement daily at 125% (MP125) or 150% (MP150) of National Research Council (NRC) MP requirements. After a 20-day supplement adaptation period, cows were synchronized for ovulation. After 10 days of synchronization, follicular growth was reset with gonadotropin releasing hormone. Daily thereafter, transrectal ultrasonography was performed to diagram ovarian follicular waves, and blood samples were collected for hormone, metabolite and AA analyses. After 7 days of observation of estrus, corpus luteum (CL) size was determined via ultrasound. Data were analyzed using the MIXED procedures of SAS. No differences (P⩾0.21) in BW and BCS existed throughout the study; however, plasma urea N at ovulation was greater (P=0.04) in MP150. Preovulatory ovarian follicle size at dominance, duration of dominance, size at spontaneous luteolysis, length of proestrus and wavelength were not different (P⩾0.11) between treatments. In the dairy industry, excess dietary CP is consistently correlated with decreased conception rates. However, the source from which excess CP is derived and how it affects reproductive function in beef cattle is largely undefined. The objective of this experiment was to determine the effects of feeding excess metabolizable protein (MP) from feedstuffs differing in rumen degradability on ovulatory follicular dynamics, subsequent corpus luteum (CL) development, steroid hormone production and circulating amino acids (AA) in beef cows. Non-pregnant, non-lactating mature beef cows (n=18) were assigned to 1 of 2 isonitrogenous diets (150% of MP requirements) designed to maintain similar BW and body condition score (BCS) between treatments. Diets consisted of ad libitum corn stalks supplemented with corn gluten meal (moderate rumen undegradable protein (RUP); CGM) or soybean meal (low RUP; SBM). After a 20-day supplement adaptation period, cows were synchronized for ovulation. After 10 days of synchronization, gonadotropin releasing hormone (GnRH) was administered to reset ovarian follicular growth. Starting at GnRH administration and daily thereafter until spontaneous ovulation, transrectal ultrasonography was used to diagram ovarian follicular growth, and blood samples were collected for hormone, metabolite and AA analyses. After 7 days of visual detection of estrus, CL size was determined via ultrasound. Data were analyzed using the MIXED procedures of SAS. As designed, cow BW and BCS were not different (P⩾0.33). Ovulatory follicular wavelength, antral follicle count, ovulatory follicle size at dominance and duration of dominance were not different (P>0.13) between treatments. Cows supplemented with CGM had greater post-dominance ovulatory follicle growth, larger dominant follicles at spontaneous luteolysis, shorter proestrus, and larger ovulatory follicles (P⩽0.03) than SBM cows. No differences (P⩾0.44) in peak estradiol, ratio of estradiol to ovulatory follicle volume, or plasma urea nitrogen were observed. While CL volume and the ratio of progesterone to CL volume were not affected by treatment (P⩾0.24), CGM treated cows tended to have decreased (P=0.07) circulating progesterone 7 days post-estrus compared with SBM cows. Although total circulating plasma AA concentration did not differ (P=0.70) between treatments, CGM cows had greater phenylalanine (P=0.03) and tended to have greater leucine concentrations (P=0.07) than SBM cows. In summary, these data illustrate that excess MP when supplemented to cows consuming a low quality forage may differentially impact ovarian function depending on ruminal degradability of the protein source.

Resource efficient winter-feeding of mature pregnant beef cows requires knowledge of how different roughage-based feeding strategies affect cow intake and energy status. Four diets based on traditional timothy-meadow fescue silage (TM), festulolium silage plus urea (FE), reed canarygrass silage (RC) or barley straw supplemented with urea and rapeseed meal (BR), were fed ad libitum for 16 weeks prepartum to 36 Hereford cows. Postpartum, cows were fed the same diet before release on pasture. Individual data on cow intake, changes in body weight (BW), body condition score (BCS) and plasma metabolites, calf birth and weaning weights were recorded. The TM and FE diets resulted in increased BW and BCS prepartum (p < 0.001), while the RC and BR diets resulted in a catabolic state, as indicated by a loss of BCS, lower insulin levels and higher non-esterified fatty acid levels in cows fed BR (p < 0.001). There were no dietary effects on calf parameters (p > 0.29). Feeding RC or BR prepartum might be a possible alternative to traditional timothy-meadow fescue silage if cows are allowed to regain lost BCS during the grazing period. The influence on cow reproductive- and calf performance should be considered before making this management change.

