Background: Pulmonary arterial hypertension (PAH) is a serious disease characterized by the progressive elevation of the pulmonary arterial resistance, leading to the right ventricular failure and death. Objective: To evaluate the effect of rapamycin (RAPA), a potent cell-cycle inhibitor, on exercise capacity, right ventricular hypertrophy and pulmonary vascular remodelling on rats. Methods: A total of 39 nine-week-old male Wistar rats (160-240 g) were divided into three groups: the control (n = 10), PAH control (n = 15) and PAH-RAPA (n = 14) groups. On the 1st day, 60 mg/kg monocrotaline was injected intraperitoneally to induce PAH in the PAH control group and PAH-RAPA groups. On the 21st day, 3 mg/kg/day RAPA was started orally, and the animals were followed for 35 days. On the 35th day, the exercise capacity of the rats was analysed through a modified forced swimming test. After measuring their right ventricular systolic pressure using an open-chest method, their hearts and lungs were excised and analysed histopathologically for right ventricular hypertrophy and pulmonary vascular remodelling. Results: Rapamycin treatment provided limited and insignificant improvements in exercise capacity, right ventricular systolic pressure and right ventricular hypertrophy of the rats. However, there was significant recovery in the rats' pulmonary artery muscular layer thickness with the RAPA treatment (p < 0.049). On the 35th day, the mortality rate was 0% in the control group, 53.1% in the PAH control group and 42.9% in the PAH-RAPA group. No statistically significant decrease was observed in their mortality rates with the RAPA treatment (p > 0.16); however, a significant recovery was noted in terms of the rats' median life span (p < 0.006). Conclusion: Pulmonary artificial hypertension is a progressive disease that is not curable with current therapies. Rapamycin may have the potential to reverse vascular remodelling and prolong life expectancy in cases of pulmonary hypertension. [ABSTRACT FROM AUTHOR]

The cortical bone is the most severely affected site in patients with primary hyperparathyroidism (PHPT) and thus, a low bone mineral density (BMD) is predominantly observed in distal forearm. Several studies have investigated potential associations between the weight of the gland and bone mineral loss. In this study, we wanted to investigate the relationship between parathyroid adenoma (PTA) volume and bone mineral loss.

Defects/imperfections can occur during manufacturing, assembly, welding, and other processes, which can reduce the critical buckling load. However, the axial buckling load is beyond the scope of this work, and there are many studies on the stiffening effect of longitudinal dents. This concept combined the idea of the dent-to-stiffener evaluation concept for thin-walled steel cylinders. This study aims to transform the dents into artificial dents for a stiffening effect on the buckling phenomena. For this purpose, 37 thin-walled steel cylinder models, including the perfect model, were designed for varying dent shapes, dent widths, dent depths, dent lengths, and dent angles. The study also contributed to the effect of dent parameters on the critical buckling load of thin-walled steel cylinders. In particular, increasing the initial buckling will motivate the industry to convert dents into stiffeners with small artificial touches to enhance the longevity of the structure. The results showed that the introduction of certain artificial dents can significantly increase the critical buckling load of cylinders, thus improving their resistance against buckling, which has significant implications for various industries that use thin-walled steel cylinders in their structures. The proposed simulations for transforming dents into artificial stiffeners can be a valuable tool for enhancing the longevity and safety of thin-walled steel cylinders and other structures.

