9. Metal organic frameworks for bacterial detection in environmental samples

Abstract

Pathogenic bacteria pose significant threats to public health. Early and accurate detection of bacteria is crucial for infection control and prevention of outbreaks. This paper reviews recent advances in metal organic framework (MOF)-based methods for bacterial detection. MOFs are promising materials for biosensing due to their large surface areas, customizable properties, and ability to integrate various sensing capabilities. Several colorimetric and electrochemical detection techniques that utilize MOFs are discussed. Colorimetric assays combining MOFs with gold nanoparticles, peroxidase-mimicking activity, and aptamer recognition have achieved sensitive and specific detection of bacteria such as E. coli and S. aureus. Electrochemical biosensors integrating MOFs, antibodies, aptamers and quantum dots have also demonstrated low detection limits for various bacteria. Notably, techniques using MOF peroxidase-like activity coupled with magnetic separation or competitive binding assays show potential for point-of-care pathogen detection. Furthermore, optimization of MOF properties and integration within microfluidic platforms may lead to portable, low-cost and rapid methods suitable for on-site bacterial analysis in diverse settings. Continued exploration of MOF-based sensing strategies holds promise for improved bacteria monitoring and control of infectious diseases.

Abstract

Carbon Quantum Dots from Natural Sources (NACQDs) is a novel type of carbon-based material that have garnered significant attention due to their remarkable features, including luminescence, photostability, nanoscale size, water solubility, low toxicity, biocompatibility, and cost-effectiveness. The synthesis of NACQDs involves a diverse range of natural sources, such as fruits, foods, beverages, human and animal derivatives, vegetables, leaves, and waste materials. Various synthesis methods, including electrochemical approach, chemical oxidation, hydrothermal carbonization, ultrasonic techniques, microwave-assisted synthesis, solvothermal method, laser ablation technique, thermolysis, and atmospheric plasma-based synthesis, have been explored to tailor the size and properties of NACQDs. Characterization techniques like Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), transmission electron microscopy (TЕM), ultraviolet absorption, fluorescence properties, and nuclear magnetic resonance (NMR) have provided invaluable insights into the physical and chemical characteristics of NACQDs. This review highlights the immense potential of NACQDs in metal ion sensing applications and underscores the need for further investigation to enhance their reproducibility and precise control over their properties. NACQDs hold great promise as versatile nanomaterials for metal ion sensing and are poised to revolutionize diverse fields, ranging from environmental monitoring to biomedical diagnostics and chemical analysis.

Abstract

Symplocos, a genus rich in endemic and keystone species in China's subtropical forests, plays a vital role in maintaining biodiversity. Climate change impacts on Symplocos trees affect ecosystem composition, dynamics, and functions. This study delves into the potential ramifications of climate change on the distribution and richness of Symplocos species across China under different climatic conditions. A comprehensive analysis was conducted using occurrence records of 29 species of Symplocos, focusing on habitat suitability modeling and spatial analysis, to demonstrate the possible effects of climate change on the distribution and diversity patterns of Symplocos species in China. Environmental data, including bioclimatic variables, were collected, and the future climate scenarios were projected using the combined average of the two global climate models. Four modeling algorithms were applied in an ensemble species distribution models to assess the distribution of Symplocos species and decipher the pattern of species richness and endemism. The results indicate high accuracy and performance of the models with a mean AUC of 0.94 and TSS of 0.77, particularly the random forest. The analysis identified key bioclimatic variables influencing Symplocos species distribution, with precipitation patterns (precipitation of driest month and precipitation of warmest quarter) and temperature variability (temperature mean diurnal range) playing significant roles. Predictions suggest a reduction in Symplocos species richness over the next six decades, particularly in southern and southwestern China, with potential gains in other regions. Overall, this study highlights the vulnerability of Symplocos species to climate change and underscores the importance of proactive conservation efforts and forest plantations to mitigate future loss in Symplocos species diversity.