Multiparous Angus×Simmental cows (n=54, 5.22±2.51 years) with male progeny were fed one of two diets supplemented with either dried distillers grains with solubles (DDGS) or soybean meal (CON), from calving until day 129 postpartum (PP) to determine effects of excess protein and fat on cow performance, milk composition and calf growth. Diets were formulated to be isocaloric and consisted of rye hay and DDGS (19.4% CP; 8.76% fat), or corn silage, rye hay and soybean meal (11.7% CP; 2.06% fat). The objective of the study was to determine whether oversupplying MP prepartum affects postpartum cow BW, colostrum composition, milk production and composition, protein catabolism in the dam, and calf growth. Crossbred Hereford heifers were individually fed a control treatment designed to meet MP requirements (CON ; n = 10) or 133% of the MP requirement (HMP ; n = 11) from day −55 ± 4 until parturition. All cows were provided a common postpartum diet. Cow BW was measured on days 7 ± 1, 14 ± 2, 28 ± 3, 57 ± 4, 82 ± 5, and 111 ± 3 relative to parturition. DMI and ruminal pH were measured daily and summarized by week until day 33. Milk yield was estimated based on a 12-h two-quarter milk yield on days 7 ± 1, 12 ± 1, 28 ± 3, 33 ± 3, 70 ± 3, and 112 ± 3. Urine samples were collected from cows over a 6-d period starting on days 7 ± 1 and 28 ± 3 and the composited samples were analyzed for 3-methylhistidine (3-MH) and creatinine. Muscle samples were collected from cows on day 13 ± 1 while calf muscle samples were collected on days 2 and 111 ± 3 of age. Muscle samples from cows were analyzed for markers of protein catabolism, and calf muscle samples were analyzed for genes regulating cell growth and differentiation. Data were analyzed as a randomized complete block design using the MIXED procedure of SAS accounting for repeated measures when necessary. Postpartum BW did not differ (P ≥ 0.30) by treatment, day, or the interaction of treatment and day (T × D), but rump fat decreased (P = 0.011) as lactation progressed. DMI decreased during weeks 2 and 3 compared to 1 and 4, whereas ruminal pH was less during weeks 2, 3, and 4 relative to week 1. Colostrum fat concentration was less (P = 0.003) for HMP than CON; but, milk production was not affected by treatment. Milk yield was greatest from days 7 to 33 and decreased thereafter (P < 0.01). Urinary 3-MH and the 3-MH:creatinine ratio did not differ by treatment, day, or the T × D (P ≥ 0.22) interaction, nor was there a difference (P ≥ 0.13) in the abundance of catabolic proteins. The objective of the study was to determine the effect of oversupplying MP during late gestation on maternal BW, ruminal fermentation, nitrogen balance, and skeletal muscle catabolism. Crossbred Hereford heifers (n = 24) were assigned to a control treatment designed to meet MP requirements (CON) or a treatment providing 133% of the MP requirement (HMP). Heifers were individually fed their treatment from day −55 ± 3 relative to parturition and DMI was summarized by week. BW was measured on day −55 ± 3, −41 ± 3, −27 ± 3, and −8 ± 3. Ruminal digesta samples were collected on day −34 ± 5 and −15 ± 4 for short-chain fatty acid and ammonia-N (NH 3 -N) concentration. Plasma was collected the day prior to ruminal digesta samples and analyzed for plasma urea-N. Nitrogen balance was measured over a 6-d period starting on day −34 ± 4 and −15 ± 4. Following completion of the N balance periods, muscle biopsies were collected from the longissimus dorsi and analyzed for abundance of proteins relating to skeletal muscle catabolism. Data were analyzed as a randomized complete block (date of parturition) design with repeated measures using the MIXED procedure of SAS. Heifers fed HMP increased conceptus-corrected BW by a greater magnitude than CON at day −8 relative to −55 and −41 (treatment × day, P < 0.01). DMI increased (P < 0.01) by 18% on week −2 compared to −8, but then decreased (P < 0.01) by 8.0% for week −1. N-intake, apparent N digestion, N excretion, and N retention (g/d) were all greater (P < 0.01) for HMP heifers than CON but did not differ when expressed as a proportion of N intake. Ruminal NH3-N decreased (treatment × day, P < 0.01) as parturition approached for HMP (10.1 to 8.6 mg/dL); whereas, NH3-N was not affected for CON (1.0 to 1.3 mg/dL). Consequently, plasma urea-N was greater (P < 0.01) for HMP heifers (15.0 vs. 7.5 mg/dL). Heifers fed HMP had improved (P < 0.01) DM, OM, and NDF digestibility relative to CON heifers. Commercial Angus heifers (n = 190; body weight (BW) = 315 ± 49.3 kg) were used to determine the effects of trace mineral injections during gestation on heifer and subsequent calf performance. Heifers received three previous subcutaneous trace mineral (Multimin 90 [ MM ]; n = 93) or sterilized physiological saline (CON ; n = 97) injections approximately 90 d apart. These treatments were maintained and subsequent injections were given 205, 114, and 44 ± 26 d prepartum. Heifers were provided free-choice inorganic minerals. Heifer BW and body condition scores (BCS) were collected at trial initiation (296 ± 26 d prepartum) and 5- to 10-week intervals thereafter. Liver samples were collected at trial initiation, 5 and 176 ± 3 d postpartum from a subset of cows to determine trace mineral status. Milk production was assessed on 80 cow–calf pairs (40/treatment) at 71 ± 15 d postpartum. Cows were artificially inseminated (AI) 82 d postpartum and then exposed to bulls for 38 d. Data were reported from 174 calves (n = 87 calves/treatment). Calf liver samples were collected 5 and 147 ± 3 d postpartum to determine trace mineral status. Calf weaning BW was collected at 159 ± 26 d postpartum. Calf performance including calving date, birth BW, weaning BW, average daily gain (ADG), and health data were collected. Heifer BW and BCS did not differ (P ≥ 0.72) throughout the experiment. Multimin heifers tended (P = 0.08) to have greater initial liver Se and tended to have decreased (P = 0.08) initial liver Zn compared with CON. At calving, MM cows had increased (P ≤ 0.01) liver Cu and Se. There was no difference (P ≥ 0.47) in Julian calving date, calving percent, or unassisted births. Calf birth BW was lesser (P = 0.02) for MM than CON calves, and