Abstract

To separate and optimize the bioactive fraction of Hemidesmus indicus as an antimicrobial lead using in silico QSAR model development. : Hemidesmus indicus was extracted by soxhalation. The crude extract is fractionated using solvents of different polarity and assessed for its antimicrobial activity. The bioactive fraction is further analysed by GC-MS to analyse the constituents. These moieties were studied for their molecular interaction using CoMFA based QSAR model development. : The bioactive fraction found to have antibacterial and antifungal activities. GC-MS revealed the presence of eicosane, pthalic acid, oleanen 3 -yl acetate and substituted alkanes. Force field analysis followed by simulation revealed that pthalic acid was found to have interaction with the receptors. : Thus the integration of activity guided fractionation with cheminformatics may reveal the putative leads in drug design. Pthalic acid analogues can thus be optimized further by subjecting to preclinical drug studies.

Abstract

Objective: The aim of this study was to develop a three-dimensional quantitative structure–activity relationships (3D-QSARs) model for studying theinteraction of different phytochemicals with nuclear factor kappa B (NF-ĸB) inducing kinase, a major regulator in tumor inflammation. Methods: Different phytochemicals (ligands) from Brassicaceae were selected and tested for Lipinski’s rule of five and further analyzed using interaction studies (docking) to identify the binding site in the target protein. Ligands with best fit were made to pass through ADMET filter, and the nontoxic ligands were selected based on the pIC50 values. R esults: The 3D-QSARs of the ligands were designed using comparative molecular field analysis, and glucoraphanin was found to be stable and fit after subjecting for molecular dynamics simulation with annealing studies. C onclusion: Thus, the model may be prospectively used in drug design to find possible inhibitors of NF-ĸB, which plays a key prominent role in cancer inflammation. 13. Evaluation of Heavy Metal Removal of Nanoparticles Based Adsorbent Using Danio rerio as Model.

Abstract

Nanoparticles are potential candidates for wastewater treatment especially for the removal of heavy metals due to their strong affinity. Many biopolymers are used as adsorbents and encapsulation of nanoparticle onto them can increase their efficiency. In this study, SPIONs, alginate, and SPIONs incorporated on alginate beads have been synthesized and characterized both microscopically and spectroscopically. These were then used for the removal of chromium metal and the percentage of removal was evaluated using a batch adsorption study. The percent removal of chromium using SPIONs, alginate and alginate-SPIONs beads were recorded to be 93%, 91% and 94%, respectively. The adsorption of chromium using SPIONs and alginate-SPIONs beads followed the Tempkin isotherm, whereas adsorption of chromium metal by alginate beads was found to be homogeneous in nature and followed the Langmuir isotherm with an R 2 value of 0.9784. An in-vivo study using Danio rerio as a model organism was done to examine the toxicity and the removal efficiency of the samples. It was observed that chromium water treated with alginate-SPIONs beads, which were removed after water treatment showed less damage to the fishes when compared to SPIONs and alginate beads treated with chromium water where the SPIONs and alginate beads were not removed after the treatment period.
