Madariaga virus (MADV) is a member of the eastern equine encephalitis virus (EEEV) complex that circulates in Central and South America. It is a zoonotic, mosquito-borne pathogen, belonging to the family Togaviridae. Disturbances in the natural transmission cycle of this virus result in outbreaks in equines and humans, leading to high case fatality in the former and acute febrile illness or neurological disease in the latter. Although a considerable amount of knowledge exists on the eco-epidemiology of North American EEEV strains, little is known about MADV. In Brazil, the most recent isolations of MADV occurred in 2009 in the States of Paraíba and Ceará, northeast Brazil. Because of that, health authorities have recommended vaccination of animals in these regions. However, in 2019 an equine encephalitis outbreak was reported in a municipality in Ceará. Here, we present the isolation of MADV from two horses that died in this outbreak. The full-length genome of these viruses was sequenced, and phylogenetic analyses performed. Pathological findings from postmortem examination are also discussed. We conclude that MADV is actively circulating in northeast Brazil despite vaccination programs, and call attention to this arbovirus that likely represents an emerging pathogen in Latin America.

Background: West Nile virus (WNV) was first sequenced in Brazil in 2019, when it was isolated from a horse in the Esp?rito Santo state. Despite multiple studies reporting serological evidence suggestive of past circulation since 2004, WNV remains a low priority for surveillance and public health, such that much is still unknown about its genomic diversity, evolution, and transmission in the country. Methods: A combination of diagnostic assays, nanopore sequencing, phylogenetic inference, and epidemiological modeling are here used to provide a holistic overview of what is known about WNV in Brazil. Results: We report new genetic evidence of WNV circulation in southern (Minas Gerais, S?o Paulo) and northeastern (Piau?) states isolated from equine red blood cells. A novel, climate-informed theoretical perspective of the potential transmission of WNV across the country highlights the state of Piau? as particularly relevant for WNV epidemiology in Brazil, although it does not reject possible circulation in other states. Conclusion: Our output demonstrates the scarceness of existing data, and that although there is sufficient evidence for the circulation and persistence of the virus, much is still unknown on its local evolution, epidemiology, and activity. We advocate for a shift to active surveillance, to ensure adequate preparedness for future epidemics with spill-over potential to humans.

Equine coital exanthema is a venereal infectious disease poorly reported in horses in Brazil and was never described in the northeastern region of the country. This work aims to describe the clinical and pathological aspects of an outbreak of equine coital exanthema caused by equid alphaherpesvirus 3, occurred in a herd of horses at the semiarid region of the State of Rio Grande do Norte. Main clinical signs consisted of anorexia, hiporexia, fibrinous or purulent secretion in the penis mucosa and vagina. Two mares presented mild to minimal lesions that consisted of scars in the mucosa of the vagina and in the perivulvar region. In a stallion the disease consisted of severe, multifocal, umbilicated-exanthematous ulcers of approximately 1cm in diameter on the penis mucosa. Other areas where ulcers and crusts were focally observed included the skin of the scrotum and on the lips and mucocutaneous junctions of the oral cavity. Histologically, the main lesion consisted of multifocal severe ulcerative and fibrinous necrotizing balanoposthitis and mild multifocal necrotizing, lymphocytic dermatitis in the lips and scrotum. The equide alphaherpesvirus 3 DNA was amplified in blood samples and penis mucosa using the PCR technique. This is the first report of molecular diagnosis of equine coital exanthema affecting horses in northeastern Brazil. Further studies should be carried out in order to investigate the epidemiology and the importance of this herpetic disease in the country. RESUMO: O exantema coital equino é uma doença infecciosa venérea pouco relatada em equinos no Brasil e nunca descrita na região Nordeste do país. Este trabalho tem como objetivo descrever os aspectos clínicos e patológicos de um surto de exantema coital equino causado pelo alphaherpesvirus equídeo 3, que ocorreu em um haras na região semiárida do Estado do Rio Grande do Norte. Os principais sinais clínicos consistiram em anorexia, hiporexia, secreção fibrinosa ou purulenta na mucosa do pênis e vagina. Duas éguas apresentavam lesões discretas que consistiam em cicatrizes na mucosa da vagina e na região perivulvar. Em um garanhão, a doença consistia em úlceras umbilicadas-exantematosas severas, multifocais, de aproximadamente 1 cm de diâmetro na mucosa do pênis. Outras áreas onde úlceras e crostas foram observadas focalmente incluíram a pele do escroto, lábios e junções mucocutâneas da cavidade oral. Histologicamente, as principais lesões consistiam em balanopostite multifocal ulcerativa e necrosante fibrinosa grave e dermatite linfocítica necrosante multifocal leve nos lábios e escroto. O DNA do alphaherpesvirus equídeo tipo 3 foi amplificado em amostras de sangue e mucosa do pênis pela técnica de PCR. Este é o primeiro relato de diagnóstico molecular de exantema coital equino afetando cavalos no nordeste do Brasil. Novos estudos devem ser realizados a fim de investigar a epidemiologia e a importância dessa doença herpética no país.