MM calves had greater (P = 0.03) liver Cu concentrations at birth than CON calves. Despite MM cows having increased (P < 0.01) milk production, calf weaning BW and ADG were not different (P ≥ 0.87). In addition, calf morbidity and mortality were not different (P ≥ 0.43) between treatments. Calf mineral status was not different (P ≥ 0.57) at the time of weaning regardless of treatment; however, MM cows had decreased (P = 0.03) liver Zn. Multimin cows had decreased (P = 0.05) AI pregnancy rates, yet there was no difference (P = 0.34) in overall pregnancy rate. The present study was conducted to investigate the effect of subclinical mastitis on clinic-pathological changes and public health importance in Mastitic cow. A total of 100 individual milk samples from clinically normal udder quarters of 100 dairy cow were collected and examined microbiologically as well as by using California Mastitis Test (CMT) for detection of subclinical mastitis and designing rapid diagnostic tests for other infection. The prevalence of Streptococcus species in the examined samples was (50%), where,17% S. agalctiae, 10 % S. dysgalactiae, 10 %., S. pyogenes, 2%, S.Salivaris, 4%., S. mitis, 3% S. equi, 2% S. mutans and 2%, S. angionosus, respectively. Streptococcus dysgalactiae are Gram-positive bacterial pathogens that affect cows in dairy herds and considered as the major causes of economic losses of dairy producers without a control program, somatic cell count revealed highly significant increase in count ≥ 145.000 cell than normal cows ˂ 100.000cells. Two types of blood samples were analyzed for hematology and biochemical analysis (total protein, inorganic phosphorous and calcium. Also LDH (Lactate dehydrogenase enzyme) was detected, where the results indicated that there is highly significant decrease in HB (Hemoglobin) concentration, significant decrease of PCV (packed cell volume) and RBCs (Red blood cells) and significant increase in WBCs (White blood cells) in hematological analysis, while biochemical analysis revealed that, highly significant increase in LDH activity while a notable decrease in total protein and serum calcium were observed. Hereford and Charolais beef cows (n = 24 per breed) were used to study the effect of breed and to evaluate late-cut reed canarygrass (RC) and whole-crop oats plus urea (WCO) compared with late-cut timothy (TG) with respect to feed intake and digestibility, rumination time, fecal particle size (PS) distribution, N excretion, and ruminal microbial CP production (MCP). The TG and RC were cut at flowering and WCO at hard dough stage of maturity. Cows were grouphoused, 6 groups per breed, and fed 3 diets ad libitum in 3 periods. The study was designed as two 3 × 3 Latin squares amalgamated to form a 3 × 6 rectangle for each breed. All data were statistically analyzed on group level. Indigestible NDF (iNDF) and urinary creatinine excretion were used as markers to estimate apparent diet digestibility and daily urine volume, respectively. Fecal PS distribution was determined by dry sieving, and ruminal MCP synthesis was estimated based on urinary output of purine derivatives. The TG diet had a higher apparent digestibility of OM and NDF (P < 0.001) than RC and WCO, which did not differ. The TG diet resulted in the greatest daily DMI, followed by WCO and RC (P < 0.001). Intake of NDF (NDFI, kg/d and % of BW) was greatest for TG, followed by RC and WCO (P < 0.001). Rumination time per kg DMI was longest for RC (P < 0.001), and RC and WCO resulted in longest rumination time per kg NDFI (P < 0.001). The WCO diet resulted in the largest geometric mean fecal PS and proportion of large particles and in the smallest proportion of small particles, whereas the opposite was found for RC, with TG being intermediate (P < 0.001). Intakes in kg per day were higher for Charolais than for Hereford (P = 0.002), but no breed effect was detected when intake was expressed in relation to BW. Charolais ruminated longer per kg NDFI corrected for BW (P = 0.02) and had smaller mean fecal PS (P = 0.049) than Hereford. Total N excretion was highest for RC and lowest for WCO (P < 0.001). The TG diet stimulated MCP production to a greater extent than RC and WCO (P < 0.001). The results indicate that late-cut RC and WCO could be suitable alternatives to late-cut TG for ad libitum feeding of early pregnant beef cows, and that intake was associated with cow BW, but not with breed. Synchronization programs using progesterone-releasing intravaginal devices that allow for fixed time artificial insemination are still finding increasing application in bovine reproduction. This practice is useful for rationalizing livestock management because an increased number of cows can be inseminated in one session without the need for estrus detection. Although much of the innovation related to the design and development of intravaginal devices for use in cattle took place in the previous century, progress in understanding the physiology of the bovine estrous cycle resulted in shorter treatment durations, a trend which is still continuing. In this competitive market, with little functional differentiation between the existing devices, the shorter treatment duration prompted for optimization of the progesterone content in the device, as the cost of the drug significantly contributes to the price per unit. For CIDR ® a reduction of the progesterone content of about 30 per cent was realized. Price reduction remained an important target for further device development. Next to reduction of