Venezuela is a country where indigenous and local communities use more than 1500 species of plants for medicinal purposes. Anticancer drugs derived from plants may act through a direct cytotoxic effect on tumor cells or through other mechanisms such as inhibitory effects on the proteases involved in tumor growth and spread. From our databases of botanical collections and ethnobiological usage, we selected 17 species to conduct an initial survey of cytotoxicity on eight human tumor cell lines (representing lung, breast, colon, and pancreas) and activity against four proteases. Thirteen extracts from 10 species were cytotoxic, at 100mg=ml or less, on one or more cell lines. Extracts of Jacaranda copaia and Tapirira guianensis were active against several cell lines, whereas extracts of Gnetum nodiflorum, Protium heptaphyllum, Protium unifoliolatum, Costus scaber ,a ndCroton cuneatus were potent and selectively inhibitory for one or two cell lines. Antiprotease activity against one or more enzymes was detected in extracts from 14 plant species, with most activity being directed against leucine aminopeptidase (LAP). The activities observed are discussed in relation to the chemical constituents reportedly isolated from these plants and=or other species of the genus and their traditional uses.

The olfactory-trigeminal response produced by two odor stimuli, isopropyl alcohol and octane, was studied in awake, restrained rabbits with recording electrodes permanently implanted in one olfactory bulb and in the anterior sensory motor cortex for measurement of spontaneous activity. Surface electrodes were used to measure respiration and EKG. Xylocaine hydrochloride injected via cannulae permanently implanted at the root of both trigeminal nerves was used as a blocking agent. That the results obtained with the xylocaine-treated rabbits after odor stimulation were due to trigeminal block, and not to changes in the central nervous system by infiltration of the drug to adjacent tissue, was proven by the results obtained with an animal whose trigeminal nerves were lesioned with electrical current. The response to odorants was characterized by changes in respiratory and heart rates, by a typical cortical “arousal” reaction, and by changes in bulbar activity. Blocking the trigeminal nerves by injection of xylocaine or by lesioning produced changes only in the cortical EEG and bulbar spontaneous activity, according to stimulus concentration. Respiration and heart rate remained unchanged during odor presentation. These data support the hypothesis that the olfactory response to many odorants is really an “olfactory-trigeminal” response. Further, the aversive response of the rabbit to high concentrations of odor stimuli is inhibited when the trigeminal nerves are blocked.

The optic tectum of Eugerres and Holocentrus was electrically stimulated, and field and unitary potentials were recorded from nucleus prethalamicus and nucleus isthmi; field potentials were also recorded from dorsolateral tegmental nuclei and corpus glomerulosum. Recording sites were verified by dye markings. The responses found are characteristics of, and limited to, each cell group in particular, and can thus be used for accurate location of such cell groups in the course of recording, stimulating or injecting experiments. In general, tectal stimulation leads to postsynaptic excitation at target nuclei. This is followed by further excitation mixed with inhibition in the case of nucleus prethalamicus. In the case of nucleus isthmi, excitation leads to repetitive firing which appears to arise at the dendrites and to be synchronized with firing of neighboring neurons.

RELATIVELY few techniques are available for the investigation of the long-term effects of physiological variables on conduction times and synaptic delay in the brain. The lateral olfactory tract (LOT) and prepyriform cortex provide a system whereby these parameters may be measured in rats chronically implanted with electrodes1. Fibres of the LOT synapse with dendrites of the prepyriform pyramidal cells2. When the LOT is electrically stimulated by a single shock, both the presynaptic LOT potential and the postsynaptic prepyriform response may be recorded from electrodes placed in the prepyriform cortex (Fig. 1). As the LOT potential first approaches and then invades the recording area, the adjacent cortex acts first as a current source and then as a sink; hence the presynaptic potential is biphasic—first positive, then negative. The time from stimulus artefact to negative crest of the presynaptic potential is a measure of conduction time along the LOT and the time from the negative peak to beginning of the prepyriform response is a measure of synaptic delay (Fig. 1).

The effect of reversible blocking of the trigeminal nerve on olfactory stimulation was studied in rabbits with chronically implanted electrodes. In one series of experiments, blocking the trigeminal nerves at the level of the ganglia increased olfactory bulb excitability, but no cortical desynchronization was evident on odor presentation. Respiration and heart rate remained relatively unchanged during odor presentation. In the trigeminally blocked animal, olfactory bulb-induced sinusoidal wave activity was significantly increased in frequency and amplitude when compared with this activity in the unblocked animal following odor presentation. The present evidence supports the concept of a central regulatory control over olfactory afferent inputs; the trigeminal nerve plays an important role in this system.

The focal stimulation technique applied to the squirrel monkeys revealed two areas that yield maximal respiratory responses with threshold intensity of stimulation: one area is located within the limits of the medulla oblongata and the other within the dorsal part of the pons. In dogs, only the medullary area gave maximal respiratory responses (except for two expiratory responses in the pons of one animal). By focal recording, numerous respiratory neurons were found in the medulla of both species, but the same technique failed to disclose any such neurons in the pons of either. It was concluded that the dorsal pons of the squirrel monkey probably plays a more active role in the control of respiration than its counterpart in the dog.