O vírus do Nilo Ocidental, responsável pela doença chamada Febre do Nilo Ocidental (FNO) foi identificado no Brasil pela primeira vez em 2009 e, desde então, a presença de anticorpos contra o vírus e/ou do seu ácido nucleico foi identificado no homem, equídeos e aves. O vírus é responsável por infecções neurológicas em aves susceptíveis, em equídeos e em seres humanos, sendo considerado um problema global de saúde pública. O controle da FNO está intimamente relacionado com a atuação dos médicos veterinários na vigilância de casos nos equídeos, animais considerados sentinelas na identificação da doença.

The swine influenza A virus (SIAV) subtypes/lineages H1N1pdm09, H3N2, H1N2, and H1N1 of seasonal human origin are widespread in Brazilian swine herds. A monovalent inactivated H1N1pdm09 vaccine was licensed in Brazil in 2014. However, there are concerns about its efficacy due to the limited vaccine cross-protection against heterologous viruses and the potential for exacerbated reactions against vaccine strains. Thus, monitoring SIAVs subtypes/lineages that are circulating in the Brazilian swine population is important, by applying a fast and efficient diagnostic test in herd field samples. A RT-PCR assay was developed, using primers specific for HA subtyping of Brazilian SIAV, and was used to evaluate the occurrence of subtypes from samples collected between 2012 and 2019. From 167 field samples positive for influenza A, 117 were subtyped by nested RT-PCR assay. A higher occurrence of H1N1pdm was observed from 2012 to 2015, H3N2 in 2017, and H1hu in 2017 to 2019. A hemagglutination inhibition test was performed in serum samples received from 2017 to 2019, confirming these data. The molecular data highlights the importance of H1hu and H3N2 detection since there are no vaccines available for the subtypes/lineages and raises an alert of H1hu for its potential to infect humans. Serological data suggest a cyclical profile of occurrence between the H3N2 and H1N1pdm over time. Monitoring SIAVs circulating in Brazilian swine herds is necessary, which provides the relevant information for field veterinarians to apply effective control measures on the properties. The breeding practices adopted and the equipment typically found in animal facilities produce sounds at frequencies within the auditory range of the mice (1 to 100 kHz), which can cause hearing and other non-hearing effects. Another aspect that could potentially affects the welfare of experimental animals would be their impaired health condition, since in addition to all the variables present in the environment, some of them are deliberately infected with pathogens. This study aimed to evaluate the possible effects of the chronic exposure of C57BL/6 and Tlr2/Tlr9(−/−) mice, uninfected and infected, with a low m.o.i. of Herpes simplex-1, at different noise levels present in the “Quiet” and “Noisy” rooms. Considering all procedures, a total of 51 male mice were used, 27 of the C57BL/6 and 24 of the Tlr2/Tlr9(−/−) strains. Physiological parameters such as weight gain, hemogram, cholesterol, glucose,corticosterone and cytokines involved in the immune response were evaluated together with the animals’ behavioral responses in Open Field and Light/Dark tests. COVID-19 patients can excrete viable SARS-CoV-2 virus via urine and faeces, which has raised concerns over the possibility of COVID-19 transmission via aerosolized contaminated water or via the faecal-oral route. These concerns are especially exacerbated in many low- and middle-income countries, where untreated sewage is frequently discharged to surface waters. SARS-CoV-2 RNA has been detected in river water (RW) and raw wastewater (WW) samples. However, little is known about SARS-CoV-2 viability in these environmental matrices. Determining the persistence of SARS-CoV-2 in water under different environmental conditions is of great importance for basic assumptions in quantitative microbial risk assessment (QMRA). In this study, the persistence of SARS-CoV-2 was assessed using plaque assays following spiking of RW and WW samples with infectious SARS-CoV-2 that was previously isolated from a COVID-19 patient. These assays were carried out on autoclaved RW and WW samples, filtered (0.22 µm) and unfiltered, at 4°C and 24°C. Linear and nonlinear regression models were adjusted to the data. The Weibull regression model achieved the lowest root mean square error (RMSE) and was hence chosen to estimate T90 and T99 (time required for 1 log and 2 log reductions, respectively). SARS-CoV-2 remained viable longer in filtered compared with unfiltered samples. RW and WW showed T90 values of 1.9 and 1.2 day and T99 values of 6.4 and 4.0 days, respectively. When samples were filtered through 0.22 µm pore size membranes, T90 values increased to 3.3 and 1.5 days, and T99 increased to 8.5 and 4.5 days, for RW and WW samples, respectively. Remarkable increases in SARS-CoV-2 persistence were observed in assays at 4°C, which showed T90 values of 7.7 and 5.5 days, and T99 values of 18.7 and 17.5 days for RW and WW, respectively. These results highlight the variability of SARS-CoV-2 persistence in water and wastewater matrices and can be highly relevant to efforts aimed at quantifying water-related risks, which could be valuable for understanding and controlling the pandemic., Graphical Abstract Image, graphical abstract

West Nile virus (WNV) is a neurovirulent mosquito-borne Flavivirus that is maintained in nature by a zoonotic transmissioncycle between avian hosts and ornithophilic mosquito vectors, mostly from the Culex genus. Until the 1990s, WNV wasconsidered to be an old-world arbovirus, but in 1999, WNV emerged in the United States (US) and spread rapidly, becoming amajor threat to public health. WNV adapted to the transmission cycle involving American mosquitoes and birds and reachedCentral and South America in subsequent years. In 2003, the National West Nile Fever Surveillance System was created in Brazilbased on serological screening of animals and sentinel vectors, as recommended by the Pan American Health Organization(PAHO) and the World Health Organization (WHO). Since 2008, serological evidence of WNV infection in Brazilian horseshas been reported, and the circulation of WNV has been monitored through the regular serological screening of sentinel horsesand reporting of encephalomyelitis cases. Horses are highly susceptible to WNV infection, and outbreaks of neurologicaldisease among horses often precede human cases. In this regard, equine surveillance has been essential in providing earlywarning to public and animal health authorities in several countries, including Brazil. This demonstrates the need for animaland public health intervention programs to allocate resources to make veterinarians aware of the role they can play in thehuman surveillance processes by monitoring horses. This review discusses the importance of equine surveillance and the gapthat veterinarians can fill on the front line in human surveillance, in Brazil and worldwide, in the context of “One Health”

Background Herpes simplex virus type 1 (HSV-1) cause not only mild symptoms but also blindness and encephalitis. It was previously shown that the immune response against HSV-1 occurs mainly in the trigeminal ganglia (TG) and that Toll-like receptors 2 and 9 (TLR2/9) are important in mediating this response. It was also demonstrated that iNOS (nitric oxide synthase) and interleukin 1 beta (IL-1β) play an essential role in the defense against HSV-1 infection. Importantly, the present work aimed to identify the primary cells responsible for iNOS and IL-1β production and search for other important molecules and cells that might or might not depend on TLR2/9 receptors to mediate the immune response against HSV-1. Methods C57BL/6 (wild type, WT) and TLR2/9−/− mice were infected by the intranasal route with HSV-1 (1 × 106 p.f.u.). Cells were obtained from the TG and spleen tissues and the profile of immune cells was determined by flow cytometry in infected and mock infected WT and knockout mice. The percentage of cells producing iNOS, IL-1β, granzyme B and perforin was also determined by flow cytometry. Chemokine monocyte chemoattractant protein-1 (MCP1) was measured by Cytometric Bead Array (CBA) in the TG, spleen and lung. Expression of type I interferons (IFNs), interleukins (IL) 5 and 10, IL-1β and granzyme B were quantified by real time PCR. Results The results indicate that