progesterone content, cheaper and easier to process materials like polyethylene vinyl acetate (EVA) copolymers have been explored to replace the commonly used silicone elastomers. The reengineering effort of CIDR ® demonstrated that knowledge of release kinetics and insight into gradual depletion patterns in the device is critical for optimization of drug content without compromising performance (blood levels). More recent publications related to the use of alternative polymers like EVA and polyisoprene (IP) indicated encouraging results regarding further reduction of progesterone content. The use of EVA seems most promising, because it is in principle a low-cost polymer available in many grades and this thermoplastic polymer can be processed easily by means of commonly used techniques like injection molding and extrusion. The use of thermoplastic polymers, however, requires insight into the physical-chemical phenomena related to drug dissolution and re-crystallization taking place in the polymer during processing at high temperatures. These aspects, which may critically affect product stability, are often overlooked and are prompting to cover some of the background in this review. Finally, two different innovative approaches are discussed, one related to programable electronic devices for tailored simultaneous drug release and the other is a flexible drug-loaded helix, which is retained very well in several species without causing the usual inflammatory response. Beef cow stayability is a complex, economically important trait often used as an indicator of a cow's potential lifetime productivity. Stayability is defined as capability of a cow to maintain a perfect record up to 6 yr of age. This age is commonly cited as a financial break-even point, where initial costs of cow development and maintenance are recovered by her cumulative net income from yearly calf receipts. Later-maturing Bos indicus-Bos taurus crossbred cows may experience reproductive difficulty early in life but have a high potential for a long reproductive life span. It was the objective of this study to identify genetic variants associated with measures of beef cow stayability. A population of B. indicus-B. taurus crossbred cows (n = 305) from central Texas was used. Phenotypes for various measures of stayability to 6 yr of age were produced by artificially imposing five different culling criteria on data from the population. Cows were scored either as a 1 (indicating a perfect record through 6 yr) or a 0 (indicating failure at or before 6 yr), under each criterion. Genome-wide association studies (GWAS) were conducted for each criterion using univariate procedures and prefitting the fixed effect of cow contemporary group. SNP associations for two criteria surpassed the false discovery threshold of 0.15, when a cow was scored as 0 upon her first failure to wean a calf, regardless of reason, through 6 yr (criterion 2), and when a cow was scored as 0 upon her first failure to give birth to a calf, through 6 yr (criterion 3). Associated SNP were found on bovine chromosomes (BTA) 1, 2, 5, 9, 18, and 21 for criterion 2 and on BTA 1, 5, 11, 15, and 24 for criterion 3. A critical region on BTA 5: 43-50 Mb was identified for each criterion. Due to the similarities to prior work, the tendency for B. indicus cattle to experience reproductive difficulties early in life, and due to the large proportion of cows that left the herd at an early age under these criteria, these results suggest that the associations are likely driven by an early life trait such as age at puberty or rate of heifer development. Contents: Two experiments were conducted to determine (i) factors influencing calf temperament at weaning, (ii) association between heifer–calf temperament at weaning and temperament at breeding and (iii) effect of heifer–calf temperament on pregnancy rate per artificial insemination (P/AI). In experiment 1, beef cows and their calves (n = 285) from three farms were used. Sire docility estimated progeny difference (EPD) score, birth type (normal or assisted), calf gender, calf behaviour (during 1st 4 weeks) and calf health status (until weaning) were recorded. Cows and calves were assigned a temperament score (0—calm; 1—excitable), and all cows were given a body condition score (BCS, 1–9; 1—emaciated; 9—obese) at weaning. Calf's illness (p < .05), low sire docility EPD score (p < .05), altered gait (p < .05), altered resting behaviour (p < .01), reduced/no play behaviour (p < .05) and cow excitable temperament (p < .001) increased calf excitable temperament at weaning. In experiment 2, replacement heifer–calves (n = 758) from 12 farms were assigned a temperament score at weaning and later at breeding. Blood from 40 calves at weaning and 31 heifers at initiation of synchronization (same animals) was collected by coccygeal venipuncture for determination of circulating cortisol and substance P concentrations. Heifers were assigned a BCS and reproductive tract score (RTS, 1–5; 1—immature, acyclic; 5—mature, cyclic), synchronized for fixed time AI, observed for oestrus and were artificially inseminated. Cortisol concentrations were increased in excitable heifer–calves compared to calm heifer–calves at weaning (p < .05), and substance P was increased in excitable compared to calm females both at weaning and breeding (p < .05). Low sire EPD docility score (p < .01), heifer–calf excitable temperament at weaning increased excitable temperament at breeding (p < .01). Controlling for BCS categories (p < .01), oestrous