Recycling and re-usability of waste materials are of great importance in terms of ecological order. Furthermore, cold-formed steel has recently drawn considerable attention. The objective of the present study was to investigate the bending and shear behavior of the composite formed by pouring waste polymer into the cold-formed I and U profile melds after homogenous pulping. The best results for shear and bending strengths were obtained using melted polypropylene (PP). The enhanced adherence between the steel and molted PP increased both shear and bending capacity. Moreover, it was reinforced by Carbon Fiber Reinforced Polymer (CFRP) and Glass Fiber Reinforced Polymer (GFRP) bars to increase the bending and shear behavior of I and U profiles filled with melted waste polymer. Changing the cross-sectional area in I and U beams under the bending moment had a considerable effect on the load at yielding, ultimate strength, displacement values corresponding to these loads, ductility, and energy dissipation capacity. Addition of CFRP to I beams could also significantly increase displacement capacity in free end regions under the shear force. Aspartame (APM) is a non-nutritive artificial sweetener that has been widely used in many products since 1981. Molecular studies have found that it alters the expression of tumour suppressor genes and oncogenes, forms DNA-DNA and DNA-protein crosslinks, and sister chromatid exchanges. While these results confirm that aspartame is a carcinogenic substance, other studies have failed to detect any negative effect. The present work was aimed to reveal the molecular mechanisms of APM's effects in the simpler model organism, Schizosaccharomyces pombe, which has cellular processes similar to those of mammals. The human HP1 (heterochromatin protein 1) family ortholog swi6 was selected for the evaluation because swi6 expression is downregulated in cancer cells. Swi6 is a telomere, centromere, and mating-type locus binding protein which regulates the structure of heterochromatin. To verify whether the carcinogenic effects of APM are linked with Swi6, S. pombe parental and swi6Δ strains were analysed through a number of tests, including cell viability, intracellular oxidation, glucose consumption, nucleus DAPI (4',6-diamidino-2-phenylindole) staining, and quantitative real time polymerase chain reaction (qRT-PCR) methods. Based on the results, the S. pombe parental strain adapts to APM effects by activating the stress response pathway, while swi6Δ did not show a meaningful response. Thus, it is proposed that there is a relationship between APM and Swi6, and that APM may be carrying out its effects through Swi6. Nevertheless, it is understood that aspartame is not an effective carcinogenic agent since its effects are weak when compared with the swi6Δ phenotype. [ABSTRACT FROM AUTHOR]

Purpose: The implantable bladder cancer (BC) models allow the researchers to perform rapid and useful experiments for BC. We investigated the implantation success of BC cells obtained from Wistar rats (grown in vitro), into bladders of syngeneic Wistar rats, which are commonly used in the laboratories.

Objective: The purpose of this study is to provide reference material for further regulations which are to be made concerning the opinions of youth about the possible impact of various warning labels. Material and Method: 494 high school students in Ankara were included in the study. A questionnaire, which measures the impact of text-only and graphic warning labels, was distributed under supervision. Participants were asked to evaluate the efficiency of 16 text-only warning labels with the Likert scaling and state the most efficient graphic amongst three for each of these text-only labels. Results: 'Smoking during pregnancy harms your baby' was chosen as the most efficient text-only warning label by 70%of the participants. 