Abstract

Abstract A novel two tri-thiosemicarbazones derivatives, namely 2,2',2''-((2-Hydroxybenzene-1,3,5-triyl)tris(methanylylidene))tris(N-benzylhydrazine-1-carbothioamide) (HBC) and 2,2',2''-((2-hydroxybenzene-1,3,5-triyl) tris (methanylylidene)) tris (N-allylhydrazine-1-carbothioamide) (HAC), have been synthesized and their chemical structures were determined using different spectroscopic and analytical approaches. Then, utilizing methods including open circuit potential, potentiodynamic polarization, and electrochemical impedance spectroscopy, the inhibitory effect of the synthesized thiosemicarbazones on mild steel (MS) in an acidic environment (0.5 M H2SO4) was thoroughly investigated. Remarkably, raising the concentration of our recently synthesized tri-thiosemicarbazones HBC and HAC increased the inhibitory efficiency values. The η values of the two investigated tri-thiosemicarbazones derivatives (HAC and HBC), at each concentration are extremely high, and the maximum values of the efficiencies are 98.5% with HAC and 98.8% with HBC at the 800 ppm. The inhibitors adsorbed on the mild steel surface and generated a charge and mass movement barrier that protected the metal from hostile ions. According to polarization curves, HBC and HAC act as mixed-type inhibitors. Electrochemical impedance testing revealed a notable rise in charge transfer resistance (Rct) readings to 4930-Ω cm2, alongside a reduction in the Constant Phase Element (CPE) value to 5.81 μF, suggesting increased effectiveness in preventing corrosion. Also, density functional theory (DFT) was applied to investigate the assembled tri-thiosemicarbazones HBC and HAC. Moreover, the adsorption mechanism of HBC and HAC on the mild steel surface was explored using Monte Carlo simulation. Finally, the theoretical outputs were discovered to support the practical outcomes.

Abstract Purpose Numerous research has reported that total knee arthroplasty (TKA) cannot reproduce axial tibial rotations of normal knees. The objective of this study was to measure the tibiofemoral articular contact motions and axial tibial rotations of TKA knees to investigate the mechanism causing the knee kinematics change of after TKAs. Methods Eleven patients with unilateral cruciate retaining (CR) TKA were tested for measurements of knee motion during a weight-bearing flexion from 0° to 105° using an imaging technique. The tibiofemoral contact kinematics were determined using the contact points on medial and lateral surfaces of the tibia and femoral condyles. Axial tibial rotations were calculated using the differences between the medial and lateral articulation distances on the femoral condyles and tibial surfaces at each flexion interval of 15°. Abstract

Condensation of pyrazine-2,3-dicarboxylic acid anhydride with aminoacetophenones gave the corresponding N-(acetylphenyl)pyrazine-2-carboxamide. Their reactions with some electrophilic (carbonyl group, bromine) and nucleophilic (malononitrile hydrazine) reagents are discussed. N-[3-(2,2-Dicyano-1-methylethenyl)phenyl]pyrazine- 2-carboxamide underwent Michael addition reaction with an activated double bond yielding 3-amino-5-aryl-3’-(pyrazine-2-carboxamido)biphenyl-2,4-dicarbonitrile derivatives and 2-imino-4-(3-(pyrazine-2-carboxamido)phenyl)-6-(4-methoxyphenyl)cyclohexa 3,5-diene-1,1,3-tricarbonitrile. The structures of the newly synthesized compounds were confirmed by their IR, 1H NMR, 13C NMR and mass spectra data. Antimicrobial acitivites and Leuconostoc sp. growth inhibition of the newly synthesized compounds were studied and some of these were found to exhibit a pronounced effect.

Abstract

Ethylidene hydrazine (4a,b) and thiazolidin-4-one (5) derivatives were synthesized by the reaction of ethylidenethiosemicarbazide derivative (3a) with α-haloketone/ethyl bromoacetate, respectively. Hetrocyclization of ethylideneacetohydrazide derivative (7) with o-phenolic aldehydes gave the corresponding coumarin derivatives (8,9). The interaction of 7 with acetylacetone afforded the corresponding pyridine derivative (10). Treatment of the arylidene derivative 11b with malononitrile afforded the corresponding pyran derivative (12). The new products 3-12 were subjected to IR, 1H NMR and mass spectra studies.

