Abstract

Taxol and other antimitotic agents are frontline chemotherapy agents but the mechanisms responsible for patient benefit remain unclear. Following a genome-wide siRNA screen, we identified the oncogenic transcription factor Myc as a taxol sensitizer. Using time-lapse imaging to correlate mitotic behavior with cell fate, we show that Myc sensitizes cells to mitotic blockers and agents that accelerate mitotic progression. Myc achieves this by upregulating a cluster of redundant pro-apoptotic BH3-only proteins and suppressing pro-survival Bcl-xL. Gene expression analysis of breast cancers indicates that taxane responses correlate positively with Myc and negatively with Bcl-xL. Accordingly, pharmacological inhibition of Bcl-xL restores apoptosis in Myc-deficient cells. These results open up opportunities for biomarkers and combination therapies that could enhance traditional and second-generation antimitotic agents.

In this paper we describe qualitatively some aspects of the holographic QCD. Inspired by a successfull 4D description, we try to separate the Confinement and the Chiral Symmetry Breaking dynamics. We also discuss the realization of the baryons as skyrmions in Soft Wall Holographic QCD, and the issue of the Vector Meson Dominance.

14. Statistical Hadronization and Holography

Abstract

In this paper we consider some issues about the statistical model of the hadronization in a holographic approach. We introduce a Rindler like horizon in the bulk and we understand the string breaking as a tunneling event under this horizon. We calculate the hadron spectrum and we get a thermal, and so statistical, shape for it.

Abstract

We consider a five dimensional SU(2) gauge theory with fermions in the bulk and with additional SU(2) and U(1) kinetic terms on the branes. The electroweak breaking is obtained by boundary conditions. After deconstruction, fermions in the bulk are eliminated by using their equations of motion. In this way Standard Model fermion mass terms and direct couplings to the internal gauge bosons of the moose are generated. The presence of these new couplings gives a new contribution to the epsilon_3 parameter in addition to the gauge boson term. This allows the possibility of a cancellation between the two contributions, which can be local (site by site) or global. Going back to the continuum, we show that the implementation of local cancellation in any generic warped metric leaves massless fermions. This is due to the presence of one horizon on the infrared brane. However we can require a global cancellation of the new physics contributions to the epsilon_3 parameter. 20. Visual imagery ability in the continuum of Alzheimer's disease: the mind's eye compared in patients with MCI and early AD.

Abstract

Background: Visual Mental Imagery (VMI) is the ability to represent stimuli in the mind without sensory visual input. Previous studies have shown alterations in visual imagery in Alzheimer's disease (AD). However, VMI has not been investigated in the AD prodromal stage, i.e., the Mild Cognitive Impairment (MCI). In this study, we aimed to investigate the VMI ability in AD and MCI patients. We hypothesized that VMI ability could be compromised from the early disease stage. Method: For our purpose, we enrolled 14 patients with a diagnosis of probable typical AD, 19 amnestic MCI (aMCI), and 23 healthy control subjects (HC), matched for sex, age, and education; MMSE scores in AD patients significantly differed from those in aMCI and HC, that not differed between them. The extensive assessment of VMI included: 1) the O'Clock test that allows disentangling the possible role of visuo‐perceptual difficulties in the VMI task's performance; 2) a modified version of The Complete Visual Mental Imagery Battery (CVMIB), including tasks evaluating the different VMI processes (Kosslyn, 1980; 2005), that is generation, maintenance, inspection and transformation of different types of visual mental images (i.e., objects, buildings and faces). Result: Results indicated that AD patients performed worse than HC in both perceptual and imaginal tasks of the O'Clock test and in all CVMIB's tasks but maintenance. On the contrary, aMCI patients showed difficulties in the generation process and the imaginal task of the clock test. Conclusion: Our findings suggested that generation, inspection and transformation processes of VMI are impaired since the early phase of Alzheimer's disease. Moreover, we found that the generation process is selectively impaired in aMCI patients, demonstrating that VMI deficits are already present in the prodromal stage of the disease. Since alterations in VMI processing might reflect the AD neuropathological progression, according to Braak and Braak's model, VMI abilities might represent a useful neuropsychological marker of dementia to identify AD prodromal stage. 25. Type 1 diabetes and inborn errors of immunity: Complete strangers or 2 sides of the same coin?

Abstract

Type 1 diabetes (T1D) is a polygenic disease and does not follow a mendelian pattern. Inborn errors of immunity (IEIs), on the other hand, are caused by damaging germline variants, suggesting that T1D and IEIs have nothing in common. Some IEIs, resulting from mutations in genes regulating regulatory T–cell homeostasis, are associated with elevated incidence of T1D. The genetic spectrum of IEIs is gradually being unraveled; consequently, molecular pathways underlying human monogenic autoimmunity are being identified. There is an appreciable overlap between some of these pathways and the genetic variants that determine T1D susceptibility, suggesting that after all, IEI and T1D are 2 sides of the same coin. The study of monogenic IEIs with a variable incidence of T1D has the potential to provide crucial insights into the mechanisms leading to T1D. These insights contribute to the definition of T1D endotypes and explain disease heterogeneity. In this review, we discuss the interconnected pathogenic pathways of autoimmunity, β-cell function, and primary immunodeficiency. We also examine the role of environmental factors in disease penetrance as well as the circumstantial evidence of IEI drugs in preventing and curing T1D in individuals with IEIs, suggesting the repositioning of these drugs also for T1D therapy.