'Smoking can damage sperm and decrease fertility' and 'Smoking may reduce the blood flow and cause impotence' were the second and third most efficient text-only warning labels amongst all participants, with a selection percentage of 61.9 and 61.1 respectively. The first message above was considered as the most efficient by more female participants than males, whereas the second and third labels were considered the most efficient by more males than females. The pictures with the most graphic content seemed to be preferred. Conclusion: Participants found the esthetic and sex content messages more efficient than death and disease content ones, which they felt were not yet their concern . Objectives: This study was conducted to determine the prevalence and risk factors of low back pain in pregnancy period. Methods: The study group was a cross sectional pregnant population admitted to health centers and hospitals for routine pregnancy controls. A structured questionnaire, disability indexes and Zung depression scale were administered to pregnant women. Nonparametric tests were used to determine pain sensation before and during the pregnancy. Logistic regression analysis was performed to determine the probable factors which may lead to pain sensation and to specify its severity. Results: This study was completed with 1357 pregnant women with a mean age of 27.1 +/- 9.9 years. The frequency of LBP in our study group was 734 (54.1%). Pain related restriction in daily activities were evaluated by Quebec and Oswestry indexes (respectively Kruskal Wallis; 74.561, p<0.001, Kruskal Wallis; 70.981, p<0.001) and a significant difference was found between those who suffered from pain and who did not. Logistic regression analysis revealed that in subjects who had pain before pregnancy had increased weight, smoking, increased maternal age, the highest Qswestry score and no assistance in their daily activities had both pain and more severe pain. A predominant decrease in disability was observed between fourth and fifth months of pregnancy. Conclusions: Low back pain prevalence in Turkish pregnant women is similar to the prevalence of pain in other countries. Oswestry disability index can be used to detect low back pain related to pregnancy and early physiotherapeutic treatment may be initiated. [ABSTRACT FROM AUTHOR]

The present study was conducted on specific skeletal muscles of six weaned male kids from each of the Angora, Hair, Honamlı, and Kilis goat breeds. The relationships between the expression of myogenic factor 5 (Myf5) and myogenic factor 6 (Myf6) genes and muscle fibre characteristics were analysed. Muscle samples from the longissimus dorsi (LD) and semitendinosus (ST) were collected from six 90-day-old weaned male kids of each breed. Muscle fiber characteristics were assessed through histochemical staining, while expression levels of Myf5 and Myf6 genes were quantified using real-time PCR. Total RNA content in the LD and ST muscles was significantly higher (p < 0.05) in Honamlı kids compared to those of the other breeds. Similarly, the expression of Myf5 gene in Honamlı kids was significantly higher (p < 0.05) than that observed in kids from the other breeds in LD muscle. Conversely, in the ST muscle, Hair kids exhibited a significantly lower expression of Myf5 (p < 0.05) when compared to both Honamlı and Kilis kids Additionally, Kilis kids demonstrated a significant reduced expression of Myf6 gene (p < 0.05) relative to the other breeds. The highest expression levels of the Myf6 gene (p < 0.05) were detected in the ST muscle of Honamlı and Angora kids, significantly surpassing those observed in Hair and Kilis kids. Moreover, significant correlations (p < 0.05) were observed among Myf5 and Myf6 gene expression levels and various muscle fiber characteristics differing across breeds. The results of this study underscore the pivotal role of these myogenic regulatory factors in muscle development, offering insights into the molecular mechanism driving breed-specific muscle growth. Background: Perivascular epithelioid-cell tumour (PEComa) is a rare mesenchymal tumour with low malignant potential. PEComa can be found in many organs throughout the body. In the urinary system, it can be found in the prostate, bladder, and kidney. Given the rarity of bladder PEComas, a multidisciplinary approach is often required for diagnosis, encompassing radiological imaging, histopathological examination, and immunohistochemical staining., Methods: A 3 cm-diameter mass outside the bladder but related to the anterolateral wall of the bladder was excised by laparoscopic method. A minimally invasive approach was selected to minimise the potential for complications and to preserve bladder function., Results: The case was diagnosed as PEComa based on the findings of the histopathological examination. Immunohistochemical analysis demonstrated robust positivity for melanocytic and myogenic markers, thereby substantiating the diagnosis. No clear algorithm for disease management was found. However, the patient, who did not have any malignancy criteria specified in literature, was followed up for about 19 months without any problems., Conclusions: This case highlighted the importance of considering