expression (p < .0001) and temperament at breeding by oestrous expression (p < .05), the calf's excitable temperament at weaning (p < .001) reduced P/AI (Calm, 62.7 (244/389) vs. Excitable, 53.4% (197/369); p < .01). In conclusion, selection of docile cows and sires with greater docility EPD score should be given consideration to reduce calf excitement. The objectives of this study were to evaluate the impact of vaccine program and progression of grazing season on beef cow milk fatty acid composition. In 2015, Fifty-one Angus-crossed beef heifers were randomly assigned to receive a modified-live viral vaccine (MLV; Bovishield Gold 5 FP) or killed viral vaccine (KV; Cattlemaster Gold 5 FP) against BVDV beginning at 3 months of age and for the duration of their lifespan in the herd. In 2019, 2020, and 2021, milk samples were collected on days 1, 63, and 205 of lactation. Milk samples were subjected to fatty acid analysis using Gas Chromatography. Data were analyzed as a completely randomized design with repeated measures using SAS. Milk concentrations of 16:0, 18:0, 18:1n-9, 18:2n-6, 18:3n-3, or total fatty acids when expressed as mg/g were not affected by vaccine treatment (P = 0.58). However, milk 16:0 concentrations decreased linearly (P < 0.0001) as the grazing season progressed from day 1 to day 205 of lactation. This was despite a linear increase (P = 0.05) in forage concentrations of 16:0. Milk concentrations of 18:0 decreased linearly throughout lactation (P = 0.002). Likewise, milk concentrations of 18:1n-9 decreased throughout lactation (P < 0.0001). Milk concentrations of 18:2n-6 declined linearly (P < 0.0001) as the grazing season progressed into the fall weaning period. Interestingly, milk concentration of 18:3n-3 did not change (P = 0.62) with time. Biohydrogenation intermediates were reduced in concentration and did not vary across grazing season (P = 0.06) Total fatty acids decreased throughout lactation (P < 0.0001). Total fatty acid content of forage did increase linearly (P = 0.02), which would suggest milk production (volume) and de novo milk fatty acid synthesis altered milk composition more so than forage fatty acid composition. In conclusion, vaccine treatment did not affect milk composition. However, grazing season progression resulted in decreased milk fatty acid concentrations from day 1 to day 205 of lactation. Cows fed diets consisting primarily of stored forages and concentrates may be at risk for vitamin A deficiency because these feedstuffs are low in ß-carotene. Furthermore, dietary vitamin A concentrations of the cow during late gestation will affect vitamin A concentrations in colostrum, which calves rely on at birth to supply vitamin A. The objective of this study was to evaluate how level of vitamin A supplementation from mid-gestation to early lactation affected cow and subsequently, liver retinol concentrations of their calf. Multiparous beef cows (n = 54) were stratified by body condition score and previous source and assigned to pen (n = 9); 3 pens were assigned to each of the following treatments: current NASEM recommendation (31,000 IU/d; 1X), 3 times (93,000 IU/d; 3X), or 5 times (155,000 IU/d; 5X) the current NASEM recommendation for supplemental vitamin A. Cows were limit-fed a diet consisting of wheat straw, corn silage, and wet distillers grains. Liver biopsies were collected on cows 3 weeks before treatment initiation, d 40 and d 81 of supplementation, and both cows and calves were sampled 32 days post-calving (165 ± 22 d SD of supplementation). No differences (P=0.86) in initial cow liver retinol (mean 186 μg/g DM) were observed between treatments. A significant treatment × day interaction (P < 0.01) was observed for cow liver retinol. On d 40, cows in 1X had liver retinol concentrations (178 μg/g DM) that were not different (P = 0.12) from 3X (213 μg/g DM) but less (P = 0.02) than 5X (241 μg/g DM), while 3X and 5X did not differ (P = 0.21). Liver retinol on d 81 was less (P < 0.05) in 1X (189 μg/g DM) compared with 3X (334 μg/g DM) and 5X (412 μg/g DM), which did not differ (P = 0.20). For cow liver retinol post-calving, 1X (187 μg/g DM) was less (P< 0.05) than 3X and 5X, and 3X (454 μg/g DM) was less (P< 0.05) than 5X (674 μg/g DM). Liver retinol concentrations of 1X cows remained below adequate reference ranges (300-700 μg/g of DM) throughout the study, whereas 3X and 5X were increased into the adequate range by d 81. Calf liver retinol concentration also differed among treatments (P = 0.01), as calves of cows in 1X had less (P < 0.05) liver concentrations than 3X and 5X calves which did not differ (P=0.12). Liver retinol concentrations considered adequate for calves at 32 days of age (100-350 μg/g of DM) were not observed in 1X calves (51 μg/g DM) but were observed in calves from 3X and 5X cows (119 and 165 μg/g DM, respectively). Optimizing reproductive efficiency is crucial in sustaining a successful cattle operation. However, optimizing reproductive performance in young range beef cows is often not captured due to an inability to meet the increased physiological demands. The objective of this study was to determine the impact of feed additives on reproductive performance, cow body weight (BW), body condition score (BCS), and calf growth in young, range beef cows. Data were collected from 2- and 3-year-old range cows (n = 189) from the March-calving herd at the University of Nebraska Gudmundsen Sandhills Laboratory (GSL) located near Whitman, NE over a three-year period (2019-2021). Cows were