Abstract

Introduction: Psoriasis is a chronic inflammatory autoimmune skin disease. Several treatment options are available including topical and systemic options. Methotrexate was the main systemic medication in treating severe psoriasis, yet adverse events can limit its use. Leflunomide is an isoxazole derivative that inhibits the synthesis of pyrimidines, and subsequently inhibits RNA and DNA synthesis. Objective: As available data directly comparing MTX to leflunomide in psoriasis are lacking, this double blinded study was designed to compare the efficacy of methotrexate versus leflunomide in the treatment of moderate to severe psoriasis. Methods: The study included 40 patients (25 males and 15 females) with chronic plaque psoriasis, s. Patients were randomly assigned to one of two equal groups, group A for subcutaneous methotrexate injections and group B for leflunomide (loading dose 100mg daily for the first 3 days then 20 mg daily for 3 months. Disease severity was determined by psoriasis area and severity index (PASI) score before and at the end of treatment The treatment response was evaluated at the baseline and weeks 4, 8 and 12 PASI score. Results: Both groups were matching at the baseline in aspects of gender, age, disease duration and PASI scores Both medications yielded comparable results with no significant difference between both groups in PASI score neither in side effects.. Conclusion: Leflunomide can be as effective as methotrexate in treatment of moderate to severe psoriasis.

47. A microcrystalline cellulose/metal-organic framework hybrid for enhanced ritonavir dispersive solid phase microextraction from human plasma

Abstract

A hybrid of microcrystalline cellulose (MCC) with metal organic frame (MOF) was developed as an efficient adsorbent for dispersive solid phase microextraction (dSPME) of ritonavir from spiked human plasma. The developed sorbent was characterized by a high-resolution scanning electron microscope (HR-SEM), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR). The MCC was immersed in a solution of CaCl2, stirred with soda water and 1,3,5-benzenetricarboxylic acid, resulting in white MCC/MOF composites, which were separated, washed, and dried. The MCC/MOF hybrids were examined under the HR-SEM and showed morphological features different form the MCC and Ca-1,3,5-benzenetricarboxylic acid (Ca-BTC) MOF. The diffraction patterns of Ca-BTC/MCC composites clearly displayed the characteristic Ca-BTC MOF diffraction bands, indicating that MCC was successfully incorporated in the formation of crystalline MOF hybrids. The FTIR spectra exhibited the bands of MCC, as well as the bands of Ca-BTC MOFs. The prepared Ca-BTC/MCC MOF was applied for dSPME of ritonavir from human plasma, before the determination by high performance liquid chromatography with UV detection. After method optimization, the best extraction efficiency was achieved by using 12.50 mg of Ca-BTC/MCC MOF as a sorbent and 250 µL of acetonitrile as an eluent. This novel sorbent (Ca-BTC/MCC) merges the advantages of the high surface areas of MOFs 820 m2 g−1 with the biodegradability and sustainability of MCC. Besides, Ca-BTC/MCC exhibited higher extraction efficiency compared with unmodified Ca-BTC MOF. To the best of our knowledge, this work applies the Ca-BTC/MCC sorbents for dSPME of ritonavir in human plasma, for the first time. These results open the door to more applications of these composites in sample preparation for biomedical analysis.

48. Bee venom: A potential natural alternative to conventional preservatives for prolonging the shelf-life of soft cheese ‘Talaga’

Abstract

The study aims to explore bee venom (honey-BV) as a potential natural preservative for ''Tallaga'' soft cheese. Characterization of the active compounds in honey-BV was conducted via chromatographic analyses. Antimicrobial efficacy against pathogenic bacteria and fungi was evaluated, and minimum inhibitory concentration (MIC) was determined. Subsequently, honey-BV was applied to Tallaga cheese at 15 mg/g concentrations. The main active ingredients identified in bee venom were apamin (2%) and melittin (48.7%). Both concentrations of bee venom (100 and 200 mg/mL) exhibited significant antifungal and antibacterial properties against tested organisms, with MIC values varied from 0.2 to 0.5 mg/mL for bacteria to 3–13 mg/mL for fungi. Application of honey-BV in Tallaga cheese resulted in complete elimination of Staphylococcal populations after 2 weeks of cold storage, with no detectable growth of molds or yeasts throughout the storage period. Additionally, a steady decrease in aerobic plate count was observed over time. In summary, honey-BV holds promise as a natural preservative for soft cheese, however, more investigation is required to optimize the concentration for economic viability, taking into account health benefits and safety considerations.

In this process, many algal bioactive compounds, such as proteins, carbohydrates, lipids, alkaloids, flavonoids and phenols, can catalyze the reduction of metal ions to NPs. In addition, many driving factors, including pH, temperature, duration, static conditions and substrate concentration, are involved to facilitate the green synthesis of algal-based NPs. Here, the biosynthesis, mechanisms and applications of algal-synthesized NPs in cancer therapy have been critically discussed.