PEComa in the differential diagnosis of bladder masses, even in asymptomatic patients. Further research and case studies are needed to establish a clear management algorithm for PEComa in the urinary system. Understanding the pharmacodynamics of ritonavir through metabolomics offers insights into its side effects and helps in the development of safer therapies. This study aimed to investigate the effects of ritonavir treatment on the metabolic profiles of rabbits via a metabolomics approach, with the objective of elucidating its impact on various biochemical pathways and identifying relevant biomarkers. The rabbits were divided into control and ritonavir-treated groups, and their plasma samples were analyzed via ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC/Q-TOF/MS/MS). Metabolites were identified on the basis of the masscharge ratio (m/z) and validated via XCMS software. Metabolites with a fold change ≥ 1.5 and P ≤ 0.01 were analyzed via principal component analysis (PCA) and orthogonal partial least squares discrimination analysis (OPLS-DA) to distinguish between the groups. MetaboAnalyst 6.0 was used for pathway analysis to identify metabolic pathways affected by ritonavir. The PCA and OPLS-DA models revealed clear separation between the control and ritonavir-treated groups, with high R² and Q² values indicating robust model performance. Pathway analysis revealed that ritonavir treatment significantly affected several metabolic pathways, including those related to ether lipid, phenylalanine, sphingolipid, and glycerophospholipid metabolism. Particularly significant changes were observed in metabolites related to lipid metabolism, oxidative stress responses and cellular signaling. Ritonavir significantly impacts metabolic pathways, particularly those involved in lipid metabolism, and oxidative stress responses, which may influence immune responses and drug interactions. This study also highlights the potential of integrating metabolomics with personalized medicine approaches to optimize ritonavir treatment strategies and reduce adverse effects. These findings indicate that ritonavir significantly influences cellular homeostasis and metabolic processes in addition to its antiviral properties. This highlights the necessity of comprehending the metabolic effects of ritonavir to enhance its clinical application, especially in the management of COVID-19. Background: Parotid gland masses encompassing both neoplastic and non-neoplastic conditions present diagnostic challenges. Fine-needle aspiration cytology (FNAC) is commonly used to differentiate between benign and malignant lesions but has limitations such as variable sensitivity and potential sampling errors. This study assesses FNAC's accuracy for parotid gland lesions, evaluates the Milan Salivary Gland Cytopathology Reporting System (MSRSGC), and compares FNAC results with histopathological findings., Methods: This retrospective study at our medical center analyzed data from 2321 patients who underwent parotid gland FNAC followed by surgical resection over 20 years. Patients were included if they had both cytological analysis and surgery between January 2004 and July 2024. Surgical procedures varied based on mass characteristics and FNAC results, and all samples were assessed using MSRSGC. Statistical analysis compared FNAC results with histopathology findings, calculating sensitivity, specificity, and accuracy., Results: This study included 352 patients who underwent both FNAC and surgical resection. FNAC had an overall accuracy of 93.9%, with a sensitivity of 78.3% and specificity of 91.5%. The risk of malignancy was 30% for Milan Category I (nondiagnostic), 6.3% for Category II (non-neoplastic), 0% for Category III (Atypia of Undetermined Significance), 2.3% for Category IVa (benign neoplasm), 5.8% for Category IVb (neoplasm of uncertain malignant potential), 58.8% for Category V (suspicious for malignancy), and 95% for Category VI (malignant). The false negative rate was 2.4%, while the false positive rate was 3.4%. The malignancy risk increased with age, and malignant masses were larger and more likely to cause facial paralysis., Conclusion: FNAC is a key diagnostic tool for parotid gland masses, offering high specificity and accuracy despite some limitations. Background: Periodic fever, aphthous stomatitis, pharyngitis, and adenitis (PFAPA) syndrome is a recurrent fever syndrome. The exact etiopathogenesis of PFAPA syndrome remains unknown. Biological fluids or tissues may provide disease-specific biomarkers that may help