individually supplemented 908 g/d with one of two 30% crude protein supplement treatments with the addition of either: 1) 160 mg·cow-1·d-1 of rumensin (MON; Elanco Animal Health) or 2) 40 g·cow-1·d-1 of propionate salt (CAP; NutroCal 100; Kemin Animal Nutrition and Health). Supplementation was initiated approximately 7 d after calving and continued for an average of 70 d. Cow BW and BCS were collected weekly from calving to the start of breeding and then again at weaning. On approximately d 60 postpartum, milk production was measured by a modified weighsuckle- weight technique. Cows were exposed to fertile bulls for a 45-d breeding season starting in June of each year. Cow BW (P ≥ 0.19) and BCS (P ≥ 0.11) were not affected by postpartum supplementation during the study. However, cows receiving CAP did lose more BW (P = 0.04; -7 vs -2 kg for CAP and MON; respectively) from calving to breeding than their counterpart. Calf BW at birth, weaning, or 205-d weight were not impacted (P ≥ 0.68) by the postpartum supplementation strategy. Twenty-four-hour milk production and milk components (fat, solids-non-fat, urea nitrogen) were not influenced (P > 0.21) by postpartum supplementation strategy. However, cows receiving CAP supplement had an increase (P = 0.04; 5.40 vs 5.31% for CAP and MON; respectively) in milk lactose percentage and tended (P = 0.06; 2.56 vs 2.68% for CAP and MON; respectively) to have lower milk protein percentage than their counterpart. Cows receiving CAP supplementation had an increase (P = 0.04; 89 vs 80% for CAP and MON; respectively) in overall pregnancy rate. This study implies that young postpartum cows fed additional glucogenic precursors in the form of propionate salts may increase reproductive efficiency without impacting a change in body weight or body condition score. The objective of this study was to explore the potential of transmission infrared (TIR) spectroscopy in combination with partial least squares regression (PLSR) for quantification of dairy and beef cow colostral immunoglobulin G (IgG) concentration and assessment of colostrum quality. A total of 430 colostrum samples were collected from dairy (n = 235) and beef (n = 195) cows and tested by a radial immunodiffusion (RID) assay and TIR spectroscopy. Colostral IgG concentrations obtained by the RID assay were linked to the preprocessed spectra and divided into combined and prediction data sets. Three PLSR calibration models were built: one for the dairy cow colostrum only, the second for beef cow colostrum only, and the third for the merged dairy and beef cow colostrum. The predictive performance of each model was evaluated separately using the independent prediction data set. The Pearson correlation coefficients between IgG concentrations as determined by the TIR-based assay and the RID assay were 0.84 for dairy cow colostrum, 0.88 for beef cow colostrum, and 0.92 for the merged set of dairy and beef cow colostrum. The average of the differences between colostral IgG concentrations obtained by the RID- and TIRbased assays were -3.5, 2.7, and 1.4 g/L for dairy, beef, and merged colostrum samples, respectively. Further, the average relative error of the colostral IgG predicted by the TIR spectroscopy from the RID assay was 5% for dairy cow, 1.2% for beef cow, and 0.8% for the merged data set. The average intra-assay CV% of the IgG concentration predicted by the TIR-based method were 3.2%, 2.5%, and 6.9% for dairy cow, beef cow, and merged data set, respectively. The utility of TIR method for assessment of colostrum quality was evaluated using the entire data set and showed that TIR spectroscopy accurately identified the quality status of 91% of dairy cow colostrum, 95% of beef cow colostrum, and 89% and 93% of the merged dairy and beef cow colostrum samples, respectively. The results showed that TIR spectroscopy demonstrates potential as a simple, rapid, and cost-efficient method for use as an estimate of IgG concentration in dairy and beef cow colostrum samples and assessment of colostrum quality. Background: The pregnancy rate in fixed-time insemination (FTAI) programs in beef cattle is affected by several factors, including the size of ovulatory follicle (OF). Induced ovulation of small OF (=11 mm) reduces the chance of pregnancy after FTAI when compared to ovulation of larger follicles (=12 mm). Hormonal treatments to enhance the follicle growth have been widely used in comercial beef cattle systems, aiming to improve the development of a dominant follicle. The objective of this experiment was to evaluate the effect of treatment with eCG or FSH at the time of progesterone implant removal on the diameter of OF, final follicle growth and pregnancy rate in beef cows. Materials, Methods & Results: Three hundred and seven suckled beef cows with body condition score 2.80 ± 0.24 (1 to 5 scale) and 45 to 70 days postpartum were synchronized to FTAI. On day 0, intravaginal implants containing 1 g of progesterone, were inserted and administered 2 mg of estradiol benzoate (EB), i.m. On day 7, intravaginal implants were removed and 150 µg of D-cloprostenol were administered i.m. At this time, three groups were assigned according to the treatments: eCG (400 UI eCG, i.m.), FSH (25 UI FSH, i.m.) and Control (1 mL Saline, i.m.). On day 8, all cows were injected with 1mg EB i.m. and were inseminated 52 to 56 h after the implant removal. Transrectal ultrasonography exams of ovaries were performed at the time of implant removal and before FTAI to determine the diameter of the largest follicle