dendritic cells (DCs) and monocytes/macrophages (Mo/Mϕ) were the main sources of IL-1β and iNOS, respectively, which, together with type I IFNs, were essential for the immune response against HSV-1. Additionally, we showed that granzyme B produced by CD8+ T and NK lymphocytes and MCP-1 were also important for this immune response. Moreover, our data indicate that the robust production of MCP-1 and granzyme B is either TLR-independent or down regulated by TLRs and occurs in the TG of TLR2/9−/− infected mice. Conclusion Taken together, our data provide strong evidence that the responses mediated by DCs, Mo/Mϕ, NK and CD8+ T lymphocytes through IL-1β, iNOS and granzyme B production, respectively, together with the production of type I IFN early in the infection, are crucial to host defense against HSV-1. Foi demonstrado anteriormente que a resposta imune contra o HSV-1 ocorre principalmente nos gânglios do trigêmeo (TG) e que os receptores Toll-like 2 e 9 (TLR2/9) são importantes na mediação dessa resposta. Também foi demonstrado que a iNOS (óxido nítrico sintase) e a interleucina 1 beta (IL-1β) desempenham um papel essencial na defesa contra a infecção pelo HSV-1. É importante ressaltar que o presente trabalho teve como objetivo identificar as células primárias responsáveis ​​pela produção de iNOS e IL-1β e buscar outras moléculas e células importantes que podem ou não depender de receptores TLR2/9 para mediar a resposta imune contra o HSV-1. Métodos Camundongos C57BL/6 (tipo selvagem, WT) e TLR2/9−/− foram infectados pela via intranasal com HSV-1 (1 × 106 p.f.u.). As células foram obtidas dos tecidos TG e do baço e o perfil das células imunes foi determinado por citometria de fluxo em camundongos WT e knockout infectados e simulados. A porcentagem de células produtoras de iNOS, IL-1β, granzima B e perforina também foi determinada por citometria de fluxo. A proteína quimioatraente de monócitos quimiocina-1 (MCP1) foi medida por Cytometric Bead Array (CBA) no TG, baço e pulmão. A expressão de interferons tipo I (IFNs), interleucinas (IL) 5 e 10, IL-1β e granzima B foram quantificados por PCR em tempo real. Resultados Os resultados indicam que células dendríticas (DCs) e monócitos/macrófagos (Mo/Mϕ) foram as principais fontes de IL-1β e iNOS, respectivamente, que, juntamente com os IFNs tipo I, foram essenciais para a resposta imune contra HSV-1. Adicionalmente, mostramos que a granzima B produzida por linfócitos T CD8+ e NK e MCP-1 também foram importantes para esta resposta imune. Além disso, nossos dados indicam que a produção robusta de MCP-1 e granzima B é independente de TLR ou regulada negativamente por TLRs e ocorre no TG de camundongos infectados com TLR2/9−/−. Conclusão Em conjunto, nossos dados fornecem fortes evidências de que as respostas mediadas por DCs, Mo/Mϕ, NK e linfócitos T CD8+ através da produção de IL-1β, iNOS e granzima B, respectivamente, juntamente com a produção de IFN tipo I no início da infecção, são cruciais para a defesa do hospedeiro contra o HSV-1.

Complexes of Eu3+, Gd3+, and Tb3+ with tropolone (tropolone=2-hydroxy-2,4,6-cycloheptatrien-1-one) were synthesized and characterized by C and H elemental analysis, infrared vibrational spectroscopy, X-ray powder diffraction analysis and Raman vibrational spectroscopy. The results show that the complexes are isostructural and have the formula [Ln(trop)3], where Ln=Eu3+, Gd3+ or Tb3+ and trop=tropolone. The characteristic luminescence of the Eu3+ and Tb3+ ions was not observed either upon ligand excitation (300–380nm) or upon direct f–f excitation, not even at 4.2 K. In order to understand this phenomenon the spectroscopic properties of the Gd(trop)3 complex (absorption spectra at 298 K, excitation and emission spectra from 4.2 to 298 K and decay time measurements from 20 to 298 K) were investigated. The emission spectra show three distinct bands, which are ascribed to ligand electronic states: a singlet state, S1 (π–π*), at 24 270 cm−1, and two triplet states, T1 and T2, at 15 630cm−1 and 16 610cm−1, respectively. Both triplet states are situated below the emitting states of the Eu3+ and Tb3+ ions (5Do and 5D4, respectively), thus suppressing the luminescence from these states by back-transfer.