clinicians to find new pathogenic pathways., Methods: Tonsil tissues of seven patients with PFAPA were collected during the tonsillectomy. Seven patients who underwent tonsillectomy for reasons other than chronic tonsillitis were enrolled as a control group. The nHPLC LC-MS/MS system was used for protein identification and label-free quantification. Bioinformatics analysis was carried out using the UniProt accession numbers of the identified proteins., Results: Proteomics analysis revealed to identity of proteins of which at least 23 were up and 57 were downregulated. Bioinformatics analysis of differentially regulated proteins by STRING indicated that protein folding and clearance machinery were interrupted in PFAPA patients compared to the controls. The affected pathways underlined the importance of the mitochondrial electron transport chain and ATP biosynthesis process., Conclusion: Although it is not clear that changes in tonsil protein expression whether directly related to pathogenesis or simply result of chronic inflammation, the identification of tonsil biomarkers for PFAPA may provide clinicians an opportunity to understand disease pathogenesis or develop new molecular targets for treatments., Impact: Proteomics analyses of tonsils revealed the identity of 80 proteins of which at least 23 were up and 57 were downregulated. Background/objectives: At present, there are few biomarkers used to predict the prognosis of uterine serous carcinoma (USC). Netrin-1 may be a promising biomarker candidate. We investigated netrin-1 expression in USC tissues and healthy endometrial tissues to determine its relevance to disease prognosis., Materials and Methods: Netrin-1 expression was examined in the tissues of 48 patients with USC and 30 patients with healthy benign endometrial tissues via immunohistochemistry., Results: None of the healthy tissues were stained with netrin-1. In tumor tissues, the overall positivity rate of netrin-1 was 75%, detected as high expression in 17 patients (35%) and low in 19 (40%). Patients who had tumors with no netrin-1 expression (n = 12) had a median overall survival (OS) of 60.0 months (95% confidence interval [CI], 47-98), whereas patients who had tumors with low to strong netrin-1 expression (n = 33) had a lower median OS of 50 months, but the difference was not statistically significant (95% CI, 58-108; P = 0.531). Disease-free survival (DFS) was not statistically significant between the groups (95% CI, 67.7-115.9; P = 0.566). Patients with a tumor diameter ≥2 cm had higher netrin-1 expression than those with a tumor diameter of 2 cm (P = 0.027). We did not find any difference in overall and DFS when age, tumor stage, histology, tumor diameter, p53 status, lymphovascular space invasion, myometrial invasion, and lymph node metastasis were compared according to netrin-1 expression (P > 0.05)., Conclusion: Netrin-1 was expressed in USC but not in healthy tissues. Objectives: Nasal cavity schwannomas are exceedingly rare, benign neoplasms that pose challenges in clinical differentiation from other nasal tumors. Methods: This study presents 5 cases of nasal cavity schwannoma treated surgically over a 10 year period, along with a review of the literature. Results: The most prevalent symptoms included unilateral nasal obstruction and intermittent nosebleeds. Tumors originated from various nasal sites, including the septum, middle conchae, lateral nasal wall, and alar mucosa. All surgeries were conducted transnasally, with 3 tumors excised en bloc, and the remaining two subjected to piecemeal resection. Ancient schwannoma was identified in 4 cases. No instances of recurrence were observed during the average 61 month follow-up period. Conclusions: The definitive diagnosis of schwannomas necessitates histopathological examination. There is a limited amount of data on the role of programmed cell death ligand (PD-L) -1 and PD-L2 in salivary gland carcinomas. We aimed to evaluate the prognostic value of PD-L1 and PD-L2 expressions, which are closely related to immune mechanisms, with respect to salivary gland tumor types and stages. Data from patients with salivary gland masses surgically removed between 2006 and 2021, diagnosed with a malignant salivary gland neoplasm, were retrospectively analyzed. Immunoreactivity for PD-L1 and PD-L2 was performed on resection materials. The mean age of 90 patients was 52.1±18.8 and 46.7% were male. Overall, 55.6% of patients were diagnosed with adenoid cystic carcinoma (ACC), 23.3% with mucoepidermoid carcinoma (MEC), 16.7% with acinic