and the follicular daily growth. Pregnancy exams were conducted by transretal ultrasonography 30 days after FTAI. Statistical analyzes were performed by Chi-square test for pregnancy rate. Ovarian diameters and follicular daily growth were analyzed by ANOVA with means compared by Tukey test considering significant P value < 0.05. The diameter of the largest follicle measured on day 7 (10.07, 9.64 and 10.36 mm) and on day 9 (12.73, 11.53 and 12.57 mm), to eCG, FSH and Control respectively, did not differ within treatments (P > 0.05). Final follicular growth was greater (P < 0.05) on eCG group (1.33 mm/day) compared to Control (1.1 mm/day) and FSH (0.94 mm/day). The pregnancy rates of FTAI were 56.6% (60/106), 52% (51/98) and 50.4% (52/103) to eCG, FSH and Control, respectively, with no statistic difference (P > 0.05). Discussion: Suckled beef cows managed under grazing systems frequently have reduced follicular development and longer postpartum anestrous. In this situation, protocols to FTAI combining progesterone, estradiol and eCG or FSH at the time of progesterone device removal are encouraged to improve final follicular growth, ovulation and to increase conception rate. The hypotesis of similar follicular growth and pregnancy rate in suckled beef cows treated with eCG or FSH was not confirmed in the field trial. eCG treated cows had greater follicular growth, follicular diameter at day 9 and higher pregnancy rate compared to FSH and Control cows. FSH produced similar follicular growth rates to Controls and both were significantly smaller than observed in cows receiving eCG. Also FSH supplementation failed to enhance pregnancy rate compared to Control cows. The longer half-life of eCG and its greater LH-like effects could explain the difference in final follicular stimulation when compared to FSH in FTAI protocols. An experiment was designed to evaluate endocrine parameters, ovarian dynamics, and pregnancy rates to fixed-time artificial insemination (FTAI) following the 9-d CIDR-PG protocol in comparison to the 14-d CIDR-PG protocol. While both are long-term protocols using CIDR treatment for presynchronization, the 9-d CIDR-PG protocol differs from the 14-d CIDR-PG protocol in that prostaglandin F 2α (PG) is administered at CIDR insertion and removal to facilitate a decreased length of progestin treatment and potentially enhance response to the presynchronization treatment. Estrus was synchronized for 393 mature beef cows across five locations. Treatments were represented in each location, and cows within each location were randomly assigned to one of the two protocols based on age, days postpartum (DPP), and body condition score (BCS). Cows assigned to the 14-d CIDR-PG treatment received a CIDR insert (1.38 g progesterone) on Day 0 with removal of CIDR on Day 14, and 25 mg PG 16 d after CIDR removal on Day 30. Cows assigned the 9-d CIDR-PG treatment received 25 mg PG and a CIDR insert (1.38 g progesterone) on Day 5; 25 mg PG and removal of CIDR on Day 14; and 25 mg PG 16 d after CIDR removal on Day 30. In both treatments, cows received FTAI on Day 33, 72 h after PG. All cows were administered 100 μg gonadotropin-releasing hormone (GnRH) concurrent with insemination. For a subset of animals in each treatment, ovarian ultrasound was performed and blood samples were collected for determination of serum estradiol concentrations at CIDR removal, PG administration, and FTAI. Protocols were compared on the basis of estrous response and pregnancy rate resulting from FTAI. Serum estradiol concentrations, follicle size, and estrous response did not differ based on treatment. However, cows assigned to the 9-d CIDR-PG protocol tended to achieve greater FTAI pregnancy rates than cows assigned to the 14-d CIDR-PG protocol (62% versus 52%; P = 0.07). Across treatments, greater pregnancy rates tended ( P = 0.10) to be achieved by cows that expressed estrus prior to FTAI (69% for 9-d CIDR-PG, 58% for 14-d CIDR-PG) than by cows that failed to express estrus (55% for 9-d CIDR-PG, 47% for 14-d CIDR-PG). In summary, the 9-d CIDR-PG protocol is an effective protocol for synchronization of estrus among mature beef cows, and pregnancy rates to FTAI tended to be improved through use of the 9-d CIDR-PG compared to the 14-d CIDR-PG protocol. The objectives of the current experiment were to determine if delaying insemination by 8 h in a FTAI protocol would alter estrus expression and pregnancy rates in cows inseminated with sex-sorted semen, characterize bull variation in pregnancy rates to sex-sorted semen and examine the impact of repeated years of FTAI to sex-sorted semen on calving distribution. Over three breeding seasons, postpartum cows (n = 839) were estrous synchronized using the 5-day CO-Synch + CIDR system. Cows were given GnRH (100 μg i.m., Factrel) at time of insertion of a controlled internal drug releasing device (CIDR; Eazi-Breed CIDR). Five d later CIDR was removed and PGF 2α (25 mg i.m., Lutalyse) was given at removal and 8 h later. Estrus detection aids were applied at CIDR removal. Cows were inseminated with X-sorted or Y-sorted sex-sorted semen at 72 h (NORM) or 80 h (DELAY) after CIDR removal, and GnRH was administered at AI. At insemination, estrus status was categorized as positive (YES), partial (QUES), unknown (NR) or negative (NO). Bulls were introduced to cows at 14 d and removed at 60 d after FTAI. Pregnancy diagnosis was performed by ultrasound at d 60 after FTAI and via palpation at 60 d after bull removal. There was no difference (P > 0.05) in