CAPES - Coordenação de Aperfeiçoamento de Pessoal de Nível Superior A dengue é uma das principais arboviroses que afeta o ser humano e um grave problema de saúde pública. Os quatro sorotipos do Dengue virus (DENV1, 2, 3 e 4) são transmitidos através da picada de mosquitos fêmea do gênero Aedes. A vacina disponível atualmente apresenta lacunas imunológicas, o que motiva a continuidade de pesquisas em busca de uma vacina eficaz. O vírus Vaccinia Ankara Modificado (MVA) está entre os vetores virais mais promissores e intensamente estudados mundialmente e é considerado como uma excelente ferramenta para a geração de vacinas. O objetivo dessa pesquisa foi construir MVA recombinantes expressando a proteína E de DENV1, 2 e 4 e avaliar o potencial protetor dessas construções em modelo murino. Genes sintéticos da proteína E de DENV1, 2 e 4 foram desenhados, otimizados, obtidos comercialmente e subclonados no plasmídeo pLW44, dando origem aos plasmídeos de transferência, utilizados na construção dos vírus recombinantes. Culturas de fibroblasto de embrião de galinha (CEF) ou BHK-21 foram infectadas por MVA e transfectadas pelos plasmídeos de transferência pLW44-DENV1, pLW44-DENV2 ou pLW44-DENV4, separadamente, para a construção dos MVAs recombinantes (rMVAs). Os rMVAs gerados tiveram a sua construção confirmada por PCR, sequenciamento e citometria de fluxo. O potencial protetor do rMVA-DENV1 foi testado em camundongos C57/BL6 em esquema de imunização dose-reforço homólogo, seguido de desafio intracranial com o DENV correspondente. Os animais foram imunizados com 107 pfu ou 108 pfu de rMVA-DENV1 e desafiados com DENV1 Mochizucki. Durante o desafio os animais foram monitorados e avaliados quanto ao seu estado geral de saúde pelo protocolo SHIRPA. Com base nos resultados obtidos podemos concluir que as construções dos diferentes vírus recombinantes se deram de maneira eficaz, sobretudo a de rMVA-DENV1, a única testada quanto à capacidade protetora. Os dados gerados nessa pesquisa serão somados aos obtidos anteriormente para DENV3 visando à obtenção de uma vacina tetravalente contra DENV. Dengue is the main arbovirus that affecting humans and a serious public health problem. The four dengue virus serotypes (DENV1, 2, 3 and 4) are transmitted through the bite of female Aedes mosquitoes. A vaccine available worldwide has immunological gaps, which motivates further research for an effective vaccine. The Modified Vaccinia Ankara Virus (MVA) is among the most advanced and intensely studied viral vectors worldwide, and is considered as an excellent tool for vaccine generation. The aim of this work was to construct recombinant MVAs expressing the protein E of DENV1, 2 and 4 and to evaluate the protective potential of these constructs in a murine model. Synthetic genes coding for protein E from DENV1, 2 and 4 were designed, optimized, commercially obtained and subcloned into plasmid pLW44, giving rise to transfer plasmids used in the construction of recombinant viruses. Chicken embryo fibroblast cultures – CEF, or BHK-21 were infected with MVA and transfected with the transfer plasmids pLW44-DENV1, pLW44-DENV2 and pLW44-DENV4 for the construction of recombinant MVAs (rMVA). The generated rMVAs had their correct construction confirmed by PCR, sequencing and flow cytometry. The protective potential of rMVA-DENV1 was tested in C57 / BL6 mice on a homologous dose-boosting immunization schedule, followed by intracranial challenge with the corresponding DENV. Animals were immunized with 107 pfu or 108 pfu of rVA-DENV1 and challenged with DENV1 Mochizuki. During the challenge, they were monitored evaluated through a SHIRPA protocol. Based on the results we can conclude that the construction of the different recombinant viruses was effective, especially rMVA-DENV1, the only one tested for protective capacity. The data generated in this research can be added to those previously obtained for DENV3 in order to obtain a tetravalent vaccine against DENV.