cell carcinoma (AciCC), 3.3% with ductal carcinoma (DC), and 1 patient with pleomorphic adenoma ex carcinoma (PA-ex-CA). In all, 52% of ACC, 12% of AciCC, 24% of MEC, and 12% of DC cases were at stage IV. The tumor diameter, frequencies of lymphovascular invasion, metastasis, positive surgical margin, recurrence, and mortality rates of patients at stages III and IV were significantly larger than those at stages I and II ( P <0.05). The percentages of tumor cell score (TCS) and immune cell score (ICS) for PD-L1 were significantly higher among patients with MEC compared with those with other types of tumors ( P =0.0011). However, the percentages of combined score (CS) for PD-L1 and tumor cell score for PD-L2 were comparable among tumor types ( P >0.05). No significant difference was found in these scores for PD-L1 between tumor stages ( P >0.05), but for PD-L2, all patients at stage I had TCS <1% for PD-L2, while all patients at stages II and III, and 92% of patients at stage IV had TCS ≥1% ( P <0.0001). High expression of PD-L1 was mostly observed in MEC cases ( P =0.0016), while all patients with AciCC had a low PD-L1 expression level ( P =0.0206). The mean tumor diameter, rate of lymphovascular invasion, perineural invasion, metastasis, positive surgical margin, recurrence, type of treatment, mortality, and TILs ratio did not differ significantly according to PD-L1 expression level ( P >0.05). The percentage of tumor-infiltrating lymphocytes was comparable among negative and positive PD-L1 scores according to both 1% and 5% threshold values ( P >0.05). High PD-L1 expression is rare in AciCC, while PD-L1 expression is high in MEC. Platelet-rich fibrin (PRF) is a widely used autologous blood concentrate in regenerative medicine. This study aimed to characterize the cellular composition and distribution of different PRF matrices generated by high (710 g) and low (44 g) relative centrifugal forces (RCFs) and to analyze their bioactivity on human primary osteoblasts (pOBs). PRF was separated into upper layer (UL) and buffy coat (BC) fractions, and their cellular contents were assessed using histological and immunohistochemical staining. The release of platelet-derived growth factor (PDGF) and transforming growth factor (TGF-β) was quantified using an ELISA. Indirect PRF treatment on pOBs was performed to evaluate cell viability and morphology. A histological analysis revealed higher quantities of leukocytes and platelets in the low-RCF PRF. TGF-β release was significantly higher in the low-RCF PRF compared to the high-RCF PRF. All PRF fractions promoted pOB proliferation regardless of the centrifugation protocol used. The low-RCF PRF showed higher TGF-β levels than the high-RCF PRF. These findings contribute to understanding the cellular mechanisms of PRF and provide insights into optimizing PRF protocols for bone regeneration, advancing regenerative medicine, and improving patient outcomes.

Today, new energy sources alternative to fossil fuels are needed to meet the increasing energy demand. It is becoming increasingly important to constitute new energy sources from waste biomass through the liquefaction process. In this study, walnut shells (WS) were liquefied catalytically and non-catalytically under different parameters using the liquefaction method. In this process, the effect of silica fume/nano zero-valent iron (SF/NZVI) catalysts on the conversion rates was investigated. The catalyst was synthesized by reducing NZVI using a liquid phase chemical reduction method on SF. The SF/NZVI catalyst was characterized by scanning electron microscopy- energy dispersive X-ray (SEM-EDX), transmission electron microscope (TEM), Brunauer-Emmett-Teller (BET), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and X-ray photoelectron spectroscopy (XPS) analysis. The effect of various process parameters on the liquefaction process was investigated. In this context, the reaction temperature ranged from 300 to 400 °C, the solid/solvent ratio ranged from 1/1 to 1/3, the reaction time ranged from 30 to 90 min, and the catalyst concentration ranged from 1 to 6%. According to the results obtained, the most suitable operating conditions for non-catalytic experiments in liquefaction of WS were found to be temperature of 400 °C, reaction time of 60 min, and solid/solvent of 1/3. In catalytic conditions, the optimum values were obtained as temperature of 375 °C, reaction time of 60 min, solid/solvent ratio of 1/3, and catalyst concentration of 6%. The highest total conversion and (oil + gas) % conversion were 