pregnancy rates to sex-sorted semen or final pregnancy rates between NORM and DELAY cows. Pregnancy to sex-sorted semen averaged 35.2% whereas final pregnancy rates were 90.6%. More cows (P < 0.05) in the DELAY group expressed estrus before FTAI, but this increase did not alter pregnancy rates to sex-sorted semen. Expression of estrus before FTAI increased (P < 0.02) pregnancy rates to sex-sorted semen across treatments with differences being YES > QUES or NR > NO. There was considerable variation in pregnancy rate by bull (P < 0.05) with pregnancy rates ranging from 55.6% to 19.3%. Whole herd calving distribution was altered (P < 0.05) after 3 y of use of sex-sorted semen compared to the previous 3 y when conventional semen was used. We conclude that delaying insemination by 8 h in an FTAI protocol did not improve pregnancy rates to sex-sorted semen despite more cows exhibiting estrus before FTAI. In addition, a high bull to bull variation in pregnancy rates to sex-sorted semen is a limitation in FTAI systems. Further research into FTAI strategies for use with sex-sorted semen is warranted. This experiment was designed to compare pregnancy rates in postpartum beef cows following split-time (STAI) or fixed-time (FTAI) artificial insemination. Estrus was synchronized for 671 cows at seven locations following administration of the 7-d CO-Synch + CIDR protocol (100 μg GnRH + CIDR insert [1.38 g progesterone] on d 0; 25 mg prostaglandin F 2α [PG] at CIDR removal on d 7). Cows were assigned to treatments that were balanced across locations based on age, body condition score, and days postpartum at the time treatments were initiated. All cows in treatment 1 (n = 333; FTAI) were inseminated at 66 h after PG and GnRH was administered concurrent with insemination regardless of estrus expression. For cows in treatment 2 (n = 338; STAI), inseminations were performed at 66 or 90 h after PG, and estrous status was recorded at these times. Cows in the STAI treatment that exhibited estrus by 66 h were inseminated at that time and did not receive GnRH, whereas AI was delayed 24 h until 90 h after PG for cows that failed to exhibit estrus by 66 h. Gonadotropin-releasing hormone (100 μg) was administered concurrent with AI at 90 h only to cows failing to exhibit estrus. Estrus expression that occurred during the 24 h delay period among cows assigned to the STAI treatment increased the total proportion of cows that expressed estrus prior to insemination (1 = 60%; 2 = 86%; P < 0.001). Pregnancy rates for cows inseminated at 66 h that exhibited estrus did not differ between treatments (1 = 58%; 2 = 58%; P = 0.93); however, pregnancy rates among non-estrous cows at 66 h were improved (1 = 35%; 2 = 51%; P = 0.01) among cows assigned to the STAI treatment when insemination was postponed by 24 h. Consequently, total AI pregnancy rate tended to be higher for cows that received STAI (1 = 49%; 2 = 56%; P = 0.06). In summary, following administration of the 7-d CO-Synch + CIDR protocol, total estrous response increased and pregnancy rates resulting from AI tended to be higher among cows assigned to STAI versus FTAI treatments. This study compared intake of alternative roughage-based diets and of common late-cut grass silage and related intake to urinary nitrogen (N), urea-N and purine derivative (PD) excretion, where PD is an indicator of rumen microbial crude protein (MCP) synthesis. Total urine was collected from 36 Hereford cows, blocked into three groups based on expected calving date. Cows within calving groups were randomly assigned to one of four roughage diets: common mixed grass silage (MGS), festulolium silage plus urea (FLS), reed canarygrass silage (RCS) and barley straw plus urea and rapeseed meal (BRM). Diet crude protein (CP) content was classified into five fractions (A, B 1 , B 2 , B 3 and C), based on degradability characteristics. Feed intake and urinary excretion data were analysed by ANOVA in a randomised block design. To further explain the ANOVA results, multiple regression analyses were conducted to study relationships between intakes of total N (g/d); sum of the CP fractions A, B 1 and B 2 (AB 1 B 2 ; g/d), most of which is considered rumen-degradable; digestible organic matter (DOMI; kg/d); protein balance in the rumen (g/kg dry matter); and urinary excretion of N, urea-N (g/d) and PD (mmol/d). Urinary N and urea-N excretion was positively related to N intake and was better explained by N intake than intake of AB 1 B 2 . Feeding BRM resulted in the lowest N intake and urinary N output ( P < 0.001). Cows fed MGS, FLS and RCS had similar N intake, but urinary N and urea-N excretion was significantly higher ( P < 0.001) in cows fed RCS, which probably was attributable to the significantly lower DOMI of this diet ( P < 0.001). Furthermore, addition of DOMI to N intake in the multiple regression analysis increased the proportion of explained variation in urinary N and urea-N excretion. The MGS and FLS diets stimulated rumen MCP production to a greater extent than the BRM diet, as indicated by the higher urinary output of PD in cows fed the grass silage-based diets ( P < 0.001). Diet had no significant effect on urinary PD excretion when expressed per kg DOMI. Overall mean urinary creatinine excretion was 0.197 ± 0.047 mmol/kg body weight, with no significant effect of diet. This study showed that intake of both N and DOM need to be assessed when choosing a suitable alternative roughage diet for suckler cows, in order to prevent undesirable losses of urinary N.