90.4% and 46.7% under non-catalytic conditions and 90.7% and 62.3% under catalytic conditions, respectively. Gas chromatography/mass spectrometry (GC/MS) analysis revealed the bio-oil was mainly composed of aromatic compounds (benzene, butyl-, indane and their derivatives,) and polyaromatic compounds (naphthalene, decahydro-, cis-, naphthalene, 1-methyl-.). Purpose: Green Fluorescent Protein is widely used as a cellular marker tool, but its potential influence on cells has been questioned. Although the potential off-target effects of GFP on tumor cells have been studied to some extent, the findings at the molecular level are insufficient to explain the effect of GFP expression on the tumorigenic capacity of cancer cells. Here, we aimed to investigate the effect of GFP expression on the tumorigenicity of PC3 prostate cancer cells., Methods: Using GFP-expressing and wild-type PC-3 cells, xenograft models were generated in athymic BALB/C mice. To identify differentially expressed proteins, the change in cells proteome was investigated by label-free quantification with nano-high performance liquid chromatography to tandem mass spectrometry (nHPLC-MS/MS). Proteins that showed significantly altered expression levels were evaluated using the bioinformatics tools., Results: Unlike the wild-type PC-3 cells, GFP-expressing cells failed to develop tumor. Comparative proteome analysis of GFP-expressing cells with WT PC-3 cells revealed a total of 216 differentially regulated proteins, of which 98 were upregulated and 117 were downregulated., Conclusion: Upon GFP expression, differential changes in several pathways including the immune system, translational machinery, energy metabolism, elements of cytoskeletal and VEGF signaling pathway were observed. Therefore, care should be taken into account to prevent reporting deceitful mechanisms generated from studies utilizing GFP., (© 2024. Objective: The basis for current and future lung cancer immunotherapy depends on our knowledge of molecular mechanisms of interactions between tumor and immune system cells. Interactions that occur between different intratumoral populations of the same cells are important. In our study, we aimed to evaluate relationship between the clinical and prognostic features and T lymphocyte subgroups of patients with lung tumors after neoadjuvant treatment., Methods: A total of 72 patients were included in our study, including study group, 39 of whom received neoadjuvant chemotherapy. Clinical/radiological/pathological findings of patients and CD4/CD8 staining rates in peritumoral/intratumoral areas were recorded., Results: Our study revealed significantly lower intratumoral CD4 + T cell density and lower intratumoral CD4/CD8 ratio in primary tumor after neoadjuvant therapy (respectively, 0.012 and 0.016). Considering tumor types, when control-study groups were compared, inflammation was statistically significant only in adenocarcinoma subtype; intratumoral CD4/CD8 ratio was statistically significant only in squamous-cell carcinoma subtype (respectively, p = 0.0008 and p = 0.0139). When CD4 + T lymphocytes and CD8 + T lymphocytes and CD4/CD8 ratio were compared between control and study groups in low-stage patients according to clinical stages, only intratumoral CD4 + T lymphocyte values and intratumoral CD4/CD8 ratio were significant (respectively, p = 0.0291 ve p = 0.0154)., Conclusion: All cell types of innate and adaptive intratumoral immunity can affect lung cancer tissues simultaneously, and these interactions have a very complex structure. Understanding the tumor microenvironment and the different roles of associated cancer immune cells may lead to the discovery of new targets for immunological therapies and increased survival times in lung cancer., (© 2024. Introduction and Hypothesis: To evaluate the effect of AF219, a P2X3 receptor antagonist, in animal models of interstitial cystitis/bladder pain syndrome (IC/BPS) induced by cyclophosphamide (CYP) or water avoidance stress (WAS)., Methods: Thirty-two adult female Wistar albino rats were used in each IC/BPS model. Assessment of nociception and anxiety and severity of inflammation in the bladder were assessed by behavioral experiments and histopathological examinations respectively. The contraction responses of the bladder were evaluated in vitro and protein levels of P2X3, P2X7, Trk-A, TRPV1, and TRPA1 were analyzed by Western blot., Results: The IC/BPS groups had shorter response times to noxious stimuli, exhibited more anxiety-like behavior, had higher inflammation-based histological scores, and showed greater increased contraction responses to carbachol, adenosine triphosphate, and electrical field stimulation in in vitro bladder strips than controls for both models (p < 0.05). The improvements in behavioral and bladder contraction responses and inflammation scores in the IC/BPS + AF219 groups were similar to control findings (p > 0.05). Exposure to WAS or CYP increased P2X3 expression in the bladder compared with the controls (p < 0.05). Apart from TRPA1, the levels of P2X7, Trk-A, and TRPV1 were also higher in the IC/BPS groups than in the controls (p < 0.05). No significant differences were observed between IC/BPS + AF219 and controls regarding P2X3, P2X7, Trk-A, and TRPV1 in the WAS model (p > 0.05). Moreover, P2X3 and P2X7 levels were significantly lower in IC/BPS + AF219 than in the AF219-untreated WAS model (p < 0.05)., Conclusions: These findings suggest that P2X3 receptors play a significant role in bladder functional responses, nociception, and also the pathogenesis of IC/BPS. AF219 may be a promising therapeutic strategy for IC/BPS. Comparing AF219 with current IC/BPS treatment agents in future studies may yield valuable insights into its efficacy., (© 2024. Background: N-Nitrosomorpholine (NMO) is one of the most common N-nitroso compounds. An oncocytic transformation has been demonstrated in renal tubules of NMO-treated rats. In our study, we aimed to investigate the potential transformation of oncocytic cells in 6 endocrine organs, i.e., thyroid, adrenal and pituitary glands, pancreas, testis, and bone, of NMO-exposed rats., Methods: Thirty male rats were born and raised. Fifteen of them were given a single dose of 320 mg NMO per kg body weight, dissolved in drinking water, by a gavage tube. At the end of 52 weeks, the animals in both series were killed. Right after the killing, 6 different endocrine organs (hypophysis, thyroid, pancreas, adrenal gland, bone [femur], and testicles) of each animal were excised., Results: There was no evidence of oncocytic cell development in the control group. In contrast, oncocytes were observed in 8 out of 13 NMO-treated rats: 2 in the adrenal sections, 1 in the thyroid sections, 3 in the pituitary sections, and 2 in the pancreas sections. Thesticle and bone sections were completely normal., Conclusions: We showed that NMO induced an oncocytic change in pancreas, thyroid, pituitary, and adrenal glands. To date, no identified specific environmental risk factors that lead to an oncocytic transformation in endocrine glands have been reported previously. Given the increasing prevalence of endocrine-disrupting chemicals in the environment, personal care products, manufactured goods, and food sources, there is a need to advance our understanding of the pathological mechanisms underlying oncocytosis in endocrine organs., (© 2024. The cytologic diagnosis of acinic cell carcinoma (ACC) can be challenging due to its polymorphous appearance and sharing cytomorphologic characteristics with other benign and malignant neoplasms as well as non-neoplastic diseases, even though various histomorphological aspects of ACC have been documented. We presented a 39-year-old female patient applied with right pre-auricular parotid swelling spreading infra-auricular region which was gradually increased in size for 3 months. Ultrasonographic examination revealed hypoechoic well-circumscribed mass with 17 × 22 × 29 mm size. Magnetic resonance imaging revealed intra-parotid solid lesion with cystic areas, slightly hypointense on T1 and hyperintense on T2 weighted images. The mass was pushing the retromandibular vein medially, still lateral to it in the caudal images, but in dumbbell-shape spreading through parapharyngeal space in superiorly cranial images. Fine-needle aspiration cytology was also performed with guidance of ultrasonography. The cytological examination of the lesion was characterized by the predominance of heterogeneous lymphoid cells, clusters of epithelial cells with a variety of cytologic appearances, including granular, transparent, vacuolated, and oncocytic, and the presence of numerous naked nuclei with a protein-like foamy background. Due to intense lymphocytic inflammation, it was considered as benign primary parotid tumor such as Warthin's tumor. The excision material was examined histopathologically. Immunohistochemical analysis showed that this carcinoma was positive for DOG1, SOX10, cytokeratin 7 and negative for mammaglobin. This salivary gland tumor was reported as a rare variant of ACC with lymphoid-rich stroma.
